ABSTRACT

OBJECTIVE To establis h the fingerprint of Cynanchum auriculatum,to conduct its chemical pattern recognition analysis, and to determine the contents of four components at the same time. METHODS High performance liquid chromatography (HPLC)method was adopted. The determination was performed on ACE UExcel C 18 column with mobile phase consisted of acetonitrile-0.1% phosphoric acid solution (gradient elution )at the flow rate of 1.2 mL/min. The determination wavelength was set at 210 nm,and the column temperature was 40 ℃. The sample size wa s 10 μL. Taking qingyang shengenin as the reference ,HPLC fingerprints of 16 batches of C. auriculatum medicinal materials were drawn and similarity was evaluated by using the Similarity Evaluation of Chromatographic Fingerprints of Traditional Chinese Medicine (2012 edition), and the common peaks were determined. SPSS 26.0 software andSIMCA 14.0 software were used for cluster analysis ,principal component analysis and orthogonal partial least squares- discriminant analysis. The differential components affecting the quality of C. auriculatum were screened by taking the value of variable importance in projection (VIP)greater than 1 as the standard ;same HPLC method was used to determine the contents of syringic acid ,acyl asclepiadelenin ,baishouwubenzophenone and qingyang shengenin. RESULTS There were 29 common peaks in 16 batches of C. auriculatum ,with a similarity of 0.723-0.998. Four common peaks were identified ,namely syringic acid (peak 7),acyl asclepiadoidin (peak 9),baishouwubenzophenone(peak 13)and qingyang shengenin(peak 15). The results of cluster analysis showed that 16 batches of C. auriculatum could be clustered into three categories ,among which S 1 were grouped into one category,S3 were grouped into one c ategory,S2,and S 4-S16 were grouped into one category. The results of principal component analysis showed that the cumulative variance contribution rate of the five principal components was 88.706%,and the classification results were consistent with the results of cluster analysis. The results of orthogonal partial least squares-discriminant analysis showed that the common peaks (from large to small )with VIP value greater than 1 were peak 20,peak 10,peak 25,peak 12, peak 15(qingyangshengenin),peak 21,peak 14,peak 16,peak 26,peak 22 and peak 17. The linear ranges of syringic acid, acyl asclepterin, baishouwubenzophenone and qingyangshengenin were 0.715 3-45.778 0,2.379 4-152.281 0,0.642 0- 41.085 0,14.541 6- 930.662 0 µg/mL respectively (all R 2>0.999). The quantitative limits were 0.357 7,0.475 9,0.642 0 and 2.423 6 μg/mL;the detective limits were 0.146 0,0.164 1,0.248 8 and 0.833 3 μg/mL,respectively. RSDs of precision ,repeatability and stability (24 h)tests were less than 3%;the average recoveries were 99.11%(RSD=2.00%,n=9),98.54%(RSD=2.21%,n=9), 96.33%(RSD=2.54%,n=9)and 95.96%(RSD=2.93%,n=9);the contents were 17.12-147.80,95.23-583.10(S8 below the quantitative limit ),16.91-210.88 and 211.68-3 587.15(S1 below the quantitative limit )μg/g,respectively. CONCLUSIONS Established HPLC fingerprint and the method of content determination are stable ,reliable, accurate and reproducible. Combined with analysis of chemical pattern recognition ,it can be used for the quality control of C. auriculatum .

