ABSTRACT

@#【Objective】To establish a cell model of radiation injury of human bone marrow mesenchymal stem cells(hBMMSC),and to provide experimental basis for further study on the pathogenesis of osteoradionecrosis of the jaws(ORNJ).【Methods】After X-ray irradiation,the gene expression change of Beclin1,Sox2,Nanog,RUNX2 and OGN in hBMMSC were evaluated by Western blot and RT-qPCR. The apoptosis rate change was detected by Annexin- V/PI double staining method. The proliferation rate change was determined by clone formation experiment. The alkaline phosphatase activity change was detected by microplate method.【Results】The gene expressions of Beclin1 increased with the doses increased,while Sox2,Nanog,RUNX2 and OGN in hBMMSC were all down- regulated after irradiation. The apoptotic rate increased,the colony formation rate decreased and the alkaline phosphatase activity decreased also.【Conclusion】X-ray irradiation can cause ionizing radiation injury of hBMMSC. We successfully established a cell model of hBMMSC radiation injury,the model can be applied to the experimental study of the pathogenesis of ORNJ.

ABSTRACT

[Objective]To explore the effect of alpha2-macroglobulin(α2M)on superoxide anion(.O2-)content, superoxide dismutase(SOD)activity and the process of cell-to-myofibroblast transformation in human skin fibroblasts (HSF)after X-ray irradiation.[Methods]HSF cells were irradiated with 0,5,10,15 and 20 Gy X-ray.The change of. O2- content and SOD activity in the supernatant of cell culture medium were measured on the first day after irradiation. The protein expression of alpha-smooth muscle actin(α-SMA)was detected by Western blot on the fifth day after irradia-tion.The most sensitive radiation dose is selected.HSF cells were irradiated with the above sensitive dose.Respectively, 1h before irradiation,1 h after irradiation,the experimental group cell culture medium was added to a final concentration of 0.25 mg/mL,0.5 mg/mL of α2M.The change of.O2-content,SOD activity and the protein expression of α-SMA were detected.[Results]HSF cells were irradiated with 5~20 Gy doses of X-ray..O2- content increased,SOD activity de-creased and α-SMA protein expression increased gradually(P<0.05).The addition of α2M at 1 h after 10Gy X-ray irradi-ation reduced the.O2- content,increased the SOD activity and downregulated the protein expression of α-SMA in HSF cells(P<0.05). There was no significant change in the administration at 1 h before irradiation.[Conclusion]HSF cells increased.O2-content significantly,while SOD activity decreased,and the tendency to transform myofibroblasts after X-ray irradiation.α2M can reduce the.O2-content,increase the SOD activity in HSF cells and inhibit the transformation of fibroblasts into myofibroblasts after irradiation.Indicating that α2M can play a role in radiation protection by anti-oxida-tion and anti-fibrosis.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the relationship between recurrent spontaneous abortion (RSA) and the level of interferon-γ (IFN-γ) and interleukin-4 (IL-4) in peripheral blood and gingival crevicular fluid of patients with chronic periodontitis.</p><p><b>METHODS</b>Fifty pregnant women with recurrent spontaneous abortion (gestational age: 5 - 28 weeks) and 40 pregnant women (gestational age: 39 - 41 weeks) were included and their periodontal status was examined. The level of IFN-γ and IL-4 in gingival crevicular fluid and peripheral blood was detected by enzyme-linked immunosorbent assay.</p><p><b>RESULTS</b>Nine RSA women and 5 in control group had periodontitis. The prevalence of periodontitis in RSA group was 18% (9/50) and 13% (5/40) in the control group (P < 0.05). The probing depth, attachment loss and bleeding index in RSA group were higher than those in the control group (P < 0.05). The level of IFN-γ was (52.98 ± 17.56) ng/L in gingival crevicular fluid of RSA group and (25.25 ± 7.93) ng/L in control group (P < 0.01). The level of IL-4 was (15.43 ± 1.77) ng/L in gingival crevicular fluid of RSA group and (19.62 ± 4.04) ng/L in control group (P < 0.01). The content of IFN-γ was (27.79 ± 3.59) ng/L in peripheral blood of RSA group and (18.39 ± 2.65) ng/L in control group (P < 0.05). The content of IL-4 was (15.88 ± 0.95) ng/L in peripheral blood of RSA group and (22.98 ± 4.30) ng/L in control group (P < 0.001). The content of cytokines in gingival crevicular fluid and peripheral blood in the same group was positively related (r > 0.8, P < 0.001).</p><p><b>CONCLUSIONS</b>Pregnant women of RSA have a higher chance of getting periodontitis. The change in the level of cytokines in periodontal tissue could result in excursion of maternal intrauterine immune environment to T-helper 1 (Th1). Periodontitis was related to RSA.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the clinical value of the mass screening by analyzing the features of prostate cancer between mass screening patients and clinical patients.</p><p><b>METHODS</b>From January 2000 to January 2008, 441 cases of prostate cancer (including 122 patients from clinical diagnosis and 319 patients from mass screening 23 183 men who were more than 50 years old) were analyzed from age, digital rectal examination (DRE), serum prostate specific antigen (PSA) levels and range, the Gleason's score and grade, clinical staging and therapy.</p><p><b>RESULTS</b>42.0% of mass screening patients were inspected by DRE, it was lower than that (79.5%) in the clinical patients. The percent of patients with serum PSA levels of less than 10.0 microg/L in mass screening group was higher than in clinical group, while the percent of patients with serum PSA levels of more than 20.0 microg/L in mass screening group was lower than in clinical group. The percent of moderately differentiated degree of prostate cancer in mass screening group was higher than in clinical group, but it was on the contrary for poorly differentiated degree of prostate cancer. The percent of T1-2 prostate cancers in mass screening group was 56.1%, which was higher than 25.4% in clinical group. While, the percent of T3-4 tumors in mass screening group was lower than in clinical group. The percent of men undergoing radical prostatectomy in mass screening group was 18.2%, which was higher than 9.8% in clinical group. The percent of men of locally advanced and far metastasis in mass screening group was 26.0%, while was lower than 46.0% in clinical group.</p><p><b>CONCLUSION</b>General investigation for prostate cancer is benefit to find asymptomatic cancer of early stage.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To discuss the diagnosis and treatment of non-specific granulomatous prostatitis (NSGP).</p><p><b>METHODS</b>Thirty-two cases of NSGP were diagnosed by puncture biopsy under transrectal ultrasound (TRUS) and treated with antibiotics and other medicines from September, 2000 to May, 2006.</p><p><b>RESULTS</b>Pathomorphologically, NSGP was basically characterized by granuloma with vessels or grand alveoli in the center. The mean follow-up was 24 months. Urination irritation and obstruction were improved. Q(max) was increased to 15.0-24.0 ml/s, and in 3 cases of urinary retention, to 12.0, 14.5 and 16.5 ml/s, respectively. Digital rectal examination (DRE) indicated a reduced size and softened texture of the prostate induration. PSA was decreased to 1.3-11.5 microg/L. Four cases experienced relapse but were cured after retreated. No prostate cancer was observed.</p><p><b>CONCLUSION</b>NSGP can be definitely diagnosed by puncture biopsy under TRUS and effectively relieved by antibiotics with the alpha-receptor blocker. In case of serious obstruction complicated by urinary retention, transurethral electrotomy can be considered.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To assess the application value of transrectal ultrasound (TRUS) in the diagnosis of chronic prostatitis.</p><p><b>METHODS</b>TRUS and examination of prostatic secretion (EPS) were used in the diagnosis of 3 500 cases of chronic prostatitis from September, 2000 to May, 2006.</p><p><b>RESULTS</b>Lower resonance of the inner gland, low-level echo, uneven echo light spots, incomplete outlines and unsmooth borderlines were found in 2279 cases (65.1%), and the enlarged prostate in 1 084 cases (31.0%), with clear integrated amicula and enhanced echogenic spots at the juncture of the external and inner gland. No obvious changes were noted in 137 cases (4.0%), and in another 391 cases (11.2%) were detected alteration of the acoustic image of cystospermitis and blurred margins and uneven echoes of the seminal vesicle. The WBC count in EPS was < 10/HP in 132 cases (3.8%), 10-19/HP in 2 156 cases (61.6%) and > or =20/HP in 1212 cases (34.6%).</p><p><b>CONCLUSION</b>TRUS, as a diagnostic means for chronic prostatitis, can be easily performed and causes little pain and therefore is readily accepted by patients. Combined with EPS, TRUS can provide more definite diagnostic evidence, and for those who are afraid of pain and reject EPS, it is a desirable alternative in the diagnosis of chronic prostatitis.</p>

