ABSTRACT
Transketolase (EC 2.2.1.1, TK) is a thiamine diphosphate-dependent enzyme that catalyzes the transfer of a two-carbon hydroxyacetyl unit with reversible C-C bond cleavage and formation. It is widely used in the production of chemicals, drug precursors, and asymmetric synthesis by cascade enzyme catalysis. In this paper, the activity of transketolase TKTA from Escherichia coli K12 on non-phosphorylated substrates was enhanced through site-directed saturation mutation and combined mutation. On this basis, the synthesis of tartaric semialdehyde was explored. The results showed that the optimal reaction temperature and pH of TKTA_M (R358I/H461S/R520Q) were 32 ℃ and 7.0, respectively. The specific activity on d-glyceraldehyde was (6.57±0.14) U/mg, which was 9.25 times higher than that of the wild type ((0.71±0.02) U/mg). Based on the characterization of TKTA_M, tartaric acid semialdehyde was synthesized with 50 mmol/L 5-keto-d-gluconate and 50 mmol/L non-phosphorylated ethanolaldehyde. The final yield of tartaric acid semialdehyde was 3.71 g with a molar conversion rate of 55.34%. Hence, the results may facilitate the preparation of l-(+)-tartaric acid from biomass, and provide an example for transketolase-catalyzed non-phosphorylated substrates.

OBJECTIVE:To systematically evaluate the efficacy and safety of Xianling gubao capsule combined with glucosamine in the treatment of knee osteoarthritis (KOA),and to provide evidence-based reference for clinical treatment of KOA. METHODS:Retrieved from Cochrane library ,PubMed,Embase,CNKI,Wanfang database ,CBM and VIP during the inception to Nov. 2020,randomized controlled trials (RCTs)about glucosamine (control group )combined with Xianling gubao capsule (trial group)in the treatment of KOA were collected. After data extraction ,Cochrane 5.1.0 system evaluator manual was used to evaluate the quality of the included clinical studies met inclusion criteria ,and Rev Man 5.4 software was used for Meta-analysis. RESULTS : A total of 11 RCTs with 970 patients were included in this study. Results of Meta-analysis showed that cure rate [OR =2.41,95%CI (1.48,3.93),P<0.001],marked effective rate [OR =1.95,95%CI(1.49,2.56),P<0.001],total effective rate [OR =4.00,95%CI (2.57,6.24),P<0.001],visual analogue scale (VAS)score [MD =-2.34,95%CI(-2.51,-2.17),P<0.001],knee function score [MD =31.32,95%CI(27.89,34.75),P<0.001],Japanese Orthopaedic Association (JOA)low back pain score [MD = 12.22,95%CI(9.68,14.76),P<0.001] and pain relief time [MD =-1.55,95%CI(-1.84,-1.25),P<0.001] of trial group were all significantly better than those of control group. The incidence of ADR in trial group was significantly lower than control group [OR =0.04,95%CI(0.02,0.12),P<0.001]. CONCLUSIONS :Xianing gubao capsule combined with glucosamine in the treatment of KOA is significantly better in cure rate ,marked effective rate ,total effective rate ,VAS,score,knee function score,JOA low back pain score and pain relief time ,and could significantly reduce the incidence of ADR.

Objective:To investigate the effect of Chinese reading aloud training (c-RAT) on cognitive function and activities of daily living in patients with vascular cognitive impairment no dementia (VCIND).Methods:From January 2018 to January 2019, totally 80 patients with VCIND in the General Hospital of Xingtai Mining Group were enrolled.Subjects were randomized grouped into c-RAT group ( n=43) and control group ( n=37). The c-RAT group was asked to read aloud the Chinese paper for 5 days a week for 12 weeks.At the time of enrollment and 12 weeks after the intervention, the Montreal cognitive assessment (MoCA), trail making test (TMT), auditory verb learn test (AVLT), digit symbol substitution test (DSST) and modified Barthel index (MBI) were filled out. Results:Compared with control group, the difference of D-value in c-RAT group was statistically significant in MoCA total scores(4.00(2.00), 1.50(0.50), Z=3.012, P=0.003), scores of MoCA-Visual space execution (2.00(1.00), 0.00(2.00), Z=2.787, P=0.008), MoCA-attention (1.00(1.00), 0.00(0.50), Z=2.369, P=0.022), MoCA-language (1.00(1.00), 0.00(0.75), Z=3.049, P=0.000)and MoCA-delayed recall(2.00(2.00), 0.00(1.00), Z=2.043, P=0.014), TMT-A scores (-8.00(23.00), 10.50(30.25), Z=2.120, P=0.039), AVLT scores (1earning)(3.00(2.00), 0.50(0.75), Z=2.266, P=0.039) , AVLT scores (recall)(2.00(1.00), 0.00(1.00), Z=2.974, P=0.003)、AVLT scores (recognition) (2.00(0.00), 0.50(1.50), Z=3.054, P=0.000)and DSST scores ((4.96±0.71), (2.39±0.78), t=2.572, P=0.014), while there were no significant differences in the rest parts of MoCA, TMT-B and MBI( P>0.05). The increased scores in DSST were positively correlated ( r=0.205, P=0.006) with the number of reading aloud tasks finished. Conclusion:C-RAT can improve general cognition, especially in information processing speed, executive function, attention and auditory memory.

