ABSTRACT

Objective:Auxiliary Verse on Drugs and methods for Zang-Fu Organs (Fuxingjue Wuzangyongyao Fayao) written by TAO Hong-jing from the Liang dynasty covered many contents from a missing prescription book Classic of Decoction (Tangye Jingfa), including a map revealing the compatibility principle of Chinese herbs, namely the Tangye Jingfatu. Represented by a centrosymmetric pentagon, the map describes a unique theoretical system for deficiency-excess syndrome differentiation of five Zang organs (liver, heart, spleen, lung and kidney) and the tonification-purgation and compatibility theory of five flavors (pungent, salt, sweet, sour and bitter). Each Zang organ fixedly corresponds to one "property" flavor (purgation), one "function" flavor (tonification) and one "transformation" flavor (harmonization) resulting from the combination of the former two. For example, the liver can be purged by sour, tonified by pungent, and moderated by sweet transformed by the combination of sour with pungent. The heart can be purged by bitter, tonified by salt, and astringed by sour transformed by the combination of bitter with salt. The spleen can be purged by pungent, tonified by sweet, and dried by bitter transformed by the interaction between pungent and sweet. The lung can be purged by salt, tonified by sour, and dispersed by pungent transformed by the combination of salt with sour. The kidney can be purged by sweet, tonified by bitter, and moistened by salt transformed by the combination of sweet with bitter. This study selected appropriate mathematical tools to analyze the fixed relationship between "property" flavor, "function" flavor and "transformation" flavor among the five Zang organs in “Tangye Jingfatu” and establish a mathematical model revealing the compatibility-transformation relationship among five flavors. Method:Based on the group representation of five elements (wood, fire, earth, metal, and water), the correlations of "property" flavor, "function" flavor, and "transformation" flavor with five elements’ generation-restriction were deduced based on matrix calculation and group theory. The three-dimensional calculation method for vector product was expanded for establishing the mathematical operator of five flavors' compatibility-transformation. Result:and Conclusion:There is a mapping relationship of the purging, tonifying, and harmonizing functions represented by the "property" flavor, "function" flavor, and "transformation" flavor of the five zang organs in the "Tangye Jingfatu" with the five elements' generation-restriction. The established mathematical operator contributes to explaining the fixed collocations of five flavor transformation. Based on such algorithm, the tonifying and purging characteristics of five flavors in 10 representative classic prescriptions have been clearly expounded.

ABSTRACT

Objective::To observe the intervention effect of Yiqi Huoxue recipe (YQHX) on ventricular remodeling in rats with chronic heart failure, in order to explore its mechanism. Method::Among 40 male SD rats, 10 were randomly selected as the sham operation group. The left anterior descending coronary artery ligation was performed to construct the chronic heart failure(CHF) rat model. After modeling, they were randomly divided into model group, captopril group(13.5 mg·kg-1·d-1) and YQHX group (20 g·kg-1·d-1), and orally given the corresponding drugs. After 8 weeks of intervention, cardiac tissues were collected, body mass and heart mass were weighed, and echocardiography were performed to detect the changes in cardiac structure. Masson staining was performed to determine the myocardial interstitial collagen volume fraction. Western blot was used to detect the expression levels of mitochondrial fusion protein optic atrophy 1 (Opa1) and cleavage protein dynamic-related protein 1 (Drpl). The quantitative real-time fluorescence polymerase chain reaction(Real-time PCR)was applied to detect the expressions of Wnt/β-catenin pathway-related factors such as lipoprotein receptor-related protein 6 (LRP6), glycogen synthase kinase-3β (GSK-3β) and β-catenin. Result::Compared with the sham group, the left ventricular wall of the model group was significantly thickened (P<0.05), the cardiac cavity was significantly enlarged, and the content of collagen in the myocardial interstitium was increased (P<0.01). The expression level of Opal decreased, the expression level of Drp1 increased (P<0.05), the mRNA expression level of LRP6, GSK-3, and β-catenin increased (P<0.01). Compared with the model group, YQHX group can reduce ventricular wall thickening, heart chamber enlargement, myocardial interstitial collagen content, up-regulate the low expression of Opa1, but down-regulate the high expressions of Drpl, LRP6, GSK-3β, β-catenin(P<0.05, P<0.01). Conclusion::YQHX can effectively alleviate ventricular remodeling and improve mitochondrial energy metabolism in rats with CHF. The mechanism may be related to the inhibition of Wnt/β-catenin related factors.

