Colorectal cancer is a common malignant tumor in gastrointestinal tract. Its onset and development are associated with its own characteristics as well as the tumor microenvironment (TME) in which tumor-associated macrophages (TAMs) are the most abundant immune cells. After being recruited to the tumor site and stimulated by different signals in TME, TAMs can grow into two different subtypes, namely M1 and M2. TAMs are mainly manifested as M1 macrophages in the early stage of colorectal cancer, mediating the immune response to inhibit tumor growth. In the late stage, TAMs mainly grow into M2 macrophages, showing the ability to suppress immunity, stimulate the proliferation of tumor cells and tumor angiogenesis, and promote the invasion and metastasis of tumor cells. It has been found that intervention in TAMs polarization can regulate its relationship with the onset and development of colorectal cancer.

Objective:To study the effects of Zuogui Pills on the expressions of miR-133b-3p and RhoA in osteoclasts of postmenopausal osteoporosis rats; To discuss its potential mechanism.Methods:SD female rats were randomly divided into normal group, model group, sham-operation group, and Zuogui Pills group using a random number table method, with 6 rats in each group. The model group and Zuogui Pills group were treated with oophorectomy to construct a rat model of osteoporosis. Zuogui Pills group was orally administered with Zuogui Pills decoction at a concentration of 10 g/kg for 12 consecutive weeks. Colorimetric method was used to measure the serum calcium and phosphorus levels of rats, and ELISA method was used to detect ALP levels. Bone density meter was used to measure the bone density of the femurs of rats in each group. The osteoclast of each group were cultured, and the expressions of RANKL and RUNX2 protein were detected by Western blot. MiRNA sequencing and differential expression analysis were performed on bone tissues of rats. Osteoclasts were treated with miR-133b-3p mimic and its negative control. The cell proliferation activity of osteoclasts was detected by cell counting kit-8 (CCK-8). The osteoclast differentiation activity was detected by the tartrate-resistant acid phosphatase staining. The dual-luciferase reporter assay was used to detect the relationship between miR-133b-3p and RhoA. The "rescue" experiment of miR-133b-3p mimic and RhoA co-expression were used to study the molecular regulatory mechanism of Zuogui Pills on osteoclast activity.Results:Compared with the model group, the bone mineral density of Zuogui Pills group significantly increased ( P<0.05, P<0.01), the levels of calcium and phosphorus in serum increased, the level of alkaline phosphatase ALP decreased ( P<0.05), the expression of RANKL protein decreased, and the expression of RUNX2 protein increased. Sequencing results showed that rno-miR-133b-3p was down-regulated in osteoclasts of postmenopausa osteoporosis rats treated with Zuogui Pills with the maximum difference ( P<0.01). Q-PCR results showed that the expression of miR-133b-3p in osteoclasts of Zuogui Pills group was significantly lower than that of the model group. The upregulation of miR-133b-3p could significantly promote the cell proliferation and differentiation of osteoclasts. RhoA overexpression could reverse the excessive proliferation and differentiation of osteoclasts caused by miR-133b-3p overexpression. Conclusions:RhoA is the target gene regulated by miR-133b-3p. Zuogui Pills can inhibit the activity of osteoclasts by regulating miR-133b-3p/RhoA axis, relieving the symptoms of osteoporosis.

