ABSTRACT

As a model industrial host and microorganism with the generally regarded as safe (GRAS) status, Corynebacterium glutamicum not only produces amino acids on a large scale in the fermentation industry, but also has the potential to produce various new products. C. glutamicum usually encounters various stresses in the process of producing compounds, which severely affect cell viability and production performance. The development of synthetic biology provides new technical means for improving the robustness of C. glutamicum. In this review, we discuss the tolerance mechanisms of C. glutamicum to various stresses in the fermentation process. At the same time, we highlight new synthetic biology strategies for boosting C. glutamicum robustness, including discovering new stress-resistant elements, modifying transcription factors, and using adaptive evolution strategies to mine stress-resistant functional modules. Finally, prospects of improving the robustness of engineered C. glutamicum strains ware provided, with an emphasis on biosensor, screening and design of transcription factors, and utilizing the multiple regulatory elements.

ABSTRACT

OBJECTIVE:To investigate the effects of the third batc h of Shanghai drug quantity purchasing and national “4+7” drug quantity purchasing in a Shanghai hospital and its impact on the use of statins. METHODS :Consumption amount (tablets) and sum of statins were collected from information system of this hospital before (Feb. 10th-Aug. 31th,2018)and after (Sept. 1th,2018-Mar. 19th,2019)the implementation of the third batch of Shanghai drug quantity purchasing ,after the implementation of national “4+7”drug quantity purchasing (Mar. 20th-Oct. 9th,2019). The defined drug frequency (DDDs)and average defined daily cost (DDC) of above 3 periods were calculated according to drug defined daily dose (DDD) recommended by New Pharmacology and related drug instructions ,so as to analyzethe influence of the third batch of Shanghaidrug quantity purchasing and national “4+7”drug quantity purchasing on the use of statins in this hospital and predict the market share trend of each specification in this hospital. RESULTS & CONCLUSIONS :Before the implementation of the third batch of Shanghaidrug quantity purchasing, there were 5 varieties and 8 specifications of statins in this hospital ;after the implementation of quantity purchasing ,it was adjusted to 5 varieties and 9 specifications. The amount of Simvastatin tablets was small in general ,and its consumption sum also continued to decline after the implementation of quantity purchasing. The amount of Rosuvastatin calcium tablets accounted for 49.49% before the implementation of quantity purchasing ,ranking the first ;after the implementation of 3 batches of Shanghai drug quantity purchasing , the amount proportion decreased to Δ 基金项目:上海市科学技术委员会科研计划项目(No.19ZR1450- 600) below 30%,and consumption sum proportion decreased from *本科 。研究方向 :医药市场研究与分析 。电话 :021- 37.82% to 18.77%;after the implementation of national “4+ 54237308。E-mail:2123398711@qq.com 7”drug quantity purchasing ,the consumption sum proportion # 通信作者 :副主任药师 ,博士。研究方向 :临床药学 。电话: continued to decline to 17.38% .The amount of Atorvastatin 021-65690520。E-mail:Hongjun.chen@tongji.edu.cn calcium tablets increased significantly after the implementation 中国药房 2021年第32卷第2期 China Pharmacy 2021Vol. 32 No. 2 ·247· of the thi rd batch of Shanghaidrug quantity purchasing ,from 41.55% to 54.09%,and decreased slightly after the implementation of the national “4+7”drug quantity purchasing ,but the amount proportion was still as high as 53.06%,ranking the first ;before and after the implementation of quantity purchasing ,the proportions of its consumption sum were ranked the first ,which remained at about 50%,being 52.87%,61.18% and 46.50%,respectively. Compared with the amount before the implementation of the third batch of Shanghaidrug quantity purchasing ,the amount of its generic drugs increased by 40.32%,but the consumption sum decreased by 78.18%. Any one type of Pravastatin sodium tablets and Pitavastatin calcium tablets was not selected ;the proportion of them was increased continuously ,but the overall proportion was relatively small. The price of statins decreased significantly after included in quantity purchasinglist. It is expected that the market share of simvastatin in thise hospital will continue to shrink ,and that of atorvastatin may continue to expand. Due to more adverse reactions ,rosuvastatin has limited room to expand ,and pravastatin accounts for a small proportion ,but there is still a demand for patients. Because pitavastatin is a new drug ,its application in this hospital needs more clinical data support.

