ABSTRACT
The present study investigated the chemical constituents of Caesalpinia decapetala in the Fabaceae family. The chemical constituents were isolated and purified by chromatographies with silica gel, RP-C_(18), Sephadex LH-20, and preparative HPLC, and their structures were determined based on the spectroscopic data and physicochemical properties, as well as relevant references. Three pairs of new dibenzoxocin derivatives were isolated from 70% ethanol extract of C. decapetala and identified as protosappanoside A(1 a), isoprotosappanoside A(1 b), protosappanoside B(2 a), isoprotosappanoside B(2 b), protosappanoside C(3 a), and isoprotosappanoside C(3 b), respectively.

OBJECTIVE@#To investigate the effect of Modified Xiaochaihu Decoction (MXD, ) on collagen degradation in rats with chronic pancreatitis (CP).@*METHODS@#Rats were injected dibutyltin dichloride (DBTC, 7 mg/kg of body weight) into the right caudal vein to induce CP model. Thirty heallhy male Wistar rats were randomly divided into three groups by a random number table: the control, the model and the treatment groups. Rats of treatment group were administered MXD (10 g/kg of body weight) orally once daily starting from the day post-model establishment. Pancreatic tissues were harvested after 28-day feeding and fibrosis was evaluated by picro-sirius red staining. The contents of collagen type I and III were detected using enzymelinked immunosorbent assay (ELISA), the expression of matrix metalloproteinase 13 (MMP13) and tissue inhibitor of metalloproteinase 1 (TIMP1) was analyzed by Western blot and real-time polymerase chain reaction (PCR).@*RESULTS@#The fibrosis scoring of pancreatic tissues, the concentrations of collagen type I and III, the expression levels of MMP13 and TIMP1 proteins and mRNA in the model group were all increased compared with the control group (P0.05).@*CONCLUSIONS@#MXD could promote collagen degradation and reverse pancreatic fibrosis in CP rats via a mechanism involve up-regulation of MMP13 expression.
ABSTRACT
Modified Da-chai-hu Decoction (MDD), a traditional Chinese medicinal formulation, which was empirically generated from Da-chai-hu decoction, has been utilized to treat severe acute pancreatitis (SAP) for decades. The aim of the present study was to explore its potential organprotective mechanism in SAP. In the present study, rat SAP model was induced by retrograde injection of 3.5% sodium taurocholate into the biliopancreatic duct, MDD (23.35 g/kg body weight, twelve times the clinical dose) were orally given at 2 h before and 10 h after injection. At 12 h after model induction, blood was taken from vena cava for analysis of amylase, diamine oxidase (DAO), pulmonary surfactant protein-A (SP-A), and C-reactive protein (CRP). Histopathological change of pancreas, ileum and lung was assayed by H&E staining, myeloperoxidase (MPO) activity were determinated using colorimetric assay, and the expressions of occludin and nuclear factor-κB (NF-κB) were detected by real-time RT-PCR and western blot, respectively. In addition, the tissue concentrations of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and monocyte chemoattractant protein-1 (MCP-1) were measured by enzyme-linked immunosorbent assay (ELISA). The results showed that in SAP rats, MDD significantly alleviated histopathological damage, depressed the MPO activity and the concentrations of TNF-α, IL-1β, and MCP-1 of pancreas, ileum and lung, and reduced the serum levels of amylase [(3283.4 ± 585.5) U·Lvs (5626.4 ± 795.1)U·L], DAO [(1100.1 ± 334.3) U·Lvs (1666.4 ± 525.3) U·L] and CRP [(7.6 ± 1.2) μg·mLvs (17.8 ± 3.8) μg·mL]. However, the serum SP-A concentration [(106.1 ± 16.6) pg·mLvs (90.1 ± 14.9) pg·mL] was elevated when treated SAP rats with MDD. Furthermore, MDD increased the occludin expression and reduced the NF-κB expression in pancreas, ileum and lung of SAP rats. Our findings suggested that MDD administration was an effective therapeutic approach for SAP treatment. It could up-regulate occludin expression to protect intercellular tight junction and down-regulate NF-κB expression to inhibit inflammatory reaction of pancreas, ileum and lung.
ABSTRACT
Objective:To study the protective effect of Fuzheng Quxie prescription on liver injury by influencing the expressions of tumor necrosis factor (TNF)-α, apoptosis factor (Fas), apoptosis factor ligand (FasL), and macrophages (CD68) in the liver tissues of concanavalin A(ConA) model mice. Method:The sixty mice were randomly divided into normal group, model group,bicyclol group (62.5 mg·kg-1), and low, medium and high-dose Fuzheng Quxie prescription groups(50.3,67.0,83.8 g·kg-1). The normal group and the model group were given the equal volume of pure water for 7 d. They were fasted for 12 hours before the last administration. At 2nd hour after the last administration, phosphate buffer(PBS) was injected into the caudal vein of the normal group, and ConA (20 mg·kg-1) was injected into the caudal vein of the other groups for modeling. The animals were put to death six hours later after the injection, and the expressions of TNF-α, Fas, FasL and CD68 in liver tissues were observed by immunohistochemistry. Result:Compared with normal group, the expressions of TNF-α, Fas, FasL and CD68 in the liver tissues of the model group were significantly increased(Pα, FasL and CD68 in liver tissues of the bicyclol group were significantly decreased compared with the model group(Pα, FasL and CD68 in liver tissues were significantly decreased in medium and high-dose Fuzheng Quxie prescription groups (PPConclusion:Fuzheng Quxie prescription can effectively reduce the apoptosis of liver cells in ConA model mice by inhibiting Kupffer cells and Fas/FasL system activation.

