ABSTRACT

OBJECTIVE To analyze the clinical characteristics of liraglutide-induced pancreatitis, and to provide reference for clinical rational drug use. METHODS Retrieved from CNKI, VIP, Wanfang database, PubMed, Web of Science and Medline, case reports about liraglutide-induced pancreatitis were collected from the inception to December 31st, 2022. Demographic characteristics, drug use, clinical manifestations, intervention and outcome were analyzed using descriptive statistical method. RESULTS A total of 17 pieces of literature were collected and 17 patients were involved, including 7 males and 10 females. The patients aged from 25 to 75 years. All 17 patients had drug indications, including 14 cases of type 2 diabetes mellitus, 3 cases of obesity or overweight. Among 17 patients, liraglutide was used alone in 5 cases, and combined with other drugs in 12 cases. Time from liraglutide administration to pancreatitis occurrence ranged from 1 day to 11 months after medication in 17 patients, with 14 cases less than 6 months. The clinical manifestations mainly included abdominal pain, nausea and vomiting, etc. After the diagnosis of pancreatitis, liraglutide discontinuation occurred in 16 patients; 1 case did not receive any other interventions and the other 15 cases were managed with symptomatic supportive treatment; the symptoms of all 16 patients resolved; however, 2 patients suffered from second episode of severe pancreatitis several weeks after liraglutide discontinuation, pancreatitis recurred after liraglutide rechallenge in 1 case. The results of correlation evaluation showed that 1 case was “positive”, 4 cases were “possible”, and the remaining patients were “very likely”. CONCLUSIONS Liraglutide-induced pancreatitis mainly occurred within 6 months after drug administration. The majority of liraglutide-induced pancreatitis cases are mild to moderate, but there are also severe and even fatal cases. It is advisable to periodically monitor the level of pancreatic enzymes and closely observe patients’ clinical mani-festations. In case of suspected liraglutide-induced pancreatitis,drug withdrawal and symptomatic treatment should be taken immediately.

ABSTRACT

Objective:To explore the scientific research capacity and its influencing factors of nursing graduates in application-oriented university.Methods:A total of 272 nursing graduates from one application-oriented university of Hunan province were selected as research subjects by cluster sampling in this study. The Scientific Research Capacity Self-rating Scale and The Chinese Version of Critical Thinking Disposition Inventory were used to conduct a questionnaire survey. SPSS 22.0 was used for independent-samples t-test, ANOVA, Spearman correlation and multiple linear regression. Results:The scores of scientific research capacity were (49.27±21.26) points, and 255 (93.7%) nursing graduates showed a medium and below degree. A positive correlation was found between nursing scientific research capacity and total critical thinking disposition, certain subscales of analytical capability, critical thinking self-confidence and inquisitiveness ( r=0.18, 0.23, 0.45, 0.32, and all P<0.01), but there was a negative correlation between nursing scientific research capacity and truth seeking ( r=-0.16, P<0.05). Training mode, experience of scientific research practice, research motivation, critical thinking self-confidence and truth seeking were also found as influencing factors of nursing scientific research capacity of nursing graduates (standardized β=0.12, 0.16, 0.14, 1.31, -0.12 , and all P<0.05). Conclusion:The scientific research capacity, especially the data analysis and study design capacity of nursing graduates in application-oriented university need to be improved, which suggests that targeted training can be carried out by establishing correct research motivation, strengthening the training of critical thinking ability in school and encouraging scientific research practice.

