ABSTRACT
Objective To study the inhibitory mechanism of soliquiritigenin on tyrosinase. Methods The inhibition effect and inhibitory kinetics of tyrosinase induced by oliquiritigenin were investigated. The effect of isoliquiritigenin on A375 melanoma cell were preliminarily indicated. Results The inhibitory effect of isoliquiritigenin on monophenolase and diphenolase of tyrosinase was good, IC50 was (11.22 ±0.92) μM and (48.53 ±4.75) μM, isoliquiritigenin was a competitive inhibitor, the value of inhibition constant ( Ki ) was ( 12.14 ±0.54 ) μM.Isoliquiritigenin could inhibit the prolifevation of A375 cell significantly, and the tyrosinase activity and melanin synthesis decreased. Conclusion Isoliquiritigenin as a tyrosinase inhibitor, plays an important role in the regulation of melanin, which provides the theoretical basis for the clinical anti skin cancer.
ABSTRACT
Objective To investigate the role of a small RNA interference against VEGF in radiation sensitivity in melanoma .Methods A375 human melanoma cell lines were transplanted into nude mice ,which with malignant melanoma were randomly divided into control group , VEGF negative plasmid group and VEGF positive plasmid group, followed by 4Gy irradiation twice a week for 2 weeks. The volume of tumor was calculated twice a week, the area of tumor necrosis was assayed by HE,the expression of VEGF in tumor was determined by Western-blot and Immunohistochemical. ResuIts The expression of VEGF in VEGF positive plasmid group decreased significantly (P<0.05), VEGF positive group had more tissue necrosis, tumor growth was significantly inhibited (P<0.05).ConcIusion siRNA-VEGF in tumor injection liposome encapsulated in malignant melanoma has a role in the radiation sensitization, which provides an experimental basis for the clinical development of targeted therapy combined with radiotherapy for the treatment of VEGF gene.
ABSTRACT
Purpose: To investigate therapeutic methods and results of comprehensive convalescent method of acupoint-pressing, Tuina, acupoint-injection and exercise in the treatment of childhood cerebral palsy. Methods: Six hundreds and forty children under 7 years old with cerebral palsy were divided randomly into treatment and control groups, and they were both given routine acupoint-injection and functional exercise, children in treatment groups were also given Tuina and acupoint-pressing therapies following acupoint-injection and functional exercise. Results: Therapeutic effect was significantly better in treatment group than that in control group (P< 0.05). Conclusion: Comprehensive convalescent method, mainly by acupointpress, Tuina, and acupoint-injection is one of premium therapy for treatment of childhood cerebral palsy, and this comprehensive convalescent therapy is really full of Chinese features and worthy of popularization.
ABSTRACT
Objective The purpose of this study was to investigate the effects of interleukin 8 (IL 8) and interferon (IFN) alpha on proliferation of ectopic endometrial cells in vitro . Methods Ectopic endometrial cells from women suffering from endometriosis were cultured, purified, and identified by immunocytochemistry. Cultured endometrial cells were treated with IL 8 and IFN ? 2b in various concentrations in 96 well plates (IL 8 in 0 1ng/ml, 1ng/ml, 5ng/ml, 10ng/ml, 50ng/ml, IFN ? in 10U/ml, 50U/ml, 100U/ml, 1 000U/ml, 10 000U/ml). The effects of IL 8 and IFN ? 2b on endometrial cell proliferation were assessed using MTT. The ultrastructural changes in endometrial cells, which were incubated in medium containing 1ng/ml of IL 8, were observed with transmission electron microscopy. Results The data suggested that there was a dose dependent stimulatory effect of IL 8 on cell proliferation. IL 8 could stimulate proliferation of endometrial cells, and the most effective concentration was 10ng/ml ( P
ABSTRACT
[ Objective ] To observe the effect of relaxing operation with small needle knife and acupoint injection of Compound Osteotide Injection (COI) for suprascapular nerve compression syndrome (SNCS) . [Methods] Two hundred and twenty cases of SNCS were randomized to three groups: group A ( re = 120) was treated with small needle knife for relaxation and acupoint injection of COI, group B ( n = 50) with small needle knife for relaxation and group C ( re = 50) with acupoint injection of COI. The short-term and long-term effects were compared in the three groups. [Results] Total effective rate was 98.3% in group A, 88.0% in group B and 74.0% in group C ( P
ABSTRACT
Objective To investigate the correlation between methylation status and death-associated protein kinase 1 (DAPK1) in SiHa and Hela cell line of cervical carcinoma and intervention of DNA methyltransferase inhibitor 5-azacytidine on the expression of DAPK1 and the proliferation of the cells. Methods DAPK1 methylation status was analyzed using methylation-specific PCR methods. The expression of mRNA and protein of DAPK1 were analyzed by RT-PCR and SABC methods after the treatment with 5-azacytidine. MTT assay was used to observe the changes of proliferation activity of the cells after 5-azacytidine treatment. Results DAPK1 genes were methylated and did not express in SiHa cells in the cervical carcinoma. Its expression could be restored by 5-azacytidine. MTT assay showed 5-azacytidine could weaken the proliferation of cancerous cells. Conclusion DAPK1 methylation plays an important role in the carcinogenesis of cervical cells and can reexpress after the treatment with 5-azacytidine which also restored its inhibitory function on carcinoma.
