ABSTRACT

Objective:To explore the diagnosis and treatment of pure red cell aplastic anemia (PRCA) caused by parvovirus B19 (HPV-B19)infection after liver transplantation (LT).Methods:Three adult PRCA patients caused by parvovirus B19 infection after LT were reviewed retrospectively.The relevant literatures were collected to sort out the detection methods and treatment of parvovirus B19 infection after LT.Results:All three patients received liver transplantation due to end-stage liver disease with massive intraoperative blood transfusion and smooth postoperative recovery.Severe anemia occurred at 1-2 Months after discharge.Hemorrhagic anemia was excluded after re-admission and PRCA was diagnosed by bone marrow aspiration and next generation sequencing (NGS). After tapering the intensity of immunosuppressive therapy, intravenous immunoglobulin (IVIG) was administered for 7-10 days and hemoglobin soon normalized.A review of 15 recent literatures on HPV-B19 infection after LT revealed that the diagnosis and treatment of parvovirus B19 infection after LT gradually were became same.Conclusions:HPV-B19 infection causes PRCA after LT in adults.Diagnosing with NGS, intravenous injection of immunoglobulin and modification of immunosuppressant regimen may achieve excellent therapeutic efficacies.

ABSTRACT

Objective:To investigate the role of microRNA (miRNA)-101a in the liver fibrosis induced by activated hepatic stellate cell (HSC), through upregulating IRE1α signaling pathway.Methods:Carbon tetrachloride (CCl 4) induced liver fibrosis model of mice was established. RT-PCR was used to measure the mRNA level of miRNA-101a in liver tissue of mice. Protein level of α smooth muscle actin(αSMA), collagen I and IRE1α were investigated by Western blot. It was divide into Vehicle, TGFβ1, TGFβ1+ miRNA-NC and TGFβ1+ miRNA-M groups. TGFβ1+ miRNA-NC and TGFβ1+ miRNA-M group were transfected with miRNA-101a mimic negative control and miRNA-101a mimic, respectively. After the corresponding treatments, mRNA level of miRNA-101a was detected by RT-PCR, the protein level of αSMA、collagen I and IRE1α were measured by Western blot. Results:Compared to normal mice, the fibrotic deposition in liver tissue of CCl 4 group was increased significantly [(0.17±0.06) vs. (2.09±0.39), P<0.001)]. Protein level of αSMA, collagen I and IRE1α was increased significantly in the model group [(1.00±0.23) vs. (4.09±0.80), (1.00±0.21) vs. (4.98±1.19), (1.00±0.24) vs. (3.27±0.65), all P<0.001)]. While the mRNA level of miRNA-101a was decreased (1.00±0.05) vs. (0.43±0.05), P<0.001). In vitro study, we found that TGFβ1 could inhibit the mRNA expression of miRNA-101a, induced HSC-T6 activation and then up-regulated protein expression of αSMA, collagen I and IRE1α. Compared to TGFβ1+ miRNA-NC group, the expression of miRNA-101a in TGFβ1+ miRNA-M group increased significantly [(0.59±0.19) vs. (1.89±0.20), P<0.001)]. The protein levels of αSMA, collagen I were reduced by over expression of miRNA-101a [(2.65±0.69) vs. (0.84±0.13), (3.15±0.59) vs. (1.31±0.25), all P<0.05)], and the protein content of IRE1α was down-regulated [ (2.63±0.47) vs. (1.03±0.15), P<0.001)]. Conclusion:miRNA-101a may play a critical role in the inhibition of HSC activation and liver fibrogenesis by blocking IRE1α signaling pathway.

