ABSTRACT

Aim To analyze the molecular mechanism of rhubarb in the treatment of aeute pancreatitis ( AP) by network pharmacology and molecular docking.Methods TCMSP,TCMID and Swiss target predic¬tion databases were used to screen the active compo¬nents and targets of rhubarb,and genecards and OMIM databases were used to screen the targets of AP.Then the active ingredient drug target network of rhubarb and theactive ingredient disease target network of rhubarb for AP were constructed by using Cytoscape software.PPI network was constructed in string database, and go and KEGG enrichment analysis was performed in metascape database and R language.Finally,molecular docking was used to verify the possibility of binding the core active components to the core target.Results A total of 192 active components and 1 882 AP targets were obtained.The first three active components of rhubarb in the treatment of AP were beta sitosterol, aloe emodin and eupatin.The core target of rhubarb in the treatment of AP was hsp90aal.Go enrichment analysis focused on reaction to toxic substances, while KEGG enrichment analysis was significantly enriched in p53 signaling pathway closely related to AP.Molecular docking showed good binding and stable conformation.Conclusions Rhubarb can affect the expression of AP related genes and proteins through p53 signaling pathway, thereby inhibiting cell apoptosis and allevia¬ting the inflammatory injury of AP.

ABSTRACT

Objective To explore the performance of mobile health platform for standardized management of pregnant women with gestational diabetes mellitus(GDM). Methods A randomized controlled trial was conducted,in which 295 women with GDM were randomized into two groups(traditional management group and mobile health management group)by a computer-generated sequence.The traditional management group accepted standardized GDM management,and the mobile health management group was supplemented by mobile health management based on the standardized management.The glycemic control rate and the incidences of low birth weight,macrosomia,preterm birth,premature rupture of membranes,postpartum hemorrhage after cesarean section,neonatal asphyxia,malformation,and admission to the neonatal intensive care unit were compared between the two groups. Results The glycemic control rate in mobile health management group was significantly higher than that in the traditional management group [(67.22±22.76)%

ABSTRACT

Polycomb group (PcG) proteins are transcriptional repressors that control cell fate and the development of embryo, and they elicit function mainly in the form of polycomb repressive complex (PRC). Chromobox protein homolog 6 (CBX6) is one of the core protein subunits of PRC1, which plays an important role in gene expression regulation, cell renewal and differentiation, tumorigenesis and development, and stem cell maintenance. In this study, CBX6 was found to be degraded through a ubiquitin-proteasome dependent pathway. Then the gene expression library containing 92 deubiquitinating enzymes (DUB) was used to screen DUB targeting CBX6 and results found that ubiquitin-specific protease 29 (USP29) could obviously stabilize CBX6 protein level and extend its half-life (P < 0.05). Immunoprecipitation experiments found that CBX6 interacted with USP29 through its C-terminal domains; Further studies found that USP29 regulated the protein stability of CBX6 by deubiquitination in an enzymatic-activity dependent manner. Cell proliferation assay also found that USP29 inhibited the proliferation of MCF7 cells (P<0.0001). Taken together, through screening, this study found that USP29 could stabilize CBX6 protein level through deubiquitinating CBX6 and inhibit the cell proliferation of MCF7.

ABSTRACT

OBJECTIVE@#To study the expression of Peroxiredoxin-6 (Prdx6) gene in elderly patients with acute myeloid leukemia (AML) and its clinical significance.@*METHODS@#The expression level of Prdx6 in bone marrow cells of 33 cases of AML, 8 cases of CML and 11 cases of other blood diseases was detected by PCR. The correlation of the abnormal expression of Prdx6 with patient age and blood routine parameters was further analyzed.@*RESULTS@#the expression level of Prdx6 in elderly patients with AML (≥60 years) was significantly lower than that in younger patients (<60 years) (P<0.05); the expression level of Prdx6 in low WBC (≤1×10/L) group was lower than that in medium WBC (1-10×10/L) group or high WBC (>10×10/L) group (P<0.05); the proportion of WBC count (≤1×10/L) in elderly patients with AML reached 38.5%, which was significantly higher than that in younger patients (5%) (P<0.05); the overall survival (OS) rate of elderly patients was lower than that of younger patients (P<0.05).@*CONCLUSION@#The expression of Prdx6 in elderly patients with AML is low, moreover, it relates with low value of WBC in peripheral blood, suggesting that it may be one of poor prognostic factors for the elderly patients with AML.

