ABSTRACT

The research on endophytes of medicinal plants mainly relies on the traditional culture and isolation methods. Because of their functions such as promoting host growth, improving stress resistance, promoting the accumulation of medicinal active ingredients or directly producing medicinal active ingredients, the endophytes of medicinal plants have gradually attracted wide attention. However, it was found that the strains isolated by traditional methods were not the true dominant endophytes of medicinal plants by comparing the results of traditional culture isolation with high-throughput sequencing. The blind and random nature of traditional methods leads to the lack of standards in terms of medium selection, culture time and interaction between species. On the contrary, high-throughput sequencing technology is an emerging molecular biology technology developed in recent decades. Due to its high resolution level and indepen-dent culture, it can be used for thorough analysis of the community structure and diversity of environmental microorganisms. Therefore, we proposed the strategy of using high-throughput sequencing technology to guide the traditional culture and isolation of endophytes from medicinal plants. Firstly, the endophytic structure and diversity of medicinal plants were completely clear by high-throughput sequencing technology, and the dominant endophytes of the host were unequivocal. Then according to the characteristics of each dominant endophytes design or query suitable medium for its growth to culture and isolation. Finally, the function of the isolates was studied. This method can prevent researchers from missing out on the important functional strains of the host, expand the research scope of endophytes of medicinal plants, and facilitate the in-depth excavation and utilization of endophytes of medicinal plants.

ABSTRACT

Intercropping farming system is one of the essence of traditional agriculture in China and one of the most common and basic patterns of modern ecological planting. Intercropping system uses the principle of species diversity to create reasonable interspecific interaction conditions with obvious productivity advantages. In this paper, the interspecies interaction is divided into aboveground and underground parts from the space view, and its influence and mechanism on the yield and secondary metabolites of medicinal plants are elaborated.The interspecific interaction in the aboveground part mainly introduces the distribution and utilization of space resources among plants. The interspecific interaction in the underground part mainly introduces the soil rhizosphere effect and related mediating factors, root exudates, soil microorganisms, root space structure and soil environmental factors. On the basis of understanding the mechanism of interspecific interaction, this paper further discusses the application of intercropping in traditional Chinese medicine ecological agriculture, taking the effective control of diseases and insect pests, the increase of medicinal material yield and the improvement of medicinal material quality as the benefit index, so as to seek better advantages of intercropping and provide ideas for the utilization of intercropping production mode in traditional Chinese medicine ecological agriculture.

ABSTRACT

Rhizosphere is the main place for the communication between medicinal plants and rhizosphere microorganisms. Medicinal plants are closely related to the diversity and richness of rhizosphere microorganisms, and rhizosphere microorganisms in the rhizosphere of medicinal plants have important effects on the growth and development, yield, quality and resilience of medicinal plants. The reasonable and effective utilization of the principle of interaction between medicinal plants and rhizosphere microorganisms has practical guiding significance for promoting the growth of medicinal plants, enhancing the ability of resistance to diseases and resisting the invasion of pathogens. This paper reviewed the research status of medicinal plants and rhizosphere microorganisms in recent years, including the influence of medicinal plants on rhizosphere microorganisms, the influence of rhizosphere microorganisms on medicinal plants and the mechanism of interaction between medicinal plants and rhizosphere microorganisms. The problems existing in the study of medicinal plants and rhizosphere microorganisms and the direction for further study were also pointed out.

ABSTRACT

In this paper, Chrysanthemum morifolium was used as the experimental object, eight different planting periods were set up in field plot experiment from April to August, which were 04-15, 05-19, 05-30, 06-09, 06-19, 07-20, 07-31, 08-15. The effects of different treatments on the occurrence of root rot, agronnmic traits, mineral element absorption and content of effective components of Ch. morifolium in Macheng country of Hubei province were studied. The results showed that delaying the planting time could effectively reduce the diseases occurrence of root rot of Ch. morifolium. With the advance of transplanting period, the plant height, the weight of one hundred flowers and the number of flowers of Ch. morifolium showed a trend of gradual decrease, while the number of primary branches and the thickness of main stem and the primary branch increased first and then decreased. The yield of Ch. morifolium per plant and per mu increased with the advance of the planting period, and the yield per mu increased during the planting period on June 19, which was 91.96% higher than that on April 15. And with the delay of the planting period,the absorption and accumulation of potassium(K) elements was promotes. The content of active ingredients such as chlorogenic acid, rutin, luteolin, and 3,5-O dicoffeoacy lquinic acid in the Ch. morifolium increased significantly and then gradually decreased with the delay of the planting period, which indicates that late planting can significantly improve the quality of Ch. morifolium. Considering factors such as the occurrence of root rot disease, yield and active ingredient content, combined with climatic conditions in the Dabie Mountains in eastern Hubei, the optimum planting period of Ch. morifolium was determined from mid-late June to early July.

