ABSTRACT

OBJECTIVE@#To evaluate the clinical efficacy of acupuncture at different timings in acute stage for limb dysfunction in patients with cerebral infarction.@*METHODS@#A total of 101 patients with cerebral infarction limb dysfunction were divided into an early exposure group (@*RESULTS@#Compared before treatment, the mRS grade at 30 and 60 days after onset in the early exposure group was improved (@*CONCLUSION@#The timing of acupuncture is an independent factor affecting the disability status and limb motor dysfunction in patients with cerebral infarction, and the effect of early intervention may be better than late intervention.

ABSTRACT

Objective To analyze the intake of arsenic in Chinese materia medica by Chinese population; To assess its health risk. Methods Totally 2056 Chinese materia medica samples were collected from representative manufacturers and markets in China from 2008 to 2016 and their arsenic contents were detected. The consumption data of Chinese materia medica were collected among population aged 18 and above from 5739 individuals by face to face questionnaire surveys in 5 provinces (Heilongjiang, Liaoning, Jiangxi, Guizhou and Gansu) with the method of multistage stratified random sampling. A deterministic estimate was used to assess the exposure of arsenic by Chinese materia medica. At the same time, the Chinese standard limit of arsenic in Chinese materia medica was used to do the theory assessment. Results The average concentration of arsenic in Chinese materia medica was 0.81 mg/kg. The residue level of arsenic in animal Chinese materia medica (average concentration: 3.29 mg/kg) was higher than that in medicinal plants (average concentration: 0.71 mg/kg). The consumption data showed that average intake of Chinese materia medica as medicines was 207 g, and P95was 540 g; the average intake of Chinese herbals as medicinal food was 43 g, and P95was 500 g. The assessment using arsenic content data showed that common use of Chinese materia medica had low level of risk. However, the population with chronic and high consumption of Chinese materia medica as medicine or as food had certain level of health risk. Arsenic content in animal herbals was about 5 times higher than that of medicinal plants. But according to the consumption, the risk was not higher than that of medicinal plants. The assessment using the standard limit of arsenic (2 mg/kg) in herbals showed that recent standard could protect most people, but risk remained in those with chronic consumption of Chinese materia medica as medicineand high consumption of Chinese materia medica as medicinal food. Conclusion The current standard limit of arsenic (2 mg/kg) in Chinese materia medica is appropriate. The health risk of arsenic exposure from Chinese materia medica among Chinese population is at a low level, but those with chronic and high Chinese materia medica consumption need more attention.

ABSTRACT

Objective To simplify and optimize the method for isolation and culture of primary mouse hepatocytes on the basis of conventional extraction method,and to provide a reference for related research. Methods Mouse liver was reversely perfused with the isolation solution(i.e.,through the vena cava inferior in,and portal vein out),cut into small pieces and digested with enzymes. Then the hepatocytes were isolated by density gradient centrifugation and transferred into culture medium. The cell viability was detected by trypan blue staining. The purity of the hepatocytes was analyzed by flow cytometry and the cell morphology was observed with an inverted microscope. Results The hepatocytes obtained by this improved method showed high viability and purity,with typical characteristics of cell morphology. Conclusions The liver perfusion is facilitated by reversed perfusion, and the isolated hepatocytes are with high viability and purity confirmed by many times of experiments. This optimized procedure is an easy and efficient method for isolation of primary mouse hepatocytes.

ABSTRACT

Purpose To study the role of lnc-HOTAIR and its target-controlled miR-21 on invasion and metastasis of hepatocellular carcinoma cells (HCC) and its related mechanisms.Methods The expression of miR-21 in 30 cases of HCC was detected by quantitative real-time PCR (qRT-PCR),and correlation with clinical pathology was also analyzed.siRNA of lncHOTAIR was transfected to HCC HepG2 cell (HepG2),invasive activities of HepG2 was detected by Transwell,the expression of miR-21 was determined by qRT-PCR,the expression of protein MMP-2 was determined by Western blotting,and the correlation with miR-21 expression was analyzed.Results Compared with pericarcinous tissue,the expressions of miR-21 in tumor tissue increased apparently (P < 0.05).The expressions of miR-21 correlated positively with malignancy degree,differentiation degree and invasion and metastasis respectively,and correlated negatively with tumour size,tumour number and level of AFP respectively.After silencing the HOTAIR gene,the invasive activities of HepG2 were suppressed,the expression of miR-21 gene and MMP-2 protein were down regulated,and both of them were positively correlated.Conclusion High expressions of miR-21 in liver cancer correlate positively with malignancy degree,differentiation degree,intrahepatic and extrahepatic metastases and portal vein tumor thrombus respectively.HOTAIR may inhibit the invasion and metastasis of HCC by controlling the expression level of miR-21.

