ABSTRACT

Triterpenoid saponins are widely used in medicine, health cares, cosmetics, food additives and agriculture because of their unique chemical properties and rich pharmacological activities. UDP-dependent glycosyltransferases (UGTs) are the key enzymes involved in triterpenoid saponin biosynthesis, and play important roles in the diversity of triterpenoid saponin structures and pharmacological activities. This review summarized the UGTs involved in plant triterpenoid saponin biosynthesis based on the sources of UGTs and the types of receptors. Moreover, the application of UGTs in heterologous biosynthesis of triterpenoid saponins based on synthetic biology was also discussed.

ABSTRACT

OBJECTIVE@#This study aimed to assess the associations between maternal drug use, cytochrome P450 ( CYP450) genetic polymorphisms, and their interactions with the risk of congenital heart defects (CHDs) in offspring.@*METHODS@#A case-control study involving 569 mothers of CHD cases and 652 controls was conducted from November 2017 to January 2020.@*RESULTS@#After adjusting for potential confounding factors, the results show that mothers who used ovulatory drugs (adjusted odds ratio [a OR] = 2.12; 95% confidence interval [ CI]: 1.08-4.16), antidepressants (a OR = 2.56; 95% CI: 1.36-4.82), antiabortifacients (a OR = 1.55; 95% CI: 1.00-2.40), or traditional Chinese drugs (a OR = 1.97; 95% CI: 1.26-3.09) during pregnancy were at a significantly higher risk of CHDs in offspring. Maternal CYP450 genetic polymorphisms at rs1065852 (A/T vs. A/A: OR = 1.53, 95% CI: 1.10-2.14; T/T vs. A/A: OR = 1.57, 95% CI: 1.07-2.31) and rs16947 (G/G vs. C/C: OR = 3.41, 95% CI: 1.82-6.39) were also significantly associated with the risk of CHDs in offspring. Additionally, significant interactions were observed between the CYP450genetic variants and drug use on the development of CHDs.@*CONCLUSIONS@#In those of Chinese descent, ovulatory drugs, antidepressants, antiabortifacients, and traditional Chinese medicines may be associated with the risk of CHDs in offspring. Maternal CYP450 genes may regulate the effects of maternal drug exposure on fetal heart development.

ABSTRACT

ObjectiveTo investigate the clinical features of patients with severe alcoholic hepatitis (AH) with different underlying liver diseases and the influencing factors for short-term prognosis. MethodsA retrospective analysis was performed for the clinical data of 170 patients with severe AH who were admitted to Tianjin Third Central Hospital from August 2004 to August 2018, and according to the underlying liver disease, they were divided into group A (27 patients without liver cirrhosis), group B (52 patients with compensated liver cirrhosis), and group C (91 patients with decompensated liver cirrhosis). Related scores were calculated, including Maddrey’s discriminant function (MDF) score, Chronic Liver Failure-Sequential Organ Failure Assessment (CLIF-SOFA) score, Model for End-Stage Liver Disease (MELD) score, age-bilirubin-international normalized ratio-creatinine (ABIC) score, and Glasgow alcoholic hepatitis score (GAHS). An analysis of variance or the Kruskal-Wallis H test was used for comparison of continuous data between multiple groups, and the chi-square test was used for comparison of categorical data between multiple groups. Univariate and multivariate Cox regression analyses were used to screen out the independent influencing factors for the short-term prognosis of patients with severe AH. The Kaplan-Meier method was used to plot survival curves, and the log-rank test was used for comparison of survival rate between groups. The receiver operating characteristic (ROC) curve was used to calculate the area under the ROC curve (AUC) and 95% confidence interval (CI), sensitivity, and specificity for each predictive model, and the DeLong method was used for comparison. ResultsThe 28-day survival rates of patients in groups A, B, and C were 88.9%, 80.8%, and 51.6%, respectively, with a significant difference between the three groups (χ2=1983, P<0.001). The AUCs (95% CIs) of MELD score, MDF score, GAHS score, ABIC score, and CLIF-SOFA score were 0.584 (0.493-0.676), 0.696 (0.605-0.786), 0.644 (0.554-0.735), 0.745 (0.662-0.827), and 0.795 (0.726-0.863), respectively, in predicting 28-day mortality rate, and there were significant differences between CLIF-SOFA score and MDF, MELD, and GAHS scores (all P<0.05); CLIF-SOFA score had a sensitivity of 79.0% and a specificity of 67.9% at the optimal cut-off value of 850 points in predicting 28-day mortality rate. Different underlying liver diseases (hazard ratio [HR]=2.296, 95% CI: 1.356-3887, P=0.002) and hepatic encephalopathy (HR=1.911, 95% CI: 1.059-3.449, P=0.031) at disease onset were risk factors for 28-day prognosis. ConclusionPatients with severe AH with different underlying liver diseases have different clinical features and short-term prognoses. Different underlying liver diseases and hepatic encephalopathy at disease onset are closely associated with the 28-day prognosis of patients with severe AH. CLIF-SOFA score can predict the 28-day prognosis of patients with severe AH.

