ABSTRACT

Objective:To investigate the correlation between the number of circulating tumor cells (CTC) in peripheral blood and clinicopathological features of patients with breast cancer. Methods:The clinical data of 104 breast cancer patients at Guangzhou Panyu Central Hospital between January 2017 and May 2020 were retrospectively analyzed. The number of CTC in peripheral blood, the levels of serum tumor markers [alpha-fetoprotein (AFP), carcinoembryonic antigen (CEA), carbohydrate antigen (CA)125, CA153] were detected. In blood samples, the number of CTC ≥ 2/ml was defined as CTC positive. Immunohistochemistry was used to analyze the protein expression of Ki-67 in tumor tissues. The association of CTC with clinicopathological features, serum tumor markers and Ki-67 protein expression was also analyzed. Results:The CTC positive rate was 80.77% (84/104). There were statistically significant differences in composition of whether there was vascular tumor thrombus (χ 2 = 0.860, P = 0.009), axillary lymph node metastasis (χ 2 = 12.382, P<0.01), N staging ( P = 0.002) and TNM staging (χ 2 = 7.698, P = 0.006) between patients with CTC positive and negative. However, there were no statistically significant differences in composition of age ( t = 0.634, P = 0.528), tumor quadrant (χ 2 = 6.523, P = 0.163), molecular subtyping (χ 2 = 4.164, P = 0.384), histological grade (χ 2 = 1.901, P = 0.387), T staging ( P = 0.099) and whether there was nerve invasion (χ 2 = 0.092, P = 0.761). The levels of serum CEA and CA125 in CTC positive patients were higher than those in CTC negative patients [median ( P25, P75): 2.50 ng/ml (2.21 ng/ml, 2.92 ng/ml) vs. 1.89 ng/ml (1.61 ng/ml, 2.35 ng/ml); 13.81 U/ml (11.79 U/ml, 16.28 U/ml) vs. 11.17 U/ml (8.91 U/ml, 12.80 U/ml); all P < 0.05], and CTC was positively correlated with serum CEA and CA153 levels ( r = 0.520, P<0.01; r = 0.497, P<0.01); CTC was not related to Ki-67 protein expression (χ 2 = 0.512, P = 0.474). Conclusion:The number of CTC in peripheral blood is closely related to clinical staging, lymph node or hematogenous metastasis, tumor markers CEA and CA153 levels of breast cancer. The increased number of CTC may cause tumor progression and metastasis.

ABSTRACT

Objective:To analyze the risk factors for postoperative pulmonary venous obstruction after correction of total anomalous pulmonary venous connection(TAPVC).Methods:We retrospectively reviewed all patients undergoing operative repair of TAPVC in our institution from December 2013 to January 2018. Patients with functionally univentricular circulations or atrial isomerism were excluded. Patients were divided into two groups according to whether there was pulmonary vein obstruction. The clinical variables of the two groups were compared. Variables for the multivariable analysis were chosen if there was statistical significance on univariable analysis.Results:145 patients were included, 91(63%) males, aged 4(2, 8)months and weight 5.5(4.5, 7.5)kg. Mean follow-up interval was(51±23) months. Postoperative obstruction developed in 27 patients(18.6%). The differences of anatomic type[supracardiac 18(67%) vs.59(50%), cardiac 4(15%) vs. 50(42%), infracardiac 3(11%) vs. 1(1%), mixed 2(7%) vs. 8(7%), P=0.003], preoperative obstruction[yes 19(70%) vs. 37(31%), no 8(30%) vs. 81(69%), P<0.001], associated cardiac lesions[yes 13(48%) vs. 27(23%), no 14(52%) vs. 91(77%), P=0.008] and bypass time[109(89, 129)min vs. 88(70, 110)min, P=0.002] between two groups were statistical significant. A multivariable model showed preoperative obstruction( P<0.001) and bypass time( P=0.009) were associated with postoperative obstruction. Conclusion:The incidence of pulmonary vein obstruction after correction of TAPVC was still high. If there was preoperative obstruction, or the bypass time was too long during operation, the surveillance of pulmonary vein obstruction should be strengthened after operation.