ABSTRACT

Posttranslational modifications of antibody products affect their stability,charge distribution,and drug activity and are thus a critical quality attribute.The comprehensive mapping of antibody modifications and different charge isomers(CIs)is of utmost importance,but is challenging.We intended to quanti-tatively characterize the posttranslational modification status of CIs of antibody drugs and explore the impact of posttranslational modifications on charge heterogeneity.The CIs of antibodies were fraction-ated by strong cation exchange chromatography and verified by capillary isoelectric focusing-whole column imaging detection,followed by stepwise structural characterization at three levels.First,the differences between CIs were explored at the intact protein level using a top-down mass spectrometry approach;this showed differences in glycoforms and deamidation status.Second,at the peptide level,common modifications of oxidation,deamidation,and glycosylation were identified.Peptide mapping showed nonuniform deamidation and glycoform distribution among CIs.In total,10 N-glycoforms were detected by peptide mapping.Finally,an in-depth analysis of glycan variants of CIs was performed through the detection of enriched glycopeptides.Qualitative and quantitative analyses demonstrated the dynamics of 24 N-glycoforms.The results revealed that sialic acid modification is a critical factor ac-counting for charge heterogeneity,which is otherwise missed in peptide mapping and intact molecular weight analyses.This study demonstrated the importance of the comprehensive analyses of antibody CIs and provides a reference method for the quality control of biopharmaceutical analysis.

ABSTRACT

Objective:To explore the application effects of mixed courses based on E-disciplinary platform on the teaching of nutrition and food hygiene.Methods:The study included in 31 preventive medicine students from group 1 to 3 Batch 2015 in Binzhou Medical University, who were divided into control group adopting (lecture-based learning, LBL) teaching model. While another 30 students from group 4-6 were divided into the experimental group adopting mixed teaching based on E-disciplinary platform. Analysis and assessments were done by course examinations and questionnaires. SPSS 20.0 was used for t test, rank sum test and chi-square test. Results:According to the course examinations, students in the experimental group got higher scores than those in the control group in terms of usual, examination and overall test scores [(46.17±8.84) points vs. (42.35±8.28) points; (37.31±6.58) points vs. (33.61±6.38) points; (81.97±9.18) points vs. (74.32±8.32) points, ( t=4.69, t=3.16, t=5.16, P< 0.01)]. Furthermore, the experimental group were better than the control group in the number distribution of excellent performance in usual, examination and overall test ( Z=2.71, Z=2.13, Z=4.57, P<0.05). The questionnaires demonstrated that the overall satisfaction rate of the experimental group (77.8%) was higher than that of the control group (47.8%) ( χ2=34.98, P<0.001). Conclusion:Student learning effect and the teaching quality can be improved by applying mixed teaching based on E-disciplinary platform.

