ABSTRACT

ObjectiveTo compare the four preparation methods of Rehmanniae Radix juice described in ancient literature and find the method that is most suitable for the preparation of Rehmanniae Radix juice used in Baihe Dihuangtang. MethodThe ancient medical books record four methods for preparing Rehmanniae Radix juice: crushing fresh Rehmanniae Radix for juice, steaming fresh Rehmanniae Radix for juice, boiling fresh Rehmanniae Radix for juice, and boiling dry Rehmanniae Radix for juice. Ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) was employed to detect the compounds in the four juice samples, followed by principal component analysis (PCA). Result① Totally 27 compounds were identified in the juice samples, including 10 iridoid glycosides, 14 phenylethanoid glycosides, 2 phenolic acids, and 1 irisone. Among them, 15 common compounds were shared by the four juice samples, including 7 iridoid glycosides, 7 phenylethanoid glycosides, and 1 phenolic acid. ② Five common compounds in the four juice samples can be matched with the reference standards, which were catalpol, aucubin, rehmannioside D, ajugol, and purpureaside C. ③ Verbascoside and isoacteoside were not detected in the juice prepared by crushing fresh Rehmanniae Radix, while it was detected in the other three juice samples, which indicated that the two components were produced after heating rather than being the original components in fresh Rehmanniae Radix. ④ The comparison of the ion fragments demonstrated that verbascoside was produced from purpureaside C after the cleavage of the glycosidic bond and removal of a molecule of mannose. ⑤ Isoacteoside could be isomerized from verbascoside, and its relative content increased with the extension of heating time. However, the relative content of verbascoside and purpureaside C did not decrease significantly. Therefore, it was hypothesized that purpureaside C was produced from its upstream component. ConclusionThe juice prepared by crushing fresh Rehmanniae Radix has the chemical composition significantly different from the juice samples prepared with the other 3 methods, while the latter 3 juice samples had similar chemical composition. Although all the four methods can be used, it is more suitable to prepare Rehmanniae Radix juice by steaming fresh Rehmanniae Radix, boiling fresh Rehmanniae Radix, and boiling dry Rehmanniae Radix.

ABSTRACT

A boy, aged 2 years and 5 months, had recurrent epistaxis, and the coagulation function examination showed that activated partial thromboplastin time (APTT) was significantly prolonged. Further laboratory examinations showed that the prolonged APTT was not immediately corrected in the APTT correction test, with positive lupus anticoagulant and low prothrombin activity. The boy was diagnosed with hypoprothrombinemia-lupus anticoagulant syndrome. The condition was improved after treatment with glucocorticoid, immunoglobulin, and vitamin K1. The boy has been followed up for 6 months, and no epistaxis was observed. Prothrombin activity returned to normal, and lupus anticoagulant remained positive. This is a relatively rare disease, and for patients with bleeding symptoms and coagulation disorders, it is recommended to perform the tests such as APTT correction test, lupus anticoagulant testing, and coagulation factor dilution test, which can improve the detection rate of this disease, so as to achieve early diagnosis, provide rational treatment in the early stage, and improve the prognosis.

ABSTRACT

Objective@#This study aimed to investigate the effects of @*Methods@#In this study, 0.1% DMG was supplemented in 20% casein diets that were either folate-sufficient (20C) or folate-deficient (20CFD). Blood and liver of rats were subjected to assays of Hcy and its metabolites. Hcy and its related metabolite concentrations were determined using a liquid chromatographic system.@*Results@#Folate deprivation significantly increased pHcy concentration in rats fed 20C diet (from 14.19 ± 0.39 μmol/L to 28.49 ± 0.50 μmol/L; @*Conclusion@#DMG supplementation exhibited hypohomocysteinemic effects under folate-sufficient conditions. By contrast, the combination of folate deficiency and DMG supplementation has deleterious effect on pHcy concentration.

