ABSTRACT
The GapC protein of Streptococcus uberis located on the surface of bacteria is a protein with glyceraldehyde-3-phosphate dehydrogenase activity. It participates in cellular processes and exhibits a variety of biological activities. In addition, it has good antigenicity. The aim of this study was to predict the possible B-cell epitopes of the GapC protein and verify the immunogenicity of candidate epitope peptides. The gapC gene of S. uberis isolate RF5-1 was cloned into a recombinant expression plasmid pET-28a-GapC and inducibly expressed. The purified protein was used to immunize experimental rabbits to produce anti-GapC polyclonal antibodies. The three-dimensional structure and three-dimensional location of the GapC B-cell epitopes and the homology comparison of the GapC protein and its B-cell epitopes were carried out using bioinformatics softwares. The results showed that the 44-kDa GapC protein had a good immunological reactivity. Six linear and 3 conformational dominant B-cell epitopes against the GapC protein were selected and synthesized. Three dimensional analysis indicated that the selected peptides have better antigen epitope formation potential. Rabbit anti-GapC polyclonal antibodies were generated after immunized with the purified GapC protein, and the polyclonal antibodies were used to identify the epitope peptide by an indirect ELISA. The ELISA results showed that all of the 9 epitope peptides could react with anti-GapC polyclonal antibodies with varying titers. Among them, the epitope polypeptide 266AANDSYGYTEDPIVSSD282 reacted with the polyclonal antibodies significantly stronger than with other epitope peptides. This study laid an experimental foundation for in-depth understanding of the immunological properties and utilizing effective epitopes of the GapC protein of S. uberis.

Objective@#To understand mental health status of middle school students in Weifang, and to analyze its association with health risk behaviors, and to provide a reference basis for the effective prevention of mental health problems.@*Methods@#Using the self-designed questionnaire and Kessler l0 (K10), a total of 3 185 middle school students in Weifang selected by using stratified clustered sampling were investigated. Chi-square test and binary Logistic regression was used for statistical analysis.@*Results@#Among the middle school students in Weifang, 1 634(51.3%) reported good mental health, 854(26.8%) in moderate level, 394(12.4%) reported poor mental health, and 303(9.5%) reported very poor mental health. Sleep duration and smoking were associated with mental health of middle school students (P<0.05). Logistic regression analyses showed that, compared with adolescents with sleep duration less than 8 h, mental health among students whose sleep duration ≥8 h was better(OR=0.36); compared with non-smokers, students who smoked showed worse mental health status (OR=3.53).@*Conclusion@#Health-risk behaviors are closely related to mental health of middle school students. Relevant departments should strengthen education and publicity of healthy lifestyles, and pay close attention to the impact of health risk behaviors on mental health.

Objective@#To explore the changes of awareness of tuberculosis and access to tuberculosis-related information among students in Zhejiang Province.@*Methods@#There were 554 students younger than 30 y were included in the study during April to sept. of 2014 and during sept. to Nov. of 2018. Univariate and multivariate Logistic regression were conducted to find the related factors of knowing about tuberculosis and related knowledge. Chi-square test was used to analyze the changes of ways access to tuberculosis-related information.@*Results@#There were 440 participants (79.4%) aware of tuberculosis, including 215(85.7%) in 2014 and 225(74.3%) in 2018. The factors affecting awareness on tuberculosis were female ( AOR=1.60, 95%CI =1.04-2.47) vs male; and junior middle school( AOR=1.99, 95%CI =1.03-3.85), high school( AOR=3.27, 95%CI =1.57-6.81), college or higher ( AOR=6.73,95%CI =2.55-17.76) vs primary school or lower; year 2018 ( AOR=0.50, 95%CI =0.31-0.80) vs 2014. There was no significant statistical difference in the overall knowledge scores of tuberculosis between the students in the two surveys. In 2018, the proportion of tuberculosisrelated information accessed from the Internet (forum, WeChat, Weibo.) and community activities (assembly promotion)(75.10%, 26.70%) was higher than that of 2014(36.20%, 13.10%)( χ 2=68.88, 12.79, P <0.05).@*Conclusion@#The proportion of students aware of tuberculosis and TB control in Zhejiang Province were not significantly improved. Higher proportion of students get tuberculosis-related information from the new media. Therefore, it is necessary to strengthen health education of tuberculosis in schools, especially through the usage of new media apart from traditional pathways.
