ABSTRACT

Objective To observe the effects of Huatan Quyu Decoction on cognitive function and the expressions of GABA and VILIP-1 in brain tissue of rats with cerebral small vessel disease;To discuss its mechanism for treatment on cerebral small vessel disease.Methods Totally 48 male SD rats were randomly divided into blank group,model group,Huatan Quyu Decoction low-and high-dosage groups,with 12 rats in each group.Except for the blank group,a rat model of cerebral small vessel disease was prepared by in vitro injection of homologous microemboli.Huatan Quyu Decoction low-and high-dosage groups were given Huatan Quyu Decoction 1.25 and 2.5 g/kg by gavage,the blank group and model group were gavage with equal amounts of distilled water for 28 consecutive days.Morris water maze experiment was conducted on day 1,7,14,and 28 after administration to evaluate the learning and memory abilities of rats,HE staining was used to observe pathological changes in hippocampal tissue,and immunohistochemical staining was used to detect the expressions of GABA and VILIP-1 proteins in brain tissue.Results Compared with the blank group,the escape latency of Morris water maze experiment in model group significantly prolonged(P<0.05),and the number of crossing platforms was significantly reduced(P<0.05);the arrangement of hippocampal tissue cells was disordered,gaps widen,and nuclei atrophy and necrosis,the GABA expression in brain tissue significantly decreased(P<0.05),while the VILIP-1 expression significantly increased(P<0.05).Compared with the model group,the escape latency of Morris water maze experiment in the Huatan Quyu Decoction low-and high-dosage groups significantly shortened(P<0.05)on day 7,14,and 28 of administration,and the number of crossing platforms significantly increased(P<0.05),GABA expression significantly increased(P<0.05),while VILIP-1 expression significantly decreased(P<0.05).Compared with the Huatan Quyu Decoction low-dosage group,the escape latency of Morris water maze experiment in Huatan Quyu Decoction high-dosage group decreased at various time points,and the number of crossing platforms increase,the pathological damage of hippocampal tissue was reduced,the expression of GABA in brain tissue increased,and the expression of VILIP-1 decreased,with statistical significance(P<0.05).Conclusion Huatan Quyu Decoction can increase the expression of GABA in brain tissue and inhibit the expression of VILIP-1,thereby improve the cognitive function of rats with cerebrovascular disease.

ABSTRACT

OBJECTIVES@#This study investigate the clinical and imaging features of Ewing sarcoma (ES) of the jaw.@*METHODS@#Eight cases of pathologically diagnosed ES of the jaw from January 2010 to June 2022 were included in the study. Clinical and radiological features were retrospectively analyzed.@*RESULTS@#Among the eight cases, the mean age at onset was 29.4 years, and the male to female ratio was 7∶1. The predilecting site was the posterior part of mandible, accounting for 75% of the cases. The lesions often exhibited early numbness of the lower lip and lymphadenopathy. The main radiographic manifestation of mandibular lesions was ill-defined radiolucency, mixed with fibrous or brush-like tumor matrix, and soft tissue mass. The maxillary ES lesions mainly presented as lytic bone destruction accompanied by adjacent soft tissue mass. Periosteal ossification was rarely seen.@*CONCLUSIONS@#The clinical and imaging characteristics of ES in the jaw are helpful for its diagnosis.

