ABSTRACT

ObjectiveTo investigate the effect of Zhishi Xiebai Guizhitang (ZXGT) on isoproterenol (ISO)-induced myocardial infarction (MI) in rats through the tumor necrosis factor/nuclear factor-κB (TNF/NF-κB) signaling pathway. MethodForty-eight SD rats were randomly divided into control group (blank), model group, perindopril group (4 mg·kg-1), ZXGT group (24.4 g·kg-1), ZXGT +inhibitor group (ZXGT, 24.4 g·kg-1, TNF-α receptor inhibitor R7050, 5 mg·kg-1), and an inhibitor group (R7050, 5 mg·kg-1), with eight rats in each group. The rats in each group were orally administered with their respective drugs for 7 days. Additionally, in the ZXGT + inhibitor group and the inhibitor group, R7050 was injected intraperitoneally at a dose of 5 mg·kg-1 on the 6th and 7th days. Except for the control group, all other groups were given intraperitoneal injections of ISO for 2 consecutive days to induce MI in rats. On the 7th day of the experiment, the rats were anesthetized 30 min after ISO injection, and their electrocardiograms (ECGs) were recorded to observe ST-segment elevation. Small animal echocardiography was used to measure global longitudinal strain (GLS) and cardiac synchrony. Blood samples were collected from the abdominal aorta to measure the levels of serum cardiac troponin T (cTnT), creatine kinase-MB (CK-MB), lactate dehydrogenase (LDH), tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β). Histopathological changes in myocardial tissue were observed using hematoxylin-eosin (HE) staining. Immunohistochemistry (IHC) was used to detect the expression of TNF-α, NF-κB p65, and p-NF-κB p65 proteins in myocardial tissue. Western blot was performed to measure the expression of tumor necrosis factor receptor 1 (TNFR1), tumor necrosis factor receptor-associated factor 2 (TRAF2), transforming growth factor-beta-activated kinase 1 (TAK1), NF-κB inhibitory protein alpha (IκBα), phosphorylated (p)-IκBα, NF-κB p65, and p-NF-κB p65 proteins in myocardial tissue. ResultCompared with the control group, the model group showed significant ST segment elevation on the ECG (P<0.01), increased GLS, and reduced cardiac synchrony on echocardiography (P<0.01). Histopathological examination revealed extensive myocardial necrosis. Furthermore, the serum levels of cTnT, CK-MB, LDH, TNF-α, and IL-1β were significantly increased (P<0.01), and the expression levels of TNF-α, TNFR1, TRAF2, TAK1, p-IκBα, and p-NF-κB p65 proteins in myocardial tissue were significantly elevated (P<0.01), while the expression level of IκBα was significantly decreased (P<0.01). Compared with the model group, the perindopril group, the ZXGT group, the ZXGT + inhibitor group, and the inhibitor group rats showed a significant reduction in ST-segment elevation on the ECG (P<0.05, P<0.01), improvement in GLS and cardiac synchrony (P<0.05, P<0.01), a decrease in the area of myocardial necrosis, and reduced serum levels of cTnT, CK-MB, LDH, TNF-α, and IL-1β (P<0.01). Additionally, the ZXGT group, the ZXGT + inhibitor group, and the inhibitor group downregulated the increased TNF-α, TNFR1, TRAF2, TAK1, p-IκBα, and p-NF-κB p65 protein expression levels and upregulated IκBα expression levels in the myocardial tissue (P<0.05, P<0.01). No significant differences were observed between the ZXGT group and the ZXGT + inhibitor group or the inhibitor group. ConclusionZXGT can protect against ISO-induced myocardial injury in rats and improve cardiac function, and its mechanism of action may be related to the regulation of the TNF/NF-κB signaling pathway.

ABSTRACT

In this research,a new phospholipid based monolith was fabricated by in situ co-polymerization of 1-dodecanoyl-2-(11-methacrylamidoundecanoyl)-sn-glycero-3-phosphoethanolamine and ethylene dimethacrylate to mimick bio-membrane environment.Excellent physicochemical properties of this novel monolith that were achieved included column efficiency,stability,and permeability.Moreover,the biomimetic monolith showed outstanding separation capability for a series of intact proteins and small molecules.In particular,it exhibited good potential as an alternative to the commercial immobilized artificial membrane(IAM)column(IAM.PC.DD2)for studying drug-membrane interactions.This study not only enriched the types of IAM stationary phases,but also provided a simple model for the prediction of phosphatidylethanolamine related properties of drug candidates.