ABSTRACT

As a common soft tissue disease, the mechanism of tendinopathy has not been clarified and is lack of effective treatment method. Change of tissue fibrosis is the one of the main pathological features. Transforming growth factor beta 1 (TGF-β1), which is one of the important factor, participated in fibrosis. Inconsonant expressions of TGF-β1 could be found in tendinopathy. The studies are still controversial, but the vast majority of studies had showed that TGF-β1 was abnormal, and it is given priority to increase, which means that TGF-β1 plays an important role in the process of tendinopathy. In the process of tendon injuries and repairs, the time of TGF-β1 increasing is inconsistent. The time for TGF-β1 plays a significant role has not been determined. TGF-β1 has abnormal expressions in both tendinopathy and tendon repairs, which are two opposite processes. Thus, it may not be a one-way adjustment factor, but has a pleiotropic. Recent studies showed that TGF-β1 was considered as binding to receptor and transferring signal into the cell. Now there are three different receptors are found. The classical pathway of TGF-β1 in intracellular signal transduction is mainly through activation of Smad pathway. In the same time, there are also some non-classical pathways. TGF-β1 could break balance of extracellular matrix, which may be a reason to cause tendinopathy. But the regulations of TGF-β1 on the extracellular matrix are complex and diverse, further studies are required. Existing researches showed that the performance of treatments on tendinopathy is unsatisfied by blocking TGF-β1 downstream pathway. Therefore, it is a good way to study the upstream mechanism of produce TGF-β1. It may be an effective method to find new targets to inhibit the development of tendinopathy better by finding the original source of TGF-β1.

ABSTRACT

<p><b>OBJECTIVE</b>To study the clinical effect and mechanism of hemoperfusion (HP) in the treatment of children with severe abdominal Henoch-Schönlein purpura (HSP).</p><p><b>METHODS</b>A total of 24 children with severe abdominal HSP were divided into two groups: conventional treatment and HP (n=12 each). Ten healthy children who underwent physical examination were enrolled as the control group. Before and after treatment, chemiluminescence was used to measure the serum levels of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α); thiobarbituric acid colorimetry was used to measure the plasma level of malondialdehyde (MDA); the hydroxylamine method was used to measure the plasma level of superoxide dismutase (SOD); chemical colorimetry was used to measure the plasma level of total anti-oxidant capability (T-AOC).</p><p><b>RESULTS</b>Compared with the control group, the conventional treatment and HP groups had significantly higher IL-6, TNF-α, and MDA levels and significantly lower SOD and T-AOC levels before treatment (P<0.05), but there were no significant differences between the conventional treatment and HP groups (P>0.05). After treatment, the conventional treatment and HP groups had significant reductions in IL-6, TNF-α, and MDA levels and significant increases in SOD and T-AOC levels (P<0.05). The HP group had significantly greater changes than the conventional treatment group; however, there were still significant differences in these indices between the HP and control groups (P<0.05). Compared with the HP group, the conventional treatment group had a significantly lower percentage of children with disappearance of digestive tract symptoms at 4 days after treatment and significantly longer time to disappearance of rash and digestive tract symptoms (P<0.05). Compared with the conventional treatment group, the HP group had a significantly lower amount of glucocorticoid used during treatment and a significantly lower percentage of children who experienced hematuria and/or proteinuria within 6 months of the disease course (P<0.05). There were no significant differences between the two groups in length of hospital stay and recurrence rates of rash and abdominal pain within 6 months of the disease course.</p><p><b>CONCLUSIONS</b>HP can reduce the amount of glucocorticoid used during treatment and the incidence rate of kidney injury in children with severe abdominal HSP, possibly by eliminating IL-6, TNF-α, and MDA.</p>

ABSTRACT

Angiogenesis involves the occurrence and development of serious diseases such as cardiovascular and cerebrovascular diseases and malignant tumors. Promoting or inhibiting angiogenesis is considered to be a key factor in controlling the progression of the disease. Chinese materia medica (CMM) has become a research hotspot because of its good clinical therapeutic effect and significant intervention on angiogenesis. The zebrafish model as an ideal animal model also provides a good experimental basis for the study of angiogenesis. CMM monomers, extracts, prescriptions and Chinese patent medicine, and other forms of intervention were selected as the research object. Based on zebrafish animal model experiments, we summarize domestic and foreign relevant research and review the research progress of the effect of CMM on angiogenesis in zebrafish, aiming to provide reference for further development and application of zebrafish as a model and further study of CMM intervention in angiogenesis.

