ABSTRACT
Objective To investigate the value of multiparametric CT features for predicting the risk classification of gastric stro-mal tumor(GST).Methods The clinical data from 139 patients with GST were retrospectively collected.According to the patho-logical risk results, the patients were divided into two groups:a low-risk GST group(including very low-and low-risk)with 75 patients and a high-risk GST group(including medium and high-risk)with 64 patients. The CT features between low-risk GST group and high-risk GST group were compared using chi-squared test or t-test.The risk factors of high-risk GST were identified by univariate analysis. The prediction models were built by multivariate logistic regression. The performance of models were evaluated by receiver oper-ating characteristic(ROC)curve. Results There were significant differences in the maximum tumor diameter, minimum tumor diameter, arterial phase enhancement degree, venous phase enhancement rate, arterial phase enhancement degree rate, venous phase enhance-ment degree rate, cystic, and necrosis between low-risk GST group and high-risk GST group, which were associated with the risk classification of GST.The area under the curve(AUC)of the quantitative features-based model that combined maximum tumor diam-eter,minimum tumor diameter, arterial phase enhancement degree, venous phase enhancement rate, arterial phase enhancement degree rate and venous phase enhancement degree rate, showed a significantly higher performance than the qualitative features-based model that incorporated cystic and necrosis(0.981 vs 0.850,P<0.001).Conclusion Maximum tumor diameter, minimum tumor diameter, arterial phase enhancement degree, venous phase enhancement rate, arterial phase enhancement degree rate, venous phase enhance-ment degree rate,as well as cystic and necrosis, are associated with the risk classification of GST and can predict the high-risk GST.
ABSTRACT
The theory of membrane anatomy has been widely used in the field of colorectal surgery. The key point to perform high quality total mesorectal excision (TME) and complete mesocolic excision (CME) is to identify the correct anatomical plane. Intraoperative identification of the various fasciae and fascial spaces is the key to accessing the correct surgical plane and surgical success. The landmark vessels refer to the small vessels that originate from the original peritoneum on the surface of the abdominal viscera during embryonic development and are produced by the fusion of the fascial space. From the point of view of embryonic development, the abdominopelvic fascial structure is a continuous unit, and the landmark vessels on its surface do not change morphologically with the fusion of fasciae and have a specific pattern. Drawing on previous literature and clinical surgical observations, we believe that tiny vessels could be used to identify various fused fasciae and anatomical planes. This is a specific example of membrane anatomical surgery.

The regulation of spermatogonial proliferation and apoptosis is of great significance for maintaining spermatogenesis. The single-cell RNA sequencing (scRNA-seq) analysis of the testis was performed to identify genes upregulated in spermatogonia. Using scRNA-seq analysis, we identified the spermatogonia upregulated gene origin recognition complex subunit 6 (Orc6), which is involved in DNA replication and cell cycle regulation; its protein expression in the human and mouse testis was detected by western blot and immunofluorescence. To explore the potential function of Orc6 in spermatogonia, the C18-4 cell line was transfected with control or Orc6 siRNA. Subsequently, 5-ethynyl-2-deoxyuridine (EdU) and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assays, flow cytometry, and western blot were used to evaluate its effects on proliferation and apoptosis. It was revealed that ORC6 could promote proliferation and inhibit apoptosis of C18-4 cells. Bulk RNA sequencing and bioinformatics analysis indicated that Orc6 was involved in the activation of wingless/integrated (Wnt)/ β-catenin signaling. Western blot revealed that the expression of β-catenin protein and its phosphorylation (Ser675) were significantly decreased when silencing the expression of ORC6. Our findings indicated that Orc6 was upregulated in spermatogonia, whereby it regulated proliferation and apoptosis by activating Wnt/β-catenin signaling.
ABSTRACT
Polysialic acid(PSA)is a homopolymer consisting of N-acetylneuraminic monomers linked by α-2,8 and(or)α-2,9 glucoside bonds. As an endogenous polysaccharide,PSA has good biocompatibility, biodegradability, high hydrophilicity,non-immunogenicity,long-term circulation, easy modification and specific targeting to selectins. In the field of drug delivery research,PSA not only can be connected with small molecule drugs, active peptides or proteins, but also can be grafted or electrostatic cross-linked with polymers to build a variety of drug delivery systems, such as nanogels, polymer micelles, liposomes, etc. It has shown great potential value in the treatment of various disease models such as tumors, inflammatory diseases and neurological diseases. In this paper, the biological functions of PSA,the classification of drug delivery systems based on PSA and its application progress were reviewed,in order to provide reference for further application and research of PSA.