ABSTRACT

Hydroxytyrosol as one of natural anti-oxidants,extracted from the fruits and leaves of Olea europaea,is a natural polyphenol compound in the form of esters. Recently, considerable studies showed that hydroxytyrosol demonstrated intrinsic biological activity for metabolic syndromes, cardiovascular- and neurodegenerative-related diseases, and it was revealed to play the roles in the anti-activities of cancerous, inflammatory as well as depressing issues. In addition, hydroxytyrosol is an oleophilic and hydrophilic compound with high bioavailability and low cellular cytotoxicity. It could be absorbed by various tissues and could easily passe through blood brain barrier. Therefore, hydroxytyrosol was introduced as one of the key subjects targeted by innovative drug development. However,it has a short half-life in vivo and non-tissue specific, which lead to its limitation in clinical application, so further in-depth studies are still needed. The authors had a literature review of hydroxytyrosol, and summarized the basic properties of its pharmacokinetic, pharmacological effects and molecular mechanisms. This article mainly focused on it’s pharmacological activity and the mechanism involved in treating damages induced by the oxidative stress,in alleviating cardiovascular diseases and in inhibition of neurodegenerative diseases. In this article, its anti-inflammatory,anti-tumor,anti-depressant effects, other biological activities, and pharmacokinetics were also briefly reviewed. The authors put forward some personal thoughts on its future research direction, hoping to provide ideas and inspirations for the vast number of researchers, and provide references for its further development, research and application.

ABSTRACT

Objective:To explore the effects and mechanism of Chinese classical prescription Dahuang Zhechongwan on silicosis in mice. Method:Thirty-six male Kunming mice of SPF grade were randomized into the normal control group, model control group, tetrandrine (Tet, 0.039 mg·kg-1) group, as well as high- (1.560 g·kg-1), medium- (0.780 g·kg-1), and low-dose (0.390 g·kg-1) Dahuang Zhechongwan groups, with six mice in each group. Mice in all groups except for the normal control group underwent static inhalation of silica (SiO2) dust for 40 consecutive days to induce fibrosis. After 28 days of intervention with corresponding drugs, the mice were sacrificed to collect the serum and lung tissues, with the former used for detecting tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), IL-6, and hydroxyproline (HYP) levels by enzyme-linked immunosorbent assay (ELISA) and the latter for observing the pathological changes. Meanwhile, the protein and mRNA expression levels of p38 mitogen-activated protein kinase (p38 MAPK), nuclear transcription factor-κB (NF-κB), transforming growth factor-β1 (TGF-β1), α-smooth muscle actin (α-SMA), Smad2, Smad3, and Smad7 in the lung tissues were determined by Western blot and real-time polymerase chain reaction (Real-time PCR). Result:Compared with the normal group, the contents of TNF-α, IL-1β, IL-6 and HYP in the model group were significantly increased, the difference was statistically significant(P<0.05,P<0.01); compared with the model group, the high-dose group of Dahuang Zhechongwan could significantly reduce the contents of TNF-α, IL-6 and HYP in the serum of mice(P<0.05, P<0.01), indicating that Dahuang Zhechongwan could reduce the lung inflammation of silicosis mice. At the same time, compared with the normal group, the protein and mRNA expression levels of p38 MAPK, NF-κB p65, TGF-β1, α-SMA, Smad2 and Smad3 in the model group were significantly increased(P<0.05,P<0.01), while the protein and mRNA expression levels of Smad7 were significantly decreased(P<0.01); compared with the model group, the protein and mRNA expression levels of p38 MAPK, NF-κB p65, TGF-β1, α-SMA, Smad2 and Smad3 in the high-dose Dahuang Zhechongwan group were significantly increased the protein and mRNA expression levels were significantly decreased(P<0.05,P<0.01), while Smad7 protein and mRNA expression levels were significantly increased(P<0.05,P<0.01). Conclusion:Dahuang Zhechongwan ameliorates the alveolar inflammation, extracellular matrix (ECM) deposition, and fibrosis in mice with silicosis possibly by regulating the p38 MAPK/NF-κB/TGF-β1 pathway.

ABSTRACT

Dahuang Zhechongwan (DHZCW) is a classic prescription from the Jingui Yaolue(《金匮要略》) by ZHANG Zhong-jing,with the effects of tonifying deficiency, relaxing the middle, promoting regeneration, and resolving stasis. It has been widely used in the clinical treatment of various diseases with definite efficacy achieved. The research on multiple organ fibrosis has shown that DHZCW can slow down the development of organ fibrosis in the heart, liver, kidney, lung, etc., and good results in both clinical practice and experimental research have been obtained. The present study reviewed the previous investigations on the experimental mechanism of DHZCW in the treatment of multiple organ fibrosis and revealed that the pathogenesis was closely related despite different disease sites. From the perspective of traditional Chinese medicine (TCM),these diseases shared a common pathogenesis, which was manifested by deficiency. Long-term diseases led to the formation of "dried blood". From the perspective of modern medicine, the diseases all showed pathological changes in the deposition of extracellular matrix (ECM), and their occurrence and development were all based on certain effector cells [such as hepatic stellate cell (HSC) and pancreatic stellate cell (PSC)], with same cytokines [such as tumor necrosis factor-α (TNF-α),interleukin-6 (IL-6),IL-1β,and α-smooth muscle actin (α-SMA)] and some key pathways [transforming growth factor-β1 (TGF-β1)/Smad, phosphatidylinositol-3-kinase (PI3K)/protein kinase B (Akt), and lipopolysaccharide (LPS) paracrine and autocrine mechanisms] involved. As a classic prescription for "deficiency-induced dry blood", DHZCW was effective in treating fibrosis, which was presumedly related to the inhibition of ECM deposition by intervening in the above-mentioned mechanisms, thereby delaying the disease progression. This study is expected to provide literature support to clarify the scientific connotation of DHZCW in the treatment of multiple organ fibrosis and lay a foundation for further experimental and clinical research.

