ABSTRACT

Abstract Cancer is a fatal malignancy and its increasing worldwide prevalence demands the discovery of more sensitive and reliable molecular biomarkers. To investigate the GINS1 expression level and its prognostic value in distinct human cancers using a series of multi-layered in silico approach may help to establish it as a potential shared diagnostic and prognostic biomarker of different cancer subtypes. The GINS1 mRNA, protein expression, and promoter methylation were analyzed using UALCAN and Human Protein Atlas (HPA), while mRNA expression was further validated via GENT2. The potential prognostic values of GINS1 were evaluated through KM plotter. Then, cBioPortal was utilized to examine the GINS1-related genetic mutations and copy number variations (CNVs), while pathway enrichment analysis was performed using DAVID. Moreover, a correlational analysis between GINS1 expression and CD8+ T immune cells and a the construction of gene-drug interaction network was performed using TIMER, CDT, and Cytoscape. The GINS1 was found down-regulated in a single subtypes of human cancer while commonly up-regulated in 23 different other subtypes. The up-regulation of GINS1 was significantly correlated with the poor overall survival (OS) of Liver Hepatocellular Carcinoma (LIHC), Lung Adenocarcinoma (LUAD), and Kidney renal clear cell carcinoma (KIRC). The GINS1 was also found up-regulated in LIHC, LUAD, and KIRC patients of different clinicopathological features. Pathways enrichment analysis revealed the involvement of GINS1 in two diverse pathways, while few interesting correlations were also documented between GINS1 expression and its promoter methylation level, CD8+ T immune cells level, and CNVs. Moreover, we also predicted few drugs that could be used in the treatment of LIHC, LUAD, and KIRC by regulating the GINS1 expression. The expression profiling of GINS1 in the current study has suggested it a novel shared diagnostic and prognostic biomarker of LIHC, LUAD, and KIRC.


Resumo O câncer é uma doença maligna fatal e sua crescente prevalência mundial exige a descoberta de biomarcadores moleculares mais sensíveis e confiáveis. Investigar o nível de expressão de GINS1 e seu valor prognóstico em cânceres humanos distintos, usando uma série de abordagens in silico em várias camadas, pode ajudar a estabelecê-lo como um potencial biomarcador de diagnóstico e prognóstico compartilhado de diferentes subtipos de câncer. Além disso, também previmos poucos medicamentos que poderiam ser usados ​​no tratamento de LIHC, LUAD e KIRC, regulando a expressão de GINS1. O perfil de expressão de GINS1 no estudo atual sugeriu que é um novo biomarcador de diagnóstico e prognóstico compartilhado de LIHC, LUAD e KIRC.

ABSTRACT

The treatment of ST elevation myocardial infarction (STEMI) has undergone significant advances over the past three decades. Current practice guidelines raise the importance of promptly restoring normal coronary blood flow and myocardial perfusion in the infarct zone after the onset of chest pain, through either pharmacologic or mechanical reperfusion strategies. Fibrinolytic therapy remains the most widely used reperfusion strategy worldwide. With the development of newer fibrinolytic agents and adjuvant potent anti-platelets therapies, this approach carries an increased risk of bleeding complications. The current research present up-date review of the use of reperfusion strategies for the treatment of STEMI, using data through the search of MEDLINE, PubMed, EMBASE, as well as related extracts from the annual report of the American Heart Association, the American College of Cardiology, and the European Society of Cardiology. We summarized data from the available studies conducted over the past last 30 years in relation to pharmacologic reperfusion therapy in regards to risks and benefits. Conclusion: Fibrinolytic therapy remains the main reperfusion strategy used for the treatment of STMI worldwide. In the current era, there is a lack of fibrinolytic therapy trials, mainly because of increased focus in mechanical reperfusion therapies’ studies in the developed world. Clinical trials on the use of the fibrinolytics with newer platelet agents are urgently needed.

ABSTRACT

Lipase was partially purified from the culture supernatant of Mucor racemosus NRRL 3631. In an attempt to increase the enzyme stability, the enzyme was immobilized on poly (vinyl alcohol) PVA, radiation cross liked poly (vinyl alcohol/ vinyl pyrrolidone) PVA / PVP and poly (vinyl alcohol/ hydroxyethylmethacrylate) PVA/ HEMA hydrogels. The maximum immobilization yield (31.74 %) was obtained using PVA/ HEMA copolymer. The effect of the immobilization parameters on the enzyme such as the hydrogel composition, irradiation dose and the immobilization technique was performed. An optimum radiation dose of 15 kGy and hydrogel composition of 10 % PVA/ HEMA (9.6: 0.4 v/v) increased the immobilization yield to 60.3 %. Diffusion phenomena can be markedly increased the enzyme immobilization on the surface of the hydrogel. In this case the retained activity was approximately 81.5 % of that of the free enzyme. The profiles of immobilized enzyme activities at various pH values (4-9) and temperatures (30-80 °C) showed an overall higher stability for the immobilized enzyme than that for the free one. The half life values of the immobilized and free enzymes at 60 °C were 3.3 h and 1.73 h, respectively. The immobilized enzyme retained 69.2 % of its initial activity after three cycles.