ABSTRACT

Objective:To investigate the effect of combining aerobic exercise with hyperbaric oxygen therapy on the cognition and oxidative stress of patients with post-stroke cognitive impairment (PSCI).Methods:Forty-five PSCI patients were divided randomly into a hyperbaric oxygen therapy (HBO) group (of 21) and a combination group (of 24). Both groups received HBO therapy, but the combination group also performed 30min of aerobic exercise using a cycle ergometer 5 times a week for 8 weeks. The HBO therapy involved 20min for pressurization, 20min of decompression, 60min of pressurized oxygen inhalation and 10min of air inhalation. The treatment pressure was 0.2MPa (2.0ATA). Before and after the intervention, cognitive functioning was measured using the mini-mental state examination (MMSE) and the Montreal cognitive assement (MoCA). The Barthel index quantified ability in the activities of daily living. Serum glutathione (GSH) and malondialdehyde (MDA) were measured spectrophotometrically. Results:The average MMSE scores of both groups had improved significantly after the treatment, with significantly better improvement in the combination group. After the treatment, the average MoCA score of the combination group had improved significantly while that of the HBO group had not. The average Barthel index scores of both groups improved significantly and there was no significant difference between them after the treatment. The average serum GSH increased after the treatment in the combination group, and the increase was weakly positively correlated with the increase in MMSE scores. The average MDA content of the HBO group decreased after the treatment, and the decrease was positively correlated to the increase in the MMSE scores, but again weakly. Conclusions:Aerobic exercise combined with hyperbaric oxygen therapy can effectively improve cognition among PSCI patients. Increases in serum GSH and decreases in serum MDA are weakly positively correlated with cognitive functioning.

ABSTRACT

Objective:To explore the correlation analysis between psychological status and blood pressure of residents in medium-risk areas during the coronavirus disease 2019 (COVID-19) epidemic, and provide references for early psychological intervention and hypertension management during the COVID-19 epidemic. Methods:From January 4 to 8, 2021, the convenience sampling method was used to conduct psychological questionnaire surveys on residents and employees of enterprises in the medium-risk area and surrounding areas of Jiuxianqiao, Chaoyang District, Beijing. General data (gender, age, height, weight, history of smoking and drinking, previous medical history), and home blood pressure monitoring were collected. Pearson′s χ 2 test was used to compare different factors (anxiety, depression, age, gender, body mass index, smoking, drinking, and history of chronic diseases) in normal blood pressure or not. A multivariate logistic regression analysis model was established to explore independent risk factors of abnormal blood pressure. Results:No anxiety accounted for 75% (684/912), and anxiety accounted for 25% (228/912). No depression accounted for 67% (611/912), depression accounted for 33% (301/912). Both anxiety and depression accounted for 23% (210/912). The substandard blood pressure accounted for 10.3% (94/912). The incidence of poor blood pressure control in participants with anxiety was 30.7% (70/228), which was significantly higher than that of participants without anxiety 3.5% (24/684) (χ 2=136.779, P<0.001). The incidence of poor blood pressure control in participants with depression 27.2% (82/301) was significantly higher than that of participants without depression 2.0% (12/611) (χ 2=139.388, P<0.001). Multivariate logistic regression analysis showed anxiety ( OR=3.261, P<0.001), depression ( OR=7.928, P<0.001), and complications of chronic diseases ( OR=3.207, P<0.001) were closely related to whether blood pressure was under control. Conclusions:During the COVID-19 epidemic, the psychological status of residents and employees in medium-risk areas should be screened and interventions implemented accordingly to prevent unstable blood pressure caused by anxiety or depression.