ABSTRACT
Background@#Co-infections of the porcine reproductive and respiratory syndrome virus (PRRSV) and the Haemophilus parasuis (HPS) are severe in Chinese pigs, but the immune response genes against co-infected with 2 pathogens in the lungs have not been reported. @*Objectives@#To understand the effect of PRRSV and/or HPS infection on the genes expression associated with lung immune function. @*Methods@#The expression of the immune-related genes was analyzed using RNA-sequencing and bioinformatics. Differentially expressed genes (DEGs) were detected and identified by quantitative real-time polymerase chain reaction (qRT-PCR), immunohistochemistry (IHC) and western blotting assays. @*Results@#All experimental pigs showed clinical symptoms and lung lesions. RNA-seq analysis showed that 922 DEGs in co-challenged pigs were more than in the HPS group (709 DEGs) and the PRRSV group (676 DEGs). Eleven DEGs validated by qRT-PCR were consistent with the RNA sequencing results. Eleven common Kyoto Encyclopedia of Genes and Genomes pathways related to infection and immune were found in single-infected and co-challenged pigs, including autophagy, cytokine-cytokine receptor interaction, and antigen processing and presentation, involving different DEGs. A model of immune response to infection with PRRSV and HPS was predicted among the DEGs in the co-challenged pigs. Dual oxidase 1 (DUOX1) and interleukin-21 (IL21) were detected by IHC and western blot and showed significant differences between the co-challenged pigs and the controls. @*Conclusions@#These findings elucidated the transcriptome changes in the lungs after PRRSV and/or HPS infections, providing ideas for further study to inhibit ROS production and promote pulmonary fibrosis caused by co-challenging with PRRSV and HPS.

ABSTRACT
Objective To examine the method of least-square fitting for calibrating the palatal rugae boundary. Methods According to the distribution of the teeth, the feature points were selected; when they were fit, the boundary of the palatal rugae area was created, thereby constructing a mask. The mask was used to remove the part located outside the boundary and filter out the interference. Three samples were utilized for the ex-periments and analyses to come. Results To evaluate the quantitative results of the fitting curves, the correla-tion coefficients(r)of the samples and the relationship between the actual mean value and ideal mean value was obtained through six fitting processes. The differences between the actual mean and ideal mean were found to be significantly small (from 0.2857 to 2.5000) in the six fitting processes, with the range of r close to 1(from 0.9896 to 0.9995). Conclusion The effect of the cubic polynomial fitting method adopted in this study was stable. The proposed boundary calibration method can effectively locate the palatal rugae boundary and remove the interference area, further promoting the practice of forensic identification.

The content of hydroxyproline in lungs, TGF-β1 and IL-6 in serum were examined.@*Results@#The lung's histopathological examination showed no obvious inflammatory cell and no fibrosis in the lung tissue of the control group, there were a lot of inflammatory cell aggregation and collagen fiber deposition in silica model group, while in the UC-MSCs intervention group and treatment group, there were less inflammatory cells and collagen fiber. The rats from silica model groups had higher HYP, TGF-β1 and IL-6 than the rats from UC-MSCs treatment group and control group. Lung fields of rats in the control group were clear and no obvious high-density shadow. Different-sized granular high-density shadows or reticular fibrous shadows were found diffusely distributed in the lungs of the rats in silica model group. Lung field of rats in UC-MSCs intervention group and treatment group were less high density shadows, and more clear.@*Conclusion@#UC-MSCs can alleviate the pulmonary fibrosis in silica model rats through regulating the secretion of some fibrosis related cytokines.
ABSTRACT
OBJECTIVE: To evaluate the effects of jejunal-ileal bypass(JIB) in addition to sleeve gastrectomy(SG) on glycemic regulation and body weight in Zucker Diabetic Fatty(ZDF) rats and underlying mechnisms. METHODS: Twelve ZDF(fa/fa) rats were randomly divided into two groups based upon the procedure performed including sleeve gastrectomy plus jejunal-ileal bypass surgery group(SG+JIB,n=6) and sleeve gastrectomy plus sham procedure group(SG+Sham,n=6). In addition, five healthy ZDF(fa/+) rats were employed as normal controls. Body weight, fasting blood glucose, food intake, oral glucose tolerance test and plasma GLP-1 levels were measured before surgery and at 2 and 4 weeks after surgery. RESULTS: Preoperatively, all baseline parametes had no statistical difference between SG+JIB and SG+Sham groups. In comparison with SG+Sham group, fasting plasma glucose levels at postoperative week 2(17.3 mmol/L vs. 8.7 mmol/L) and 4(21.4 mmol/L vs. 11.9 mmol/L)were significantly lower in SG + JIB group(P<0.0001). Oral glucose tolerace was significantly improved, represented by glucose excursion area under curve for 180 minutes period(4221 vs.2964,P<0.0001 at postoperative week 2;and 4104 vs. 3388,P<0.01 at postoperative week 4). Simutaneously, plasma GLP-1 levels were significantly higher in SG + JIB group than SG + Sham group(64.58 pmol/L v.s.30.52 pmol/L,P<0.001). However,SG + JIB had less food intake and body weight only decreased at postoperative week 2,but not at week 4. CONCLUSION: By adding JIB onto SG,the metabolic regulation capability is able to be furtherly enhanced. However,it seems that there is not additional benefit in weight loss. Therefore, based upon these results,we suggest that SG + JIB should be recommended to the patients who needs additional metabolic effects, but not to the patients for the purpose of additional weight loss above SG alone.

