ABSTRACT

Objective:To investigate the mechanism of Shugan Bushen Yulin decoction in inhibiting voltage-dependent anion-selective channel protein 2 (VDAC2) gene methylation, affecting sperm mitochondrial function, and improving sperm motility through the cyclic adenosine monophosphate/protein kinase A (cAMP/PKA) pathway. Method:Forty male SD rats were randomly divided into the blank group, model group, high- and low-dose Shugan Bushen Yulin decoction groups, and L-carnitine group, with eight rats in each group. Adenine (0.05 g·kg-1) was administered by gavage for 14 d for inducing oligospermia and asthenospermia. Rats in the Shugan Bushen Yulin decoction groups were treated with intragastric administration of 32.4, 8.1 g·kg-1 Shugan Bushen Yulin decoction, respectively, while those in the L-carnitine group received 0.27 g·kg-1 L-carnitine by gavage. Following the measurement of sperm motility using an automatic sperm analyzer, the pathological changes in testicular tissue were observed by hematoxylin-eosin (HE) staining. Sperm mitochondrial membrane potential was detected by flow cytometry. The expression of VDAC2 in the testicular tissue was determined by immunofluorescence assay. Real-time polymerase chain reaction (Real-time PCR) was conducted for detecting VDAC2 mRNA expression in testicular tissue. The methylation of VDAC2 gene was examined using bisulfite sequencing. The cAMP expression in testicular tissue was detected by enzyme-linked immunosorbent assay (ELISA), and the PKA protein expression in testicular tissue by Western blot. Result:Compared with the blank group, the model group exhibited significantly decreased sperm density and motility (P<0.01), increased mitochondrial membrane potential (P<0.01), down-regulated VDAC2 mRNA and protein expression, PKA protein expression, and cAMP content in testicular tissue (P<0.01), and elevated VDAC2 gene methylation (P<0.01). Compared with the model group, L-carnitine and Shugan Bushen Yulin decoction at the high and low doses all remarkably increased the sperm density and motility and mitochondrial membrane potential (P<0.01), up-regulated VDAC2 mRNA and protein expression, PKA protein expression, and cAMP content in the testicular tissue (P<0.01), and lowered the methylation of VDAC2 in testicular tissue (P<0.01). The comparison with the L-carnitine group showed that the sperm density and motility and mitochondrial membrane potential in the low-dose Shugan Bushen Yulin decoction group declined significantly (P<0.01). The VDAC2 mRNA and protein expression, PKA protein expression, and cAMP content in the testicular tissue were significantly down-regulated (P<0.01), while the methylation of VDAC2 was significantly enhanced (P<0.01). Conclusion:Shugan Bushen Yulint decoction may inhibit VDAC2 gene methylation, increase VDAC2 expression, regulate cAMP/PKA pathway, and change mitochondrial membrane potential to enhance the sperm motility.

ABSTRACT

Objective:To explore a new method of diagnosing and identifying renal transplantation clinical almost tolerance through a diagnostic model using plasma proteomics. Methods:From November 2011 to November 2012, plasma samples from 43 subjects were collected and divided into the groups of clinical almost tolerance(18 cases), rejection(12 cases)and healthy control(13 cases). Protemic analysis of plasma samples was performed by surface enhanced laser desorption/ionization time of flight mass spectrometry (SELDI-TOF-MS). Differential mass spectrometry peaks were screened and diagnostic model was established by Biomarker Wizard and Biomarker Pattern software. Identification of mass spectrometric peaks of nodes in the diagnostic model was carried out by searching the databases of SWISS-PROT and TrEMBL using ExPASy's TagIdent tool. Results:A total of 21 differential proteins peaks were obtained( P<0.05). Diagnostic model was composed of five mass spectrum peaks of 2 565.15, 1 966.28, 6 674.78, 1 103.27, 1 716.69 and 1 966.28.The sensitivity, specificity and area under the ROC curve of model were 83.3%, 92.0% and 0.951 respectively for diagnosing clinical almost tolerance.Bioinformatic identification results of mass spectrometric peaks of nodes in model were proteins of ANFB, MCH, TFF1, PDYN and PSME3. Conclusions:Establishing a diagnostic mode by plasma proteomics may be effectively employed for diagnosing clinical almost tolerance in kidney transplant.

