ABSTRACT

OBJECTIVE@#To investigate the expression of CD56 in multiple myeloma (MM) cells and its relationship between extramedullary disease and extramedullary relapse.@*METHODS@#Clinical data of 99 patients with MM treated in our hospital from January 2015 to December 2019 was retrospectively analyzed. The patients were divided into positive group and negative group according to the expression of CD56. The relationship between CD56 and multiple myeloma extramedullary disease, extramedullary relapse was analyzed.@*RESULTS@#Among 99 newly diagnosed patients with MM, the positive rate of CD56 was 65%, and the incidence of extramedullary disease of patients in the CD56 positive group was lower than that in the CD56 negative group (17.19% vs 48.57%) (P<0.01). Meanwhile, the incidence of extramedullary relapse of patients in the CD56 positive group was lower than that in the CD56 negative group (1.56% vs 34.29%) (P<0.01).@*CONCLUSION@#CD56 is highly expressed in MM, and its low expression is associated with the occurrence of extramedullary disease and extramedullary relapse, which suggests that CD56 may be an important indicator for predicting the occurrence of extramedullary disease and extramedullary relapse.

ABSTRACT

OBJECTIVE@#To analyze the influence of serum homocysteine (Hcy) levels to the prognosis of newly diagnosed multiple myeloma (MM) patients, and to explore related factors affecting the prognosis of the patients.@*METHODS@#The clinical pathological data of 180 newly diagnosed MM patients treated in our hospital from March 2013 to February 2015 were collected, and the patients were divided into high and low Hcy groups based on the median Hcy. The survival curves of the patients in the two groups were drawn to compare the differences of the survival; univariate and multivariate survival analysis was used to observe the influence of serum cysteine to the prognosis of newly diagnosed MM patients; the clinicopathological data of the patients with high and low Hcy in the two groups was compared, Pearson test was used to further analyzes the relationship between Hcy and different factors, and explores the related factors of Hcy affecting the prognosis of the patients.@*RESULTS@#The median survival times of patients in the high and low Hcy groups were 32 (5-59) and 41 (7-71) months, respectively. The 3-year survival rate of the patients in high Hcy group was significantly lower than those in low Hcy group, and the difference shows statistically significant (P<0.05). The results of univariate survival analysis showed that the OS of newly diagnosed MM patients whom with advanced age, high bone disease grade, high-level bone marrow plasma cell count, LDH, C-reactive protein, Cr, β@*CONCLUSION@#Serum Hcy level has a correlation trend with the survival of newly diagnosed MM, which is affected by factors such as Hb.

ABSTRACT

OBJECTIVE: To establish a solvent desorption-gas chromatography method for determination of methyl pentane in workplace air. METHODS: Methyl pentane in workplace air was collected with activated carbon tube and desorbed with carbon disulfide, then separated by DB-1 capillary column, detected by flame ionization detector, and quantified by standard curve method. RESULTS: Good linearity was obtained in the range of 1.98-6 600.00 mg/L with the correlation coefficient of 0.999 9. The minimum detection limit and the minimum quantification limit were 0.06 and 0.20 mg/L, respectively. The minimum detection concentration and minimum quantification concentration was 0.04 and 0.14 mg/m~3, respectively(calculated by collecting 1.5 L of air sample). The average desorption efficiency was 97.3%-102.2%. The within-run relative standard deviation(RSD) and the between-run RSD were 0.4%-0.9% and 0.3%-3.0%, respectively. The sampling efficiency was 96.7%-100.0% and the penetration capacity was 8.68 mg. The samples can be stored at room temperature for at least 14 days. CONCLUSION: This method is suitable for methyl pentane detection in workplace air.