ABSTRACT

Sepsis, a common critical disease in the intensive care unit(ICU), features high morbidity and mortality. At present, it is mainly tackled with western medicine, which may trigger a series of problems like antibiotic resistance, adverse hormonal reactions, and high cost after a long-term use. Therefore, exploring new efficient, safe, and cheap drugs and treatment modes has become the focus of our research at this stage. By virtue of unique advantages including "the concept of holism and individualized treatment based on syndrome differentiation", Chinese medicine has accumulated quite rich experience in the prevention and treatment of sepsis. In recent years, research on the regulation of Chinese medicine on nuclear transcription factor-κB(NF-κB) signaling pathway in sepsis has kept emerging. On this basis, this paper reviewed the etiology and pathogenesis of sepsis, syndrome differentiation and treatment, NF-κB signaling pathway, and its intervention with Chinese medicine. It has been found that some single Chinese herbs and their extracts, Chinese herbal compounds, and Chinese herbal injections effectively inhibit the expression of such inflammatory factors as NF-κB-mediated tumor necrosis factor-α(TNF-α), interleukin-1(IL-1), and IL-6 as well as the related proteins, reduce the systemic inflammatory response and organ injury, and improve the prognosis by regulating the activation of NF-κB signaling pathway and the immune function of macrophages. However, due to the limitations of objective conditions, some studies also have the problems of fuzzy pro-inflammatory anti-inflammatory balance mechanism, unclear pharmacokinetics and low drug safety evaluation, which need to be further studied and explored in order to provide a new theoretical basis and diagnosis and treatment thinking for the treatment of sepsis with traditional Chinese medicine.

ABSTRACT

Objective:To investigate the effect of Jianpi Bufei prescription (JPBFP) on airway inflammation, airway hyperresponsiveness (AHR), and cyclic adenosine monophosphate (cAMP) signaling pathway activity in ovalbumin (OVA)-sensitized and challenged juvenile asthma rats. Method:Seventy-five male SD rats were randomly divided into a blank group (n=15) and an experimental group (n=60). The rats in the experimental group were sensitized by aluminum hydroxide gel containing 0.2% OVA and stimulated by aerosol inhalation of normal saline containing 1% OVA to induce an asthma model, followed by assignment into the following groups: a model group (n=15), a JPBFP group (n=15, 8.37 g·kg-1·d-1), an aminophylline group (n=15, 40 mg·kg-1·d-1), and a dexamethasone group (n=15, 0.1 mg·kg-1·d-1). AHR was detected by the pulmonary function analyzer, changes in inflammatory cells by white blood cell (WBC) count and differential blood count in bronchoalveolar lavage fluid (BALF), and pathological changes of lung tissues by hematoxylin-eosin (HE), Masson, and periodic acid-schiff (PAS) staining. The interleukin (IL)-4, IL-5, IL-13, interferon (IFN)-γ, and tumor necrosis factor (TNF)-α levels in serum and the cAMP level in plasma were tested by the enzyme-linked immunosorbent assay (ELISA). Protein kinase A (PKA) expression in lung tissues was detected by immunohistochemistry. The cAMP-response element-binding protein (CREB) mRNA and protein expression in lung tissues was detected by the real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) and Western blot. Result:Compared with the blank group, the model group showed increased lung resistance, decreased pulmonary compliance (P<0.05), elevated WBC count and proportion of eosinophils in BALF (P<0.05), up-regulated levels of IL-4, IL-5, IL-13, and TNF-α in peripheral blood, declining IFN-γ level (P<0.01), severe pathological changes of lung tissues, dwindled cAMP, and down-regulated PKA and CREB expression (P<0.01). Compared with the model group, JPBFP inhibited AHR, reduced WBC count and proportion of eosinophils in BALF and lung resistance (P<0.05), improved pathological changes of lung tissues, increased pulmonary compliance, and up-regulated cAMP in serum and PKA and CREB expression in lung tissues (P<0.01). Conclusion:JPBFP can improve AHR, inhibit airway inflammation, and alleviate lung injury in asthma rats. Its mechanism may be related to the up-regulation of the activity of the cAMP/PKA/CREB signaling pathway.

