ABSTRACT

OBJECTIVE@#To explore the effect of circ-SFMBT2 on the biological behavior of non-small cell lung cancer (NSCLC) cells and its regulatory role on the miR-7-5p/ADAM10 axis.@*METHODS@#qRT-PCR and Western blotting were used to determine the expression of circ-SFMBT2, miR-7-5p, and ADAM10 in NSCLC tissues and adjacent tissues. Pearson analysis was used to analyze the correlation between circ-SFMBT2 and miR-7-5p, and between miR-7-5p and ADAM10. In vitro cultured human bronchial epithelial-like cells (HBE) and lung cancer cell lines H1650, H460, A549, H1299. CCK-8 and EdU methods were used to assess the ability of cell proliferation. Plate experiment was used to detect the clone formation ability. Flow cytometry was used to detect the apoptosis rate. Transwell experiment was used to detect cell invasion ability. Dual luciferase reporter experiment detects the targeting relationship between circ-SFMBT2 and miR-7-5p, and between miR-7-5p and ADAM10. Transplanted tumor experiment in nude mice assessed the effect of knocking down circ-SFMBT2 on the growth of transplanted tumor. Immunohistochemical experiments were performed to detect the positive rates of ADAM10 and Ki67 proteins in transplanted tumor tissues.@*RESULTS@#The expression levels of circ-SFMBT2 and ADAM10 were increased in NSCLC tissues and cell lines, while decreased the expression of miR-7-5p. circ-SFMBT2 was negatively correlated with miR-7-5p, while miR-7-5p was negatively correlated with ADAM10. Silencing the overexpression of circ-SFMBT2 and miR-7-5p could inhibit cell proliferation, clone formation and invasion, and also promote apoptosis. circ-SFMBT2 could target miR-7-5p, and ADAM10 was the target gene of miR-7-5p. The combined effect of silencing circ-SFMBT2 and inhibition of miR-7-5p, as well as miR-7-5p overexpression and ADAM10 overexpression could promote cell proliferation, clone formation and invasion, and also suppress cell apoptosis. Silencing circ-SFMBT2 could inhibit the growth of transplanted tumors.@*CONCLUSION@#Silencing circ-SFMBT2 can suppress the proliferation, clone formation, invasion ability and induce apoptosis of NSCLC cells by regulating the miR-7-5p/ADAM10 axis.

ABSTRACT

Objective To evaluate treatment and operation timing of anal fistula in acute leukemia patients.Methods Clinical data of 42 acute leukemia patients with anal fistula from 2010 to 2015 in First Hospital of Xinjiang Medical University were evaluated according to the choice of operation time.Patients were divided into two groups undergoing surgery before chemotherapy, (group A).and surgery after chemotherapy (group B).Clinical analysis was performed usingx2 or t test and correlation index.Results Neutrophil count in group B was lower than that in group A (x2 =8.077, P =0.004).Chemotherapy before surgery regime reduces the incidence of infection.Conclusions Chemotherapy before surgery may improve the prognosis of anorectal fistula patients of acut leukemia.

ABSTRACT

Objective To evaluate the clinical characters, management and prognostic factors of patients with differentiated invasive thyroid carcinoma (DITC). Methods The data were analyzed retrospectively for 114 DITC patients treated at Department of Head and Neck Surgery of Sun Yat-sen University Cancer Center. Survival analysis was performed by Kaplan-Meier method, comparison among/between groups was performed using log-rank test, and multivariate analysis was carried out using Cox proportional hazard model. Results After surgery, 68 patients were with tumor residue. The 5-year and 10-year overall survival rate were 91.9% and 80.1% respectively in all patients, while the 10-year overall survival rate were 88.5% 、78.5% and 53.1% in no tumor residue group, micro-residue group and grossresidue group respectively. This study failed to prove that radiotherapy might improve the survival rate in patients with postoperative tumor residue. Multivariate analysis indicated that age, invasion to esophagus and recurrence predict the prognosis. Conclusion DITC may be treated mainly by surgical operation. Radical resection is the key factor in the treatment of DITC. Patients with DITC have a relatively poor prognosis.Age, esophagus invasion and status of tumor residue are the most important factors affecting the prognosis.

BACKGROUND: Methods to effectively overcome re-stenosis or atresia following reconstruction of external auditory canal are explored.OBJECTIVE: To explore the effect of NiTi shape memory alloy netlike bracket to prevent the restenosis or atresia of newly constructed external ear canal in external canal atresia surgery. DESIGN, TIME AND SETTING: Case analysis of aural atresia patients from Department of Otorhinolaryngology Head and Neck Surgery, Third Hospital of Sun Yat-sen University was performed from January 2001 to June 2003. PARTICIPANTS: Eight cases of aural atresia were selected, including 5 patients with congenital aural atresia, and 3 with postnatal aural atresia. All cases had different degrees of conductive hearing loss. METHODS: All cases underwent external canals reconstruction by using NiTi shape memory alloy netlike brackets. MAIN OUTCOME MEASURES: Diameter of newly constructed aural canal and pure tone auditory threshold. RESULTS: All the eight ears had a smooth and wide external ear canal during follow up. No restenosis of canal or cicatricial contracture was found. Pure tone auditory threshold of 7 ears had improved postoperatively. CONCLUSION: NiTi shape memory alloy netlike bracket has definite effect to prevent the restenosis or atresia of new constructed external ear canal.

ABSTRACT

Objective:The therapeutical effects of salpingocatheterism and the states of tubal function were studied after different middle ear operations and within different middle ear pathological changes.Method:Total 71 patients with tubal occlusion, 18 of myringoplasty, 20 of reconstruction of ossicolar chain followed tympanoplasty, 14 of combined approach tympanoplasty, 10 of radical mastoidectomy followed tympanoplasty, 9 of re-radical mastoidectomy followed tympanoplasty were implemented salpingocatheterism. The patients were followed up for evaluation of the postoperative therapeutical effects and the state of tubal function. Result:The causes of operation failure in different middle ear operations or different middle ear pathological changes had no significantly difference. The major cause for postoperative tympanic membrane perforation and tubal occlusion was the pathogenic damage being not cleared up thoroughly and/or there being nasal cavity or pharyngeal cavity pathogenic changes accompanied.Conclusion:Salpingocatheterism is effective for preventing eustachian tube postoperative adhesion.