ABSTRACT

Zwitterionic sulfobetaine-based monolithic stationary phases have attracted increasing attention for their use in hydrophilic interaction chromatography.In this study,a novel hydrophilic polymeric monolith was fabricated through photo-initiated copolymerization of 3-(3-vinyl-1-imidazolio)-l-propanesulfonate(SBVI)with pentaerythritol triacrylate using methanol and tetrahydrofuran as the porogenic system.Notably,the duration for the preparation of this novel monolith was as little as 5 min,which was significantly shorter than that required for previously reported sulfobetaine-based monoliths prepared via conventional thermally initiated copolymerization.Moreover,these monoliths showed good morphology,permeability,porosity(62.4%),mechanical strength(over 15 MPa),column efficiency(51,230 plates/m),and reproducibility(relative standard deviations for all analytes were lower than 4.6%).Mechanistic studies indicated that strong hydrophilic and negative electrostatic interactions might be responsible for the retention of polar analytes on the zwitterionic SBVI-based monolith.In particular,the resulting monolith exhibited good anti-protein adhesion ability and low nonspecific protein adsorption.These excellent features seem to favor its application in bioanalysis.Therefore,the novel zwitterionic sulfobetaine-based monolith was successfully employed for the highly selective separation of small bioactive compounds and the efficient enrichment of N-glycopeptides from complex samples.In this study,we prepared a novel zwitterionic sulfobetaine-based monolith with good performance and developed a simpler and faster method for preparation of zwitterionic monoliths.

ABSTRACT

Intraoperative hypothermia can lead to a lot of complications during peri-operative period, that may affect the prognoses of patients and has attracted clinical doctors and nurses to pay attention to. During the operation period, proper hypothermia can reduce tissue metabolism, reduce oxygen consumption, enhance tolerance to injury and protect the body; while excessive hypothermia can increase the incidences of wound infection, acidosis, blood coagulation dysfunction, arrhythmia and other complications, that may seriously threaten the life and health of operative patients. In this article, the causes of excessive hypothermia during operation were mainly analyzed, effectively monitoring temperature changes in intra-operative patients was proposed and carried out to find out the main causes leading to the body temperature lowering, and then comprehensive, reasonable and safe nursing methods were applied to effectively prevent the occurrence of excessive hypothermia during operation, so that the hospitalization time can be shortened.

ABSTRACT

Objective To investigate the effect of transitional care of physical and mental health of the thoracic spinal cord injury and paraplegic patients discharged on its recovery. Methods Randomized controlled analysis of patients in Spine and Spinal Cord Injury Surgery between June 2014 to October 2015, a total of 65 patients with thoracic spinal cord injury and paraplegic were studied, according to hospital mantissa odd and even numbers divided into observation group (35 cases) and control group (30 cases). The observation group was instituted to extending posthospital care, not the control group. The self-care ability and psychological state were evaluated by Function Independent Measure (FIM), Hamilton Anxiety Scale(HAMA), Hamilton Depression Scale (HAMD)within discharge and in 3 months. And the incidence of complications between the two groups were compared. Results At discharge, the difference of FIM, HAMA, HAMD score between the observation group and the control group was not statistically significant (P>0.05). Discharged after 3 months, the score of FIM, HAMA, HAMD in observation group was(72.35 ± 4.35),(4.80 ± 4.50),(12.13 ± 7.25)points, (63.95 ± 5.02),(10.15 ± 4.91), (19.48 ± 8.31)points in control group. The difference between the two groups was statistically significant (t=-4.798, 3.524, 3.110, P 0.05); The incidence of urinary tract infection, pressure ulcers, constipation were 8.57% (3/35), 5.71% (2/35), 17.14% (6/35) in observation group, 40.00% (12/30), 33.33%(10/30), 63.33%(19/30) in control group, there were significant differences between, two groups (χ2= 4.324, 4.219, 5.337, P < 0.05). Conclusions Transitional care can significantly improve the psychological state of patients with thoracic spinal cord injury and paraplegia discharged, reduce the incidence of complications and improve quality of life.

