Objective To investigate the epidemiological characteristics of varicella public health emergency events (PHEEs) in Xinwu District from 2016 to 2020, and to provide a scientific basis for formulating prevention and control measures. Methods Data was retrieved from “Public Health Emergency Management Information System” in "Chinese Disease Control Information System" and “Jiangsu Provincial Vaccine Management Information System”. Descriptive epidemiological methods were employed to analyze the data. SPSS software was used for statistical analysis. Results A total of 35 varicella PHEEs were reported in Xinwu District from 2016 to 2020, accounting for 57.38% of all PHEEs of infectious diseases in the same period. A total of 1 033 cases were reported, with an attack rate of 1.91%. The events showed a bimodal distribution. A small peak occurred from April to June, and the number of events accounted for 17.14%. A large peak occurred from November to January of the following year, and the number of events accounted for 82.86%. All events occurred in schools. Kindergartens were the most dangerous places, and the number of events accounted for 45.71%. Preschoolers aged 3-6 years accounted for 48.83% of the total cases, and the male-to-female ratio of students was 1.40:1. Among 234 cases with a history of immunization with Varicella Attenuated Live Vaccine (VarV), the longer the time interval from the date of VarV inoculation to the data of onset, the higher the proportion of varicella cases. Varicella PHEEs reporting time was positively correlated with duration (rs=0.391 , P<0.05). Conclusion Early detection, early reporting and early isolation can effectively control the duration of the event. School varicella surveillance should be strengthened during the epidemic peak period. Preschoolers are vulnerable groups. It is suggested that VarV should be included in the inspection work for enrollment in kindergartens.

Glioblastoma (GBM) is the most common primary brain tumor, which is prone to recurrence and metastasis with poor prognosis. In recent years, immunotherapy has prolonged the survival of patients with GBM, providing a new option for the treatment of GBM. Target selection is very important for immunotherapy. Epidermal growth factor receptor variant III (EGFRvIII) is highly expressed on the surface of GBM cells in some patients, and EGFRvIII was not expressed in normal tissues. EGFRvIII are pivotal for the occurrence and progression of GBM, various targeted therapy including immunotherapy is promising to improve the efficacy of GBM. Currently, there are various approaches to target EGFRvIII, including humanized monoclonal antibodies, adoptive cell therapies and therapeutic vaccines. In this review, we focus on the preclinical and clinical findings of targeting EGFRvIII for GBM.

OBJECTIVE To investigate the immunogenicity of Isatis indigotica polysaccharides, IIP-A-1 and IIP-2 as vaccine adjuvants. MRTHODS Keyhole limpet hemocyanin (KLH) was linked to IIP-A-1 and IIP-2 respectively to prepare immuno-antigen KLH-IIP-A-1 or KLH-IIP-2. Bovine serum albumin (BSA) was also linked to them respectively to prepare BSA-IIP-A-1 or BSA-IIP-2 as the detecting-antigen. Rabbits were twice immunized with KLH-IIP-A-1 or KLH-IIP-2 (300 pg per rabbit) plus Freund adjuvant (0.65 mL per rabbit) ) by intradermal injection at a 28 d interval. The plates were coated with protein KLH, BSA, BSA-IIP-A-1 or BSA-IIP-2 (5 mg.L") and serum specific antibody titers were determined by ELISA. In the sera of mice immunized with Aftosa plus IIP-A-1, anti-IIP-A-1 IgG, was not found. However, anti-IIP-2 IgG, with a titer of 1:400 was found in the serum samples of mice immunized with Aftosa or H1N1 plus IIP-2 respectively. A titer of 1 = 100 was found in the sera of immunized mice co-immunized with HBsAg. CONCLUSION As haptens, IIP-A-1 and IIP-2 could induce the production of antibodies against IIP-A-1 and IIP-2 after being linked to KLH. As an adjuvant, IIP-A-1 doesn't present any immunogenicity, while IIP-2 can induce the production of anti-IIP-2 IgG, in mice when co-immunized with Aftosa, H1N1 or HbsAg.

