ABSTRACT

In recent years, significant progress has been achieved in heart transplantation, perioperative management and application of immunosuppressants. Nevertheless, complications after heart transplantation are still main risk factors affecting the survival of recipients. Among them, acute kidney injury is a common complication during the early stage following heart transplantation. It may lead to secondary chronic kidney diseases after heart transplantation and progress into end-stage renal diseases, severely affecting the quality of life and long-term survival of recipients. Therefore, it is of significance to identify the risk factors of kidney injury after heart transplantation and deliver prompt interventions to improve clinical prognosis of heart transplant recipients. In this article, research progress on the incidence, risk factors, prevention and treatment strategies of kidney injury-related complications after heart transplantation was reviewed, aiming to provide reference for the prevention, diagnosis and treatment of kidney injury-related complications after heart transplantation and enhance clinical prognosis of recipients undergoing heart transplantation.

ABSTRACT

OBJECTIVE:To estab lish the quality standard of Tetrastigma hemsleyanum stem and leaves dispensing granules , and to determine the contents of 5 active ingredients simultaneously. METHODS : Chlorogenic acid , isoorientin and isoorientin-2′-O-rhamnoside in T. hemsleyanum stem and leaves dispensing granules were identified by TLC. The total flavonoids of the 3 batches granules were determined by UV spectrophotometry (by isoorientin ). The granule sizes ,moisture contents , dissolvability,and content uniformity were determined. Using isoorientin as internal reference ,relative correction factors of other components were established. QAMS method was adopted to determine the contents of neochlorogenic acid ,chlorogenic acid , orientin,isoorientin and isoorientin- 2′-O-rhamnoside,which were compared with the results of ESM method. RESULTS :TLC spots of chlorogenic acid ,isoorientin and isoorientin- 2′-O-rhamnoside were clear and well-separated ,without interference from negative control. The linear range of isoorientin were 7.73-61.82 μ g/mL(r=0.999 9). RSDs of precision ,stability and reproducibility tests were all lower than 2%. The recoveries were 93.75%-97.85%(RSD=1.41%,n=6). The average percentages that the 3 batches granules could not pass through sieve No. 1 - moisture contents were 4.63%,5.18% and 4.03%(n=3). were dissolved within 5 min ,and content uniformity were 4.8%-5.0%. Which were all in line with the relevant provi-sions of granules in 2015 edition of Chinese Pharmacopoeia . The linear range of 5 ingredients were 2.325-93 μg/mL(r=0.999 9),5.125-205 μg/mL(r=0.999 9),1.150- 46 μg/mL(r=0.999 3),2.625-105 μg/mL(r=0.999 9),4.725-189 μg/mL(r=0.999 9),respectively. RSD of precision ,stability and reproducibility tests were all lower than 3%. The recoveries were 99.78%-106.13%(RSD=2.33%,n=6),95.07%-103.32% (RSD=2.72%,n=6),97.17%-105.43%(RSD=2.98%,n=6),95.52%-101.33%(RSD=2.46%,n=6),99.42%-105.56% (RSD=2.34%,n=6). Using isoorientin as internal reference ,relative correction factors of neochlorogenic acid ,chlorogenic acid , isoorientin and isoorientin- 2′-O-rhamnoside were 0.731,0.805,0.821,0.590,respectively. The contents were 0.828-1.123, 2.379-3.118,0.281-0.880,1.039-1.393,2.121-3.209 mg/g by QAMS method ,while the contents were 0.803-1.099,2.345-3.085, 0.269-0.872,1.309-1.393,2.113-3.201 mg/g by ESM method ,there was no significant difference in the content determination results between QAMS and ESM method (P>0.05). CONCLUSIONS :Established quality standard is simple and rapid ,and can be used for quality control of T. hemsleyanum stem and leaves dispensing granules. Established QAMS method is accurate and efficient,and it can be used for simultaneous determination of 5 active ingredients of T. hemsleyanum stem and levels dispensing granules.