ABSTRACT

Objective:To explore the diagnostic value of the serial position effect combined with delayed recall of auditory verbal learning test in diagnosing the elderly patients with different levels of cognitive impairment.Methods:A total of 310 subjects were enrolled and divided into three groups: the normal control group(NC group, n=128), the mild cognitive impairment group(MCI group, n=133)and the mild Alzheimer's disease group(mild AD group, n=149)matched for gender, age and education level.Comprehensive neuro-psychological tests were performed on all subjects, and the Auditory Verbal Learning Test-Huashan version(AVLT-H)was used to analyze the serial position effect.Results:There were significant differences in the primacy effect of the second and third time of immediate recall, short-term and long-term delayed recall among the three groups( P<0.05), but no difference was found in the recency effect between the MCI and mild AD groups( P>0.05). The long-term delayed primacy effect combined with the short-term delayed recall showed a better validity to distinguish those elderly subjects with different levels of cognitive impairment than other memorial indexes, and the correlative sensitivity and specificity were 83.21% and 71.43%(NC group vs.MCI group), 85.71% and 92.86%(NC group vs.mild AD group), 66.67% and 75.57%(MCI group vs.mild AD group), respectively. Conclusions:The long-term delayed primacy effect combined with the short-term delayed recall can assist in distinguishing elderly patients with different levels of cognitive impairment.

ABSTRACT

METHODS@#The clinical data of 15 patients with primary diffuse large B-cell lymphoma treated in our hospital from January 2013 to December 2020 were collected, the clinical data and prognosis of the patients were analyzed retrospectively.@*RESULTS@#The median age of the 15 patients was 59 (19-89) years old; among the patients, 7 were males and 8 were females, ostealgia was the initial symptom. The pathological types of the 15 patients were diffuse large B-cell lymphoma (DLBCL), 5 cases of Has type GCB subtype (5/15), and 10 cases of Non-GCB subtype (10/15). After 15 patients were diagnosed, 11 patients (11/15) received chemotherapy, 3 patients (3/15) received surgery, and 1 patient was untreated (1/15), median chemotherapy courses was 5 (1-9). 8 patients have achieved complete remission (8/15), 3 patients achieved partial remission (3/15), and 1 patient achieved stable disease (1/15), 1 patient was lost to follow-up (1/15), 1 patient was untreated (1/15), and 1 patient was progression of disease (1/15). Age, pathological subtype, sex, stage, β2-MG level, LDH level, and the using of rituximab were not correlated with the complete remission rate of the patients(P>0.05), while the IPI score was correlated with the recent complete remission rate (P<0.05). The median follow-up time was 19 (1-38) months, 10 patients survived, in which 6 cases were still in complete remission, and the median time to progression-free survival was 15 (1-38) months.@*CONCLUSION@#The first symptom of primary bone diffuse large B-cell lymphoma is bone pain, the main pathological subtype is Non-GCB, the optimal treatment is combined chemotherapy, and the IPI score is related to the prognosis of the treatment.

ABSTRACT

OBJECTIVE@#To investigate the genotypes and clinical characteristics of thalassemia on children in Wuhan region.@*METHODS@#A total of 159 patients diagnosed as thalassemia in Maternal and Child Health Hospital of Hubei Province, Tongji Medical College, Huazhong University of Science and Technology from December 2017 to December 2019. The patients were retrospectively analyzed for their types of mutations, detection rates and clinical characteristics.@*RESULTS@#Among the 422 samples, 159 samples were finally diagnosed as thalassemia through genetic testing, the total detection rate was 37.68%. The detection rate of α, β and αβ-thalassemia was 17.30%, 20.14% and 0.24% respectively. Among α-thalassemia, αα/-SEA was the most common one, with a composition ratio of 68.49%(50/73), followed by αα/-α3.7 (19.18%), αα/-α4.2 (6.85%) and αα/ QS (1.37%). 9 types of β-thalassemia gene mutations were detected, and the most common three mutations were IVSII-654(C→T), with a composition ratio of 40.00%, CD41-42(-TTCT) (20.00%) and CD17(A→T)(16.47%). Two novel mutations of β-thalassemia, HBB: c.92-2A>T and HBB:c.-23A>G were detected. Among all the positive patients, 134 (84.28%) were 0-3 years old, 19 (11.95%) were 4-6 years old, and 6 (3.77%) were 7 years of age or older. There were 147 patients with mild anemia (92.45%), 11 patients with moderate anemia (6.92%), and 1 patients with severe anemia (0.63%). The MCV of 94(59.12%) patients was lower than 65 fL, and that of 51(32.08%) patients was between 65 fL and 80 fL, while 14(8.81%) patients was higher than 80 fL. MCV in β-thalassemia group was lower than that in α-thalassemia group, and the difference showed statistically significant (P<0.05).@*CONCLUSION@#The genotypes of thalassemia in children in Wuhan area are diverse, and most of them are mild thalassemia, and diagnosed under 3 years old. Children with β-thalassemia have smaller red blood cell volumes than those with α-thalassemia.