ABSTRACT

Objective: To investigate the effects and mechanism of hydrogen peroxide (HP) pretreatment with low molarity on oxidative stress induced apoptosis of mouse bone marrow mesenchymal stem cells (BMSCs). Methods: The experimental research methods were used. BMSCs were isolated and cultured from two 2-week-old male BALB/c mice by the whole bone marrow culture method. The 3rd-7th passages of cells in logarithmic growth phase were used for the experiments after identification. According to the random number table (the same grouping method below), the cells were divided into 0 μmol/L HP group (without HP, the same below), 25 μmol/L HP group, 50 μmol/L HP group, 100 μmol/L HP group, 150 μmol/L HP group, 200 μmol/L HP group, 250 μmol/L HP group, and 300 μmol/L HP group in which cells were treated by the corresponding final molarity of HP, respectively. The apoptosis rate was detected by flow cytometry (n=4) after 24 hours of culture. The cells were divided into 0 μmol/L HP group, 25 μmol/L HP group, 50 μmol/L HP group, and 100 μmol/L HP group in which cells were treated by the corresponding final molarity of HP, respeclively. After 24 hours of culture, the protein expressions of B-lymphoma-2 (Bcl-2) and Bcl-2-related X protein (Bax) were detected by Western blotting, and the Bcl-2/Bax ratio was calculated (n=3). The cells were divided into 0 μmol/L HP group, 25 μmol/L HP group, 50 μmol/L HP group, 100 μmol/L HP group, 200 μmol/L HP group, and 300 μmol/L HP group in which cells were treated by the corresponding final molarity of HP, respectively. After 24 hours of culture, the protein expressions of glycogen synthase kinase-3β (GSK-3β) and phosphorylated GSK-3β (p-GSK-3β) were detected by Western blotting (n=3). The cells were divided into 0 μmol/L HP group, 50 μmol/L HP group, and 300 μmol/L HP group in which cells were treated by the corresponding final molarity of HP, respeclively, and HP pretreatment group with 50 μmol/L HP being added in advance for 12 h and then 300 μmol/L HP being added. After 24 hours of culture, the morphology and growth of cells were observed by inverted fluorescence microscopy (non-fluorescent condition) and immunofluorescence method, the apoptosis rate was detected by flow cytometry, the protein expressions of Bcl-2, Bax, cysteine aspartic acid specific protease-3 (caspase-3), caspase-9, cleavage caspase-3, cleavage caspase-9, GSK-3β, and p-GSK-3β were detected by Western blotting, and the Bcl-2/Bax ratio was calculated, with all the number of samples being 3. Data were statistically analyzed with one-way analysis of variance and Bonferroni test. Results: After 24 hours of culture, compared with that in 0 μmol/L HP group, the apoptosis rate of cells did not change significantly in 25 μmol/L HP group, 50 μmol/L HP group, or 100 μmol/L HP group (P>0.05) but increased significantly in 150 μmol/L HP group, 200 μmol/L HP group, 250 μmol/L HP group, and 300 μmol/L HP group (P<0.01). After 24 hours of culture, compared with that in 0 μmol/L HP group, the Bcl-2/Bax ratio of cells increased significantly in 25 μmol/L HP group and 50 μmol/L HP group (P<0.05 or P<0.01) but decreased significantly in 100 µmol/L HP group (P<0.05). After 24 hours of culture, compared with those in 0 μmol/L HP group, the protein expression of GSK-3β in cells showed no significant change in 25 μmol/L HP group and 50 μmol/L HP group (P>0.05), the protein expressions of p-GSK-3β in cells significantly increased in 25 μmol/L HP group and 50 μmol/L HP group (P<0.01), the protein expressions of GSK-3β and p-GSK-3β in cells in 100 μmol/L HP group showed no significant change (P>0.05), the protein expressions of GSK-3β in cells in 200 μmol/L HP group and 300 μmol/L HP group were significantly increased (P<0.05). but the protein expression of p-GSK-3β in cells in 200 μmol/L HP group and 300 μmol/L HP group was significantly decreased (P<0.05). After 24 hours of culture, the morphology and growth of cells in 0 μmol/L HP group and 50 μmol/L HP group were similar and normal; in contrast, the cells in 300 µmol/L HP group became smaller and round, with the cell protrusions being shorter or disappeared, the nucleus being cavitated, and the cell abscission being increased significantly; the morphology of most cells in HP pretreatment group was normal, with the shedding of cells being less than that in 300 µmol/L HP group, and the morphology of nucleus being normal. After 24 hours of culture, the protein expression of caspase-9 was similar among the four groups (P>0.05). Compared with that in 0 μmol/L HP group, the apoptosis rate and the protein expressions of cleavage caspase-9, caspase-3, and cleavage caspase-3 of cells in 50 μmol/L HP group showed no significant changes (P>0.05), the Bcl-2/Bax ratio of cells in 50 μmol/L HP group increased significantly (P<0.05), the apoptosis rate and the protein expressions of cleavage caspase-9, caspase-3, and cleavage caspase-3 of cells in 300 μmol/L HP group were significantly increased (P<0.01), while the Bcl-2/Bax ratio of cells in 300 μmol/L HP group was significantly decreased (P<0.05). Compared with those in 300 μmol/L HP group, the apoptosis rate and the protein expressions of cleavage caspase-9, caspase-3, and cleavage caspase-3 of cells were significantly decreased in HP pretreatment group (P<0.05 or P<0.01), while the Bcl-2/Bax ratio of cells was significantly increased in HP pretreatment group (P<0.01). After 24 hours of culture, the protein expressions of GSK-3β and p-GSK-3β of cells in 0 μmol/L HP group, 50 μmol/L HP group, 300 μmol/L HP group, and HP pretreatment group were 1.09±0.14, 0.62±0.17, 1.35±0.21, 0.74±0.34, 0.68±0.03, 0.85±0.08, 0.38±0.10, and 0.54±0.09, respectively. Compared with those in 0 μmol/L HP group, the protein expression of p-GSK-3β of cells was significantly increased in 50 μmol/L HP group (P<0.05) but significantly decreased in 300 μmol/L HP group (P<0.01), while the protein expression of GSK-3β of cells was significantly increased in 300 μmol/L HP group (P<0.05). Compared with those in 300 μmol/L HP group, the protein expression of GSK-3β of cells was significantly decreased in HP pretreatment group (P<0.01), while the protein expression of p-GSK-3β of cells was significantly increased in HP pretreatment group (P<0.01). Conclusions: The molarity of 50 μmol/L may be the optimal molarity of HP to pretreat mouse BMSCs, and 50 μmol/L HP pretreatment can antagonize mitochondrial pathway of oxidative stress induced apoptosis by inhibiting the activity of GSK-3β.