ABSTRACT

Objective To investigate the level of serum superoxide dismutase(SOD),high-sensitive C-reactive protein (hsCRP)and myocardial injury markers in acute coronary syndrome(ACS)and evaluate their relationship and diagnostic values.Methods Case control study was conducted.This study enrolled 64 ACS patients and 50 non-ACS patients from the 181st Hospital of Chinese PLA in 2015.Serum SOD,hsCRP were tested and myocardial injury markers such as cardiac tro-ponin I(cTnI),creatin kinase MB(CK-MB)were also tested.Student t test and Pearson test were used as statistical meth-ods.Results Compared with control group,SOD of ACS group were significant lower(t=4.136,P<0.001)and hsCRP, Mb,cTnI,CK and CK-MB were significant higher(t=-5.396,-3.495,-5.578,-4.655 and -4.614,all P<0.001). The area under ROC curve of SOD,hsCRP,MYO,cTnI,CK and CK-MB was 0.713,0.758,0.699,0.879,0.841 and 0.802 respectively.After pearman test,the serum SOD were correlated to hsCRP,total cholesterol(TC)and low density lipopro-tein(LDL-C)(r=-0.493,0.548 and 0.404,all P<0.01).Serum hsCRP was correlated to cTnI,triglyceride(TG)and SOD(r=0.671,-0.417 and -0.493,all P<0.01).Conclusion cTnI was positively correlated to hsCRP and markers of myocardial injury.hsCRP was negatively correlated with SOD.It implicated that oxidative stress,inflammatory response and serum lipid deposition may act an important role in the occurrence and development of ACS.

Objective@#To analyze liquid milk exposure of thiocyanate among Chinese population and preliminarily assess its health risk.@*Methods@#A total of 2 059 raw milk samples were collected during 2013 and 2014 from 12 Chinese provinces, New Zealand and Netherlands. Farms were chosed according to the main sources of dairy companies, the distribution of farms and the yield of milk. Content of thiocyanate were detected by ion chromatography. Liquid milk consumption data were taken from Chinese beverage and alcoholic beverage consumption survey in 18 cities or counties in 9 provinces, including 16 775 subjects older than 3. A simple distribution model was used to estimate the exposure of thiocyanate from liquid milk. The tolerable daily intake (TDI) of thiocyanate was made 0.08 mg·kg-1·d-1. Then the exposures of different age groups were compared with TDI.@*Results@#Finally, 1 331 samples out of 2 059 were detected to contain thiocyanate. The detection rate was 65%. The average concentration of thiocyanate was 2.11 mg/kg, with a range of 0.10-16.20 mg/kg. The general population's consumption of thiocyanate by drinking liquid milk was 0.001 mg · kg-1 · d-1, which was lower than TDI. The P95 of general population and consumers were 0.009 mg · kg-1 · d-1 and 0.020 mg·kg -1·d-1 respectively, which were also lower than TDI. Mean exposures of population aged 3-6, 7-12, 13-17, 18-59 as well as elderly aged 60 and above were 0.007, 0.003, 0.002, 0.001 and 0.001 mg · kg-1·d-1 respectively, which were all lower than TDI.@*Conclusion@#The results suggested that the health risk of thiocyanate exposure by drinking liquid milk among Chinese population was at a low level. However, milk products for children deserve more concern.