ABSTRACT

Oncogene StarD4 had the function of promoting proliferation and metastasis of triple-negative breast cancer (TNBC), but its clinical value and molecular mechanism are unknown. This paper found that StarD4 was highly expressed in cancer tissues of TNBC patients, and higher expression level of StarD4 in TNBC patient resulted in poorer prognosis. Based on transcriptomics of MDA-MB-231 cell model, the results of bioinformatics analysis showed that down-regulated expression level of StarD4 led to overall downregulation of cholesterol-relative genes and significant enrichment of cancer mechanism and pathway. Further analysis and investigation verified that StarD4 might cross-promote the protein stability of receptor ITGA5 through the cholesterol pathway to enhance TNBC progression, which provides guidance for clinical application of TNBC diagnosis and treatment.

ABSTRACT

NAC(NAM/ATAF/CUC) protein plays an important role in plant growth and development, secondary cell wall formation and stress response. In this study, based on the sequencing data of Angelica dahurica, the NAC family was systematically analyzed using bioinformatics methods and its expression pattern was analyzed. Studies showed that 75 candidate genes had been selected from the NAC transcription factor family of A. dahurica, with the protein size of 148-641, all of which were unstable hydrophilic proteins. Most NAC proteins were localized in the nucleus, and had complete NAC domain. Phylogenetic analysis of NAC family proteins of A.dahurica and Arabidopsis thaliana showed that among the 17 subfamilies, NAC members were unevenly distributed in each subfamily, indicating that the evolution of species is developing in multiple directions. Among them, ANAC063 subfamily contained no NAC sequence of A. dahurica, which might be due to the functional evolution of the species. Analysis of protein transmembrane structure and signal peptide showed that NAC transcription factor could carry out transmembrane transportation, but its signal peptide function had not been found. Expression analysis showed that most transcription factors responded to abiotic stress and hormones to varying degrees, and the effects of hormones were obvious, especially ABA and IAA. In different organs of A. dahurica, most members of the NAC family had higher expression in root phloem, followed by root xylem. This study lays a foundation for further research on the function of A. dahurica NAC transcription factor and for solving the biological problems of A. dahurica.

ABSTRACT

Remitting seronegative symmetrical synovitis with pitting edema(RS3PE),the inflammatory arthritis attacking mainly elderly males,is characterized by symmetrical synovitis with pitting edema of the dorsum of hands and feet and the absence of rheumatoid factor.RS3PE commonly accompanies malignant tumor,infections and other diseases.Here we report a case of RS3PE associated with lung malignancy and review other six cases to summarize the clinical features,treatment and prognosis.

ABSTRACT

OBJECTIVE:To analyze the metabolites of tetrahydroxystilbene glucoside (THSG)and speculate its metabolism pathway in rats. METHODS :Male SD rats were randomly divided into plasma group (n=3),urine group (n=3),bile group (n=3),and tissue group (n=9). Each group was given single dose of THSG 200 mg/kg intragastrically. Plasma samples 10,30 min and 1,1.5,2,4 h after medication ,the unrine 0-6 h after medication ,the bile 0-4 h after medication ,the tissue of heart , liver,spleen,lung,kidney and stomach 30 min and 1,2 h after medication (3 at each time point )were collected respectively.After precipitated with methanol ,the metabolites of samples were analyzed and identified by UHPLC-Q-Exactive Orbitrap MS and mass loss filtration (MDF). Its metabolism pathway was speculated. RESULTS:In the blood ,urine,bile,heart,liver,spleen, lung,kidney,stomach samples ,6,7,11,1,5,1,3,4,4 metabolites were detected ,including two phase Ⅰ(hydrolysis, hydrogenation and hydroxylation )metabolites,18 phase Ⅱ(glucuronic acid binding and sulfation )metabolites. There were 12 glucuronic acid binding products. CONCLUSIONS:Most of the metabolites of THSG are found in bile ,mainly glucuronic acid binding products of phase Ⅱ metabolite THSG ; main metabolic pathways involve glucose hydrolysis , hydrogenation, hydroxylation,glucuronic acid binding and sulfation.