ABSTRACT

Objective:To investigate the clinical value of electrocardiogram in risk stratification of patients with non-ST segment elevation acute coronary syndrome(ACS).Methods: According to the change of electrocardiogram, 260 patients with non-ST segment elevation ACS were divided into four groups: ECG-undiagnosed group (n=22), T-wave inversion group (ST-segment depression group), ST-segment depression group (n=118) and normal electrocardiogram group(n=80). All patients were treated with conventional 18-lead electrocardiogram and troponin I(TnI)detection in the four groups. The incidence of cardiovascular events during and after hospitalization was observed. Results: The complex cardiovascular events and recurrent angina attacks in ST segment depression group increased significantly than that in normal ECG group. The difference was statistically significant (x2=5.723,x2=15.680;P<0.05). Multivariate logistic regression analysis showed that TnI positive factors predict the independent value of composite cardiovascular events higher.Conclusion:The patients with ST segment elevation acute coronary syndrome can effectively carry out risk stratification by ECG ST-segment changes and TnI levels, and predict the cardiovascular events. It has higher clinical application value and independent predictive value of TnI positive.

ABSTRACT

BACKGROUND:Hip segmentation based on CT image has been widely used in computer-assisted surgery planning, prosthesis design and finite element analysis. OBJECTIVE:To explore application effects of automatic segmentation method for hip joint based on Bayesian Decision Theory in computer-assisted hip surgery. METHODS:An accurate outer surface segmentation and extraction remain chal enging due to deformed shapes and extremely narrow inter-bone regions. In this paper, we present an automatic, fast and accurate approach for segmentation of femoral head and proximal acetabulum. The outline of the femur was segmented and extracted by contrast enhancement, thresholding algorithm and region growth algorithm. The boundaries of the bone regions are further refined based on Bayes decision rule. RESULTS AND CONCLUSION:Automatic segmentation method for hip joint based on Bayesian Decision Theory is an accurate segmentation technique for femoral head and proximal acetabulum and it can be applied in computer-assisted hip surgery and prosthesis design.

ABSTRACT

ObjectiveTo investigate the effect of arterial stiffness on systolic deformation in hypertensive disease.MethodsSixty essential hypertensive patients were enrolled, including 25 cases with left ventricular normal geometric (group LVN) and 35 cases with left ventricular hypertrophy (group LVH) in the Affiliated Hospital of Qingdao University during July 2013 to March 2014. Thirty patients in the control group were enrolled in the same period. The peak systolic strains and strain rates were determined by using velocity vector imaging. Stroke volume was obtained by using real-time three-dimensional echocardiography. And pulse pressure/stroke volume was used as a surrogate index of arterial stiffness. Pulse pressure/stroke volume, the differences of strain and strain rate in three groups were compared by analysis of variance, and SNK-q test was used for further comparison between two groups. Multiple linear regression was performed to estimate predictors for systolic longitudinal deformation. Pearson?s correlation was used to analysis the relevance of systolic longitudinal strain and body mass index, triglyceride, left ventricular ejection fraction, age, left ventricular mass index, pulse pressure/stroke volume.ResultsPulse pressure/stroke volume were (1.26±0.45) mmHg·m2·ml-1, (1.53±0.59) mmHg·m2·ml-1, (1.82±0.43) mmHg·m2·ml-1 (1 mmHg=0.133 kPa) in the control group, LVN, LVH respectively. The systolic strains and strain rates in the control group, LVN, LVH were recorded as follows:systolic longitudinal strains were (23.60±1.94)%, (19.69±2.56)%, (17.34±2.48)%, the systolic longitudinal strain rates were (1.64±0.17) s-1, (1.52±0.14) s-1, (1.38±0.18) s-1; the systolic radial strains were (28.69±5.2)%, (30.81±4.14)%, (26.53±3.50)%, the systolic radial strain rates were (2.51±0.56) s-1, (2.60±0.45) s-1, (2.00±0.41) s-1; the circumferential strains were (24.50±5.21)%, (24.01±4.60)%, (21.00±3.70)%, the circumferential strain rates were (1.38±0.38) s-1, (1.30±0.30) s-1, (1.10±0.26) s-1. Pulse pressure/stroke volume was higher in LVN and was more pronounced in the LVH group compared with the control (LVN/LVH with the control group:q=2.90, 6.56, LVN with LVH:q=3.22, allP<0.05). The strains and strain rates in LVH were lower than those of LVN and the control group, and the differences were statistically significant. (longitudinal strains:q=15.22, 5.43; longitudinal strain rates:q=8.88, 4.54; radial strains:q=2.85, 5.36; radial strain rates:q=6.10, 6.81; circumferential strains:q=4.42, 3.61; circumferential strain rates:q=5.04, 3.42; allP<0.05). The strains and strain rates in LVN were lower than the normal group, the signiifcant differences of the longitudinal strains and longitudinal strain rates were found (q=8.73, 3.77, bothP<0.05) while there were no statistically signiifcant differences of radial strains and radial strain rates, circumferential rates and circumferential strain rates. In a multivariate analysis, LVMI and AS were found to be predictors for systolic longitudinal strain. Body mass index, triglyceride, left ventricular ejection fraction, age, left ventricular mass index and pulse pressure/stroke volume were negatively related to systolic longitudinal strain (r=-0.10,-0.09,-0.14,-0.42,-0.56, allP<0.05) by Pearson?s correlation, while LVEF was positively related to mean systolic longitudinal strain (r=0.13,P<0.05).ConclusionArterial stiffness is suitable as an predictor for left ventricular systolic deformation in hypertensive disease.