ABSTRACT

Objective@#To investigate the surgical options for splenic lymph node dissection in patients with advanced gastric cancer undergoing radical total gastrectomy, and to evaluate the sentinel effect of No. 4s lymph node on splenic lymph node metastasis.@*Methods@#A prospective, single-center, randomized and controlled study was carried out (Trial registration, No.NCT02980861). Enrollment criteria: (1) >18 years old and <65 years old; (2) gastric adenocarcinoma locating in the proximal or corpus; (3) preoperative clinical staging as cT2-4aN0-3M0; (4) D2 radical total gastrectomy feasible judged before operation; (5) physical ability score 0 to 1; (6) I to III of ASA classification. Pregnant or lactating women, patients with severe mental illness or previous history of upper abdominal surgery, those suffered from other malignant tumors in the past 5 years, or heart and lung system diseases judged to affect surgery before operation, those receiving preoperative chemotherapy, radiotherapy or targeted therapies, and distant metastases being found during surgery were excluded. According to above criteria, 222 patients at The First Medical Center of Chinese PLA General Hospital from December 2016 to December 2017 were enrolled prospectively and were randomly divided into the laparoscopic splenic hilar lymph node dissection group (laparoscopic group, n=114) and the open splenic hilar lymph node dissection group (open group, n=108). The result of rapid frozen immunohistochemistry of harvested No.4s lymph nodes was used to evaluate the sensitivity and specificity of sentinel effect on splenic hilar lymph node metastasis. The surgical parameters, postoperative recovery parameters, and complication rates were compared between the two groups.@*Results@#There were 80 males and 34 females in the lapascopic group with a mean age of (56.1±10.2) years, and 69 males and 39 females in the open group with a mean age of (58.4±10.9) years. There were no significant differences in baseline data between the two groups (all P>0.05). Total blood loss was less in the laparoscopic group [(96.3±82.4) ml vs. (116.6±101.9) ml, t=1.124, P<0.001], and the amount of bleeding from the splenic hilar lymph nodes dissected was also less than that in the open group [(25.3±17.8) ml vs. (59.5±36.4) ml, t=1.172, P<0.001]. However, the operation time, the time of splenic hilar lymph node, the number of lymph node dissected and number of splenic hilar lymph node dissected were not significantly different between the two groups (all P>0.05). As compared to the open group, the laparoscopic group had shorter time to the first flatus [(1.3±1.2) days vs. (1.6±1.5) days, t=1.665, P=0.021], shorter time to fluid diet [(4.6±1.4) days vs. (4.9 ± 1.6) days, t=1.436, P=0.007], shorter time to remove nasogastric tube [(3.9±2.6) days vs. (4.3±2.4) days, t=0.687, P<0.001] and shorter hospital stay [(10.3±6.6) days vs. (12.1±7.2) days, t=0.697, P<0.001]. Complication rate was 14.0% (16/114) and 12.0% (13/108) in the laparoscopic group and the open group, respectively, without significant difference (χ2=6.723, P=0.331). The sensitivity of the No. 4s lymph node for the prediction of splenic hilar lymph node metastasis reached 89.5%, and the specificity reached 99.6%.@*Conclusions@#Laparoscopic technique is safe and feasible in the treatment of splenic hilar lymph node dissection in advanced gastric cancer. The No.4s lymph node examination has good sentinel effect on predicting the metastasis of splenic hilar lymph nodes.