Objective To explore the effect of the combination of bandage contact lenses and ar-tificial tears on corneal epithelial injury , due to correction of blepharoptosis in patients .Methods Thirty-five patients (56 eyes) were included in this randomized controlled trial during May 2013 to May 2018 ,and randomly divided into observation group (combination of bandage contact lenses and artificial tears) and control group (combination of erythromycin eye ointment and artificial tears ) ,and followed up for 3 months .The corneal wound healing ,corneal fluorescent staining (FL ) ,Schirmer test (Slt) ,break-up time (BUT ) and the severity of eye pain were documented and compared by sta-tistical method .Results The scores of FL in the observation group were reduced (t = - 3 .211 , P <0 .05 ;t = - 3 .312 , P < 0 .05) ; the scores of eye pain in the observation group were reduced (t =- 7 .105 ,P < 0 .05 ; t = - 6 .543 , P < 0 .05) 7 days and 1 months postoperatively as compared with controls .The corneal wound healing of the observation group was improved during the same time peri-od (χ2 = 10 .286 ,P < 0 .05 ;χ2 = 4 .918 ,P < 0 .05) ,The BUT score of the observation group were im-proved (t = 3 .763 ,P < 0 .05 ;t = 3 .833 ,P < 0 .05) .However ,all above differences failed to reach sta-tistical significance 3 months postoperatively (the scores of FL ,t = 0 .328 ,P > 0 .05 ;the scores of eye pain ,t = - 1 .800 , P > 0 .05 ;the corneal wound healing ,χ2 = 1 .018 , P > 0 .05 ;the BUT score ,t =1 .661 ,P > 0 .05) .Conclusions The combination of bandage contact lenses and artificial tears is rec-ommended for the early use after correction of blepharoptosis ,to alleviate eye symptoms and promote corneal wound healing .

Background Pregnancy-induced hypertension syndrome (PIHS) appears retinopathy.Optical coherence tomography (OCT) and contrast sensitivity (CS) has been widely used in ophthalmology, but the application of OCT and CS in PIHS fundus lesions is less reported.Objective This study was to assess fundus changes of PHIS by using OCT and CS.Methods Two hundred and thirty-three eyes of 116 PIHS patients with fundus diseases were enrolled from January 2011 to January 2014 in the Cangzhou Central Hospital.One hundred eyes of fifty normal pregnant women were collected at the same period.Best corrected visual acuity (BCVA), fundus examination, OCT, CS were performed on these subjects.The CS values and BCVA between normal pregnant woment and PIHS patients were compared by independent samples t test.The correlation between BCVA grade and CS grade, OCT grade and CS grade, OCT grade and BCVA grade was calculated by trend x2 test and Gamma rank correlation analysis.The study was approved by the Ethics Committee of Cangzhou Central Hospital, all the subjects signed informed consent.Results OCT results showed that 172 eyes were abnormal (65.15%) ,including 94 eyes with neurosensory serous retinal detachment (54.65%) ,40 eyes with pigment epithelium and the ellipsoid zone change (23.26%) and 38 eyes with optic disc edema,retinal hemorrhage and other changes (22.09%).The CS values in each spatial frequency and the BCVA in the PIHS group were significantly lower than those in the normal control group (all at P < 0.05).The OCT examination result was positively correlated with BCVA in the PIHS group (G=0.807,x2=93.418,both at P =0.000).The CS value under the 6.0 c/d had the best correlation with BCVA (G=0.820,x2=97.397, both at P=0.000).The OCT examination result was positively correlated with CS under the 6.0 c/d(G=0.932,x2 =126.376,both at P=0.000).Conclusions Combination of fundus examination,OCT and CS can objectively and comprehensively assess the morphology and function of retinopathy in PIHS patients,which provide the basis for the clinical diagnosis and timely treatment.
ABSTRACT
Objective To explore effect of rehabilitation training on deglutition disorders of children with cerebral palsy . Methods Twenty-seven children patients from January to June in 2013 were set as control group and thirty-one patients from July 2013 to January 2014 as experiment group. The children in the control group were treated with tube-feeding combined with spoon feeding and bottle-feeding by professional nurse. Children in the experiment group were treated with oral rehabilitation training by professional therapists and nurses apart from the same feeding as in the control group. The two groups were compared in terms of effect of deglutition within 4 weeks, time and rate of removing the stomach tube. Results The recovery of deglutition function in the experiment group was much better than that in the control group , the total effective rate and rate of removing the stomach tube within 3 months were higher and the intubation duration was significantly lower as compared with those of the control group (P < 0.01). Conclusion Rehabilitation training can improve the recovery of deglutition disorders, improve active feeding ability of children with cerebral palsy, shorten time of tube feeding and improve their life quality.