ABSTRACT

Objective@#To analyze the incidence and mutation characteristics of FLT3 gene mutation and clinical efficacy of tyrosine kinase inhibitor (TKI) in patients with mixed phenotype acute leukemia (MPAL).@*Methods@#A total of 48 patients with MPAL who were admitted to Hebei Yanda Lu Daopei Hospital from June 2015 to February 2018 were retrospectively analyzed. The common mutated 58 genes in hematologic malignancies were detected by using amplicon-targeted next generation sequencing, of which internal tandem duplication (ITD) and point mutation occurred in the hotspot region of exon 14, 15 and 20 in FLT3 gene. Multiplex polymerase chain reaction (PCR) analysis was used to detect 35 gene fusions in hematological neoplams.@*Results@#There were 7 cases of FLT3 mutation in 48 MPAL patients, which were all ITD mutations. The median length of the inserts of FLT3-ITD was 48 bp, and one MPAL patient carried 2 multiple length inserts simultaneously, and the median variant allele frequency (VAF) was 40.5% (7.9%-84.7%). There were no statistically significant differences in clinical and genetic characteristics between FLT3 mutation-positive and FLT3 mutation-negative MPAL patients (both P > 0.05). Among 7 FLT3 mutation-positive MPAL patients, 4 cases were often accompanied with RUNX1 mutation. A total of 4 MPAL patients with FLT3-ITD-positive received sorafenib or sunitinib combined chemotherapy, and 3 of them achieved complete remission.@*Conclusions@#ITD mutation is the main part in the FLT3 mutation of MPAL patients. FLT3-ITD-positive MPAL patients are often accompanied with RUNX1 mutation, which may benefit from targeted therapy with FLT3 kinase inhibitor.

ABSTRACT

A QuEChERS-ultra high performance liquid chromatography-tandem mass spectrometry method was developed for qualitative screening of 169 veterinary drug residues in bear bile powder, including β-agonists and inhibitors, antibiotics (penicillins, β-lactams, sulfomamides, quinolones, chloramphenicals, tetracyclines, nitroimidazoles, macrolides, polyethers, etc.), antiviral drugs, anthelminitics, steroid hormones, nonsteroidal antiinflammatory drugs (NSAIDs) and sedatives. The samples were extracted by Na2EDTA-McIlvaine buffer solution and 5% fomic acid-acetonitrile solution, then purified by dispersive solid phase extraction. Detection of veterinary drug residues by ultra high performance liquid chromatography-triple quadrupole mass spectrometry was conducted and qualitative confirmed by ion ratios. The limits of detection of 169 veterinary drugs were 1-1 000 μg·kg-1. The method is simple and fast, which had been used for the analysis of actual samples, and can be extended to the detection of similar matrix.

ABSTRACT

Glutamic acid is an important amino acid with wide range of applications and huge market demand. Therefore, by performing transcriptome sequencing and re-sequencing analysis on Corynebacterium glutamicum E01 and high glutamate-producing strain C. glutamicum G01, we identified and selected genes with significant differences in transcription and gene levels in the central metabolic pathway that may have greatly influenced glutamate synthesis and further increased glutamic acid yield. The oxaloacetate node and α-ketoglutarate node play an important role in glutamate synthesis. The oxaloacetate node and α-ketoglutarate node were studied to explore effect on glutamate production. Based on the integrated strain constructed from the above experimental results, the growth rate in a 5-L fermenter was slightly lower than that of the original strain, but the glutamic acid yield after 48 h reached (136.1±5.53) g/L, higher than the original strain (93.53±4.52) g/L, an increase by 45.5%; sugar-acid conversion rate reached 58.9%, an increase of 13.7% compared to 45.2% of the original strain. The application of the above experimental strategy improved the glutamic acid yield and the sugar-acid conversion rate, and provided a theoretical basis for the metabolic engineering of Corynebacterium glutamicum.

ABSTRACT

A tadalafil analogue was detected during routine screenings from two "fatigue reliever, immunity enhancer" dietary supplements by using UHPLC/Q-TOF HRMS. The MS2 spectrum of this compound was almost identical to that of 2-hydroxypropylnortadalafil. However, the retention time of this analogue was different from that of the 2-hydroxypropylnortadalafil isomers. The analogue was purified by using preparative HPLC and the structure was elucidated by mass spectrometric and NMR spectroscopic experiments. The spectral data suggested that the analogue bore a 3-hydroxypropyl group instead of the N-methyl group in tadalafil. The structure was further confirmed by comparison of the 1H NMR spectra data with those of the reference standard, and thus named as 3-hydroxypropylnortadalafil. The structure is first reported in China.