Bone mesenchymal stem cells (BMSCs), which are force-sensitive cells, have potentials for differentiation into cells with various types. Their biological characteristics can change functionally according to the appropriate stimulation in vitro, in order to reach the optimal demand of the stimulation. Many signal pathways are involved in osteogenesis. Signal transducers and activators of transcription 3 (STAT3) is a ubiquitously expressed transcription factor, mediating cell proliferation, differentiation, survival, apoptosis and cellular immunity. It has been reported that STAT3 can regulate the differentiation process of BMSCs into osteoblasts. This paper summarizes the recent progress about effect of STAT3 on bone differentiation of BMSCs and the possible mechanism.

ABSTRACT

Objective To explore the clinical effect of Weikangling and Weifuchun treatment of chronic atrophic gastritis and precancerous lesions.Methods 85 cases of CAG precancerous lesions treated in our hospital from Jan.2012 to Dec.2016 were selected as the research object,and were randomly divided into control group (n =41) and study group (n =44).The control group were given Weifuchun,and the study group were given Wei Kangling.The pathological score and clinical efficacy were compared between the two groups before and after treatment.Results After treatment,inflammation,atrophy,intestinal metaplasia scores of the two groups were significantly decreased than before treatment in addition to dysplasia (P < 0.05),and inflammation and intestinal metaplasia in the study group were significantly lower than those in the control group (P < 0.05).The total effective rate of the study group was 90.91%,which was higher than75.61% of the control group (P < 0.05).Conclusions The effect of Wei Kangling in treating precancerous lesions of CAG is significant,especially for the histological changes of inflammation and intestinal metaplasia.

ABSTRACT

The method of physical fingerprint spectrum for Reduning injection (RI) was proposed in this paper to improve its quality standards based on the strong correlation between physicochemical properties of drugs, their safety, effectiveness and stability. The quality of RI was studied by the thought and method of physical chemistry. The physical fingerprint spectrum was visually showed by the radar map, and consisted of eight indexes (pH, conductivity, turbidity, refractive index, osmolarity, surface tension, relative density, and kinematic viscosity). Then 12 batch of samples were verified. It was found that the physical fingerprint spectra of 3 batches of RI were in line with the standards within their validity time, with similarity above 0.999; in addition for the expired 9 batches of RI, their physical fingerprint spectra did not meet the standards. The results showed that physical fingerprint spectrum can be used for the quality control of RI, with a certain exemplary role in the quality evaluation of traditional Chinese medicine injection.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the clinical outcomes of internal fixation or replacement for the treatment of radial head fractures through the extensor digitorum communis splitting approach.</p><p><b>METHODS</b>From July 2012 to May 2015, 25 patients with radial head fractures were reviewed. There were 17 males and 8 females, ranging in age from 20 to 67 years old, with a mean age of 39 years old. Twenty-one patients were treated with reconstruction of plate internal fixation, and 4 patients were treated with radial head replacement. According to Mason classification, 19 cases were type II and 6 cases were type III. All the patients underwent internal fixation or replacement through the extensor digitorum communis splitting approach. The patients were followed up clinically and radiographically until the beginning of fracture union and the entrance of function recovery of elbow motion into a plateau. The functional status of the elbow was evaluated using the Mayo Elbow Performance Index(MEPI). Radiographic signs of post-traumatic arthritis were rated according to the Broberg and Morrey system.</p><p><b>RESULTS</b>All the patients were followed up, and the average duration was 29 months (ranged, 12 to 56 months). The average range of flexion and extension was 120°, the extension was limited by 10°, and the flexion was 135°. The average forearm rotation range was 142°, pronation was 75°, supination was 67°. The mean MEPI was 93±7(ranged, 80 to 100 scores); according to the MEPI scoring criceria, 19 patients got an excellent functional result, 6 good. According to the Broberg and Morrey systems of traumatic arthritis, 19 patients were in grade 0, 6 in grade 1, and no patients in grade 2 or 3. No patients with nonunion of the radial head and failure of internal fixation were found. There were no complications of nerve or vascular injuries, and obvious limitation of elbow rotation. Heterotopic ossification around the elbow occurred in 4 cases, but the motion was not significantly limited. There were no complications such as prosthesis loosening and infection in 4 cases after radial head prosthesis replacement.</p><p><b>CONCLUSIONS</b>The extensor digitorum communis splitting approach is an effective exposure method for internal fixation or replacement in the treatment of radial head fractures.</p>