ABSTRACT

Objective:To explore the mechanism of gentiopicroside (GPS) in preventing acute liver injury induced by carbon tetrachloride (CCl4) in mice and its effect on the Toll-like receptor 4 (TLR4)/myeloid differentiation factor 88 (MyD88)/nuclear factor-κB (NF-κB) signaling pathway. Method:Sixty mice were randomly divided into a normal control group, a model group, a silymarin group (150 mg·kg-1), and high- (200 mg·kg-1), medium- (100 mg·kg-1), and low-dose (50 mg·kg-1) GPS groups, with 10 in each group. The mice in the groups with drug intervention were administered correspondingly by gavage at 10 mL·kg-1, and those in the normal control group and the model group receive an equal volume of distilled water, once per day. Ten days after administration, mice in the normal control group were subjected to the intraperitoneal injection of peanut oil (10 mL·kg-1) and those in other groups were injected with peanut oil (10 mL·kg-1) containing 0.12% CCl4 for the induction of acute liver injury model. After fasting for 16 hours, blood was collected from eyeballs and liver tissues were collected. Hematoxylin-eosin (HE) staining was used to observe the pathological changes of liver tissues. The content or activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TBIL), alkaline phosphatase (ALP), total superoxide dismutase (T-SOD), and γ-glutamyl transpeptidase (γ-GT) in the serum, malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) in liver tissues were determined by biochemistry techniques. The levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) in liver tissues were measured by enzyme-linked immunosorbent assay (ELISA). Western blot was used to detect the protein expression of TLR4, MyD88, and NF-κB in liver tissues. The expression of phosphorylated NF-κB (p-NF-κB) was detected by immunohistochemistry. Result:Compared with the normal control group, the model group showed increased levels of ALT, AST, ALP, TBIL, γ-GT, and MDA (P<0.01), and blunted activities of T-SOD and GSH-Px (P<0.01). Compared with the model group, the high- and medium-dose GPS groups exhibited declining levels of ALT, AST, ALP, TBIL, γ-GT, and MDA (P<0.05, P<0.01) and potentiated T-SOD and GSH-Px activities (P<0.05, P<0.01). Compared with the normal control group, the model group displayed elevated levels of TNF-α, IL-1β, and IL-6 in liver tissues (P<0.01) and increased protein expression of TLR4, MyD88, and p-NF-κB (P<0.01). Compared with the model group, the high- and medium-dose GPS groups showed decreased TNF-α, IL-1β, and IL-6 content in liver tissues (P<0.05, P<0.01) and dwindled TLR4, MyD88, and p-NF-κB protein expression (P<0.05, P<0.01). Conclusion:GPS possesses a protective effect on mice with acute liver injury induced by CCl4, and its mechanism of action may be related to the regulation of TLR4/MyD88/NF-κB signaling pathway and inhibition of oxidative stress.

ABSTRACT

Brain-computer interface (BCI) technology can activate the plasticity of the nervous system and induce the reconnection of nervous system information pathways. Techniques for recording brain activity include invasive methods and non-invasive methods. Non-invasive methods are safer, more portable and cheaper, but are prone to signal pollution. For upper limbs rehabilitation in patients with severe chronic stroke, BCI of electroencephalogram and robotic arm is usually used, almost combined with other rehabilitation approaches, and seems to improve the effectiveness. It is necessary to improve the accuracy of BCI motion intention decoding, carry out hierarchical and customized treatment for patients, develop hybrid and portable BCI system.