ABSTRACT

This study aims to explore the relationship of DNA methylation with the contents of the index components as well as the growth and development of Pogostemon cablin. The demethylation reagent 5-azacytidine(5-azaC) was used to treat the tissue culture seedlings of patchouliol-type P. cablin. High performance liquid chromatography was employed to evaluate the changes of DNA methy-lation in P. cablin, and GC-MS to detect the contents of index components in P.cablin. The agronomic characters of P.cablin were measured using the common methods. The results showcased that DNA methylation of P.cablin was significantly reduced by 5-azaC in a concentration-dependent manner. Thirty days after treatment with 5-azaC at different concentrations, the content of patchouli alcohol changed slightly; compared with that in the control group, the content of pogostone in 50 μmol·L~(-1) and 100 μmol·L~(-1) 5-azaC groups was significantly up-regulated. The 100 μmol·L~(-1) 5-azaC group had the largest differences in contents of pogostone and patchouli alcohol compared with the control group, followed by the 50 μmol·L~(-1) 5-azaC group. Ninety days after disinhibition, the content of pogostone in the treatment group was significantly increased and the content of patchouli alcohol was significantly decreased. In addition, 5-azaC significantly inhibited the growth and development of P.cablin in a dose-dependent manner. These results indicate that DNA methylation regulates the biosynthesis of the index components in patchouliol-type P.cablin and proper demethylation can directly promote the synthesis of pogostone and indirectly affect the accumulation of patchouli alcohol.

ABSTRACT

OBJECTIVE:To prelimi narily investigate the possible mechanism of orazamide to prevent anti-tuberculosis drug-induced liver injury (ATB-DILI). METHODS :A total of 60 Kunming mice were randomly divided into blank group ,model group,positive control group [diammonium glycyrrhizinate 60 mg/(kg·d)],orazamide low-dose ,medium-dose and high-dose groups [ 80,160,320 mg/(kg·d)],with 10 mice in each group. Except for blank group ,other groups were given isoniazid [ 75 mg/(kg·d)]+rifampicin [ 75 mg/(kg·d)] for 14 days intragastrically to induce ATB-DILI model. At the same time ,administration groups were given relevant medicine intragastrically ,blank group and model group were given normal saline intragastrically. The administration volume was 20 mL/(kg·d),once a day ,for consecutive 14 days. The general conditions of the mice were observed and recorded every day ,such as growth and development ,mental and diet state. After last medication ,liver index was calculated , and HE staining was adopted to observe pathological changes of liver tissue of mice. The positive expression of high mobility group protein B 1 (HMGB1) and NF-κ B in liver tissue were detected by streptavidin biotin-peroxidase complex (SABC) immuno- histochemistry. The serum levels of liver function indexes in serum ,the protein expression of advanced glycation end product receptor(RAGE)and TNF-α in liver tissue were detected by ELISA. RESULTS:Compared with blank group ,the growth and development of mice in the model group were slow ,and their appetite and spirit were poor. The liver index ,serum levels of TBIL , DBIL,ALT,AST,ALP,TBA and γ-GT were increased significantly (P<0.05). Structural disorder of liver lobules ,degeneration and necrosis of liver cells and inflammatory cell infiltration were observed. The expression of HMGB 1,NF-κB,RAGE and TNF-α in liver tissue were elevated significantly (P<0.05). Compared with model group ,the general condition of mice were all improved to different extents in orazamide low-dose ,medium-dose and high-dose groups ,positive control group ,while liver index and above serum indexes were all decreased significantly (P<0.05). The pathological changes of liver tissue were all improved to different extents ,while the protein expression of HMGB 1,NF-κB,RAGE and TNF-α were all decreased significantly(P<0.05). The improvement of above indexes in orazamide high-dose group were all significantly better than orazamide low-dose and medium-dose groups (P<0.05);the levels of ALP and TBA in orazamide high-dose group were significantly lower than positive control group (P<0.05). CONCLUSIONS :Orazamide can prevent ATB-DILI induced by isoniazid combined with rifampicin in mice,the mechanism of which may be associated with down-regulating the protein expression of HMGB 1 and RAGE in liver tissue and inhibiting the secretion of inflammatory factors.

