ABSTRACT
Objective To develop a pharmacy device for drug counting and dispensing.Methods The device was composed of four parts of dispensing tank, hook, support frame and separator, which was made of bamboo and metal. The multi application of the separators contributed to fulfilling the functions of the device.Results The device gained advantages over the traditional mode in time,accuracy,drug loss and the staff's efficiency.Conclusion The device is easy to manufacture and operate and behaves well in feasibility, practicability and innovation, and thus is worthy promoting practically. ABSTRACT
Objective To fabricate DVDMS-Mn-LPs and evaluate its potential on fluorescence mo-lecular imaging, MRI and sonodynamic therapy ( SDT) . Methods DVDMS was used to chelate with Mn2+, and then encapsulated into nanoliposomes by a typical thin-film rehydration method to fabricate DVDMS-Mn-LPs. The particle morphology, average diameter, zeta potential, encapsulation efficiencies and Mn2+content were determined. Fluorescence molecular imaging was performed on different concentrations of DVDMS-Mn-LPs (0, 40, 80, 120, 160 μg/ml) with the small animal fluorescence imaging system. T1WI was per-formed with 3.0 T MR. The SDT effect of DVDMS-Mn-LPs was verified by CCK8 assay among the following groups:control group, ultrasound group ( experimental group 1, EG1) , DVDMS-Mn-LPs group ( EG2) and DVDMS-Mn-LPs + ultrasound group ( EG3) . One-way analysis of variance and the least significant differ-ence t test were used to analyze the data. Results DVDMS-Mn-LPs exhibited a well-defined spherical mor-phology and homogeneous distribution. The encapsulation efficiency was (65.56±1.47)%. ICP-AES meas-urement revealed that the chelation of Mn2+ with DVDMS occurred at a molar ratio of 1. 6:1. The fluores-cence intensities progressively increased with the elevated concentrations of DVDMS-Mn-LPs. The r1 value for DVDMS-Mn-LPs was 23.74 mmol·L-1·s-1. The cell viabilities of EG3 was (54.82±8.55)%, which were significantly lower than those of EG2 and EG1 ((86.54±2.67)% and (83.76±6.48)%;F=6011, t values: -8.35, -9.15, all P<0.001). Conclusion DVDMS-Mn-LPs is successfully fabricated and has good potential on fluorescence molecular imaging, MRI and SDT, which provides a promising imaging-guided modality for glioma treatment in vivo.
ABSTRACT
<p><b>OBJECTIVE</b>To prepare internalized RGD (iRGD) modified echogenic liposomes containing methotrexate (MTX) and indocyanine green (ICG) (iRGD MTX ICG ELIP) and evaluate its targeting efficiency and inhibitory effect combined with ultrasound on synovial cells.</p><p><b>METHODS</b>iRGD MTX ICG ELIP was prepared by the thin film rehydration and freeze-lyophilization method and its general characteristics and acoustic responsiveness were assessed. The targeting effect of the prepared liposome was observed by assessing its cell uptake in vitro. In a mouse model of rheumatiod arthritis, the targeting effect of the prepared liposome was determined by detecting the fluorescence intensity of the drug in arthrosis. The inhibitory effect of iRGD MTX ICG ELIP combined with ultrasound on synovial MH7A cells in vitro were investigated using CCK8 test.</p><p><b>RESULTS</b>The average diameter and zeta potential of iRGD MTX ICG ELIP was 134.4∓17.61 nm and 10.07∓4.28 mV, and the entrapment efficiency of MTX and ICG was (62.56∓0.77)% and (95.13∓0.82)%, respectively. With ultrasound exposure, the release of MTX and ICG from iRGD MTX ICG ELIP increased with the ultrasound intensity and with the exposure time. In HUVECs, the uptake efficiency of iRGD MTX ICG ELIP was 1.89 times higher than that of non targeted MTX ICG ELIP (P<0.05). In vivo imaging of mouse joint with rheumatiod arthritis showed that the fluorescence intensity of iRGD MTX ICG ELIP was significantly stronger than that of the non targeted liposome. CCK8 assay showed that iRGD MTX ICG ELIP combined with ultrasound resulted in a survival rate of MH7A cells of (32.49∓3.04)%, significantly lower than the rate of cells treated with iRGD MTX ICG ELIP but without ultrasound (P<0.05).</p><p><b>CONCLUSIONS</b>iRGD MTX ICG ELIP has a suitable particle size and can effectively target HUVECs and the joints with rheumatiod arthritis. With a good drug entrapment efficiency and acoustic responsiveness, the drug loaded liposome shows enhanced inhibitory effect on MH7A cells combined with ultrasound in vitro, suggesting its potential in the treatment of rheumatoid arthritis.</p>
ABSTRACT
OBJECTIVE: To investigate the current status of quality of life( QOL) in patients with occupational pneumoconiosis,and explore the effects of the social demographic data and disease-related information on their QOL.METHODS: By the convenience sampling method,144 male patients with occupational pneumoconiosis were selected. The St. George's Respiratory Questionnaire( SGRQ) was used in the study and the stepwise regression analysis method was used to analyze the influencing factors of QOL in these patients. RESULTS: The SGRQ total score was( 60. 0 ± 18. 0),respiratory symptom dimension score was( 61. 2 ± 19. 7),limitation of motion dimension score was( 64. 0 ± 21. 5) and the disease influence dimension score was( 57. 7 ± 19. 5). The QOL of 52. 08%( 75 /144) of the patients were below the average level. The stage of pneumoconiosis was positively correlated with SGRQ total score and the above three dimension scores( P < 0. 01). The length of disease was negatively correlated with respiratory symptom dimension score( P < 0. 05);the educational level was negatively correlated with limitation of motion dimension score( P < 0. 01); the lung function was positively correlated with limitation of motion dimension score( P < 0. 05). CONCLUSION: The QOL of patients with pneumoconiosis was below the average level. The stage of pneumoconiosis,length of diagnosis,the educational level and lung function are the important factors affecting the QOL of these patients.
