ABSTRACT

Background@#Porcine parvovirus (PPV) is a single-stranded DNA virus that causes porcine reproductive failure. It is of critical importance to study PPV pathogenesis for the prevention and control of the disease. NS1, a PPV non-structural protein, is participated in viral DNA replication, transcriptional regulation, and cytotoxicity. Our previous research showed that PPV can activate nuclear factor kappa B (NF-κB) signaling pathway and then up-regulate the expression of interleukin (IL)-6. @*Objectives@#Herein, the purpose of this study is to determine whether the non-structural protein NS1 of PPV also has the same function. @*Methods@#Real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR), enzyme-linked immunosorbent assay, western blot, immunofluorescence assay and small interfering RNA (siRNA) were used. @*Results@#Our findings demonstrated that PPV NS1 protein can up-regulate the expression levels of IL-6 and tumor necrosis factor-alpha in a dose-dependent manner. Moreover, PPV NS1 protein was found to induce the phosphorylation of IκBα, then leading to the phosphorylation and nuclear translocation of NF-κB. In addition, the NS1 protein activated the upstream pathways of NF-κB. Meanwhile, TLR2-siRNA assay showed TLR2 plays an important role in the activation of NF-κB signaling pathway induced by PPV-NS1. @*Conclusions@#These findings indicated that PPV NS1 protein induced the up-regulated of IL-6 expression through activating the TLR2 and NF-κB signaling pathways. In conclusion, these findings provide a new avenue to study the innate immune mechanism of PPV infection.

ABSTRACT

Objective To explore the clinical effect of rosuvastatin in treatment of unstable angina and the influence of levels on blood lipids,hypersensitive C-reactive protein(hs-CRP),homocysteine(Hcy)and cardiac troponin(cTnl).Methods 200 patients with unstable angina pectoris(UAP)in our hospital from March 2014 to March 2016 were selected as the subjects of the study,they were randomly divided into control group and experimental group,each group had 100 cases.The control group was given conventional treatment,the experimental group was given rosuvastatin on the basis of conventional treatment,the clinical efficacy between two groups was observed and compared,and the levels of blood lipid and C reactive protein(hs-CRP),homocysteine(Hcy)troponin I(cTnl)pre-and post-treatment were detected.Results The total effective rate of the experimental group was 92%and the control group was 80%,the difference was statistically significant(P<0.05).Before treatment,the blood lipid,hs-CRP,Hcy and cTnl between two groups showed no significant difference; after treatment,the blood lipid,hs-CRP,Hcy and cTnl in both two groups were decreased,high density lipoprotein cholesterol(HDL-C)levels than before treatment significantly increased,and the experimental group decreased or increased significantly compared with the control group,the difference was statistically significant(P<0.05).Conclusion Rosuvastatin has remarkable curative effect in clinical treatment for UAP,not only can significantly reduce the blood lipid,and reduce inflammation in patients,the clinical treatment of unstable angina patients have help,it is worthy of clinical application.

ABSTRACT

Objective To analyzethe expression level of peripheral blood mononuclear cell Toll like receptor 4(TLR4)in patients with acute coronary syndrome(ACS)undergoing elective percutaneous coronary intervention(PCI),and intensive atorvastatin on Toll like receptor 4 in clinical treatment.Methods 100 cases of patients with ACS were selected in our hospital from May 2014 to May 2016 were randomly divided into control group and experimental group,control group were given atorvastatin treatment after admission,20mg/d PCI postoperative continuous,after one month still maintain dose of 20mg/d.The experimental group were given atorvastatin treatment after admission,40mg/d,PCI postoperative continuous,after one month reduced to 20mg/d.After admission and a course of treatment,the expression level of peripheral blood mononuclear cells TLR4 was detected by flow cytometry and the Gensini scoring system on coronary lesion degree.At the same time,50 healthy subjects were selected as normal group.Results Compared with healthy subjects,the peripheral blood mononuclear cells TLR4 level in ACS patients has been significantly increased,and the level in patients with unstable angina pectoris is far lower than the level increased in patients with acute myocardial infarction,triple branch lesions groups were significantly higher than them in single and double branch lesion groups,the difference between the three obviously,with statistical significance(P<0.01); According to stepwise multiple linear regression and othercorrelation analysis showed that ACS category and the severity of coronary artery stenosis,coronary artery stenosis score were positively associated with the levels of TLR4(P<0.05); After treatment for one month,the levels of TLR4,the expression of low density lipoprotein cholesterol and total cholesterol levels were significantly lower,high density lipoprotein Cholesterol levels were significantly increased,the differences were statistically significant(P<0.01); compared with the normal group,the experimental group peripheral blood mononuclear cells of patients with Toll like receptor 4 expression level,total cholesterol,low density lipoprotein cholesterol level in the experimental group is higher than the normal group,high density lipoprotein cholesterol increased significantly,comparison between groups the differences were statistically significant(P<0.01).Conclusion The degree of lesions and the occurrence of acute coronary syndrome in patients with peripheral blood mononuclear cells of Toll like receptor 4 expression level increase has certain effect,increase the dosage of atorvastatin can significantly reduce the ACS after PCI in patients with peripheral blood mononuclear cells of patients with Toll4 like receptor expression.