ABSTRACT

OBJECTIVE@#To evaluate the clinical efficacy of Huangqi Sijunzi decoction (HQSJZD) for treating cancer-related fatigue (CRF) of spleen and stomach Qi deficiency type after chemotherapy in patients with breast cancer.@*METHODS@#A total of 94 breast cancer patients who developed CRF of spleen and stomach Qi deficiency type after chemotherapy were randomized into chemotherapy group (n=47) and traditional Chinese medicine (TCM) + chemotherapy group (n=47). The patients in chemotherapy group received the AC or EC regimen and non-drug interventions including psychological counseling, and those in TCM + chemotherapy group received oral administration of HQSJZD in addition to chemotherapy for 21 days as a treatment cycle, after which improvement of fatigue was assessed using Modified Piper Fatigue Scale. The active ingredients and targets of HQSJZD were screened using the TCM System Pharmacology Analysis Platform (TCMSP); the CRF- and breast cancer-related disease targets were retrieved based on data from the GeneCards, NCBI gene and OMIM databases to construct the component-target network and the protein-protein interaction (PPI) network. GO functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes KEGG pathway enrichment analysis of the target genes were performed to construct the component-disease-pathway-target biological network. The binding strength of the major drug ingredients and CRF key targets were predicted using AutoDock software.@*RESULTS@#The scores for somatic fatigue, emotional fatigue and cognitive fatigue, along with the overall fatigue score, showed more significant improvements in TCM+chemotherapy group than in chemotherapy group (P < 0.001), and the response rate reached 89.4% in the combined treatment group. We identified 250 targets for HQSJZD, 2653 CRF-related genes, 15 329 breast cancer-related genes and 161 prescription-disease intersected targets, from which topological analysis identified 66 potential key targets. GO and KEGG enrichment analyses predicted multiple pathways related with the disease. Molecular docking results suggested that the core ingredients of HQSJZD showed high affinities to the key targets AKT1, CASP3, IL6, JUN and VEGFA, among which AKT1 might be the most important target for HQSJZD to treat CRF.@*CONCLUSION@#HQSJZD can obviously improve CRF symptoms in breast cancer patients possibly by regulating multiple signaling pathways including PI3K-Akt through AKT1.

ABSTRACT

Neuropsychiatric disorders are the leading cause of mental and intellectual disabilities worldwide. Current therapies against neuropsychiatric disorders are very limited, and very little is known about the onset and development of these diseases, and their most effective treatments. MIR137 has been previously identified as a risk gene for the etiology of schizophrenia, bipolar disorder, and autism spectrum disorder. Here we generated a forebrain-specific MIR137 knockout mouse model, and provided evidence that loss of miR-137 resulted in impaired homeostasis of potassium in mouse hippocampal neurons. KCC2, a potassium-chloride co-transporter, was a direct downstream target of miR-137. The KCC2 specific antagonist VU0240551 could balance the current of potassium in miR-137 knockout neurons, and knockdown of KCC2 could ameliorate anxiety-like behavior in MIR137 cKO mice. These data suggest that KCC2 antagonists or knockdown might be beneficial to neuropsychiatric disorders due to the deficiency of miR-137.