Objective To investigate the results of MRI and electroencephalogram examination and pathological examination of drug refractory epilepsy. Methods 70 cases with drug refractory epilepsy were treated with systemic antiepileptic drugs, but they still could not control their condition, and they were given MRI, EEG and pathological examination respectively. MRI, EEG and pathological findings of 70 cases were recorded, and the conclusion was drawn after statistical analysis. Results Compared with pathological examination, the localization accuracy of MRI for epileptogenic foci and related lesions was 88.57%, the accuracy of localization of EEG for epileptogenic foci and related disease was 51.43%, the difference was statistically significant (P<0.05); MRI examination of the epileptogenic lesions and qualitative accuracy the rate of up to 75.71%. Conclusion According to the patients with drug intractable epilepsy, the coincidence rate of MRI examination and pathological examination is higher. Compared with EEG, it can provide clinicians with more reliable basis for the diagnosis and treatment of drug refractory epilepsy. It is helpful to protect the curative effect of these patients.

OBJECTIVES@#To investigate the population genetic polymorphisms of 24 Y-STR loci in unrelated individuals in Eastern Chinese Han population, and to compare the difference of Han group between Eastern China and Guangdong.@*METHODS@#The population genetics of 24 Y-STR loci in 268 unrelated Han individuals from Eastern China were analyzed by GFS 24 Y-STR amplification kit. The allele frequencies in Eastern Chinese Han population were compared with the data in Guangdong Han population, and the difference analysis between two groups was performed.@*RESULTS@#Among the 24 Y-STR loci of 268 unrelated Han individuals from Eastern China, 235 alleles and 267 haplotypes were observed. GD value ranged from 0.564 9 to 0.966 8. The difference between 12 loci (DYS622, DYS552, DYS443, et al.) of Han population in Eastern China and in Guangdong was statistically significance.@*CONCLUSIONS@#GFS 24Y STR amplification system shows favorable polymorphisms, which can be used in patrilineal genetic relationship identification.

The present study isolated 17 compounds from the tubers of Bletilla striata (Orchidaceae), using various chromatographic techniques. Their structures were identified based on their physical-chemical properties and spectroscopic analyses. Among them, two new 2-isobutylmalates, named bletimalates A (1) and B (2), together with other fifteen known compounds (3-17), were isolated and identified. Additionally, compounds 3, 4, and 8 were isolated from this plant for the first time.