Objective:To study therapeutic effect of cilazapril and amlodipine on QT interval dispersion and arrhyth-mia in patients with essential hypertension (EH) .Methods:A total of 120 EH patients complicated with ventricular arrhythmia in our hospital from Mar 2011 to Mar 2014 were enrolled ,randomly and equally divided into cilazapril group and amlodipine group ,the two groups all received routine antihypertensive therapy .Blood pressure ,QT in-terval and arrhythmias were compared between two groups .Results:Compared with amlodipine group ,there were significant reductions in 24h mean systolic blood pressure [24hSBP ,(123.81 ± 17.95) mmHg vs .(104.92 ± 14.25) mmHg] ,24h mean diastolic blood pressure [24hDBP ,(88.31 ± 10.28) mmHg vs .(72.24 ± 8.45) mmHg] ,24hSBP variability [ (11.62 ± 1.34) mmHg vs .(7.34 ± 0.97) mmHg] ,24hDBP variability [ (9.47 ± 1.18) mmHg vs .(5.13 ± 0.72) mmHg] ,QT interval [ (385.1 ± 50.4) ms vs .(342.7 ± 48.5) ms] ,corrected QT interval [ (362.7 ± 50.2) ms vs .(321.8 ± 45.1) ms] and QT interval dispersion [ (34.1 ± 5.1) ms vs .(22.5 ± 3.7) ms] in cilazapril group , P<0.05 or <0.01;ventricular arrhythmia class in cilazapril group was significantly better than that of amlodipine group , P<0. 05. Conclusion:Therapeutic effect of cilazapril is more helpful to reduce blood pressure ,QT interval dispersion and improve ventricular arrhythmia than amlodipine in EH patients complicated with arrhythmia .

AIM:Todiscusstherelationshipbetweensmallubiquitin-relatedmodifier4(SUMO4)expression and papillary thyroid carcinoma (PTC).METHODS:The mRNA and protein levels of SUMO4 in PTC and normal thyroid tissues were determined by the methods of real-time PCR, Western blot and immunohistochemistry .The relationship be-tween SUMO4 expression and clinicopathologic parameters in PTC was evaluated .RESULTS:The results showed that both the mRNA and protein levels of SUMO 4 expression in PTC were obviously higher than those in normal thyroid tissues ( P<0.01).CONCLUSION:The high level of SUMO4 expression in PTC suggests that SUMO4 plays an important role in PTC development and it is probably a molecular mechanism of PTC .