OBJECTIVE@#To study the efficacy and safety of intensity-modulated radiotherapy (IMRT) in children with high-risk neuroblastoma (NB).@*METHODS@#A retrospective analysis was performed on the medical data of 24 children with high-risk NB who were diagnosed and treated with IMRT in the Department of Hematology and Oncology, Hunan Provincial People's Hospital, from April 2018 to December 2020. The medical data included age, radiotherapy dose, times of radiotherapy, laboratory examination results, adverse reactions, and survival.@*RESULTS@#All 24 children (14 boys and 10 girls) received IMRT, with a mean age of (65±23) months and a median age of 59 months. The primary tumor was located in the abdomen in 23 children and 1 child had primary tumor in the mediastinum. The median age was 41.5 months at the time of radiotherapy. The radiation dose of radiotherapy ranged from 14.4 to 36.0 Gy, with a mean dose of (22±3) Gy and a daily dose of 1.8-2.0 Gy. The radiotherapy was performed for a total number of 8-20 times, with a mean number of 11.9 times. Among these children, 6 received radiotherapy for the residual or metastatic lesion. Of all the 23 children, 3 experienced cough, 2 experienced diarrhea, and 1 experienced vomiting during radiotherapy. At 2 weeks after radiotherapy, serum creatinine ranged from 2.3 to 70.1 μmol/L and alanine aminotransferase ranged from 9.1 to 65.3 μ/L. Ten children experienced grade Ⅲ bone marrow suppression and 2 experienced grade Ⅳ bone marrow suppression 1 to 2 weeks after radiotherapy. Four children experienced grade Ⅲ bone marrow suppression and 1 experienced grade Ⅳ bone marrow suppression 3 to 4 weeks after radiotherapy. During a median follow-up time of 13.5 months, 23 children (96%) achieved stable disease and 1 died. Up to the follow-up date, second malignant tumor or abnormal organ function was not observed.@*CONCLUSIONS@#IMRT can improve the local control rate of NB. IMRT appears to be safe in the treatment of children with NB.

OBJECTIVE@#To study the long-term clinical effect of multicenter multidisciplinary treatment (MDT) in children with renal malignant tumors.@*METHODS@#A retrospective analysis was performed on the medical data of 55 children with renal malignant tumors who were diagnosed and treated with MDT in 3 hospitals in Hunan Province from January 2015 to January 2020, with GD-WT-2010 and CCCG-WT-2016 for treatment regimens. A Kaplan-Meier survival analysis was used to analyze the survival of the children.@*RESULTS@#Of the 55 children, 10 had stage I tumor, 14 had stage Ⅱ tumor, 22 had stage Ⅲ tumor, 7 had stage IV tumor, and 2 had stage V tumor. As for pathological type, 47 had FH type and 8 had UFH type. All children underwent complete tumor resection. Of the 55 children, 14 (25%) received preoperative chemotherapy. All children, except 1 child with renal cell carcinoma, received postoperative chemotherapy. Among the 31 children with indication for radiotherapy, 21 (68%) received postoperative radiotherapy. One child died of postoperative metastasis. The incidence rate of FH-type myelosuppression was 94.4%, and the incidence rate of UFH-type myelosuppression was 100%. The median follow-up time was 21 months and the median survival time was 26 months for all children, with an overall survival rate of 98% and an event-free survival rate of 95%.@*CONCLUSIONS@#Multicenter MDT has the advantages of high success rate of operation and good therapeutic effect of chemotherapy in the treatment of children with renal malignant tumors, with myelosuppression as the most common side effects, and radiotherapy is safe and effective with few adverse events. Therefore, MDT has good feasibility, safety, and economy.

