ABSTRACT

Through interviews with 30 pregnant nurses, this paper found that pregnant nurses needed humanistic care in four aspects of worrying about their own health and their fetus' health, psychological pressure, career planning and interpersonal relationships. And this paper put forward humanistic care countermeasures, including that nursing administrators should remind pregnancy nurses to pay attention to the health of themselves and their fetal, and pay attention to the psychological health of the pregnancy nurses, increasing the humanistic care to the pregnancy nurses can reduce the turnover rate of nurses, and family and society should provide humanistic care support for pregnant nurses.

ABSTRACT

Objective:To observe the clinical effect of liver-regulating and blood-tonifying acupuncture in the treatment of cervical spondylosis of vertebral artery type. Methods:A total of 60 cases meeting the inclusion criteria were randomized into an observation group and a control group, 30 cases in each group. Cases in the observation group received liver-regulating and blood-tonifying acupuncture, in which Jiaji (EX-B 2) points of C3-C7, Ganshu (BL 18) and Geshu (BL 17) were used; cases in the control group received routine acupuncture treatment in which jiaji (EX-B2) points of C3-C7 were used. Both groups received 5 times of treatment in a week with a 2-day rest, symptoms evaluation was done after 4 weeks of treatment, and then therapeutical effect was evaluated. Results:The total effective rate was 90.0% in the observation group, 73.3% in the control group, and inter-group comparison showed a statistical significance (P<0.05). After treatment, the symptoms and signs scores in both groups all substantially dropped, showing statistical significances (both P<0.05); inter-group comparison showed that the improvements in subscales of dizziness, pressing pain along vertebrae and revolve-cervix test in the observation group were superior than those in the control group, showing statistical significances (all P<0.05), while the inter-group comparisons of subscales of headache and vomiting and nause didn't show any statistical significances after treatment (both P>0.05). Conclusion: Liver-regulating and blood-tonifying acupuncture is effective in treating cervical spondylosis of vertebral artery type, and it can significantly improve patients' symptoms and signs.

ABSTRACT

OBJECTIVE@#To evaluate the therapeutic effects of recombinant Exendin-4 and double-stranded adeno-associated virus (Exendin-4/dsAAV) on SD rats with type 2 diabetes (T2DM) through injecting it into submandibular gland (SG).
@*METHODS@#The Exendin-4/dsAAV was injected into submandibular gland of diabetic rat. The insulin and α-amylase were detected by real-time PCR at the 2nd, 4th and 8th weeks. The immunohistochemisty was used to detect the insulin contents in SG at the 8th week. The concentration of blood glucose and levels of insulin secretion were detected after pancreatectomy.
@*RESULTS@#The SG gland was bigger in Exendin-4/dsAAV group than that in the control group, but the changes in α-amylase were not significant. The Exendin-4 and insulin gene expression was increased in the Exendin-4/dsAAV group (P<0.05). The Exendin-4 and insulin were positive in the SG. The blood glucose was lower and insulin concentration was higher in the Exendin-4/dsAAV group than those in the control group after pancreatectomy (P<0.05), and the insulin content was also increased in the dsAAV groups.
@*CONCLUSION@#Continuous expression of Exendin-4 in SG may improve glucose control and insulin secretion in T2DM rats through inducing expression of insulin.

ABSTRACT

OBJECTIVE: To explore the feasibility of the reversal of multidrug resistance in breast cancer using vector-based small interference RNA(siRNA) and to solve the problems of siRNA transient expression and siRNA delivery in vivo. METHODS: Based on the siRNA sequence which was screened in previous studies that could effectively inhibit the expression of MDR1 gene, the expression plasmid was constructed. The siRNA expression plasmid was then encapsulated in new nano-sized stealth cationic liposomes. Pharmacodynamic studies of the liposomes were carried out in vitro and in vivo. RESULTS: The results showed that the cationic liposomes loaded with MDR1 siRNA expression plasmid could effectively inhibit the expression of MDR1 gene both in vitro and in vivo. CONCLUSION: Multi-drug resistance of breast cancer cells is reversed to a great extent by this siRNA-containing cationic liposomes. Nano-sized cationic liposomes are ideal delivery vehicle of siRNA, which could protect the siRNA from degradation and deliver siRNA into the tumor region where it could exert functions.