ABSTRACT

Objective To study the remodeling of alveolar bone and change in expression of forkhead box O1 (FOXO1) during orthodontic tooth movement (OTM) in rat, so as to preliminarily investigate the role of FOXO1 in alveolar bone remodeling induced by orthodontic force. Methods The rat OTM models were established and the left maxillary 1st molars were moved with force of 50 g. The rats were executed on the 1st, 3rd and 7th day of OTM, respectively. HE staining and immunohistochemical staining were used to observe the remodeling of alveolar bone in the inter-radicular region of the 1st molars and expression of FOXO1 at different time points during OTM. Results The 1st molars were constantly moved mesially under orthodontic force. There were more osteoclasts in the alveolar bone of OTM group than that in non-OTM group, and the osteoclasts on the 3rd day of OTM showed the highest activity. The number of active osteoblasts gradually increased in the inter-radicular region of alveolar bone under orthodontic force, with the enhanced osteoblast activity. Expression of FOXO1 in OTM group was elevated compared with non-OTM group. Most osteoblasts in alveolar bone during OTM were FOXO1 positive, and the expression of FOXO1 was gradually increased with the number of osteoblasts increasing. Conclusions Orthodontic force induces bone remodeling of alveolar bone in the inter-radicular region during OTM, and the change in FOXO1 expression may be related to alveolar bone remodeling during OTM.

ABSTRACT

Objective To analyze the effect of different bone defect types on stress distributions of the implant- bone interface and bone-Bio-oss interface under dynamic loads by finite element method. Methods The finite element models for four types of bone defects (fenestrative bone defect, dehiscent bone defect, circular bone defect, vertical bone defect) without the first molars were established, then the models of grafting bone and implants corresponding to each model were established. The vertical load of 100 N and Oblique load of 200 N were used to simulate the dynamic chewing process of 5 stages in a masticatory cycle, and the maximum Von Mises stresses at the implant-bone interface and bone-Bio-oss interface were analyzed. Results In a masticatory cycle, from stage 2 to stage 4, the stresses on implant-bone interface increased by 81.6%, 90.7%, 182%, 106%, respectively, while the stresses on Bio-oss bone and natural bone interface increased by 26%, 13%, 6%, 56.25%, respectively. Conclusions The sequence of bone graft stability for the models with four types of bone defects is: circular bone defect> dehiscent bone defect>fenestrative bone defect>vertical bone defects. In clinic, doctors should screen the high-risk patients according to the sites of the bone grafts, and choose different methods for fixing the bone grafts so as to improve the stability of bone grafting. The axial loading will be more beneficial to the stress distributions on the interface, and the oblique loading should be decreased or avoided in clinical design for the superstructure of implant denture.

ABSTRACT

Objective To study the effect of continuous tensile stress on expression of Forkhead box protein O1 (FoxO1) in MC3T3-E1 cells in vitro during osteogenic differentiation, and explore the role of FoxO1 in the mechanism of continuous tensile stress induced-osteogenic differentiation. Methods MC3T3-E1 cells were seeded and applied with tensile stress at the frequency of 1 Hz and amplitude of 10% by FX-4000TTM mechanical loading system. MC3T3-E1 cells were divided into control, 1 h, 4 h, 6 h, 12 h, 24 h, 48 h, 72 h group, respectively, according to the time subjected to the tensile stress. Alkaline phosphatase (ALP) staining, real-time PCR, Western blotting and immunofluorescence were applied to detect the effects of continuous tensile stress on osteogenesis ability of MC3T3-E1 cells, mRNA and protein expression of FoxO1, and allocation of FoxO1 in MC3T3-E1 cells. Results (1) Continuous tensile stress could promote the osteogenic differentiation of MC3T3-E1 cells. Compared with the control group, the mRNA expression of ALP increased significantly at 24 h, 48 h, and the mRNA expression level of osteocalcin (OCN) reached the peak value at 72 h, which was significantly higher than that in the control group. The mRNA expression of runt-related transcription factor-2 (Runx2) significantly increased at 4 h as compared to the control group, and Runx2 protein level changed accordingly. The ALP staining results of the stress group and control group were significantly different. (2) Continuous tensile stress could increase mRNA and protein expression of FoxO1. The mRNA expression of FoxO1 markedly increased at 24 h, and its protein expression significantly elevated at 12 h. (3) FoxO1 was expressed in the nucleus and cytoplasm at 6 h, and then significantly increased in the cytoplasmat at 24 h. Conclusions 10% continuous tensile stress can stimulate the osteogenic differentiation of MC3T3-E1 cells, up-regulate the mRNA and protein expression of FoxO1 and change the allocation of FoxO1 in MC3T3-E1 cells. The investigation on the change rules of FoxO1 expression and allocation under mechanical stimulation will provide the experimental basis for studying the role of FoxO1 in mechanical stimulation.

