ABSTRACT

ObjectiveTo observe the effect of acupuncture “Zhibian (BL 54)-to-Shuidao (ST 28)” on the reproductive function of asthenospermia model mice and to explore the possible mechanism. MethodsForty-two male C57BL/6 mice were randomly divided into blank group, model group, and acupuncture group, with 14 mice in each group. Cyclophosphamide 30 mg/(kg·d) was injected intraperitoneally for 7 days to establish model of asthenospermia for the model group and the acupuncture group, while 0.9% sodium chloride solution 10 ml/(kg·d) was injected intraperitoneally for 7 days in the blank group. After successful modeling, mice in the acupuncture group received acupuncture at “Zhibian (BL 54)-to-Shuidao (ST 28)” once a day for 20 minutes, 6 times a week for 3 weeks; the remaining two groups were fixed without acupuncture. Daily observations were made on the general conditions of mice in all groups, and changes in body weight after intervention were recorded. On the next day after completing the treatment, 6 male mice selected randomly from each group to test the sperm quality as well as testicular and epididymal weights, and calculate the corresponding indices; ELISA was used to test the levels of testosterone (T), follicle-stimulating hormone (FSH), and luteinizing hormone (LH) in serum; HE staining and TUNEL staining were performed to observe the pathological morphology and apoptosis of testicular and epididymal tissues; fluorescence quantitative PCR and western blot were used to detect changes in the expression of apoptosis-related factors (Fas), Fas-associated death domain protein (FADD), B-cell lymphoma 2 (Bcl-2), Bcl-2-associated X protein (Bax), and cysteine-aspartic acid protease (caspase)-3 mRNA and protein in testicular tissue. The remaining 8 male mice in each group were housed with estrus-cycling mice of the same strain at 1∶1 ratio, and the pregnancy rate and number of embryos per litter in each group were determined after mating. ResultsIn comparison with the blank group, mice in the model group exhibited reduced body weight, decreased testicular mass, testicular index, epididymal mass, and epididymal index. Additionally, there was a decrease in total sperm count, sperm motility, and sperm viability. Serum levels of T, FSH, and LH were reduced. The apoptosis rate increased, and the expression levels of Fas, FADD, Bax, and Caspase-3 mRNA and proteins were elevated, while Bcl-2 mRNA and protein expression levels decreased (P<0.01). Pathological abnormalities were observed in testicular and epididymal tissues, with disrupted arrangement of seminiferous tubules and a decreased number of spermatogenic cells within the tubular lumen. Furthermore, the pregnancy rate and the number of embryos per litter decreased significantly after mating (P<0.01). In comparison with the model group, mice in the acupuncture group showed improvements in testicular mass, testicular index, epididymal mass, epididymal index, total sperm count, sperm motility, and sperm viability. Serum levels of T, FSH, and LH increased. The apoptosis rate decreased, and the expression levels of Fas, FADD, Bax, and Caspase-3 mRNA and proteins decreased, while Bcl-2 mRNA and protein expression levels increased (P<0.05 or P<0.01). Morphological improvements were observed in testicular and epididymal tissues, with a compact arrangement of seminiferous tubules and an increased number of spermatogenic cells within the tubular lumen. Furthermore, the pregnancy rate and the number of embryos per litter increased significantly after mating (P<0.01). ConclusionAcupuncture “Zhibian (BL 54)-to-Shuidao (ST 28)” can improve testicular tissue apoptosis and enhance reproductive function in a mouse model of asthenospermia. Its mechanism may be associated with the modulation of key factors in the extrinsic membrane receptor pathway (Fas-mediated pathway) and the intrinsic mitochondrial pathway (Bcl-2/Bax-regulated pathway) in testicular tissue.

ABSTRACT

During interventional procedures,subjects are exposed to direct and scattered X-rays.Establishing diagnostic reference levels is an ideal way to optimize the radiation dose and reduce radiation hazard.In recent years,diagnostic reference levels in interventional radiology have been established in different countries.However,because of the too many indicators for characterizing the radiation dose,the indicators used to establish diagnostic reference levels vary in different countries.The research achievements in this field remain to be reviewed.We carried out a retrospective analysis of the definition,establishment method,application,and main factors influencing the dose difference of the diagnostic reference level,aiming to provide a basis for establishing the diagnostic reference level for interventional procedures in China.