Objective: To compare the clinical effectiveness and safety of axillary vein catheterization [AVC] and scalp vein catheterization [SVC] in the neonatal intensive care unit [NICU]

Design: Retrospective study

Setting: The Central Hospital of Xinxiang, China

Subjects: One hundred and fifteen cases from the NICU were enrolled from June 2012 to June 2013

Interventions: Fifty-four patients underwent AVC and 61 underwent SVC

Main outcome measure[s]: Catheter-related bloodstream infection, catheter obstruction, ectopia, phlebitis, and local leaking

Results: No significant difference between groups was observed for catheter related bloodstream infection [SVC: 4.4%, AVC: 3.7%, p = 0.56] and catheter obstruction [9.8% vs. 9.3%, P = 0.87] between the two catheterization methods. However, the rate of catheter ectopia, phlebitis and leaking in SVC were significantly higher than those for AVC [18.0% Vs. 3.7%, p = 0.02, 18.0% Vs. 3.7%, p = 0.02 and 16.4% Vs. 3.7%, p = 0.03, respectively]

Conclusions: Although both SVC and AVC could be the choice for intravenous infusion in the NICU, AVC was safer than SVC in newborn infants

Objective: To investigate the inhibition of cell proliferation and induction of apoptosis of flavonoids [diosmetin-7-O-β-D-xylopyranosyl-(1 → 6)-β-D-glucopyranoside, DXG)] from Gallium verum on liver cancer cell HepG2 and to study its mechanisms. Methods: Cell growth was detected with Trypan blue staining. The proliferation inhibition rates of DXG on HepG2 were measured by MTT assay. The morphology changes of apoptotic cell were obserbed by AO-EB double staining. Agarose gel electrophoresis was used to detect the DNA fragmentation. The expression of apoptosis-related genes bax and bcl-2 was examined by RT-PCR. Results: DXG (50, 100, and 200 μg/mL) could significantly inhibit the proliferation of HepG2 cells by Trypan blue staining and MTT assay compared with the control group (P < 0.05). AO-EB double staining showed that DXG could induce the apoptosis and morphological change of HepG2 cells and the ratios of apoptosis were obviously high when the concentration of DXG increased. The laddering pattern was clearly presented in the cells treated with 100 and 200 μg/mL DXG for 48 h. RT-PCR showed that DXG could up-regulate the mRNA level of bax and down-regulate the mRNA levels of bcl-2 in HepG2 cells. Conclusion: DXG could obviously inhibit the proliferation and induce the apoptosis in HepG2 cells. The mechanism is related to the modulation of the expression level of bax/bcl-2 mRNA.

<p><b>OBJECTIVE</b>To observe the effect of Yiqixue Buganshen recipe(, YBR) on the expression of integrin ανβ3 in the endometrium of controlled ovarian hyperstimulation mice.</p><p><b>METHODS</b>A total of 180 mice were divided into three groups: model group, treatment group and control group. The treatment and model groups were intraperitoneally injected with gonadotropin-releasing hormone analogue for 7 days; pregnant mare serum gonadotropin was also injected on the 7th day. After 48 h, human chorionic gonadotropin was injected. The control group was injected with an equal volume of saline at the same time. From the start of the experiment, the treatment group was intragastrically administered Jinghouzengzhi Recipe() and Cuhuangti Recipe(). The model group and the control group were intragastrically administered an equal volume of saline. Real-time reverse transcription polymerase chain reaction and Western blotting were used to detect the mRNA and protein expression of integrin ανβ3 in mouse endometrium.</p><p><b>RESULTS</b>Integrin ανβ3 was expressed in mouse endometrium in all groups. Integrin αββ3 expression increased gradually along with pregnancy, progressing from pregnant day (Pd) 1. Integrin ανβ3 expression significantly increased on Pd 4, then began to decrease on Pd 6. Integrin ανβ3 expression in the treatment group was higher than in the model group, and the difference was statistically significant (P <0.05). The difference between the treatment group and the control group was not statistically significant (P >0.05).</p><p><b>CONCLUSION</b>YBR improves endometrial receptivity, and may play an important role in embryonic implantation.</p>