<p><b>OBJECTIVE</b>To improve the diagnosis and treatment of testicular teratoma in children by analysis of clinical data.</p><p><b>METHODS</b>We retrospectively analyzed the clinical data about 64 cases of testicular teratoma treated in the Children's Hospital of Chongqing Medical University from 1995 to 2014.</p><p><b>RESULTS</b>Sixty-one of the cases presented painless scrotal mass with a sense of bearing down and the other 3 cases were confirmed because of empty scrotum diagnosed as cryptorchidism. The level of serum alpha fetal protein ( AFP) was obviously increased in 46 cases but normal in the other 18 preoperatively. Ultrasonography manifested abnormal inhomogeneous echo zones with calcification or necrosis. X-ray examination presented patchy or curvilinear high-density shadows in 28 cases. Forty-one of the patients underwent testis-sparing surgery (TSS) , 20 received high inguinal orchiectomy, and 3 refused surgical treatment. Pathological examination revealed 3 mature germinal layers in the 49 cases of mature teratoma and immature germinal tissue, including the original neural tube, and 11 cases of immature teratoma. The mature cases were exempted from chemotherapy, while the immature cases received the combination of cisplatin, etoposide, and bleomycin (PEB). The patients were followed up for 2 years postoperatively, which revealed no recurrence or metastasis.</p><p><b>CONCLUSION</b>Most children with testicular teratoma presented painless scrotal mass with a sense of bearing down and with abnormal serum AFP in most cases. Ultrasonography and plain radiography of the scrotum contribute to the diagnosis of the tumor. TSS is the main treatment option and intraoperative frozen-section can help the surgeons decide on the surgical mode. Postoperative chemotherapy is necessitated for immature teratoma but not for mature cases.</p>

OBJECTIVE@#To assess the patterns of linkage disequilibrium (LD) of 16 STR loci on X chromo- some and investigate the genetic stability.@*METHODS@#Genomic DNA samples extracted from blood stains from 500 unrelated individuals and 885 lineage members from Eastern Chinese Han population were genotyped through multiplex amplification using IDtyperX-16 kit by our independent research followed by capillary electrophoresis. LD was assessed by PowerMarker v3.25 software and mutation rate of every locus was analyzed.@*RESULTS@#LD were not found at the 16 X-STR loci. Allele mutations were observed at 10 loci. Among them, mutation rates of DXS6809 and DXS7132 were both up to 0.0048.@*CONCLUSION@#When the 16 X-STR loci included in IDtyperX-16 kit were used for parentage testing, product princi- ples can be applied to calculate the likelihood, but mutation should be taken into consideration in the case that the genotypes do not meet the genetic law (especially at DXS6809 and DXS7132).

Objective To investigate the expression of interleukin-1 β(IL-1 β) and apoptosis in the hippocampus of rats with prolonged febrile seizures(PFS),and to provide the theoretical basis for exploring the brain damage of PFS and the effective prevention and control measures.Methods Ninety-six 14-day-old Sprague-Dawley rats were randomly divided into 3 groups:normal control group(NC group,n =32),hyperthermia group(HC group,n =32) and PFS group(n =32).Lipopolysaccharide combined low-dose Kainic acid of intraperitoneal injection were used to induce PFS.Then the histopathologic changes in hippocampus were viewed by HE staining and electronmicroscope,the expression of IL-1β protein in hippocampus of rats was detected with immunohistochemistry methods,the neuron apoptosis was observed by TdT-mediated dUTP nick end labeling,and the relationship of both was researched.Results 1.Thirty-two rats all appeared seizures in PFS group,the average rectal temperature was (39.3 ± 0.4) ℃ ; while the rats in other groups were not convulsions.2.The TUNEL positive cells in hippocampus CA1 of PFS group were more than that of NC group and HC group at 6 h after the PFS(all P <0.01),reached its highest level at 24 h.3.Measured by immunohistochemistry,the level of IL-1β in the hippocampus of PFS group was significantly higher than that of the NC group and HC group at 6 h after the PFS(all P < 0.01).There was a positive correlation between the expression of IL-1 β and the apoptotic index in hippocampus(r =0.789,P < 0.01).Conclusions IL-1 β expression in the hippocampus increases following PFS in rats,and the increased IL-1 β expression was correlated with neurocyte apoptosis.