To optimize the formula of levodopa microcapsules floating tablets. Methods:The contents of levodopa and benserazide in the microcapsules floating tablets were determined by HPLC simultaneously. The release rate as the index, an orthogonal design was used to optimize the formula and preparation technology of levodopa microcapsules floating tablets. The release property of the microcapsules floating tablets was evaluated. Results:The HPLC method for the in vitro determination of levodopa and benserazide in the floating tablets met the methodological requirements. The selected formula was as following:the amount ratio of stearic acid, the drugs, acrylic resin and HPMC was 2∶5∶2∶1. The average weight of the tablets was 550 mg. The results of validated tests showed that the microcapsules floating tablets had floating and sustained release property, which could be used by divided dose. Conclusion: The optimized formula of the microcapsules floating tablets is reasonable, and the production process is stable and feasible.

Objective To study the effects of different sources of dendritic cells (DCs) on root resorption and healing after allo-geneic tooth transplantation in rats .Methods The BN rats and Lewis rats were respectively used as the donor and recipient teeth for establishing the animal experimental transplantation models and randomly divided into 4 groups ,20 cases in each group :the syn-geneic transplantation group(A) and the allogeneic transplantation groups(B ,C ,D) .The group A and B were infused with PBS buffer solution 0 .5 mL on 7 d before operation .The group C and D were infused with 1 × 106 donor or recipient tolerogenic DCs on 7 d before operation .5 rats randomly selected from each group were sacrificed for performing the pathological examinations of transplanted teeth at the end of 1 ,2 ,4 and 8 weeks .Results The inflammatory cellular infiltration was most severe in the group B , slightest in the group A and moderate in the group C and D .The root resorption was minimal in the group A ,maximal in the group B and moderate in the group C and D (P0 .05) ,but which at 8 weeks in the group D was lower than that in the group C .The periodontal membrane healing points at 8 weeks in the group C and D was less than that in the group A ,but more than that in the group B (P<0 .05);the inflam-matory absorption was highest in the group B(P<0 .05);the alternative absorption was lowest in the group A (P<0 .05) .Conclu-sion The two different sources of tolerogenic DCs all could reduce the rejection reaction of allogeneic tooth transplantation ,reduce the root resorption and promote the healing of periodontal membrane .Recipient tolerogenic DCs could reduce the root resorption in late stage even more .

To get the tolerability and consumption of Klebsiella oxytoca on major inhibitors in lignocelluloses hydrolysate, we studied the effect of acetic acid, furfural and 5-hydroxymethylfurfural on production of 2,3-butanediol by Klebsiella oxytoca. The metabolites of furfural and 5-hydroxymethylfurfural were measured. The results show that when acetic acid, furfural and 5-hydroxymethylfurfural was individually added, tolerance threshold for Klebsiella oxytoca was 30 g/L, 4 g/L and 5 g/L, respectively. Acetic acid was likely used as substrate to produce 2,3-butanediol. The yield of 2,3-butanediol increased when acetic acid concentration was lower than 30 g/L. In the fermentation, more than 70% 5-hydroxymethylfurfural was converted to 2,5-furandimethanol. All furfural and the rest of 5-hydroxymethylfurfural were metabolized by Klebsiella oxytoca. It showed that in the detoxification process of 2,3-butanediol production using lignocelluloses hydrolysate, furfural should be given priority to remove and a certain concentration of acetic acid is not need to removal.