Objective To explore the application and evaluation of S-P chart in analyzing examination papers of nutrition and food hygiene and improve the quality of examination.Methods 15 papers were randomly selected from 56 examination papers of nutrition and food hygiene and 5,4,5,1,0 were randomly selected from the students with scores of <60,60-69,70-79,80-89,and 90-100 with stratified samplingmethod.S-P chart and fuzzy S-P chart were drawn according to objective and subjective questions.Results In the S-P chart of multiple choice,both the P-curve and S-curve were in the lower right corner.There was no obvious fault on the S-curve,while P-curve had obvious faults.There were 2 questions and 1 student with alarm coefficient > 0.75.The difference,difficulty coefficient,stability coefficient and suitability coefficient were 0.13,0.66,0.55 and 0.58 respectively.In the S-P chart of multiple choices,both the P-curve and S-curve were in the upper left corner.There was no obvious fault on the S-curve,while P-curve had obvious faults.There were 1 question and 1 student with alarm coefficient > 0.75.The difference,difficulty coefficient,stability coefficient and suitability coefficient were 0.10,0.30,0.64 and 0.60 respectively.In the S-P chart of subjective questions,the difference was 0.275,and both the P-curve and S-curve were in the upper left corner and had no obvious fault.Conclusion The application of S-P chart can help teachers to analyze the difference between the quality of test questions and the grade of students,thereby making teaching more targeted and improving the quality of teaching and examination.

ABSTRACT

With Bupleurum smithii var. parvifolium and B. scorzonerifolium as test objects, in order to provide a theoretical basis for the introduction and domestication of B. smithii var. parvifolium, the growth and development dynamics of seedlings, biomass accumulation, the content of malonaldehyde(MDA), the activity of antioxidase such as SOD, POD, CAT and APX between them were comparatively analyzed by direct sowing culture in the open field. The results indicated that the morphological index and the biomass accumulation of B. smithii var. parvifolium such as root diameter, root length, plant height and leaf number were inferior to B. scorzonerifolium, the antioxidase SOD and POD activity of B. smithii var. parvifolium was significantly inferior to B. scorzonerifolium (<0.05), the antioxidase CAT and APX activity of B. smithii var. parvifolium was inferior to B. scorzonerifolium but the difference wasn't significant, while MDA content was superior to B. scorzonerifolium(<0.05). Thus, compared with cultivated B. scorzoneri folium, the plant growth velocity of wild B. smithii var. parvifolium was relatively slower and its resistance was relatively weaker after introduction and domestication.

ABSTRACT

Objective To analyze the related rules and prognosis analysis for lymphatic metastasis in patients for Siewert Ⅱ and Ⅲ adenocarcinoma of the gastroesophageal junction.Methods This study retrospectively reviewed 65 patients who underwent radical surgery for Siewert Ⅱ and Ⅲ adenocarcinoma of the gastroesophageal junction in Beijing Friendship Hospital between July 2013 and March 2017.The primary indicators were sex,age,tumor site,tumor size,macroscopic type,histological type,depth of invasion,operation,tumor emboli and carcinoma nodes.The clinical characteristics and pathological features were analyzed by using Logistic regression and Kaplan-Meier survival analysis.Follow-tup using outpatient examination and telephone interview was performed to detect patients' diseases and postoperative survival up to April 2017.Results Lymph node metastasis was observed in 41 of 65 patients (63.1%).Lymph node metastasis frequency was found in No.1,No.2,No.3,No.7,No.11 and No.110 lymph nodes.The incidences of those lymph node metastasis were 45.3%,32.5%,28.8%,22.5%,19.4% and 8.2%.The metastasis rate of the tumors with diameter over 2 cm was higher than the tumors which were less than 2 cm (71.9% vs 0;P =0.000);The observed rate of lymph node metastasis was 0,12.5%,69%,95.2% in early cancer (T1) and advanced cancer (T2-T4) (P=0.000) respectively.The metastasis rate of highly differentiated Siewert Ⅱ and Ⅲ adenocarcinoma of the gastroesophageal junction was 45.2% lower than poorly differentiated tumors 79.4% (P =0.009).Thirty-three cases were found with lymphatic embolus,28 of them (84.8%) were found to be combined with lymph node metastasis among them (P =0.001).All the differences were statistically significant (P < 0.05).The results of multivariable analyzed by binary logistic regression analysis revealed that neoplasms histologic type and lymphatic embolus were independent risk factors for lymph node metastasis of Siewert Ⅱ and Ⅲ adenocarcinoma of the gastroesophageal junction.Sixty-five patients were followed up for 1-45 months,with a median time of 18.81 months.Although there was no statistically significant difference,3 years-overall survival rate showed tumor without lymph node metastasis has better prognosis than with lymph node metastasis and early stage tumor also has better prognosis than advanced tumor.Conclusions Lymph node metastasis of Siewert Ⅱ and Ⅲ adenocarcinoma of the gastroesophageal junction is related with histological type of tumor and lymphatic embolus.For Siewert Ⅱ and Ⅲ adenocarcinoma of the gastroesophageal junction,the incidence of lymphatic metastasis were higher appear in abdominal cavity,which included No.1,No.2,No.3,No.7 and No.11;Meanwhile,the metastasis of mediastinal lymph node is given priority to No.110 lymph node.Therefore,radical total gastrectomy combined with standard D2 lymphadenectomy and No.110 lymphadenectomy are performed in order to improving the prognosis.For patients with lymph node metastasis and tumor in advanced stage,long-term prognosis remains to be further study.