ABSTRACT
Objective: To investigate the current practice of extracorporeal cardiopulmonary resuscitation (ECPR) for Chinese cardiac arrest patients after the publication of 2015 American Heart Association guidelines for cardiopulmonary resuscitation. Methods: A questionnaire was distributed to healthcare providers of emergency departments (EDs) and/or Intensive Care Units (ICUs) across 52 hospitals in China from August to November 2016. Data collection ended in February 2017. The questionnaire included three parts: (1) characteristics of the departments and the respondents; (2) knowledge about ECPR; (3) practice of ECPR in cardiac arrest patients (case volume, inclusion/exclusion criteria, ECPR procedure). The characteristics of the departments/hospitals were only answered by the head of the department. Results: A total of 1 952 (86.8%) respondents fulfilled the survey. Only 2.5% of the respondents from 3 of 52 hospitals performed ECPR. Among the three hospitals, the case number of ECPR were ≤5 per year and none of them had written ECPR procedures. Only one hospital had formal inclusion/exclusion criteria. The inclusion criteria included age between 18 to 60 years, suspected cardiogenic cardiac arrest, beginning of cardiopulmonary resuscitation 10 min. The top three reasons for the nonuse of ECPR were unknown fields (31.2%), potential ECMO-related side effects (26.9%) and cost (18.7%). Conclusions: ECPR for cardiac arrest patients are not well understood by healthcare providers in the emergency department or ICUs and its application is still in the early stage in China. Educational training and other interventions are needed to promote the clinical practice.
ABSTRACT
In this study, the anti-inflammatory mechanism of Ginkgo biloba extract 50 (GBE50) and its mechanism of action on NLRP3 inflammatory corpuscles were observed by primary microglia cells. LPS/ATP was used to stimulate microglia, and the best time for stimulation and optimal concentration of GBE50 were screened. Pro-inflammatory cytokine IL-1 and TNF-α was determined by ELISA. Western blot was performed to observe the protein expression of NLRP3, ASC, caspase-1 and IL-1 in cultured primary rat microglia. Effect of GBE50 on NLRP3 inflammatory corpuscle aggregation was detected by CO-IP. GBE50 (10 mg·L⁻¹) preconditioning could significantly inhibit the expression of LPS (100 μg·L⁻¹,12 h), ATP (5 mmol·L⁻¹, 30 min) induced primary microglia corpuscle associated protein, inflammatory corpuscle aggregation, and the release of inflammatory factors IL-6 and TNF-α. These results indicated that GBE50 could inhibit the secretion of inflammatory factors after microglia activation, which is related to down regulating the protein expression and activity of NLRP3 inflammatory corpuscle.
ABSTRACT
Objective To observe the effect of resveratrol(Res)on myocardial ischemia/reperfusion(I/R)injury in rats and to explore its potential mechanisms. Methods Sixty male SD rats were selected and divided into the sham operation group,I/R group,I/R+ Res group and I/R+ Res +LY294002(I/R+Res+LY)group.The rat myocardial I/R injury model was established by the ligation of left anterior descending coronary artery for 30 min and then by reperfusion for 2 h,and the injection of Res(20 mg/kg)or LY294002(5 mg/kg)was given via sublingual vein at 1 min before reperfusion. The levels of malondialdehyde(MDA) and superoxide dismutase(SOD)in plasma and myocardial tissue, and level of heme oxygenase 1(HO-1)in myocardial tissues were determined in each group;Western blot was used to detect the expression level change of PI3K,Akt(p-Akt),Nrf2 and HO-1 protein in myocardial tissue. Results Compared with the I/R group, the plasma and myocardial MDA levels in I/R+ Res group were re-duced,the SOD activity level was elevated, and the activity and expression of HO-1 was enhanced(P<0.05);compared with the I/R gourp, the expression levels of PI3K,Akt(p-Akt),nrf2 and HO -1 protein levels in myocardial tissue of I/R+ Res group were increased(P<0.05).But these effects in the I/R+Res+LY group could be partially eliminated by PI3K/Akt inhibitor LY294002. Conclusion Resveratrol post-treatment can enhance the activity and expression of HO-1 protein through up-regulating the activity of PI3K/Akt/Nfr2,and then promotes the activity and expression of HO-1,thus plays the HO-1 anti-oxidative role, and alleviates myocardial I/R injury.