ABSTRACT

OBJECTIVE:To establ ish the fingerprint of Sojae Semen Nigrum and content determination method of 5 kinds of isoflavones,so as to provide reference for controlling its quality better. METHODS :HPLC method was adopted to establish the fingerprint and detect the contents of 5 kinds of isoflavones. The determination was performed on Phenomenex C 18 column with mobile phase consisted of acetonitrile- 0.12% formic acid solution (gradient elution )at the flow rate of 1 mL/min. The detection wavelength was set at 260 nm;the column temperature was 30 ℃ and sample size was 10 μL. Using daidzin as reference,HPLC fingerprints of 12 batches of samples were determined. The similarity of 12 batches of samples was evaluated by TCM Chromatographic Fingerprint Similarity Evaluation System (2012A) to confirm common peak. Cluster analysis and principal component analysis were performed by using SPSS 20.0 software and SIMCA 13.0 software. RESULTS :There were 19 common peaks in HPLC fingerprints of 12 batches of samples ,the similarity of which was higher than 0.94. Totally 5 components were identified,such as daidzin ,glycitin,genistin,daidzein,genistein. Cluster analysis showed that 12 batches of Sojae Semen Nigrum were clustered into 2 categories,i.e. S 1-S3 clustered into one category ,and S 4-S12 clustered into the other category. By principal component analysis ,the contribution rates of two principle components were 53.261% and 40.715%;accumulative contribution rate was 93.976%. The linear range of above 5 components were 5.97-191.00 µg/mL(r=0.999 9),1.05-33.46 µg/mL(r=0.999 9), 8.93-285.61 µg/mL(r=0.999 5),0.82-26.33 µg/mL(r=0.999 9),0.93-29.64 µg/mL(r=0.999 7),respectively. The limits of quantitation were 0.881 1,0.611 6,0.078 6,0.243 3,0.511 6 μg/mL,respectively. The limits of detection were 0.264 3,0.244 7, 0.021 4,0.124 8,0.106 7 μg/mL,respectively. RSDs of precision ,stability,reproducibility and durability tests were all lower than 5%. Recoveries were 95.15%-96.56%(RSD=0.51%,n=6),98.52%-103.45%(RSD=1.88%,n=6),95.37%-97.91% (RSD=0.95%,n=6),99.75%-102.00%(RSD=0.78%,n=6),100.26%-103.65%(RSD=1.21%,n=6). Among 12 batches of Sojae Semen Nigrum ,the contents of above 5 components were 0.178 3-0.265 9,0.021 7-0.096 2,0.288 5-0.597 2,0.014 1- 0.058 8,0.012 9-0.082 9 mg/g. CONCLUSIONS :Established HPLC fingerprint and content determination method of 5 kinds of isoflavones can be used for quality control of Sojea Semen Nigrum. The Isoflavone components are similar ,but the contents are different among Sojae Semen Nigrum from different producing areas.

OBJECTIVE@#to explore the value of capillary electrophoresis in screening β- thalassemia of children, and to establish the cutoff values of HbA2 and HbF in our laboratory.@*METHODS@#The data of hemoglobin capillary electrophoresis and genetic diagnosis of β- thalassemia from 886 examined children were retrospectively analyzed. The cutoff values of HbA2 and HbF were determined by ROC curve.@*RESULTS@#The cutoff value of HbA2 screening minor β- thalassemia was 3.65%, the specificity was 0.996, and the sensitivity was 0.995. The cut-off value of HbF for screening minor β- thalassemia was 1.45%, specificity was 0.751 and sensitivity was 0.675. Thus, 1 case with codon5 (CCT→C) mutation, 1 case with SEA -HPFH β deletion, 1 case with - 28 (A→G) merger IVS-Ι-128 (T→G) double heterozygous mutations yet were found out, 1 case with 47 bp β gene missing has not yet been reported in literature.@*CONCLUSION@#Capillary electrophoresis has more high sensitivity and specificity in the screening of β- thalassemia in children, especially for the detection of rare β- thalassemia.