ABSTRACT

OBJECTIVE Programmed death ligand-1 (PD-L1) and indoleamine 2, 3-dioxygenase 1 (IDO1) are immune checkpoints which can be induced by interferon-γ(IFN-γ) in the tumor microenvironment, leading to immune escape of tumors. Myricetin (MY) is a flavonoid distributed in many edible and medicinal plants. The aim of this study is to clarify the effect and the mechanism of MY on inhibiting IFN-γ-induced PD-L1 and IDO1 in lung cancer cells. METHODS Expressions of PD-L1 and major histocompatibility complex-I (MHC-I) were evaluated by flow cytometry and Western blotting, and the expression of IDO1 was measured by Western blotting. qRT-PCR was used to detect their mRNA levels. The function of T cells was evaluated using a co-culture system consist of lung cancer cells and the Jurkat-PD-1 T cell line that overexpressing PD-1. Molecular docking analysis, Western blotting and immunofluorescence were used for mechanism study. RESULTS MY potently inhibited IFN-γ-induced PD-L1 and IDO1 expression in human lung cancer cells, while didn't show obvious effect on the expression of MHC-I. In addition, MY restored the survival, proliferation, CD69 expression and interleukin-2 (IL-2) secretion of Jurkat-PD-1 T cells suppressed by IFN-γ-treated lung cancer cells in the co-culture system. Mechanistically, IFN-γ up-regulated PD-L1 and IDO1 at the transcriptional level through the JAK-STAT-IRF1 axis, which was targeted and inhibited by MY. CONCLUSION Our research revealed a new insight into the anti-tumor effects of MY which inhibited IFN-γ-induced PD-L1 and IDO1 expression, supporting the potential of MY in anti-tumor immunotherapy.

ABSTRACT

Objective:To investigate the concentration level of chloroform in the water of swimming pool in Baoan District of Shenzhen City,and determine the risk factors. Methods:During May and July,2019,a total of 110 water samples from 40 swimming pools were collected in Xin’an subdistrict of Bao’an District for the examination of chloroform routine indicators. In addition, 38 pipe water samples were collected for the examination of chloroform and free residual chlorine. Results:The concentration of chloroform in the swimming pools was determined to be (43.400±27.802) μg/L with the median of 37.343 μg/L. Chloroform was correlated positively with total bacterial count(P<0.05),turbidity, free chlorine residual, and PH value(P<0.01). Conclusion:The disinfection quality of swimming pool water in Bao’an District remains low. It is necessary to determine the risk factors associated with chloroform in the swimming pool and further reduce the concentration level of disinfection by-products.

ABSTRACT

Atherosclerosis is one of the most common diseases that threaten human health. How to effectively inhibit atherosclerosis, extend the survival time and improve the quality of life has become one of the most urgent issues to be solved clinically. Mongolian medicine, with a long history of managing human diseases, is an important part in traditional Chinese medicine (TCM) and has distinct ethnic characteristics. It has been gradually formed and developed by absorbing some theories of Tibetan medicine, Indian medicine and relevant knowledge of TCM. Mongolian medicine has many advantages, including but not limited to, low toxicity and diverse structure. However, the action mechanism of Mongolian medicine in preventing and managing atherosclerosis has yet to be fully clarified, which has been a major obstacle for further promotion and application of Mongolian medicine in clinical settings. In this review, the up-to-date research findings on Mongolian medicine were collected, analyzed and summarized, and the anti-atherogenic action mechanism of Mongolian medicine were reviewed from the aspects of anti-inflammatory, lipid-lowering, anti-oxidative stress, vascular endothelial cell protection, and inhibition of vascular smooth muscle cell proliferation and migration.