ABSTRACT

Objective:To compare clinical characteristics and results on parameters for muscle measurement in elderly sarcopenia patients of different age groups.Methods:This was a cross-sectional study, and 404 outpatients aged 60 years and over were enrolled and received a long-term follow-up.According to the World Health Organization classification of the elderly, the subjects were divided into a youngest-old group, a middle old group and an oldest-old group; and according to the diagnostic criteria of the Asian Working Group for Sarcopenia in 2019, elderly people of all ages were divided into a sarcopenia group and a non-sarcopenia group.General clinical data were recorded, and the upper arm circumference, leg circumference, skeletal muscle index, grip strength and walking speed, the Short Physical Performance Battery and timed up and go were conducted.At the same time, the body composition was measured by multifrequency bioelectrical impedance analysis.Results:The subjects had a mean age of(85.24 ± 8.23)years, including 90(22.28%)youngest-old, 165(40.84%)middle old and 149(36.88%)oldest-old.123 patients(30.45%)were diagnosed with sarcopenia, with a prevalence of 27.78%(25)in the youngest-old group, 28.48%(47)in the middle old group and 34.23%(51)in the oldest-old group.Compared with those without sarcopenia, the basal metabolic rate in sarcopenia patients of all age groups decreased significantly, the levels of hemoglobin and albumin in youngest-oldsarcopenia patients decreased, the waist circumference and body mass index in middle old and oldest-old sarcopenia patients decreased, and the body fat rate in middle old sarcopenia patients increased.In the subjects with sarcopenia, the proportion of men in the oldest-old group was higher than in the other two groups.Compared with the youngest-old group, the levels of alanine aminotransferase and prealbumin decreased, serum creatinine increased, and skeletal muscle index values did not change in the middle old and longest-old groups, but handgrip strength(27.7±5.9 vs.23.2±6.9 vs.21.4±5.8, F=5.81, P=0.004), gait speed(0.8±0.2 vs.0.7±0.2 vs.0.5±0.2, F=11.88, P=0.000)and calf circumference(33.4±3.3 vs.15.9± 8.4 vs.31.5±3.1 vs.30.9±3.3, F=3.58, P=0.031)significantly decreased, and time up and go values(10.0±2.1 vs.15.9±8.4 vs.20.8±12.8, F=6.98, P=0.001)increased in the middle old and longest-old groups.Partial correlation analysis showed that age had a significant negative correlation with handgrip( r=-0.374, P=0.001), daily gait speed( r=-0.441, P=0.000), and calf circumference( r=-0.223, P=0.017), but a significant positive correlation with timed up and go( r=0.319, P=0.009). Conclusions:Compared with youngest-old sarcopenia patients, middle old patients show significant decline in muscle function, muscle strength and muscle mass in the lower limb.With increasing age, the impact of changes in muscle function, muscle strength and regional muscle mass on adverse events should receive heightened attention.

ABSTRACT

OBJECTIVE@#To assess the impact of nonsteroidal anti-inflammatory drugs (NSAIDs) on clinical outcomes of patients receiving anti-PD-1 immunotherapy for hepatocellular carcinoma.@*METHODS@#We conducted a retrospective study among 215 patients with primary liver cancer receiving immunotherapy between June, 2018 and October, 2020. The patients with balanced baseline characteristics were selected based on propensity matching scores, and among them 33 patients who used NSAIDs were matched at the ratio of 1∶3 with 78 patients who did not use NSAIDs. We compared the overall survival (OS), progression-free survival (PFS), and disease control rate (DCR) between the two groups.@*RESULTS@#There was no significant difference in OS between the patients using NSAIDs (29.7%) and those who did not use NSAIDs (70.2%). Univariate and multivariate analyses did not show an a correlation of NSAIDs use with DCR (univariate analysis: OR=0.602, 95% CI: 0.299-1.213, P=0.156; multivariate analysis: OR=0.693, 95% CI: 0.330-1.458, P=0.334), PFS (univariate analysis: HR=1.230, 95% CI: 0.789-1.916, P=0.361; multivariate analysis: HR=1.151, 95% CI: 0.732-1.810, P=9.544), or OS (univariate analysis: HR=0.552, 95% CI: 0.208-1.463, P=0.232; multivariate analysis: HR=1.085, 95% CI: 0.685-1.717, P=0.729).@*CONCLUSION@#Our results show no favorable effect of NSAIDs on the efficacy of immunotherapy in patients with advanced primary liver cancer, but this finding still needs to be verified by future prospective studies of large cohorts.

