ABSTRACT

Objective To investigate the effects of Akkermansia muciniphila (AKK) on azomethane-oxide (AOM)/glucan sodium sulfate (DSS)-induced inflammatory colorectal cancer mouse model and intestinal stem cells. Methods AOM/DSS-induced mouse models of inflammatory-associated colorectal cancer were randomly divided into three groups, namely, model, AKK and aspirin groups, based on different administration of drugs by gavage. The tumor number, size, distribution, and burden were observed 10 weeks after intervention. Immunohistochemical method was used to analyze the expressions of Ki67 and Lgr5 proteins, which are utilized to characterize tumor malignancy and stem cells. The mRNA expressions of Lgr5, CD133, Nanog, and ALDH1 were detected by qRT-PCR. Results Compared with those of the model group, the tumor number, size, and burden of the AKK group were significantly reduced (P < 0.01). The expressions of Ki67 and Lgr5 in the AKK group of tumor tissues were significantly decreased (P < 0.05), and the mRNA expressions of CD133, Nanog and ALDH1 were significantly down-regulated. Conclusion AKK is effective against AOM/DSS-induced colitis-associated colorectal cancer in mice, and its mechanism of action may be closely related to colorectal stem cell activity.

ABSTRACT

Granzyme exists in the cytotoxic granules and belongs to a highly homologous serine protease family. Granzyme is synthesized and released from natural killer cells and cytotoxic lymphocytes. It induces multiple programmed death with the participation of perforin and is an important medium for immune cells to exert anti-tumor effects. This article reviews the granzyme-mediated tumor cell death and the relationship between granzyme and kinds of ways of cell apoptosis, in order to provide new theoretical basis and therapeutic strategies for tumor therapy.

ABSTRACT

Programmed death factor-1 (PD-1) is a promising target molecule for clinical tumor immunotherapy in recent years. Recent studies suggest that PD-1 and related signaling pathways (PI3K/AKT, JAK/STAT3, p38MAPK, ERK, etc.) played a key regulatory role in the process of pulmonary fibrosis. Silicosis is a systemic disease caused by inhalation of free silicon dioxide dust, which is mainly characterized by extensive pulmonary nodular fibrosis and seriously endangers the health of patients. Dissecting the role of PD-1 in the pathogenesis of silicosis may be of great significance in the mechanism research and clinical diagnosis and treatment of silicosis. This paper reviews the regulation of PD-1 molecule on related signaling pathways and its role in pulmonary fibrosis, and looks forward to the potential application of these mechanistic studies in silicosis research.

ABSTRACT

Objective:To investigate the risk factors of hyperglycemia during pregnancy and its correlation with adverse pregnancy outcomes based on the retrospective analysis of glucose metabolism of pregnant women in Chongming area. Methods:A total of 604 singleton pregnant women who underwent prenatal examination and delivered normally in the Chongming branch of Xinhua Hospital from September 2019 to May 2021 were enrolled in the study. All subjects were divided into normal glucose tolerance gestation (NGTG) group and gestational diabetes mellitus (GDM) group. Pregnant women whose blood glucose exceeded normal but did not meet the diagnostic criteria of GDM were classified into the intermediate state gestational blood glucose (ISGBG) group. Questionnaire, physical examination, and relevant laboratory tests were completed. Data were analyzed using the Statistical Product and Service Solutions 13.0 (SPSS, Chicago, IL).Results:The incidence rate of GDM was 20.86% (126/604), ISGBG was 40.39% (244/604), and NGTG was 38.74% (234/604) in 604 pregnant women. Multivariate logistic regression analysis showed that gestational age ( OR=1.092, P<0.001), serum triglyceride ( OR=1.625, P=0.001) and free T 3 levels ( OR=1.995, P=0.002) were independent risk factors for GDM. The incidence of pregnancy-induced hypertension, cesarean section, macrosomia, the total incidence of adverse pregnancy outcomes and fetal birth weight in ISGBG and GDM were significantly higher than those in NGTG ( P<0.05 or P<0.01). Conclusion:The incidence of GDM in Chongming area is high, especially higher in that of ISGBG. As both GDM and ISGBG lead to increased adverse pregnancy outcomes, early monitoring should be paid more attention to pregnant women in ISGBG in addition to the early intervention of GDM.

ABSTRACT

Objective:To screen the early biomarkers associated with lung carcinoma by using proteomic method. Methods:The proteins of early lung squamous cell carcinoma tissues and adjacent normal tissues were separated by two-dimensional gel electrophoresis (2-DE) and differentially expressed proteins were screened by mass spectrometry (MS). The differential expressions of part proteins were verified by immunohistochemical method. Results:The proteomic profiles of lung squamous cell carcinoma and the adjacent normal tissues were established and the average matched protein spots in the two group were 626 and 602, respectively. Ten protein spots with higher expression in lung squamous cell carcinoma tissues were identified as Annexin 1(Anx-A1),heat shock proteins (HSP27), etc. The proteins were mainly related with cell cycle and signal transduction. The results of immunohistochemistry showed that the positive expression rates of Anx-A1 and HSP27 in lung squamous cell carcinoma tissues were significantly increased (P<0.05). Conclusion:Proteomic technology was an effective method for preliminary identification of the proteins associated with early lung carcinoma. The identified proteins laid a solid foundation for further screening the molecular markers for early diagnosis and treatment of lung cancer.