ABSTRACT

The physiological and bio-marker function of D-acidic amino acids is now becoming the hot topic on metabolomics study and new drug discovery. A fully automated two-dimensional high performance liquid chromatography (2D-HPLC) system was established by using monolithic ODS column as the first dimension column, acetonitrile-trifluoro acetic acid-water (9: 0. 05: 92, V/ V) as the mobile phase; micro Chiralpak QD-1-AX column as the enantiomer separation column, 10 mmol/ L citric acid in methanol-acetonitrile (50: 50, V/ V) as the mobile phase for the second dimension, 4-fluoro-7-nitro-2,1,3-benzoxadiazole (NBD-F) as the fluorometrical derivative reagent. The separation efficiency ( Rs > 2. 5), determination sensitivity ( LOD =1 fmol) of acidic amino acids enantiomers were higher than those of existing methods, and an online confirmation of the enantiomers amounts was also achieved using this system. The recoveries were around 97-104% , RSD values for intra-day and inter-day precision were less than 5% for the acidic amino acids enantiomers in the biological samples. Furthermore, by analyzing the aging model senescence accelerated mouse prone 1 (SAMP1) mice which have low immunocompetence, the amounts of D-aspartic acid in thymus and spleen were determined as (206±18) and (264±21) nmol/ g, respectively. It is the first time that an obvious trend of the increasement of D-aspartic acid (p<0. 01) was observed in thymus and spleen of SAMP1 mice compare to senescence accelerated mouse resistant 1 (SAMR1) mice.

ABSTRACT

Objective To investigate the cross-talk between extracellular signal-regulated kinase (ERK) and nuclear factor κB (NF-κB) signal transduction pathways in A375 human melanoma cells.Methods Cultured A375 cells were randomly divided into 5 groups:control group receiving no treatment,two U0126 (a selective inhibitor of the ERK signaling pathway) groups treated with U0126 of 10 and 5 μmol/L,and two BMS-345541 groups treated with BMS-345541 of 10 and 5 μmol/L.After 24-hour treatment,Western blot and reverse transcription PCR were performed to measure the protein expressions of NF-κB P65,phosphorylated IκBα (p-IκBα),ERK1/2,as well as p-ERK1/2,and the mRNA expressions of NF-κB P65 and ERK1,respectively.One-way analysis of variance and least significant difference (LSD)-t test were carried out for statistical analysis.Results After 24 hours of treatment with U0126 of 10 and 5 μmol/L,a significant decrease was noted in the relative expression level of NF-κB p65 protein (0.60 ± 0.04 and 0.56 ± 0.06 vs.1.54 ± 0.15,both P< 0.01) and mRNA (0.79 ± 0.05 and 0.75 ± 0.04 vs.0.86 ± 0.05,both P < 0.01),but a statistical increase in that of p-IκBα protein (0.90 ± 0.05 and 0.70 ± 0.02 vs.0.61 ± 0.03,both P < 0.01) in the two U0126 groups compared with the control group; significant differences were observed in the expression level of p-IκBo protein (P < 0.01) but not in that of NF-κB p65 protein (P > 0.01) between the two U0126 groups.The relative expression levels of ERK1/2 and p-ERK1/2 proteins as well as ERK1 mRNA were significantly higher in the control A375 cells than those in the cells treated with BMS-345541 of 10 μmol/L (0.73 ± 0.07,0.75 ± 0.09,1.51 ± 0.02,all P < 0.01),but similar to those treated with BMS-345541 of 5 μmol/L (0.94 ± 0.11,0.99 ± 0.04,1.62 ± 0.03,all P > 0.05).Conclusion There is a cross-talk between ERK and NF-κB signal transduction pathways in A375 melanoma cells.

ABSTRACT

An integrated segmentation method for 3D ultrasound carotid artery was proposed. 3D ultrasound image was sliced into transverse, coronal and sagittal 2D images on the carotid bifurcation point. Then, the three images were processed respectively, and the carotid artery contours and thickness were obtained finally. This paper tries to overcome the disadvantages of current computer aided diagnosis method, such as high computational complexity, easily introduced subjective errors et al. The proposed method could get the carotid artery overall information rapidly, accurately and completely. It could be transplanted into clinical usage for atherosclerosis diagnosis and prevention.