ABSTRACT

Objective:To improve the diagnosis of anti-synthetase antibody syndrome(ASS) by analyzing the clinical features of 6 patients. Methods:Six cases of ASS with complete data were included in this study as they were diagnosed or other CTD during the period of hospitalization in Wuxi People's Hospital from January 2016 to February 2020. Their clinical and laboratory characteristics, and follow-up information were analyzed. Features and changes in the course of disease were analyzed. Results:Four out of 6 patients were females, with age of disease onset as 34-72 years, and an interval of 4-59 months from the first diagnosis to the diagnosis of ASS. The first diagnosis was Sjogren's syndrome (SS) in 2 cases, rheumatoid arthritis (RA) in 1 case, mixed connective tissue disease (MCTD) in 1 case, systemic sclerosis (SSc) in 1 case, and undifferentiated connective tissue disease (UCTD) in 1 case. At the first diagnosis, 5 cases had dry cough and/or dyspnea, followed by fever (4 cases), arthritis and Raynaud's phenomenon (3 cases). Anti-nuclear antibody(ANA), which was more common in cytoplasmic type, and anti-SSA/52 000 antibody were mostly positive(5 cases). The presence of non-specific interstitial pneumonia (NSIP) pattern (6 cases) in high resolution CT(HRCT) was found at the initial diagnosis. During follow-up, patients developed repeated liver function or muscle enzyme abnor-malities (3 cases), mechanic's hand (MH) (3 cases), and lung interstitial disease progression (4 cases). The myositis antibodies were found to be positive. Conclusion:ASS can occur in the course of or at the same time as other CTDs. ASS should be considered in patients with interstitiallung disease (ILD) (especially NSIP pattern) in HRCT, and/or positive of cytoplasmic type ANA and/or anti-SSA/52 000 antibodies At the same time, if repeated liver function or muscle enzyme abnormalities, new onset of MH, and lung interstitial disease progresses during treatment, consideration may be given to the possibility of complicated ASS. Myositis-specific antibodies are helpful in the diagnosis of ASS.

ABSTRACT

Objective:To investigate the effect of fine anatomy combined with exposure of recurrent laryngeal nerve in cricothyroid space in differentiated thyroid cancer surgery. Methods:Sixty patients with differentiated thyroid cancer who received treatment in Ningbo Hospital of Traditional Chinese Medicine from January 2017 to April 2020 were included in this study. They were randomly assigned to receive either fine anatomy combined with exposure of recurrent laryngeal nerve in cricothyroid space (observation group, n = 30) or routine surgery + inferior thyroid artery exposure of recurrent laryngeal nerve (control group, n = 30). Operative time, the volume of intraoperative blood loss, and the number of lymph node dissected were compared between the two groups. Parathyroid hormone (PTH) and blood calcium (Ca 2+) concentrations in the two groups were determined. The adverse reactions such as recurrent laryngeal nerve and parathyroid injury were observed in each group. Results:Operative time, the volume of intraoperative blood loss, and the number of lymph node dissected in the observation group were (71.28 ± 10.08) minutes, (16.69 ± 4.65) mL, and (4.78 ± 1.26), respectively, which were significantly different from those in the control group [(69.27 ± 11.89) minutes, (17.14 ± 4.52) mL, (4.76 ± 1.22), t = 0.706, 0.380, 0.062, all P > 0.05]. Before surgery, there were no significant differences in PTH and Ca 2+ concentrations between the two groups [PTH: (50.24 ± 11.21) g/L vs. (50.21 ± 11.19) g/L; Ca 2+: (2.18 ± 0.08) mmol/L vs. (2.17 ± 0.09) mmol/L, t = 0.010, 0.454, both P > 0.05]. After surgery, PTH and Ca 2+ concentrations in the observation group were significantly higher than those in the control group [PTH: [(31.44 ± 6.09) g/L vs. (16.57 ± 2.35) g/L; Ca 2+: (2.07 ± 0.10) mmol/L vs. (1.23 ± 0.12) mmol/L, t = 12.477, 29.454, both P < 0.01]. Total incidence of adverse reactions in the observation group was significantly lower than that in the control group [10.00% (3/30) vs. 46.67% (14/30), χ2 = 9.931, P < 0.01). Conclusion:Application of fine anatomy combined with exposure of recurrent laryngeal nerve in cricothyroid space in differentiated thyroid cancer surgery is highly effective in the clinic. Accurate identification of recurrent laryngeal nerve in cricothyroid space is helpful to preserve the parathyroid gland and blood supply in situ, leads to a small range of changes in PTH and Ca 2+ concentrations, reduces postoperative complications, so as to protect the recurrent laryngeal nerve and parathyroid gland and thereby promote postoperative recovery.