ABSTRACT

Objective:To investigate the effect of different cleaning methods on the cleaning effect of Da Vinci robotic electrosurgical instruments and the feasibility of adenosine triphosphate(ATP) fluorescence detection technology for evaluating the cleaning effect of Da Vinci robotic electrosurgical instruments.Methods:A total of 180 electrosurgical instruments were divided into control group and test group by random digits table method from June 2018 to June 2019 after robotic surgery. For the control group, 90 cases were manually cleaned to complete the device cleaning process, and for the test group, 90 cases were mechanically cleaned to complete the device cleaning process. Through visual observation, magnifying glass examination and ATP fluorescence detection technology, the cleaning effect of the two methods and the correlation between the evaluation results obtained by ATP fluorescence detection technique and traditional inspection method were compared.Results:The qualified rate of visual observation, magnifying lens examination and ATP fluorescence detection in the control group were 95.56%(85/90), 86.67% (78/90) and 84.44% (76/90), respectively, and that in the test group were 100.00% (90/90), 97.78% (88/90) and 98.89% (89/90), respectively. The test group was higher than the control group. The difference between the two groups was statistically significant ( χ2 values were 4.310, 7.745, 12.291, P<0.05 or 0.01). In the control group, there was a positive correlation between the qualified rate of ATP fluorescence detection and visual observation ( r value was 0.374, P < 0.01), and a significant positive correlation between the qualified rate of magnifying lens examination ( r value was 0.538, P < 0.01). In the test group, There was also a significant positive correlation between the qualified rate of ATP fluorescence detection and visual observation ( r value was 0.754, P < 0.01), and between the qualified rate of magnifying lens examination( r value was 0.556, P < 0.01). Conclusions:Mechanical cleaning is used to clean Da Vinci robotic electrosurgical instruments, which is superior to traditional manual cleaning. ATP fluorescence detection technology is feasible for evaluating the cleaning effect of Da Vinci robotic electrosurgical instruments.

ABSTRACT

Objective To investigate the feasibility of sodium glycididazole(CMNa)on the radio-sensitization of esophageal cancer(EC)by 18F-fluoromisonidazole(FMISO)PET/CT.Methods A total of 60 patients with EC from December 2016 to December 2017 were enrolled prospectively and were divided into control group(n = 30;21 males,9 females;age:(56.6±10.0)years)and experimental group(n=30;20 males,10 females;age:(59.3±9.0)years)using completely randomized grouping design.Patients in the control group received radiotherapy alone,and those in the experimental group were treated with conventional radiotherapy and CMNa.All patients underwent 18F-FMISO PET/CT imaging 1 week before and after radiotherapy.The imaging results were visually and semi-quantitatively analyzed.The maximum standardized uptake value(SUVmax),tumor/muscle ratio(T/M),and hypoxia volume(HV)were calculated.Paired t test,two-sample t test and/x2 test were used to analyze the data.Results T/M and HV in the control group before and after radiotherapy were 3.92±0.57 vs 1.66±0.35,(1.84±0.31)vs(1.04±0.15)mm3,respectively;T/M and HV in the experimental group before and after radiotherapy were 4.01±0.68 vs 1.27±0.11,(2.01±0.22)vs(0.90±0.09)mm3,respectively.The primary tumor T/M,HV after radiotherapy in 2 groups were significantly lower than those before radiotherapy(t values:12.15-24.43,all P<0.05)and the amplitudes of T/M and HV in the experimental group were significantly higher than those in the control group(?T/M:2.77±0.60vs 2.25±0.49,?HV:(1.12±0.18)vs(0.81±0.26)mm3;t values:3.00 and 1.80,both P<0.05).Meanwhile,the local control rate of EC after 3 months in the experimental group was higher than that in the control group(80.0%(24/30)vs 53.3%(16/30);x2=4.80,P<0.05).Conclusion CMNa has the radiosensitizing effect on EC and the 18F-FMISO PET/CT can evaluate the radiosensitization effect.

