ABSTRACT

OBJECTIVE To study the improvement effects and mechan ism of thalidomide on Alzheimer ’s disease (AD)model of Caenorhabditis elegans . METHODS In this study ,the BR 5270 strain of C. elegans was used as AD model and BR 5271 strain as the control. The effects of thalidomide (0.5,2.0,6.0,15.0 mg/mL)on the motility of BR 5270 strains of C. elegans were studied by the basal slowing response assay ;the effects of thalidomide (0.5,2.0,6.0,15.0 mg/mL)on the survival time of BR 5270 strain of C. elegans were studied by life assay ;the effects of thalidomide (0.5,2.0,6.0 mg/mL)on learning and memory ability of BR 5270 strain of C. elegans were studied by short-term and long-term learning and memory assay. RT-PCR technology was used to study the effects of thalidomide (0.5,2.0,6.0 mg/mL)on mRNA expression of phosphatidylinositol 3-kinase(PI3K)/protein kinase B (Akt)signal pathway related genes (Age-1,Akt-1,Gsk-3)and calpain homologous gene (Clp-1)in BR 5270 strain of C. elegans . RESULTS After the intervention of thalidomide ,oscillation times of BR 5270 strain of C. elegans increased significantly within 30 s (except for 0.5 mg/mL group ),and the 10% of maximum life span was prolonged significantly (only 0.5 mg/mL group );the short-term and long-term learning indexes were improved significantly (only 6.0 mg/mL group );mRNA expression of Age-1 and Akt-1(except for 0.5,2.0 mg/mL groups )were increased significantly ,mRNA expression of Gsk-3(except for 0.5 mg/mL group ) and Clp-1 were decreased (P<0.05 or P<0.01). CONCLUSIONS Thalidomide can ameliorate the dyskinesia of AD model of C. elegans,prolong the lifespan of this strain ,and enhance its learning and memory ability. Its mechanism of action may be related to activation of PI 3K/Akt signaling pathway and inhibition of calpain.

ABSTRACT

Objective:To investigate the effect of flipped classroom in functional experiment teaching.Methods:A total of 86 medical students from 4 classes of Tongji University School of Medicine were selected as subjects. 44 students were randomly selected from 2 classes as experimental group using flipped classroom as teaching approach; 42 students were randomly selected from the other 2 classes as control group using traditional teaching method. The teaching effect was assessed by objective evaluation and subjective evaluation. SPSS 22.0 was used for data analysis. The t-test was used for objective evaluation, and the chi-square test was used for subjective evaluation.Results:There were significant differences in the scores of experimental reports [(45.00±2.63) vs. (41.05±3.62), t=5.766, P<0.01] and experimental operation [(48.27±2.31) vs. (44.79±3.04), t=5.962, P<0.01] between the experimental group and the control group. The statistical analysis of subjective evaluation showed that compared with the control group, the experimental group had a significantly higher number of students who selected "helpful" or "very helpful" in improving self-learning ability, innovation ability, understanding of theoretical knowledge, and communication ability ( P<0.05). Conclusion:The teaching method of flipped classroom applied in medical functional experiments can improve students' operation ability, self-learning ability, innovation ability, and communication ability and deepen their understanding of theoretical knowledge.

ABSTRACT

Lycophytes and seed plants constitute the typical vascular plants. Lycophytes have been thought to have no paleo-polyploidization although the event is known to be critical for the fast expansion of seed plants. Here, genomic analyses including the homologous gene dot plot analysis detected multiple paleo-polyploidization events, with one occurring approximately 13-15 million years ago (MYA) and another about 125-142 MYA, during the evolution of the genome of Selaginella moellendorffii, a model lycophyte. In addition, comparative analysis of reconstructed ancestral genomes of lycophytes and angiosperms suggested that lycophytes were affected by more paleo-polyploidization events than seed plants. Results from the present genomic analyses indicate that paleo-polyploidization has contributed to the successful establishment of both lineages-lycophytes and seed plants-of vascular plants.

ABSTRACT

Objective To investigate the clinical efficacy of hysteroscope combined with laparoscopic fallopian tube dredging on female infertility .Methods 64 patients with tubal infertility were selected ,and they were randomly divide into two groups according to the random number table method ,32 cases in each group .The control group was only given hysteroscope , while the observation group was treated with hysteroscope combined with laparoscope .The dredge situation after treatment and pregnancy results 6-12 months after treatment were compared . Results After treatment,the patency rate of the observation group was 93.75%,which was higher than 75.00% of the control group(χ2 =4.27,P<0.05).The successful pregnancy rate of the observation group 6-12 months after treatment was 90.63%,which was obviously higher than 68.75% of the control group (χ2 =4.73,P <0.05). Conclusion Hysteroscope combined with laparoscopic fallopian tube dredging is very effective in treating female infertility,and it is worthy of clinical promotion .