ABSTRACT

Objective To evaluate the effect of mild hypothermia on the renal ischemia-reperfusion injury (IRI), and the expression profile of RNA-binding motif protein 3(RBM3) and its downstream effector molecules during this process. Methods Eighteen healthy SD male rats were randomly divided into the normal control (NC) group, IRI group and mild hypothermia pretreat (MHP) group, with 6 rats in each group. Serum creatinine level was measured to evaluate the renal function. Hematoxylin-eosin (HE) staining was performed to assess the renal tissue injury. Western blot was used to determine the relative expression levels of RBM3, Yes-associated protein 1(YAP1), nuclear factor E2-related factor 2(Nrf2), B cell-lymphoma-2(Bcl-2) and Bcl-2-associated X protein (Bax) in the kidney tissues. Immunohistochemical staining was employed to further detect the expression levels of RBM3 and YAP1 proteins. Terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL) assay was adopted to detect the cell apoptosis of kidney tissues. Malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were evaluated to determine the oxidative stress level of kidney tissues. Results Compared with the NC group, the serum creatinine level, the pathological injury score of kidney tissues and the expression levels of RBM3, YAP1 and Nrf2 proteins were significantly up-regulated, the Bcl-2/Bax ratio was considerably lower, the apoptosis rate was remarkably elevated, the MDA content was significantly increased and the SOD activity was dramatically reduced in the IRI and MHP groups (all P < 0.05). Compared with the IRI group, the serum creatinine level and the pathological injury score of kidney tissues were significantly decreased, the expression levels of RBM3, YAP1 and Nrf2 proteins were significantly up-regulated, the Bcl-2/Bax ratio was considerably higher, the apoptosis rate was significantly decreased, the MDA content was significantly decreased and the SOD activity was considerably elevated in the MHP group (all P < 0.05). Conclusions Mild hypothermia may exert protective effect upon renal IRI and it could alleviate cell apoptosis and oxidative stress injury induced by IRI, probably by up-regulating the expression level of RBM3 and its downstream effector molecules of YAP1 and Nrf2.

ABSTRACT

Objective:To explore the tracing effect of nano-carbon in the cervical lymph nodes of papillary thyroid carcinoma (PTC) and the guiding role of the dissection strategy of the contralateral lymph nodes.Methods:The medical records of 516 patients with PTC in Department of Endocrine and Breast Surgery, the First Affiliated Hospital of Chongqing Medical University from Jan. 2013 to Dec. 2017, including cN0 patients (436 cases) and cN1 patients (80 cases) , were retrospectively analyzed. There were 137 males and 379 females, the male to female ratio was 1.00:2.76. During the operation, nano-carbon was used to trace lymph nodes, and the number of lymph nodes in each lateral area (area II, III, IV) was collected, and the rate of black stained and non-black stained lymph node metastasis (LNM) was calculated based on the postoperative pathological results. SPSS 22.0 statistical software was used for analysis, t test was used for measurement data, and χ2 test was used for count data. Results:In 436 patients with cN0, the black-stained LNM rate in zone II ( P=0.002) , the black-stained LNM rate in zone III ( P=0.000) , and the black-stained LNM rate in zone IV ( P=0.002) were higher than those of non-black stained LNM. The rate of black-stained LNM in 80 cN1 patients (Ⅱ, Ⅲ, Ⅳ) was also higher than that of non-black-stained LNM (0.011, 0.019, 0.015) . The rate of black-stained LNM in cN0 patients affected the LNM in areas Ⅱ and Ⅳ ( P=0.000, P=0.000) . In patients with cN1, the black-stained LNM rate in zone Ⅲ had an effect on zone Ⅱ (0.030) ; it had no effect on the black-stained LNM rate in zone Ⅳ (0.315) . Conclusion:The black-stained LNM rate of the posterior zone (zone Ⅱ, Ⅲ, Ⅳ) with nano-carbon can represent the LNM rate of the lateral zone and help guide the lymph node dissection in the lateral zone. In addition, the black-stained LNM rate of zone Ⅲ can affect zone Ⅱ. The LNM rate in zone IV has the role of sentinel lymph nodes in the lateral zone.