ABSTRACT

Objective:To investigate the effects of artesunate (ART) on epithelial-mesenchymal transformation (EMT) of colorectal cancer HCT-8 cells, and explore the effects of ART on cell migration, invasion,EMT ability, and protein kinase B (Akt)/Snail signaling pathway of colorectal cancer. Method:3-(4-5-Dimethylthiazol-2-yl)2,5-diphenyltetrazolium bromide (MTT) assay was used to detect the effects of ART at different concentrations on the proliferation of HCT-8 cells. Wound healing assay and Transwell assay were used respectively to detect the effects of ART on migration and invasion of colorectal cancer cells. The effects of different concentrations of ART on the distribution of EMT-related proteins vimentin and E-cadherin in HCT-8 cells were detected by double-immunofluorescent staining. The effects of ART on protein expression levels of EMT markers E-cadherin,vimentin and N-cadherin in HCT-8 cells and the expression of Akt1, p-Akt1, and Snail1 in the Akt/Snail signaling pathway were determined by Western blot. Result:The dose-dependent inhibitory effects of ART on the proliferation of HCT-8 cells were determined and the inhibition rate was calculated. A dose-response curve was plotted accordingly. The half-maximal inhibitory concentration (IC50) of ART on HCT-8 cells was (16.67±1.95) μmol·L-1. The following four groups were set up: a control group (0 μmol·L-1),and low-, medium-, and high-dose ART groups(2, 10, 50 μmol·L-1). Compared with the results in the control group,ART inhibited the migration and invasion of HCT-8 cells(P<0.05). Specifically, the expression of E-cadherin in HCT-8 cells was significantly up-regulated,and that of vimentin and N-cadherin was significantly down-regulated (P<0.05). The expression levels of p-Akt1 and Snail1 were significantly decreased after ART treatment, thus inhibiting EMT(P<0.05). Conclusion:The findings of this study suggested that ART inhibited the EMT-triggered migration and invasion of HCT-8 cells presumedly by inhibiting the activation of the Akt/Snail pathway to reverse EMT.

ABSTRACT

Objective:To explore the effect of chemical compound of aconitum alkaloid on the lipopolysaccharide (LPS)-induced inflammatory response of RAW264.7 macrophages and investigate its mechanism. Method:The chemical compounds of Aconitum Kusnezoffii Reichb were collected from TCMSP database with consideration of oral bioavailability (OB)≥30% and drug-likeness (DL)≥0.18. The potential targets of each chemical component were predicted with use of Pubchem database and Swiss Target Prediction database. Rheumatoid arthritis (RA) targets were collected from GeneCards database and selected by intersection screening. Gene ontology (GO) classification enrichment and Pathway enrichment analysis were carried out with use of DAVID database. Cytoscape was used to construct "Chemical Compound-Potential Targets-Pathway-Disease" network. Protein-protein interaction (PPI) network was constructed by using STRING database and Cytoscape software. RAW264.7 macrophages were stimulated by LPS to establish macrophage inflammation model in vitro. Western blot was used to detect the effects of chemical compounds on the expression of tumor necrosis factor-α (TNF-α) and cyclooxygenase-2 (COX-2) in RAW264.7 cells induced by LPS, as well as on the expression of JAK kinase and nuclear transcription factor- kappa B (NF-κB) signal pathway. Result:A total of 27 chemical compounds were obtained by searching TCMSP database and consulting literature (OB≥30%, DL≥0.18). 12 chemical compounds were obtained after screening. 177 potential targets were obtained after database prediction and screening, and 97 targets were obtained as potential targets for the treatment of RA after intersection between 177 potential targets and 4 329 RA targets. A total of 32 biological processes (BP), 5 cellular components (CC), and 12 molecular functions (MF) were enriched by DAVID database. The construction of network topology map showed that different chemical compounds can act on the same target and the same chemical compound can also act on different targets in the treatment of RA. Aconitum alkaloid can be connected with the same pathway through different targets or with different pathways through the same target, indicating that different targets may have synergistic effect, which fully reflected the complex multi-compound, multi-targets and multi-pathways mechanism. Different concentrations of LPS in stimulation (0-200 μg·L-1) can significantly up-regulate the expression of COX-2 protein in RAW264.7 macrophages (P<0.05), indicating that the inflammatory model was successful. Compared with the normal group, the expression of TNF-α and COX-2 protein in the inflammatory model of RAW264.7 cells increased significantly(P<0.05), while the expression of TNF-α and COX-2 protein in bulleyaconitine A(BLA), songorine, yunaconitine and karacoline groups decreased in varying degrees compared with the model group (P<0.05). Compared with the normal group, the expression of IRAK4, NF-κB, JAK1 and STAT3 in the inflammatory model of RAW264.7 cells were significantly increased (P<0.05), while such levels in BulleyaconitineA, songorine, yunaconitine and Karacoline groups were significantly lower than those in the model group(P<0.05). Conclusion:Based on systematic pharmacology and in vitro experiments, the related targets and signal pathways were analyzed to provide new insights into the pathogenesis of RA, reveal the molecular mechanism of aconitum alkaloid in the treatment of RA, and provide new ideas for the application of Mongolian medicine in modern medicine.