ABSTRACT

Objective: To observe the clinical efficacy of herb-partitioned spreading moxibustion at Baliao points plus climen for diminished ovarian reserve (DOR). Methods: A total of 60 patients with DOR were randomized into a spreading moxibustion group and a Western medicine group by the random number table method, with 30 cases in each group. The Western medicine group was treated with climen, starting from the 5th day of the menstrual cycle for 21 d. The spreading moxibustion group was treated with herb-partitioned spreading moxibustion at Baliao points on the basis of the medication in the Western medicine group, 1 h per time, once a week. The treatment was performed for 1 month as one treatment course in both groups, for 3 courses in total. The serum follicle-stimulating hormone (FSH), luteinizing hormone (LH) and estradiol (E2) in the patients were measured before and after treatment. The peak systolic velocity (PSV) and resistance index (RI) were also detected. The traditional Chinese medicine (TCM) symptom score was evaluated. The clinical efficacy was evaluated after treatment. Results: The total effective rate in the spreading moxibustion group was 93.3%, which was significantly higher than 80.0% in the Western medicine group, and the difference between the groups was statistically significant (P<0.05). After treatment, the TCM symptom scores, the serum FSH levels, FSH/LH ratios and RI in both groups decreased, and the intra-group differences were all statistically significant (all P<0.05). The serum E2 level and PSV increased compared with those in the same group before treatment, and the intra-group differences were statistically significant (all P<0.05). After treatment, the TCM symptom score, the serum FSH level, FSH/LH ratio and RI in the spreading moxibustion group were lower than those in the Western medicine group, while the serum E2 level and PSV were higher than those in the Western medicine group, and the differences between the groups were statistically significant (all P<0.05). Conclusion: Herb-partitioned spreading moxibustion at Baliao points plus climen can produce valid therapeutic efficacy for DOR. It can improve the clinical symptoms, regulate serum hormone levels and increase ovarian blood perfusion, thus improving ovarian reserve function, producing more significant efficacy than climen alone.

ABSTRACT

Objective:s To evaluate the impacts of prior surgical scores(PSS) on the clinical efficacy and perioperative safety of cytoreductive surgery(CRS) and hyperthermic intraperitoneal chemotherapy(HIPEC) for pseudomyxoma peritonei(PMP).Methods:From the comprehensive PMP database, we collect the cases treated for the first time by CRS+ HIPEC, to form this study cohort. The clinicopathological features, PSS, CRS+ HIPEC details, overall survival(OS), and serious adverse events(SAEs) are systematically analyzed, to study the correlations between PSS and OS or SAEs.Results:335 PMP cases received standardized CRS+ HIPEC in this study. The median OS is 58.2 months for PSS-0 patients, 63.7 months for PSS-1, and 55.4 months for PSS-2/3, with no statistically significant differences in OS among the different PSS groups(χ 2=0.499, P=0.779). Subgroup analysis by pathologic types also found no statistically significant differences among the different PSS groups. Moreover, no significantly statistical differences are observed in overall SAEs(χ 2=0.625, P=0.722), CRS-related SAEs(χ 2=0.267, P=0.901), and non-CRS-related SAEs(χ 2=0.677, P=0.715), among the different PSS groups. Conclusions:PSS does not pose significant impacts on the efficacy and safety of CRS+ HIPEC for PMP patients at experienced treatment center.

ABSTRACT

Objective • To provide guidance for the rational usage of antibiotics, reduction of drug-resistant strains and hospital infection control by investigating the distribution and drug resistance of pathogens isolated from burned patients. Methods • Isolates from burned patients of Ruijin Hospital, Shanghai Jiao Tong University School of Medicine between Jan. 2016 and Dec. 2017 were collected. The VITEK 2 Compact automatic microbial analysis system and K-B disc diffusion method were used for antimicrobial susceptibility test. Statistical analysis was performed to investigate the type of specimens and strains and the resistance rates of major pathogens. Chi-square test was used to compare the changes of the detection rates and drug resistance rates of major pathogens between 2016-2017 and 2013-2014. Results • A total of 1 053 isolates were isolated, and most of them (73.88%) were from wounds. Among them, 609 (57.83%) isolates were gram-negative bacilli (G-B), 422 (40.08%) were gram-positive cocci (G+C) and 9 (0.85%) were fungi. The most common pathogen of G-B was Klebsiella pneumoniae (218, 20.70%) in 2016-2017, the proportion of which was significantly higher than that in 2013-2014 (114, 10.72%) (P=0.000). The most common pathogen of G+C was Staphylococcus aureus (210, 19.94%), the proportion of which was significantly higher than that in 2013-2014 (200, 18.81%) (P=0.009). K. pneumoniae showed low resistance only to piperacillin/tazobactam, cefoperazone/sulbactam, imipenem, meropenem, fosfomycin and tigecycline (90%). The resistance rates of K. pneumoniae to ceftazidime, cefepime, imipenem, meropenem and amikacin in 2016-2017 were significantly higher than those in 2013-2014 (P<0.05). S. aureus was only resistant highly to penicillin (94.63%) and 100% susceptible to vancomycin and linezolid. The proportion of methicillin-resistant S. aureus (MRSA) was 59.05% in 2016-2017, which had no statistical difference compared with that in 2013-2014 (P=0.412). Conclusion • The most two prevalent isolates from burned patients were K. pneumoniae and S. aureus with multi-drug resistance. Improved management and rational use of antibiotics can reduce the incidence of antibiotic resistant pathogens, and it's still important to keep routine surveillance on pathogens isolated from burned patients for control and prevention of nosocomial infections.