ABSTRACT

Objective:To investigate the current status of primiparas′ postpartum fatigue and paternal involvement, and to explore the relationship between primiparas′ postpartum fatigue and paternal involvement, and to provide reference basis for developing targeted intervention measures to alleviate postpartum fatigue of primiparas.Methods:A cross-sectional survey was conducted on 347 primiparas from Affiliated Hospital of Yangzhou University, Yangzhou Maternal and Child Health Care Hospital from September to December 2020 by convenience sampling. The survey instruments included the general information questionnaire, the Parenting Alliance Inventory (PAI), and the Postpartum Fatigue Scale (PFS).Results:The total score of PAI was (86.51 ± 12.07) points, and the level of paternal involvement was high. The total score of PFS was (16.68 ± 4.12) points. 95.97% (333/347) of primiparas had varying degrees of postpartum fatigue. There was a significant negative correlation between paternal involvement and primiparas′ postpartum fatigue ( r=-0.327, P<0.01). The results of multiple stratified regression analysis showed that paternal involvement was included in the influencing factor model of primiparas′ postpartum fatigue, which could independently explain 9.7% variation of primiparas′ postpartum fatigue. Conclusions:The higher level of paternal involvement could predict the lower level of primiparas′ postpartum fatigue. Medical staff should pay attention to the participation level of the spouses of primiparas in childcare, and improve the participation level of the spouses of primiparas in scientific ways to alleviate the postpartum fatigue of primiparas.

ABSTRACT

Objective@#To investigate the change in intestinal flora in Mongolian students with anxiety,so as to provide basis for exploring the relationship between flora and secretion expression in vivo.@*Methods@#The Self rating Anxiety Scale(SAS)was used to assess anxiety in medical college students; then a semi structured interview was administered. Fecal samples that met the inclusion criteria were collected and divided into anxiety (SAS score≥50) and control groups (no anxiety, SAS score<50) according to the standard score of SAS. Samples provided by Mongolian female students were selected from each group. The total bacterial DNA was extracted from the fecal samples for PCR amplification and NovaSeq 2x250bp high throughput sequencing was performed for the V3- V4 region of 16S rDNA gene to obtain the biological information of the intestinal flora. The intergroup OTU, structural diversity, significant difference, and LEfSe analyses were performed with information mining of the literature think tanks.@*Results@#Anxiety existed in 23.86% of the Mongolian students,and 16.96% of the Han people. A Chi square test showed no significant difference in detection of anxiety between Mongolian and Han college students ( P =0.07). Analysis of the alpha diversity index showed that the Shannon index, Simpson index, Chao1 index, and Observed species did not differed significantly between the two groups( t =8.0, 9.0 ,6.0,6.5). The difference in abundance of some bacteria was significant at the Class, Order, Family, and Genus levels between the two groups( t =-2.26-2.57,-5.08-3.58,-2.65-2.09, P <0.05).@*Conclusion@#The alpha diversity index showed that there was no significant difference in the abundance and diversity of intestinal flora between the two groups. While there were significant differences at different classification levels, the results suggest that the structure of intestinal flora can change in students with anxiety.