<p><b>OBJECTIVE</b>To establish a detection method based on gas chromatography-mass spectrometry (GC-MS) for concentrations of volatile nitrosamine compounds in urine, and apply it to the test of real samples.</p><p><b>METHODS</b>Target compounds dichloromethane in urine samples was extracted with dichloromethane through liquid-liquid extraction, then the dichloromethane extract was filtrated, evaporated with nitrogen at 40°C to dryness, and the volume was set with 0.2 ml dichloromethane. Analysis of nine volatile nitroso-compounds were performed with GC-MS under selected ion monitoring mode, external reference method was used for quantification, and the detection limit, repeatability and sensitivity were evaluated. In addition, nine volatile nitroso-compounds of 92 urine samples in a town of Anhui province were measured.</p><p><b>RESULTS</b>A good linear range of 2 - 200 ng/ml (with correlation coefficient 0.9985 - 0.9999) were obtained for the above mentioned nine kinds of analyte, and the lowest examination concentration was 0.05 - 0.50 ng/ml. The addition standard recoveries were 68%-102% with the RSD of 0.4% - 5.5% (n = 3). The detection limits were 0.001 - 0.013 ng/ml urine. The detection rate of N-nitrosodimethylamine (NDMA), N-nitrosomethylethylamine (NMEA), N-nitrosodiethylamine (NDEA), N-nitrosodi-n-propylamine (NDPA), N-nitrosopyrrolidine (NPYR), N-nitrosomorpholine (NMOR), N-nitrosopiperidine (NPIP), N-nitrosodi-n-butylamine (NDBA) and N-nitrosodiphenylamine (NDPhA) were 71% (65), 74% (68), 65% (60), 80% (73), 92% (85), 78% (72), 76% (70), 87% (80), 98% (90), respectively, with the results (0.27 ± 0.12), (0.75 ± 0.29), (0.06 ± 0.02), (0.16 ± 0.07), (23.66 ± 5.18), (1.01 ± 0.35), (0.38 ± 0.11), (2.47 ± 0.52) and (15.13 ± 3.48) nmol/g creatinine.</p><p><b>CONCLUSIONS</b>A gas chromatography-mass spectrometry detect method was developed for low level volatile nitrosamines in urine samples.</p>

<p><b>OBJECTIVE</b>To evaluate the antitumour efficacy of shRNA plasmid specifically targeting Jab1 gene.</p><p><b>METHODS</b>The nude mouse tumor model was made by subcutaneous injection of human laryngeal carcinoma Hep-2 cells. The tumor growth was monitored after intratumoral injection of pJab1, pKB plasmids and saline. Jab1 and p27 expressions in tumour tissues were examined by immunohistochemistry staining and RT-PCR.</p><p><b>RESULTS</b>Mean volume of the pJab1-treated tumors was (267.60 ± 88.19) mm(3), significantly less than that of tumors treated with pKB plasmids (832.20 ± 140.39) mm(3) or saline (895.40 ± 145.93) mm(3) (F = 36.73, P < 0.001). Immunohistochemistry showed that the expression of Jab1 protein was significantly reduced in the pJab1-treated group (32.40% ± 5.59%) compared to the control groups, whereas the expression rate of p27 protein in the pJab1 group (76.80% ± 6.30%) was significantly increased compared to the control groups (P < 0.001). The down regulation of Jab1 protein by pJab1 plasmid was consistent with mRNA expression confirmed by RT-PCR. The level of Jab1 mRNA level in the pJab1-treated group (0.65 ± 0.03) was significantly lower than the control groups (F = 558.00, P < 0.001), however, p27 mRNA, was 0.80 ± 0.02, had no significant alteration (F = 1.52,P > 0.05).</p><p><b>CONCLUSIONS</b>The pJab1 plasmid results in downregulation of Jab1 in an xenograft tumour model of human laryngeal carcinoma Hep-2 cells and significantly inhibits the tumour growth in vivo. This suggests that pJab1 plasmid specifically targeting Jab1 gene expression could be an effective therapy for human laryngeal carcinoma.</p>