ABSTRACT
Objective To investigate the feasibility and preventive effect of benign esophageal restenosis by binding 125I seeds to upper esophageal stent through animal experiments.Methods Eight 125I seeds were evenly bound to upper normal esophageal stents for the animal experiments.A total of 32 beagle dogs were randomly assigned to 2 groups:experimental group,implanted with esophageal stents with eight 125I seeds (33.3 MBq),and the control (stents without 125I seeds).Four dogs of each group were killed at the 1 st,2nd,4th,and 8th week after imaging studies.The tissue of 2 cm upper stent underwent pathology analysis.Results All the novel stents were successfully implanted.No radioactive leak was detected by ECT.The lumen diameter of the top stent showed the tract gradually narrowed and at the 4th and 8th weeks,the experiment group narrowed more seriously compared with the control group and the difference was statistically significant (P < 0.05).PCNA,ot-SMA mean optical density were significantly different in the 4th week.Both hydroxyproline and total amino acid increased progressively,with significant difference at the 4th and 8th weeks.The macroscopic and optical findings of the trachea and major vessels were basically the same in both groups.Conclusion The novel stent is feasible and safe for preventing benign esophageal restenosis and preventing benign esophageal stent restenosis.

Objective To evaluate the effects of low-dose 125I seeds to the esophagus fibroblast cell proliferation in vitro.Methods The titanium wire was implanted in the beagle dog's esophagus to induce fibroblast proliferation,and the esophageal tissue was removed after two weeks and cultured in vitro.The 125I seeds with different dose commonly used were chosen to irradiation fibroblast in group B (11.1 MBq × 9)、C (22.2 Mbq ×9) 、D (33.3 MBq ×9) for 72 hours,while in control group no 125I seeds were used.After irradiation,cells were collected.MTT and AnnexinV/PI double staining were performed respectively to evaluate the effects of 125I seeds in cell proliferation and apoptosis.Results The inhibition rate respectively:(26.81 ± 1.96) %、(34.52 ± 3.21) % and (45.33 ± 2.59) % ; the apoptosis rate respectively:(6.73 ±0.57)% 、(13.11 ± 1.39)% and (15.23 ± 0.90)%.There were significant differences among the experimental groups and between every experimental group and the control group.Conclusion The three doses of 125I seeds could significantly inhibit the fibroblast proliferation and promote cells apoptosis,of which 33.3 MBq 12sI seed was the most significant.

@#ObjectiveTo observe the effect of exercise on expression of protein kinase B (PKB) in adipose tissue of insulin resistant rats fed by high fat diet.Methods30 male Wistar rats were randomly divided into the control group (n=10),given basic diet; the model group (n=20), given fat diet. After 4 weeks, the model group was randomly divided into 2 subgroups, the insulin resistant group was continually given high fat diet, the exercise treated group accepted high fat food and swimming training. After 6 week intervention, the expression of PKB stimulated by insulin in adipose tissue was determined with Western blotting at the end of experiment.ResultsAfter long term high fat diet, expression of PKB in adipose tissue of the insulin resistant group decreased by 23.5% comparing with the control group (P<0.01). After 6 weeks swimming training, the expression of PKB of the exercise treated group was increased by 19.2% comparing with the insulin resistant group (P<0.01).ConclusionExercise treatment can significantly elevate the expression of PKB and ameliorate the state of insulin resistance.