ABSTRACT

Objective:To analyze the source of infection, clinical symptoms and epidemiological characteristics of pertussis surveillance cases in Minhang District of Shanghai from 2017 to 2019,so as to provide the basis for pertussis surveillance and immunization strategy. Methods:Descriptive epidemiological method was used to analyze the pertussis surveillance cases. Results:A total of 334 cases were included in the pertussis active surveillance system, 237 cases were confirmed cases. There were 13 towns or streets in Minhang District, except for Pujin Street, all the other 12 towns or streets had cases. The high risk group (131/237, 55.27% of the cases) were the infants under five months old who did not reach the age of immunization or did not complete the basic immunization of diphtheria, tetanus, and acellular pertussis combined vaccine (DTaP). The cases mainly occurred in March to July, accounting for 71.31% (169/237). The highest culture-positive rate of Bordetella pertussis was in August (82.76%, 24/29); the highest positive rate of polymerase chain reaction (PCR) detection was in March (58.82%, 20/34). All the monitored cases had cough symptoms with an average cough days of 22.05±9.81 in confirmed cases. A total of 64 clustered outbreaks were detected, with an attack rate of 37.45% (94/251). 32.91% (78/237) of the cases had contact history with coughing patients 21 days before the onset, and 94.87% (74/78) of them were family members. The aggregation of pertussis was mainly transmitted from adults to infants and infants to adults. Conclusion:Children under five months old are the high incidence population of pertussis, and family members are the main source of infection. It is suggested that pertussis monitoring and prevention should be carried out in adolescents and adults.

ABSTRACT

Objective:To analyze the collected data of adverse events (AEFI) of pneumonia vaccine in Shanghai from 2010 to 2020,, so as to provide the appropriate immunization strategy and popularization of science for pneumonia vaccine . Methods:the data of suspected adverse events were collected from National AEFI Surveillance System during 2010 to 2020, and the vaccination information of pneumonia vaccine was obtained from Shanghai Vaccination Information System. Descriptive epidemiological method was used to analysis. Results:The total 18 446 AEFI cases were reported in Shanghai from 2010 to 2020, and the incidence rate was 425.0/100 000 doses. The general reactions were reported as 18 003 cases (97.6%), with 393 cases of abnormal reactions (2.0%), 57 cases of coincidences (0.3%), 2 cases of psychogenic reactions (<0.1%) and no vaccine quality accident or vaccination accident was reported. There was no significant difference in the incidence rate between male and female; The incidence rate of AEFI decreased with the age of the old. The incidence rate of 23 valent pneumococcal polysaccharide vaccine (PPV23) AEFI was 476.5/100 000 doses, which was significantly lower than that of other kinds of pneumonia vaccine. The first dose was also significantly higher than that of non-first dose. Conclusion:The various types of pneumonia vaccines being used in Shanghai have high safety.

ABSTRACT

A case report of pneumonia vaccination followed by death due to Guillain-Barré syndrome was fully presented and discussed. The association between the vaccination and Guillain-Barré syndrome could not be completely ruled out; however, the diagnosis of coincidental event was preferred, which may provide a reference for future similar case investigation.

The Singapore Health Services cluster (SingHealth) radiology film archives are a valuable repository of local radiological cases dating back to the 1950s. Some of the cases in the archives are of historical medical interest, i.e. cerebral angiography in the workup of patients with hemiplegia. Other cases are of historical social interest, being conditions seen during earlier stages of Singapore's development, i.e. bound feet. The archives form a unique portal into the development of local radiology as well as the national development of Singapore. A selection from the archives is published in commemoration of the International Day of Radiology in 2020, as well as the 200th anniversary of the Singapore General Hospital in 2021. This pictorial essay comprises gastroenterology, musculoskeletal and obstetrics and gynaecology cases from the archives.

ABSTRACT

Aim To investigate the effects of schisandrin C on arterial inflammation and atherosclerosis of ApoE-/-mice fed with high-fat diet. Methods ApoE-/-mice were divided into high fat diet group (HFD) and high fat diet group + schisandrin group (HFD + Sch C). High fat diet and Sch C were given at the same time. After 12 weeks, mice were executed and arterial tissues were collected. RT-q PCR and Western blot were respectively used to detect the expressions of IL-6, TNF-α, ICAM-1, β-actin and the phosphorylation and expression of IκB-α in arteries. Oil red O staining and biochemical kit were respectively used to detect the lipid changes in aortic root and the blood lipid levels. Results Compared with HFD group, the area of atherosclerotic plaque in HFD + Sch C was reduced (P < 0. 05), but Sch C did not affect blood lipid levels. Compared with HFD group, the mRNA expression of TNF-α, IL-6 and ICAM-1 and the phosphorylation of IκB-α of arterial tissue in HFD + Sch Cgroup decreased (P < 0. 05). Conclusions Sch C could effectively alleviate atherosclerosis induced by high-fat diet in ApoE-/-mice, accompanied by alleviation of arterial inflammation.