ABSTRACT

Exosomes are spherical microvesicles(30-100 nm)secreted by a variety of cells and contain a variety of proteins, lipids, mRNAs, and microRNAs.These molecules into exosomes can be transferred to adjacent cells and distant tissues or cells by exosomes to develop full effects.Exosomes-released microRNAs are involved in the pathogenesis of Alzheimer's disease(AD)by regulating the abnormal expression of amyloid β-protein(Aβ)and p-Tau protein, initiating the inflammatory response by interacting with toll-like receptors.In addition, exosomal microRNAs can be used as a potential therapeutic target for AD.Exosomes as good carriers have great research values.This review summarized the literatures related to the role of exosomal microRNAs in the development of AD and the potential therapeutic effect on AD.A large amount of evidence indicates that expression disorder of exosomal microRNA plays an important role in the pathogenesis of AD and can be used as a new potential biomarker and therapeutic target for the diagnosis and treatment of AD.

ABSTRACT

Objective:To investigate the relationship between atherosclerotic plaque characteristics of the middle cerebral artery(MCA)and acute cerebral ischemic symptoms by using high-resolution magnetic resonance imaging(HR-MRI).Methods:A retrospective analysis of clinical and imaging data was performed in 136 patients[100 males, aged(60.8±11.4)years]with MCA atherosclerotic plaque who underwent HR-MRI.Patients were divided into the acute symptomatic group(n=73)and the non-acute asymptomatic group(n=63)according to whether the patients had acute ischemic symptoms in the MCA territory within two weeks.The basic demographic characteristics, clinical risk factors, MCA plaque enhancement, maximum plaque length, maximum wall thickness, ratio of maximum plaque length to maximum wall thickness and luminal stenosis were compared between the two groups.Binary Logistic regression was used to estimate the association between MCA plaque characteristics and acute ischemic symptoms in the corresponding vascular territory.Results:Compared with the non-acute symptomatic group, the acute symptomatic group had a greater prevalence of diabetes mellitus(50.7% or 30/73 vs. 30.2% or 19/63, χ2=5.882, P=0.015), a higher occurrence of enhanced plaques(87.7 % or 64/73 vs.46.0% or 29/63, χ2=27.118, P<0.001), greater maximum plaque length[(6.7±3.7 mm) vs.(4.9±2.3)mm, t=-3.330, P=0.001], greater maximum wall thickness[(1.5±0.8)mm vs.(1.0±1.0)mm, t=-2.231, P=0.027]and more severe luminal stenosis[(66.4±31.3)% vs. (30.7±25.3) %, t=-7.354, P<0.001]. Logistic regression analysis showed that MCA enhanced plaque( OR=4.492, 95% CI: 1.762-11.449, P=0.002)and luminal stenosis( OR=1.032, 95% CI: 1.018-1.047, P<0.001)were independently associated with acute ischemic symptoms. Conclusions:Compared with luminal stenosis, MCA enhanced plaque has a stronger correlation with acute ischemic symptoms and significant plaque enhancement may be an important indication of its instability.