ABSTRACT

Objective To explore the application effects of blending learning mode which is based on SPOC in preventive medicine teaching. Methods 92 students of Grade 2013 majoring in medical imag-ing were divided into two classes. 47 students in the first class adopted LBL teaching method. Another class was experimental group, teaching the remaining 45 students by blending learning mode based on SPOC method. Analyses and assessments were done by course examinations and questionnaires. Results The experimental groups were better than the control groups not only in the average score of synthetic scores (t=5.068, P=0.000) but also in the 80-89 and 90-100 number distribution (Z=3.713, P=0.000). The question-naires showed that 72.6%students of the experimental group accepted blending learning mode which was based on SPOC. Conclusion Ability of explorative thinking, independent learning and effect of teaching can be achieved by blending learning model which is based on SPOC.

ABSTRACT

Objective To explore the effects of hawthorn proanthocyanidins (HPC) and vitamin C (VC) on kidney protection in insulin-resistance (IR) rats. Methods The IR rats were induced by high-fat diet in order to establish the model of nephropathy in type II diabetic rats, and the high-fat feeding were continued for 2 weeks after the IR rats were made. The contents of fasting blood glucose, serum insulin, glucose and microabuminuria (mAlb) in urine were tested. Fifty IR model rats were divided into model, HPC (56 mg/kg), VC (180 mg/kg), combined application of HPC (56 mg/kg) and VC (180 mg/kg), and rosiglitazone (2 mg/kg) groups. Another 10 normal rats were selected randomly to be control group. After continuous ig administration for 20 weeks in each group, these indexs, such as the levels of fasting blood glucose (FBG), serum insulin (SI), glucose in urine (24 h), creatinine (Cr), urea nitrongen (UN), uric acid (UA), were all detected. The excretion of urinary total protein (UTP), microabuminuria (mAlb), N-acetyl-β-D-glucosaminidase (NAG) were also measured. Moreover, the histopathological microstructure of kidney was observed by light microscope. Results After modeling, the levels of FBG, SI and glucose in urine increased significantly (P < 0.01) in comparation with the control group, so does the levels of UN, Cr, UA (P < 0.01) and the excretion rates of UTP, mAlb, NAG (P < 0.01). However, all these indexs were decreased significantly (P < 0.01) after the combined use of HPC and VC, which were more notable than that in HPC and VC group (P < 0.01) and equivalent to that in rosiglitazone group. Conclusion The combined application of HPC and VC can improve the renal function of IR rats and have protective effects on kidney injury.