ABSTRACT

Seven local fungal isolates of Pyricularia grisea were purified from infected rice leaves. The total proteins were extracted and SDS-PAGE was carried out to differentiate between the expression of proteins in infected and healthy plants. SDS-PAGE analysis revealed the accumulation of 35-kDa chitinase after 16, 20, 24 and 48 hr [hours post inoculation]. Rice chitinase gene [1.023 bp] was successfully amplified from the total RNA extracted from infected rice using RT-PCR. The amplified fragment was cloned and overexpressed in E. coli BL21 cells as 6x-His- fusion protein. Recombinant chitinase fusion protein was successfully purified using Ni-NTA affinity column chromatography. Two chitinase activity assays against P. grisea were carried by the filter disc and the dissimilar concentrations plates method. The results indicated that the expressed chitinase protein had an antifungal activity against P. grisea

ABSTRACT

Seventeen local bacterial isolates which can hydrolyze a wide range of organophosphate [OP] insecticides were purified. They were reduced to ten different isolates based on their RAPD pattern and protein profile and termed as ASM-1 through ASM-10. Chemical assay and bioassay revealed that the isolate ASM-5 was the best isolate in chlorpyrifos degradation. The morphological and molecular identification characterized ASM-5 as Agrobacterium tumefaciens. A gene encoding a protein involved in OP hydrolysis was cloned and sequenced from A. tumefaciens ASM-5. This gene [named opdA] had sequence similarity about 98.7% with the A. tumefaciens opd gene. The coding sequence of the gene was sub cloned down stream an inducible expression promoter to evaluate the gene-enzyme system responsible for its OP-hydrolyzing activity. The biological activity of OPDA protein became more efficient compared to OPDA native protein

ABSTRACT

Clinical and histopathological evaluation of acute and chronic ordinary urticaria. Eighty-two patients having ordinary urticaria, 57 [69.5%] acute and 25 [30.5%] chronic, 24 [29.27%] males and 58 [70.73%] females, had been recorded from the out patient clinic-department of dermatoIog and venerolog-Baghdad Teaching Hospital-Baghdad. Data abstracted from the patients by a questionnaire and clinical assessment. All patients were sent for laboratory investigations including, blood examination, general urine and stool examination. Antinuclear factor was carried out for those patients with chronic urticaria. The mean age of the patients was 24 +/- 4.6 years. Females were more affected than males. Regarding atopy, there was no much difference from control. Night exacerbation was noticed in ordinary urticaria 43 [52.4%]. Emotional tension 32 [39%], food 25 [30.5%], heat 25 [30.5%]. drugs 16 [19.5%] and infections 14 [17%] were the most provoking factors. Gastrointestinal disturbances were the most common associated symptoms and noticed in 23.2% of patients. These symptoms were more in acute urticaria [29.8%] when compared with chronic urticaria [8%]. Parasitic infestations were noticed in chronic urticaria [20.4%], while eosinophilia was more evident in acute urticaria [16.1%]. Positive ANF, leukocytosis, high ESR, anemia and pyuria were observed in some patients. Tuberculin testing was less positive in patients with urticaria. Acute and chronic ordinary urticaria constitute a common type of urticaria affect predominantly females. Good questionnaire and full investigations are essential for the detection of their provoking factors

ABSTRACT

The present study was carried out on 40 female albino rabbits [1 kilogram in weight] in the animal laboratory of the National Cancer Institute. General anaesthesia was administered to all rabbits. The abdomen was entered through an upper midline incision and the liver was exposed. The scalpel was used to inflict a 2 cm long wound in the liver parenchyma deep enough but short of the posterior Glisson's capsule. The rabbits were divided into 2 equal Groups; in group [1] fibrin glue made from platelets and bovine thrombin was applied to the hepatic tear; while in group [2] fibrin glue made from platelets and human thrombin was applied to the liver tear. The time needed to achieve haemostasis was recorded in each group. Both types of fibrin sealants were successful to achieve haemostasis well as healing of the liver tear. However, the time needed to achieve haemostasis was significantly shorter in group [1] [bovine thrombin group]. It was concluded that both agents could be an attractive solution in the management of hepatic injuries especially in the modern era of surgical laparoscopy