ABSTRACT

Objective:To observe the efficacy of drug combined with red-blue light irradiation, chemical peeling by glycolic acid and intense pulsed light (IPL) in the treatment of moderate acne vulgaris.Methods:A total of 187 patients with moderate acne vulgaris were divided into five groups. There were 59 males and 128 females, aged from 16 to 29 years (21.79±2.52). In group A, patients were treated with oral medicines; the patients were treated with oral medicines combined with red-blue light irradiation, chemical peeling by glycolic acid and IPL respectively in group B, group C and group D. In group E, patients were treated with oral medicines combined with chemical peeling by glycolic acid and IPL. The number of different types of skin lesions (comedoes, papules, pustules) were compared between before and after treatment in five groups. The clinical effect of five treatment groups was evaluated by comparing regression rates of different types of skin lesions and total skin lesions and treatment efficiency.Results:All the patients with moderate acne vulgaris were brought into this study. The regression rates of comedoes, papules and total skin lesions were (86.37±9.64)%, (94.25±9.79)% and (88.80±9.40)% respectively in group E, and significantly higher than that of other four groups ( P<0.05). The treatment efficiency of group E also were significantly higher than that of other four groups ( P<0.05). Then, the regression rates of comedoes (70.91±18.52) in group C was significantly higher than that of group A and group B ( P<0.05). The regression rates of papules (91.42±13.86) in group D was significantly higher than that of group A and group B ( P<0.05). Conclusions:Oral medicines combined with chemical peeling by glycolic acid and IPL has obvious clinical efficacy in treatment of patients with moderate acne vulgaris. Oral medicines combined with chemical peeling by glycolic acid can obviously improve comedoes, and oral medicines combined with IPL can obviously improve papules.

ABSTRACT

Objective@#To understand the prevalence of HIV (human immunodeficiency virus) testing and associated factors among sexual active college students in Zhuhai City.@*Methods@#From November to December, 2019, an anonymous electronic questionnaire was administered among 12 235 students in six colleges and universities in Zhuhai City by multistage sampling. A total of 1 789 college students ever had sex were selected. Pearsons Chisquare test and nonconditional Logistic regression model were applied to analyze the factors associated with uptake of HIV testing.@*Results@#Among these students ever had sex, 7.55% (135/1 789) had been tested for HIV mainly through hospitals (71.85%). The main reasons for testing were regular testing (50.37%) and intending to know their infection status (23.70%). Multivariable Logistic regression showed that homosexual individuals (OR=4.62, 95%CI=1.07-19.95) and those who had heterosexual commercial sex in the past year (OR=3.77, 95%CI=1.96-7.26) were more likely to test for HIV, while female (OR=0.41, 95%CI=0.24-0.69) were less likely to test for HIV.@*Conclusion@#The proportion of HIV testing was low among sexual active college students in Zhuhai City. Interventions should be tailored including strengthening the HIV testing propaganda education and enhancing students awareness of HIV testing, and the influencing factors such as gender, sexual orientation and commercial sexual behavior should be taken into consideration, so as to improve the HIV testing coverage of this population.

The difficulty of neural localization during operation often leads to the increased risk of nerve injury, which results in neurological dysfunction, so how to accurately locate the nerves in surgeries, such as operations for patients with recurrent parotid tumors and prostate surgery, has become one of the key factors for the success of the operation. There are many methods to locate nerves in real time during operation, among which fluorescence imaging has attracted more and more attention due to its unique advantages of high sensitivity, easy to use, low cost and no radiation. In this article, the related research progresses of fluorescence imaging in neural localization are reviewed.

Objective@#To investigate the soft and hard tissue morphology in the infrazygomatic crest zone by observing the cone-beam computed tomography (CBCT) scans in patients with mini-implants.@*Methods@#CBCT scans of 43 patients performed from January 2014 to December 2016 in the Department of Stomatology, Beijing Friendship Hospital, Capital Medical University, with 84 existing mini-implants in the infrazygomatic crest region were collected. The bone thickness and cortical bone thickness were measured in the palatal and buccal side of the mini-implant. The maxillary sinus membrane thickness, sinus septa, reverse fold, and the sinus opening angle were also determined and analyzed in the maxillary posterior region.@*Results@#The bone thicknesses in the buccal and palatal side of the existing mini-implant were 2.5 (1.5, 3.2) and 5.2 (4.0, 6.4) mm, respectively. However, the corresponding cortical bone thicknesses were only 2.1 (1.3, 2.8) and 1.5 (1.0, 1.9) mm, respectively. The prevalences of the septa and the reverse fold were 33% (28/84) and 45% (38/84), respectively. The sinus opening angle was the largest in the mid-coronal plane of the maxillary first molar (71.6°±15.6°). In the coronal plane located at 10 mm mesially, the angle was the smallest (46.1°±18.0°), and in this area, 20% (16/82) of the angle was less than 30°.@*Conclusions@#The outer cortical plate of the infrazygomatic crest along with the cortical plate of sinus floor could be related to the initial stability of mini-implant anchorage. The anatomic variation such as the reverse fold indicated thorough consideration of insertion depth and angle to avoid unexpected sinus injury.

