ABSTRACT

Diosgenin is widely distributed in many plants, such as Polygonatum sibiricum, Paris polyphylla, Dioscorea oppositifolia, Trigonella foenum-graecum, Costus speciosus, Tacca chantrieri, which has good anti-tumor activity and preferable effects on preventing atherosclerosis, protecting the heart, treating diabetes, etc. This review combed through the anti-tumor mechanisms of diosgenin encompassing lung, breast, gallbladder, liver, oral cavity, stomach, bladder, bone marrow, etc. Besides, it was discovered that diosgenin mainly exerts its effect by inhibiting tumor cell migration, suppressing tumor cell proliferation and growth, and inducing cell apoptosis. However, problems like low yield and bioavailability frequently exist in natural diosgenin. This review introduced methods such as structural modification, dosage form optimization and combination medication to improve the yield and anti-tumor activity of diosgenin. Via the summary of this paper, it is expected to provide theoretical basis for the rational exploitation and utilization of diosgenin.

ABSTRACT

Objective:To observe the effect of tetrahydroxy stilbene glycoside (TSG) on the expression of glycogen synthase kinase 3β (GSK3β), cyclic adenosine monophosphate-dependent protein kinase (PKA) and Serine/threonine phosphatase 2A(PP2A) in the brain of amyloid precursor protein/presenilin-1/Tau (APP/PS1/Tau) triple-transgenic mice dementia model. Method:A total of forty-five 8-month-old APP/PS1/Tau transgenic mice were randomly divided into model group, positive control group (Huperzine-A, 0.15 mg·kg-1), low, medium and high dose TSG groups (TSG, 0.033,0.1,0.3 g·kg-1), with 9 mice in each group, and another nine C5B7L/6J mice of the same age were selected as normal control group. After 60 days of intragastric administration, the general structure of hippocampal neurons was observed by hematoxylin-eosin (HE) staining, immunohistochemical (IHC) was used to detect the expression of PKA protein in the brain of mice in each group, the mRNA expression levels of GSK3β, PKA and PP2A were detected by real time quantitative reverse transcription polymerase chain reaction (Real-time PCR), and protein expression levels of GSK3β and PP2A were detected by Western blot. Result:Compared with the normal control group, the apoptosis level of neurons in the model group was significantly increased, the protein and mRNA expression levels of GSK3β and PKA were significantly increased (P<0.05, P<0.01), and the protein and mRNA expression levels of PP2A were significantly decreased (P<0.05, P<0.01). Compared with the model group, the apoptosis level of neurons in each treatment group was significantly down-regulated, the protein and mRNA expression levels of GSK3β and PKA were significantly down-regulated (P<0.05, P<0.01), and the protein and mRNA expression levels of PP2A were significantly increased (P<0.05, P<0.01). Conclusion:The mechanism of TSG in the treatment of Alzheimer's disease (AD) may be related to lowering the transcription and expression of GSK3β and PKA, increasing the transcription and expression of PP2A.

ABSTRACT

Rhododendron molle G. Don is first recorded in Shengnong's Herbal Classic, and its fruits, which are termed as Liuzhouzi, are often used to treat rheumatoid arthritis in Chinese folk. During our ongoing investigation to develop a safer and potential new arthritis therapy, a process for the preparation of diterpenoid fraction from Rhododendron mollefruits was established. In order to evaluate the main components and the anti-rheumatoid arthritis effect of the diterpenoid fraction, phytochemical and pharmacological experiments were used. As the result, the main components of diterpenoid fraction were identified as rhodojaponin III (1), rhodojaponin VI (2), 2-O-methylrhodojaponin (3), and 5'-β-D-glucopyranosy-loxyjasmonic acid (4). These four components constitute greater than 95% of diterpenoid fraction using area normalization method of HPLC-ELSD. The results of CIA rat experiment showed that high dose of diterpenoid fraction (0.6 mg·kg

