ABSTRACT
ObjectiveTo investigate the therapeutic effect of Yupingfeng San on allergic rhinitis (AR) and its effect on Reactive oxygen species (ROS)/NOD-like receptor thermal protein domain associated protein 3 (NLRP3)/cysteinyl aspartate specific proteinase-1 (Caspase-1) pathway. MethodSPF mice were randomly divided into control group, model group, loratadine group (0.9 mg·kg-1), and low, medium, and high dose Yupingfeng San groups (6, 12, 24 mg·kg-1), with 10 mice in each group. The control group was given routine feeding, and the other groups were intraperitoneally injected with [ovalbumin(OVA) + Al(OH)₃ + phosphate buffer solution(PBS)] suspension once every other day for seven consecutive times. After seven days, 10% OVA solution was instilled in the nose, two times each day for seven consecutive days. After successful modeling, each administration group was intraperitoneally injected with different doses of Yupingfeng San Decoction, and the control group and model group were intraperitoneally injected with an equal volume of normal saline. Symptoms of sneezing, scratching, and runny nose were recorded and scored daily. The levels of ovalbumin specific immunoglobulin E (OVA-sIgE), histamine, eosinophil cationic protein (ECP), prostaglandin D2 (PGD2), interleukin-1β (IL-1β), interleukin-18 (IL-18), interleukin-4 (IL-4), and γ interferon (IFN-γ) in nasal lavage solution and serum of mice were detected by enzyme-linked immunosorbent assay (ELISA). The damage status of the nasal mucosa was observed by hematoxylin-eosin (HE) staining. The number of goblet cells in the nasal mucosa of mice was observed by periodic acid-Schiff (PAS) staining. The expression of NLRP3 protein in the nasal mucosa of mice was detected by immunohistochemistry. Western blot was used to detect the expressions of NLRP3, cleaved Caspase-1, and cleaved gasdermin D (GSDMD) proteins in the nasal mucosa. The test kit was used to detect the changes in ROS in the nasal cavity of mice in each group. ResultCompared with the control group, the nasal symptoms of the model group were significantly aggravated, and the levels of inflammatory factors OVA-sIgE, histamine, ECP, PGD2, IL-1β, IL-18, and IL-4 in serum and nasal lavage solution were increased (P<0.05,P<0.01). The levels of IFN-γ were decreased (P<0.05,P<0.01). The histopathological score, goblet cell number, and ROS content were significantly increased (P<0.01), and the expressions of pyrodeath-related proteins NLRP3, cleaved Caspase-1, and cleaved GSDMD were increased (P<0.01). Compared with the model group, the nasal symptoms of the loratadine group and Yupingfeng San groups were significantly relieved, and the levels of inflammatory factors OVA-sIgE, histamine, ECP, PGD2, IL-1β, IL-18, and IL-4 in serum and nasal lavage solution were decreased (P<0.05,P<0.01). The levels of IFN-γ were increased (P<0.05,P<0.01). The histopathological scores, goblet cell number, and ROS content were significantly decreased (P<0.05,P<0.01), and the expressions of pyrodeath-related proteins NLRP3, cleaved Caspase-1, and cleaved GSDMD were decreased (P<0.05,P<0.01). Compared with the loratadine group, the curative effect of the high dose Yupingfeng San group was further increased (P<0.05,P<0.01). ConclusionYupingfeng San has a therapeutic effect on AR, and its specific effect may be related to the inhibition of ROS/NLRP3/Caspase-1-induced cell pyroptosis.

MethodAfter the treatment of CNE1 by Scutellariae Barbatae Herba extract, cell counting kit-8 (CCK-8) was used to detect cell proliferation, and Giemsa staining was used to detect the clone formation rate. Flow cytometry was used to detect cell cycle distribution, and reverse transcription-polymerase chain reaction ( RT-PCR) assay and Western blot assay were used to detect the relative expression of messenger ribonucleic acid (mRNA) by small interfering RNA (siRNA) or overexpression. ResultAs compared with the blank group, the proliferation and colony formation rate of CNE1 in the Scutellariae Barbatae Herba extract group significantly decreased (P<0.05, P<0.01) in a dose and time-dependent manner, whereas the percentage of cells in the presynthetic phase (G0/G1) increased (P<0.05, P<0.01). The expression level of S-phase kinase associated protein 2 (SKP2) in the Scutellariae Barbatae Herba extract group significantly decreased (P<0.01) as compared with the blank group. As compared with the Scutellariae Barbatae Herba extract group, the protein levels of p21 and p27 significantly decreased in the overexpressed SKP2+ Scutellariae Barbatae Herba extract group (P<0.01). As compared with the blank group, the signal activation and the phosphorylation level of signal transducer and activator of transcription 3 (STAT3) of CNE1 in the S. barbata extract group significantly decreased (P<0.05, P<0.01). ConclusionScutellariae Barbatae Herba extract effectively inhibits the proliferation of CNE1, and the mechanism may be related to its action on the STAT3/SKP2 pathway.