ABSTRACT

Objective To investigate the correlation of spinal mechanical imbalance and thoraco-dorsal pain of ankylosing spondylitis (AS). Methods The clinical data of 90 patients with AS were collected. Patients were divided into two groups according to the presence of thoracodorsal pain: the AS with thoraco-dorsal pain group (30 cases) and the AS without thoraco-dorsal pain group (60 cases). Clinical symptoms, Bath ankylosing spondylitis disease activity index (BASDAI), Bath ankylosing spondylitis function index (BASFI), Bath ankylosing spondylitis measurement index (BASMI), ankylosing spondylitis disease activity (ASDAS), and spinal mechanical function and nuclear myocardial force test were compared using t-test, one-way analysis of variance (ANOVA) analysis and Spearman correlation analysis. Results ① There were differences between thoraco-dorsal pain group and patients without thoracodorsal pain group at the time of back muscle strength [(0.82±0.41) min vs (1.33±0.74) min, F=12.372, P=0.001]; ②Thoraco-dorsal pain in the AS group was mainly the middle and lower thoracic vertebrae, such as the inflammation of rib head and rib transverse process, facial arthritis, and spinous ligaments, etc. And the missed diagnosis rate of magnetic resonance imagin (MRI) was high. ③ In healthy control group, the anterior flexion strength of thoracodorsal pain group was signific-antly different from that of patients without thoracodorsal pain [(92.1 ±46.3) Nm vs (126.6±35.7) Nm, F=6.440, P=0.002]. ④ There was significant difference in spinal strength as well as left and right rotation strength between the thoracodorsal pain group and patients without thoracodorsal pain [(1.18 ±0.22) vs (1.05 ±0.17), F=10.044, P<0.01];⑤In the thoraco-dorsal pain group, the right/left index was related to BASDAI (r=-0.522, P=0.004). For spinal mobility, the right/left index was related to cross cutting faces to right ( r=0.435, P=0.021), cross cutting faces to left (r=0.528, P=0.004). In spinal strength, the right/left index was related to left turn (r=0.57, P=0.001); right lateral flexion (r=0.368, P=0.049) and left lateral flexion (r=0.369, P=0.049). Conclusion The thoracodorsal pain of AS is dominated by the middle and lower thoracic vertebrae, and the missed diagnosis rate of MRI is high. The imbalance of the left and right side of the spine is one of the factors of the thoracic back pain in AS.

ABSTRACT

Objective To investigate the effect of irisquinone (IR) on the radiosensitization of MDA-MB-231 breast cancer in nude mice using 18F-fluoroerythronitroimidazole (18F-FETNIM) microPET/ CT.Methods Thirty-two nude mice bearing MDA-MB-231 breast cancer were divided into 4 groups by random number table method (n =8 for each group):group A was treated with radiotherapy alone,group B was treated with radiotherapy and IR,group C was treated with IR alone,and group D was fed with distilled water.18F-FETNIM microPET/CT images were obtained to monitor the maximum standardized uptake value (SUVmax) of tumor before radiotherapy and 24 h after radiotherapy.The mice were sacrificed and tumor tissues were removed for HE staining.The expression of hypoxia inducible factor-1α (HIF-1α) was detected by immunohistochemical (IHC) method.Data were analyzed by paired t test,one-way analysis of variance,the least significant different t test and Pearson correlation analysis.Results There was no significant difference in SUVmax among the 4 groups before radiotherapy (1.429±0.090,1.430±0.076,1.445±0.071,1.432± 0.074;F=0.072,P>0.05).At 24 h after radiotherapy,the SUVmax of group A and B decreased significantly (1.075±0.098,0.890±0.076;t values:12.888,33.217,both P<0.05),and there was significant difference between group B and group A (t =4.197,P<0.05).However,the SUVmax of group C and D increased significantly (1.617±0.090,1.644±0.063;t values:-11.009,-16.061,both P<0.05).Pathological results showed that tumor cells in group A and B were reduced.IHC results showed that the expression of HIF-1α was lower in group B than others ((26.75±7.19)%;F=46.745,t values:2.898-9.743,all P< 0.05).The expression of HIF-1α was positively correlated with SUVmax(r =0.89,P<0.05).Conclusion 18F-FETNIM microPET/CT could evaluate the radiosensitization effect of IR on MDA-MB-231 breast cancer in nude mice.