ABSTRACT

Objective To explore the value of superb microvascular imaging (SMI) for differentiating benign and malignant breast lesions. Methods Totally 281 patients (300 lesions) confirmed with pathology were enrolled, and the data of 2D gray-scale, CDFI and SMI were analyzed retrospectively. Adler's grading was applied to classify blood flow in breast lesions, and the ability of CDFI and SMI in detecting blood flow was compared. The vascular morphology features of breast lesions were also observed with CDFI and SMI, and then were classified and compared between benign and malignant lesions. Results The detection rate of blood flow with CDFI was 78.33% (235/300), with SMI was 90.33% (271/300), and the difference was statistically significant (χ2=116.066, P<0.001). Both CDFI and SMI showed statistically significant differences in blood flow Adler grading between benign and malignant lesions (both P<0.001). The difference of Adler grades with CDFI and SMI was significant (Z=-9.722, P<0.001). The differences in vascular morphology between benign and malignant breast lesions detected with CDFI and SMI were statistically significant (both P<0.001). The benign lesions mostly presented non-vascular, linear or curvilinear and tree-like, whereas enlarged and twisted penetrating and radial vessels were found more common in malignant lesions. Conclusion SMI can increase blood flow detection of breast lesions. SMI Adler grading of blood flow and vascular morphology are helpful to identifying benign and malignant breast lesions.

Objective The relationship between osteoblast autophagy induced by wear particles and secretion of inflammatory cytokines has not been illuminated. The purpose of this study was to investigate the effect of wear particles on the secretion of TNF -α and IL-6 by autophagy of osteoblasts in aseptic loosening. Methods The mouse MC3T3-E1 osteoblasts in logarithmic growth phase were divided into 5 groups:control group(in α-MEM medium),CoCrMo particle group(100 μg/mL CoCrMo particles was added in α-MEM medium),3-MA group(10 mmol/L autophagy inhibitor 3-MA prestimulated osteoblasts in α-MEM medium,3h later 100 μg/mL CoCrMo particles were added),the expression of autophagy marker protein LC-3 in osteoblasts was detected by Western blotting, and the levels of TNF-αand IL-6 were detected by ELISA. Results Compared with the control group, the expression level of auto-phagy marker protein LC-3 was significantly increased in CoCrMo particles group(P<0.05).Compared with CoCrMo particles group, the expression level of LC-3 in 3-MA group was significantly decreased(P<0.05).Compared with control group TNF-α[(0.95± 0.00)pg/mL]and IL-6[(4.67±0.23)pg/mL], CoCrMo particles group TNF-α[(10.35±3.63)pg/mL]and IL-6[(14.27±1.07)pg/mL]secretion level was significantly increased(P<0.05),compared with CoCrMo particles group, the effects of 10 mmol/L 3-MA group to TNF-α[(13.13±5.08)(14.71±5.46)pg/mL]and IL-6[(15.31±0.88),(16.75±3.87)pg/mL]secretion were not significant, while 10 mmol/L 3-MA group TNF-α[The secretion levels of(302.61±45.63)pg/mL]and IL-6[(216.67±27.71)pg/mL]in-creased significantly(P<0.01). Conclusion The osteoblast autophagy induced by CoCrMo particles can inhibit the secretion of in -flammatory cytokines TNF-αand IL-6 in osteoblasts.

ABSTRACT

BACKGROUND:Great development has been made in the research of various repair materials and tissue-engineered bone, and different materials exhibit a strong ability to repair bone defects. OBJECTIVE:To review the research status and progress of repair materials for bone defects. METHODS:PubMed, PMC, CBM, CNKI and WanFang databases were used online to retrieve the relative articles regarding repair materials for bone defects published from June 2007 to June 2017. The key words were "biomaterials, bone repair materials, bone substitute, bone tissue engineering, bone defect, bone repair" in English and Chinese, respectively. Finally, 66 eligible articles were included, while the unrelated and repeated ones were excluded. RESULTS AND CONCLUSION:The main repair materials for bone defects include autologous bone, allogeneic bone, xenogeneic bone, demineralized bone matrix, bioceramics, metal materials, polymer materials,tissue-engineered bone and relative derived composites. Different materials have shown certain advantages and disadvantages. Clinicians should make an optimal choice in view of the characteristics of materials. Although there is a great progress in thestudies of repair materials for bone defects, further investigations are warranted on how to produce highly biomimetic materials, optimize cell adhesion and migration, precisely control the expression and release of related genes and growth factors, validate clinical safety, and provide medical evidence in clinical practice.