Objective:To observe the clinical efficacy of modified Buyang Huanwutang combined with electroacupuncture (EA) in the treatment of traumatic spinal cord injury (TSCI) due to Qi deficiency and blood stasis. Method:Eighty-seven TSCI patients who met the inclusion requirements were randomly divided into an observation group (n=44) and a control group (n=43). On the basis of comprehensive western medical treatments, patients in the control group were further provided with Wuwei Tongshuan oral liquid,10 mL per time, three times per day, while those in the observation group received modified Buyang Huanwutang, one bag per day,for 12 consecutive weeks. Besides, EA was performed in both groups in the same way, once per day, six times per week, for six weeks in total. The American Spinal Injury Association (ASIA) motor score, modified Barthel index (MBI),visual analog scale (VAS) pain score,Berg balance scale (BBS) score, modified Ashworth scale (MAS) score, spinal cord independence measure-Ⅲ(SCIM-Ⅲ) score, lower limb range of motion (ROM), and Qi deficiency and blood stasis syndrome score before and after treatment were evaluated, followed by the recording of the occurrence of complications during treatment. The brain-derived nerve growth factor (BDNF), nerve growth factor (NGF), vascular endothelial growth factor (VEGF), neurotrophic factor-3 (NT-3), malondialdehyde (MDA) and superoxide dismutase (SOD) levels before and after treatment were determined. Result:The motor, light touch, needling sensation, MBI, and BBS scores of the observation group were higher than those of the control group (P<0.01), while the AS and MAS scores were lower(P<0.01). The angles of adductor and straight leg raising in the observation group were greater than those of the control group (P<0.01),but the Qi deficiency and blood stasis syndrome score was lower(P<0.01). Both the scores of self-care, respiration, and sphincter management in SCIM-Ⅲ and the total score in the observation group were elevated as compared with those of the control group (P<0.01). The cumulative incidence of complications in the observation group was 34.09%,significantly lower than 55.81% in the control group (χ2=4.149,P<0.05). Compared with the control group, the observation group exhibited remarkably increased BDNF, NGF, VEGF, NT-3, and SOD (P<0.01) and decreased MDA (P<0.01). Conclusion:Modified Buyang Huanwutang combined with EA is effective in alleviating spinal cord injury, promoting neural functional recovery, improving independence in activities of daily living, reducing the incidence of complications of patients with TSCI, which may be related to the amelioration of ischemia and hypoxia, inhibition of lipid peroxidation, and acceleration of nerve cell repair and regeneration.

ABSTRACT

The aim of this paper was to explore the effect and mechanism of Jiawei Baitouweng Decoction(JWBTW) against ulcerative colitis(UC) from the perspective of intestinal mucosal tight junction proteins. From 60 SPF-grade male SD rats, 10 were randomly selected as the blank control, and the remaining 50 were treated with 3% dextran sodium sulfate(DSS) solution to induce UC and then randomized into the model group, mesalazine group, and low-, medium-, and high-dose JWBTW( L-JWBTW, M-JWBTW and H-JWBTW) groups, with 10 rats in each group. After successive medication for 14 days, the rat general conditions like body weight and stool were observed and the disease activity index(DAI) was calculated. The pathological changes in colon tissue was observed under a microscope for injury severity scoring and histopathological scoring. The serum endotoxin content was determined by limulus assay, followed by the measurement of protein expression levels of ZO-1, occludin, claudin-1, p38 MAPK, MLCK, MLC2 and p-MLC in colon tissue by Western blot. The results showed that compared with the blank group, the model group exhibited significantly reduced body weight, elevated DAI, injury severity and histopathological scores and serum endotoxin content, up-regulated protein expression levels of p38 MAPK, MLCK, MLC2 and p-MLC, and down-regulated ZO-1, occludin and claudin-1. Compared with the model group, mesalazine and JWBTW at each dose obviously increased the body weight, lowered the DAI, injury severity and histopathological scores and serum endotoxin content, down-regulated the protein expression levels of p38 MAPK, MLCK, MLC2 and p-MLC, and up-regulated the ZO-1, occludin and claudin-1, with the most obvious changes noticed in the H-JWBTW group. All these have indicated that JWBTW exerts the therapeutic effect against UC by inhibiting the activation of p38 MAPK/MLCK pathway, reversing the protein expression levels of occludin, claudin-1 and ZO-1, decreasing the serum endotoxin content, promoting the repair of intestinal mucosal mechanical barrier, maintaining the integrity of tight junctions, and reducing the permeability of intestinal mucosa.