ABSTRACT
Objective:The consensus molecular subtype (CMS) classification is based on the gene expression profiles, This article attempts to conduct a preliminary exploration and analysis of the clinical features of different CMS patients. We can give an individualized diagnosis and treatment for the patients.Methods:Seven GSE series of colorectal cancer gene expression profiles were downloaded by R software from the GEO database. A total of 1414 patients were included. Using the specific computational method published by Peter in 2017, the patients were divided into four groups: CMS1, CMS2, CMS3 and CMS4. The measurement data is expressed by Mean± SD, and the count data is expressed by n(%). The software of SPSS 18.0 was used to conduct analysis. Results:CMS1 tumors originated in the right colon (77.4%), while CMS2 mostly originated in the left colon (72.8%). The proportion of T 4 stage in CMS2 was 16.6%, while in the other three types was 23.3%, 29.3% and 24% respectively; the proportion of distant metastasis of CMS1 was the lowest (3.5%), while the distant metastasis rate of CMS4 was 18.2%. The KRAS mutation rates in CMS1 and CMS2 were 25.6% and 30.3% respectively, while in CMS3 it was up to 73.9%; the BRAF mutation rate in CMS1 was 45.5%, while the other three mutation rates were 0.6%, 6.2% and 5.9%, respectively; The average mutation rate of APC was 59.45%. Overall survival and progression-free survival analysis showed that the CMS4 interstitial type was the worst, while the CMS2 classic type had the best relative prognosis. Conclusions:CMS1 immunotype, the tumor has origin from right colon in female patients. MSI-H is often accompanied by BRAF gene mutation, this type patients has a refractory treatment response and poor prognosis. The classic CMS2 type is characterized by APC deletion mutation and Wnt activation, with good therapeutic effect and good prognosis. CMS3 metabolic type often harbor KRAS mutation, anti-EGFR treatment is not sensitive. Although this type is prone to relapse, but the chemotherapy is effective, so the overall survival prognosis is acceptable. CMS4 interstitial type with left colon tumor is prevalence. due to the activation of TGF-β and enhanced angiogenesis, this type tumor is prone to metastasis to distant site and has the worst prognosis.
ABSTRACT
OBJECTIVE: To investigate the effects of total flavonoids of Jasminum nudiflorum (TFJN) on pancreatic islet function in type 2 diabetes mellitus (T2DM) model mice and its possible mechanism. METHODS: Totally 10 Kunming mice were included in blank group and given normal diet. Other 75 mice were included in model group and given high-lipid and high-glucose diet for 4 weeks combined with streptozotocin once via sublingual vein to induce T2DM model. After modeling, the mice were randomly divided into model group, metformin hydrochloride group (positive control, 350 mg/kg), TFJN low-dose, medium-dose and high-dose groups (100, 200, 400 mg/kg, by crude drug), with 10 mice in each group. Blank group and model group were given constant volume of normal saline intragastrically; administration groups were given relevant medicine intragastrically; once a day, for consecutive 6 weeks. The level of fasting dynamic blood glucose was measured after modeling (before medication) and 2, 4, 6 weeks after medication. The histomorphological characteristics of pancreatic tissue were observed by HE staining. The contents of serum insulin (INS), insulin cell antibody (IAA), TNF-α, IL-1β, IL-6, IL-18, CRP, leptin (LP) and adiponectin (ADPN) were measured by ELISA. RESULTS: Compared with blank group, islet atrophy, blurred boundary, insufficient cytoplasm and smaller volume of pancreatic tissue were observed in model group. The fasting dynamic blood glucose level (at different time points after medication), the contents of IAA, TNF-α, IL-1β, IL-6, IL-18, CRP and LP were increased significantly, while the contents of INS and ADPN were decreased significantly (P<0.01). Compared with model group, above symptoms of pancreatic tissue in administration groups were relieved, and the fasting dynamic blood glucose level (2, 4, 6 weeks after administration in metform in hydrochloride group, 4, 6 weeks after administration in TFJN groups), the contents of IAA, TNF-α, IL-1β, IL-6, IL-18, CRP and LP were decreased signifi- cantly, while the contents of INS and ADPN were increased significantly (P<0.05 or P<0.01). CONCLUSIONS: TFJN can repair the damaged pancreas of mice with T2DM, reduce the level of blood glucose, and improve the sensitivity of the body to insulin, the mechanism of which may be associated with reducing the secretion of inflammatory factors.