ABSTRACT

We report a case of a 3-year-old boy with dysplasia epiphysealis hemimelica [DEH] who presented with a hard painless swelling on the medial side of his left ankle joint. Plain X-rays, CT and MRI showed a bony tumor and associated soft-tissue abnormalities. At age one year, he had been diagnosed with an osteochondroma and underwent incomplete surgical excision at a local hospital. Due to joint impingement and restricted movement, we chose to completely excise the tumor. Pathologically, the specimen showed evidence of an osteochondroma. From its clinical, imaging, and pathological features and reports in the medical literature, a diagnosis of DEH was established. DEH is a rare variant of osteochondroma, resulting in many patients being incorrectly diagnosed or diagnosed with osteochondroma alone. It is important to low-grade malignant tumor or osteochondroma-like parosteal osteosarcoma from DEH of the talus, especially when accompanied by soft tissue abnormalities. Surgical treatment is mandatory in patients with symptoms such as pain, joint impingement and deformation. Incomplete excision of an articular lesion may lead to recurrence and additional surgery, making it less than complete excision

ABSTRACT

<p><b>OBJECTIVE</b>Tuberculosis remains a severe public health issue, and the Beijing family of mycobacterium tuberculosis (M. tuberculosis) is widespread in East Asia, especially in some areas in China, like Beijing and Tianjin. This study aimed at determining the mutation patterns of drug-resistant Beijing strains of M. tuberculosis isolated from Tianjin, China.</p><p><b>METHODS</b>A total of 822 M. tuberculosis isolates were screened for drug resistance by an absolute concentration method and the genotype was identified by PCR. 169 drug-resistant isolates of the Beijing family were analyzed for the potential mutations in the rpoB, katG, inhA promoter region and in rpsL, rrs and embB genes, which are associated with resistance to rifampin (RFP), isoniazid (INH), streptomycin (SM) and ethambutol (EMB) respectively by PCR and DNA sequencing.</p><p><b>RESULTS</b>Fifty-eight out of 63 RFP-resistant isolates were found to carry the mutations within the 81-bp RFP resistance determining region (RRDR) of the rpoB gene and the most frequent mutations occurred at codon 531 (44.4%), 526 (28.6%), and 516 (7.9%) respectively. 16 mutation patterns affecting 12 different codons around the RRDR of rpoB were found. Of 116 INH-resistant isolates, 56 (48.3%) had the mutation of katG 315 (AGC-->ACC) (Ser-->Thr), 3 (2.6%) carried S315N (AGC-->AAC) and 27 (16.0%) had the mutation of inhA-15A-->T. 84 out of 122 SM-resistant isolates (68.9%) displayed mutations at the codons 43 or 88 with AAG-->AGG (Lys-->Arg) of the rpsL gene and 22 (18.0%) with the mutations at positions 513A-->C, 516C-->T or 905 A-->G in the rrs gene. Of 34 EMB-resistant isolates, 6 had mutation with M306V (ATG-->GTG), 3 with M306I (ATG-->ATT), 1 with M306I (ATG-->ATA), 1 with D328Y (GAT-->TAT), 1 with V348L (GTC-->CTC), and 1 with G406S (GGC-->AGC) in the embB gene.</p><p><b>CONCLUSION</b>These novel findings extended our understanding of resistance-related mutations in the Beijing strains of M. tuberculosis and may provide a scientific basis for development of new strategies for diagnosis and control of tuberculosis in China and other countries where Beijing strains are prevalent.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effects on the expression of mGluR5 mRNA and protein levels in primarily cultured hippocampal neurons after lead exposure.</p><p><b>METHODS</b>Primary embryonic rat hippocampal neuronal culture was prepared. On the 3(rd) day of incubation, lead chloride solution was added into medium to produce four different lead exposure levels: 0, 1 x 10(-8) mol/L, 1 x 10(-6) mol/L, 1 x 10(-4) mol/L Pb(2+). After 10 days of incubation, the neurons were collected to measure the alteration of mGluR5 mRNA expression by real-time fluorescent quantity PCR and the expression of mGluR5 in protein level by Western blot.</p><p><b>RESULTS</b>The studies revealed that mGluR5 mRNA expression was down-regulated after lead exposure in a dose-dependent manner. The mGluR5 mRNA expression of the lower lead-exposed neurons (Pb(2+) 10(-8) mol/L), the medium lead-exposed neurons(Pb(2+) 10(-6) mol/L), the higher lead-exposed neurons(Pb(2+) 10(-4) mol/L) were 0.724, 0.421, 0.321 times less than that of the controls, respectively. The Western blot demonstrated that mGluR5 expression in protein level should be decreased after lead exposure.</p><p><b>CONCLUSION</b>The expression of mGluR5 in mRNA and protein levels should be down-regulated after lead exposure at different lead levels in a dose-dependent manner.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the status of mercury exposure of neonates and their mothers in Zhoushan City, and analyze the related influencing factors.</p><p><b>METHODS</b>From Aug to Sep, 2004, 408 neonate-mother pairs were cluster sampled at the Third People's Hospital in Dinghai region, Zhoushan City, and a further investigation for the neurobehavioral development of these children was followed up in a year. The contains of mercury of 14 kinds of fish, 5 pieces of each kind bought in markets of Dinghai region were detected.</p><p><b>RESULTS</b>The geometric mean of cord-blood Hg was 27.81 nmol/L (5.58 microg/L), while that of hair Hg of mothers was 1246.56 microg/kg. 69.9% of pregnant women had surpassed the RfD (reference dose, 0.1 microg/kg/day) for Hg intake. There was a strong correlation between hair and cord blood Hg levels (r = 0.821). The factors influencing cord-blood Hg levels were fish dinners, dwelling time, and fathers smoking or not, while those of hair Hg were fish dinners and dwelling time. The correlation between fish dinners and the cord blood Hg as well as hair Hg were of significance (r = 0.539, 0.481 respectively), the Hg levels were higher in more fish dinners (H = 119.805, 94.384 respectively). The mean fish Hg level at Dinghai region was low (< 0.1 mg/kg), and no samples were higher than 0.5 mg/L. Two samples of cutlassfish and dried eel contained higher Hg than the others, belonging to a middle level (0.1-0.5 mg/kg).</p><p><b>CONCLUSIONS</b>Hg levels in neonates and pregnant women were generally below those considered hazardous, but the safety margin was narrow for some women and supporting efforts should be made to reduce mercury exposure. The main resource of Hg was from eating fish, and it is necessary to investigate the Hg levels in large samples, as to developing the fish advisories for susceptible people.</p>

