ABSTRACT
ObjectiveTo provide a new idea for exploring the molecular genetic approach to the pathogenesis of schizophrenia via construction of microRNA-messenger RNA (miRNA-mRNA) regulatory network in schizophrenia. MethodsThe microarray datasets of GSE54578 miRNA expression profiles in peripheral blood and GSE145554 mRNA expression in the anterior cingulate in postmortem brain of schizophrenic subjects were downloaded from Gene Expression Omnibus (GEO) database since July 2021. The GEO2R was used to identify the differentially expressed miRNAs and mRNAs, screen the miRNA with target differentially expressed mRNA, and predict their potential upstream transcription factors. The overlapping genes from the mRNA targeted by the differentially expressed miRNA and the mRNA differentially expressed in GSE145554 dataset were collected. Then the biological features of hub genes were analyzed via Gene Ontology (GO) analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis, and the protein-protein interaction (PPI) network and miRNA-mRNA regulatory network of hub genes were constructed. ResultsA total of 8 up-regulated differentially expressed miRNAs with targeted mRNA were screened out in GSE54578 datasets regarding schizophrenia, which involved in the regulation of 10 transcription factors, 247 down-regulated differentially expressed mRNAs were screened out in GSE145554 datasets, and 17 overlapping mRNAs were obtained. GO analysis showed that the target mRNAs were mainly involved in astrocyte differentiation and development. KEGG pathway enrichment analysis showed that the target mRNAs were mainly involved in Rap1 and Ras signaling pathways. PPI network analysis showed that the mRNAs (KRAS and CD28) might be key genes in schizophrenia. ConclusionThe integrated bioinformatics analysis based on GEO database can identify potential susceptibility genes in schizophrenia, and it also contributes to the construction of miRNA-mRNA regulatory network in schizophrenia.

OBJECTIVE@#To explore the clinical features and genetic basis for a patient diagnosed with creatine deficiency syndrome (CDS).@*METHODS@#The patient was subjected to whole exome sequencing (WES). Candidate variant was verified by Sanger sequencing. The level of creatine was determined by using a magnetic resonance spectrum (MRS) method.@*RESULTS@#The patient presented with development delay and poor response to stimuli. No obvious abnormality was found with his muscle tone and strength of his limbs. Borderline EEG was detected. MRI showed abnormal development of the white matter and dysplasia of corpus callosum. Urine organic acid screening has shown increased glycerin-3-phosphate. WES revealed that the patient has carried compound heterozygous variants of the GAMT gene, namely c.412C>T and IVS4-1G>A, which were respectively derived from his mother and father. MRS showed reduced creatine in bilateral basal ganglia. Functional study of the splicing site suggested that the IVS4-1G>A variant has resulted skipping of exon 5 upon splicing.@*CONCLUSION@#The compound variants of the GAMT gene probably underlay the disease in this child. Above finding has enriched the spectrum of GAMT gene variants.

OBJECTIVE: To strengthen the occupational protection of pharmacy intravenous admixture service (PIVAS) staff, and to ensure the safe and timely delivery of drugs. METHODS: The cytotoxic drug packing bag was made by oxford cloth, velvet cotton, pearl cotton, silica gel, activated carbon, rope, etc. Taking Paclitaxel liposomes for injection as an example, the application effect of packing bags was observed. A total of 156 medical orders of PIVAS formulated Paclitaxel liposomes for injection were selected and divided into test group and control group aoccording to random number table. The dispensing drugs were packed with cytotoxic drug packing bag in trial group, and packed with previous plastic packing bag in control group. The packing time of each drug, the delivery time, leaking and shattering, the breakage processing time and loss amount were compared between 2 groups. RESULTS: There was no statistical significance in the delivery time between 2 groups (P>0. 05). The packing time of trial group was significantly longer than control group; there were 16 bottles of leaking in the control group, and 8 bottles of shattering; In trial group, 3 bottles were leaking but not spilled over (infiltrated in the silica gel layer), and there was no shattering bottle; the breakage processing time of trial group was significantly shorter than that of control group; the loss amount was significantly less than that of control group, with statistical significance (P<0. 05). CONCLUSIONS: The cytotoxic drug packaging bag developed in this study can play a spill-proof, anti-seismic role, reduce the loss of valuable drugs, and also strengthen the occupational safety and health protection of medical staff.

Objective Through two nationwide sero-epidemiological survey programs on hepatitis B virus (HBV),hepatitis B markers in Yunnan province were compared between the findings in 2014 and in 2006.Results were used to understand the HBV epidemic status in Yunnan province and to estimate the efficiency of measures on prevention and control of the disease.Methods People at the age of 1-29 years were sampled from 6 counties of the National Disease Surveillance sites in Yunnan province,by multi-stage random sampling method.Demographic information was collected by questionnaire,and 2-4 ml serum sample was taken to detect HBsAg,anti-HBs,anti-HBc,HBeAg and anti-HBe,by ELISA method.Results were logged-in the database and analyzed by SPSS 17.0.Results Between the two studies,no statistically significant differences were observed on the overall HBV infection rates and the positive rates of HBsAg,HBeAg,anti-HBe,anti-HBc.Positive rate of anti-HBs in 2014 was significantly higher than that in 2006.The main two combination profiles of hepatitis B markers were through the fifth model which was positive only for anti-HBs and the sixth model which was negative for all markers in 2006,and the same in 2014.Detection rate through the fifth profile in 2014 was significantly higher than the rate in 2006,while the detection rate of the sixth profile in 2014 was significantly lower than the one in 2006.When analyzing the differences in the same age group,between the two surveys,results showed that the positive rate of anti-HBs in 2014 was significantly higher than that in 2006,in both the10-14 and the 15-19 years-old groups.Positive rate of anti-HBc in the 20-29 year-olds in 2014 was significantly lower than the one of 2006.The positive rates of HBsAg in the age groups of 1-4,5-14 and 15-29 were 1.12%,1.61% and 1.25% respectively,in 2014.Conclusion Prevention and control measures on HBV seemed in great progres.However,more effective prevention and control measures should be taken continually among people under the age of 15,in Yunnan province.