ABSTRACT

Objective:To identify new host substrate of SseK3 and study its biological function. Methods:A yeast two hybrid system (Y2H) was used to identify the potential binding proteins of SseK3 from the Hela cDNA library; the arginine N-acetylglucosamine (Arg-GlcNAc) modification of the substrate protein by SseK3 was detected by co-expression in 293T cells and in vitro activity test; the modification sites of the substrate protein by SseK3 were detected by point mutation; the effect of Arg-GlcNAc modification of the substrate protein on its interaction protein binding ability was detected by immunoprecipitation test. Results:Results of Y2H and gene sequencing showed that Snapin was a new substrate of SseK3. Snapin could be Arg-GlcNAc-modified by SseK3 in vivo and in vitro; the modification sites of Snapin were arginine 119 and arginine 120; Arg-GlcNAc-modified Snapin inhibited its binding with SNAP25. Conclusions:Snapin, a new host substrate protein of SseK3, was successfully screened in this study. The Arg-GlcNAc modification of Snapin by SseK3 was studied, and the effect of this modification on Snapin function was preliminarily studied, which provided theoretical basis for further understanding the function of Arg-GlcNAc modification of bacteria and the mechanism of action in the process of pathogen infection.

ABSTRACT

Terpenes are the largest group of natural products and contain the widest assortment of structural types. Terpene cyclization is also the most complex reaction found in nature. For a long time, terpenoids with diverse structures have attracted natural product chemists to explore their biosynthesis mechanism. Such a large number of terpene skeletons are catalyzed by enzymes called terpene synthase. Sesquiterpene synthase is a kind of terpene synthase, which can catalyze the cyclization of linear precursor farnesyl pyrophosphate(FPP) to sesquiterpene skeletons. Sesquiterpene synthase cyclize a single precursor FPP into many sesquiterpene skeletons. With the continuous discovery of sesquiterpene synthase, the cyclization mechanism of sesquiterpene synthase has been studied deeply. In recent years, with the development and improvement of isotope labeling of substrate FPP and structural analysis of sesquiterpene synthase, the structure and cyclization mechanism of sesquiterpene synthase have been studied more systematically and accurately. In this review, we reviewed the progress of the research methods on the mechanism of sesquiterpene cyclization by substrate isotope labeling and protein structure, as well as the summary and prospect of sesquiterpene synthase research.

ABSTRACT

Cadinanes are a class of bicyclic sesquiterpenes with complex stereochemistry and broad pharmacological activities, such as antibacterial, anti-inflammatory, and hypoglycemic activities. To date, structurally diverse and bioactive cadinane sesquiterpenes have been isolated and identified from a variety of plants and microorganisms. Moreover, deeper understandings on cadinane sesquiterpene synthases have been made. This article categorized the 124 new cadinanes which were published in the literatures in the past four years (2017-2020) into five structural types, and presented their pharmacological activities. We also illustrated the elucidation of the biosynthetic pathways for typical cadinanes, summarized the research progress on cadinane sesquiterpene synthases. Finally, current challenges and future prospects were proposed and discussed.

ABSTRACT

Medicinal natural products derived from plants are usually of low content and difficult to extract and isolate. Moreover, these compounds are structurally complex, making it difficult to obtain them by environmental unfriendly chemical synthesis. Biosynthesis of medicinal natural products through synthetic biology is a novel, environment-friendly and sustainable approach. Taking terpenoids (ginsenosides, paclitaxel, artemisinin, tanshinones), alkaloids (vincristine and morphine), and flavonoids (breviscapine) as examples, this review summarizes the advances of the biosynthetic pathways and synthetic biology strategies of plant-derived medicinal natural products. Moreover, we introduce the key technologies and methods of synthetic biology used in the research of medicinal natural products, and provide future prospects in this area.