ABSTRACT

The ticks feed large amount of blood from their hosts and transmit pathogens to the victims. The salivary gland plays an important role in the blood feeding. When the female ticks are near engorgement, the salivary gland gradually loses its functions and begins to rapidly degenerate. In this study, data-independent acquisition quantitative proteomics was used to study changes in the phosphorylation modification of proteins during salivary gland degeneration in Haemaphysalis longicornis. In this quantitative study, 400 phosphorylated proteins and 850 phosphorylation modification sites were identified. Trough RNA interference experiments, we found that among the proteins with changes in phosphorylation, apoptosis-promoting Hippo protein played a role in salivary gland degeneration.

ABSTRACT

Objective@#To investigate the feasibility and accuracy using digital surgical guide to position the jaws in bimaxillary orthognathic surgery.@*Methods@#16 patients with dento-maxillofacial deformity were included from January to December 2018 in Nanjing Stomatological Hospital. There were 10 males and 6 females, all of which were skeletal Ⅲ deformity. The mean age was 21.3 years old (16 to 28 years). They were divided into digital guide (group A) and traditional model surgery (group B) according to the treatment sequence. All patients underwent both Lefort Ⅰ osteotomy and bilateral ascending sagittal split. Virtual surgery was performed and digital three-dimensional surgical template was printed before surgery in group A. During the operation, digital surgical templates were used to position maxilla and mandible. The accuracy was evaluated by generating color distribution map of deviation grade, which was imaging fusion of virtual surgical jaw position and actual surgical jaw position to measure the deviation distance of each marker point one week after surgery. The deviations between the preoperative design and the actual operation were compared with the t-test respectively in group A and group B, and the independent sample t test was chosen to compared the accuracy of reduction between the two groups in sagittal, horizontal, and vertical directions.@*Results@#All 16 patients had primary healing. The maximum deviation of the patients in group A was at U6. The deviation direction was vertical, and the deviation distance was (1.67±0.32) mm. In group A, the deviation distances of all marker points in the sagittal, horizontal and vertical directions were (0.55±0.26) mm (t=2.985, P=0.205), (0.62±0.20) mm (t=2.672, P=0.193) and (1.07±0.62) mm (t=5.122, P=0.823), respectively. The result in group B was (0.63±0.28) mm(t=3.397, P=0.307), (0.71±0.42) mm (t=4.438, P=0.541) and (1.82±0.36) mm (t=1.115, P=0.021) in these directions. There was no significant difference between two groups in the sagittal (t=4.704, P=0.763) and lateral (t=4.008, P=0.615) directions, and the difference in the vertical (t=1.432, P=0.037) direction was statistically significant.@*Conclusions@#The digital guides were feasible to position jaws accurately in orthognathic surgery.

ABSTRACT

Objective To systematically review the obstacles that hindered participants to attend at or adhere to cardiac rehabilitation program. Methods We searched databases including PubMed, Medline, Ovid, EBSCO host, The Cochrane Library, CNKI and Wanfang Data, to find out qualitative and mix-method studies published from establish of data base to April 2017, which explore the barriers patients encountered when they decided to attend at a cardiac rehabilitation program or comply with cardiac rehabilitation suggestions. At the same time, quality of included studies was assessed. Data was synthesized using qualitative ethnography. Results A total of 13 qualitative researches were included in this systematic review. Synthesis of study yields three themes and nine sub-themes, which indicates that "personal factors" "program factors" and"contextual factors" interact and influence patients′ access to cardiac rehabilitation. Conclusions In order to improve participation rate in and firm adherence to cardiac rehabilitation, health workers should help patients to get deep insight into their conditions and cardiac rehabilitation; besides, cardiac rehabilitation prescription should be individually designed and take contextual factors into consideration.