ABSTRACT

Objective: To investigate the effect and mechanism of cyclanoline on nitrosamine (BBN)-induced bladder cancer in rats. Methods: Male SD rats were randomly divided into control group, model group, Cyc-low dose (20 mg/kg) group, Cyc-high dose (40 mg/kg) group and cisplatin (5 mg/kg) group. Rats were intragastrically administered with 0.5 mL BBN (0.4 kg/L, twice a week for 8 weeks) to establish bladder cancer model. In addition to control and model group given with normal saline, the other groups were intraperitoneally injected with drugs (once a day for 8 weeks). The bladder tissue was collected after experiment. HE staining was used to investigate the histopathological changes of the bladder in rats. The expressions of MMP9 and Ki67 were observed by immunohistochemistry method, and the expressions of KLF4, p21, CyclinD1, E-cadherin, N-cadherin, Vimentin, Wnt, β-catenin in the bladder tissues were detected by Western bloting. Results: Compared with the model group, cyclanoline effectively inhibited the infiltration of cystitis cells, promoted the degeneration of cancer cells, and reduced the proportion of cytoplasm. Cyclanoline significantly decreased the expressions of MMP9 and Ki67 (P < 0.05, 0.01), up-regulated the expressions of KLF4, p21 and E-cadherin (P < 0.05, 0.01), down-regulated the expressions of CyclinD1, Wnt, β-catenin, N-cadherin and Vimentin in bladder cancer tissues (P < 0.05, 0.01). Conclusion: Cyclanoline promotes the expression of KLF4, inhibits Wnt/β-catenin signaling transduction and the epithelial cell-mesenchymal transition of bladder cancer cells, thereby inhibiting the migration and invasion of bladder cancer cells. Meanwhile, cyclanoline regulates the expressions of p21 and CyclinD1 by up-regulating KLF4, affects the proliferation of bladder cancer cells, and thereby delays the pathological process of BBN-induced bladder cancer in rats.

ABSTRACT

Objective To compare.the therapeutic effect of claw-type bone plates and anatomical plates in fixation of traumatic multiple rib fractures.Methods Clinical data of 70 patients with traumatic multiple rib fractures between January 2016 and November 2017 was analyzed retrospectively.Based on the treatnents,patients were assigned to two groups:the claw-type bone plates fixation group (n0=33) and anatomical plates fixation group (n =37).The variables including operation time,intraoperative blooding,postoperative thoracic hemorrhage rate,wound drainage,plate loosening and breakage,postoperative chest pain were compared.Data were analyzed using SPSS17.0 statistical software.Student's t test and rank sum test were used to compare the measurement data,and x2test was used to compare the counting data.Results The operation time after the full exposure of fractures were recorded.Operation time for each fracture [(11.04± 1.81)min vs (12.30±0.47) min],intraoperative bleeding [(120.76±7.19) mL vs (110.00±7.45) mL] and wound drainage [(114.06±7.24) mL vs (107.98±7.23) mL] in the claw-type rib bone plates fixation group were statistically significant different compared with the anatomical plates fixation group (P<0.05).There were lcase of screw abscission in the anatomical plates fixation group (suffering trom osteoporosis) and 2 cases in the claw-type bone plates fixation group (without osteoporosis and other complications).Two cases suffered from chest pain and 2 cases suffered from massive hemorrhage in the chest in the claw-type bone plates fixation group,with secondary operation removing internal fixations.Conclusions The two kinds of internal fixation have their own advantages and disadvantages.The claw-type bone plates fixation is cheaper,easier to shape and has shorter operation time,which is especially suitable for comminuted fracture.Although the anatomical plates fixation is more expensive than claw-type bone plates fixation,the anatomical plates fixation on multiple fractured ribs has less postoperative blood loss,lower postoperative thoracic hemorrhage rate,plate loosening,chest pain,which significantly reduces second operation and possesses a higher clinical application value.