Objective To observe the changes of serum ET-1,TNF-α and VEGF in patients with hypertensive disorder complicating pregnancy ( HDCP),explore the significance of cytokines in HDCP and the relationship between cytokines and HDCP.Methods The serum levels of ET-1,TNF-α and VEGF in 60 patients with HDCP and 30 normal pregnant women were detected by ELISA.Results The serum levels of ET-1,TNF-o in HDCP group were higher than those of the control group( all P <0.05).The serum level of VEGF in HDCP group was lower than that of the control group ( P < 0.05).Negative correlation was observed between VEGF and ET-1,TNF-α in HDCP group (P < 0.05).Conclusion The serum levels of ET-1,TNF-α increased significantly,while the serum level of VEGF decreased.ET-1,TNF-α and VEGF may play important roles in ipathogenesis and development of HDCP.

Objective To investigate the effect of folic acid on the DNA methylation of tumorrelated genes promoters in healthy human peripheral blood mononuclear cells(PBMC). Methods Ten healthy volunteers were divided into two groups, and were randomized to receive either 5 mg folic acid (n=5)or placebo(n = 5) , one time per day for 3 months. The serum folic acid concentration was detected with chemiluminescence enzyme immunoassay kit before and after the intervention. The methylation statuses of five tumor-related genes promoter, including oncogenes c-myc, c-Ha-ras,tumor suppressor genes p16INK4A, E-cadherin and mismatch repair gene hMLH1 in PBMC were detected by bisufite sequencing. Results After folic acid intervention, the level of serum folic acid increased significantly in intervention group (t= -4. 739,P<0. 05) , however no significant difference in control group. After three-month folic acid intervention, the level of methylation of oncogene c-myc promoter increased from 4%, 3. 3%, 4. 1% before intervention, one week after intervention, one month after intervention respectively to 8%(t= -4. 079,P<0. 05), while no significant change in placebo taken group. Before and after the folic acid intervention, there was no significant difference of DNA methylation of other tumor-related genes promoter, including c-Ha-ras、E-cadherin、p16INK4Aand hMLH1. Conclusion Folic acid intervention can up-regulate DNA methylation of oncogene c-myc promoter, but can not affect the promoter methylation status of tumor suppressor genes E-cadherin,p16INK4Aand hMLH1.

Objective To study the effects of extracellular-signal regulated kinase mitogenactivated protein kinase (ERK-MAPK) signaling pathway inhibition on histone phosphorylation and the related gene expression in human colorectal cancer cells.Methods Two human colorectal cancer cell lines (SW1116 and HCT116) were cultured and treated with gradient(0,20,40/μmol/L) doses of ERK-MAPK signaling pathway inhibitor U0126.Cell viability was determined by cell counting kit 8 (CCK-8) assay.Cell cycle distribution was assessed by flow cytometry.The expression levels of histone H3 kinases including ribosomal S6 serine-threonine kinase (RSK-2) and mitogen-and stressactivated protein kinase 1 and 2 (MSK1 and MSK2),and the levels of histone H3 (Ser10) phosphorylation and c-Fos protein were detected using Western blotting.Results Treatment of these two human colorectal cancer cell lines with ERK-MAPK inhibitor resulted in a time and dose-dependent inhibition of cell proliferation significantly. Proliferation rate of HCT116 was reduced to 47% at 72 hours after 40/μmol/L U0126 treatment. Cell cycle analysis showed that the percentage of phase G0/G1 cells significantly increased (P<0. 01) and the percentage of phase S cells decreased (P<0.01) after treatment with ERK-MAPK inhibitor. The expression of MSK1 and RSK2 reduced obviously in both of human colorectal cancer cell lines treated with U0126, which resulted in a 28% and 40% reduction of levels of MSK1 and RSK2 as compared with control HCT116 cells respectively,while no detectable change in the expression of MSK2 was found. Consistent with this, the expression level of histone H3 (ser10) phosphorylation was markedly down-regulated by ERK-MAPK inhibitor, and the related protein c-Fos expression decreased accordantly. Conclusions Decreased ERK-MAPK signaling pathway may reduce histone H3 (Ser10) phosphorylation via suppression of the activity of histone H3 kinase including MSK1 and RSK2, but not MSK2, consequently decrease the expression of c-Fos protein, which results in the inhibition of colorectal cancer cells proliferation.