ABSTRACT

Objective:To analyze the risk factors of systemic inflammatory response syndrome (SIRS) caused by wasp stings. Methods:A prospective case-control study was conducted to analyze the clinical data of 225 patients with wasp stings admitted to Taihe hospital of Shiyan City and Ankang Central Hospital from January 2014 to December 2018, including 131 males and 94 females, with the age of 49 (41, 60) years. The patients were divided into SIRS group ( n=62) and non-SIRS group ( n=163) according to the SIRS diagnostic criteria. The data were collected including gender, age, sting in the head, sting in waist and back, sting in the abdomen, sting in the limbs, number of stings, admission time, hospitalization day, and mortality. The plasma levels of interleukin(IL)-6 and IL-8 in peripheral blood of the patients were analyzed by ELISA method. The whole genome DNA was extracted from white blood cells, and the IL-6: -174G/C, -572G/C, -597G/A, -634C/G and IL-8: -251A/T, -738T/A, -845T/C, + 396T/G were selected for research. The PCR method was used to perform bidirectional sequencing and comparison after amplification, and record the genotyping and frequency. The risk factors of SIRS caused by wasp stings were investigated by univariate analysis and multivariate Logistics regression analysis. Results:(1) The two groups showed significant differences in sting in the limbs, sting in waist and back, sting in the abdomen, number of stings, hospitalization day and mortality ( P<0.01), while there were no significant differences in gender, age, sting in the head and admission time ( P>0.05). (2) ELISA test showed the plasma levels of IL-6 and IL-8 in SIRS group were higher than those in non-SIRS group ( P<0.01). (3) Three genotypes of CC, GC and GG were observed in IL-6-572G/C. There were significant differences in the frequencies of genotypes and alleles between SIRS group and non-SIRS group ( P<0.01). Three genotypes of AA, AT and TT were observed in IL-8 -251A/T. There were significant differences in the frequencies of genotypes and alleles between SIRS group and non-SIRS group ( P<0.01). (4)Univariate analysis showed sting in waist and back, sting in the abdomen, sting in the limbs, number of stings, IL-6-572G allele, IL-8-251T allele were related to SIRS caused by wasp stings ( P<0.01). (5)Multivariate Logistics regression analysis showed limb stings ( OR=2.15), number of stings ≥10 ( OR=11.10), IL-6-572G allele ( OR=3.91) and IL-8-251T allele ( OR=3.97) were significantly related to SIRS caused by wasp stings ( P<0.05 or 0.01). Conclusions:The plasma levels of IL-6 and IL-8 are increased in patients with SIRS after wasp stings. The limbs stings, number of stings ≥10, IL-6 -572G and IL-8 -251T are all independent risk factors for SIRS caused by wasp stings.

ABSTRACT

Objective:To explore the mechanism of Bushen Huoxue prescription in regulating the related factors in phosphatidylinositol 3-kinase (PI3K)/ protein kinase B (Akt)/ mammalian target of rapamycin (mTOR) pathway and improving ovarian reserve function of rats with diminished ovarian reserve (DOR). Method:Sixty DOR model rats were duplicated by Ataya method (intraperitoneal injection of cyclophosphamide) and then randomized into the model group, estradiol valerate (0.000 9 g·kg-1) group, and high- (33 g·kg-1), middle- (16.5 g·kg-1) and low-dose (8.25 g·kg-1) Bushen Huoxue prescription groups, with 12 rats in each group. Another 12 healthy rats were classified into the blank control group. The rats in each group were given the corresponding drugs by gavage, while those in the blank control group and model group received the same volume of normal saline, once per day, for 14 successive days. After treatment, the ovarian tissue was stained with hematoxylin-eosin (HE) for observing the changes in quantities of primary follicles, mature follicles, and total follicles under a light microscope, followed by the detection of vascular endothelial growth factor (VEGF) expression in the ovarian tissue by immunohistochemistry. The protein expression levels of PI3K, Akt, mTOR, and cysteine-dependent aspartate-directed protease-3 (Caspase-3) in the ovarian tissue were assayed by Western blot, whereas the mRNA expression levels of PI3K, Akt, and mTOR were measured by Real-time polymerase chain reaction (Real-time PCR). Result:As revealed by comparison with the blank control group, the quantities of mature follicles and total follicles in the ovarian tissue of model group were significantly reduced (P<0.05, P<0.01). The protein expression levels of VEGF and Caspase-3 in the ovarian tissue were increased (P<0.05), while the protein and mRNA expression levels of PI3K, Akt, and mTOR were decreased (P<0.05, P<0.01). Compared with the model group, Bushen Huoxue prescription at the high and middle doses elevated the quantities of mature follicles and total follicles to varying degrees (P<0.05, P<0.01), and VEGF increased most significantly in the middle-dose Bushen Huoxue prescription group (P<0.05). Caspase-3 in the low-, middle-, and high-dose Bushen Huoxue prescription groups and the western medicine group declined. The protein and mRNA expression levels of PI3K, Akt, and mTOR were up-regulated in the middle- and high-dose Bushen Huoxue prescription groups (P<0.05, P<0.01), and the levels in the middle-dose Bushen Huoxue prescription group were closer to those in the blank control group. Conclusion:Bushen Huoxue prescription effectively improves the ovarian reserve function of rats with DOR and increases the number of follicles possibly by up-regulating VEGF expression in the ovarian tissue, activating the PI3K/Akt/mTOR signaling pathway, and regulating the content of Caspase-3.