ABSTRACT

Objective@#To determine the influence of donor-recipient sex matching on outcome of haploidentical hematopoietic stem cell transplantation (haplo-HSCT) for acute leukemia in the setting of T-cell-replete transplants.@*Methods@#The retrospective study is based on 1 160 consecutive patients who received their first haplo-HSCT for acute leukemia between April 2002 and December 2014 at Peking University Institute of Hematology. The patients were divided into the sex-matched group and sex-mismatched group in terms of the recipient and donor sex. Then we conducted an analysis in four subgroups, male patients with male donors (M→M), male patients with female donors (F→M), female patients with female donors (F→F), and female patients with male donors (M→F). Transplant outcomes were measured in terms of hematopoietic recovery, acute graft versus host disease (aGVHD), relapse, nonrelapse mortality (NRM), disease-free survival (DFS) and overall survival (OS) in the above four subgroups. Then univariate and multivariate analysis were conducted.@*Results@#There was a higher 3-year and 5-year NRM but no difference in other transplant outcomes in sex-mismatched group when compared with the sex-matched group. F→M was compared with M→M, and the former group had higher 3-year and 5-year cumulative incidences of NRM (25.5% vs 16.1%, P=0.002; 27.1% vs 17.3%, P=0.002), decreased 5-year DFS (56.9% vs 64.4%, P=0.044), decreased 3-year OS (62.6% vs 69.8%, P=0.045). There was no significant difference in 3-year DFS and 5-year OS. There was no significant difference in grade Ⅱ-Ⅳ aGVHD and cGVHD incidence. When F→F group was compared with M→F group, only a higher grade Ⅱ-Ⅳ aGVHD incidence (43.9% vs 34.6%, P=0.047) existed. F→M was proved to be the independent risk factor influencing NRM and OS in multivariate analysis.@*Conclusion@#In haplo-HSCT for acute leukemia, the donor-recipient sex combination of male patients with female donors was of a poorer prognosis, so a male donor was a better choice for a male patient.

ABSTRACT

Objective To study the radiosensitization of irisquinone on the Warburg effect of MDA-MB231 cells.Methods MDA-MB231 cells in the exponential growth phase were divided into 6 groups:control group,irisquinone group,radiation group,irisquinone plus radiation group(Irisquinone + RA),negative control group,and experimental group (siRNA).Colony formation assay was used to measure cell survival fraction of MDA-MB231 cells.Single-hit multi-target model was used to fit the survival curve and calculate the sensitive enhancement ratio (SER).Flow cytometry was used to measure cell apoptosis.The relative expression levels of HK Ⅱ mRNA and protein in each group were detected by qRT-PCR and Western blot respectively.Results The values of D0,Dq and SF2 in the irisquinone plus radiation group were obviously lower than those in the radiation group.The SER of irisquinone was 1.52.Compared with the other groups,the cell apoptosis rate was increased (t =13.29,12.09,5.90,3.83,P < 0.05),while the relative expression levels of HK Ⅱ mRNA (t =9.14,10.48,3.40,P<0.05) and protein (t=13.39,16.08,5.81,P < 0.05) were decreased in the irisquinone plus radiation group significantly.Conclusions Radiosensitization function of irisquinone inhibits the Warburg effect of MDA-MB231 cells by down-regulating the expression of HK Ⅱ.