Objective:To preliminarily study the feasibility, safety and efficacy of transcatheter arterial chemoembolization (TACE) combined with selective portal vein embolization (SPVE) before surgical resection in the treatment of large liver cancer.Methods:A retrospective study was conducted on the clinical data of 17 patients with large liver cancer treated with TACE combined with SPVE from January 2016 to December 2019 at the Department of Hepatobiliary Surgery, the Sixth Medical Center of PLA General Hospital. The study included 15 males and 2 females, aged (59.17±10.30) years. The levels of alanine aminotransferase, tumor changes and patient survival were analyzed before operation, after TACE, and after SPVE.Results:Among the 17 patients, the levels of alanine aminotransferase on the 1st and 3rd day after SPVE was significantly higher than those after TACE [191.4 (30.5-1966.4) IU/L vs 125.3 (35.7-846.2) IU/L on the first day, and 298.5 (24.6-1334.2) IU/L vs 208.6 (21.6-775.6) IU/L on the 3rd day], all P<0.05. One month after the two combined embolism, among the 6 patients with a tumor diameter of 5-10 cm, 2 patients (33.3%) had complete remission, 3 patients (50.0%) had partial remission, and 1 patients (16.6%) had stable disease. For the tumor’s longest diameter, among the 11 patients with tumors >10 cm, 1 patient had complete remission (9.1%), 4 patients had partial remission (36.4%), 5 patients had stable diseases (45.5%), and 1 patient had disease progression (9.1%). Eventually, 11 patients underwent surgical exploration. The median residual liver volume before treatment was 329.5 (284.9-365.7) ml, and after the combined procedure 415.6 (354.7-718.8) ml. The median hyperplasia ratio was 28.1% (14.1%-51.3%). Eight patients finally underwent surgical resection. There was no death in the perioperative periods. The median tumor-free survival time was 17 (7-42) months, and the median survival time was 27 (7-42) months. Conclusion:For patients with large liver cancer with insufficient remnant liver volume, preoperative TACE+ SPVE has certain value in controlling tumor progression, promoting remnant liver hyperplasia, increasing surgical resection rate and improving prognosis.

Objective:To predict the expression and molecular regulatory mechanisms of βⅢ-tubulin (TUBB3) in non-small cell lung cancer (NSCLC) using bioinformatics methods.Methods:The GEO profile, TransmiR, miRWalk and ConSite database were employed in the present study. The upstream transcription factor, co-expression gene and the interactive miRNA of TUBB3 were studied to determine the molecular regulatory mechanisms and possible role of TUBB3 in NSCLC.Results:The promoter region of TUBB3 could be recognized by transcription factors associated with NSCLC including Snail and n-MYC and so on. At the post-transcription level, the TUBB3 could be regulated by miRNAs including miR-200 family, miR-342-3p, miR-410 and so on. These miRNAs were associated with proliferation and invasion of NSCLC. TUBB3 was co-expressed with HDGF, GMPS, MRPL9, PMAIP1 and SLBP, and they were co-regulated by the transcription factor Snail. SLBP, PMAIP1 and transcription factor Snail were related to cell cycle and apoptosis.Conclusion:The expression of TUBB3 can be regulated at multiple levels in NSCLC. It may take part in cell cycle and apoptosis regulation in NSCLC, and further influence proliferation and invasion of cancer cells.

Objective To evaluate the characteristics of arterial elasticity changes of abdominal aorta atherosclerosis in type 2 diabetic rats by using velocity vector imaging(VVI).Methods A rat model of diabetes mellitus(DM)were successfully established by a high-fat diet combined with the streptozotocin injection in thirty healthy male sprague-dawley rats.DM rats were randomly divided into five groups (n =6,each):0-week group (rats before treatment),4-week group (rats treated for 4 weeks),8-week group(rats treated for 8 weeks),the 12-week group(rats treated for 12 weeks),the 16-week group(rats treated for 16 weeks).The intima-media thickness(IMT)of abdominal aorta,the difference of end-systolic and end-diastolic vessel diameter(△D),resistance index(RI),pulsatility index (PI)and peak systolic velocity(Vs)were detected by ultrasound every 4 weeks.Peak radial velocity (PRy),PCs rate (PCs),peak circumferential strain (PCsr) and peak radial displacement (PRd) of abdominal aorta in short axis view were determined by VVI technique.Rats were sacrificed in different stages.Then,the specimens of abdominal aorta were collected,and observed by light microscope and electron microscope for the histopathological and ultrastructural changes in the atherosclerosis,respectively.Results Compared with 0-week group(0.25±0.03) mm and 4-week group(0.26 ±0.02)mm,IMT was thickened in the 16 weeks group(0.30±0.03)mm,(F=3.718,P=0.017).PRv,PCs,PCsr and PRd were lower after 8 weeks of treatment than in 0-week group.PRy,PCs,PCsr and PRd were lower in 16-week group than 0-week,4-week and 8-week groups (F =15.565,14.754,25.015 and 19.326,P<0.01).Under the light microscope in abdominal aorta of rats in 12-week group and 16-week group,the stromal layer was thickened,smooth muscle cells were deformed and migrated to the tunica intima,and foam cells were formed.And under electron microscope,endothelial cells had a large amount of lipid attachment,and smooth muscle cells had vacuolar degeneration.Conclusions As compared with the two-dimensional ultrasound,velocity vector imaging can earlier find the elasticity change and intimal damage in abdominal aortic wall of diabetic rat,and can better analyze a motor function of the vascular wall.