ABSTRACT

Objective:To observe the effect of Jinxiangdan (JXD) on NOD-like receptor pyrin domain-containing-3 (NLRP3)/cysteine-dependent aspartate-directed protease-1 (Caspase-1)/interleukin-1β (IL-1β) signaling pathway in myocardium of rats with myocardial ischemia-reperfusion injury (MIRI) and explore the protective effect and mechanism of JXD against MIRI. Method:Fifty male SD rats were randomly divided into the sham operation group, model group, high- and low-dose JXD groups, and positive drug (Di'ao Xinxuekang) group, with 10 rats in each group. Seven days before modeling, rats in the JXD groups were separately treated with intragastric administration of 0.72 and 0.18 g·kg-1 JXD tablets, the ones in the sham operation group and model group with the same volume of normal saline, and those in the positive drug group with 1.29 g·kg-1 Di'ao Xinxuekang. Twelve hours after the last intragastric administration, the anterior descending branch of the left coronary artery was ligated for 30 min and then re-perfused for 60 min for inducing MIRI. ST segment elevation was detected by electrocardiogram(ECG) for model evaluation. The contents of creatine kinase (CK) and lactate dehydrogenase (LDH) in cardiac tissue were measured by colorimetry. Hematoxylin-eosin (HE) staining was conducted for observing myocardial histopathological changes, followed by the detection of cardiomyocyte apoptosis by DNA in situ nick end-labeling (TUNEL) assay. The protein and mRNA expression levels of NLRP3, Caspase-1, and IL-1β were detected by Western blot and real-time polymerase chain reaction (Real-time PCR), respectively. Result:Compared with sham operation group, the model group exhibited obviously elevated ST segment (P<0.01), enhanced CK and LDH activities in the myocardium (P<0.01), increased apoptotic cardiomyocytes (P<0.01), and up-regulated NLRP3, Caspase-1, and IL-1β protein and mRNA expression (P<0.01). Compared with model group, JXD at both the high and low doses and Di'ao Xinxuekang significantly lowered the ST segment (P<0.05,P<0.01), diminished the CK and LDH activities in myocardial tissue (P<0.05,P<0.01), improved the apoptosis of cardiomyocytes (P<0.05,P<0.01), and down-regulated the mRNA and protein expression levels of NLRP3, Caspase-1, and IL-1β in myocardial tissue (P<0.05,P<0.01). The ST segment of ECG in the low-dose JXD group was increased as compared with that in the Di'ao Xinxuekang group (P<0.05), while the ST segment in the high-dose JXD group was obviously elevated (P<0.05). Besides, the green fluorescence intensities in the low- and high-dose JXD groups and the Di'ao Xinxuekang group remarkably declined (P<0.05,P<0.01). The mRNA and protein expression levels of NLRP3, Caspase-1, and IL-1β in the high-dose JXD group were down-regulated (P<0.05). Conclusion:JXD alleviates MIRI possibly by lowering NLRP3 and IL-1β expression and inhibiting cardiomyocyte apoptosis.