Objective To assess the value of magnetic resonance (MR) imaging (MRI) in depicting the depth of myometrial infiltration, cervical invasion and lymph nodes metastasis in patients with endometrial carcinoma compared with surgicopathologic findings. Methods Thirty-eight patients with endometrial carcinoma diagnosed by pathology were inspected by MRI in this prospective study. MR images were analyzed by two radiologists to report the depth of myometrial infiltration, infiltration of the uterine cervix and lymph nodes metastasis. MRI and surgicopathologic staging of endometrial carcinoma were based on FICO classification. MRI findings were compared with surgicopathologic findings. Results The overall accuracy rate of MRI in staging endometrial carcinoma was 77.1 %. The results of Kappa statistics showed that there was strong consistency between MRI and surgicopathology for staging of endometrial carcinoma (k= 0.677). Respective diagnostic accuracy, sensitivity, specificity, negative and positive predictive values in assessing myometrial infiltration(>l/2 depth) were 91.4 %, 83.3 %, 95.6 %, 91.7 % and 90.9 %; those in cervical infiltration were 94.2 %, 80.0 %, 96.7 %, 96.7 %, and 80.0 %; and those in lymph node assessment were 96.2 %, 66.7 %, 100 %, 95.8 % and 100 %. Conclusion MRI is highly accurate in depicting the depth of myometrial infiltration, cervical invasion, lymph nodes metastasis and local-regional staging of endometrial carcinoma.

The aim of the study was to explore the safety management strategies for homeless persons with psychotic disorders.The following nursing management measures were implemented in 127 psychiatric patients,including precaution of aggression and other accidents,treatment of physical diseases,prevention and control of infectious diseases,intensive managementof key areas,key time and key patients,formulation and implementation of safety management regulations and emergency response system,nurse training on professional diathesis,skills and safety awareness.These measures effectively avoided the risksin psychiatric nursing,reduced nursing errors and ensured patient safety.

Objective We observed the effect of dexamethasone ointment on preventing phlebitis induced by vinorelbine. Methods Patients with malignant tumor who received chemotherapy of vinorelbine through peripheral superficial vein injection were divided into the observation group (70 cases) and the control group (72 cases) according to the date of hospitalization. All patients received vinorelbine four times averagely. Patients in the observation group was given dexamethasone ointment along punctured superficial vein. Patients in the control group received routine nursing measure. The incidence rate, time and degree of phlebitis was compared between these groups. Results The incidence rate and degree of phlebitis was lower than those of the control group (P< 0.01, P< 0.05). The incidence time of phlebitis in the observation group was also later than that of the control group (P<0.05). Conclusion Local application of dexamethasone ointment could effectively reduce the incidence of superficial phlebitis caused by vinorelbine chemotherapy.

Objective:To systematically analyze the ADRs caused by azithromycin,and provide references for its safe and rational meidication.Method:Based on the literature search in Chinese Journal Full-text Database(CNKI) between 1994 and 2007,139 cases of adverse drug reactions caused by azithromycin were collected and analyzed.Result:The ADRs caused by azithromycin could be categorized into 7 types,and the most frequent one was allergy.Conclusion:Adverse reactions monitoring of azithromycin should be strengthened to ensure its safe and rational meidication.

@# ObjectiveTo observe the effect of monosialo gangliosides-1 (GM-1) and Radix salviae miltiorrhizae (RSM) on neonatal hyposemic ischemic encephalopathy (HIE).Methods62 cases of HIE were randomly divided into treatment group and control group, and both groups received conventional therapy. Injection of RSM and GM-1 were added into the treatment group in 7~10 days therapy. ResultsHigh curative effect, average curative effect and total curative effect in the treatment group of 31 cases were 80.6%, 16.1% and 96.8%, while that in control group of 31 cases were 32.3%, 51.6% and 83.9% respectively. There was significant difference in the high curative effect and total effect between the two groups (P<0.05). ConclusionThe treatment combined with RSM and GM-1 can enhance the curative effect on HIE without any side effect.

Objective:To demonstrate the effect of cytokine with different concentration in mouse bone marrow-derived immature DCs and find the optimal cytokine dose for DC culture.Methods:mouse bone marrow-derived immature DCs were cultured in the presence of low,moderate and high dose of GM-CSF cytokine with or without IL-4.The morphology of cells were observed and the phenotype were detected.Results:On day 7,DCs in low dose group had the least quantity,small cell process and low expression of CD80,CD86 and MHC class II.The quantity of DCs increased if cultured with IL-4;DCs in moderate dose group and high dose group were more than that in low dose group and the expression of CD80,CD86 and MHC classⅡwere higher,but it is no difference between moderate dose group and high dose group,however cultured with or without IL-4.Conclusion:Higher purity of immature DCs could be get when cultured with low dose of GM-CSF.It was no difference of yield and purity of immature DCs when cultured with moderate or high dose of GM-CSF whether or not combine IL-4.