<p><b>BACKGROUND</b>The equations for estimating glomerular filtration rate (GFR) based on creatinine have been found to have limitations and have not been generalizable across all populations. Equations based on cystatin C provide an alternative method to estimate GFR. Whether the equation based on cystatin C alone or combined creatinine would improve GFR estimates has not been validated among Chinese patients with chronic kidney disease (CKD) and diabetes. The aim of this study was to compare the performance of the modification of diet in renal disease (MDRD) equation based on creatinine with the five cystatin C-based formulae for estimation of GFR in patients with CKD and diabetes.</p><p><b>METHODS</b>A total of 166 patients with CKD and 91 patients with type 2 diabetes were enrolled in this study. Cystatin C was measured by using the particle-enhanced immunonephelometric method and estimated formulae proposed by five different investigator teams (Stevens, Ma, Rule, Macisaac and Perkins). The plasma clearance of (99m)Tc-DTPA was determined as measured GFR (mGFR).</p><p><b>RESULTS</b>For CKD patients, the bias and accuracy for the Ma and Macisaac equations were superior compared with the MDRD, and the mean results for the Ma formula were closer to mGFR than the other equations in CKD stages 2 - 5. The differences between Macisaac and mGFR in CKD stages 2 - 4 were significantly less than those in CKD stage 1 or 5. Stevens and Rule's formulae revealed a similar bias and accuracy compared with the MDRD equation. The MDRD formula had a higher accuracy in CKD stages 3 - 5 as compared with the results in other stages. For diabetic patients, the mean results between Macisaac and mGFR were closer than those of other equations in mGFR >or= 90 mlxmin(-1)x1.73 m(-2) stage. In GFR 60 - 89 mlxmin(-1)x1.73 m(-2) stage, the MDRD formula showed the smallest difference compared with other equations. All equations overestimated GFR in the cases with GFR < 60 mlxmin(-1)x1.73 m(-2) stages. The MDRD formula had a greater accuracy within 50% of mGFR than the equations based on cystatin C in diabetic patients. Perkins formula showed a large positive bias and low accuracy, therefore it may not be suitable for assessing GFR in patients with CKD and diabetes.</p><p><b>CONCLUSIONS</b>The formulae for estimating GFR based on cystatin C or creatinine have different trends and accuracies in patients with CKD and diabetes, especially in patients with various GFR levels. The equations based on cystatin C provide less accurate results than MDRD formulae, at least in the diabetic patients. Therefore, whether the formulae based on cystatin C are superior to MDRD formula requires further investigation in large diverse populations.</p>

<p><b>OBJECTIVE</b>To investigate the genetic polymorphisms and their forensic application of 9 non-combined of DNA index system (CODIS) short tandem repeat(STR) loci in Guangdong Han population.</p><p><b>METHODS</b>DNA samples from 500 unrelated individuals were extracted and amplified with fluorescence labeled multiplex PCR system. PCR products were separated and genotyped with capillary electrophoresis.</p><p><b>RESULTS</b>One hundred and fifteen alleles and 160 genotypes were observed in the 9 STR loci, respectively. The heterozygosity was 0.824-0.884, the discrimination power (DP) was 0.925-0.969 and the polymorphism information content (PIC) was 0.77-0.86, respectively. The distribution met the Hardy-Weinberg equilibrium (P > 0.05). The total discrimination power was 1.00 x 10(-13), the combined probability of exclusion for trio-paternity testing was 0.999989488. The combined probability of exclusion for duo-paternity testing was 0.873436.</p><p><b>CONCLUSION</b>The 9 STR loci are powerful and reliable for personal identification and paternity testing. They can be used as supplementary loci in fatherless (motherless) testing or cases with mutation events.</p>

To establish a dynamic connecting and comprehensive thinking mode as well. They also pointed out that to integration of traditional Chinese and Western medicine is the necessity for the development of society and science. More achievements of integrative medicine are expectable in future.