ABSTRACT

The aim of the study was to determine the feasibility of the Vitellaria paradoxa nutshell as a new medicinal resource for treating diabetes. A total of forty-one compounds were identified by HPLC-DAD-Q-TOF-MS and phytochemical methods in V. paradoxa nutshell methanol extract. Based on HPLC fingerprints, four characteristic constituents were quantified and the origin of twenty-eight V. paradoxa nutshells from seven sub-Saharan countries was compared, which were classified into three groups with chemometric method. Twenty-eight samples contained high total phenolic content, and exhibited moderate-higher antioxidant activity and strong α-glucosidase inhibitory activity. Furthermore, all fractions and isolated compounds were evaluated for their antioxidant and α-glucosidase inhibitory activities, and α-glucosidase inhibitory action mechanism of four characteristic constituents including protocatechuic acid, 3, 5, 7-trihydroxycoumarin, (2R, 3R)-(+)-taxifolin and quercetin was investigated via molecular docking method, which were all stabilized by hydrogen bonds with α-glucosidase. The study provided an effective approach to waste utilization of V. paradoxa nutshell, which would help to resolve waste environmental pollution and provide a basis for developing potential herbal resource for treating diabetes.

ABSTRACT

The incidence of thyroid carcinoma is increasing.Because of the special location,patients usually have cosmetic requirements for thyroid surgery.Compared with open surgery,improved Miccoli surgery,total endoscopic surgery,radiofrequency ablation and other minimally invasive surgery have obvious advantages in cosmetology,but the surgical indications are limited.Through incision design and protection,appropriate suture materials,standard suture methods and negative pressure drainage measures,we can effectively reduce the scar formation of open surgery,and improve the cosmetic results.Preoperative individualized treatment should be formulated according to the patient's actual situation,tumor nature,cosmetic needs,hospital conditions and other comprehensive factors.

Objective To construct a mutant strain of Nocardia farcinica ( N. farcinica ) IFM10152 with mammalian cell entry 4A gene (mce4A) deletion and to analyze the function of that gene dur-ing infection. -ethods The mutant strain of N. farcinica was constructed through in-frame deletion without antibiotic labeling and verified by PCR and sequencing analysis. To analyze the function of mce4A gene in the interaction between N. farcinica and host cells, in vitro growth experiment, macrophage killing experi-ment using THP-1 ( a human leukemia mononuclear cell line) as the model and adhesion and invasion exper-iments using HeLa cells ( cervical cancer epithelial cells) were carried out. Results The mutant strain with mce4A gene deletion was successfully constructed and named △mce4A. No significant difference in growth rate was observed between the mutant and the wild-type strains. After knocking out the mce4A gene, the ability of N. farcinica to resist macrophage killing was obviously weakened as well as its ability to adhere and invade. Conclusions The mutant strain of N. farcinica with mce4A gene deletion was successfully construc-ted. The mce4A gene might play an important role in the adhesion and invasion of N. farcinica to host cells and its survival in macrophages.

ABSTRACT

Abstract Purpose: To investigate the apoptotic mechanisms in rabbits with blast-induced acute lung injury (ALI). Methods: A total of 40 rabbits were randomly divided into a blank control group (A, n=10) and an experimental group (EXP, n=30). Explosion-induced chest-ALI models were prepared and sampled at different time points (4, 12, and 24h after modeling, T1-T3) to test the lung dry weight/wet weight ratio (W/D) and arterial oxygen pressure (PaO2), apoptosis of lung tissue by the TUNEL assay, and Caspase-3, Bax, and Bcl-2 levels by immunohistochemical analysis. Furthermore, lung tissue was sampled to observe pathological morphology by microscopy. Results: Under a light microscope, Group EXP exhibited obvious edema in the pulmonary interstitial substance and alveoli, a large number of red blood cells, inflammatory cells, and serous exudation in the alveolar cavity, as well as thickening of the pulmonary interstitial fluid. Compared to Group A, the W/D ratio was significantly increased in Group EXP (P<0.01), while PaO2 was significantly reduced (P<0.01). The apoptosis index was significantly increased (P<0.01), and caspase-3 and Bax/Bcl-2 levels were increased (P<0.01). Conclusion: Apoptosis plays an important role in the occurrence and development of acute lung injury in rabbits by participating in lung injury and promoting the progression of ALI.