Objective To explore the serum lipid metabolism and immunoregulation of patients with primary nephritic syndrome (PNS) .Methods 50 PNS patients were served as the test group and 50 healthy people the control group .Automatic biochemical analyzer was utilized to detect their serum low density lipid-cholesterol (LDL-C ) ,high density lipid-cholesterol (HDL-C ) , triglyceride(TG) ,total cholesterol(TC) ,apolipoprotein(Apo)A1 ,ApoB ,and lipoprotein (a) .Immunization rate nephelometry was employed to measure their serum IgG ,IgA ,IgM ,complement C3 ,complement C4 ,CD3+ ,CD4+ ,CD8+ and CD4/CD8 .Results Compared with the control group ,serum levels of TC ,TG ,LDL-C ,ApoB ,ApoA1 ,IgM and CD4/CD8 of patients in the test group were significantly higher ,while those of HDL-C ,IgG ,complement C3 ,complement C4 ,CD3+ ,CD4+ ,CD8+ were obviously lower , with both statistically significant differences (P0 .05) .Conclusion Serum lipid level of PNS patients is higher than healthy people ,and considerable loss of Ig and complements and T cell subsets disproportion results in humoral and cellular immune dysfunction .
ABSTRACT
<p><b>OBJECTIVE</b>To establish an everted intestinal sac method for determining absorption ingredients of yuanhuzhitong pill and study absorption characteristics of major chemical constituents of yuanhuzhitong pill.</p><p><b>METHOD</b>The everted intestinal sac model was adopted. Intestinal sac fluid samples at different time points after administration of three concentrations of Yuanhuzhitong pill were collected and determined by HPLC. The accumulative absorbed doses of active constituents were calculated, while the proportion between samples of yuanhuzhitong pill and absorption ingredients was compared.</p><p><b>RESULT</b>Eight ingredients of Yuanhuzhitong pill can be detected in intestinal sac, they are protopine, palmatine, coptisine, imperatorin, berberine, byakangelicin, alpha-allocryptopine and tetrahydropalmatine. The absorption rate constants (Ka) of eight constituents increased in jejunum and ileum with the increase in concentration of Yuanhuzhitong pill extracts (P < 0. 05), indicating a passive absorption.</p><p><b>CONCLUSION</b>As ingredients are selectively absorbed in intestinal sac, the everted intestinal sac method is selected to assess the intestinal absorption characteristics of ingredients of Yuanhuzhitong prescription.</p>

Objective To study on the significance of accompany relatives of children with access to pediatric transplantation ward on clinical treatment and nursing.Methods 37 children undergoing autologous peripheral blood stem cell transplantation (APBST) were collected from January 2007 to April 2010.All cases were divided into the no tending group(7 cases)and the tending group (30 cases).The accompany relatives of the tending group accepted standard training before entering the ward.The compliance with basic nursing,acute side-effect and hemopoiesis rebuild period of marrow during APBST in two groups were compared.Results The compliance with basic nursing of the no tending group was only 28.6%,but was 73.3% in the tending group.According to Bearman standard,the acute side-effect of the no tending group:1 case of grade Ⅳ,1 case of grade Ⅲ,4 cases of grade Ⅱ,1case of grade Ⅰ; in the tending group:1 case of grade Ⅳ,1 case of grade Ⅲ,6 cases of grade Ⅱ,22 case of grade Ⅰ.The marrow-reconstitution took place in 36 children.The most long time was 41 days in the no tending group,the average time was (28.14+7.98)days.The time took for marrow-reconstitution was 22 days in the tending group,the average time was ( 14.61+2.33)days.Conclusions It is safe and feasible for accompany relatives of children to participate in nursing during transplantation.Relatives accompany is beneficial for psychology adaptability,nursing compliance and success of APBST.