Objective To evaluate the performance of Cys C results among two detection system.Methods The particle-enhanced immunonephrometic assay was used in Dade Behring BNII. Immunoturbic assay was used in Hitachi 7170 to evaluate the JING' YUAN reagents.We compared the precison,linearity,interference,correlation,and calibrators agreement with Dade Behring BNII.Results The total CV of the samples that contain 0.6-5.0 mg/L was less than 10%.The Dade Behring and JING'YUAN method showed good linearity.Haemoglobin(10 g/L),Bilirubin(300 mg/L), Vitamin C(5 g/L)in the tested sample had no significant interference in the assay(interference 0.05) between JING' YUAN and Dade Behring reagents.Values were slightly lower than that from the Dade Behring BNII method,the mean bias was-0.16.The bias range was 1.1%-23% between JING'YUAN and Dade Behring for one sample.Conclusions The precision,linearity and interference test were suitable for routine Cys C measurement on automated biochemistry analyzer,but results has bias.

Objective To compare the coherence of serum creatinine,creatinine clearance(Ccr), Cystatin C,and estimated glomerular filtration rate(eGFR)in each stage of chronic kidney disease(CKD) patients.Methods Creatinine in serum and urine were determined by Jaffe method;serum Cystatin C was measured by particle enhanced turbidimetric method,while eGFR was calculated using the abbreviated Modification of Diet in Renal Disease(MDRD)equation which was mainly based on the serum creatinine concentration.According to the American national kidney foundation-Kidney Disease Outcome Quality Initiative(NKF-K/DOQI)guideline,all cases were grouped by eGFR into 5 stages.Results In these 228 cases,as eGFR decreased gradually,the average levels of creatinine and Cystatin C increased,while Ccr decreased.The level of each items showed a statistic difference among each stage(P0.05);in eGFR 60-89 ml/min group,the average level of creatinine was 83.3 ?mol/L,the abnormal rate was only 6.8%,it was not a sensitive marker to detect the slightly damaged GFR,the levels of Ccr and Cystatin C showed a marginal decrease and increase,with an abnormal rates of 70% and 86%,there was a statistic difference among the three abnormal rates(P

Objective To investigate plasma levels of total carnitine (TC) and free camitine (FC) in patients undergoing hemodialysis and peritoneal dialysis.Methods 200 cases of normal group came from physical examination in this hospital,all testing cases were the in-hospital patients in the department of nephropathy.TC and FC were determined by use of an enzymatic cycle assay on Hitachi 7170 automatic biochemical analyzer.Results In 200 cases of normal group,TC level was (56.52?9.61) ?mol/L,and FC was (46.60?8.23) ?mol/L.In 37 hemodialysis patients,TC and FC levels were (41.47?13.22) ?mol/L and (24.58?8.91)?mol/L before dialysis,a statistic difference was observed against the control group (P0.05).Conclusions Carnitine deficiency was seen in most patients undergoing hemodialysis and peritoneal dialysis.Furthermore,the deficiency status got worse along with the dialysis course in hemodialysis patients.Carnitine infusion can effectively improve the status of these patients.

Objective To investigate the efficacy of using a calibrator with values assigned with the reference method for improving the comparability of serum gamma-glutamyltransferase (GGT) measurements.Methods The IFCC reference method for GGT was established and the performance was verified by testing a certified reference material (CRM).A calibrator was prepared and its value for GGT was assigned with the reference method.Forty serum samples were measured on different (including HITACHI 7600,7060,7170,7180 and BECKMAN LX20,OLYMPUS AU 400) chemistry analyzers with Zhongsheng GGT reagent kits calibrated with the calibrator.The samples were also measured on the same analyzers using a theoretical factor.Biases of results obtained with the calibration and with the theoretical factor based calculation were compared.Results The reference method resuhs on the CRM agreed the certified value within the stated uncertainty.Serum results calculated from the theoretical factor showed various biases and inter-analyzer variations.When the analyzers were calibrated with the calibrator,the number of results with biases less than 10% became significantly higher and those with biases more than 20% significantly lower.The variation of the results on 5 serum samples was reduced from 11.0%~14.0% to less than 5% by using the calibrator.Conclusion Accuracy and comparability of GGT measurements with of ZhongSheng GGT kits can be improved by using a calibrator that has a value assigned with the reference method.