ABSTRACT

Objective To evaluate the effectiveness of a rat model for comorbidity of Tourette syndrome and anxiety with empty water bottle stimulation plus iminoodipropionitrile(IDPN) injection.Methods The 48 male SD rats were randomly divided into four groups:the blank control group,the TS group,the anxiety group and the comorbidity group.The blank control group was injected with saline for 7 days.The TS groop was injected with 3,3-iminodipropionitrile (IDPN) with 250 mg/kg once a day for 7 consecutive days.The anxiety group was given empty water bottle stimulation for 21 consecutive days.The comorbidity group was given empty water bottle stimulation plus IDPN injection.At the end of the 3rd week,the behavioral changes of the stereotyped movement,elevated plus-maze and open field of the rats in each group were measured,and the contents of monoamine neurotransmitters in striatum and hippocampus were determined by HPLC.Results The results of stereotyped movement showed that there was no significant difference between the groups except for the blank control group.The elevated plus-maze test showed that the 0E/TE values of the comorbidity group (21.33±11.35) % and the anxiety group (17.68±16.89) % were significantly decreased,lower than that of the blank control group (73.24± 19.33) % and TS group(61.43±21.84) %.The results of open field test showed that the total scores of open field in the comorbidity group(15.22±9.87)and anxiety group (11.17±10.76) were lower than that of the blank control group (41.86±33.30) and TS group(48.83± 17.65) (P<0.01).However,there was no significant difference between the comorbidity group and the anxiety group.The test of monoamine neurotransmitters in striatum showed that the content of HIAA in the comorbidity group(0.03±0.00) ng/mg was the highest,and that of the TS group and anxiety group (0.02±0.00) ng/mg was higher than that of the blank control group (0.01±0.00) ng/mg (P<0.01).The DA test showed that the content of DA in the comorbidity group (0.03±0.00) ng/mg was the highest,and that of the comorbidity group,TS group(0.02±0.00) ng/mg and anxiety group was higher than that of the blank control group(0.01±0.00) ng/mg (P<0.01).The expression of 5-HT was most significant among the groups (P<0.01),and there was significant difference between the anxiety group ((0.011 ± 0.001) ng/mg)and the comorbidity group ((0.014±0.002) ng/mg) (P<0.01).The expression of HVA in the three model groups ((0.05±0.00) ng/mg) was higher than that in the blank group ((0.02±0.00) ng/mg) (P< 0.01).The expression of DOPAC in the TS group ((0.23±0.02) ng/mg) was higher than that in the blank control group((0.16±0.01) ng/mg) and comorbidity group ((0.16±0.02) ng/mg) (P<0.01).The test of monoamine neurotransmitters in hippocampus showed that the content of 5-HT in the comorbidity group ((0.14±0.02) ng/mg) was the highest,followed by the anxiety group ((0.1 ± 0.03) ng/mg) and the TS group ((0.07±0.04) ng/mg),which were all higher than the blank control group((0.04±0.03) ng/mg) (P<0.05,P<0.01),and there were significant differences between the comorbidity group and the TS group or anxiety group (P<0.01).The expressions of HIAA and HVA were higher in the comorbidity group((0.44±0.04)ng/mg,(0.01±0.00) ng/mg),TS group ((0.46±0.15) ng/mg,(0.01 ±0.01) ng/mg) and anxiety group ((0.46±0.08)ng/mg,(0.01±0.00) ng/mg) than that in the blank control group((0.21±0.10)ng/mg,(0±0) ng/mg) (P<0.05,P<0.01).Conclusion This study confirms the reliability of the model and it is an ideal animal model for the study of TS with comorbidity of anxiety,which can be used for follow-up research.

ABSTRACT

Objective To investigate APP mobile information technology innovation methods in preoperative visits to patients in the clinical application effect. Methods A total of 840 patients undergoing elective surgery were divided into control group and experimental group by random digits table method with 420 cases each and control group by adopting the method of the nurse oral interpretation of the preoperative visit list and health education leaflets oral interpretation information, experimental group using mobile medical technology innovation preoperative visit method, combining with specialized subject characteristic in APP software design way of cartoon animation and video surgery in patients with preoperative visit. The comparison of two groups of patients with the patients know about the operation, cooperate with surgery, perioperative anxiety levels, hospitalized experience satisfaction four dimensions of experience were performed. Results The patients know about the operation was (7.6 ± 2.3) points in control group and(8.5 ± 2.5)points in experimental group, and there was significant difference between two groups (t=5.430,P<0.01). The cooperate with surgery was 92.62%(389/420) in experimental group and 75.71%(318/420) in control group, and there was significant difference between two groups(χ2=45.032, P<0.01). The perioperative anxiety levels was (38.27 ± 2.50) points in experimental group and (47.21 ± 3.84) points in control group, and there was significant difference between two groups (t=8.420, P<0.01). The hospitalized experience satisfaction was (96.41±2.30) points in experimental group and (93.70±1.51)points in control group, and there was significant difference between two groups (t=3.297, P <0.01). Conclusions Using APP for clinical preoperative visit mobile information technology, which can effectively improve the patients know about the operation and the matching degree, reduce the perioperative patients with anxiety, improve surgical patients during hospitalization medical treatment satisfaction.

This paper summarized nursing points for caring 23 cases undergoing pediatric heart transplantation,including:characteristics of accessing and protection of hearts,collaboration in pediatric heart transplantation,nursing of capacity management,temperature controlling,extracorporeal membrane oxygenation and delayed sternal closure during surgeries.The average length of hospitalization for 23 cases was 24.5±8.3 days,1 case died from primary graft failure after 19 months,the rest of 22 cases survived,and cardiac function recovered to NYHA class Ⅰ-Ⅱ.