OBJECTIVE:To pre pare Glycyrrhetinic acid-modified docetaxel magnetic nanoparticles (GA-DTX-NGO/IONP- NPs),and to evaluate its physicochemical properties. METHODS :Magnetic nano graphene oxide (NGO/IONP)was chosen as the anti-tumor drug carrier ,docetaxel(DTX)as the model drug and glycyrrhetinic acid (GA)as the target molecule. Firstly ,NGO/ IONP was synthesized by hydrothermal method and GA-CS was synthesized by amidation reaction. Fourier IR spectrometer ,DSC and vibration sample magnetic measuring instrument were used to characterize NGO/IONP and GA-CS. GA-DTX-NGO/IONP-NPs Huperzine A in the nicotinic acetylcholine receptor alleviates Aβ -induced 1-42 treatment of Alzheimer ’s disease and vascular dementia :a neurotoxicity via downregulation of p 38 and JNK MAPK meta-analysis[J]. Evid Based Complement Alternat Med , signaling pathways[J]. Neurochem Int ,2018. DOI :10. 2014. DOI :10.1155/2014/363985. 1016/j.neuint.2018.09.005. were prepared by the ion gelation method. TEM and particle size analyzer were used to observe and determine the morphology , particle size and Zeta potential of GA-DTX-NGO/IONP-NPs ;the ultrafiltration-centrifugation method was used to determine encapsulation efficiency and drug loading amount ;the magnetic properties were investigated by investigating the state with or without external magnetic field ;the photothermal conversion test was carried out with laser irradiation of 808 nm. RESULTS :NGO/ IONP and GA-CS were successfully synthesized ,and NGO/IONP exhibited superparamagnetism characteristics. GA-DTX-NGO/ IONP-NPs were spherical under TEM ,the particle size was (262.8±4.23)nm and the Zeta potential was (13.6±1.51)mV. The encapsulation rate and drug loading amount were (94.29±0.50)% and(17.12±0.12)%,respectively. GA-DTX-NGO/IONP-NPs were black in appearance and evenly dispersed. Under the external magnetic field ,the magnetic nanoparticles could move directionally,showing good magnetic properties. GA-DTX-NGO/IONP-NPs showed a good concentration- and time-dependent photothermal conversion effect under 808 nm laser irradiation. CONCLUSIONS :GA-DTX-NGO/IONP-NPs are successfully prepared. This study could provide some theoretical basis for the combined treatment of magnetic heating-chemotherapy for liver tumors.

OBJECTIVE:To summarize the role of clinical pharmacists on the prevention of medicine-induced epilepsy and common epilepsy-inducing drugs in clinic. METHODS:The role of clinical pharmacists in whole process of pharmaceutical care for an epilepsy patient was introduced,and clinical pharmacists assisted physicians to reduce the risk of drug-induced epilepsy. The role of clinical pharmacists on the prevention of epilepsy-inducing medicine during pharmaceutical care was explored through reviewing literature,summarizing epilepsy-inducing drug and analyzing mechanism. RESULTS:The role of clinical pharmacists in diagnosis and treatment of epilepsy included that analyzing the risk of medicine-induced epilepsy by inquiring about the history of drug use, guiding patients to gradually stop taking risk drugs,participating in the formulation of clinical medication plan and drug risk during diagnosis and treatment,assisting physicians to adjust medication plan,monitoring the change of patient's symptoms after drug,"special medication notification for epilepsy patient",etc. The types of common epilepsy-inducing medicine included antibiotics (fluoroquinolone,β-lactam,etc.),drugs for central nervous system(sedative and antiepileptic drug,mental disorders drug,etc.), Chinese herbal drug,Chinese patent drug,antipyretic- analgesic and anti-inflammatory drug,circulatory system drug and other drugs. The mechanisms of drug effect included entering the brain through the blood cerebrospinal fluid barrier,inhibiting the binding of γ-aminobutyric acid to the receptor site,producing amino or free radicals and affecting a part of the cerebral cortex. CONCLUSIONS:Clinical pharmacists play an irreplaceable role in the safety of drug use in epilepsy patient. Their focuses to epilepsy-inducing drug are of significance to clinical practice.

Hyperuricemia is closely associated with abdominal obesity .The prevalence of hyperuricemia combined with abdominal obesity has been increased significantly in recent years , along with the improvement of daily life and the changes in dietary structure .The state of hyperuricemia combined with abdominal obesity is most harmful , and becomes a common and high risk metabolic disease .Animal model with hyperuricemia combined with abdominal obesity is very impor-tant for the research of pathomechanism and treatment of this disease .

Objective Deletion detection of Duchenne muscular dystrophy(DMD).Methods Totally 135 patients were tested through polymerase reaction of amplification with 12 dystrophin exons,polyacrylamide gel electrophoresis make gene analysis.Results The deletion of different region was found in 54 patients.Conclusion The deletion region is concentrated in 45~53 exons,the deletion of 48 exons is the peak.