ABSTRACT

Objective To study the expression of WWOX, ErbB2 and Ki67 protein in non-small cell lung cancer(NSCLC) and the relationships with cell proliferation. Methods WWOX and ErbB2 protein expressions were evaluated by immunohistochemistry in 81 NSCLC patients (50 squamous cell carcinomas,31 adenocarcinomas and 20 adjacent normal lung tissues) and the correlations with histopathologic cell proliferation were analyzed. Results WWOX expression was absent/reduced in 72.8% of NSCLC, while it was normal in 80.0% of adjacent normal lung tissues. WWOX expression was strongly associated with tumor histology, histologic grade and lymph node metastasis ( X2 = 5.44, P = 0.019 ; X2 = 4.740, P = 0.029, X2 = 4.51, P = 0.034 ), reduced WWOX expression was higher in squamous cell carcinomas and in poorly differentiated tumors. The overexpression of ErbB2 was associated with TNM stage and lymph node metastasis ( X2 = 6.90, P = 0.009 ; X2= 5.68, P=0.017). WWOX expression was negatively related to the overexpression of ErbB2 (r=-0.239, P <0.05 ). WWOX expression intensity was nega-tively related to the high index of cell proliferation (r=-0.252, P<0.05 ). Conclusion The loss of WWOX ex-pression plays different roles in tumorigenesis of NSCLC and is related to high aggressiveness of tumors. The loss of WWOX expression and the overexpression of ErbB2 can estimate the prognosis of NSCLC.

ABSTRACT

Objective To study the best combination of respiratory parameters that can maintain good ventilation with a low airway pressure under general anesthesia and CO2 pneumoperitoneum (12 mm Hg) during laparoscopy, which is important for lung protections. Methods Basic respiratory parameters of anesthesia machine, respiratory frequency (f), tidal volume (VT), and respiration ratio (I∶E), were used as three factors A, B, and C. The there levels were set as f=15, 12, and 9 bpm; VT =8, 10, and 12 ml/kg body weight; and I∶E =1∶2.5, 1∶2.0, and 1∶1.5. L9(34) K=3 was adopted for repeated orthogonal experimental design. The effect of different combinations of respiratory parameters on peak inspiratory pressure (PIP), mean airway pressure (Pmean),and end-tidal carbon dioxide partial pressure (PETCO2), were analyzed statistically. Results During the laparoscopy, the vital signs of 27 patients were stable under general anesthesia with 9 combinations of the respiratory parameters, the SpO2 was maintained at 100%, and the PEEP was kept at 1 hPa. The effect of the three levels of VT (factor B) on PIP was not significant (P=0.074). While the effects of f (factor A) on PETCO2 and I∶E on Pmean were significantly different among the three levels (P=0.002 and P=0.017, respectively). Conclusion The best combination of three factors-levels respiratory parameters is not small tidal volume with fast frequency or large tidal volume with slow frequency, but is A2B2C2 (f=12 bpm, VT=10 ml/kg body weight, and I∶E=1∶2.0).

ABSTRACT

Objective: To investigate the expression of mage-a mRNA in lung cancer tissues of mice induced by coal tar pitch(CTP) fume and to discuss the above model for lung cancer immunotherapy with mage-a. Methods: Tumor tissue samples of lung cancer and paired non-tumor tissues were obtained from 8 lung cancer mice. Total RNA was extracted and cDNA was synthesized. Nested polymerase chain reaction amplification using mage-a specific primer was performed to detect the expression of mage-a. The 2 clones of mage-a mRNA positive PCR products were sequenced by DNAs sequencer (PE-377). Results: Of 29 mice in the experimental group, 8 were induced to lung cancer.Among which 5 (5/8) expressed mage-a mRNA. The expression of mage-a gene was not found in adjacent lung tissues. The DNA sequencing confirmed that the target gene fragments in 2 samples of PCR products were mage-a cDNA. Conclusion: The mage-a gene is highly expressed in lung cancer in mice induced by CTP fume, suggesting that CTP-induced lung cancer in mice may be an ideal animal model for lung cancer therapeutic experiment with MAGE-A.

ABSTRACT

Objective To construct an eukaryotic expression vector carrying human glutathione-S-transferase(GST)A1gene and to provide study materials for toxicology and pharmacogenetic. Methods The GST A1cDNA was amplified and separated from human liver total RNAs by RT-PCR approach and recombined with eukaryotic expression vector pcDNA3.The recombined plasmid pcDNA3-hGSTA1was verified using PCR,restriction analysis and sequencing determination. Results Human GST A1gene was recombined correctly with pcDNA3,compared with Genbank,in code152T→C,amino acid Met→Thr.Conclusion The eukaryotic expression vector pcDNA3-hGSTA1was constructed for research on toxicology and pharmacogenetics.
