ABSTRACT

Objective To investigate the expressions of HOXA11 and β-catenin proteins in colorectal serrated lesions and their roles in carcinogenesis. Methods A total of 252 cases of colorectal biopsy specimens in Shanxi Dayi Hospital from January 2012 to December 2016 were analyzed retrospectively, including 97 serrated lesions, 46 common adenomas, 109 adenocarcinomas, and 24 normal colorectal mucosa tissues were selected as controls. The expressions of HOXA11 and β-catenin proteins were detected by immunohistochemical EnVision method. Methylation of HOXA11 gene was detected by specific methylation polymerase chain reaction (MSP) in 25 paraffin-embedded adenocarcinoma tissues. Results In 97 serrated lesions, 29 (29.9%) occurred in the left colon;in 46 common adenomas, 27 (58.7%) occurred in the left colon;in 109 adenocarcinomas, 76 (69.7%) occured in the left colon. The difference of the occurrence location among three groups was statistically significant (χ2 = 34.75, P< 0.01). The heterotopic expression rate ofβ-catenin protein in serrated lesions, common adenomas and adenocarcinomas was significantly higher than that in normal mucosae [96.9% (94/97), 82.6% (38/46), 86.2% (94/109) vs. 0 (0/24), P < 0.01]. The heterotopic expression of β-catenin protein was found in serrated lesions and the co-expression of cytoplasm and nucleus was found in common adenomas and adenocarcinomas. The normal expression rate of HOXA11 protein in serrated lesions, common adenomas and adenocarcinomas was lower than that in normal mucosae [33.0% (32/97), 67.4% (31/46), 48.6% (53/109) vs. 100.0% (24/24), all P< 0.05]. The methylation rate of HOXA11 gene was 84.0% (21/25). Conclusion The ectopic expression of β-catenin in colorectal serrated lesions suggests that it is associated with serrated lesions, and the low expression of HOXA11 may be an early event in the carcinogenesis of serrated lesions.

ABSTRACT

Objective To study the expression of HPV、p16 and Her-2 in the squamous cell cervical carcinoma and its chnical significance. Methods Expression of HPV16/18、p16, and C-erbB-2 and the amplification of Her-2 gene were examined using situ hybridization technique,SP immunohistochemistry,and FISH imaging analysis system in 60 cases of cervical cancer, 61 cases of CIN, and 21 cases of normal cervical tissue,respectively.Results The positive rates of HPV16/18 and p16 in the normal tissue,CIN Ⅰ -Ⅱ,CINⅢ and the squamous cell cervical carcinoma were gradually increased, they wereo (0/21),9.68 % (3/31),46.67 % (14/30),71.67 % (43/60);0 (0/21),19.35 % (6/31),93.33 % (28/30),96.67 % (58/60),respectively,and there were significant differences among the groups (P<0.05),but no significant difference was found between the normal tissue and the CIN Ⅰ - Ⅱ. The positive rates of Her-2 and Her-2 in the CIN Ⅲ and the carcinoma were 13.33 % (4/30),31.67 % (19/60),3.33 % (1/30),21.67 % (13/60),respectively,but in the normal group and the CIN Ⅰ - Ⅱ wereo,and the differences between the carcinoma group and the CIN group,the carcinoma group and the normal group were significant(P<0.05).The expression of Her-2 and the amplification of Her-2 were closely related to the stage, degree of differentiation and metastasis of lymph node in the squamous cell cervical carcinoma (P<0.05). Conclusion The infection of HPV is one of thetriggers for the squamous cell cervical carcinoma.The expression of p16 and the amplification of Her-2 may be closely correlated with tumor development and high expression of p16 and Her-2 indicates poor prognosis.

