ABSTRACT
ObjectiveTo explore the efficacy of Solifenacin in the treatment of children with idiopathic overactive bladder (OAB). MethodsThe study included 67 children with idiopathic OAB treated in the urology clinic of our hospital from March 2022 to March 2023. After at least 2-week-long behavioral therapy showed no significant therapeutic effect, 52 of the cases in the trial group were given oral Solifenacin 5 mg once daily and the other 15 cases in the control group continued the behavioral therapy. The cases in trial group were subdivided into OAB-dry group (27 cases without urinary incontinence) and OAB-wet group (25 cases with urinary incontinence). The 3-day micturition diary, OAB Symptom Scores (OABSS) and the adverse reactions were recorded and analyzed before, at Weeks 2, 4, 8 and 12 after the treatment. ResultsOf all the 67 cases who completed a 3-month follow-up, 44 were cured including 41 in the trial group and 3 in the control group, 4 presented with adverse reactions. After the 3-month follow-up, the OABSS declined markedly in trial group than in control group (Z=4.524, P<0.001); the cure rates in trial group and control group are 78.8% and 20% respectively, with significant difference (χ2=15.367, P<0.001). At each follow-up,we found increased mean voided volume (F=9.707, P<0.001), reduced mean voiding frequency during daytime (F=3.837, P=0.009) and decreased voiding urgency (χ2=482.835, P<0.001). After the 3-month follow-up, the cure rates in OAB-dry group and OAB-wet group are 81.5% and 76% respectively, with no significant difference (χ2=0.234, P=0.629). ConclusionsIn children with idiopathic OAB, oral Solifenacin 5 mg once daily could significantly increase mean voided volume, reduce mean voiding frequency during daytime, relieve symptoms of urinary urgency and lead to fewer adverse reactions, but is not significantly effective for the treatment of urinary incontinence in OAB-wet children .
ABSTRACT
Astrocytes are the most abundant cell type in the central nervous system (CNS). They provide trophic support for neurons, modulate synaptic transmission and plasticity, and contribute to neuronal dysfunction. Many transgenic mouse lines have been generated to obtain astrocyte-specific expression of inducible Cre recombinase for functional studies; however, the expression patterns of inducible Cre recombinase in these lines have not been systematically characterized. We generated a new astrocyte-specific Aldh1l1-CreER knock-in mouse line and compared the expression pattern of Cre recombinase between this and five widely-used transgenic lines (hGfap-CreER from The Jackson Laboratory and The Mutant Mouse Resource and Research Center, Glast-CreER, Cx30-CreER, and Fgfr3-iCreER) by crossing with Ai14 mice, which express tdTomato fluorescence following Cre-mediated recombination. In adult Aldh1l1-CreER:Ai14 transgenic mice, tdTomato was detected throughout the CNS, and five novel morphologically-defined types of astrocyte were described. Among the six evaluated lines, the specificity of Cre-mediated recombination was highest when driven by Aldh1l1 and lowest when driven by hGfap; in the latter mice, co-staining between tdTomato and NeuN was observed in the hippocampus and cortex. Notably, evident leakage was noted in Fgfr3-iCreER mice, and the expression level of tdTomato was low in the thalamus when Cre recombinase expression was driven by Glast and in the capsular part of the central amygdaloid nucleus when driven by Cx30. Furthermore, tdTomato was clearly expressed in peripheral organs in four of the lines. Our results emphasize that the astrocyte-specific CreER transgenic lines used in functional studies should be carefully selected.

Objective To optimize the processes of extraction and inclusion of volatile oil from Citri Grandis Exocarpium. Methods With yield ratio of volatile oil as evaluation index, single factor experiments were used to study the extraction process of volatile oil. With the inclusion rate of the volatile oil and the yield of inclusion as evaluation indexes, saturated aqueous solution was used to the L9(34) orthogonal experiments to reach the best inclusion process optimization. And the microscopic imaging analysis and X-ray scattering technology were adopted to character the inclusion compound. Results The optimum extraction process of volatile oil was extracted for 10 hours with 10 folds the amount of water, and without soaking. The optimum conditions of inclusion process were as follows: volatile oil (mL): β-CD (g): water (mL) ratio was 1:8:80; inclusion temperature was 50 ℃; inclusion time was 3 hour. The validation experiments of the inclusion rate of the volatile oil and the yield of inclusion were 91.50% and 88.36%. Microscopic imaging analysis and X-ray scattering technology proved the inclusion compound had been formed. Conclusion Optimal extraction and inclusion process are feasible and stable, which can provide certain supporting data for preparation and production.