Objective@#To evaluate the effect of acteoside on learning, memory and neurotransmitter in SAMP8 mice.@*Methods@#The 6-month-old rapidly aging SAMP8 mice were randomly divided into model group, namenda group, low-dose acteoside group(30 mg·kg-1 ·d-1), medium-dose acteoside group(60 mg·kg-1 ·d-1) and high-dose acteoside group(120 mg·kg-1 ·d-1) according to the digital table method, with 12 in each group.And 12 SAMR mice with the same age resistance were used as the control group.After 75 days of continuous intragastric administration, Morris water maze method and spontaneous activity experiment were used to investigate the effects of acteoside on learning, memory and anxiety of mice.The levels of neurotransmitters acetylcholine(ACh), serotonin(5-HT), norepinephrine(NE) and dopamine(DA) in mouse brain tissue(cortex and hippocampus) were detected by ELISA.@*Results@#(1)In the Morris water maze test, compared with the model group, the acteoside significantly reduced the escape latency of SAMP8 mice in training period.(2)In the experiment of autonomic activity, compared with the model group, the average speed and total distance of the low-dose acteoside group were significantly increased(t=15.0, 20.8, both P<0.05); the number of vertical movements and the average speed of the medium-dose acteoside group were significantly increased(t=15.8, 13.6, both P<0.05). The average speed and total distance of the high-dose acteoside group were remarkarbly increased(t=30.9, 29.7, both P<0.05), and the number of defecations in the mice of the lav-dose acteoside group, medium-dose acteoside group, high-dose acteoside group were((2.83±1.19)particles, (3.25±1.29)particles, (2.58±1.16)particles), they were obviously lower than that of the model group ((5.25±1.48) particles)(t=15.7, 20.1, 13.5, all P<0.01). (3)Simultaneously, the contents of ACh, NE and DA in the hippocampus and cortex of the model group were significantly lower than those in the normal group (hippocampus: t=10.3, 12.7, 13.2, all P<0.05; cortex: t=11.7, 10.5, 12.4, all P<0.05). Compared with the model group, the contents of ACh, NE, DA and 5-HT in the hippocampus and cortex of the low, medium and high doses of acteoside were significantly increased(hippocampus: t=31.4, 20.3, 10.7, 12.9, all P<0.05; cortex: t=33.7, 29.4, 14.5, 12.7, all P<0.05).@*Conclusion@#The acteoside can enhance the ability of spatial learning and memory of SAMP8 mice, and can regulate the depression and anxiety of animals.The mechanism may be related to the ACh content and the monoamine neurotransmitters increase in the brain.

ABSTRACT

Objective@#To evaluate the safety of population based sequential vaccination schedule of inactivated poliovirus vaccines prepared with different strains.@*Methods@#This randomized, parallel-group controlled trial was conducted from March, 2017 to May, 2018, in Shanghai. Adverse reaction data of Sabin strain inactivated polio vaccine (sIPV), wild strains inactivated polio vaccines (wIPV) and bivalent types Ⅰ and Ⅲ oral poliomyelitis vaccine (bOPV) were systematically collected through active observation in 1 917 infants in Shanghai after the vaccination at 2, 3, 4 months old. The eligible infants aged 2 months were divided into 4 groups: ①sIPV+sIPV+bOPV group; ②sIPV+wIPV+bOPV group; ③wIPV+sIPV+bOPV group; ④wIPV+wIPV+bOPV group.@*Results@#The incidence of adverse reaction 30 days later after 3 basic dose vaccinations was 16.79% (946/5 633). No serious adverse reaction was reported. Local and systemic reactions were mainly mild. Common local reactions were pain, erythema, cutaneous nodule, etc.; and common systemic reactions were abnormal crying, drowsiness, diarrhea and appetite lost, etc.. The incidence of local reactions 30 days later after 3 basic dose vaccinations was 1.65% (93/5 633), and the incidence rates of grade 1-3 reactions were1.26% (71/5 633), 0.21% (12/5 633) and 0.20% (11/5 633) respectively. The incidence rate of systemic reactions 30 days later after 3 basic vaccinations was 15.14% (853/5 633), and the incidence rates of grade 1-3 reactions were 11.33% (638/5 633), 3.18% (179/5 633) and 0.64% (36/5 633) respectively. There were no significant differences in the rate of grade 3 reaction among different groups (χ2=4.17, P=0.24).@*Conclusions@#No severe adverse reactions related to sequential vaccination of different strain inactivated polio vaccines were observed, most of reactions were mild and all of them were cured. It is safe to use sIPV and wIPV simultaneously or alternately for childhood sequential vaccination.