ABSTRACT

Objective:To study the effect of Aidi injection (AD) on the expression of cytochrome P450 isoenzyme 1A2,2E1,3A2,2C11(CYP1A2,2E1,3A2,2C11)mRNA and protein in rats with N-nitrosodiethylamine (DEN) chemically induced primary hepatocellular carcinoma(HCC). Method:Three healthy SD male rats were randomly selected as the blank group, and the remaining rats were treated with DEN intermittently induced primary hepatocellular carcinoma rat model. After success of the model, the rats were randomly divided into model group and AD group, with 3 rats in each group. The rats in the blank group and model group were intraperitoneally injected with 10 mL·kg-1 saline, while those in the AD group were intraperitoneally injected with 10 mL·kg-1 AD once a day, a total of 14 d intervention. Real-time quantitative polymerase chain reaction(Real-time PCR) and Western blot were used to detect the mRNA and protein expressions of CYP1A2, CYP2E1, CYP3A2 and CYP2C11, respectively. Result:Real-time PCR results showed that after 14 days of drug administration, compared with the blank group, the mRNA expressions of CYP1A2, CYP2E1, CYP3A2 and CYP2C11 were all down-regulated in para-cancerous tissue (PCT) and cancerous tissue (CT) in model group, and there were significant differences (P<0.05,P<0.01). Compared with the model group, the mRNA expressions of the four subtype enzyme were significantly down-regulated in PCT in the AD group(P<0.05,P<0.01), while the mRNA expressions of the four subtype enzyme were significantly up-regulated in CT (P<0.05), and the expression was down-regulated overall. Western blot results showed that compared with the blank group, the protein expressions of CYP1A2, CYP2E1, CYP3A2 and CYP2C11 in CT of the model group were significantly down-regulated (P<0.01), and the protein expressions of CYP3A2 and CYP2C11 were significantly down-regulated in PCT (P<0.01). Compared with the model group, the protein expressions of CYP1A2, CYP2E1, CYP3A2 and CYP2C11 in CT and PCT were down-regulated in the AD group, but the differences were not statistically significant. Conclusion:AD can down-regulate the mRNA and protein expressions of CYP1A2, CYP2E1, CYP3A2 and CYP2C11 in rat liver tissues. In clinical use of AD, attention should be paid to drug interactions that may be caused by CYP450 enzyme inhibition.

