ABSTRACT

Lamin B1 (LMNB1) is highly expressed in liver cancer tissues, and its influence and mechanism on the proliferation of hepatocellular carcinoma cells were explored by knocking down the expression of the protein. In liver cancer cells, siRNAs were used to knock down LMNB1. Knockdown effects were detected by Western blotting. Changes in telomerase activity were detected by telomeric repeat amplification protocol assay (TRAP) experiments. Telomere length changes were detected by quantitative real-time polymerase chain reaction (qPCR). CCK8, cloning formation, transwell and wound healing were performed to detect changes in its growth, invasion and migration capabilities. The lentiviral system was used to construct HepG2 cells that steadily knocked down LMNB1. Then the changes of telomere length and telomerase activity were detected, and the cell aging status was detected by SA-β-gal senescence staining. The effects of tumorigenesis were detected by nude mouse subcutaneous tumorigenesis experiments, subsequent histification staining of tumors, SA-β-gal senescence staining, fluorescence in situ hybridization (FISH) for telomere analysis and other experiments. Finally, the method of biogenesis analysis was used to find the expression of LMNB1 in clinical liver cancer tissues, and its relationship with clinical stages and patient survival. Knockdown of LMNB1 in HepG2 and Hep3B cells significantly reduced telomerase activity, cell proliferation, migration and invasion abilities. Experiments in cells and tumor formation in nude mice had demonstrated that stable knockdown of LMNB1 reduced telomerase activity, shortened telomere length, senesced cells, reduced cell tumorigenicity and KI-67 expression. Bioinformatics analysis showed that LMNB1 was highly expressed in liver cancer tissues and correlated with tumor stage and patient survival. In conclusion, LMNB1 is overexpressed in liver cancer cells, and it is expected to become an indicator for evaluating the clinical prognosis of liver cancer patients and a target for precise treatment.

ABSTRACT

Objective To study and correlate serum bilirubin and regulatory T cell (Treg) levels in patients with bile duct stone.Methods Flow-cytometry and Enzyme-linked immunosorbent assay (Elisa) were used to study the peripheral blood expression level of Tregs and the bilirubin level in 27 patients with bile duct stones and jaundice.The changes in the expression level of Tregs and the bilirubin level were studied and correlated before and after treatment.Results After treatment,both the peripheral blood bilirubin level,the Tregs expression level and the cell cytokines decreased significantly.The total bilirubin level decreased from (102.8 ± 33.1) mmol/L to (15.3 ± 5.7) mmol/L (P < 0.05),the direct bilirubin level decreased from (38.1 ± 12.8) mmol/L to (5.0 ± 1.6) mmol/L (P <0.05);the percentages of CD4+ CD25 +Foxp3 + T cells in CD4+ T decreased from (4.2 ± 2.0) % to (2.4 ± 1.0) % (P < 0.05).Before treatment,the levels of IL-10 and TGF-β were 171.4 ± 13.7 and 2016 ±657 pg/ml but after treatment,the two cytokines decreased to 92.1 ± 7.4 and 1 686 ± 168 pg/ml,respectively (P < 0.05).Conclusions Patients with bile duct stones and jaundice presented with high expressions of bilirubin and Tregs level.These expressions returned to normal after effective treatment.The Tregs expression level was positively correlated with the bilirubin level.

ABSTRACT

Objective To investigate the effects and safety of needle?knife precut papillotomy in diffi?cult common bile duct cannulation of endoscopic retrograde pancreatic duct radiography ( ERCP ) . Methods The data of 104 cases treated with needle?knife precut papillotomy were retrospectively analyzed. The effect of pre incision and short?term complications were observed,and compared with 1326 cases( conventional group) of con?ventional selective intubation in the same period of ERCP. Results Successful cannulation of the bile duct was achieved in 97 of 104 patients who underwent needle?knife precut papillotomy, the total success rate was a?chieved in 93. 3%. The complications of needle?knife precut papillotomy occurred in 8 patients(7. 7%),inclu?ding mild bleeding in 4 cases,mild pancreatitis in 2 cases and billiary infection in 2 cases. There were 1326 ca?ses conventional selective intubation in the same period of ERCP,and 98 cases(7. 4%) with complication. There was no significant difference in the incidence of complications between the needle knife cut group and the con?ventional selective intubation group(χ2=0. 055,P>0. 05 ) . Conclusion In case of difficult common bile duct cannulation of ERCP,needle?knife precut sphincterotomy surgery can be safely and effectively catch the high ?ERCP the success of treatment,is worthy of wider application.