ABSTRACT

Objective:To investigate the protective effect of glycyrrhizic acid (GA) from tumor necrosis factor-α+actinomycetes ketone (TNF-α+CHX) induced apoptosis in rat small intestine crypt epithelial cell line (IEC-6) via AU rich element mRNA binding protein HuR mediated posttranscription of p21 and the potential mechanism. Method:The cultured IEC-6 cells were observed. The experiment was divided into blank group, GA (60 μmol·L-1) group, TNF-α+CHX group and GA+TNF-α+CHX group. Cytoplasmic and nuclear HuR were measured by Western blot. The interaction of HuR and p21 mRNA was detected by biotin pull down and RNA IP. Luciferase activity was measured after transfection with construct with p21 3'-UTR cloned into downstream of luciferase reporter. Cell apoptosis was detected by real-time dynamic cell analyzer, p21 and cysteine proteinas-3 precursor protein(proCaspase-3) association was analysised by CO-IP. Result:After GA treatment for 48 h, cytoplasmic HuR protein expression increased(P<0.05),the binding between HuR and p21 mRNA expression up regulated(P<0.05), luciferase activity increased(P<0.01), and p21 mRNA and protein expression also increased(P<0.05), while these results were abolished by HuR silencing with siRNA. GA enhanced p21 and procaspase3 interaction(P<0.05), and attenuated TNF-α+CHX induced apoptosis in IEC-6 cells. Conclusion:GA protected IEC-6 cells from TNF-α/CHX induced apoptosis via HuR mediated p21 posttranscription, which due to GA enhanced HuR binding to endogenous and recombinant p21 mRNA and increased p21 interaction with proCaspase3.

ABSTRACT

Objective:To report our experience in diagnosis and treatment of posterior atlantoaxial dislocation with odontoid retrolisthesis. Methods:A retrospective study was conducted of the 5 patients who had been treated from July 2012 to August 2018 at Department of Orthopaedics, General Hospital of Southern Theater Command for posterior atlantoaxial dislocation. They were 4 men and one woman, aged from 34 to 67 years (average, 47 years). All of them had a history of trauma. Of them, 4 were complicated with odontoid fracture and one with congenital free os odontoideum. Their posterior atlantoaxial dislocation ranged from 3 to 9 mm (average, 6 mm). By the American Spinal Injury Association (ASIA) grading system, their preoperative spinal injury was rated as grade B in one, as grade C in 3 cases and as grade D in one. All the 5 patients underwent skull traction at 10° flexion. Surgical trans-oralpharyngeal atlantoaxial reduction and internal fixation was performed for the one patient whose reduction had not been achieved by traction while posterior atlantoaxial screw-rod fixation or anterior odontoid screwing was conducted for the 4 patients whose reduction had been achieved by traction. The distance of posterior atlantoaxial dislocation was measured to evaluate their reduction and ASIA grade system was used to assess their spinal function after operation. Results:The postoperative distance of posterior atlantoaxial dislocation was 0 mm, showing a reduction rate of 100%. The 5 patients were followed up for 6 to 36 months (average, 15 months). By the ASIA grade system, the postoperative functional recovery of the spine was grade D in 4 cases and grade C in one. No implant loosening or breakage occurred. Conclusion:As a kind of high-energy hyperextension injury, posterior atlantoaxial dislocation is rare in clinic, but an appropriate treatment can be adopted to deal with its different clinical types to achieve good outcomes.