ABSTRACT

Objective:To observe the clinical efficacy of modified Huanglian Wendantang and Shaofu Zhuyutang in the treatment of ovulation disorder in patients with polycystic ovary syndrome (PCOS) due to the combined phlegm and stasis-induced and its influence on chronic inflammation. Method:According to the random number table, 100 patients were divided into a control group (50 cases) and an observation group (50 cases). Apart from lifestyle intervention and oral administration of clomiphene citrate (CC) capsules to induce ovulation, patients in the control group further received Guizhi Fulingwan, 6 g/time, 2 times/day, while those in the observation group were treated with the modified Huanglian Wendantang and Shaofu Zhuyutang, 1 dose/day, for six menstrual cycles. The ovulation, endometrial thickness, proportion of type A endometrium, as well as the pulsatility index (PI) and resistance index (RI) of uterine artery were monitored before and after treatment. The fasting blood glucose (FBG), fasting insulin (FINS), luteinizing hormone (LH), follicle stimulating hormone (FSH), serum testosterone (T), estradiol (E2), dehydroepiandrosterone sulfate (DHEAS), tumor necrosis factor-α (TNF-α), high-sensitivity C-reactive protein (hs-CRP), adiponectin (APN), and interleukin-6 (IL-6) levels before and after treatment were detected, followed by the calculation of homeostasis model assessment-insulin resistance (HOMA-IR) value and the evaluation of ovarian volume and combined phlegm and stasis-induced syndrome score. Result:The overall response rate of the observation group was (44/47) 93.62%, which was higher than (36/46) 78.26% of the control group (χ2=4.802, P<0.05). The ovulation rate in the observation group was (199/264) 75.38%, higher than (173/272) 63.60% in the control group (χ2=8.714, P<0.01). The clinical pregnancy rate of the observation group was (11/47) 23.40%, higher than (5/46) 10.87% of the control group, but the difference was not statistically significant (χ2=2.564, P>0.05). Compared with the control group, the observation group exhibited reduced PI, RI, LH, T, DHEAS, FINS, FPG, HOMA-IR, TNF-α, hs-CRP, and IL-6 (P<0.01), but elevated E2, FSH, and APN (P<0.01). Besides, the bilateral ovarian volume and combined phlegm and stasis-induced syndrome score of the observation group were smaller than those of the control group (P<0.01), while the endometrial thickness and proportion of type A endometrium were higher (P<0.01). Conclusion:On the basis of CC treatment, the modified Huanglian Wendantang and Shaofu Zhuyutang alleviates the ovulation disorder in PCOS patients of combined phlegm and stasis-induced syndrome and regulates IR and chronic inflammation, thus creating a favorable condition for clinical pregnancy, which is worthy of further research.

ABSTRACT

Chronic liver diseases have various etiologies and often have poor long-term prognosis in clinical practice. Y-box binding protein-1 (YB-1) is a multifunctional protein, and in-depth studies in recent studies have found that it plays a key role in the development and progression of chronic liver diseases such as liver fibrosis and hepatocellular carcinoma (HCC). This article summarizes the role of YB-1 in chronic liver diseases such as liver fibrosis, HCC (proliferation, apoptosis, metastasis, prognosis, and drug resistance), and liver failure, so as to provide a theoretical basis for the diagnosis and treatment of chronic liver diseases.

ABSTRACT

Phelan-McDermid syndrome (PMS)(OMIM#606232) is a rare genetic disorder caused by a deletion of the distal long arm of chromosome 22q13 involving a variety of clinical features with considerably heterogeneous degrees of severity. This syndrome is characterized by global developmental delay, intellectual disability, hypotonia, absent or severely delayed speech, minor dysmorphic features and autism spectrum disorder. PMS is easy to be misdiagnosed due to the lack of specific clinical manifestations. SHANK3 has been identified as the critical candidate gene for the neurological features of this syndrome. However, some studies have shown that other genes located in the 22q13 region may have a role in the formation of symptoms in individuals with PMS. This article provides a review for recent progress made in research on PMS including etiology, clinical manifestation, diagnosis, and treatment, with a particular emphasis on clinical diagnosis and treatment.

