ABSTRACT

Objective To assess the safety and efficacy of extended liver resection using preoperative PTCD (percutaneous transhepatic cholangial drainage) and PVE (portal vein embolization) to treat patients with locally advanced hilar cholangiocarcinoma.Methods We collected and analyzed the clinical data of 27 patients with Bismuth types Ⅲ and Ⅳ hilar cholangiocarcinoma who underwent extended hepatecomy using preoperative PTCD and PVE in our hospital.Results There were 21 patients with R0 resection and 6 patients with R1 resection.The mortality rate was 0%.Eight patients died of cancer recurrence.Conclusion Preoperative PTCD and PVE combined with extended hepatectomy were safe and efficacious in treating patients with locally advanced hilar cholangiocarcinoma,which resulted in potential cure.

ABSTRACT

Objective To study the role of Ⅲb isoform of human fibroblast growth factor receptor 1 (FGFR1-Ⅲb) in PANC-1 pancreatic cancer cells. Methods The plasmid of human full-length FGFR1-Ⅲb isoform,pSVK4/FGFR1-Ⅲb, was stable transfected into cultured PNAC-1 pancreatic cancer cell lines facilitated by lipofectamine. The function of FGFR1-Ⅲb in transfected pancreatic cancer cells were examined by MTT assay, soft agar assay, cell migration assay, single cell movement assay, In vivo tumorigenicity assay. Results The basal anchorage-dependent and -independent cell growth was significantly inhibited. Additionally, FGFR1-Ⅲb expression inhibited single cell movement and in vitro invasion as determined by time-lapse microscopy and boyden chamber assay as well as in vivo tumor formation and growth in nude mice. Microscopic analysis of the xenograft tumors revealed a reduced Ki-67 labelling, lower amount of tumor necrosis and higher grade of differentiation in FGFR1-Ⅲb expressing tumors. Conclusion We identified a functional human FGFR mRNA splice variant that inhibits the transforming potential of pancreatic cancer cells.

ABSTRACT

Analysis is carried out on millimeter-wave (MMW) dosimetry in culture dishes used in experiments on MMW biological effects at the cellular level. Finite-difference time-domain (FDTD) technique is employed to compute the 6 mm MMW power density (PD) irradiated into cells in a typical culture dish and the MMW power absorption density (PAD) of cells, followed by the qualitative and quantitative analyses on the outcomes. Indicated in the results, distributions of MMW PD irradiated into cells and the PAD of cells are complicated with evident inhomogeneity so that MMW dosimetry varies a lot in different position whose influence on the experimental outcomes is not neglectable. Consequently, the precise determination of dosimetry is of great importance to be conducted in related experiments.

ABSTRACT

Biological effects of millimeter waves (MMWs) at the cellular level are explored in experiments using culture dishes containing cell monolayers with MMW irradiated from the underneath. Analysis is carried out for culture dishes with diameters much larger than the MMW wavelength to discover the relation between the portion of the incident MMW power density (PD) irradiated into the cell monolayer and the interfering factors such as the bottom thickness of the culture dish, the MMW wavelength, and the electromagnetic parameters of the dish and culture solution. With the help of the finite-difference time-domain (FDTD) technique, another analysis is performed to display the effect of the dish geometries upon the MMW PD irradiated into the cell monolayer in a specific culture dish with a diameter similar to the MMW wavelength. The results indicate that rigorous analysis, precise measurement and accurate calculation of MMW PD are essential accompaniments for these experiments, and large-diameter culture dishes are more preferred in experiments to small-caliber ones.

Objective To test the expression of cyclooxygenase 2 (COX 2) gene in colorectal adenomas, cancers in contrast to that of normal mucosa.[WT5”HZ] Methods [WT5”BZ] With reference to the expression of ? actin gene, the expression of COX 2 mRNA was examined in colorectal adenomas from 10 patients, cancerous tissues and adjacent noncancerous mucosa from 30 patients by reverse transcription polymerase chain reaction (RT PCR). Quantitation of relative band densities was performed using densitometry scanning techniques.[WT5”HZ] Result [WT5”BZ] Strong expression of COX 2 mRNA was detected in ninety percent of cancers with range of 0 02~0 85 (median 0 5). ABSTRACT

Objective To investigate the surgical measures for relieving biliary malignancies that occur in bile duct cysts (BDCs). Methods Twenty four cases of biliary carcinoma occurring in BDCs treated in our Department from October 1965 to June 1999 were reviewed. Results Of the patients, 5 were male and 19 female. The average age at the time of being diagnosed was 38. Nine patients had undergone cystoenterostomy before. The mean duration between operation of drainage and detection of carcinoma was 10 years. Operations included excision of the extrahepatic bile duct plus pancreatoduodenectomy in 5, excision of extrahepatic bile duct in 6, and various palliative procedures because of extensive spread of tumor in 13. Follow up data were obtained in all but 2 patients who died in hospital. The mean survival time for the 16 death cases after discharge was 11 months. Four patients have been alive and free from disease for 12, 7, 6.5 and 0.5 years, respectively. Conclusions 1) Early diagnosis and curative resection for the biliary carcinoma in BDCs are the key points to achieve better outcomes. 2) Prophylactic excision of cyst in adults with BDCs, especially those previously treated with cystoenterostomy, can effectively reduce the risk of malignant changes.

ABSTRACT

Objective To investigate the correlation between VEGF-C and COX-2 expression in human rectal cancers and its significance in cancer metastasis. Methods VEGF-C expression was detected with Western blot in LOVO cells treated with NS-398 or PGE2. VEGF-C and COX-2 expression in 45 rectal adenocarcinomas was tested with immunohistochemistry. Results NS-398 inhibited the VEGF-C expression, and PGE2 up-regulated the expression of VEGF-C in a dosage-dependent way in LOVO cells. VEGF-C expression was significantly higher in adenocarcinomas with lymph node metastasis, and was related with the expression of COX-2 in 45 rectal adenocarcinomas. Conclusion COX-2 up-regulates VEGF-C and VEGF-C plays an important role in lymphatic metastasis of rectal cancers.