ABSTRACT

Objective To evaluate the feasibility of 18F-FDG PET/CT in assessing the radiosensitizing effect of oleanolic acid (OA) in C6 rat glioma model,and to explore the possible mechanism of radiosensitizing effect of OA.Methods Thirty-two C6 glioma-bearing SD rats were divided into 4 groups by random number table:group A without OA and radiotherapy,group B with OA alone,group C with radiotherapy alone,group D with OA and radiotherapy.18F-FDG PET/CT was performed before radiotherapy,at 24 h and 7 d after radiotherapy.The tumor SUVmax was measured.All rats were sacrificed and tumor tissues were excised for HE and immunohistochemistry staining.Paired t test,one-way analysis of variance,LSD-t test and Spearman correlation analysis were performed to analyze the data using SPSS 13.0.Results There was no statistically significant difference in SUVmax among the 4 groups (SUVmax of groups A,B,C,D:5.252± 0.536,5.261±0.544,5.273±0.520,5.232±0.507) before treatment (F=0.008,P>0.05).At 24 h postradiotherapy,SUVmax of groups C and D (4.766±0.511 and 4.403 ±0.486) decreased significantly (t =14..788,13.366,both P<0.05);while groups A and B (5.680±0.635 and 5.763±0.689) increased significantly (t =-11.578,-8.651,both P<0.05;F=10.550,P<0.05).However,there was no statistically significant difference between groups C and D (t =1.453,P>0.05).At 7 d post-radiotherapy,SUVmax of groups C and D decreased significantly (t =9.750,10.530,both P<0.05);while SUVmax of groups A and B (t=-35.353,-6.884,both P<0.05) increased significantly (F=97.691,P>0.05).SUVmax of Group D decreased significantly compared with that of group C (t =5.329,P>0.05).Less tumor cells and more areas of necrosis were observed in group D compared with those in other groups by HE staining.The expression of HIF-1α was lower in group D than that in other groups by immunohistochemistry staining.HIF-1α expression was positively correlated with SUVmax(r=0.853,P<0.05).Conclusion 18F-FDG PET/CT has potential to evaluate the radiosensitizing effect of OA in vivo,and the radiosensitization of OA might be related to the down regulation of HIF-1α.

ABSTRACT

Aim To investigate the effect of PLK1 on epithelial-mesenchymal transition (EMT)of human e-sophageal squamous cell carcinoma (ESCC)cells TE-1 5 and its relevant molecular mechanisms.Methods PLK1 overexpressed ESCC cells and control vector were used as the experimental cells.The expression of EMT-related protein markers E-cadherin and vimentin were measured by Western blot.vimentin mRNA was measured by Real-time PCR.Total cellular protein and nuclear protein were respectively extracted,and then they were used to detect the expression of β-catenin by Western blot.β-catenin siRNA and non-specific siR-NA were transiently transfected into the cell clones overexpressed PLK1 ,and then vimentin was detected by Western blot.β-catenin protein degradation com-plex was detected by immunoprecipitation and Western blot.Results The mesenchymal marker vimentin was distinctively upregulated and the epithelial marker E-cadherin was distinctively downregulated in the cell clones overexpressed PLK1 ,compared with those in the vector clones.This indicated that EMT occurred in ESCC cells.vimentin mRNA was also markedly in-creased.In the cell clones overexpressed PLK1 ,β-catenin were both elevated from the total cells and the nucleus.The expression of vimentin was reduced whenβ-catenin was knocked down.APC and GSK-3βwere both reduced from Axin immunoprecipitate in the cell clones overexpressed PLK1 .Conclusion PLK1 up-regulates vimentin and promotes EMT in ESCC cells probably by inhibiting the formation of protein degrada-tion complex and stabilizing β-catenin.

