ABSTRACT

Psoriasis is a chronic inflammatory skin disorder in humans, and the inflammatory reaction plays an important role in development and onset of psoriasis. 4'-O-β-D-glucosyl-5-O-methylvisamminol (4GMV) is one of the major active chromones isolated from Saposhnikoviae divaricata (Turcz.) Schischk, which has been reported to exhibit excellent anti-inflammatory activities. However, the possible therapeutic effect on psoriasis and underlying mechanism has not been reported. Thus, the aim of this study was to investigate the protective effect of 4GMV on the imiquimod (IMQ)-induced psoriasis-like lesions in BALB/c mice and the anti-inflammatory effect on the lipopolysaccharide (LPS)-induced inflammation in RAW264.7 macrophages. The results demonstrated that 4GMV decreased IMQ-induced keratinocyte proliferation and inflammatory cell infiltration. Moreover, 4GMV treatment significantly inhibited the production of NO, PEG 2, and cytokines such as interleukin (IL)-1β, IL-6, interferon (IFN)-γ, and IL-22 in LPS-stimulated RAW264.7 macrophages. 4GMV also suppressed the LPS-upregulated protein expressions of iNOS and COX-2 in a dose-dependent manner. Furthermore, qRT-PCR analysis showed that 4GMV down-regulated the mRNA level of IL-1β and IL-6 expression. Further studies by western blot indicated that 4GMV inhibited the activation of upstream mediator NF-κB by suppressing the expression of TLR4 and the phosphorylation of IκBα and p65. The phosphorylation of JNK, p38, and ERK were also markedly reversed by 4GMV in LPS-treated RAW264.7 macrophages. Taken together, these results demonstrated that 4GMV showed a protective effect in IMQ-induced psoriasis-like mice and inhibited inflammation through the NF-κB and MAPK signaling pathways, indicating that 4GMV might be a potential therapeutic drug for psoriasis.

ABSTRACT

Sugar-poison caused blood-heat is the pathological basis of many complications of diabetes. Advanced glycation end products( AGEs) are considered as the potential glycotoxic factor that can cause blood-heat. Sophorae Flos hold the effect of removing pathogenic heat from blood. In this study,chromatographic non-enzymatic glycation reaction system of bovine serum albumin( BSA)/methylglyoxal( MGO) and Sophorae Flos was established to identify active components in Sophorae Flos inhibiting AGEs formation. The HPLC was used to analyze chromatograms before and after the incubation of Sophorae Flos and methylglyoxal. Changes of chromatographic peaks of eight compounds was found. It is speculated that this change may be due to new substance produced by the reaction of active components in Sophorae Flos and methylglyoxal,and these active components may be flavonoid component rutin. Further investigation for the effects of rutin and MGO reaction( 1 ∶ 1,1 ∶ 3,3 ∶ 1) for 6 days on the formation of AGEs was performed. The results showed that the inhibition activity of rutin on AGEs production was most obvious when the reaction ratio was 1 ∶3,and the most inhibition was in 24 h and stabilized after 3 d. The product of the reaction of rutin with MGO was identified by LC-ESI-MS/MS,which indicated that the newly formed seven substances were the mono-and di-MGO adducts of rutin. This study showed that rutin is the active component on Sophorae Flos for removing pathogenic heat from blood by forming new compounds to inhibit the formation of sugar poison products,which provides reference for rational application of Sophorae Flos.

ABSTRACT

Objective To assess the right ventricular function in pulmonary hypertension (PH) patients according to their tricuspid annular plane systolic excursion.Methods Ninety PH patients admitted to our hospital from October 2014 to September 2015 were divided into mild PH group (n=30),moderate PH group (n=30) and severe PH group (n=30) with 30 individuals undergoing physical examination served as a control group.Their right ventricular transverse diameter,right atrial transverse diameter,main pulmonary artery diameter,and respiration variations of inferior vena cava,tricuspid annular plane systolic excursion of right ventricular free wall were measured by ultrasonography.Results The right ventricular transverse diameter,right atrial transverse diameter,main pulmonary artery diameter were significantly longer and the suction collapse rate was significantly lower in mild PH group,moderate PH group and severe PH group than in control group (P<0.05,P<0.01).The tricuspid annular plane systolic excursion was significantly shorter in moderate PH group and severe PH group than in control group (12.9± 3.2 cm,10.1±3.1 cm vs 23.4±2.2 cm,P<0.01).However,no significant difference was found in tricuspid annular plane systolic excursion between moderate PH group and control group (P> 0.05).Conclusion The tricuspid annular plane systolic excursion is shorter and the right ventricular function is lower in PH patients.Tricuspid annular plane systolic excursion can sensitively show the right ventricular function in PH patients and is thus a reliable predictor for their right ventricular function.