ABSTRACT

Purpose To investigate the clinicopathological features of the adult rhabdomyoma (A-RM) in the oral and maxillofacial region.Methods Three cases of A-RM in the oral and maxillofacial region were analyzed for the clinical symptoms,imaging data,pathological features,differential diagnosis,treatment,and prognosis with the literatures review.Results The age arranged from 4 yea~ to 83 years in three A-RM cases.Two cases were male and one case was female.Two cases occurred in the mouth floor and one case in the tongue.Three A-RM cases presented as slowly growing,painless masses clinically.The masses were lobular,red-brown nodules with clear boundary.Histologically,they consisted of densely packed,large,round and polygonal cells with abundant granular eosinophilic cytoplasm.Cross striations and vacuoles were seen in the cytoplasm.Tumor cells expressed desmin and myoglobin,partially expressed SMA,MyoD1 and S-100 protein.PAS was also positive among tumor cells and could be digested by the diastasum diastace.Conclusion Histological examination is the main diagnostic basis for A-RM.For accurate diagnosis,cross striations and immunohistochemistry for myogenic proteins in cytoplasm are of pivotal importance.Radical resection is the main treatment with generally promising prognosis.

BACKGROUND: The target gene cannot be deleted at specific time by using conditional gene knockout system. Most gene knockout animals cannot survive during embryonic development and after birth, which limits the study of gene function. OBJECTIVE: To review the recent advances about time-specific gene knockout technology. METHODS: An online search in PubMed was performed from the initial stage of database establishment to August 2014. The key words were gene knockout, inducer and Cre/loxP system. The repeated experiment and unrelated articles were excluded during primary search. A summary was made by analyzing time-specific gene knockout technology. RESULTS AND CONCLUTION: Most important gene knockout animals could not survive after birth, or die at early embryonic stage due to serious physical defects induced by an important gene deletion. Therefore, the progress in the functionality of a gene at a particular time, the genetic characteristics of an individual organism in a particular developmental period, and the treatment of certain acquired disease at a given time have been limited because of the limitations of research methods. However, the occurrence of conditional gene knockout animals, especial y time-specific knockout animals, provides a research model for the study of these areas. This study reviewed the recent advances, development and application about time-specific gene knockout animals, and discussed the significance and broad prospects of time-specific gene knockout technology of specific genes in the field of scientific research.

Objective To analyze the characteristics of the rpoB, KatG and inhA genes mutations in rifampin and isoniazid resistant Mycobacterium tuberculosis (MTB) clinical isolates in Baise district, Guangxi autonomous region. Methods 128 MTB clinical strains were collected and isolated for drug susceptibility testing, and drug resistant strain DNA was subtracted for rpoB, KatG and inhA genes mutation analysis. Results 75%(27/36)isolates carried mutations in the rpoB gene,and 59.3%(16/27)isolates carried mutations in 531 sites. 44.1%(15/34) isolates carried mutations in KatG or inhA, and 66.7%(10/15) isolates appeared in KatG 315 site, with two new mutations found in KatG 279 and 427 site. In these mutation isolates, 13.3%(2/15) mutations appeared in inhA 5, 6.7%(1/15) in inhA 16, and 20%(3/15) in both katG and inhA. Conclusions The mutation of rpoB, katG and inhA genes in TB is highly correlated with its resistance to rifampin and isoniazid in Baise district, Guangxi autonomous region. The study will provide a basis for further understanding the anti-bacterium mechanism and quick diagnostic methods for drug-resistant tuberculosis.