ABSTRACT
Hypermetabolism is one of the remarkable characteristics of burn injury. Prolonged hypermetabolism causes insufficient energy supply, which leads to delayed wound healing, immune system dysfunction, increased infection rate, and multi-organ failure. In recent years, it is concerned that the activation of brown or beige adipose tissue may be related to hypermetabolism in severe burn patients. Brown or beige adipose tissue could be regulated by stress hormones and some cytokines which increase and persist in high level for several months after severe burn. This paper reviews the current knowledge of brown or beige adipocytes developmental lineages, molecular regulation mechanism, and regulation of brown or beige adipocytes activation after severe burn.
ABSTRACT
Objective@#To investigate the effects of dexmedetomidine on myocardium of rats at early stage after severe burn.@*Methods@#Twenty specific pathogen free male SD rats were immersed in 90 ℃ hot water for 20 s, causing 30% total body surface area (TBSA) full-thickness scald (hereafter referred to as burn) on the back. And then they were divided into burn resuscitation group (BR) and burn resuscitation+ dexmedetomidine group (BRD) according to the random number table, with 10 rats in each group. Sodium lactate Ringer′s solution (2 mL·kg-1·%TBSA-1) were intraperitoneally injected into rats of both groups after burn. Dexmedetomidine with dose of 1 μg/kg was intraperitoneally injected into rats of group BRD at the same time point. Another 5 rats in sham injury group (SI) were immersed in 37 ℃ water bath causing sham injury, and fluid resuscitation of rats in group SI was the same as that in group BR. Five rats of group BR and BRD were respectively selected at post burn hour (PBH) 6 and 24. And then left ventricular end-systolic internal diameter (LVIDs), left ventricular end-diastolic internal diameter (LVIDd), ejection fraction (EF), and cardiac output (CO) were determined with small animal ultrasonic imaging system. Plasma levels of cardiac troponin (cTn) I and cTnT were detected by enzyme-linked immunosorbent assay, and morphological changes of myocardium were observed under optical microscope and transmission electron microscope (observed only at PBH 24). In rats of group SI, morphological change of myocardium was observed at PBH 24, and the other indexes were detected as above. Data were processed with one-way analysis of variance and SNK test.@*Results@#At PBH 6, EF value of rats in group BR [(98.0±2.8) %] was obviously higher than that in group SI [(91.0±0.4)%, P<0.05]. The other 3 cardiac ultrasound indexes of rats in group BR were close to those in group SI (with P values above 0.05). Each cardiac ultrasound index of rats between groups BRD and SI was close at PBH 6 (with P values above 0.05). At PBH 24, LVIDs levels of rats in group BR [(0.66±0.59) mm] and group BRD[(0.69±0.27) mm] were obviously lower than LVIDs level of rats in group SI [(1.65±0.33) mm, with P values below 0.05]. LVIDd, EF, and CO levels of rats were close among 3 groups at PBH 24 (with P values above 0.05). At PBH 6, the plasma levels of cTnI [(17.40±1.59) ng/mL] and cTnT [(1 488±229) pg/mL] of rats in group BR were significantly higher than those in group SI [(1.84±0.92) ng/mL and (169±12) pg/mL, with P values below 0.01]. At PBH 6 in group BRD, the plasma level of cTnI of rats [(2.58±0.60) ng/mL] was close to that in group SI (P>0.05), and the plasma level of cTnT [(649±190) pg/mL] was higher than that in group SI (P<0.01). At PBH 24, the plasma levels of cTnI and cTnT of rats in group SI were close to those in groups BR and BRD (with P values above 0.05). At PBH 24, the plasma level of cTnI of rats in group BRD was obviously lower than that in group BR (P<0.01). At PBH 6, the myocardial structures of rats in group BR and group BRD were normal, which were close to myocardial structure of rats in group SI at PBH 24. At PBH 24, obviously damaged myocardial tissue, disorderly arrangement of myofilament, and seriously damaged mitochondria were observed in rats of group BR, which were significantly ameliorated in rats of group BRD.@*Conclusions@#Dexmedetomidine can protect the myocardium of rats with severe burn at early stage.