ABSTRACT

Objective:To compare and analyze the clinicopathological data of elderly patients with versus without hepatitis B virus(HBV)antigen deposition in renal tissue, and to investigate the clinicopathological features in elderly patients with HBV associated nephritis(HBV-GN).Methods:The clinicopathological data of 33 patients with HBV-GN and 27 patients with hepatitis B and nephritis(HBV-CG)aged ≥60 years in the 90 th Hospital of the United Nations Support Force were retrospectively analyzed and statistically analyzed. Results:(1)Both the HBV-GN and HBV-CG groups were often men[26(78.8%) vs. 21(77.8%)]and had high rate of nephritic syndrome[18(54.5%) vs.15(55.6%)]. And patients in the HBV-GN group had higher incidence of hypertension than that in the HBV-CG group; (2)Membranous nephropathy(MN)was the main pathological type in both groups, and the rate of membrane proliferative glomerulonephritis(MPGN)was more found in the HBV-GN group than the HBV-CG group; (3)The proportion positive HBsAg, HBeAg and HBcAb test resultsand high viral load was significantly higher in the HBV-GN group than that in the HBV-CG group; (4)Multiple immune complex deposits were observed in the renal tissues of HBV-GN group, which was called the phenomenon of "all bright" ; (5)The pathological features in the HBV-GN group showed untypical manifestation of MN, the occasionally observed crescent formation, and the thickening of basement membrane in focal segmental glomerulosclerosis; (6)The incidence and the severity of renal tubule and interstitial lesions in HBV-GN group were significantly higher than those in HBV-CG group. Conclusions:Although there are many similarities in clinical manifestations and testing indicators between the elderly HBV-GN and HBV-CG patients, HBV-GN patients are still significantly different from HBV-CG patients in terms of serum virology and pathological changes.Therefore, we should pay attention to the serum HBV replication and renal pathological changes to provide a clinical basis for treating this kind of elderly patients.

In the process of experimental teaching, anatomy teachers expand the anatomy knowledge to basic scientific research techniques, so as to enhance students' confidence in participating in innovative experimental projects. Meanwhile, the teachers combine introduction of the anatomical structures with hot medical research issues, in order to help students to select appropriate innovative experimental topics. What's more, the new media teaching is integrated into the teaching of anatomy, enlightening students to perfect the experimental designs. Finally, in the teaching practice of regional anatomy, students are guided to carry out innovative experiments and write research papers. Abovementioned measures not only improve the teaching quality of anatomy, but also improve the students' scientific research ability, laying a foundation for them to participate in innovative experimental projects.

OBJECTIVE@#To study the association between the expression of the MDR3 gene and the pathogenesis of parenteral nutrition-associated cholestasis (PNAC) in preterm infants.@*METHODS@#Among the preterm infants who were admitted to the hospital from June 2011 to November 2017 and received parenteral nutrition for more than 14 days, 80 who did not develop PNAC were enrolled as non-PNAC group, and 76 who developed PNAC were enrolled as PNAC group. On days 1, 14, 30, 60 and 90 after birth, serum hepatobiliary biochemical parameters [alanine aminotransferase (ALT), total bilirubin (TBil), direct bilirubin (DBil), total bile acid (TBA) and gamma-glutamyl transpeptidase (γ-GT)], fibrosis indices [hyaluronic acid, laminin, procollagen III N-terminal peptide and type IV collagen] and clinical manifestations were observed. Real-time quantitative PCR was used to measure the mRNA expression of MDR3 in both groups, and the correlation between the mRNA expression of MDR3 and serum hepatobiliary biochemical parameters was analyzed.@*RESULTS@#In the PNAC group, serum levels of hepatobiliary biochemical parameters and fibrosis indices increased on day 14 after birth and reached the peak on day 30 after birth, followed by a reduction on day 60 after birth. On days 14, 30, 60 and 90 after birth, the PNAC group had significantly higher serum levels of hepatobiliary biochemical parameters and fibrosis indices than the non-PNAC group (P<0.05). The PNAC group had higher relative mRNA expression of MDR3 in peripheral blood cells than the non-PNAC group (P<0.05). In the PNAC group, the relative mRNA expression of MDR3 in peripheral blood cells was negatively correlated with serum levels of hepatobiliary biochemical parameters (ALT, TBil, DBil, TBA and γ-GT) (P<0.001).@*CONCLUSIONS@#High mRNA expression of MDR3 in preterm infants may be associated with the development of PNAC, and further studies are needed to identify the mechanism.