ABSTRACT

Objective:To know the status of knowledge, attitude and behavior related to sexual and reproductive health in high school students in three regions of East China, and to provide a reference for adolescent reproductive health education. Methods:From August to October in 2019, 614 high school students were invited from 6 high schools in Jiading District and Yangpu District, Shanghai and Taicang City, Jiangsu to participate in this study. We conducted an anonymous self-questionnaire survey using structured questionnaires based on adolescent reproductive health knowledge and literacy. Results:The male to female ratio of all high school students in the study was 1∶1.25, and the average age was (16.1±0.9) years old. The score of pregnancy and abortion knowledge was the lowest among the reproductive health knowledge scores, and the differences among the three regions were statistically significant (P=0.002). Male high school students (P<0.001), students in the school with reproductive health education base (P=0.008) and students who wanted to obtain reproductive health education (P=0.002) were more acceptable to premarital sex. The high school students obtained adolescent health knowledge mainly through the internet or mobile phones, and had a demand for reproductive health related services. Conclusion:High school students in the three regions have a more open attitude towards premarital sex, but have a poor grasp of correct and efficient contraceptive knowledge. Responsible departments need to strengthen the health education of relevant knowledge, and at the same time to find new forms of education to effectively improve the level of adolescent reproductive health.

ABSTRACT

Objective:To analyze the effects of espO gene knockout on the biological characteristics of enterhemorrhagic Escherichia coli (EHEC). Methods:Two-step methods mediated by the suicide plasmid pCVD442-Δ espO and plasmid pTrc99a were used to construct the espO gene-deleted strain (Δ espO) and the complemented mutant (CΔ espO), respectively. HeLa cells were infected with different EHEC strains to analyze the biological functions and lethal effects of espO gene during infection. Results:PCR, electrophoresis and gene sequencing showed that the Δ espO and CΔ espO mutants were successfully constructed. Compared with the wild-type strain, neither the Δ espO nor CΔ espO mutant showed significant difference in growth rate, indicating that the espO gene had no influence on the growth and replication of EHEC. Furthermore, EspO could activate the tumor necrosis factor receptor (TNF)-induced NF-κB signaling pathway, while the effector protein NleB could inhibit the process. EspO could not inhibit the death of HeLa cells induced by TNF or TNF-related apoptosis-inducing ligand (TRAIL) after EHEC infection. Conclusions:In this study, we successfully constructed the espO gene-deleted and complemented mutants of EHEC and preliminarily analyzed the interaction between espO gene and host cells and the effects of espO gene on cell apoptosis during infection, which provided reference for further research on the in vitro biochemical activity and in vivo pathogenic roles of EspO.

ABSTRACT

Objective:To understand the status and knowedge, attitude, practice(KAP) of adolescent sexually transmitted disease(STD) patients in Shanghai, and provide evidence for the development of health interventions. Methods:A cross-sectional study was conducted by anonymous interview survey using self-made questionnaires in patients during 2017.10.1-2017.12.31. Consecutive variables were analyzed using Mann-Whitney U test. Classification variables were analysed using Chi-square analysis and logistic analysis. Results:Regarding to sex and reproductive health, the knowledge awareness rate of STD patients was very low. The rate of correctly answering most questions was less than 5%. The patients were more open to risky sex, while the proportion of positive attitude towards safe sex was lower. Men were more open than women to risky sex. For example, 34.7% and 35.7% of men accepted homosexuality and one-night stand, respectively, but only 1.5% and 12.4% of women showed acceptance. There was statistically significant difference in knowledge scores of STD between different genders (P=0.020). The knowledge score of female patients was higher than that of male patients. There were statistically significant differences in unsafe sex and preventive health care attitudes among patients of different gender (P<0.001) and sex orientation (P<0.001). The attitudes of female patients were better than those of male patients, and the attitudes of heterosexual patients were better than those of sexual minorities. There was statistically significant difference in sexual behavior among patients with different sexual orientation (P=0.014), and the behavior score of sexual minority patients was better than that of heterosexual patients. The influencing factors of sex and reproductive knowledge were gender (P=0.001) and native place (P=0.004).