ABSTRACT

To systematically evaluate the efficacy and safety of Ningxinbao Capsules in treatment of arrhythmia by Meta-analysis. Randomized controlled trial(RCT) or quasi-randomized control trial(Quasi-RCT) on Ningxinbao Capsules treating arrhythmia were obtained by computer-based retrieval in CNKI, Wanfang, VIP, SinoMed, PubMed, Web of Science, Cochrane Library and EMbase as well as manual retrieval, with time limit from database establishment to April 7, 2020. According to the inclusion and exclusion criteria of trials, all RCTs were screened and evaluated. Then the effective data were collected and RevMan 5.3 Meta-analysis software was used for analysis. Thirteen trials were included, involving 1 379 patients in total. Ningxinbao Capsules combined with anti-arrhythmia Western medicine were adopted as the intervention, and the patients in control group were treated with the anti-arrhythmia Western medicine alone. Meta-analysis results showed that as compared to control group, Ningxinbao Capsules combined with anti-arrhythmia Western medicine group was superior in clinical efficacy, dynamic electrocardiogram and average heart rate in patients with bradycardia, with indicated statistically significant differences. Ningxinbao Capsules had fewer adverse reactions and could relieve the toxic and side effects of anti-arrhythmia medicine possibly. The study showed that Ningxinbao Capsules played a role in treatment of arrhythmia and was relatively safe. However, due to the limited quality of the included studies, high-quality clinical trials are needed to verify the conclusions.

ABSTRACT

The potential effect mechanism of acupuncture for perimenopausal syndrome is analyzed and summarized from five aspects, i.e. the influence on reproductive endocrine system, the regulation on nervous system, the improvement of immune function, the intervention on free radical metabolism and the regulation of adverse emotions. It is believed the mechanism research of acupuncture in the treatment of perimenopausal syndrome should be deepened in terms of acupoint specificity and the influence of acupoint compatibility based on the characteristics of acupuncture therapy itself. In clinic, the research on therapeuticmethods and treatment frequency should be enhanced.

ABSTRACT

Objective:To explore and study the incidence of frailty and influencing factors in this region, so as to provide ways and targets for the intervention of frailty. Methods:From January 2018 to June 2019, 218 hospitalized patients aged over 60 years old in the Department of Geriatrics of Beijing Longfu Hospital were selected for cross-sectional survey.According to the state of frailty state, the patients were divided into frailty group and non -frailty group.Spss25.0 statistical software was used for statistical analysis. According to the data type, independent sample t-test or corrected t-test and chi square test were used for inter group comparison. Logistic regression analysis or linear regression analysis were used to analyze the risk factors. Results:The age of 218 patients was (73.3±7.5) years old, including 92 males and 126 females.According to the state of frailty, the patients were divided into debilitation group (58 cases) and non-frailty group (160 cases). The incidence of frailty was 26.6% (58/218). The incidence of frailty in age≥75 years old 45.3% (43/95). Compared with non asthenia patients, the proportion of elderly patients with coronary heart disease, cerebrovascular disease, physical activity limitation, fall history, fecal incontinence, urinary incontinence, malnutrition, audio-visual oral disease, solid tumor, anemia, dementia, depression and high-sensitivity C-reactive protein increased significantly in the frail group. However, serum albumin, walking speed, grip strength and body mass index decreased significantly (all P<0.05). After adjusting for confounding factors, binary logistic regression analysis showed that malnutrition ( OR=0.103, 95% CI 5.467-73.930, P<0.001) and physical activity limitation (poor physical function OR=98.149, 95% CI19.400-496.557, P<0.001; moderate OR=11.974, 95% CI 2.525-56.775, P=0.002) were the risk factors of frailty. The dominant grip strength was the protective factor of frailty ( OR=0.903, 95% CI 0.831-0.981, P=0.016). Conclusion:Limited physical activity and malnutrition are risk factors for frailty, and that improving balance disorders, gait speed, and five times sit to stand test may be approaches to frailty rehabilitation.