ABSTRACT

Objective:To investigate the effect of Banxia Xiexintang on the epithelial-mesenchymal transition (EMT) of human peritoneal mesothelial cell line (HMrSV5) induced by gastric cancer-derived exosomes (Exo). Method:Banxia Xiexintang-containing serum was prepared and the human gastric cancer NCI-N87-derived exosomes (NCI-N87-Exo) were extracted, followed by their identification by transmission electron microscopy and Western blotting and labeling with 1,1-dioctadecyl-3,3,3,3- tetramethylindocarbocyanine perchlorate (Dil). The cells were divided into the blank group, model group, and low-, medium-, and high-dose (13.5,27,54 g·kg<sup>-1</sup>) Banxia Xiexintang groups. HMrSV5 cells in the blank group were cultured alone, the ones in the model group with 100 mg·L<sup>-1</sup> NCI-N87-Exo, and those in the low-, medium-, and high-dose Banxia Xiexintang groups with 100 mg·L<sup>-1</sup> NCI-N87-Exo plus low-, medium-, and high-dose 10% Banxia Xiexintang-containing serum, respectively. Confocal laser microscope was used to observe the uptake of NCI-N87-Exo by HMrSV5 cells at 24 h, 48 h and 72 h. Seventy-two hours later, the morphological changes in HMrSV5 cells were observed. The protein expression levels of E-cadherin, cytokeratin 19 (CK19), <italic>α</italic>-smooth muscle actin (<italic>α</italic>-SMA), elastin, and transforming growth factor-<italic>β</italic><sub>1</sub> (TGF-<italic>β</italic><sub>1</sub>), Smad2/3, and p-Smad2/3 were assayed by Western blot. Result:It was observed under the transmission electron microscope that NCI-N87-Exo showed an oval or dish-shaped vesicle structure with a particle size ranging from 40 to 80 nm. Exo marker proteins CD9 and CD63 were highly expressed while calreticulin was not expressed, implying that the NCI-N87-Exo was confirmed. After 24 h, 48 h, 72 h of co-culture, it was observed under the fluorescence microscope that NCI-N87-Exo were taken up by HMrSV5 cells, which was positively correlated with time. Compared with the blank group, Banxia Xiexintang significantly inhibited the uptake of NCI-N87-Exo by HMrSV5 cells, with better effect noticed in the middle- and high-dose Banxia Xiexintang groups(<italic>P</italic><0.05,<italic>P</italic><0.01). After intervention with Banxia Xiexintang-containing serum, the HMrSV5 cells were arranged densely, and the intercellular space was significantly reduced, with the most obvious changes present in the high-dose Banxia Xiexintang group. Western blot revealed that the protein expression levels of E-cadherin and CK19 in HMrSV5 cells after being intervened with the medium- and high-dose Banxia Xiexintang-containing serum were increased significantly as compared with those in the blank group, whereas the levels of <italic>α</italic>-SMA and Elastin were decreased significantly (<italic>P</italic><0.01). Banxia Xiexintang-containing serum at the low, medium, and high doses remarkably down-regulated TGF-<italic>β</italic><sub>1</sub> and p-Smad2/3 protein expression(<italic>P</italic><0.05,<italic>P</italic><0.01). However, there was no significant change in Smad2/3. Conclusion:NCI-N87-Exo can be taken up by HMrSV5 cells to induce EMT. Banxia Xiexintang can inhibit the uptake of NCI-N87-Exo by HMrSV5 cells and the resulting EMT induced by NCI-N87-Exo, which is related to the regulation of TGF-<italic>β</italic><sub>1</sub>/Smads signaling pathway.

Objective:This paper aims to study the genetic diversity of <italic>Pogostemon cablin</italic> by amplified fragment length polymorphism (AFLP) markers. Method:The 12 pairs of primers were used for AFLP analysis of 212 samples from 14 varieties,and biological analysis software such as POPGENE 32,Arlequinver 3.5,MEGA 7 and NTSYSpc 2.10e were used for polymorphism parameter calculation,principal coordinate analysis and cluster analysis. Result:A total of 2 238 loci were amplified by 12 pairs of primers. 2 226 of them were polymorphic loci, accounting for 99.38%. At the inter-population level,the values of effective alleles(<italic>Ne</italic>),Nei's gene diversity index(<italic>H</italic>),Shannon polymorphic information index(<italic>I</italic>) were 1.365 6±0.066 3, 0.220 7±0.036 4, and 0.343 7±0.050 2,respectively;and 1.118 5±0.038 7,0.071 3±0.023 0,0.109 4±0.035 0,respectively at the intra-population level. Analysis of molecular variance(AMOVA)showed that 71.57% of the total variation of <italic>P. cablin</italic> was of inter-population nature, and 28.43% was of intra-population nature. The 14 populations could be divided into four groups by cluster analysis. Conclusion:The results of AFLP molecular markers showed that abundant genetic diversity was present at inter-population level of <italic>P. cablin</italic>,however,relatively low at intra-population level; the genetic differentiation at the inter-population level was significant,which could provide a reference for the subsequent study of good germplasm selection of <italic>P. cablin</italic>.