ABSTRACT

Aim To investigate the expression of CD31 , CD105 , HIF-1αand VEGF in xenografts tumor of HT-29 tumor cells during different stages of growth. Methods HT-29 tumor cells were transplanted into nude mice, the tumor was removed when the tumor volumes reached 300 mm3 respectively. Immunochemical method was a-dopted to detect the expression of CD31 , CD105 , HIF-1α and VEGF. Results HT-29 xenografts tumor vol-umes 300 mm3 showed expressions with CD31-MVD at 37.40±4.17 , 18.80±1.72 and 14.20±2.23 respectively; CD105-MVD at 22.80 ±3.54 , 15.60 ±1.35 and 10.20 ±2.48; positive expression rate of VEGF was 26.20% ±0.83%,40.73% ±6.29% and 13.41% ±1.20%respectively; while positive expression rate of HIF-1αwas 3.20% ± 2.97%, 11.89% ± 1.94% and 80.62% ±3.47% respectively. On the other hand, for different volumes group, CD31-MVD, CD105-MVD, VEGF and HIF-1αexpression ratios had signifi-cant differences ( P <0.01 ) . Conclusions The ex-pression of MVD and vascular-related factors within the tumor caused by HT-29 xenografts tumor in nude mice at different growth stages was varied. There was a cer-tain correlation between tumor volume and MVD, VEGF, HIF-1α.

ABSTRACT

<p><b>OBJECTIVE</b>To compare the ultra-microscopic changes of periodontal Ruffini's corpuscle induced by different patterns of tooth movement, and investigate the influence of different changes of the periodontal mechanical environment on the periodontal mechanoreceptor.</p><p><b>METHODS</b>Thirty-two eight-weeks-old SD rats were divided into control group (n = 4), none-extraction group (n = 12) and extraction group (n = 12), and none-extraction group and extraction group were further divided into three subgroups, namely 3 day and 14 day and 28 day. For control group, no intervention was performed. For none-extraction group and extraction group, the following interventions were conducted. In none-extraction group, the maxillary left and mandibular right third molars were moved distally. In extraction group, the maxillary left third molar was moved distally, and the bilateral mandibular third molars were extracted. The ultra-structures of periodontal Ruffini's corpuscle in the periodontal ligament of the distal root of the bilateral maxillary third molars were observed under the transmission electron microscope.</p><p><b>RESULTS</b>The ultra-structrural changes in the none-extraction group were mainly characterized by degeneration or abnormal distribution of mitochondria in the axon terminal, which were almost recovered at 28 d. In the extraction group, the changes were mainly characterized by deficiency or abnormal elongation of the Schwann sheath and were not recovered at 28 d.</p><p><b>CONCLUSIONS</b>The ultra-structures of periodontal Ruffini's corpuscle might be influenced by tooth movement and occlusal changes, and the mechanorecepting function of it might be affected by changes of the periodontal mechanical environment.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To examine the expression of nucleotide-binding oligomerization domain 1 (NOD1), nuclear factor-kappa B (NF-κB) and human beta-defensins in candidal albicans leukoplakia and to investigate the effect of candida albicans infection on key proteins in NOD1 signaling pathway and the expression of human beta-defensin.</p><p><b>METHODS</b>Forty cases of oral leukoplakia samples were collected and stained by hematoxylin-eosin staining, periodic acid-Schiff staining, silver staining and immunohistochemical methods. Nineteen samples were positive with these four methods and judged as candidal albicans leukoplakia, and the other twenty- one samples judged as leukoplakia without candidal albicans infection. Western blotting was used to detect the expressions of NOD1 and NF-κB in these forty samples. In addition, the immunohistochemical method was adopted to investigate the relationship between NOD1, NF-κB, human beta-defensin 1, 2, 3 expressions and candida albicans.</p><p><b>RESULTS</b>The positive rate of candida albicans in oral leukoplakia was 48% (19/40). The expressions of NOD1 and NF-κB in the candida albicans leukoplakia were lower than that in leukoplakia without candida albicans infection. The mean optical density value of NOD1, NF-κB, human beta-defensin 1, 2, 3 in candidal albicans leukoplakia were 0.25 ± 0.01, 0.30 ± 0.02, 0.35 ± 0.02, 0.42 ± 0.03, 0.36 ± 0.02 respectively, which were significantly lower than that in leukoplakia without candida albicans infection (0.31 ± 0.02, 0.47 ± 0.03, 0.42 ± 0.02, 0.53 ± 0.04, 0.47 ± 0.03) (P < 0.05).</p><p><b>CONCLUSIONS</b>By inhibiting the NOD1 signaling pathway, candida albicans infection may reduce the expression level of human beta-defensin 1, 2, 3 in oral leukoplakia.</p>