Objective To investigate the features of female sexual dysfunction (FSD) in type 2 diabetic patients (T2DM)and to analyze its risk factors. Methods According to FSFI indexes in the sexual function questionnaire,227 female T2DM patients were divided into DM group without FSD [n=144,FSFI≥25] and DM+FSD group[n=83,FSFI0.05). The score of sexual desire[(2.54±0.58) vs (3.76±0.90) vs (4.59±1.22) score],sexual arousal[(2.30±0.42) vs (3.57±0.85) vs (4.81±1.00) score],vaginal lubricate[(3.35±0.29) vs (4.46±1.32) vs (5.14±0.83) score],orgasm[(2.75±0.68) vs (3.40±1.86) vs (4.02±1.25)score] and total score [(17.94±4.15) vs (24.71±6.33) vs (28.13±5.78) score] were lower in DM+FSD group and DM group than those in NC group. The score of sexual satisfaction[(3.45±1.68) vs (4.94±0.93),(4.99±0.86)score] and pain[(3.55±0.50) vs (4.58±0.47),(4.58±0.62)score] were lower in DM+FSD group than those in DM group and NC group(P0.05). Logistic regression analysis showed that age,BMI and HbA1c were risk factors for FSD in T2DM patients (P=0.029,0.001,0.022,respectively). Conclusion The occurrence of FSD is common in female type 2 diabetic patients. BMI and HbA1c are the main risk factors for FSD.

Methods:Children born in the first ultrasound examination in 72h, the anterior temporal skull detect cerebral artery, middle cerebral artery blood flow dynamics, the main parameters: the peak systolic blood flow velocity(Vs), end-diastolic velocity(Vd), resistance index(RI). Results: Seventy cases in 90 cases of brain injury, to different extent in the positive rate of 77.8%.Conclusion: Cerebral ultrasound can early detect brain tissue structure change and hemodynamic changes, ultrasonic has the advantages of bedside examination.

Objective:To retrospectively analyze the CT features among gastric cancer, gastric lymphoma and gastric malignant stromal tumors, evaluated the application value of MDCT in gastric malignant tumor.Methods: Sixty two patients with gastric malignancies tumor confirmed by pathology underwent MDCT plain scanning and triphasic contrasted scanning before operation. Referring to the pathological results, to observe the CT signs, include: scope of tumors and involvement, the density of tumors and enhanced characteristics, mucosal membrane, the change of the stomach softness, peripheral lymph node enlargement, the involvement of other organs, and to make statistical analysis for imaging features of these tumors byx2 testing.Results: Sixty two cases of gastric malignant tumor included 42 cases of gastric carcinoma, 12 cases of gastric lymphoma and 8 cases of gastric malignant stromal tumor, were confirmed by postoperative pathology. The CT manifestations were as follows: the stomach mucous membrane surface involvement in all the gastric cancer, the positive rate was significantly higher than gastric lymphoma and stromal tumor. The area involved by lymphoma were more than two partitions, significantly higher than that of gastric cancer and stromal tumor (x2=36.643,x2=20.00;P<0.05). 39 cases (92.8%) of gastric cancer and 8 cases (100%) of malignant stromal tumors were significantly enhanced, but all gastric lymphoma were not obvious enhancement. The positive rate of enlarged lymph nodes in gastric cancer(61.9%) and lymphoma(83.3%) were higher than stromal tumor(12.5%)(x2=9.731,x2=6.603;P<0.05).Conclusion: The CT manifestations of the gastric cancer, lymphoma, malignant stromal tumors had different characteristics. Multislice CT for the diagnosis and differential diagnosis of gastric malignant tumor has important value.