Objective To evaluate differences in genes expression of rat bone marrow stromal cells (rBMSCs) under continuous mechanical strain by gene microarray technology.Methods rBMSCs were isolated and cultured in vitro. Continuous stresses with amplitude of 10% and frequency of 1 Hz were applied on rBMSCs for 6 hours by Flexercell mechanical loading system to investigate rBMSC gene expression profiles, and quantitative PCR was used to verify gene expression changes related to osteoblastic differentiation. Results Compared with the control group, 1 244 differentially expressed genes were found in mechanical loading group, among which 793 genes were up-regulated, while 451 genes were down-regulated.GO (gene ontology) analysis suggested that differentially expressed genes were mainly involved in multicellular organismal development, cell differentiation, chemotaxis, cell adhesion and so on. Four signaling pathways as Notch, Wnt, FGF and IGF might participate in the regulation of stress-induced osteoblastic differentiation. PCR validation results were consistent with the gene chip results. Conclusions Mechanical stress could induce osteoblastic differentiation of the BMSCs, while several differentially expressed genes screened by gene microarray may attribute to this process.

The method has been developed to accurately identify the magnitude of health risks and provide scientific evidence for implementation of risk management in food safety. It combines two parameters including consequence and likelihood of adverse effects based on risk matrix. Score definitions and classification for the consequence and the likelihood of adverse effects are proposed. The risk score identifies the intersection of consequence and likelihood in risk matrix represents its health risk level with different colors: 'low', 'medium', 'high'. Its use in an actual case is shown.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the combined subchronic toxicity of bisphenol A (BPA) and dibutyl phthalate (DBP) in male Sprague Dawley (SD) rats.</p><p><b>METHODS</b>Forty 4-week-old male rats weighing 115-125 g were randomly divided into BPA-treated, DBP-treated group, BPA+DBP-treated and control groups and fed with a soy- and alfalfa-free diet containing 285.4 ppm BPA, 285.4 ppm DBP, 285.4 ppm BPA plus 285.4 ppm DBP, and a control diet, respectively, for 90 consecutive days. At the end of the study, the animals were sacrificed by exsanguination via the carotid artery under diethyl etherane aesthesia and weighed. Organs, including liver, kidneys, spleen, thymus, heart, brain, and testis underwent pathological examination. The androgen receptor (AR), gonadotropin-releasing hormone receptor (GNRHR), and progesterone hormone receptor (PR) genes from the hypothalamus were detected by real-time PCR. The biomedical parameters were analyzed.</p><p><b>RESULTS</b>No significant difference was found in food intake, body weight, tissue weight, organ/brain weight ratio, and biomedical parameters among the four groups (P>0.05). However, BPA and DBP up-regulated AR, PR and GNRHR expression levels in rats and showed a synergistic or an additive effect in the BPA+DBP group.</p><p><b>CONCLUSION</b>The combined subchronic toxicity of BPA and DBP is synergistic or additive in male SD rats.</p>

ABSTRACT

Bone metastasis is the most common diagnosis considered for lytic lesions in the bone. Parathyroid cancer presenting as overt bone disease with diffuse lytic lesions secondary to untreated hyperparathyroidism (HPT) is a rare entity nowadays. We report the case of a 50-year-old Chinese lady who presented with diffuse bone lesions consistent with Brown tumours, vertebral body collapse and hypercalcaemia mimicking bone metastasis and was later diagnosed with parathyroid cancer.
