ABSTRACT

Hepatic ischemia-reperfusion injury (HIRI) is a common clinical problem after hepatectomy and liver transplantation and is the main cause of liver dysfunction and liver failure after transplantation. In recent years, autophagy-mediated pathways have become a research hotspot in alleviating HIRI. Autophagy refers to the process in which a large number of substrates such as cytoplasm and damaged organelles are transported into lysosomes for digestion and degradation, so as to constantly renew, reshape, and reuse cells. This article summarizes the research advances in the mechanism of targeting autophagy to alleviate HIRI from the aspects of gene, protein, signaling pathway, inflammatory response, oxidative stress, and mitochondrial and endoplasmic reticulum stress, as well as existing problems and prospects in research, in order to provide theoretical support for the future research on alleviating HIRI by targeting autophagy.

ABSTRACT

Hepatic ischemia-reperfusion injury (HIRI) is a very common complication of liver transplantation, liver resection, and shock. At present, many studies have been conducted on HIRI, but there is still a lack of drugs for radical treatment in clinical practice. Many factors, such as related cells, molecular mechanisms and signaling pathways, reactive oxygen species and oxidative stress response, nitric oxide, and mitochondria, mediate the development and progression of HIRI, which leads to the decline of patients' quality of life and even endangers their life safety. Based on the pathogenesis of HIRI and related articles, this article summarizes the research advances in the prevention and treatment of HIRI with traditional Chinese medicine components, so as to provide theoretical support for basic research and clinical research on HIRI.

ABSTRACT

Objective:To explore the influence of elastic band aerobics training on cardiovascular risk factors in adults.Methods:Twenty-four health management centers participated in the competition. Each individual participant learned elastic band aerobics through video and tested their own indexes before and after intervention. Height, body weight (BW), muscle content (MC), fat content (FC), waist circumference (WC), hip circumference (HC), body fat rate (BFR), resting systolic and diastolic blood pressure (SBP and DBP) were measured. A paired samples t-test was used to analyze pre-and post-test results. Results:After 8 weeks of elastic band aerobics training, there were significant differences in body weight, body mass index (BMI), waist circumference, body fat content, systolic and diastolic blood pressure for male contestants before and after training ( P<0.01). These decreased by 1.39%, 1.45%, 0.98%, 3.61%, 2.28%, 2.34%, and 3.05% respectively. Weight, waist circumference, hip circumference, waist hip ratio, body fat content and rate of female contestants decreased by 1.25%, 1.59%, 0.65%, 0.99%, 4.22%, and 2.38% respectively ( P<0.01). There were significant differences in absenteeism between the top 10 and bottom 14 teams ( P<0.05). Average absenteeism rates were 2.8% and 10.7% respectively. Overall attendance rate of participants was 92.6%, higher than the clinical standard of an 80% level of compliance. Conclusions:Eight-week elastic band aerobics training can significantly improve body composition of adults, reducing body fat rate and waist circumference. Using competition as an opportunity to exercise promotes implementation and persistence of sports training programs.

ABSTRACT

Objective:To explore the application value of non-invasive pressure-strain loop (PSL) in evaluating left ventricular systolic function of young strength athletes with different heart rates.Methods:Thirty-five young wrestlers were collected randomly and divided into 2 groups according to the heart rate: group 1 (heart rate of 40-59 beats/min, n=20) and group 2 (heart rate of 60-80 beats/min, n=15). Thirty healthy young males were selected as the control group at the same period. Non-invasive PSL was used to obtain left ventricular global longitudinal strain (GLS), longitudinal peak strain dispersion (PSD), global myocardial work index (GWI), global constructive myocardial work (GCW), global wasted myocardial work (GWW) and global myocardial work efficiency (GWE) in three groups, and the differences between them were measured. Results:Compared with the control group, GWE in the athlete group 1 and 2 reduced, PSD, GWW were increased, and GWI, GCW in the athlete group 1 were decreased, and all differences were statistically significant (all P<0.05). Compared with the athlete group 1, PSD, GWI, GCW, GWW in the athlete group 2 increased and GWE was reduced (all P<0.05). Conclusions:PSL can quantitatively evaluate the left ventricular myocardial work of young strength athletes with different heart rates, and then assess the effect of heart rates on the left ventricular systolic function of athletes.