Objective To discuss the experience of self -perceived burden(SPB)in AIDS patients and to provide evidence for promoting physical and mental health.Methods Interviews were conducted among 10 AIDS patients,and the data were analyzed with the Colaizzi 's method.Results 3 themes from the experience of self -perceived burden in AIDS patients were identified:self -accusation and remorse,fear and insecurity,loss of dignity. Conclusion The research results showed feelings,difficulties and needs of patients in the process of disease.Nurses should think about the influence of SPB during the nursing intervention and evaluation.Nurses should alleviate the problems faced by patients in order to increase the health level,reduce the psychological pressure and increase the medication compliance of the patients in the long -term treatment.It can be used in clinical nursing.

ABSTRACT

Objective To assess the applicative effect of the flipped classroom in nutrition and food hygiene. Methods 48 students of Class One from Grade 2011 majoring in preventive medicine were taken as the TBL group, with 49 students of Class Two as the flipped classroom group. The TBL group used TBL and the formative assessment, while the flipped classroom group used the flipped classroom and the forma-tive assessment. The teaching effect was evaluated through the unified examination and teaching satisfaction questionnaire survey. SPSS 17.0 statistical software was used for the data t test between the groups, and Wilcoxon test, chi square test were used for the linear correlation analysis of the usual grade and final grade of the flipped classroom group. Results The experimental scores (9.21 ±1.14) and the final scores (66.76±4.53) of the flipped classroom group students were significantly higher than the experimental scores (8.31±1.01) and final scores (61.31±4.37) of the TBL group and the difference was statistically significant (t=4.50, P=0.004; t=5.45, P=0.003). The excellent distribution of the flipped classroom group's overall results were higher than that of the TBL group (u=21.36, P=0.002). The usual scores and the final scores were positively correlated (r=0.960, P=0.000) in the flipped classroom group. The results of the question-naire showed that the flipped classroom group students' satisfaction with the positive impact the teaching methods had on all aspects of their own was higher than that of TBL group (P<0.05). Conclusion The flipped classroom teaching can help improve the students' learning enthusiasm, thinking activity and com-municative competence.

ABSTRACT

Objective: To explore the effect of hawthorn proanthocyanidins (HPC) and vitimin C (VC) on liver oxidative stress in insulin-resistance rats. Methods: The insulin-resistance models were prepared by high-fat diet, the weight variations of all rats were tested before and after modeling, so were the contents of fasting blood-glucose and serum insulin after modeling. The colorimetric technique was used to test the concentration of serum ALT, AST, and ALP. The rats in high-fat diet group, who were succeeded in modeling, were divided into model, HPC (56 g/kg), VC (180 g/kg), HPC (56 g/kg) + VC (180 g/kg), and rosiglitazone (122 g/kg) groups. After 12 weeks of continuous administration, these indexes, such as levels of SOD, CAT, GSH-Px, GSH, and MDA in liver homogenates and SOD, GSH-Px, Na+, K+-ATPase, Ca2+, Mg2+-ATPase, and MDA in liver mitochondria, were all tested; RT-PCR was used to test the expression of PPAR-γ mRNA; The Western blotting method was adopted to test the PPAR-γ protein expression. Results: After modeling, the body weight of rats decreased significantly (P<0.05), the concentration of blood, such as glucose, serum insulin ALT, AST, and ALP all increased significantly (P<0.01); The model group was compared with control group, the activities of SOD, CAT, GSH-Px, and GSH were significantly decreased (P<0.01), the levels of glucose, insulin, and MDA increased significantly (P<0.01) in rat liver homogenate, the activities of SOD, GSH-Px, Na+, K+-ATPase, and Ca2+, Mg2+-ATPase were significantly decreased (P<0.01) and MDA level increased significantly (P<0.01) in the liver mitochondria; PPAR-γ mRNA and protein expression decreased significantly in liver tissue (P<0.01). HPC, VC, HPC + VC, and rosiglitazone improved the above indexes, which in the HPC + VC group was better than that in the HPC group and VC group, equivalent to that in rosiglitazone group. Conclusion: Liver oxidative stress which is resulted from insulin-resistance can be improved when HPC and VC are combined.