ABSTRACT

BACKGROUND@#Radical nephrectomy and thrombectomy is the standard surgical procedure for the treatment of renal cell carcinoma (RCC) with tumor thrombus (TT). But the estimation of intra-operative blood loss is only based on the surgeon's experience. Therefore, our study aimed to develop Peking University Third Hospital score (PKUTH score) for the prediction of intra-operative blood loss volume in radical nephrectomy and thrombectomy.@*METHODS@#The clinical data of 153 cases of renal mass with renal vein (RV) or inferior vena cava tumor thrombus admitted to Department of Urology, Peking University Third Hospital from January 2015 to May 2018 were retrospectively analyzed. The total amount of blood loss during operation is equal to the amount of blood sucked out by the aspirator plus the amount of blood in the blood-soaked gauze. Univariate linear analysis was used to analyze risk factors for intra-operative blood loss, then significant factors were included in subsequent multivariable linear regression analysis.@*RESULTS@#The final multivariable model included the following three factors: open operative approach (P < 0.001), Neves classification IV (P < 0.001), inferior vena cava resection (P = 0.001). The PKUTH score (0-3) was calculated according to the number of aforementioned risk factors. A significant increase of blood loss was noticed along with higher risk score. The estimated median blood loss from PKUTH score 0 to 3 was 280 mL (interquartile range [IQR] 100-600 mL), 1250 mL (IQR 575-2700 mL), 2000 mL (IQR 1250-2900 mL), and 5000 mL (IQR 4250-8000 mL), respectively. Meanwhile, the higher PKUTH score was, the more chance of post-operative complications (P = 0.004) occurred. A tendency but not significant overall survival difference was found between PKUTH risk score 0 vs. 1 to 3 (P = 0.098).@*CONCLUSION@#We present a structured and quantitative scoring system, PKUTH score, to predict intra-operative blood loss volume in radical nephrectomy and thrombectomy.

ABSTRACT

Objective: Headspace-solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) was used to analyze the characteristic volatile compounds from 18 heat-clearing herbs. Method: HS-SPME was used to extract the volatile components from different heat-clearing herbs, and the chemical compositions were analyzed by GC-MS. The GC was performed at the injector temperature of 250℃,the high-purity helium was used as the carrier gas at a constant column flow of 1.0 mL·min-1 with no split sampling. The solvent delay time was 3 min. Mass spectrum conditions included electron bombardment ion source, electron bombardment energy of 70 eV,ion source temperature of 230℃,quadrupoles temperature of 150℃,and scanning range of m/z 29-370.The volatile components were confirmed according to NIST2013 database, and the relative content percentage of each component was calculated by peak area normalization method. PCA was used to detect clustering and discuss the characteristics of volatile compounds of 18 heat-clearing herbs. Result: A total of 328 volatile compounds were detected in 18 heat-clearing herbs, including alkanes, unsaturated hydrocarbon, alcohols, aldehyde, ketones, esters, furan and other unknown compounds. The number and relative content of volatile components in different heat-clearing herbs varied dramatically. Several important characteristic volatile compounds, such as l-camphor,torreyol,caryophyllene oxide, globulol and 2,4-cycloheptadien-1-one,2,6,6-trimethyl,were detected in the 18 heat-clearing herbs by principal component analysis. Conclusion: Heat-clearing herbs contains rich volatile components, which are the main contributors to the formation of unique aroma of herbs,as well as the main active substances.

ABSTRACT

Diabetes is a lifelong non-communicable disease caused by continuous high blood sugar, which poses a great threat to people's health. Diabetic cognitive dysfunction is the neurological impairment caused by the disorder of glucose and lipid metabolism in diabetes mellitus, which is characterized by inattention, decreased learning and memory ability, and then developed into alzheimer's disease. The pathogenesis of diabetic cognitive dysfunction is not fully understood.there are no effective drugs for this disease, hypoglycemic drugs are mainly used in the clinical treatment of diabetic cognitive dysfunction, and most drugs are accompanied by serious adverse reactions while playing a therapeutic role, drug resistance and liver injury are easy to occur. In recent years, there have been a lot of research achievements on the prevention and treatment of cognitive dysfunction in diabetes mellitus with traditional Chinese medicine(TCM).Modern medicine believes that diabetes belongs to the category of consumptive thirst of TCM, and the diabetic cognitive dysfunction is consumptive thirst combined with dementia and forgetfulness.Qi and Yin deficiency runs through the whole process of consumptive thirst, the Qi injured,Then the Qi defficiency would cause blood circulation malfunction, long-time poor blood condition would cause blood stagnating in the brain and blocking the brain, which would lead to the brain disease.TCM can improve the diabetic cognitive dysfunction by lowering blood sugar, inhibiting neuron damage, deposition of Amyloid beta(Aβ)and abnormal phosphorylation of Tau protein. This article reviews the pathogenesis of diabetic cognitive dysfunction from glucolipid metabolic disorders, disruption of blood brain barrier, inflammation, oxidative stress and non enzymatic glycosylation, insulin resistance in diabetes etc.,and explores the prevention and treatment of diabetic cognitive dysfunction by Chinese medicine,to provide a reference for the research and development of drug prevention and treatment of diabetic cognitive dysfunction.