ABSTRACT

Proteins and peptides have become a significant therapeutic modality for various diseases because of their high potency and specificity. However, the inherent properties of these drugs, such as large molecular weight, poor stability, and conformational flexibility, make them difficult to be formulated and delivered. Injection is the primary route for clinical administration of protein and peptide drugs, which usually leads to poor patient's compliance. As a portable, minimally invasive device, microneedles (MNs) can overcome the skin barrier and generate reversible microchannels for effective macromolecule permeation. In this review, we highlighted the recent advances in MNs-mediated transdermal delivery of protein and peptide drugs. Emphasis was given to the latest development in representative MNs design and fabrication. We also summarize the current application status of MNs-mediated transdermal protein and peptide delivery, especially in the field of infectious disease, diabetes, cancer, and other disease therapy. Finally, the current status of clinical translation and a perspective on future development are also provided.

ABSTRACT

We explored the pharmacodynamic material basis and network regulatory mechanism of Fufang Yuxingcao Mixture (FYM) for the treatment of fever and inflammation. Targets of the 25 compounds in FYM were predicted according to the reverse pharmacophore method and TCMSP, UniProt database. Gene ontology (GO) function enrichment and pathway analysis of the targets was analyzed by Omicsbean software and the Kyoto Gene and Genome Encyclopedia (KEGG) database. A "compound-target-pathway-pharmacological action-effect" network was established with Cytoscape 3.6.1 software. The lipopolysaccharide (LPS)-induced RAW264.7 cell inflammation model was used to verify the anti-inflammatory effects of FYM and its 10 important components. The network pharmacology experiment showed that 25 compounds affected 97 pathways through 211 targets, of which 15 key targets [including RAC-alpha serine/threonine-protein kinase (AKT1), insulin (INS), vascular endothelial growth factor A (VEGFA), interleukin-6 (IL-6), cellular tumor antigen p53 (TP53), tumor necrosis factor (TNF), transcription factor AP-1 (JUN), caspase-3 (CASP3), matrix metalloproteinase-9 (MMP9), interleukin-8 (IL-8), prostaglandin G/H synthase 2 (PTGS2), proto-oncogene c-Fos (FOS), tyrosine-protein kinase SRC (SRC), c-Jun N-terminal kinase 1 (MAPK8), estrogen receptor 1 (ESR1)] and 46 pathways (including NF-kappa B signaling pathway, Toll-like receptor signaling pathway, MAPK signaling pathway, IL-17 signaling pathway, arachidonic acid metabolism, cAMP signaling pathway, T cell receptor signaling pathway, calcium signaling pathway, inflammatory mediator regulation of TRP channels, chemokine signaling pathway, Th1 and Th2 cell differentiation, natural killer cell mediated cytotoxicity, etc.) were related to anti-inflammatory, antipyretic, immune regulation, and analgesia. In vitro cell experiments showed that FYM and the 10 components (including isoquercitrin, luteoloside, baicalein, wogonin, wogonoside, phillyrin, forsythoside A, chlorogenic acid, isochlorogenic acid A, and sweroside) could significantly reduce the expression of nitric oxide (NO), TNF-α and IL-6 in cell supernatants, indicating that the above 10 components may be the key pharmacodynamic material basis of FYM.

ABSTRACT

Liver is the main place of drug metabolism. Mitochondria of hepatocytes are important targets of drug-induced liver injury. Mitochondrial autophagy could maintain the healthy operation of mitochondria in cells and the stable proliferation of cells. Therefore, the use of mitochondrial autophagy to remove damaged mitochondria is an important strategy of anti-drug-induced liver injury. Active ingredients that could enhance mitochondrial autophagy are contained in many traditional Chinese medicines, which could regulate the mitochondrial autophagy to alleviate relevant diseases. However, there are only a few reports on how to accurately and efficiently identify and evaluate such components targeting mitochondria from traditional Chinese medicine. Liquid chromatography-mass spectro-metry(LC-MS) combined with serum pharmacology in vivo can be used to accurately and efficiently find active ingredients of traditional Chinese medicine acting on mitochondrial targets. This paper reviewed the research ideas and methods of traditional Chinese medicine ingredients for increasing the hepatotoxicity of mitochondrial autophagy, in order to provide new ideas and methods for the study of active ingredients of traditional Chinese medicine targeting mitochondria.