ABSTRACT

Objective To investigate the role of cell cycle regulatory protein CDK4,p18,p19 in the genesis and development of esophageal squamous cell carcinoma (SCC).Methods Tissue microarray and immunohistochemical method (Envision) were used to detect the protein expression of CDK4,p18,p19 in 120 cases of esophageal tissues.The results were statistically analyzed.Results The positive rate of CDK4 protein expression in normal esophageal epithelium was low [28.3 % (34/120)],it increased in esophageal intraepithelial neoplasia [32.5 % (39/120)],and it was high in esophageal SCC [84.2 % (101/120)],which increased with the degree of SCC differentiation decreasing gradually.There was significant differences between the SCC and normal esophageal epithelium or esophageal intraepithelial neoplasia (x2= 76.004,P <0.05; x 2= 65.897,P < 0.05).The expression of CDK4 in group with lymphatic metastasis [93.88 % (46/49)]was higher than without it [71.43 % (55/71)] (x2= 5.860,P < 0.05).The positive rates of p18,p19 protein expression in normal esophageal epithelium were high [34.2 % (41/120),29.2 % (35/120)],it decreased in esophageal intraepithelial neoplasia [19.2 % (23/120),15.0 % (1 8/120)] (x 2= 134.481,P < 0.05; x 2 = 141.376,P < 0.05),but it were high in esophageal SCC [63.3 % (76/120) and 61.7 % (74/120)] which decreased with the degree of SCC differentiation gradually increased.There were significant differences between the normal esophageal epithelium and esophageal intraepithelial neoplasia,esophegeal intraepithelial neoplasia and SCC,normal esophageal epithelium and SCC (p 18:x 2 = 6.903,48.296,20.429,P < 0.05; p1 9:x2 = 6.998,55.276,25.565,P< 0.05).CDK4 protein expression was correlated with both p18 and p19 (r =0.696,0.630,P <0.05),and there was significant positive correlation between the protein expression of p18 and p19 (r =0.833,P <0.05).Conclusion Cell cycle regulatory gene CDK4,p18,p19 get involved in the genesis and development of esophageal squamous cell carcinoma.Their protein expressions are closely related to canceration of esophageal epithelium.

ABSTRACT

Objective To investigate the expression of EGFR, Her-2 and TOPO Ⅱ in esophageal canceration course, analyze the correlation between the expression and clinical pathological parameters and the correlation of the three genes. Methods EGFR, Her-2 and TOPO Ⅱ were detected by Tissue microarray technology and Envision immunohistochemistry method in 107 cases of esophageal carcinoma, including normal esophageal epithelium, esophageal intraepithelial neoplasia and esophageal squamous cell carcinoma. Results The positive expression rates of EGFR and TOPO Ⅱ display an improving trend from normal esophageal epithelium, intraepithelial neoplasia to carcinoma (P =0.031) in the above four groups. The positive expression rates of EGFR were 8.41 %, 7.94 %, 27.27 %, 50.47 %, and TOPO Ⅱ were 3.74 %, 4.76 %, 20.45 %, 43.93 %. Furthermore, the expression showed gradually incresing with histological grades advance (P =0.009). There was no correlation between EGFR or TOPO Ⅱ and gender, age, lymph node or distant metastasis (P >0.05). There was a positive correlation between EGFR and TOPO Ⅱ (r 1=0.410, P 0.05), no correlation was obtained between Her-2 and gender, age, the depth of invasion or lymph node metastasis (P >0.05), no relationship between Her-2 and EGFR or TOPO Ⅱ either. Conclusion EGFR and TOPO Ⅱ are closely related to the occurrence and development of esophageal squamous cell carcinoma, their expressions all make a qualitative change in the esophageal high-grade intraepithelial neoplasia. The role of Her-2 in the development of esophageal squamous cell carcinoma is still not definite in Shanxi province.