ABSTRACT

In traditional Chinese medicine herbs (TCM), including Radix Salviae Miltiorrhizae (Danshen), Radix Puerariae Lobatae (Gegen), Radix Angelicae Sinensis (Danggui), and Rhizoma Chuanxiong (Chuanxiong) are widely used for the prevention and treatment of cardiovascular diseases and also often co-administered with Western drugs as a part of integrative medicine practice. Carboxylesterase 1 (CES1) plays a pivotal role in the metabolisms of pro-drugs. Since (S)-2-(2-(6-dimethylamino)-benzothiazole)-4,5-dihydro-thiazole-4-carboxylate (NLMe) has recently been identified by us as a selective CES1 bioluminescent sensor, we developed a rapid method using this substrate for the direct measurement of CES1 activity in rats. This bioluminescence assay was applied to determine CES1 activity in rat tissues after a two-week oral administration of each of the four herbs noted above. The results demonstrated the presence of CES1 enzyme in rat blood and all tested tissues with much higher enzyme activity in the blood, liver, kidney and heart than that in the small intestine, spleen, lung, pancreas, brain and stomach. In addition, the four herbs showed tissue-specific effects on rat CES1 expression. Based on the CES1 biodistribution and its changes after treatment in rats, the possibility that Danshen, Gegen and Danggui might alter CES1 ac-tivities in human blood and kidney should be considered. In summary, a selective and sensitive biolu-minescence assay was developed to rapidly evaluate CES1 activity and the effects of orally administered TCMs in rats.

Those thirty model mice were randomly divided into 3 groups (n=10), diabetes mellitus group (DM), low dose of BBL (DM+L) treated group, high dose of BBL (DM+H) treated group. The mice in the NC group and the DM group were given normal saline per day, and the DM+L group and DM+H group were treated with black tartary buckwheat at the doses of 0.21g/kg·d and 0.42g/kg·d respectively. After 14 days. All mice were executed by cervical dislocation, then blood samples were collected, pancreas and spleen were removed for subsequent experiments. The serum levels of fasting blood glucose (FBG), triglyceride (TG), total cholesterol (TCH) and insulin were detected. TNF-α protein in spleen tissue was detected by ELISA kit. The morphology of pancreas tissue was observed by HE staining, and the spleen coefficient was calculated. The expression levels of insulin receptor substrate-1(IRS-1) mRNA and IRS-1 protein in pancreatic tissue were detected.@*RESULTS@#Compared with the control group, the serum levels of FBG, TC and TCH in the model group were increased significantly, while the serum level of insulin was decreased significantly (P<0.05), the expression of TNF-α protein in spleen tissues was obviously raised, the expressions of IRS-1 mRNA and IRS-1 protein in pancreatic tissue in model group were decreased significantly (P<0.05). Compared with the model group, the serum levels of FBG, TC and TCH were decreased significantly in the BBL treated groups. The serum insulin level, spleen coefficient, TNF-α protein expression level in spleen tissue, IRS-1 mRNA expression and IRS-1 protein expression levels in pancreatic tissue in BBL treated group were increased significantly (P< 0.05). High-dose black tartary buckwheat leaves (0.42g/kg·d) exerted a more significant effect.@*CONCLUSION@#Stem and leaf of black bitter buckwheat has significant therapeutic effects on reducing blood sugar and blood fat in type 2 diabetic mice, and has certain protective effects on pancreas and spleen of diabetic mice.