ABSTRACT

Objective To investigate the effects of infrasound at different frequencies on the pathological morphology of mouse testes and to explore the possible involvement of β-cateinin in the mechanism of injury. MethodsSixty adult BALB/C mice were randomly divided into 2 groups.One group of 6 mice were the controls without infrasound exposure.The other group with 54 mice was evenly divided into 3 subgroups of 18 exposed to 130 dB infrasound at 4 Hz,8 Hz or 20 Hz for 2 hours daily.The pathological morphology of the mice's testes was observed 3,9 and 21 h after exposure.Immunohistochemical staining was used to examine β-catenin expression 3 h after exposure.ResultsMarked damage was observed in the morphology of the testicular cells of rats in the 8 Hz group.Cell denaturation was found in germinal cells.The cell structure was loosened and the cell junctions had decreased or disappeared.The damage was most serious at 3 h after exposure.But at the same time point,pathological damage of testicular cells was not obvious in the 4 Hz group,with only a few testes cells swollen.The morphology of testicular tissue of mice in the 20 Hz subgroup exhibited no significant difference from the control group.β-catenin was mainly found in the membranes of Leydig cells,Sertoli cells and myoepithelial cells in all of the mice,and there was no significant change in the expression level or distribution pattern between the exposed mice and the controls.No expression ofβ-catenin was found in spermatogenic cells. ConclusionInfrasound at 4 Hz or 8 Hz can damage testicular tissue,at least in mice.At 20 Hz there is no significant effect on the testes.The loosened cell structure might have been caused by the mechanical force produced by the infrasound and not by any decrease of β-catenin.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the potential role of BME-10X collagen/hydroxyapatite (HA) bone graft in periodontal tissue regeneration.</p><p><b>METHODS</b>Four 18 months old male beagles in the experiment were treated with non-surgical periodontal therapy. At the sites of mandibular 3rd and 4th premolars at the time of dogs were treated with non-surgical periodontal therapy one week later, the teeth with the same name in the same jaw were selected to the experimental group (T group) or the control group (C group) at random. The defects in T group were filled with BME-10X collagen/HA bone graft while the defects in C group were filled with nothing. The dogs were sacrificed in twelve weeks and analyzed by histopathology.</p><p><b>RESULTS</b>The defects in T group got more tissue regeneration compared with the defects in C group. The height of new bone (NB) was (3.01 +/- 0.14) mm in T group versus (1.32 +/- 0.11) mm in C group (P < 0.05). The height of new periodontal ligament (NP) was (3.12 +/- 0.19) mm in T group versus (1.35 +/- 0.12) mm in C group (P < 0.05). The height of new cementum (NC) was (3.30 +/- 0.15) mm in T group versus (2.70 +/- 0.12) mm in C group (P > 0.05). The new tissue guided by the bone graft was the same as the normal tissue in histopathology analysis.</p><p><b>CONCLUSION</b>The results of the study suggest that BME-10X collagen/HA bone graft is a good bone graft for periodontal tissue regeneration.</p>

ABSTRACT

Objective To study the genotyping of clarithromycin-resistant Helicobacter pylori(Hp) isolates.Methods From March 2007 to October 2007,247 gastric mucosa specimens were collected by the endoscopy from the patients with peptic ulcer or gastritis at the affiliated hospital of Youjiang Medical College for Nationalities.A total of 126 Hp strains were isolated.Resistance to clarithromycin in Hp was determined by E-test.All of the resistant isolates were genotyped with REP-PCR and further clustered with NTsys_2 software.The clinical data was collected for these patients with clarithromycin-resistant Hp.Results The 26 clarithromycin-resistant isolates from the west of Guangxi were divided into six genotypea including Group Ⅰ,Group Ⅱ,Group Ⅲ,Group Ⅲ,Group Ⅳ,Group Ⅴ and Group Ⅵ according to the homology of 78%.Every group have 2,11,1,8,3,1 strains Hp,respectively.Strains of group Ⅱ were isolated from patients with peptic ulcer and most of them from Chuang patients.All strains of group Ⅳ were isolated from patients with gastritis.Conclusions The clarithromycin-resistant isolates were divided into six groups by REP-PCR Disease type.nationality of patients and family history of stomach diseases were associated with the genotypes.