ABSTRACT

Objective@#To evaluate the immunogenicity of different strains of inactivated poliomyelitis vaccines (IPV) by sequential program.@*Methods@#This parallel-group controlled trial was conducted in immunization clinics in Shanghai from March 2016 to September 2017. Sabin strains inactivated poliomyelitis vaccines (sIPV), WPV strains inactivated poliomyelitis vaccines (wIPV) and live poliomyelitis Type Ⅰ Type Ⅲ vaccine (bOPV) as the investigational vaccine were used at 2, 3, 4 months old in 325 infants in Shanghai. Infants vaccinated by four sequential program were divided into 4 groups: sIPV+sIPV+bOPV, sIPV+wIPV+bOPV, wIPV+sIPV+bOPV and wIPV+wIPV+bOPV. A total of 230 investigators′ blood samples were collected before primary immunization and 163 investigators′ blood samples were collected after primary immunization. A total of 151 investigators (36, 44, 30 and 41 in each group) finished primary immunization and blood sampling before and after the primary immunization. The geometric mean titer (GMT) of poliovirus typesⅠ and Ⅲ neutralizing antibody was tested and calculated, and the positive results of antibody before and after primary immunization were analyzed.@*Results@#Among the 151 investigators, the age were (2.27±0.61) months and birth weight were (3.27±0.43) kg, and 70 were male. The positive rates of typeⅠwas 98.68% (149 cases), and type Ⅲ was 97.35% (147 cases); the number of investigators tested in each group was 36, 44, 30 and 41, respectively; the positive rates of typeⅠwas 97.22% (35 cases), 100.00% (44 cases), 96.67% (29 cases) and 100.00% (41 cases) (P=0.345); the positive rates of type Ⅲ were 97.22% (35 cases), 95.45% (42 cases), 96.67% (29 cases) and 100.00% (41 cases) (P=0.614).@*Conclusion@#Using sIPV and wIPV simultaneously or alternately for sequential immunization of poliomyelitis vaccines showed good immunogenicity for infants at appropriate age.

ABSTRACT

Objective To evaluate the effect of acteoside on learning,memory and neurotransmitter in SAMP8 mice. Methods The 6-month-old rapidly aging SAMP8 mice were randomly divided into model group,namenda group,low-dose acteoside group(30 mg·kg-1 ·d-1),medium-dose acteoside group(60 mg ·kg-1 ·d-1) and high-dose acteoside group(120 mg·kg-1 ·d-1 ) according to the digital table method, with 12 in each group. And 12 SAMR mice with the same age resistance were used as the control group. After 75 days of continuous intragastric administration,Morris water maze method and spontaneous activity experi-ment were used to investigate the effects of acteoside on learning,memory and anxiety of mice. The levels of neurotransmitters acetylcholine ( ACh), serotonin ( 5-HT ), norepinephrine ( NE ) and dopamine ( DA ) in mouse brain tissue(cortex and hippocampus) were detected by ELISA. Results (1) In the Morris water maze test,compared with the model group,the acteoside significantly reduced the escape latency of SAMP8 mice in training period. (2)In the experiment of autonomic activity,compared with the model group,the aver-age speed and total distance of the low-dose acteoside group were significantly increased(t=15. 0,20. 8,both P<0. 05);the number of vertical movements and the average speed of the medium-dose acteoside group were significantly increased(t=15. 8,13. 6,both P<0. 05). The average speed and total distance of the high-dose acteoside group were remarkarbly increased(t=30. 9,29. 7,both P<0. 05),and the number of defecations in the mice of the lav-dose acteoside group, medium-dose acteoside group, high-dose acteoside group were ((2. 83±1. 19)particles,(3. 25± 1. 29) particles,(2. 58± 1. 16) particles),they were obviously lower than that of the model group ((5. 25±1. 48) particles)(t=15. 7,20. 1,13. 5,all P<0. 01). (3) Simultaneously, the contents of ACh,NE and DA in the hippocampus and cortex of the model group were significantly lower than those in the normal group (hippocampus:t=10. 3,12. 7,13. 2,all P<0. 05;cortex:t=11. 7,10. 5,12. 4, all P<0. 05). Objective@#To access the influence of voice disorders on children′s voice-related quality of life through the parental version of pediatric voice handicap index (pVHI).@*Methods@#From April 2017 to March 2018, a total of 192 children with voice disorders (dysphonic group) and 111 children without voice disorders (control group) were enrolled in this work. Parents of children in both groups were asked for fill out the questionnaire containing the parental version of pVHI and the data of non-normal distribution were analyzed by Mann-Whitney U test. Spearman test was used for correlation analysis.@*Results@#(1)Vocal cord nodule was the most common voice disorder in children, and boys were more susceptible to voice disorder than girls in this study (70.3%(135/192) vs 29.7%(57/192)). (2)The most common voice abuse or misuse habit was "Shouting loudly". (3)In dysphonic group, the scores of function, physiology, emotion and total were higher than those in control group (all P<0.05). (4)In dysphonic group, there was a weak correlation between the parents′ overall evaluation of the children′s voice status and the three dimensions of the parental version of pVHI (function: r=0.339, physiology: r=0.334, emotion: r=0.208, all P<0.001).@*Conclusions@#Voice disorders can cause a negative impact on children′s quality of life. Parental version of pVHI can be used to assess the voice-related quality of life in children with voice disorders.