ABSTRACT

Objective To compare the therapeutic efficacy between scalp acupuncture plus rehabilitation and rehabilitation in treating children epilepsy.Method Sixty children with epilepsy were enrolled and randomized into two groups, 30 cases in the treatment group and 30 cases in the control group. The treatment group was intervened by scalp acupuncture plus rehabilitation, while the control group was treated with rehabilitation. After the intervention, the changes of Comprehensive Function Assessment for Disabled Children (CFADC) and electroencephalogram (EEG) waves were observed, and the therapeutic efficacies of the two groups were compared and analyzed.Result The comprehensive function scores were changed significantly in both groups after the treatment (P<0.05). After the treatment, the comprehensive function score was (61.15±2.65) in the treatment group versus (58.60±2.81) in the control group, and the between-group difference was statistically significant (P<0.05). There was a significant difference in comparing the EEG waves between the two groups (P<0.05). In the control group, no case was recovered, 4 cases showed marked efficacy, 21 showed effective, and 5 failed in the treatment; in the treatment group, 2 cases were recovered, 10 showed marked efficacy, 16 showed effective, and 2 failed in the treatment. There was a significant difference in comparing the therapeutic efficacy between the two groups (P<0.05), and the treatment group was superior to the control group.Conclusion Scalp acupuncture plus rehabilitation can produce a satisfactory clinical efficacy in treating children epilepsy and is thus feasible.

Objective To establish an anxiety-like gastric hypersensitivity ( GHS) rat model of functional dyspep-sia induced by new sequential stress. Methods Twenty-six male 1-day-old Sprague-Dawley rat pups were randomly di-vided into control and model groups ( n=13 in each group) . The model rats received sequential stress from postnatal day 2:neonatal maternal separation (NMS), acute gastric irritation (AGI) and restraint stress (RS). The control rats were reared freely with their mothers without undergoing the sequential stress. From postnatal 8th week all rats started to receive elevated plus maze ( EPM ) , open field test ( OF ) , abdominal withdrawal reflex ( AWR ) and electromyographic test ( EMG) . Results EPM and OF experiments depicted that the model rats showed obvious anxiety-like behavior ( P<0. 05 or P<0. 01). AWR and EMG tests exhibited that the model rats had elevated gastric hypersensitivity to gastric distention (P<0. 05 or P<0. 01). Conclusions An anxiety-like GHS rat model of functional dyspepsia can be successfully estab-lished with our new method of sequential stress.

ABSTRACT

Idiosyncratic drug-induced liver injury (IDILI) is a kind of unique adverse drug reaction with relative high morbidity compared with other idiosyncratic diseases. Its occurrence, however, has nothing to do with pharmacological effects and clinical dosage of drugs administered, and only a small number of susceptible individuals will suffer from it. Especially to deserve to be mentioned, the proportion of TCM-induced IDILI showed an ascending trend year by year. So in this article, the author has reviewed some facts related with TCM-induced IDILI, including the predisposing causes and occurrence mechanism, and tries to provide reference for the prevention, diagnosis and treatment of TCM-induced IDILI through the analysis of characteristics and research status of TCM-induced IDILI and exploration of the internal relationship between Chinese medicine constitution type and IDILI.

ABSTRACT

Amino-acid neurotransmitter system dysfunction plays a major role in the pathophysiology of depression. Several studies have demonstrated the potential of amino acids as a source of neuro-specific biomarkers could be used in future diagnosis of depression. Only partial amino acids such as glycine and asparagine were determined from certain parts of rats' brain included hippocampi and cerebral cortex in previous studies. However, according to systematic biology, amino acids in different area of brain are interacted and interrelated. Hence, the determination of 34 amino acids through entire rats' brain was conducted in this study in order to demonstrate more possibilities for biomarkers of depression by discovering other potential amino acids in more areas of rats' brain. As a result, 4 amino acids (L-aspartic acid, L-glutamine, taurine and gamma-amino-n-butyric acid) among 34 were typically identified as potentially primary biomarkers of depression by data statistics. Meanwhile, an antidepressant called Fluoxetine was employed to verify other potential amino acids which were not identified by data statistics. Eventually, we found L-alpha-amino-adipic acid could also become a new potentially secondary biomarker of depression after drug validation. In conclusion, we suggested that L-aspartic acid, L-glutamine, taurine, gamma-amino-n-butyric acid and L-alpha-amino-adipic acid might become potential biomarkers for future diagnosis of depression and development of antidepressant.