ABSTRACT

Objective: To study the characteristics of QT interval dynamicity in patients with heart failure (HF) and provide reference for predicting risk of sudden death in HF patients. Methods: A total of 80 inpatients with chronic HF from our department of cardiology were selected as HF group, and another 50 healthy volunteers were enrolled as healthy control group. All subjects received 24h ambulatory electrocardiography (ECG), surface ECG and echocardiography examination. Left ventricular ejection fraction (LVEF), left ventricular end-diastolic volume (LVEDV), left ventricular end-systolic volume (LVESV), QT dispersion (QTd), index for heart rate variability-- standard deviation of normal to normal RR intervals calculated over the 24 h period (SDNN) and slope of QT/RR correlated linear were measured. Results: Compared with healthy control group, there were significant increase in LVESV [(34.0±8.3) ml vs. (90.4±15.4) ml], LVEDV [(86.0±32.2) ml vs. (150.3±30.4) ml], QTd [(35.6±8.5)ms vs.(46.6±10.4)ms], slopes of QTe /RR [(0.136±0.021) vs. (0.175±0.023)] and QTp/RR [(0.130±0.026) vs. (0.158± 0.033)], and significant decrease in LVEF [(60.6±8.4) % vs. (38.5±8.8) %], and SDNN [(140.3±53.3)ms vs.(100.4±40.3)ms] in HF group, P<0.01 all.. Conclusions: Instability of ventricular repolarization in patients with heart failure makes their QT/RR slope significantly higher than that of normal subjects.

[Methods] By the pulse diagnosis, discern the five kings of psychological disorders, including irritable, anxiety, palpitation due to fright, depressed, and an medical record of professor Qi Xianghua is cited.[Results] The syndrome differentiation should be based on the five kings of psy-chological disorders.[Conclusions] Professor Qi Xianghua's five psychological disorders of TCM on treating insomnia are worth to extend and apply.

@#ObjectiveTo explore the effects of high fat diet and caloric restriction on brain aging as well as the activity of Acetylcholinesterase(AChE) and afford scientific evidence to rational diet and prevent brain aging.MethodsSixty male ICR mice were randomly divided into 6 groups: the D-galactose-induced brain aging, brain aging plus high fat diet, brain aging plus caloric restriction, high fat diet only, caloric restriction only and normal control groups. Mice were given 100 mg/kg·d subcutaneous injection of D-galactose to prepare brain aging model for 9 weeks. Morris water maze (MWM) test was employed to determine their spatial learning and memory ability. Acetylcholinesterase (AChE) activity in brain was determined by hydroxylaminecolorimetric assay.ResultsIn Morris water maze test, brain aging mice showed a significant longer escape latency than the normal control mice (P<0.05). There was no statistical difference in escape latency between brain aging mice plus high fat diet and brain aging mice groups (P>0.05), and between the control and high fat diet groups (P>0.05). Brain aging mice plus caloric restriction exhibited a significant shorter escape latency than brain aging mice (P<0.05), but no difference was found when compared with normal control mice (P>0.05). There were no statistical difference in escape latency between the controls and caloric restriction group (P>0.05). The AChE activity in brain aging, brain aging plus high fat diet and brain aging plus caloric restriction group were higher than those in control and caloric restriction group (P<0.05). There were no statistical difference in AChE activity between the controls and caloric restriction group (P>0.05). Brain aging plus high fat diet were higher than brain aging and other non model control groups.ConclusionHigh fat diet can raise the activity of AChE effectively, but can not influence the capacity of learning and memory in mice. Caloric restriction can improve the capacity of learning and memory in mice, but has no significant influence on the activity of AChE in brain.

ABSTRACT

Objective: To explore the underlying mechanism by which acupuncture regulates the peripheral leukocyte count and observe the relationship between the effects of electroacupuncture (EA) and the spleen ultrastructure in leukopenia. Methods: Leukopenia models of rabbits were established by injecting cyclophosphamide (CY) into rabbits' ear vein,and then the rabbits were treated with acupuncture. The peripheral leukocyte count and classification were measured daily. At last, after the animals were anesthesized, the abdominal cavity was opened, and a piece of spleen tissue was cut. The diameter of splenic sinusoid basal lamina eyehole was measured under electric microscope. Results: The peripheral blood leukocyte count in the EA group increased significantly with shift to right of the granulocyte nuclei compared with model control group (P<0.01). Moreover, the calibers of splenic sinusoid basal lamina eyehole in the EA group were larger than those in the model control group (P<0.05). Conclusion: Acupuncture can enhance the peripheral leukocyte count by promoting spleen activity in the early phase of leukopenia.