ABSTRACT

Objective To investigate the influence of RNAi gene silence on the chemosensitivity of human digestive system neo-plasms .Methods Esophageal cancer EC109 cells and gastric adenocarcinoma SGC-7901 cells were selected and divided into the transfection group and the blank control group for culturing .Two cell lines in the transfection group were transferred by siRNA and silenced the expression of Survivin ,giving cisplatin 5 mg/L to co-culture for 72 h .The blank control group was given the conven-tional culture and the same dose of cisplatin for co-culture for 72 h .The in vitro proliferation rate of cells in the two groups were measured by MTT and their sensitivities to the chemotherapy drug cisplatin were compared .Results After siRNA silencing the ex-pression of Survivin in SGC-7901 and EC109 cell lines ,the sensitivity of cells to cisplatin drugs was increased .MTT results sugges-ted that the proliferation activity of cells was significantly lower than that in the non-silence cell lines ,the difference showing statis-tical significance(P<0 .05) .Conclusion Adopting RNAi gene silencing technique can interfere the expression of Survivin gene ,in-hibit the proliferation of tumor cell ,and at the same time significantly improve the sensitivity of tumors to chemotherapy .

ABSTRACT

Objective To explore the infection rate,the correlated risk factors and knowledge of Helicobacter Pylori infection among the outpatients with 13C-urea breath test (13C-UBT).Methods 13C-UBT was performed to evaluate the HP infection status.Self-designed questionnaires were adopted to conduct investigation among 500 outpatients with 13C-UBT and then the results were analyzed.Results The HP infection rate was 46% in our research.Demographic factors of ages in different groups,on the job or not,HP testing times and different education degree as well as having susceptible factors of family history of HP infection or not,using the public chopsticks or not,sitting up late,history of digestive disease were statistically significant.Positive HP infection was statistically significant different in people with different knowledge of HP infection.71.8% of the patients had never heard of the HP could cause none digestive system diseases,61.4% of the patients didn't know how to use drug combination,64.6% of patients didn't know the review time and 53.4% of patients did not know how to prevent and control HP infection.Conclusions There is high HP infection rate in outpatients receiving 13C-UBT.The infectious sources of HP commonly exist in population,and the route of transmission was various.We should focus on the susceptible people and strengthen health education,advocate eating separately,improve sanitary consciousness,add HP-test in the health test and enhance the reasonable intervention about HP infection.

ABSTRACT

Objective To analyze the expression of IFIT4,PRKR and investigate the clinical and immunology features of different types of liver involvement in patients with systemic lupus erythematosus (SLE).Methods Clinical data of 62 cases of SLE with liver damage and 62 cases of SLE without liver damage were collected.Peripheral venous blood samples were obtained and total RNA were extracted and transcribed into cDNA.Sybr green dye based real-time quantitative PCR method was used to compare the expression levels of IFIT4,PRKR in patients with SLE.Clinical parameters were analyzed by ANOVA,Chi-square test,Pearson's or Spearman's test.Results ① The increase of γ-GT or ALP was correlated with rash and oral ulcer (x2=5.625,P=0.018),lupus nephritis (x2=5.631,P=0.019),anemia,thrombocytopenia (99±21,P=0.028; 81±45,P=0.004,),CRP (33±43,P=0.004).The positive rate of nRNP (x2=4.862,P=0.027 ) and SSA (x2=8.087,P=0.004) was higher in patients with liver damage than other groups; ② The positive rate of anti-Rib antibody in SLE with liver damage was significantly higher than SLE without liver damage (x2=19.542,P=0.000); ③ There was no difference in the expression of IFIT4 among these groups,but higher expression of PRKR was detected in the group of patients with increased γ-glutamyl transpeptidase (γ-GT) or ALP(F=3.54,P=0.018).Conclusion The different types of liver damage in SLE patients have different clinical and immunology characteristics.The expression of PRKR is higher in patients with increased γ-GT or ALP.