ABSTRACT

Objective To investigate the influence of rosuvastatin on inflammatory factors and immune func-tion in chronic obstructive pulmonary disease(COPD)with pulmonary heart disease.Methods 80 COPD patients with pulmonary heart disease were randomly divided into two groups,40 cases in each group.The control group was treated with conventional medicine comprehensive treatment,the observation group was given rosuvastatin based on the control group.The inflammatory cytokines (TNF -α,IL -1 and hs -CRP)changes,immunoglobulin IgM,IgG and IgA changes situation,and heart,lung function changes related indicators were compared between the two groups before and after the treatment.Results After treatment,the TNF -α,IL -1 and hs CRP levels of the observation group were significantly lower than those before treatment(t =145.5,35.35,57.5,all P <0.05)and those of the control group(t =160.75,11.45,32.99,all P <0.05).After treatment,the IgM,IgG and IgA levels of the observa-tion group were significantly lower than those before treatment (t =4.89 of 18.29,3.42,all P <0.05)and those of the control group (t =2.55 9.67,2.342,all P <0.05 ).After treatment,the FEV1 ,FEV1 /FVC and LVEF of the observation group were significantly higher than those before treatment(t =40.44 33.64,33.79,all P <0.05),while the PAP level was significantly lower than that before treatment (t =24.12,P <0.05),and the FEV1 ,FEV1 /FVC and LVEF of the observation group were significantly higher than those of the control group after treatment(t =16.30, 15.82,18.72,all P <0.05),while the PAP level was significantly lower than that of the control group after treatment (t =13.43,P <0.05).Conclusion Combined rosuvastatin therapy for COPD patients with pulmonary heart disease can effectively reduce body inflammatory response,improve immune function and myocardial contractility,so it can promote functional recovery of heart and lung in patients.

ABSTRACT

Objective To study effects of exectin hydrochloride and intensity modulated radiotherapy in treatment of patients with advanced lung cancer and its effect on levels of carcinoembryonic antigen (CEA), neuron-specific enolase (NSE) and serum glycoprotein antigen 199 (CA199). Methods 88 patients with advanced lung cancer were selected from June 2014 to June 2016 in our hospital were selected and randomly divided into observation group and control group, 44 cases in each group.In the control group, the patients were treated with IMRT, and the observation group was treated with exectin hydrochloride on the basis of control group.The clinical efficacy and adverse reaction were compared between the two groups.The levels of CEA, NSE and CA199 before and after treatment were analyzed.Results After treatment, the total effective rate in the observation group 84.09% was significantly higher than the control group 52.27%, the difference was statistically significant (P<0.05).Before treatment, the levels of CEA, NSE and CA199 of the two groups had no significantly different, after treatment, the levels of CEA, NSE and CA199 of the two groups were significantly lower than before treatment, the difference was statistically significant (P<0.05).CEA, NSE, CA199 levels in the observation group were significantly lower than the control group, the difference was statistically significant (P<0.05).There was no significant difference in adverse reaction rate of I-II, III-IV liver function, gastrointestinal reaction, bone marrow suppression and acute radiation pneumonitis in observation group and control group.Conclusion Ectectin hydrochloride combined with intensity modulated radiotherapy can effectively reduce the levels of CEA, NSE and CA199 in patients with advanced lung cancer, and the clinical efficacy is good, and will not increase the adverse reaction rate.

ABSTRACT

Objective To investigate the expressions of vascular endothelial growth factor (VEGF)-C,-D and VEGF receptor-3 (VEGFR-3/Flt-4) in pN0 early gastric cancer (ECG) and their relationship with lymph node micrometastasis.Methods From January 2005 to January 2010,the paraffin-embedded specimens of 61 pN0 ECG were collected.The expressions of VEGF-C,VEGF-D and VEGFR-3 in 61 pN0 early gastric cancer tissue,25 adjacent tissue and CAM5.2 expression in 868 hematoxylin-eosin staining negative lymph nodes were detected by immunohistochemical assay.The rate of lymph node micrometastasis of 61 pN0 ECG was evaluated.According to the data type,t test,x2 test or Fisher exact probability were performed for the relationship analysis between the expressions of VEGF-C,VEGF-D and VEGFR-3 in pN0 stage ECG and lymph node micrometastasis.Results The high expressions of VEGF-C,VEGF-D and VEGFR-3 in the 61 pN0 ECG were 34.4% (21/61),34.4% (21/61) and 44.3% (27/61) respectively,which were higher than those of adjacent tissues [12.0% (3/25),8.0% (2/25) and 16.0% (4/25) respectively] (x2=4.433,6.321 and 6.144 respectively,all P<0.05).There were 10 cases (16.4%) of pN0 ECG with lymph node micrometastasis.In pN0 ECG,the low expressions of VEGF-C and VEGFR-3 in negative Flt-4 vascular invasion (FVI) were common than positive FVI (x2 =15.828,6.879 and 9.244,all P<0.05).The high expressions of VEGF-C and VEGFR-3 were related to the depth of tumor invasion (x2 =5.561 and 5.678,both P<0.05),the density of VEGFR 3 positive vascular (FVD) (t=2.987and 5.652,both P<0.01) and lymph node micrometastasis (x2 =6.705 and 6.192,both P<0.05),but not related to the degree of differentiation (P>0.05).However,the high expression of VEGF-D was not related to depth of tumor invasion,FVD and lymph node micrometastasis (all P>0.05),but related to the degree of differentiation (x2 =8.472,P =0.004).The high expressions of VEGF-C,VEGF-D and VEGFR-3 were not related to gender,age,tumor location,macroscopic type and tumor size (all P>0.05).Conclusions The high expressions of VEGF-C,VEGFR-3 were related to lymph node micrometastasis in pN0 ECG.Although the high expression of VEGF-D was not related to lymph node micrometastasis,it could indirectly affect lymph node micrometastasis through VEGF-C-VEGFR-3 axis in pN0 ECG.