Recent studies have found that microRNAs (miRNAs) can be secreted via exosomes, stay outside the cells, and keep stable in circulating blood. Various studies have shown that the changes in circulating miRNAs are associated with different pathological states, suggesting that the circulating miRNAs can be used as potential molecular targets for the diagnosis and treament of diseases. Research has shown that miRNAs are closely associated with the development and progression of liver fibrosis. This article reviews the studies on the differences in the expression of circulating miRNAs in the course of liver fibrosis and points out that circulating microRNAs can provide a new research direction for early noninvasive diagnosis and treatment of liver fibrosis.

<p><b>OBJECTIVE</b>To study the effect of different concentration of phytohemagglutinin (PHA) on mouse embryo development.</p><p><b>METHOD</b>In experiment 1, crude and purified PHA extracted from Yunnan white kidney bean with different concentration were added into M16 culture medium, the final concentration of PHA were: 50, 100, 200, 500, 1 000, 2 000 and 5 000 mg x L(-1) respectively. 2-cell stage embryos were collected and cultured in PHA containing or control medium for 72-96 h and their development were recorded. In experiment 2, different stage of embryos from 1-cell to blastocyst were treated by different concentrations of PHA same as experiment 1 and 10 000 mg x L(-1) in culture medium for 24 h before washing and cultured in M16 + PVA without PHA to blastocyst or hatching blastocyst stage.</p><p><b>RESULT</b>Low concentrations PHA at 50-100 mg x L(-1) promoted embryo development and increased the number of blastocyst stage embryos. In contrast, high concentrations of PHA (> 1 000 mg x L(-1)) blocked the embryos development from 1-cell to blastocyst stage and showed apoptosis morphology or death.</p><p><b>CONCLUSION</b>Depending on the concentrations, PHA from white kidney bean shown promotion or inhibition on mouse embryo development. 1-cell stage embryo shown more sensitive to PHA treatment than that of later stage embryos. Pretreatment 24 h in PHA containing medium can influence the further development of embryos. Low concentrations of PHA is benefit to embryo development, but high concentrations of PHA (> 1 000 mg x L(-1)) will block of the development of embryos.</p>

OBJECTIVE: To investigate the effects of Shenshuning Recipe (SSNR) on gene expression of hepatocyte growth factor (HGF) in renal tissues in rats with glomerulosclerosis. METHODS: Glomerulosclerosis was induced in 42 rats by unilateral nephrectomy and intravenous injection of doxorubicin. Then these 42 rats were randomly divided into three groups: untreated group, SSNR-treated group and benazepril-treated group. Another eight rats were included into sham-operation group. The rats in the SSNR-treated group and the benazepril-treated group were fed SSNR or benazepril respectively for 8 weeks. The levels of 24 h urine protein (Upr), serum creatinine (Cr) and blood urea nitrogen (BUN) of rats in each group were examined. The renal morphological changes were observed under microscope, and the diameter of glomerular capillary, mesangial matrix and glomerulosclerosis index were analyzed by image analysis software. Reverse transcription-polymerase chain reaction (RT-PCR) method was used to detect the gene expression of HGF in the renal tissues. RESULTS: The levels of 24 h Upr, serum Cr and BUN in the untreated group were remarkably increased than those in the sham-operation group (P<0.01). The pathological morphological changes in the untreated group showed that the glomerulosclerosis was diffused around the renal tissue and the capillaries were shrunk. The expression level of mesangial matrix was up-regulated and the glomerulosclerosis index was 3.32+/-0.35. The expression level of HGF mRNA in the untreated group was obviously lower than that in the sham-operation group (P<0.05). The levels of 24 h Upr, serum Cr and BUN in the SSNR-treated group and the benazepril-treated group were remarkably decreased as compared with those in the untreated group, while the expression levels of HGF mRNA were both obviously higher than that in the untreated group (P<0.01). The pathological morphological changes in the SSNR-treated group and the benazepril-treated group were both alleviated. There was no significant difference in therapeutic effect between the SSNR-treated group and the benazepril-treated group. CONCLUSION: Shenshuning Recipe can up-regulate the expression of HGF mRNA, decrease the mesangial matrix, and improve the renal function, so that it may retard the development of glomerulosclerosis.

[Objective] To establish a method for the determination of paeoniflorin content in Kun'an Pills. [Methods] High pressure liquid chromatography (HPLC) was used. The chromatographic conditions were: Hypersil ODS column, acetonitrile-water (17 : 83) as mobile phase, detection wavelength at 230 nm and the flow rate being 1.0 mL/min. [Results] A good linearity of paeoniflorin was shown in the range of 0.096 8 - 0.484 0 ?g, r = 0.999 8; the average recovery was 97.84% and the relative standard deviation being 1.44%. [Conclusion] This method is simple and effective and can be used for the quality control for Kun'an Pills.