ABSTRACT

In order to solve the problem of selection and in vivo delivery problem in siRNA treatment, hepatitis B virus (HBV) HBx gene which could be targeted by siRNA was studied. The siRNA expression plasmid which specific inhibits HBx expression was obtained by in vitro selection via a dual-luciferase plasmid including HBx-Fluc fusion protein expression domain. The selected siRNA expression plasmid was then encapsulated in PEG-modified cationic liposome, which was devoted into pharmacodynamic studies at both cellular and animal level. The results illustrated that the cationic liposome which encapsulated siRNA expression plasmid could effectively inhibit HBx gene expression both in vitro and in vivo.

ABSTRACT

Objective To study the effect of allicin on the growth and radiosensitivity of human pancreatic carcinoma BXPC3 cells.Methods BXPC3 cells were exposed to X-rays in the presence or absence of allicin.Cell proliferation was measured by MTT assay.Cell cycle distribution and apoptosis were detected by flow cytometry assay.Cell radiosensitivity and the influence of allicin on it was evaluated by colony formation assay.The expressions of Bax and Bcl-2 proteins were assayed by RT-PCR and Western blot.Results IC50 values of allicin on cell growth were 76.24,58.34 and 43.58 μmol/L under 12,24 and 48 h treatment,respectively.Treatment of cells with allicin obviously inhibited cell growth after irradiation and hence increased radiosensitivity (t =2.74,P < 0.05).This treament also enhanced radiation-induced cell cycle arrest at G2/M phase (t =11.41,P <0.05),apoptosis induction (t =12.36,P < 0.05),and Bax expression (t =4.83,P < 0.05),but it decreased Bcl-2 expression (t =3.69,P < 0.05).Conclusions Allicin could inhibit cell growth,induce cell cycle arrest and apoptosis via Bax/Bcl-2 pathway and hence increases radiosensitivity of BXPC3 cells.

ABSTRACT

Objective To evaluate the clinical effectiveness of interventional therapy in treating thoracic aortic dissection (TAD) . Methods A follow-up visit with 24 TAD patients had been operated in our hospital from November 2008 to March 2010. The clinical data, the therapeutic measures and the follow-up results were retrospectively analyzed. Including the remission of symptom, average day in hospital,complication,the long term survival rates and so on. Results All 24 patients have survived, the symptoms have improved instantly, the hospitalization days was 7.3 days,and no complication occurred. The 3 years survival rate was 95.8%. Conclusion Interventional therapy have advantages of treating thoracic aortic dissection with less complication,more secure and higher long term survival rates.

ABSTRACT

<p><b>OBJECTIVE</b>The purpose of this article is to examine the effect of curcumin on the proliferation and metastasis of human tongue squamous cell carcinoma and analyze its mechanism.</p><p><b>METHODS</b>SCC-4 were treated with curcumin of 0, 5, 10, 20, 30, 60, 100 micromol x L(-1) in 24 h. MTT assay, Matrigel invasion assay, flow cytometry and fluorescence microscopy were used to examine the effect of curcumin on the growth and metastasis of SCC-4. cDNA microarray and RT-PCR were employed to analyze the expression of genes treated by curcumin.</p><p><b>RESULTS</b>The results showed that curcumin could concentration-dependently inhibit SCC-4 cell proliferation at the concentration range from 20 to 100 micromol x L(-1). Furthermore, Matrigel invasion assay indicated that curcumin can reduce SCC-4 cell invasion under the dosage of 20, 30, 60 micromol x L(-1). Flow cytometry also showed that curcumin can influence the distribution of cell cycle of SCC-4 cell with the dosage of 20, 30, 60 micromol x L(-1). And the dosage of 30 micromol x L(-1) curcumin could lead to the recruitment of alpha-tubulin. cDNA microarray showed that 87 genes were activated and 198 genes were inhibited with the effect of curcumin. These results were validated by the real time quantitative RT-PCR.</p><p><b>CONCLUSION</b>According to the results, it suggests that curcumin has the potential as the leading compound for anti-cancer proliferation and invasion in oral cancer treatment, and cdc27, EGFR substrate 15, PPAR-alpha and H2A histone may play an important role among this multiple anticancer-targeting ability.</p>