<p><b>OBJECTIVE</b>To explore the effect of food allergy (FA) on the development of visceralgia sensibility and the substance P (SP) expression in colon of developing rats with FA.</p><p><b>METHOD</b>Three-week old female Sprague-Dawley (SD) rats were randomly divided into two groups (n = 10 in each). The rats in FA group were sensitized with ovalbumin (OVA) 40 µg and Al(OH)3 1 mg suspension solution (0.2 ml) intraperitoneal (i.p.) injection on day 0, only OVA 40 µg solution i.p. on day 2, 4, 7, 9, 11, respectively, and the rats were challenged by gavage with OVA solution 30 mg (2 ml) on day 20, 24, 28, 30. The rats in non-sensitized (NS) group were not challenged except handled in the same ways. The serum OVA-IgE were determined by enzyme-linked immuno sorbent assay (ELISA) on day 0, 30. Jejunum segments were used to observe morphological structure, the expression of eosinophils, and the density and the percentage of degranulation of mast cells (MC). The rats were appraised for the pain sensibility of intestinal tract under colorectal distension irritation by the electrophysiological method on external oblique in the 18-24 hr after the last challenge. Colons were used to analyze the expression of SP through immunohistochemical staining and computer image analyzing system.</p><p><b>RESULT</b>The serum OVA-IgE concentration and the eosinophils, mast cell, the percentage of mast cells degranulation in FA group were more than NS group (P < 0.01). The amplitudes of spike external oblique muscle of abdomen (EOMA, µV) of the FA group under the colorectal distension (CRD) pressures at 0, 15, 30, 45, 60, 75 mm Hg were (17.74 ± 0.72), (18.63 ± 1.72), (22.55 ± 1.70), (28.63 ± 7.00), (33.97 ± 7.34), (37.26 ± 8.40), and (17.43 ± 1.18), (17.27 ± 1.16), (17.73 ± 1.42), (19.55 ± 3.54), (23.29 ± 5.46), (25.20 ± 4.75) in NS group. With the CRD pressure increased, the amplitudes of spike EOMA increased significantly. There were significant differences between groups under the CRD pressures at 30, 45, 60, 75 mm Hg (F = 47.470, 13.367, 13.317, 15.390, P < 0.01). The expressions of colons SP in FA group and NS group are 247.12 ± 90.83 and 103.90 ± 58.94, respectively (t = 4.183, P < 0.01).</p><p><b>CONCLUSION</b>Sensitization through i.p. pathway and challenge by gavage with OVA in early life could result in FA in young SD rats. FA in early life enabled the amplitudes of spike EOMA and the expression of colons SP increase significantly. It may be related to increase in amount and degranulation of MC and SP abnormal expression in colon, which could lead to the development of visceralgia sensibility.</p>

<p><b>OBJECTIVE</b>To investigate the effect of modified Xiaochaihu Decoction (, MXD) on transforming growth factor-β1/Sma- and Mad-related proteins (TGF-β1/Smads) signaling pathway in rats with chronic pancreatitis (CP) induced by dibutyltin dichloride.</p><p><b>METHODS</b>Thirty healthy male Wistar rats were randomly divided into the normal control group, CP group and CP+MXD-treated group. CP was induced by injection of dibutyltin dichloride (DBTC, 7 mg/kg of body weight) into the right caudal vein, and the control rats were treated with vehicle. MXD was given daily by gavage at a dose of 10 g/kg of body weight, starting from the day after CP induction. After 28-day treatment, the n-benzoyl-tyrosyl para-aminobenzoic acid (NBT-PABA) test was carried out to evaluate exocrine pancreatic function. Then, rats were sacrificed, and pancreatic tissues were harvested for histological evaluation. In addition, the mRNA expression of TGF-β1, TGF-β1 type II receptor (TGFβRII), Smad3 and Smad7 was determined in pancreatic tissues by using real-time polymerase chain reaction.</p><p><b>RESULTS</b>Treatment of CP with MXD improved the PABA recovery, decreased the histological lesion, and reduced the mRNA expression of TGF-β1, TGFβRII and Smad3 (P<0.05). However, MXD had no effect on Smad7 mRNA level.</p><p><b>CONCLUSIONS</b>MXD could protect the pancreas against chronic injury and improve pancreatic exocrine function in DBTC induced rat CP model. Its mechanism may involve inhibition of the TGF-β1/Smads signaling pathway.</p>