ABSTRACT

<p><b>OBJECTIVE</b>To discuss surgical treatment of right colon carcinoma of hepatic flexure invading the duodenum.</p><p><b>METHODS</b>Sixty-five patients with right colon carcinoma of hepatic flexure invading the duodenum, treated in our department from 1987 to 2007, were included in this study. Their clinicopathological data were retrospectively reviewed and analyzed. All the cases were divided into three types (local invasion, regional invasion, and cancer with internal fistula) according to duodenal defect, including local invasion (< 2.0 cm), wide invasion (> 2.0 cm) and the presence of internal fistula.</p><p><b>RESULTS</b>25 patients with local invasion underwent en bloc resection of the duodenal wall. Pedicled ileal flap was used to cover the large duodenal defect measuring 2.0 - 3.0 cm in 5 patients. Dudenojejunostomy was used to reconstruct the large defect measuring more than 5 cm in 3 patients. Conservative resection of right-sided colon was performed in 18 patients with wide invasion. 4 patients underwent pancreaticoduodenectomy combined with right hemicolectomy for colon cancer involving the pancreatic head. 10 underwent duodenal diverticularization. One patient with anastomotic leakage healed within 3 weeks. Other patients were cured without postoperative complications. The total 3-year and 5-year survival rates after surgery were 53.8% and 9.2%, respectively.</p><p><b>CONCLUSION</b>The surgical procedure to be performed is usually decided according to the cancer location, extent, and duodenal defect and invasion, which are important for prolonging life time, improving of quality of life and prognosis in these patients.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To summarize the experience in the diagnosis and surgical treatment of carcinoid tumors of the appendix.</p><p><b>METHODS</b>From 1972 to 2006, 64 patients with carcinoid tumors of the appendix received surgical treatment in our hospitals. The clinical data of those patients were retrospectively analyzed.</p><p><b>RESULTS</b>Of the 64 cases, only 6 cases (9.4%) were correctly diagnosed preoperatively, while 58 (90.6%) not confirmed, with a misdiagnosis rate of 90.6%. All patients underwent surgical treatment, including appendectomy in 54, ileocecectomy in 4, right hemicolectomy in 2 and right hemicolectomy with regional lymph node dissection in 4 cases. The operation modes were determined according to the doctor's judgments based on the age of the patients, the nature, size, location, infiltration depth and lymph node metastasis of the tumors. Of the 64 patients, 58 were followed up with a longest follow-up period of 13 years, while 6 lost follow-up. Fifty-seven of those were still surviving, only one died of liver metastasis at 13 years after operation.</p><p><b>CONCLUSION</b>Carcinoid tumor of the appendix is rare with a high rate of misdiagnosis before operation. Surgical resection is the only effective treatment for this disease and proper operation mode is the key to achieve good survival.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the expressions and clinical implications of kruppel-like factor 6(KLF-6) and APC in human colorectal carcinoma.</p><p><b>METHODS</b>The expressions of KLF-6 and APC in tumor and normal tissues from 32 patients with colorectal carcinoma were investigated by RT-PCR and immunohistochemical technique.</p><p><b>RESULTS</b>The expression rates of KLF-6 and APC mRNA were 37.5% and 34.3% in tumor tissue, 96.9% and 93.8% respectively in normal tissues (both P< 0.05). The expression rates of KLF-6 and APC protein were 28.1% and 25.0% in colorectal carcinomas, 81.3% and 84.43% respectively in normal tissues (both P< 0.05). There was a significant correlation between the expressions of KLF-6 and APC in colorectal carcinomas (P < 0.05). The expressions of KLF-6 and APC were significantly correlated with tumor differentiation, depth of infiltration, lymph node metastasis and clinical stage (P< 0.05).</p><p><b>CONCLUSION</b>Down-regulations of KLF-6 and APC might play an important role in the carcinogenesis, development, metastasis of human colorectal carcinoma.</p>