ABSTRACT

Objective To study the effects of hVEGF on the expression of fms-like tyrosine kinase-1(flt-1)and thrombospondin 1(TSP-1)during the healing process of rats' full-thickness cutaneous defect which covered by genetically membrane planted by fibroblast integrated with hVEGF recombinant. Methods Wounds with full-thickness cutaneous defect were produced on the dorsum of the SD rats.Eighty rats were divided into four groups at random,20 rats each. For the rats in the experimented group A,the wounds were covered with hVEGF genetically engineering membrane and Tegaderm membrane. In the control group B,the wounds were covered with membrane planted by the fibroblasts integrated with blank plasmid and Tegaderm membrane. In the control group C,the wounds were covered with blank membrane and Tegaderm membrane. In control group D,the wounds were only covered with Tegadern membrane.Specimens were obtained at the 3,7,14 and 29 days after injury, immunohistochemistry and image analysis were used to observe the expression of flt-1 and TSP-1.Results The expression of flt-1 in vascular endothelial cells was increased obviously in the experimental group. There was significant difference between the experimental group and other groups at 3,7 and 14d after injury(P

ABSTRACT

Objective To investigate the effect of positive pressure ventilation on radiosensitivity of the patients suffering from primary lung cancer. Methods Thirty cases of lung cancer were randomly divided into two groups: ① combining therapy group: patients treated with posi tive pressure ventilation using BiPAP respirator and radiotherapy;② simple radi otherapy group. The changes of PaO2, SaO2, blood white cells and the cellula r immunological function were observed before and after treatment. Resul ts Nasal or naso-facial positive pressure ventilation with BiPAP respi rator increased PaO2 significantly with the maximum of 7.5 kPa, SaO2 was ma inteined at above 95%. No significant change for the cellular immunological func tion was found in the combining therapy group (P>0.05)and only one patient with leukopenia(6.7%). But in the radiotherapy group, the lymphocyte transfor mation efficiency and the ratio of CD4/CD8 were obviously decreased(P<0 .01), and leukopenia was found in 5 cases(33.3%). Conclusion Nasal or naso-facial positive pressure ventilation with BiPAP respirator could improve the effect of radiotherapy for lung cancers. It could also increase PaO 2 significantly and has protective rote to the marrow and cellular immunologic al function of the patients.