ABSTRACT

Objective To investigate the expressions of p16, cyclinD1 protein in esophageal squamous cell carcinoma and its clinic significance. Methods The expressions of p16 and cyclinD1 protein in 55 esophageal carcinoma cases were detected by immunohistochemical technique. Results The positive expression of p16 protein in 55 patients was 49.1%(27/55). The loss of p16 protein was significantly related to lymph node metastasis (P <0.05), and the positive expression of p16 had the higher five-year survival rate.Conversely, the positive expression of cyclinD1 in 55 patients was 74.5 % (41/55), and had the lower five-year survival rate compared to the negative expression cases. Conclusion Loss of p16 protein in human ESCC is a frequent event and correlates significantly with the lymph node metastasis. The loss of p16 and the over expression of cyclinD 1 in esophageal carcinoma may serve as a risk prognosis factors.

ABSTRACT

Objective To analyse the expression of p16INK4A, CyclinD1, Ki-67 proteins and HPV16/18, 31/33 in invasive squamous cell carcinoma of cervix(ISCC)and to indicate the significance of them in the occurrence and development of ISCC. Methods Using tissue microarray,in Situ Hybridization (ISH) and immunohistochemical method detected the expression of HPVI6/18, 31/33 and investigated the expression of p16INK4A, CyclinD1, Ki-67 in ISCC, CIN and NCE. Results was analysed by SPSS vision 13.0. Results The positive expression rate of HPV16/18, CyelinD1, Ki-67 in ISCC was markedly higher than that in CIN and NCE. Significant relationship was observed between p16INK4A protein expression and histological grade of the cancer. No significant correlation was observed between CyclinD1 protein expression and every clinical pathological parameters of ISCC. Conclusion High-risk HPV takes an important role in the development in ISCC. p16INK4A gene might not act as a suppressor-gene only on the occurrence in ISCC. Ki-67 can be looked as a valuable clinic index of diagnosing benign and malignant tumor.

ABSTRACT

Objective To explore expression and clinical value of VEGF-C and p63 in early esophageal carcinoma and intraepithelial neoplasia. Methods 146 cases were randomized into normal esophageal mucosa, low level intraepithelial tumor, high level intraepithelial tumor and early esophageal carcinoma. The expression of VEGF-C and p63 were detected by using the immunohistochemistry dyeing.Results The expression of VEGF-C immunohistochemistry dyeing had statistical differences among different levels(X~2= 47.455, P <0.001). Normal esophageal mucosa v.s. high level intraepithelial tumor (X~2=36.721, P <0.001), Normal esophageal mucosa v.s. early esophageal carcinoma (X~2=26.483, P <0.001), low level intraepithelial tumor v.s. high level intraepithelial tumor(X~2= 10.025, P<0.0083), low level intraepithelial tumor v.s. early esophageal carcinoma(X~2=16.734, P<0.001). There was a significant correlation between pathological classification and the expression amount of VEGF-C (r = 0.462, P <0.001). The expression of p63 had statistical differences among different levels(X~2=28.962, P <0.05). There was a significant difference on normal esophageal mucosa comparing with low level, high level intraepithelial tumor or early esophageal carcinoma (X~2=12.735, P =0.005, X~2=20.421, P<0.001, X~2=20.854, P<0.001). There was a significant correlation between pathological classification and the expression of p63 (r= 0.272, P<0.05). Conclusion There is a significant correlation in the express of either VEGF-C or p63 comparing with either intraepithelial tumor or early esophageal carcinoma. It may be an early warning indicator.

ABSTRACT

Objective To evaluate the expression of the GLUT-1 glucose transporter in borderline and malignant epithelial neoplasms of the ovary. Methods Histologic sections of tumor tissues from 100 epithelial neoplasms of the ovary were stained for GLUT-1 using polyclonal GLUT-1 antibody and the labeled streptavidin procedure. Chi-square analysis was used to assess if their expression was associated with clinicopathoiogic characteristics of the tumors. Results Normal ovarian surface epithelial cell were either negative or weakly positive. The intensity of the stain was significantly stronger in malignant than borderline tumors (P<0.001). In malignant neoplasms, the intensity of the stain for GLUT-1 increased with histology. Conclusion Overexpression of the GLUT-1 transporter is associated with the histology and grade of the minors. These data suggest that the expression of this transporter may be closely related to the malignant transformation of epithelial ovarian tumors.