Objective To compare the effects of different doses of adenosine(AD)on the left coronary blood flow(CBF)and left coronary flow reserve(CFR)in mini-swine.Methods By using Doppler flow wire and intracoronary ultrasound imaging catheter at the middle segment of LAD,coronary average peak flow velocity(APV)and lumen area were measured in 10 healthy mini-swines.CBF was calculated from the equation of 0.6 APV times 0.5 lumen area.Intracoronary bolus injection of different dose AD(12μg,18μg,24 μg and 36μg,separately)was used to induce maximal coronary hyperemic reaction.CFR was defined as the ratio of hyperemic CBF to resting CBF.Results Intracoronary bolus injection of 18μg AD level induced a maximal coronary vasodilation.Significant difference in the hyperemic CBF existed between 12 μg AD dose and 24μg or 36μg AD dose(P<0.05).No significant difference in heart rate,blood pressure,rCBF and CFR was found among three different AD dose groups(18μg,24μg,and 36μg).Conclusion CBF and CFR in mini-swine can be effectively assessed by bolus injection of AD with the dose of(18~24)μg.

Objective By using percutaneous endovascular microcatheter technique to establish an animal model of coronary microembolization in mini-swine which is suitable for long-term observation.Methods Coronary microembolization was established in 10 mini-swine by infusing 15 × 10~4 microspheres (φ45μm) selectively into the left anterior descending artery (n = 10). Coronary flow reserve (CFR) was measured by Doppler wire and left ventricular eject fraction (EF) was assessed by echocardiography.Hematoxylin and eosin (HE) staining and nitroblue tetrazolium (NBT) dye were used to demonstrate the presence of microembolization after the procedure of coronary microembolization. The ultra-structures of cardiomyocyte were observed by transmission electron microscopy (TEM). Before sacrifice, the CFR measurement and coronary angiography were performed again in survival animals. Results The coronary microvascular integrity (CFR < 2.0) and left ventricular function (EF < 50% ) were damaged by coronary microembolization. One month after the procedure, all the 10 animals survived and were able to receive the angiography and CFR measurement again. HE staining and NBT dye could demonstrate the presence of microembolization. The edema and fibrosis of cardiomyocytes could be revealed with TEM. Conclusion The animal model of coronary microembolization can be established in mini-swine by using percutaneous endovascular microcatheter technique. The model is suitable for long-term observation, the preparation is technically-simple and minimally-invasive with very low mortality. Therefore, this kind of animal model is an ideal experimental form for studying the mechanism of coronary microembolization.

Objective To explore body image(BI),prosthesis satisfaction and quality of life(QOL)of amputees from Wenchuan earthquake and the relationship among them to provide evidence for the intervention.Methods A total of 21 amputees accepted a survey regarding QOL,BI,and satisfaction with prosthesis.The scale of SF-36 was used to assess QOL.Amputees Bodily Image Scale(ABIS)was;used to assess BI.A self-design questionnaire wag used to evaluate the prosthesis satisfaction of amputees.Results The scores of SF-36,ABIS,and prosthesis satisfaction wag 41.90 ±15.00,64.58 ±10.60,and 59.52±11.06,respectively.The ABIS scores in women(65.07 ±12.10)were hisher than that in men(63.20 ±5.0)(P<0.05).There was a positive correlation between score of QOL and prosthesis satisfaction,but a negative correlation between QOL and BI.The negative correlations were also observed between BI and Vitality,BI and mental health.Prosthesis satisfaction had positive correla tions with education level.QOL and mental health.Conclusion The body image disturbance(BID)and dissatis faction with prosthesis may cause negative influences on the amputees'quality of life.Relevant intervention should be provided of amputees handling their prostheses and body images correctly and raising QOL.

Objective To explore the influencing factors during long-distance transportation and medical rescue of trauma patients in Wenchuan earthquake. Methods After Wenchuan earthquake, based on prehospital care scheme, we organized medical coordinators and psychologically trained medical staff in emergency medicine, cardiovascular medicine and logistics to transport trauma patients and pro-vide early psychological intervention during whole course of transportation. Results A total of 162 trauma patients were safely transported from Sichuan province to Chongqing relying on prehospital care scheme, equipment supplies, integrated rescue mode and psychological intervention during the transporta-tion. Conclusions Medical rescue mode and psychological intervention indicate that it is necessary to conduct psychological intervention for better transportation of patients for further medical treatments. In the meantime, we should establish medical rescue system for coping with psychological stress, improve rescue preparation scheme and professional medical care team for long-distance transportation and guaran-tee sufficient medical supplies in station hospitals.

ABSTRACT
The protection of ginseno-side (GNS ) isolated from leaves and stems on cultured cell from virus were observed. The results showed that GNS, especially the Rb1 monomer could suppress replication of virus such as HSV -Ⅰ , HSV-Ⅱ ,ADV- Ⅲ and VSV, and protect the cultured cells from virus infec-tion. A cream containing GNS was used in the treatment of herpes lebialis infected by HSV- I and 27 of 31 (87. 1%) cases were cured.