Hepatic artery thrombosis (HAT) is the most frequent vascular complication following with liver transplantation,whichis the foremost cause of primary graft nonfunction,graft loss and recipient's death.Hepatic artery thrombosis after liver transplantation wasdivided into early hepatic artery thrombosis (E-HAT) and late hepatic artery thrombosis (L-HAT).And the etiologywascomplex,clinical presentations were diversity,treatment effects were controversial,therefore,the early detection,early diagnosis and early treatment of hepatic artery thrombosis after liver transplantation are very important.In this paper,the progress in the diagnosis and treatment of hepatic artery thrombosis after liver transplantation were reviewed.

ABSTRACT

Objective To systematically review the effect of donor ischaemic preconditioning in liver transplantation.Methods Databases including the Cochrane Library,PubMed,EMbase,CNKI,VIP and WanFang database were searched up to June 2016 for studies which involved donor ischaemic preconditioning (IPC) in liver transplantation.The data retrieved included 1-year mortality,incidence of Primary Graft Non-Functioning (PGNF),intensive therapy unit (ICU) hospitalization and liver function tests which were used to evaluate the treatment outcomes.The data were analyzed using both the fixed-effect and the random-effects models.For categorical outcomes,risk ratio (RR) with 95% confidence intervals (CI) were calculated.For continuous outcomes,the mean difference (MD) with 95% CI were calculated.The metaanalysis was performed using Review Manager 5.2 software.Results Six clinical studies with 322 patients were qualified for this meta-analysis.There were no significant differences in the 1-year mortality (OR =0.51,95% CI 0.24 ~ 1.05,P > 0.05),PGNF (OR =0.33,95% CI 0.08 ~ 1.40,P > 0.05) and ICU hospitalization (OR =-0.17,95 % CI-2.72 ~ 2.38,P > 0.05) between the donor ischaemic preconditioning and the control groups.There were also no significant differences in the transaminase and bilirubin levels on postoperative day 1,3 and 7 between the two groups.Conclusion There is currently not enough evidence in evidenced based medicine to recommend the routine use of ischaemic preconditioning in donor liver retrieval.

ABSTRACT

Objective: To preliminarily analyze the graft flow of right internal mammary artery (RIMA) in patients after coronary artery bypass grafting (CABG) using bilateral internal mammary artery (BIMA). Methods: A total of 52 patients received CABG by BIMA in our hospital from 2015-12 to 2016-07 were studied. The patients were younger than 65 years at the mean age of (56.6±6.8) years including 46 male. According to conduit grafting to left anterior descending artery (LAD), the patients were divided into 2 groups: RIMA anastomosed to LAD group, n=42 and LIMA anastomosed to LAD group, n=10. The immediate graft flow was measured by Veri Q system, surgical outcomes and graft flows were compared between 2 groups. Results: There were no operative death in all 52 patients, 1 had poor wound healing and received debridement and suturing, no one had operative complication in left 51 patients. The average LAD bridge flow in both groups were (28.7±11.5) ml/min and (31.8± 11.7) ml/min, the mean pulsation index (PI) were (2.3±0.7) and (2.0±0.4), P>0.05; the average RIMA graft flow were (28.7±11.5) ml/min and (21.1±11.0) ml/min, the mean PI were (2.3±0.7) and (2.6±1.1), P>0.05. Conclusion: Flows in RIMA-LAD graft and LIMA-LAD graft were similar; the flow of RIMA anastomosed to other target vessel was satisfactory which was an ideal graft for CABG.