ABSTRACT

@#AIM: To investigate the effect of connective tissue growth factor(CTGF)on the proliferation and migration and transformation in Tenon's capsule fibroblasts(Tfb)of primary open angle glaucoma(POAG)patients.<p>METHODS: Tfb were cultured from Tenon's tissues in POAG patients <i>in vitro</i>. The free-serum DMEM-F12 containing 1.0, 10.0, 100.0ng/mL of CTGF was added into medium for 24h and 48h in different experimental group respectively, and only equal volume of free-serum DMEM-F12 was added in the negative control group. After 24h, the cell proliferation was analyzed through MTT assay, and migration was evaluated by crutch method. After 48h, Semi-quantitative RT-PCR was used to observed the mRNA expression of α-smooth muscl actin(α-SMA), and expression of α-SMA protein was examined by immunochemistry.<p>RESULTS: The proliferation values <i>A</i> of the cells in 1.0, 10.0, 100.0ng/mL of CTGF group respectively were 0.436±0.009, 0.643±0.009, 0.679±0.006, and 0.423 ±0.156 in the negative control group. The migrated cell number was 34.600±3.507, 70.400±2.074, 80.000±2.915 in different concentrations of CTGF group respectively, and 31.000±3.536 in the negative control group. And also in different experimental groups respectively, the absorbance ratio of α-SMA/β-actin was 0.873±0.161, 1.213±0.312, 1.352±0.376, and 0.851±0.158 in the negative control group, the expressing levels <i>A</i> of α-SMA protein in Tfb were 0.110±0.026, 0.141±0.017, 0.175±0.027, and 0.108±0.020 in the negative control group. The statistics of the above experimental data showed that, comparing with the negative control group, the 10.0 and 100.0ng/mL CTGF groups was statistically significant different(<i>P</i><0.05), but there was no statistical different between the 1.0ng/mL CTGF group and the negative control group(<i>P</i>>0.05). <p>CONCLUSION: The proliferation, migration, and phenotypic transformation of Tfb can be promoted in CTGF group in POAG patients. These findings suggest that CTGF may play a role in the development of filtering bleb scarring.

ABSTRACT

Objective To understand detection results and difference in multidrug-resistant organisms(MDROs) in intensive care unit(ICU)and non-ICU.Methods Strains isolated from clinical specimens of hospitalized patients in a hospital from January 2015 to December 2016 were analyzed, 6 kinds of MDROs were conducted targeted monitoring, isolation and antimicrobial resistance of 6 kinds of MDROs from ICU and non-ICU patients were compared. Results A total of 1 013 strains of 6 kinds of MDROs were monitored, isolation rate was13.13%.Isolation rate of MDROs in ICU was higher than that of non-ICU (24.60%vs 5.47%, P<0.001).Carbapenem-resistant Acinetobacter baumannii(CRAB)was the main isolated MDROs, accounting for 69.40%;of different pathogenic organisms, isolation rate of CRAB was the highest(55.75%).The main MDROs detected in ICU and non-ICU were both CRAB, accounting for 76.32%and 48.62%respectively;Of isolated pathogens, isolation rate of MDROs in ICU was higher than that of non-ICU(47.95%vs 8.02%, P<0.001).Antimicrobial resistance rates of Escherichia coli isolated from ICU to ticarcillin/clavulanic acid, ceftriaxone, cefotaxime, cefepime, imipenem, meropenem, amikacin, and gentamicin were all higher than that of non-ICU, resistant to piperacillin was lower than non-ICU, difference was statistically significant(all P≤0.05);resistance rates of Klebsiella pneumoniae from ICU to common antimicrobial agents(except piperacillin)were all higher than non-ICU(all P<0.05).Resistance rates of Acinetobacter baumannii and Pseudomonas aeruginosa from ICU to common antimicrobial agents were all higher than non-ICU (all P<0.05).Resistance rates of Staphylococcus aureus isolated from ICU to oxacillin, ciprofloxacin, tetracycline, and rifampicin were all higher than non-ICU (all P<0.05), and resistance rates of Enterococcus faeciumto quinupristin/dafoeleptin and tetracycline were both lower than non-ICU (both P<0.05).Conclusion Isolation rate of MDROs in ICU is high, resistance rates to most antimicrobial agents are also higher than non-ICU, monitoring on MDROs in ICU should be strengthened, and according prevention and control measures should be formulated.

ABSTRACT

Objective: To study the clinical characteristics and diagnosis of primary pulmonary lymphoma (PPL). Methods: A retrospective review was performed based on the clinical records of 18 PPL cases treated at Affiliated Pulmonary Hospital of Tongji University from 1999 Sep to 2006 June. The clinical manifestations, imaging features, diagnosis approaches, staging, and prognosis of the 18 patients were analyzed. Results: All the eighteen cases of PPL belonged to the non Hodgkin's lymphoma. There were 13 patients with mucosa-associated lymphoid tissue (MALT) lymphoma, 3 patients with small B cell lymphoma, 1 patient with anaplastic large cell lymphoma, and 1 patient with T cell lymphoma tendency. The main symptoms were cough and weight loss. The PPL in 4 patients with non specific symptoms were diagnosed during physical examination. Pulmonary consolidation shadows and multiple nodules were the main findings of CT scan. An air bronchogram is often seen in the consolidation imaging. All the 18 patients received fiber-bronchoscopic biopsy but only 2 patients were diagnosed as PPL. Seven patients were given percutaneous needle biopsy but PPL was confirmed in only 1 patient. The diagnosis was confirmed in fifteen patients by operative pathology. Ten patients received open thoracotomy and 5 patients received video-assisted thoracoscopy. Of the 15 patients, there were 9 cases in stage E,4 cases in stage II 1 E,3 cases in stage II 2E, 1 case in stage III, and 1 case in stage IV. Surgery, radiotherapy, and chemotherapy were most common treatment modalities for PPL. Conclusion: PPL was easy to be misdiagnosed due to non specific clinical and imaging manifestations. The invasive biopsy is the main diagnosis approach. The prognosis is relatively better.