ABSTRACT

Objective To investigate the relationship and clinicopathologic significance between the expression of CD95,CD44V6 and metastasis of axillary lymph node in breast cancer.Methods Immunohistochemistry was used to detect the expression of CD95 and CD44V6 in 101 cases of breast cancer, in which, 40 cases without metastasis in axillary lymph node were detected MCK expression by IHC. The results were analyzed statistically. Results 9 cases of breast cancer with lymph node micrometastasis were observed by IHC in 40 cases without metastasis in axillary lymph node by microscope. The expression of CD95, CD44V6 in lymph node metastasis group was similar as in lymph node micrometastasis group. There was significant difference of CD95 expression between those with lymph node metastasis and those without. The positive rate of CD95 and the high expression of CD44V6 in the cases that the tumor size was over 2 cm were significandy higher than in the cases that the tumor size was less than 2 cm (P<0.05). Conclusion Detection of the expression of CD95, CD44V6 in breast cancer may be helpful to predict the lymph node micrometastasis and provide more dependable evidence for judging prognosis and selecting treatment prescription clinically.

ABSTRACT

Objective To evaluate the value and safety of transbronchial needle aspiration (TBNA) in the diagnosis of lung cancer. Methods The cytologic diagnosis of TBNA in 82 patients with enlarged hilar and/or mediastinal lymphnodes or lesions adjacent to the bronchial wall were analyzed retrospectively. All specimens were detected by the ThinPrep cytologic test. Results There were 43 positive cases in the 82 patients, and the positive rate was 52.4 %. There were 18 SCLCs,11squamous cancers, 9 adenocarcinomas and 5 undefinable cancers, respectively. There were 39 patients with local bronchial wall swelling accompanied with abnormal mucosae. TBNA, douche, brushing and forcep biopsy were applied, and the diagnostic rate was 64.1%, 7.7 %, 25.6 % and 48.7 %, respectively. The total positive rate was 76.9 %. 43 patients with normal mucous membrane only underwent TBNA. 18 cases were positive, and the positive rate was 41.9 %. There was no obvious complication in the 82 patients. Conclusion The technique of TBNA enlarged the inspection scope of bronchoscopy. It has significant meaning to the diagnosis of lung cancer. TBNA was an useful and safe method in clinical application and could be used widely.

ABSTRACT

Objective To study the valuable index for the diagnosis of cervical minimal deviation adenocarcinoma(MDA).Methods Immunohistochemical EnVisionTM two-step method was used to evaluate the expression of CEA,Ki-67,p16,MUC6,MUC5AC,ER,PR,α-SMA in 9 samples of MDA.12 normal cervical gland tissues and 21 other types of cervical adenocarcinomas were as controls. Results The expression of CEA,MUC6,α-SMA protein in MDA(positive ratio orderly 66.67%,77.78%,88.89%)was higher than that of normal cervical gland(0,8.33%).The average Ki-67 labeling index in MDA(21.03±5.46)was also higher than that of the normal group(0.12±0.08).But as to the expression of ER and PR,the former was lower than the latter.All the differences had statistical significance (P<0.05).The expression of p16,MUC5AC had no statistical significance between the two groups(P>0.1).The ratio of positive expression of MUC6 protein in MDA(77.78%)was higher than that of the other types of adenocarcinomas(14.29%),with the expression of p16(11.11%)lower than the latter(71.43%).All the differences had statistical significance(P<0.01).The expression of CEA,Ki-67,MUC5AC,ER,PR,α-SMA had no statistical significance between the two groups(P>0.05).Conclusion Immunohistochemical positive detection of CEA,Ki-67,MUC6,α-SMA perhaps make for the diagnosis of cervical MDA.The absence of ER,PR of gland cells also support MDA.Overexpression of p16 may play an impo~ant role on the occurrence and the development of some usual adenocarcinoma of cervix,but perhaps not in MDA.