ABSTRACT

Objective:To study the protective effect of Xiayuxue Tang on adenine-induced renal fibrosis model in rats and its impact on Wnt/β-catenin and transforming growth factor β1(TGF-β1)/Smad signal pathway. Method:A total of 50 SPF-grade male SD rats were randomly divided into 5 groups: the normal group, the model group, the losartan group (9 mg·kg-1) and low and high dose (2.43,4.86 g·kg-1) of Xiayuxue Tang groups. The rat model of renal fibrosis was established by ig administration adenine (250 mg·kg-1) for 24 consecutive days. The rats were then given the corresponding drugs for 30 consecutive days. The levels of serum creatinine(SCr) and blood urea nitrogen (BUN) were measured by enzyme-linked immunosorbent assay(ELISA). The histopathological changes of renal tissues in rats were observed by hematoxylin and eosin (HE) staining. The collagen deposition in rat renal tissue was observed by Masson staining; the protein expression levels of Wnt5a, Wnt5b, β-catenin, TGF-β1, Smad4, Smad7 in renal tissue were detected respectively by immunohistochemistry(IHC) and Western blot. Result:Compared with the normal group, the results of each experimental group showed that SCr and BUN levels significantly increased in the model group (P<0.01). SCr and BUN levels decreased significantly after the intervention with the Xiayuxue Tang (P<0.01). Compared with the normal group, HE and Masson staining results showed that rats in the model group had severe renal interstitial damage and massive deposition of renal interstitial collagen. The renal interstitial tubule injury was relieved after the intervention with the Xiayuxue Tang, and the renal interstitial collagen deposition decreased. The results of IHC and Western blot showed that compared with the normal group, the expressions of Wnt5a, β-catenin, TGF-β1 protein in the kidney of rats up-regulated (P<0.01), while the expressions of Wnt5b and Smad7 protein down-regulated (P<0.01). After the intervention with Xiayuxue Tang, the expressions of Wnt5a, β-catenin, TGF-β1 protein down-regulated (P<0.01), while the expressions of Wnt5b and Smad7 protein up-regulated(P<0.01). There was no significant difference between the low-dose and high-dose groups with Xiayuxue Tang. Conclusion:Xiayuxue Tang has the protective effect on RIF rats induced by adenine, and its mechanism is related to the inhibition of Wnt/β-catenin and TGF-β1/Smad signal pathway.