ABSTRACT

Objective To investigate the short-term outcome of laparoscopic adjustable gastric banding (LAGB) for morbid obesity complicated with type 2 diabetes. Methods Eight morbidly obese patients with type 2 diabetes underwent LAGB from October 2006 to August 2007. The weight parameters, fasting (FBG) and 2-hour blood glucose (2hBG), medication for diabetes were assessed 1,3, 6 and 9 months after surgery. Results All of the patients lost weight, with a mean body mass index decreased from (38.7±7.5) kg/m2 before LAGB to (30.5±4.3) kg/m2 9 months after LAGB. The FBG and 2hBG were decreased significantly at month 6 and 9 after LAGB, with normal FBG and 2hBG in 4 patients. At month 9 after LAGB, 3 of 5 patients with insulin treatment before LAGB were changed to oral hypoglycemics, 1 was continuously administered with a reduced dose of insulin, and 4 patients stopped any medication. Conclusion LAGB is an effective procedure in the treatment of morbid obesity complicated with type 2 diabetes with a favorable short-term outcome.

BACKGROUND: The biomechanical features of Swan-like memory compression connector designed for fractures in hemerus or the parts adjacent to joints are analyzed through biomechanical calculation and experimental measurements.OBJECTIVE: To analyze the relative memory biomechanieal significance through biomechanical measurement of Swan-like memory compression connectorDESIGN: Both biomechanieal calculation and the experimental measurement were conducted for mutual supplementation and confirmation SETTING: The Orthopaedic Department of Changhai Hospital Affiliated to Second Military Medical University of Chinese PLA and the Life Science and Tissue Engineering Institute of Tongji University MATERIALS: This experiment was conducted at the laboratory of Orthopaedic Department of Changhai Hospital Affiliated to Second Military Medical University of Chinese PLA and the Life Science and Tissue Engineering Institute of Tongji University between January 2001 and May 2003.Totally 20 hemerus from adult males who died of craniocerebral injury were chosen.METHODS: Humid hemerus from the adult corpse was chosen to prepare fracture. Encircling the prescale followed by fixation of Swan-like memory compression connector, then we measured the stress produced by compression part and encircle part at fixing the fracture.MAIN OUTCOME MEASURES: ①The stress of holding part; ② the stress of the compression part RESULTS: The stress range of holding part contacting with humerus was 2.42-22.68 N, and the stress in the fracture face, which was produced by compression part, was about 13.6 Mpa.CONCLUSION: The stress of holding part of Swan-like memory compression connector is useful in fixing the fracture parts of humerus, and the stress of compression part is suitable for the healing of fracture.

BACKGROUND: During the therapy of the fracture of humerus, the main problem is the loading direction and power of the stress.OBJECTIVE: To analyze the choice of the different loading manners using Swain Memory Compression Bone Reduction Apparatus so as to provide the clinical evidence for the set position of the internal fixation apparatus, the loading direction and power during the therapy of the fracture of humerus.DESIGN: To construct the three-dimensional finite element model of the fracture of humerus with different loading manners.SETTING: Department of Orthopaedics of Changhai Hospital of Second Military Medical University of Chinese PLA; College of Life Sciences and Bioengineering of Tongji University,MATERIALS: The study was conducted during the period from January 2001 to May 2003 in the Orthopedic Laboratory of Changhai Hospital of Second Military Medical University of Chinese PLA and the laboratory of Life Science and Bioengineering Academy of Tongji University. The samples of the humerus taken from the wet adult cadavers, and the Swain Memory Compression Bone Reduction Apparatuses of the corresponding size were prepared.METHODS: The CT scanning was done on the samples of the wet humerus to obtain the cross section image of each layer of the humerus.The images were analyzed by a large-scale finite element analysis software ANSYSS.6 to construct the three-dimensional model of the humerus, Swain Memory Bone Reduction Apparatus and the fixed humerus using Swain Memory Bone Reduction Apparatus.MAIN OUTCOME MEASURES: To get the reference values for the clinical therapy through the comparison of the power endurance with different loading manners.RESULTS: The three-dimensional model of the fixed humerus using Swain Memory Bone Reduction Apparatus reflects the real anatomical configuration and the biomechanics behavior; meanwhile the status of the power endurance of humerus with different loading manners is also obtained.CONCLUSION: The three-dimensional model of the fixed humerus using Swain Memory Bone Reduction Apparatus may provide an accurate model for the basic study of the biomechanics behavior of the normal and fixed humerus after the fracture.