ABSTRACT

Objecfive Most prostate cancer contains two or more widely separate turnors.To study the origin of prostate cancer based on the analysis of microsatellite alteration in separate tumors from the same prostate.Methods A polymerase chain reaction (PCR) was used to examine the allelic loss pattern of 4 microsatellite polymorphic markers on chromosome 8p (D8S133,D8S136,D8S137) and 17q (D17S855) in multifocal tumors of prostate from 19 patients.DNA samples were obtained from different regions of distinctly separate tumors on single case using microdissection technique.Results The overall frequence of LOH at D8S133,D8S136,D8S137 and D17S855 for all informative cases was 74%,38%,86% and 46%respectively in 40 separate tumors of prostate from 19 patients.The pattern of allelic loss was not identical in 15 of 18 (83%) informative cases. It showed that the different regions of prostate cancer were independent origin respectively.Discordant pattern of histology was observe in distantly separate regions.whereas the same allele was consistently lost in samples from different regions of the same tumor in 3 cases. Condusion Current data supports independent origin of multiple tumors in most prostate cancer patients.

Objective To examine the expression of COX-2, PCNA, CD44v6 protein in different procancerous lesion and esophageal squamous cell carcinoma tissue, and to investigate the relationship between their expression and the biological level,lymph node metastasis in esophageal squamous cell carcinoma tissue. Methods Immunohistochemistry was demonstrated to show these tumor markers expression in different procancerous lesions and esophageal squamous cell carcinoma tissue. Results Stepwise over-expression of COX-2, CD44v6, PCNA protein were observed in the multi-step process of esophageal tissue. Significant difference in expression of these tumor markers(P <0.05) was observed between normal esophageal epithelia and dysplasia. The expression of COX-2 (P <0.05) had statistical different in biological level and lymph node metastasis.The expression of CD44v6 (P <0.05)had statistical different in lymph node metastasis. The relationship between COX-2 and CD44v6 was statistical significant in esophageal squamous cell carcinoma tissue(P <0.01). Conclusion Stepwise over-expression of COX-2,CD44v6 PCNA protein have relation with esophageal squamous cell carcinoma. The overexpression of COX-2 and CD44v6 have effect on prognosis of esophageal squamous cell carcinoma. The expression of COX-2 have relationship with that of CD44v6 in esophageal squamous cell carcinoma.

Objective Through comparision of HER2/neu status detected by immunohistoehemistry (IHC),fluorescence insitu hybridization(FISH)and chromogenic insitu hybridization(CISH),to explore the clinical significance of FISH and CISH on detecting gene status of HER2.Methods Sixty-four paraffinembedded breast samples whose pathological types were infiltrating ductal carcinomas were tested by IHC for HER2 protein overexpression and CISH,FISH for HER2 gene status.The rate of chromosome 17 polysomy was also analyzed.Results Among the cases of HER2 protein overexpression(+++)group,the concordant ratios of IHC and CISH.FISH detecting HER2/neu status were 100%.Among the cases of HER2 protein overexpression(++)group,the concordant ratios were 95.83%and 91.67%.Among the cases of HER2 protein overexpression(+/-)group,the concordant ratios were also 100%.One case in which FISH and CISH were performed yielded the different results.so the general coincident rate was 98.41%.The incidence of chromosome 17 polysomy Was 45.16%,45.83%and 11.11%in three IHC(+++,++,+/-)groups,and the total incidence was 40.62%.ConclusionIHC is a preliminary screening method;HER2 gene status detected by CISH and FISH have high concordant ratio,CISH is a more convenience and viable method to detect HER2/neu status,when doubling interference of fichromosome 17 polysomy,we need to select FISH further.