ABSTRACT

Objective To explore the effects of acteoside on dysfunction of learning and memory and the protective effect of oxidative stress in rats with vascular dementia.Methods 30 SD rats were randomly divided into sham group,model group,nicergoline group,low-dose acteoside group and high-dose acteoside group,with 6 rats in each group.Preparation of vascular dementia model by 2-vessed occlusion.The ability of exploring and learning and memory in rats were detected by step down test and avoid dark test.Determination of malondialdehyde (MDA),reactive oxygen species (ROS) and glutathione peroxidase (GSH-PX) activity in serum and brain tissue was conducted by Elisa.Results Autonomic activity test showed that the frequency and activity of autonomous activity in the model group were significantly lower than those in the sham group(P<0.01),the frequency of autonomous activity in each administration group was significantly higher than that in the model group (P<0.01),and the central activity time in the high-dose group was significantly higher than that in the model group (P<0.01).Step down test and avoid dark test showed that the latency of the model group was significantly lower than that of the sham group.(Model group of step down test:(25.33 ± 3.01) s,Sham group of step down test:(56.83 ± 15.90)) (P< 0.01).(Model group of avoid dark test:(15.67 ± 3.61) s,Sham group of avoid dark test:(135.82±44.00) s) (P<0.01).The latency of low dose group was significantly higher than that of model group.(Low dose group of step down test:(46.40±14.32) s) (P<0.01).(Low dose group of avoid dark test (44.20± 8.26)s) (P<0.05).Step down test and avoid dark test showed that the number of mistakes in the model group was significantly higher than that in the sham operation group(P<0.01).The number of errors in nicergoline group and the low dose group was significantly lower than that in the model group (P<0.01,P<0.05).In serum,the content of MDA and ROS in model group was significantly higher than that in sham group (P<0.01) while the activity of GSH-PX in model group was significantly lower than that of sham group.The content of MDA in the other groups was significantly lower than that in the model group (P<0.01).The content of ROS in the nicergoline group and low dose group was significantly lower than that in the model group (P<0.05,P<0.0l).The activity of GSH-PX in high dose group was significantly higher than that in model group (P<0.01).In brain tissue,the content of MDA and Ros in model group was significantly higher than that in sham group (P<0.01).The content of MDA and ROS in low dose group and high dose group were significantly lower than that in model group (P<0.05,P<0.01).Conclusion Acteoside can improve the dysfunction of learning and memory and depressive mood disorder caused by vascular dementia and reduce oxidative stress injury by decreasing the content of MDA-ROS and increasing the activity of GSH-PX enzyme.