BACKGROUND: Increasing studies have shown that solid lipid nanoparticles made from traditional Chinese medicine can inhibit cancer cell proliferation and induce cell apoptosis. OBJECTIVE: To investigate the mechanisms of oridonin solid lipid nanoparticles (ORI-SLN) by the regulation of Wnt/β-catenin signaling pathway on the proliferation and apoptosis of esophageal cancer cells. METHODS: After 0, 2.5, 5, 10, 20 μmol/L ORI-SLN treated human esophageal cancer cell lines Eca-109 for 24, 48, 72 hours, the cell inhibition rate was detected by cell counting kit-8, and the half maximal inhibitory concentration (IC50) was calculated. After 0, 14 μmol/L ORI-SLN treated Eca-109 cells for 48 hours, the cell apoptosis was detected by flow cytometry. The expression of Cleaved caspase3, β-catenin, C-myc, Cyclin D1 proteins was detected by western blot assay. Wnt/β-catenin signaling pathway activator LiCl and LiCl+ORI-SLN were used to treat Eca-109 cells for 48 hours, and then the relevant indicators were reexamined. Eca-109 cells without any treatment were used as controls. RESULTS AND CONCLUSION: The cell inhibition rate of Eca-109 cells treated with different concentrations of ORI-SLN for 24, 48 and 72 hours was significantly higher than that at 0 hour, and the cell inhibition rate was found to increase with the prolongation of time and the increase of the concentration (P < 0.01). 14 μmol/L ORI-SLN was confirmed to result in the higher cell apoptosis and Cleaved caspase3 expression compared with the 0 μmol/L group, while the expression of β-catenin, C-myc, Cyclin D1 proteins were significantly lower than the 0 μmol/L group (P < 0.01). Cell inhibition rate, apoptosis rate and Cleaved caspase3 protein expression in the activator group and ORI-SLN+activator group were significantly higher than those in the control group, and the expression of β-catenin, C-myc, Cyclin D1 protein was significantly lower than those in the control group (P < 0.01). The cell inhibition rate, apoptosis rate and expression of Cleaved caspase3 in ORI-SLN+activator group was significantly lower than those in the activator group, and the β-catenin, C-myc, Cyclin D1 protein expression was significantly higher than that in the activator group (P < 0.01). To conclude, ORI-SLNs can inhibit the proliferation and apoptosis of human esophageal carcinoma cell line Eca-109, and its mechanism is related to the regulation of Wnt/β-catenin signaling pathway.

Brucella is a facultative intracellular pathogen that causes infection in domestic animal and humans,mainly parasitizing in cells and macrophages of hosts.Brucella can lead to abortion and sterility in animals.Meanwhile,it also causes arthralgia,weakness,undulat fever,hepatosplenomegaly and other symptoms in humans.Brucella relies on immune escape mechanism in the confrontation with the host.It can help Brucella "camouflage" to evade the identification of the immune system of the hosts and replicate within cells to persist to establish a long-term infection in the host.As a result of the existence of this mechanism,the treatment of Brucella infection is quite difficult.Type Ⅳ secretion system (T4SS) is a key virulence factor and it is essential for Brucella to survive in host cells.The effector proteins secreted by T4SS can help regulate the immune response against Brucella.In this article,we reviewed the studies on related proteins of the type Ⅳ secretion system of Brucella and its immune response,especially the relationship between the secretions of effector proteins mediated by VirB operon and immunity of the host.