ABSTRACT

<p><b>AIM</b>To investigate the transforming growth factor beta1 (TGF-beta1) and basic fibroblast growth factor (bFGF) expressions in benign prostatic hyperplasia (BPH) and the effect of beta-radiation.</p><p><b>METHODS</b>TGF-beta1 and bFGF expression was studied by means of an immunohistochemical method in nine normal prostatic (NP) tissues, 15 hyperplastic prostatic tissues and 35 hyperplastic prostatic tissues treated with 90Sr/90Y.</p><p><b>RESULTS</b>The TGF-beta1 expression in the epithelium and stroma of normal prostatic tissues was 68.2 % +/- 10.5 % and 29.7 % +/- 4.6 %, respectively, while it was 64.8 % +/- 9.3 % and 28.6 % +/- 4.1 %, respectively, in hyperplastic prostatic tissues. Compared with the controls, TGF-beta1 expression in the epithelia and stroma of BPH treated with 90Sr/90Y increased significantly (P <0.01). The bFGF expression in epithelia and stroma of normal prostatic tissues was 17.4 % +/- 3.7 % and 42.5 % +/- 6.8 %, respectively, and was 46.3 % +/- 8.2 % and 73.2 % +/- 12.1 %, respectively, in hyperplastic prostatic tissues. Compared with the controls, expressions of bFGF in the epithelia and stroma of BPH treated with a 90Sr/90Y prostatic hyperplasia applicator decreased significantly (P <0.01).</p><p><b>CONCLUSION</b>Exposure of beta-rays had noticeable effects on BPH tissues, enhancing TGF-beta1 expression and inhibiting bFGF expression.</p>

ABSTRACT

To achieve the goal of early detection, diagnosis and treatment of prostate cancer in China, mass screening for prostate cancer using serum prostate specific antigen (PSA) has been performed in men over 50 years. We compared this result with the clinical cases. It proved that only through the mass screening can we really find the early stage prostate cancer and be given the chance of curing the cancer. We also investigated the current treatment situation and problems and figured out the further direction on the prostate cancer research.