ABSTRACT

Objective:To study the relative molecular weight and distribution of polysaccharides in Polygonati Rhizoma (PR) before and after processing, as well as the effects of different polysaccharide fractions on immune function and inflammatory response of mouse peritoneal macrophages. Method:High-performance gel permeation chromatography (HPGPC) was used to determine the relative molecular weight and distribution of polysaccharides in PR (named SC) and polysaccharides in PR processed with wine (named JC), and polysaccharide fractions with different relative molecular weights were obtained by dialysis. Different polysaccharide fractions were applied to mouse peritoneal macrophages, which was normal or induced by lipopolysaccharide (LPS). Cell counting kit-8 (CCK-8) method was used to select the optimal administration concentration. Enzyme linked immunosorbent assay (ELISA) was used to determine the concentrations of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) in the cell supernatant. The Griess method was used to detect the concentration of nitric oxide (NO). Result:SC and JC could be divided into four parts according to relative molecular weight and its distribution range, including part Ⅰ(14 800-2 273 kDa), part Ⅱ(2 148-296 kDa), part Ⅲ(12-1 kDa) and part Ⅳ(818-362 Da). Based on the differences of part Ⅰ and part Ⅲ after processing, the SC and JC were divided into two-part according to the weight-average relative molecular weight (MW). For normal mouse peritoneal macrophages, JC could significantly promote the secretion of TNF-α (P<0.01), while SC had no significant effect. Four polysaccharide fractions, named SD (SC fraction with MW>50 kDa), JD (JC fraction with MW>50 kDa), SX (SC fraction with MW<50 kDa) and JX (JC fraction with MW<50 kDa), also could significantly promote the secretion of TNF-α (P<0.01), but only JX could significantly promote the secretion of NO (P<0.05). In addition, the effect of JX group stimulated secretion of TNF-α was better than the JD group (P<0.01). For the LPS-induced macrophage model, JC and SC group could significantly inhibit the secretion of TNF-α and IL-1β (P<0.01), and the effect of JC was stronger. To compare different polysaccharide fractions, the impact of JX on inhibiting the secretion of TNF-α and IL-1β was significantly stronger than JD (P<0.01), and SX inhibited the secretion of TNF-α was significantly stronger than SD (P<0.01). Conclusion:The relative molecular weight and distribution of polysaccharides in PR before and after processing have changed. JC and SC improve the immune regulation mainly by inhibiting the inflammatory response, the fraction of MW<50 kDa is the main effective part, and the effect of PR polysaccharides in inhibiting the inflammatory is enhanced after processing with wine.

ABSTRACT

Objective:To explore the mechanism of Sishenwan, Baitouweng Tang, and Lianlitang in the treatment of ulcerative colitis (UC), and compare their efficacies on UC in rats. Method:Ninety SD rats of SPF grade were randomly divided into blank group (distilled water, 2 mL·d-1) and experimental group. The rats in the experimental groups were administered with 2,4,6-trinitrobenzene sulfonic acid (TNBS) by clysis to induce the UC model. Subsequently, the model rats were divided into a model group (distilled water, 2 mL·d-1), positive group [sulfasalazine (SASP), 0.4 g·kg-1·d-1], Sishenwan group (1.76 g·kg-1·d-1), a Baitouweng Tang group (1.40 g·kg-1·d-1), and Lianlitang group (2.13 g·kg-1·d-1) according to the random number table. The rats in each group were dosed at 2 mL·d-1 for 14 days. The pathological score for colonic mucosa was detected. Cytokines were detected by the cytokine chip. The enzyme-linked immunosorbent assay (ELISA) was used to detect the free triiodothyronine (FT3), glucagon-like peptide-1 (GLP-1), and corticosterone (CORT) in plasma, and neurotensin (NT), substance P (SP), vasoactive intestinal peptide (VIP), and somatostatin (SST) in colon tissues. Result:Compared with the normal group, the model group showed increased colon mass-length ratio and pathological score for colonic mucosa (P<0.01), infiltration of massive lymphocytes, disordered or absent intestinal villi, elevated levels of cytokine-induced neutrophil chemoattractant-1/2α/β/3 (CINC-1/2α/β/3), interleukin-1α (IL-1α), interferon-inducible protein-10 (IP-10) and other factors in colon tissues (P<0.05), dwindled CORT and GLP-1 levels in plasma (P<0.05), and increased SP content in colon tissues (P<0.05). Compared with the results in the model group, the mucosal injury in the colon of rats in each drug group was relieved. The levels of IL-1α, IP-10, lipopolysaccharide-inducible CXC chemokine (LIX), and L-selectin of rats in the Lianlitang group and Sishenwan group were reduced (P<0.05), and the CINC-3 and IL-17 levels were diminished in the Baitouweng Tang group (P<0.05). The levels of CINC-1/3, IL-1α, and IP-10 were reduced in the SASP group (P<0.05). The plasma FT3 was up-regulated in the Lianlitang group, and the plasma GLP-1 levels were elevated in the three Chinese medicine groups (P<0.05). The VIP content in colon tissues of the Sishenwan group and Baitouweng Tang group was down-regulated (P<0.05), and the SST content in colon tissues of the SASP group was significantly up-regulated (P<0.01). Conclusion:The intervention of Lianlitang and Sishenwan on UC was significant, and the underlying mechanism of action might be related to inflammation inhibition and immune balance by regulating the cytokine network. The efficacy of Lianlitang was predominant, followed by Sishenwan and Baitouweng Tang.