ABSTRACT

Objective@#To investigate the current status and real performance of the detection of RUNX1-RUNX1T1 fusion transcript levels and WT1 transcript levels in China through interlaboratory comparison.@*Methods@#Peking University People’s Hospital (PKUPH) prepared the samples for comparison. That is, the fresh RUNX1-RUNX1T1 positive (+) bone morrow nucleated cells were serially diluted with RUNX1-RUNX1T1 negative (-) nucleated cells from different patients. Totally 23 sets with 14 different samples per set were prepared. TRIzol reagent was added in each tube and thoroughly mixed with cells for homogenization. Each laboratory simultaneously tested RUNX1-RUNX1T1 and WT1 transcript levels of one set of samples by real-time quantitative PCR method. All transcript levels were reported as the percentage of RUNX1-RUNX1T1 or WT1 transcript copies/ABL copies. Spearman correlation coefficient between the reported transcript levels of each participated laboratory and those of PKUPH was calculated.@*Results@#①RUNX1-RUNX1T1 comparison: 9 samples were (+) and 5 were (-) , the false negative and positive rates of the 20 participated laboratories were 0 (0/180) and 5% (5/100) , respectively. The reported transcript levels of all 9 positive samples were different among laboratories. The median reported transcript levels of 9 positive samples were from 0.060% to 176.7%, which covered 3.5-log. The ratios of each sample’s highest to the lowest reported transcript levels were from 5.5 to 12.3 (one result which obviously deviated from other laboratories’ results was not included) , 85% (17/20) of the laboratories had correlation coefficient ≥0.98. ②WT1 comparison: The median reported transcript levels of all 14 samples were from 0.17% to 67.6%, which covered 2.6-log. The ratios of each sample’s highest to the lowest reported transcript levels were from 5.3-13.7, 62% (13/21) of the laboratories had correlation coefficient ≥0.98. ③ The relative relationship of the reported RUNX1-RUNX1T1 transcript levels between the participants and PKUPH was not always consistent with that of WT1 transcript levels. Both RUNX1-RUNX1T1 and WT1 transcript levels from 2 and 7 laboratories were individually lower than and higher than those of PKUPH, whereas for the rest 11 laboratories, one transcript level was higher than and the other was lower than that of PKUPH.@*Conclusion@#The reported RUNX1-RUNX1T1 and WT1 transcript levels were different among laboratories for the same sample. Most of the participated laboratories reported highly consistent result with that of PKUPH. The relationship between laboratories of the different transcript levels may not be the same.

ABSTRACT

<p><b>OBJECTIVE</b>To report on prenatal diagnosis and follow up of two patients with paternal uniparental disomy of chromosome 6 (pUPD6).</p><p><b>METHODS</b>Fetal cells were subjected to in situ culturing and G-banded chromosomal analysis. DNA samples of the fetuses and their parents were also analyzed with single nucleotide polymorphism microarray (SNP array).</p><p><b>RESULTS</b>Both fetuses had a normal male karyotype. SNP array analysis showed both have carried pUPD6.</p><p><b>CONCLUSION</b>pUPD6 can lead to transient neonatal diabetes mellitus type 1. Homozygous status of recessive mutations, disorder of gene imprinting, and its influence on placental function are the main factors to be considered during prenatal diagnosis and genetic counseling for pUPD6.</p>

ABSTRACT

OBJECTIVE@#To study the effects of adiponin receptor agonist (AdipoRon) on renal injury in type 2 diabetic mice.@*METHODS@#The experiment was carried out on 40 SPF C57/BL6 male mice and they were randomly divided into normal control group (=10) and experimental group (=30). Mice in experimental group were given with high sugar and high fat feed in combination with only an intraperitoneal injection of small dose of streptozotocin to build the model of type 2 diabetes (T2DM), which were randomly divided into three groups, model control group (DM), low dose AdipoRon group (DM + L) and high dose AdipoRon group (DM+H)(=10). Then the change of blood glucose was detected. The serum levels of insulin receptor (INSR), insulin receptor substrate-1 (IRS-1) and tumor necrosis factor-α (TNF-α) in mice were measured by ELASA. Pathological changes of renal tissues were observed with a light microscope after HE staining. The expressions of pancreatic duodenal homebox-1 (PDX-1) and insulin mRNA in renal tissues were detected by RT-PCR. The content of phosphated insulin receptor substrate-1 (p-IRS-1) protein in the kidney was determined by Western blot.@*RESULTS@#Compared with DM mice, blood glucose and TNF-α levels in DM + H mice and DM + L mice were significantly reduced (<0.05), while the expressions of INSR,IRS-1 and the content of p-IRS-1 were increased markedly(<0.05), and the expressions of PDX-1 and insulin mRNA in renal tissue were increased significantly(<0.05, <0.01).@*CONCLUSIONS@#Mice treated with AdipoRon have lower blood glucose and TNF-α levels, and higher protein expression levels of INSR, IRS-1, and higher mRNA expression levels of PDX-1 and insulin, and the content of p-IRS-1. All of these indicate that AdipoRon has a certain effects on renal injury in type 2 diabetic mice.