ABSTRACT

Objective To investigate renal tissue changes in rats exposed to 16 Hz/130 dB infrasound.Methods Twenty male Sprague-Dawley adult rats were randomly divided into a control group,a pseudo-infrasound group,and two infrasound groups-A and B.Groups A and B were exposed to 16 Hz/130 dB infrasound for 2 h or4 h daily respectively over 7 days.Pathological and uhrastruetural changes in their renal tissues were observed with a light microscope and an electron microscope.Results Under the light microscope,Bowman's capsules expanded,epithelial cells were shed,and a little secretion was found in the renal tubules of infrasound group A.Slight degeneration and necrosis of the renal tubules and glomerular exudation could be observed in group B.Under the electron microscope,a large number of lysosomes displayed hyperplasia,there was interstitial edema,and leukocyte pavementing wag found in group B.Other changes such as swelling of podocytes' processes,fusion of foot processes,and vacuolization in the mitochondria could be observed in both infrasound groups.Conclusion Lengthy exposure to 16 Hz/130 dB infrasound can cause severe pathological and uhrastruetural changes in renal tissue,at least in rats.

ABSTRACT

Objective: To identify the different protein expression profiles between human oral squamous cell carcinoma (OSCC) and normal oral mucosa tissues, and provide experimental data for further study of the development mechanism of OSCC. Methods: 10 cases of OSCC and paired normal oral mucosa tissues were collected and analyzed through two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS). Results: (1) The average protein spots of OSCC were 2 325±390, while that of normal oral mucosa tissues were 2 487±281. (2) 29 differential protein spots were found between OSCC and normal oral mucosa. Moreover, these protein spots were all down- regulated in OSCC compared with normal oral mucosa. Among these spots, 3 were identified as fibrin beta, triosephosphate isomerase (TIM) and unknown protein through mass spectrometry and bioinformation. Conclusion: Down-regulation of fibrin beta, Triosephosphate isomerase(TIM) and unknown protein are found in the development of OSCC and the mechanism needs further study.

ABSTRACT

This study sought to shed light on the killing effect of peripheral blood mononuclear cells (PBMCs) irradiated by gamma ray at a dose of 1 Gy on cultured human gastric tumor cell line MKN-28. The radiation dose rate of 17 Gy/min was used. The groups in the experiment were MKN-28 cell control group, PBMCs control group, MKN-28 tumor cells with irradiated or non-irradiated PBMCs co-cultured groups. Radiation dosage was one Gray, acridine orange/ethidium bromide (AO/EB) staining was used for observation of the killing effects of PBMCs on tumor cells in different period. Cells were harvested 240 h later and observed by transmission electron microscopy. The result showed the living period of irradiated PBMCs was shorter than that of non-irradiated PBMCs. In the irradiated and non-irradiated groups,a few PBMCs were still alive after being cultured for 240 h, but the cell volume was larger than that of lymphocytes. These cells were identified as monocytes (95%) or DCs (5%) by transmission electron microscopy. The co-culture of irradiated PBMCs and MKN-28 cells showed that tumor cells were eliminated after 96 h. As compared with the non-irradiated goup, the irradiated PBMCs had more potent ability for killing tumor. The results demonstrate that 1 Gy gamma irridiation can improve the killing effect of PBMCs on the tumor cells, and that 1 Gy gamma irritation can also induce shorter living period of lymphocytes in PBMCs cultured in vitro, but such irritation has little effect on the living period of monocytes and DCs in PBMCs.