Objective To investigate the protective effects and the possible mechanisms of mild hypothermia against liver L02 cells hypoxic-ischernia reperfusion injury by using mild hypothermia pretreatment.Methods L02 cells were randomly divided into three groups:normal control group (N group),hypoxic-ischemia reperfusion group (control group) and hypoxic-ischemia reperfusion with mild hypothermia pretreatment group (experimental group).Before hypoxic-ischemia reperfusion,cells in experimental group were pretreated with mild hypothermia for 6 h,while the other groups were given the normal culture.Thereafter,the hypoxic-ischemia reperfusion models of L02 cells were performed by a tri-gas incubator to hypoxic-ischemia culture for 12 h,followed by reperfusion with normal conditions for 4 hours.Cells in N group were cultured in normal conditions.The temperature of experimental groups was set to 32 C.The samples were collected,and the cell injury,the cell vitality,the cell apoptosis and the expression of JNK in different groups were detected.Results Compared to N group,the levels of alanine aminotransferase (ALT),aspartate transaminase (AST) and lactic dehydrogenase (LDH) were significantly increased,the cell vitality was significantly decreased,the cell apoptosis and the expression of p-JNK were significantly increased in control and experimental groups (P < 0.05).Compared to control group,all these changes were significantly ameliorated in experimental group.The levels of ALT,AST and LDH in control group were (30.0 ± 4.6),(26.3 ± 3.8) and (1129.0 ± 134.3) U/L,and those in the experimental group were (21.0 ± 2.7),(18.7 ± 2.1)and (898.3 ± 79.2),respectively.The cell vitality in control group and experimental group was (64.33 ± 2.32)% and (78.17± 3.01)% respectively.The cell apoptosis in control group and experimental group was (32.4 ± 2.3) % and (18.8 ± 1.4) % respectively.The expression of p-JNK in experimental group was significantly decreased.All these differences were statistically significant (P<0.05).Conclusion Mild hypothermia pretreatment could significantly attenuate liver L02 cells hypoxicischemia reperfusion injury probably by inhibiting JNK activation.

ABSTRACT

Objective To investigate the different regulation of TLR4 between NF-κB and IRF3 signaling pathways which are triggered by LPS. Methods LPS was used to stimulate mouse peritoneal macrophages. Immunofluores-cence was performed to find the translocation of TLR4 and the transcription factors p65 and IRF3. The phosphoryla-tion level of transcription factors was examined by Western blot. Results With stimulation of LPS in 30 minutes, the fluorescence intensity of TLR4 on the cytomembrane was increased (P<0.01),while its fluorescence intensity on the endosome in cytoplasm was increased more robustly(P<0.01),and TLR4 in cytoplasm was found to co-lo-cate with EEA1. When the stimulation reached 90 minutes and 180 minutes,the fluorescence intensity of TLR4 in both cytomembrane and cytoplasm decreased obviously (P<0.01),and the co-location with EEA1 was attenuated present (P<0.01). In the rest state,the fluorescence intensity of the downstream molecule p65 and IRF3 was al-most totally in the cytoplasm. After the stimulation of LPS, fluorescence intensity both in nucleus was increased gradually (P<0.05),but the fluorescence intensity of p65 in nucleus was enhanced earlier and lasted longer than that of IRF3. The phosphorylation of p65 was also earlier and lasted longer than that of IRF3. Conclusions The changed location of TLR4 after being activated can make the transfer time of IRF3 pathway longer and the lasting time shorter than that of NF-κB robustly.

ABSTRACT

Objective To observe the effect of strengthening training of hip muscles on functional ankle instability (FAI). Methods From May, 2015 to June, 2016, 40 patients with FAI were randomly divided into control group (n=20) and observation group (n=20). Both groups were treated with routine exercise training including muscle strength and proprioceptive training of the ankle joint, while the treat-ment group received hip muscle strength training in addition, for six weeks. They were evaluated with hip force assessment, postural stabili-ty assessment, Star Excursion Balance Test (SEBT) and Cumberland Ankle Instability Tool (CAIT). Results There was no significant differ-ence in all the indexes between two groups before treatment (P>0.05). After treatment, the peak torques of hip muscles increased in the ob-servation group (t>3.528, P<0.01);the overall stability index, anteroposterior stability index, middle-lateral stability index decreased in both groups (t>2.360, P<0.05);the distance of eight direction in SEBT increased in both groups (t>2.254, P<0.05);the score of CAIT increased in both groups (t>3.268, P<0.01). All the above indexes were better in the observation group than in the control group (t>2.161, P<0.05). Conclusion Strengthening training of hip muscle, based on conventional ankle muscle strength and proprioceptive training, could facilitate to improve FAI.