ABSTRACT

ObjectiveTo investigate the correlation of the serum and synovial fluid interleukin (IL)-1β,IL-6,tumor necrosis factor(TNF)-α and bone marrow edema with osteoarthritis of the knee (KOA).MethodsThe clinical data of 331 KOA patients were included and all patients had knee MRI.Bone marrow edema were detected in 172 cases and 159 cases had non-bone marrow edema.The clinical symptoms,WOMAC score,and the serum and synovial fluid IL-1β,IL-6,TNF-α levels were compared using One-way ANOVA analysis and Spearman's correlation analysis.Results① The serum and synovial fluid IL-1β,IL-6,TNF-α in osteoarthritis was positively correlated(serum r=0.24,0.38,0.31; synovial fluid r=0.20,0.29,0.33 ; all P<0.05) ; ② The serum and synovial fluid IL-6 and TNF-α levels of osteoarthritis with bone marrow edema were significantly higher than those of the osteoarthritis without bone marrow edema(serum F=8.139,7.172; synovial fluid F=9.201,7.001; all P<0.05); ③ The serum and synovial fluid TNF-α,IL-6 levels was associated with osteoarthritis with bone marrow edema in volume (serum r,=0.27,0.26; synovial fluid rs=0.29,0.32; all P<0.05) and severity(serum rs=0.29,0.27; synovial fluid rs=0.29,0.31; all P<0.05); ④ The serum and synovial fluid IL-1β,IL-6,TNF-α of osteoarthritis with synovitis was significantly higher than that of osteoarthritis without synovitis group (group of bone edema:serum F=3.931,5.866,5.514; synovial fluid F=4.211,5.202,5.972; all P<0.05.non-bone edema patients:serum F=3.513,3.114,7.112; synovial fluid F=3.722,3.965,8.891; all P<0.05).ConclusionIL-1β,IL-6,TNF-α are elevated in osteoarthritis with synov-itis.IL-6 and TNF-α are elevated significantly in knee osteoarthritis with bone marrow edema in particular.

ABSTRACT

Objective To investigate the expression of the double-stranded RNA-dependent protein kinase (PKR) gene in the peripheral blood leukocyte of patients with systemic lupus erythematosus (SLE),and to evaluate the relationship between the gene expression and the disease activity.Methods The clinical data of 100 SLE patients,40 non-SLE patients with rheumatic diseases,and 40 normal controls were collected.Total RNA was extracted from the peripheral blood and then reverse transcribed into cDNA.Sybr green dye based real-time quantitative PCR method was used to compare the expression levels (indicated as 2-△Ct value) of PKR in the three groups.Results (1) The 2-△Ct value of PKR expression level in the SLE patients was (14.69 ± 7.62),which was significantly higher than those in the non-SLE patients (5.09 ±4.73,P =0.012) and normal controls (4.79 ± 3.49,P =0.005).(2) The 2-△Ct value of PKR expression level in the SLE patients with severe activity was (22.57 ±2.61),which was significantly higher than those in the SLE patients with mild activity and no activity(12.94±2.41,P =0.000 ;8.85 ± 2.17,P =0.000).(3) The 2 △Ct value of PKR expression level in the SLE patients with lupus nephritis was significantly higher than that in the SLE patients without lupus nephritis (16.85 ± 7.32 vs 8.35 ± 2.04,P =0.034).(4) The 2-△Ct value of PKR was correlated with the systemic lupus erythematosus index(SLEDAI) scores(r =0.32,P =0.000),WBC (r =0.46,P =0.000),Hb (r =-0.22,P =0.035),the quantitation of urine protein in 24 hours (r =0.21,P =0.000),HDL-C (r =0.21,P =0.022),and anti-RNP antibody (rs =-0.21,P =0.025).Conclusions The expression of PKR in the SLE patients is up-regulated,especially in those with severe activity.The expression level of PKR gene is associated with SLE disease activity.

ABSTRACT

ObjectiveTo identify the status of quality of life in breast cancer patients and explore the impact of environmental factors on quality of life in breast cancer patients receiving chemotherapy. MethodsTotally 201 primary breast cancer patients undergoing chemotherapy after surgery were investigated.Self-developed demographic and clinical character investigation questionnaire,WHO Disability Assessment Schedule Ⅱ(WHODAS Ⅱ),and environmental factors in International Classification of Functioning Disability and Health(ICF)core sets for breast cancer patients were adopted to collect relevant data. ResultsBreast cancer patients had different levels of difficulties on each domain of WHODAS Ⅱ.Multiple stepwise regression analysis demonstrated that“material and emotional support and interrelationship with friends”, “social security service,system and policies” and“social norms,practices and ideologies” were factors influencing quality of life of breast cancer patients. ConclusionsIn addition to the strengthen of rehabilitation exercise training,nurses should advocate more services from society through policy making,prompt public awareness and scientific cognition,as well as construct effective nursing interventions to help enlarge social support resources and improve social support level.