ABSTRACT

Gastric cancer is one of the most common malignant tumors in the digestive system, and precancerous lesion of gastric cancer (PLGC) represents a long-term stage in the process of malignant development of normal gastric mucosa into gastric cancer. Gastric cancer and precancerous lesions are difficult to cure clinically, leaving poor prognosis and a serious negative impact on the quality of daily life of patients. In recent years, studies on cell autophagy have been at the forefront of the natural life science. Regulating autophagy to treat precancerous lesions and prevent gastric cancer has become nowadays a hot topic. Autophagy is a process in which cells enclose some redundant or damaged cytoplasm, proteins and organelles to form autophagosomes, and bind to lysosomes to degrade the contents. Autophagy has bidirectional effect on different cells and different stages of the same cell. Autophagy at a lower level can kill cancer cells, while autophagy can promote the growth and proliferation of cancer cells under stress conditions such as hypoxia, hunger and infection, or when autophagy clears damaged proteins in cells and organelle function is abnormal. Traditional Chinese medicine(TCM), which has low toxicity and easy acceptance by patients, has a positive effect on the treatment of gastric cancer and PLGC. At present, studies on the prevention and treatment of gastric cancer and PLGC by TCM have been carried out in depth with cell autophagy as the breakthrough point. More and more research results have confirmed that TCM can regulate the autophagy process of gastric cancer cells and play an anti-tumor role by interfering with various autophagy related genes, signal pathways and organelles. This paper summarizes the studies on the regulation of cell autophagy by TCM in the treatment of gastric cancer and precancerous lesions, so as to provide references for future studies on the regulation of autophagy by TCM.

ABSTRACT

The β2m (Beta-2-microglobin) gene encodes a non-glycosylated protein that functions as an important component of major histocompatibility complexⅠ(MHCⅠ) for antigen presentation. To evade immune mediated clearance, human tumors and pathogens have adopted different strategies, including loss of MHCⅠexpression. Appropriate animal models are essential for understanding the mechanisms underpinning the clinical treatment of tumor and other human diseases. We constructed β2m knockout mice using CRISPR/Cas9 gene editing tool through embryo microinjection. Subsequently, genotyping and phenotyping of knockout mice were performed by PCR, qPCR, and flow cytometry. Mice genotyping showed that the coding region of the target gene was absent in the knockout mice. Real time PCR showed that mRNA level of β2m was significantly downregulated. Flow cytometry showed that the proportions of CD8+ killer T cells was significantly reduced in a variety of tissues and organs of the immune system. Taken together, we have successfully constructed a strain of β2m knockout mice, which will facilitate subsequent in vivo study on the function and mechanism of the β2m gene.

ABSTRACT

Porcine epidemic diarrhea (PED) is a major disease of pigs that inflicts heavy losses on the global pig industry. The etiologic agent is the porcine epidemic diarrhea virus (PEDV), which is assigned to the genus Alphacoronavirus in the family Coronaviridae. This review consists of five parts, the first of which provides a brief introduction to PEDV and its epidemiology. Part two outlines the passive immunity in new born piglets and the important role of colostrum, while the third part summarizes the characteristics of the immune systems of pregnant sows, discusses the concept of the "gut-mammary gland-secretory IgA(sIgA) axis" and the possible underpinning mechanisms, and proposes issues to be addressed when designing a PEDV live vaccine. The final two parts summarizes the advances in the R&D of PEDV vaccines and prospects future perspectives on prevention and control of PEDV, respectively.