ABSTRACT

Objective To compare the myocardial enzyme spectrum of native mountain yaks ( Bos grunniens) , alien mountain yellow cattle ( Bos taurus domestica) and low altitude yellow cattle breeding in Qinghai plateau, explove the mechanism that the native mountain yaks adapt to the plateau environment.Methods The samples were treated by cervical bleeding to death, and whole blood was collected.Then serum were prepared from whole blood.The myocardial enzymes including aspartate amino transferase, creatine kinase, creatinine kinase-MB isoenzyme, lactate dehydrogenase, α-hydroxybutyrate dehydrogenase and.Results In the native mountain yaks and in the alien mountain yellow cattle, levels of AST, CK, CK-MB were significantly higher than those in the low altitude yellow cattle (P<0.05);compared with the alien mountain yellow cattle, levels of AST,CK were significantly higher than those in the native mountain yaks (P<0.05),CK-MB,LDH and HBDH were lower(P<0.05).The results suggested that under the high altitude and hypoxia environment, myocardial cell injury occurred in the alien mountain yellow cattle, thus various enzymes penetrated through the cells into the blood circulation, lead to the higher serum enzymatic levels.Conclusion The phenomenon indicates that the alien mountain yellow cattle is in a state of high altitude acclimatization.But the native mountain yaks are well adapte to the high altitude and hypoxia environment.

ABSTRACT

Objective To investigate the clinical and pathological characters and seize the diagnostic main points of minimal deviation adenocarcinoma of cervix to improve the accuracy of pathological diagnosis. Methods A clinic pathological review of 6 cases with minimal deviation adenocarcinoma was analyzed retrospectively. Results The main clinical symptoms were watery leucorrhagia, enlargement of the cervix with erosin and hardening. The pathological findings included hyperplasic and mild abnormality of the glands, invasion effects into the stroma could be observed in some glands and abortive glands with desmoplastic changes, or edema and inflammatory infiltration around the glands were also observed. Conclusion The detection rate of minimal deviation adenocarcinoma may increase by triple combination of clinic, cytology and histology.

The status of bioelectrical impedance measurement especially the multifrequency segmental bioelectrical impedance measurement of liver diseases is introduced.The multifrequency segmental bioelectrical impedance can well rule out the impact from the change of body weight caused by edema and ascites,and can evaluate the volume of ascites.Compared with single frequency bioelectrical impedance,the multi frequency bioelectrical impedance measurements probably display good prospect.It helps to produce objective judgement on nutrition status and energy metabolism for patients with liver diseases.The effect,however,needs further clinical research.

ABSTRACT

Objective To investigate the changes of ultrastructure and Bax,Bcl-2 expression of cerebral cortex in lymphostatic encephalopathy(LE).Methods LE models were established by occluding and removing both the shallow and deep cervical lymph nodes in rats.Ultrastructural changes of the cerebral cortex were observed at 1 d,2 d,3 d,5 d,7 d and 14 d after operation.Expression of Bax and Bcl-2 were examined by Western blot.Results Brain edema appeared under electron microscope in LE model group.The perivascular spaces in the cortex were enlarged and full of fluid,the dropsical nerve fibers had swelling and fragmented myelin sheath.Some neurons showed apoptotic conformation and some had characters of necrosis.The changes mentioned above were the most serious at day 5.The protein expression of Bax,Bcl-2 and the ratios of Bax/Bcl-2 in LE 3 d,5 d,7 d groups were significantly higher than those in normal control group(all P

ABSTRACT

AIM: To investigate the changes of bcl-2, bax expression and neuron apoptosis of cerebral cortex in lymphostatic encephalopathy of rats. METHODS: The model of lymphostatic encephalopathy was established by occluding and removing both the shallow and deep cervical lymph nodes in rats. The animals were sacrificed at 1, 2, 3, 5, 7 and 14 days after operation. HE staining was used to observe the structure of brain tissues and TUNEL staining was used to detect in situ cell apoptosis. The expressions of bcl-2 and bax were examined by RT-PCR. RESULTS: Cerebroedema appeared at the second day and was the most serious at the 5th day after blockage of cervical lymphatics. The number of TUNEL positive cells and the expression of bax began to increase at the 2nd day, reached a peak at the 5th day and dropped to control level at the 14th day. The expression of bcl-2 began to increase at the 1st day, reached a peak at the 5th day and dropped to control level at the 7th day. The increasing extent of bax was higher than that of bcl-2. CONCLUSION: The blockage of cervical lymphatics can lead to lymp[JP2]hostatic encephalopathy. Apoptosis is the main form of neuron death in the cortex and has relation to the increasing expression of bcl-2 and bax. [JP]