ABSTRACT

The terpenoids in Pogostemon cablin have complex structures and abundant pharmacological effects. Patchouli alcohol(PA) and pogostone(PO) have a high medicinal value by virtue of anti-tumor, anti-inflammatory, antibacterial, antioxidant, and other biological activities. Due to the low content of terpenoid metabolites in P. cablin, the study of biosynthesis and metabolism regulation can provide a biosynthetic basis for obtaining high-content terpenoids. In this study, key enzyme genes in biosynthesis, transcription factors in metabolism regulation, spatio-temporal expression of terpene synthase were reviewed, aiming to provide a reference for the development, protection, and utilization of P. cablin resources.

ABSTRACT

OBJECTIVE@#To study the clinical features of neuroblastoma (NB) and the factors influencing survival rate.@*METHODS@#A total of 44 children with NB who were admitted from April 2016 to February 2020 were enrolled as research subjects. A retrospective analysis was performed on their medical data and follow-up data.@*RESULTS@#The common clinical symptoms of these 44 children were fever (10/44, 23%), mass (9/44, 20%), abdominal pain (8/44, 18%), cough (7/44, 16%), pale complexion (3/44, 7%), claudication (2/44, 5%), and abnormal activity (2/44, 5%). According to the INSS stage, 2 children (4%) had stage I NB, 5 children (11%) had stage II NB, 5 children (11%) had stage III NB, and 32 children (73%) had stage IV NB. The mean follow-up time was (15.3±1.5) months, with a recurrence rate of 20% and an overall survival rate of 82%. Among the 44 children, 29 (66%) achieved event-free survival and 7 (16%) had survival with tumor. The univariate analysis showed that a pathological type of NB and an increase in serum neuron-specific enolase (NSE) decreased the overall survival rate of children with NB (P<0.05).@*CONCLUSIONS@#The clinical symptoms of children with NB are not specific at the first visit. Fever, abdominal pain, and mass are common symptoms, and there is a high proportion of children in the advanced stage. The pathological type of NB and an increase in serum NSE may be associated with a reduction in the overall survival rate of children with NB.

ABSTRACT

Objective: To identify the anti-depressive effect of ferulic acid (FA) in mice exposed to lipopolysaccharide (LPS) and explore its molecular mechanisms. Methods: The mice were divided into 5 groups as follows: Control, LPS, LPS + SP, LPS + FA, and LPS + FA + anisomycin. The LPS + FA and LPS + FA + anisomycin groups were administered with FA (100 mg/kg, i.p.) once daily continuously for 7 days, and the other groups received an equivalent volume of saline. On the 7th day, LPS (0.1 mg/mL, i.p.) was injected in all mice except the control group 30 min after FA or saline administration. The LPS + SP and LPS + FA + anisomycin groups were intravenously administered with SP600125 [c-Jun N-terminal kinase (JNK) inhibitor] (100 μL/site, i.v.) and anisomycin (JNK activator) (100 μL/site, i.v.) 15 min before LPS, respectively. The depressive behaviors were assessed by open field test, sucrose preference test, and forced swimming test at 24 h post-LPS administration. Tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) levels in plasma were measured by ELISA. The levels of phospho-JNK, TNF-α, IL-1β, Bcl-2, Bax, cytochrome c and caspase-3 were evaluated by Western blotting. Results: FA alleviated depression symptoms caused by LPS in mice, including increasing sucrose water consumption in sucrose preference test and reducing the immobility time in forced swimming test. FA could inhibit upregulated levels of phospho-JNK, TNF-α, and IL-1β. FA also markedly decreased Bax, caspase-3, and cytochrome c, and increased Bcl-2 levels. Besides, SP600125 showed neuroprotective effect similar to FA which was attenuated by anisomycin. Conclusions: FA attenuates inflammation and apoptosis by inhibiting LPS-induced activation of JNK to alleviate depression-like behaviors.