OBJECTIVE@#To develop a PCR-based X-STR kit for typing of 16 X-STR loci and investigate the polymorphisms of the X-STR markers.@*METHODS@#Sixteen STR loci (GATA 165B12, DXS101, GATA 172D05, HPRTB, DXS981, DXS8378, DXS6795, GATA 31E08, DXS6809, DXS6803, DXS9902, DXS6807, DXS7423, DXS7133, DXS6810 and DXS7132) located on X chromosome were selected. The primers for multiplex PCR were designed by Primer Premier 5.0 software and labeled by four fluorescences (FAM, HEX, TAMRA and ROX). The developed multiplex PCR system was used for investigating the polymorphisms of the X-STR markers in Han populations.@*RESULTS@#The 16-plex amplification system named IDtyper X-16 was successfully developed and validated. Among the 16 X-STR loci, DXS7133 and DXS7423 were found to be moderately polymorphic and the other 14 X-STR markers were highly polymorphic (P1C > 0.5, H > 0.5). The cumulative discrimination power in females and in males were 0.999 999 999 999 97 and 0.999 999 993 respectively in Han population. The combined power of exclusion in trios and in duos were 0.999 999 93 and 0.999990, respectively.@*CONCLUSION@#The IDtyper X-16 kit is highly valuable in forensic science and is suitable for paternity testing in disputed cases.

<p><b>OBJECTIVE</b>To observe the hemodynamic and histological effects of unilateral testicular torsion on the contralateral testis in immature rats, and compare the results of different treatments.</p><p><b>METHODS</b>Testicular torsion models were established in 3-week-old rats and randomized into a normal control, a testicular torsion, a reposition and an orchiectomy group. The systolic peak velocity of the right testicular artery was measured by color Doppler before and 8, 12, 24 and 72 h after the operation. Histological observations of the right testes were performed 2 h after testicular torsion, 12 h after testicular reposition and orchiectomy and when the rats were 9 weeks old.</p><p><b>RESULTS</b>The blood supply of the immature right testes increased continuously after testicular torsion of the left side. Interstitial edema and ultrastructure changes were observed in the testicular torsion, reposition and orchiectomy groups. The right testis weight was significantly greater in both the testicular torsion and orchiectomy groups than in the normal control group of the 9-week-old rats (P < 0.01). No significant differences were noted in the right testicular seminiferous tubule diameter (STD) , count measure spermatogenic (CMSE) and testicular biopsy score (TBS) among the four groups (P > 0.05).</p><p><b>CONCLUSION</b>Unilateral testicular torsion increases blood supply and induces histological changes in the contralateral testis in immature rats. Reposition and orchiectomy following light injury are prognostic of similar results.</p>