ABSTRACT

ObjectiveTo investigate the expression and clinical significance of protein kinase CK2β in hepatocellular carcinoma (HCC) tissue and the association of CK2β expression with patient prognosis. MethodsHCC tissue and adjacent tissue samples were collected from 127 HCC patients Who were diagnosed in Department of Hepatopancreatobiliary Surgery, The first Affiliated Hospital of Zhengzhou University from Januany 2012 to June 2013, and immunohistochemistry was used to measure the expression of CK2β. The association of CK2β expression with clinical features of HCC patients was analyzed. A total of 20 HCC tissue samples, 20 adjacent tissue samples, 20 cirrhotic tissue samples, and 20 normal liver tissue samples were collected from March to June 2018 in our hospital, and Western blot and real-time PCR were used to measure the protein and mRNA expression of CK2β in these tissue samples. A one-way analysis of variance was used for comparison of means between multiple groups, and Bonferroni test was used for further comparison between two groups; the chi-square test was used for comparison of categorical data between groups. The Mann-Whitney U test or the Kruskal-Wallis H test was used to investigate the association of CK2β expression with clinical features. The Kaplan-Meier method was used for survival analysis, and the log-rank test was used for comparison between groups. ResultsWestern blot and real-time PCR showed that the expression rate of CK2β in HCC tissue was significantly higher than that in adjacent tissue, cirrhotic tissue, and normal liver tissue (P<0.05); there was no significant difference in the expression of CK2β between adjacent tissue and cirrhotic tissue (P>0.05), and the expression of CK2β in adjacent tissue and cirrhotic tissue was significantly higher than that in normal liver tissue (P<0.05). Immunohistochemical staining showed that there was a significant difference in the positive rate of CK2β between HCC tissue and adjacent tissue in the 127 HCC patients (85.8% vs 63.0%, P<0.001). There was a significant difference in CK2β expression distribution between HCC patients with different ages (Z=-2.277, P=0.023), presence or absence of liver cirrhosis (Z=-2.144, P=0.032), different tumor sizes (Z=-2.289, P=0.004), or different Edmondson-Steiner pathological grades (χ2=8.210, P=0.016). The Kaplan-Meier survival curve analysis showed that the strongly positive CK2β expression group had a significantly shorter postoperative survival time than the moderately positive, weakly positive, and negative CK2β expression groups (all P<0.001). ConclusionCK2β may be involved in the development and progression of HCC, and its positive expression is associated with the prognosis of HCC patients.

ABSTRACT

The aim of this paper was to investigate the mechanism and effect of psoralen and isopsoralen in the treatment of lipid accumulation in LO2 cells. Human LO2 cells nonalcoholic fatty liver models were established by using palmitic acid( PA). Then psoralen and isopsoralen were administered for intervention. Intracellular triglyceride( TG) and total cholesterol( TC) content,the cell supernatant alanine aminotransferase( ALT) and aspartate aminotransferase( AST) levels were determined by enzyme method. Cell supernatant proinflammatory cytokines( IL-6,TNF-α) and chemokines( IL-8,MCP-1) were determined by ELISA method. Western blot method was conducted to detect the protein expression of intracellular nuclear factor( NF-κB) p65 phosphorylation( p-p65),nonphosphorylated protein( p65),and transforming factor TGF-β1. Result showed that as compared with the model group,intracellular TG and TC levels,the cell supernatant ALT and AST levels,proinflammatory cytokines and chemokines were decreased( P < 0. 01,P <0. 05); the p-p65/p65 ratio and TGF-β1 protein expression were also significantly decreased( P< 0. 01,P< 0. 05) in psoralen intervention group. As compared with the model cells,intracellular TG content had no significant changes,but all the other indexes were reduced( P<0. 01,P<0. 05) in the cells of isopsoralen intervention group. Psoralen exhibited better effect than isopsoralen( P< 0. 01,P<0. 05). It is concluded that psoralen could improve the adipogenesis of LO2 cells induced by PA; both psoralen and isopsoralen are effective in ameliorating LO2 cells injury induced by PA,reducing inflammation via inhibiting the activation of NF-κB and down-regulating the expression of TGF-β1.