ABSTRACT

Objective::To investigate the effects of modified Buwangsan on the learning and memory ability of Alzheimer's disease (AD) model rats and the expression of NOD-like receptor 3 (NLRP3), cysteine-containing aspartate-specific proteases 1 (Caspase-1) and interleukin-1 beta (IL-1β) in NLRP3 inflammatory pathway in hippocampus of AD model rats, and exploring the underlying mechanism of modified Buwangsan. Method::The 52 eligible rats were randomly divided into sham control group, AD model group, low-dose modified Buwangsan group (1.5 g·kg-1) and high-dose modified Buwangsan group (3 g·kg-1). AD mouse model was established by bilateral hippocampus injection of Aβ1-425 μL (2 g·L-1). The rats in low-dose and high-dose modified Buwangsan group received low and high dose modified Buwangsan respectively within the next 4 weeks, once daily. The learning and memory ability was tested by Morris water maze. The expression of NLRP3, Caspase-1 and IL-1β mRNA was tested by quantitative PCR(Real-time PCR) and Western blot. Result::As compared with the sham group, the learning and memory ability of the rats were significantly impaired (P<0.05). Compared with AD model group, the learning and memory ability and the expression levels of NLRP3, Caspase-1, and IL-1β mRNA and protein were all no statistical differences in low-dose modified Buwangsan group, while the learning and memory ability of the rats were significantly improved and the expression of NLRP3, Caspase-1 and IL-1β mRNA in hippocampus of rats was significantly decreased in high-dose modified Buwangsan group (P<0.05). Conclusion::High-dose modified Buwangsan could attenuate neuroinflammation in the hippocampus of AD mouse model via inhibiting the expression of NLRP3, Caspase-1 and IL-1β, which may be the mechanisms of modified Buwangsan could be used to ameliorate the learning and memory ability of AD mouse model.

ABSTRACT

Objective:To observe the effect of Dabuyuan Jian on the synaptic plasticity of hippocampus and the brain derived neurotrophic factor (BDNF)/tyrosine kinase receptor (TrkB)/cyclic adenosine phosphate reactive element binding protein (CREB) signaling pathway in amyloid precursor protein/presenilil (APP/PS1) mice, and to explore its possible mechanism for improving synaptic plasticity. Method:Totally 36 APP/PS1 mice were divided into model group, donepezil group(6.5×10-4 g·kg-1·d-1) and Dabuyuan Jian group (13.2 g·kg-1·d-1), and another wild mice were set as control group. The mice in control group and model group received an equal volume of saline, and the mice in each group received drugs by gavage for 30 days. The learning ability and memory of mice in each group were detected by Morris water maze. The pathological changes of neurons and synapses in the hippocampus of each group were observed by Nissl staining and Golgi staining. The expression levels of postsynaptic density protein 95 (PSD95) and synaptophysin (SYN) in hippocampus of each group were detected by immunofluorescence (IF). The protein expression levels of BDNF, TrkB, CREB and phosphorylated CREB (p-CREB) in hippocampus were detected by Western blot. Result:As compared with the control group, the platform latency and total swimming distance of the model group were increased in the model group (P<0.01), with decreased times of crossing the platform and staying time in the target quadrant (P<0.01), the intracellular Nissl bodies of neurons in hippocampal CA3 area decreased or disappeared in model group, with decreased number of neurons and dendritic branches and decreased density of dendritic spine in hippocampal CA3 area of the mice (P<0.01), and the protein expression levels of SYN, PSD95, BDNF, TrkB and p-CREB in hippocampus of mice were also decreased in model group (P<0.05, P<0.01). As compared with the model group, the platform latency and total swimming distance were decreased in the donepezil group and Dabuyuan Jian group (P<0.05, P<0.01), with increased times of crossing platform and staying time in target quadrant (P<0.05, P<0.01), the number of Nissl bodies of neurons in hippocampal CA3 area was increased in the donepezil group and Dabuyuan Jian group, with increased number of neurons and dendritic branches and increased density of dendritic spine in hippocampal CA3 area of the mice, and the protein expression levels of SYN, PSD95, BDNF, TrkB and p-CREB in hippocampus of mice were increased in the donepezil group and Dabuyuan Jian group (P<0.05, P<0.01). Conclusion:Dabuyuan Jian can improve the synaptic plasticity of APP/PS1 double transgenic mice, and its mechanism may be related to its up-regulation of BDNF/TrkB/CREB signal pathway in mouse hippocampus.
