ABSTRACT

Objective:It introduced the application of bayesian network in real-world data and the implementation of Tetrad software, so as to provide methodological application reference for medical researchers.Methods:Real-world data for diabetes diagnosis had been taken as an example. UCI machine learning database Pima-Indian-diabetes data set was used as case data, Bayesian network ratiocination was carried out according to the construction order of bayesian network of Tetrad software.Results:When the screening age was over 30 years old, impaired glucose tolerance, abnormal serum insulin two hours after meal, body weight index and family genetic index were large, the probability of diabetes onset from 34.99% to 83.33%.Conclusions:The bayesian network realized by Tetrad software provides a powerful tool for causal inference of real-world data, and promoted the objective and quantitative interpretation of the dependence between variables.

ABSTRACT

Objective To evaluate the effect of silencing ACAT1 gene on colon cancer cells proliferation,migration,invasion and colon cancer development by using the small interference RNA (siRNA) in colon cancer cell line HT-29.Methods Acyl coenzyme A cholesterol acyltransferase 1 (ACAT1) gene was silenced in HT-29 cell lines using Hiperfect transfection reagent.The expression level of ACAT1 was detected by real time PCR.CFSE and transwell assays were used to evaluate the effect of ACAT1 gene interfering on cells proliferation,mi gration and invasion.Result ACAT1 mRNA expression decreased obviously after siRNA interference.Compared with pre-transfection,proliferation,migration and invasion of colon cancer cells have been significantly inhibited (P < 0.05).Conclusion ACAT1 gene interference reduced proliferation,migration and of invasion of HT29 cells,which provide a new potential target for colon cancer treatment.

ABSTRACT

Objective To investigate up-regulation of lncRNA-AK058003 expression in SK-Hep1 hepatocellular carcinoma cells and its effect on the growth of human hepatocellular carcinoma xenograft tumor in nude mice.Methods The recombinant plasmid of lncRNA-AK058003 and empty plasmid were transferred into SK-Hep1 human hepatocellular carcinoma cell line by the method of slow virus transfection.The overexpression of lncRNA-AK058003 cell line was screened by neomycin.Overexpression of lncRNA-AK058003 in SK-Hep1 hepatocarcinoma cells was detected by quantitative PCR.SK-Hep1 hepatocellular carcinoma cells stably overexpressing lncRNA-AK058003 and SK-Hep1 hepatocellular carcinoma cells containing empty plasmids were injected subcutaneously into armpit of nude mice.The long and short diameters of the transplanted tumor were measured regularly to calculate the volume of the xenograft tumor.After 35 days of feeding we measure the weight of the xenograft tumor.Results The recombinant plasmid of lncRNA-AK058003 was successfully constructed and stably over expressed in hepatocellular carcinoma cells.The experimental results of subcutaneous tumor bearing nude mice showed that the volume(308.4 ± 439.4mm3),weight (0.464 ± 0.518g)and growth rate of the xenograft tumor in the over expression group were lower than that of the empty plasmid group(1410.0 ± 973.0mm3,1.363 ± 0.856g,P < 0.05).Conclusion LncRNA-AK058003 has inhibitory effect on the proliferation of human hepatocellular carcinoma xenograft tumor in nude miee.

ABSTRACT

<p><b>OBJECTIVE</b>This article aims to introduce the benefits of qualitative research and to discuss how such research can be applied to the study of polycystic ovary syndrome (PCOS).</p><p><b>DATA SOURCES</b>Relevant articles were published in English as of May 2013 from Pubmed. Terms "polycystic ovary syndrome/PCOS, qualitative research and methodology" were used for searching.</p><p><b>STUDY SELECTION</b>Articles studying PCOS with qualitative methods were reviewed. Articles associated with the use of qualitative research in clinical research were cited.</p><p><b>RESULTS</b>Six qualitative studies related to PCOS were found in the literature search. These studies addressed different aspects in PCOS women including their womanhood, lived experience, information need, and experience of treatment with acupuncture. Five of these six studies used phenomenology as guiding theory.</p><p><b>CONCLUSION</b>Quantitative research has been the dominant approach in the field so far, qualitative research is relevant to the advancement of PCOS.</p>

ABSTRACT

Objective To investigate the effects of paravertebral injection of different concentrations of adriamycin on motor conduction function in rats. Methods Eighty healthy male Wistar rats weighing 250-300 gwere randomly divided into 4 groups (n = 20 each): 3 adriamycin groups receiving paravertebral injection of 0.3%, 0.5% and 1.0% adriamycin 10 μl respectively (group A1, A2, A3) and control group (group C)receiving equal volume of normal saline (NS) instead of adriamycin. The animals were anesthetized with intraperitoneal 3% pentobarbital 40 mg/kg. A right paramedian incision was made in the back from L3 to S1.L4,5,5,6 intervertebral foramina were exposed. 0.3%, 0.5% and 1.0% adriamycin and NS 10 pl were injected into the intervertebral foramina in group A1, A2, A3 and C respectively. Spinal motor evoked pontential (SMEP)was measured at 1, 2, 4 and 8 weeks (T1-4)after paravertebral injection. Results The latent period of SMEP was significantly prolonged and the amplitude decreased at T1-4 in group A3 as compared with group A1, A2 and C.Conclusion Paravertebral injection of 1.0% adriamycin can significantly depress motor conduction function.