ABSTRACT
<p><b>OBJECTIVE</b>To compare the difference of sensitivity and specificity on the results of the same samples determined by indirect ELISA and sandwich ELISA.</p><p><b>METHODS</b>A 51 anti-HCV positive serum samples obtained from donors were screened by two indirect ELISA kits initially, and then were detected using one sandwich ELISA kit, and were confirmed by recombinent immunoblot assay (RIBA) and HCV-RNA.</p><p><b>RESULTS</b>To compared with HCV-RNA, false positive rate of two kinds of indirect ELISA and one sandwich ELISA was 40.9%, 59.1% and 2.2% respectively. The positive rates of them were 100%, but the analytical sensitivity of sandwich ELISA was more than indirect ELISA 2 to 6 times.</p><p><b>CONCLUSIONS</b>The sensitivity and specificity of sandwich ELISA was significantly better than those of indirect ELISA.</p>

<p><b>OBJECTIVE</b>Occult hepatitis B infection of voluntary blood donors has been plagued in the serum screening. Determined the OBI through the highly sensitive detection methods Nest-PCR among the blood donors, and then learned occult HBV infection and analysed the genotypes of this area.</p><p><b>METHODS</b>10 080 serums of donors were determined respectively by the imported Abbott HBsAg kit and Beijing Wantai anti-HBc and anti-HBs reagents, obtained the gene and detected DNA sequences by the high sensitive Nest-PCR method.</p><p><b>RESULTS</b>Among 10 080 cases of unpaid blood donors, 108 cases were detected HBsAg positively by Abbott sensitivity kit (positive rate of 1.07%), 767 cases were anti-HBc single - positive (positive rate of 7.67%). 25 patients screened blood donors who tested negative for serum HBsAg and positive for HBV DNA in the 10 080 cases. Occult HBV infection incidence rate was 0.25%. 12 cases were HBV genotype C (48%), 13 cases were genotype B (52%), and no other genotypes. Genotype B has no statistically significant difference to genotype C (P > 0.05). Sequence analysis showed that 5 patients in the HBsAg epitope "a" (aa124 - aa147) have mutation (20%).</p><p><b>CONCLUSION</b>The high proportion of occult hepatitis B infected among voluntary blood donors in our country. Also genotype and mutation was differences in different regions.</p>

Objective To investigate the treatment of postoperative gastric fistula with combined traditional Chinese and western medicine.Methods Altogether 56 cases ofgastric fistula after gastrectomy were randomly divided into a treatment group and a control group.The control group was simply treated with conventional western medicine therapy,while treatment group was treated with self-made compound Wubei powder scattered with gelatin sponge plugging in addition to conventional western medicine treatment.Results The effective rate of the treatment group and the control group was 96.4% and 64.8% respectively.The effective rate of the treatment group was significant better than the control group (p<0.01).Conclusion The combined traditional Chinese and western medicine therapy can promote healing of the fistula,reduce pain during the process of surgery shorten the duration of hospitalization,and decrease the cost of treatment.
ABSTRACT
Objective To investigate an effective method to treat gastric ulcer perforation with integrated traditional Chinese and western medicine. Methods 117 patients were randomly divided into a treatment group and a control group.The treatment group was treated with acupuncture plus Sanbal Powder, and the control group was treated with conventional western medicine exclusively. Results The two groups showed significant difference in the cure rate, the total efficiency, the average inpatient free, and the relapse rate in three years with P<0.01. Conclusion The therapy of integrated traditional Chinese and western medicine will shorten inpatient time, alleviate pain and increase recovery rate when treating gastroduodenal ulcer perforation.