ABSTRACT

Objective To investigate the regulation of gene variation and explore the tumor associated suppressor genes on chromosome 9 during the developing of cervical squamous cell carcinoma. Methods Microdissection, PCR, electrophoresis and radiograph were selected to detect the LOH in 46 cases cervical squamous cell carcinoma with high-grade squamous dysplasia and normal tissue. The changes of LOH at chromosome 9 total seven microsatellite markers and relationship between LOH rate and clinical parameters were analysed. Results Total frequency of LOH in CIN (Ⅱ,Ⅲ) was 16 % and in cervical squamous cell carcinoma was 25 %. In the former, LOH at marker D9S171(30 %), D9S162(23 %), D9S43(20 %), D9S303(17 %), D9S753(12 %), D9S242(11 %) were found, whereas D9S1748 LOH was not detected in high-grade dysplasia. In the latter, LOH at marker D9S171(41 %), D9S43(33 %), D9S162(31 %), D9S242(24 %), D9S303(17 %), D9S753(17 %), D9S1748 13 % were found. In addition, LOH was found in high-grade dysplasia in three cases but not in cervical squamous cell carcinoma. Conclusions The study in seven microsatellite markers showed that from normal squamous tissue to dysplasia to cervical squamous cell carcinoma were accompanied with accumulation of gene errors. The p15 gene inactivation had a high relationship with the occur of cervical squamous cell carcinoma. Tumor suppressor genes associated with cervical squamous cell carcinoma may exist near or at D9S43, D9S162, D9S242. Some cases showed that high-grade dysplasia and cervical squamous cell carcinoma may came from different independent clone which provide some clue for multiple foci carcinoma at genetic level.

Objective To investigate the relationship between COX-2 and Ki-67 proliferation index, bax, microvascular density(MVD)and CD44v6 respectively. Methods The expression of COX-2,Ki-67, bax, MVD (labelled by CD34) and CD44v6 were examined by immunohistochemistry PV method and tissue chip method in 101 cases of cervical carcinomas epithelium. Results The expression of COX-2 in cervical carcinomas has positive correlation with Ki-67, MVD and CD44v6, but has no correlation with bax. Conclusion The probable mechanisms are related with neovascularization, cell proliferation and cell adherence, but not related with the expression of bax.

ABSTRACT

Objective To investigate expression of CD56, CD95(Fas), Ki-67, p53, bcl-2, HMB45 and S-100 in mucosa amelanotic malignant melanoma in order to improve the pathological diagnosis level reduce wrong diagnosis and avoid missing dignosis, and afford objective factors for prognosis and therapy. Methods The techniques of tissues chips and immunohistochemical lablling were used for analyzing 48 cases of mucosa amelanotic malignant melanoma. Results The positive rates of HMB45 and S-100 were 100 % (48/48) and 85.4 % (41/48) respectively. The positive rate of CD56 was 91.6 % (44/48), there was not statistical difference between original cases and metastatic cases. The positive rate of CD95 was 85.4 %(41/48). In which it is 100 % (11) in 11 cases of having lymphanoid metastasis. The positive rates of Ki-67 and p53 were 79.2 % (38/48) and 58.3 % (28/48) respectively. ABSTRACT

Objective To evaluate the relationship between the expressions CyclinD1 and nm23H1 and the response to neoadjuvant chemotherapy (NACT) in cervical carcinoma. Methods 100 patients with cervical cancer were enrolled in this study. These patient were randomly divided into two groups. The observed group were treated by neoadjuvant chemotherapy followed by radical hyterectomy and lymphonectomy. The other 50 cases were treated by operation as control group. The expression of CyclinD1 and nm23H1 were examined by immunohistochemistry using post-operation specimens. Results The total response rate of neoadjuvant chemotherapy was 75 %( CR+PR). The expression rate of CyclinD1 was 76 % in experimental group, and 54 % in control group(P 0.05). Conclusion The neoadjuvant chemotherapy was helpful for the patients with local advanced stage of cervical carcinoma. CyclinD1 may be one of the path of NACT, but nm23H1 may not related to NACT.