ABSTRACT

Objective:To observe the effects of a self-developed wearable intelligent upper limb rehabilitation robot system on upper limb motor function and activities of daily living of stroke patients. Methods:From August, 2018 to October, 2020, 61 stroke patients with hemiplegia from Beijing Bo'ai Hospital were randomly divided into control group (n = 30) and observation group (n = 31). The control group accepted routine rehabilitation training twice a day, while the observation group accepted routine rehabilitation training once a day and robot-assisted training once a day, for four weeks. All the patients were assessed with Fugl-Meyer Assessment-Upper Extremities (FMA-UE) and modified Barthel Index (MBI) before and after training, while the active range of motion (aROM) of affected shoulder, elbow and forearm was measured. Results:A case dropped out in the observation group. The scores of FMA-UE and MBI improved in both groups after training (|t| > 4.372, P < 0.001), and the improvement of MBI was more in the observation group than in the control group (t = 2.393, P < 0.05). The aROM of flexion/extension, internal/external rotation and abduction of shoulder, flexion of elbow, and supination/pronation of forearm increased in both groups (|t| > 3.050, P < 0.01), and the improvement of flexion and external rotation of shoulder was more in the observation group than in the control group (|t| > 2.160, P < 0.05). Conclusion:Robot-assisted training based on routine rehabilitation may promote the recovery of upper-limb function for stroke patients.

ABSTRACT

This study aimed to explore the characteristic role of Puerariae Lobatae Radix(PLR) in Gegen Decoction for the treatment of primary dysmenorrhea(PD). Estrogen(E_2) was combined with oxytocin to establish a mouse model of PD. The mice were randomly divided into a normal group, a model group, a Gegen Decoction group, a PLR-free Gegen Decoction group, a PLR group, and a positive drug group(ibuprofen). Writhing response times and writhing incubation of mice in each group were tested by behavio-ral assessment, and the serum levels of prostaglandin F_(2α)(PGF_(2α)), prostaglandin E_2(PGE_2), E_2, and progesterone(PROG) were detected by ELISA kits. Western blot method was adopted to detect cyclooxygenase-2(COX-2) and estrogen receptor alpha(ER_α) expression levels in uterine tissues. Doppler ultrasound was employed to detect changes in uterine artery blood flow in mice, including peak systolic blood flow velocity(maximum velocity), end-diastolic velocity(minimum velocity), peak systolic blood flow velocity/end-diastolic velocity(S/D), pulsatility index(PI), and resistive index(RI). Histopathological changes in the uterus were detected by HE staining. Based on the oxytocin-induced isolated uterine contraction model, the effects of Gegen Decoction, PLR-free Gegen Decoction, and PLR on the amplitude, frequency, and activity of isolated uterine contraction were compared to investigate the role of PLR in Gegen Decoction for the treatment of PD. The results showed that compared with the Gegen Decoction group, the PLR-free Gegen Decoction improved the indicators of PD except for E_2 content, ER_α expression, and uterine artery blood flow. PLR could significantly down-regulate the serum content of E_2 and the protein expression of uterine ER_α, and improve the uterine artery blood flow. The data suggested that PLR, as the sovereign drug of Gegen Decoction, might function in Gegen Decoction for the treatment of PD by mediating E_(2 )and improving the uterine artery blood flow.

ABSTRACT

This paper explores the statistical distribution characteristics of coating film thickness, so as to present a new method for determining coating endpoint based on 3σ criterion and logic regression. Firstly, the spectrum and thickness of 4 batch samples were collected. Secondly, the spectral range of normal products was obtained by 3σ criterion, with the spectral feature NI as the number of test spectrum in the above range. Then, the model based on 3σ criterion and logic regression was built according to the best condition in K-fold cross-validation and the determined threshold of qualified rate in the coating endpoint. Finally, the qualified rate of test set samples at different time points was calculated by the above model, and the above change trend and the threshold value were combined to determine the coating endpoint. The results of KS analysis showed the distribution of thickness of the qualified products followed the normal distribution(P=0.081>0.05). The accuracy of the coating endpoint determination was as high as 100% by the model based on 3σ criterion and logic regression when the determined threshold of qualified rate was 90%. Therefore, the 3σ criterion was feasible to the research of coating eligibility. This paper reveals certain random phenomena in the coating process, and the method features a high accuracy, quick analysis and a good interpretability, which provides a reference for online detection and qualify evaluation in future.

ABSTRACT

To obtain the difference of the fungal and bacterial community diversity between wild Cordyceps sinensis, artificial C. sinensis and their habitat soil, Illmina Hiseq high-throughput sequencing technology was applied. The results show that Proteobacteria was the dominant bacterial phylum in C. sinensis, Actinobacteria was the dominant bacterial phylum in soil microhabitat, Ophiocordyceps sinensis was the predominant dominant fungus of C. sinensis. The α diversity analysis showed that the fungal diversity of stroma was lower than other parts, and the fungal diversity of wild C. sinensis was lower than that of artificial C. sinensis. The β diversity analysis showed that the fungal and bacterial community diversity of soil microhabitat samples was significantly different from that of C. sinensis. The fungal community diversity was less different between wild and artificial C. sinensis, especially in sclerotia. LEfSe analysis showed a lot of species diversity between wild and artificial C. sinensis. Those different species between wild C. sinensis, artificial C. sinensis and their habitat soil provide ideas for further research on breed and components of C. sinensis.