<p><b>OBJECTIVE</b>To perform a preliminary study on the dysfunction of spermatogonial stem cells (SSCs) induced by cyclophosphamide.</p><p><b>METHODS</b>According to development stage of spermatogenesis in Wistar rat, 72 rats were divided into three groups, 1, 3 and 9 weeks groups, respectively. Then 24 rats per group were randomly divided into experimental and control groups, and 100 mg/kg cyclophosphamide was injected by intraperitoneal injection in experimental groups, with the same volume of normal saline in the control. Apoptosis of spermatogenic cells were detected after 24 hours by TUNEL. Then 60 one-week rats, whose germ cells were only SSCs, were randomly divided into experimental and control group. We detected c-Kit by immunohistochemistry and cell cycle by flow cytometry at 24 hours, 3 and 9 weeks.</p><p><b>RESULTS</b>There was no significant difference in apoptosis of germ cells in 1 week between experimental group and control group ( P > 0.05); however, there were significant differences among other groups. The ratio of S stage and c-Kit expression of spermatogonial cells were decreased in one-week rats at 24 hours, 3 and 9 weeks in experimental group compared with the control, respectively (P < 0.01).</p><p><b>CONCLUSION</b>Cyclophosphamide does not significantly induce SSCs apoptosis. It may be more important to interfere the proliferation and differentiation of SSCs.</p>

<p><b>OBJECTIVE</b>To search for an effective method for treatment of cerebral infarction.</p><p><b>METHODS</b>Sixty cases of cerebral infarction were randomly divided into an observation group and a control group, 30 cases in each group. All of them were treated with routine needling method, and "Shandong Xiaoying" needling was added to the observation group. The therapeutic effects, recovery of myodynamia and daily living ability were compared in the two groups.</p><p><b>RESULTS</b>There was no significant difference between the two groups in the total effective rate for recovery of upper limb myodynamia (P > 0.05), but there was a very significant difference in the total effective rate for recovery of lower limb myodynamia and increase of daily living ability between the two groups (P < 0.01).</p><p><b>CONCLUSION</b>"Shandong Xiaoying" needling has markedly therapeutic effect on cerebral infarction.</p>

Objective To evaluate the damage degree of diseased kidneys in children with hydrone- phrosis using urinary?2-microglobulin(?2-MG),albumin(Alb),T-H glycoprotein(THP)and immuno- globulin(IgG).Methods The?2-MG,Alb,THP and IgG in pelvis urine in diseased kidneys and in healthy kidneys(as controls)were detected in 78 children with congenital hydronephrosis by radioimmuno- assay;at the same time urinary creatinine(Cr)was detected in 36 cases of them.Pathologic changes of the diseased kidneys were graded;and the correlations of pathologic grades with?2-MG,Alb,THP,IgG and?2-MG/Cr,Alb/Cr,THP/Cr,IgG/Cr were analyzed by Spearman's test.Results In healthy and hydro- nephrotic pelvises,the relevant parameters were as follows.Urine?2-MG was(130.06?22.02)ng/ml vs(386.18?42.15)ng/ml;Alb was(80.52?9.32)ng/ml vs(275.54?69.46)ng/ml,respectively (both P<0.05).Urine?2-MG/Cr was(0.16?0.04)ng/?mol vs(1.57?0.39)ng/?mol;Alb/Cr was (0.11?0.03)ng/?mol vs(1.07?0.56)ng/?mol(both P<0.01).THP was(25.80?9.02)ng/ml vs(31.43?10.56)ng/ml;and IgG was(15.21?3.56)ng/ml vs(20.52?10.85)ng/ml(P>0.05). THP/Cr was(0.05?0.01)ng/?mol vs(0.14?0.06)ng/?mol;and IgG/Cr was(0.04?0.01)ng/?mol vs(0.12?0.05)ng/?mol(P<0.05).There was significant correlation between urine?2-MG,Alb lev- els and pathologic grades(P<0.05),also between?2-MG/Cr,Alb/Cr levels and pathologic grades(P<0.01).There was positive correlation between THP,IgG and pathologic grades but with no significance(P>0.05),also between THP/Cr,IgG/Cr and pathologic grades with significance(P<0.05).Conclu- sions For hydronephrotic kidneys,urine?2-MG/Cr can evaluate the damage degree of nephric tubules, whereas urine Alb/Cr can evaluate that of glomeruli.Increase in THP/Cr and IgG/Cr respectively indicate the impaired nephric tubules and glomeruli in children with congenital hydronephrosis.