Objective To observe the effects of soybean isoflavone in attenuating trans fatty acids-induced athero-sclerosis in rats .Methods The rats were randomly divided into normal diet group、trans fatty acids group、soybean isoflavone group and trans fatty acids +soybean isoflavone group .Research kit was used to test the amounts of TG , HDL,LDL and IL-6,TNF-αin rats'blood.Bovine aortic endothelial cells were cultured and divided into control group,trans fatty acids group and trans fatty acids group +soybean isoflavone group .Western blot was performed to test the NF-κB phosphorylation ,as well as to determine ICAM-1 and VCAM-1 protein contents in bovine aortic endo-thelial cells.Results Compared with normal diet group ,the amounts of TG,LDL,IL-6,TNF-αincreased evidently (P<0.05), whereas the amounts of HDL decreased evidently (P<0.05) in the rats'blood of trans fatty acids group . After the soybean isoflavone intervened ,the amounts of TG,LDL,IL-6,TNF-αall decreased (P<0.01), meanwhile the amounts of HDL increased significantly (P<0.01).Soybean isoflavone inhibited NF-κB phosphorylation induced by trans fatty acids ( P<0.01) .The high expression of ICAM-1 and VCAM-1 induced by trans fatty acids was de-creased by soybean isoflavone when comparing with those in the control group ( P<0.01 ) .Conclusions Soybean isoflavone has a role in attenuating trans fatty acids trans fatty acids-induced atherosclerosis in rats .

Objective To analyze the implementation effect of formative assessment in nutri-tion and food hygiene. Methods 55 students of Class One from grade 2010 majoring in preventive medicine were taken as the control group, with 56 students of Class Two as the experimental group.The control group used the summative assessment , while the experimental group used the formative assessment. Results were analyzed by course examinations and questionnaire surveys. SPSS 17.0 soft-ware was used and the measurement data were expressed by x±s. Statistical analysis was performed using t test, test, rank sum test, a Yuan linear correlation analysis, with the level of test α=0.05. Results The experimental scores and final scores of experimental group students were significantly higher than the control group and the difference was statistically significant(P values were 0.001, 0.000). The experimental group's overall result is closer to the negative skewed distribution. The ex-perimental group's average score of 89 points is significantly higher than the control group's average score of 79 points(P<0.01), and the usual results and the final results were positively correlated(r=0.865, P<0.001). The survey indicated that the students recognized the application of formative assess-ment in teaching process . Conclusion Formative assessment can help improve the teaching and learning quality in nutrition and food hygiene.

ABSTRACT

<p><b>OBJECTIVE</b>To analysis the molecular interaction network of 14-3-3 sigma in non small cell lung cancer (NSCLC) cells.</p><p><b>METHODS</b>Established stable over-expressed 14-3-3 sigma protein PG cells, MTT assay was used to assess the growth rate of PG cells. Though stable isotope labeling by amino acids in cell culture (SILAC) and Mass spectrometry (MS) technology, to identify difference expressed proteins caused by over expressed 14-3-3 sigma. The protein expressed >2 or <0.5 times was termed as the differential protein. By searching Human protein reference database (HPRD) and Kyoto encyclopedia of genes and genomes (KEGG), established the molecular interaction network of tumor suppressor gene 14-3-3 sigma.</p><p><b>RESULTS</b>The growth rate of over-expressed 14-3-3 sigma PG cell was obviously slower down compared to vector PG cells. A database including 147 differential protein was established. And a molecular interaction network of 14-3-3 sigma containing 26 protein was constructed.In this network, the expression of CSNK2A1 (casein kinase II subunit alpha), involved in numerous cellular processes, such as cell cycle progression, apoptosis and transcription, was the most significantly increased. A DNA repair protein, MEN1 (Menin) which functions as a transcriptional regulator was the most significantly decreased.</p><p><b>CONCLUSION</b>After stable transfected with 14-3-3 sigma gene, growth rate of PG cells was inhibited, the proteins associated with cell cycle, DNA damage repair mechanisms were significantly changed, and constructed the molecular interaction network.</p>