ABSTRACT

Objective@#To establish the patient derived xenograft (PDX) model of pseudomyxoma peritonei (PMP), and identify the key characteristics of tumor biology of this model, in order to provide a reliable model for studying the pathological mechanisms and new therapeutic strategies of PMP.@*Methods@#PMP tumor tissue was obtained from surgery and cut into pieces after washing. Then tumor pieces were implanted subcutaneously in BAL B/c-nu mice for 6 stable passages. In the 7th passage, tumor tissue was implanted orthotopically into abdomen. Subcutaneous tumor and orthotopic tumor were then homogenized to make tumor cell suspension, implanted into abdomen of 10 BAL B/c-nu mice through midline laparotomy, 100 μl for each. The key experimental parameters including body weight changes in the observation period, experimental peritoneal cancer index (ePCI) score at the autopsy, histopathological and immunohistochemical characteristics, and gene expression profiles by high-throughput whole-genome exon sequencing were detected and recorded.@*Results@#The successful rate of established orthotopic PDX model of human PMP was 100% (10/10). The animals showed smooth body weight increases after tumor inoculation until day 27, then the body weight began to decrease steadily. Widespread tumor dissemination of PMP tumor through the whole abdomen was found by autopsy, including the diaphragm, liver, spleen, stomach, kidney, parietal peritoneum, bowel and mesenterium. Gelatinous ascites was also observed in abdominopelvic cavity. The ePCI score ranged from 5 to 9, with a 8 of median ePCI. Histopathological studies showed peritoneal mucinous carcinomatosis accompanied with signet ring cells (PMCA-S), obvious tumor cell atypia and parenchymal invasion.Immunohistochemistry showed the expressions of MUC1, MUC2, MUC5AC, CEA, CA199, CK20, CDX-2 and Ki-67 were positive, MUC6, CK7 and p53 were negative. Whole-exome sequencing identified that the most significant genetic alteration is the exon10 missense mutation c. 1621A>C of KIT gene, the mutation abundance was 89.7%.@*Conclusion@#PDX model of PMCA-S is successfully established, which displays the characters of high-degree malignancy, high proliferation and strong aggressiveness.

ABSTRACT

OBJECTIVE: To explore the cause and treatment of bariatric and metabolic surgery reoperation. METHODS: A retrospective analysis was conducted on the clinical data of 54 patients underwent reoperative metabolic and bariatric surgery in Department of Bariatric and Metabolic Surgery,the First Affiliated Hospital of Jinan University from November 2000 to December 2018, and the reasons and surgical techniques of reoperation were analyzed. RESULTS: Causes of reoperation included inadequate weight loss/weight regain in 22 cases(40.7%),adhesive intestinal obstruction in 5 cases(9.3%),mesenteric hernia in 5 cases(9.3%),gastroesophageal reflux in 4 cases(7.4%),anastomotic leakage4 cases(7.4%),anastomotic ulcer bleeding in 3 cases(5.6%),anastomotic stenosis in 3 cases(5.6%),postoperative intraabdominal bleeding in 3 cases(5.6%),recurrent diabetes in 2 cases(3.7%),severe dumping syndrome in 2 cases(3.7%),anastomotic error in 1 case(1.9%). Reoperation methods mainly included Roux-en-Y gastric bypass in 30 cases(55.6%),sleeve gastrostomy in 10 cases(18.5%),hernia repair in 5 cases(9.3%),exploratory laparotomy hemostasis in 4 cases(7.4%),gastroscopic balloon dilatation in 2 cases(3.7%),restoration surgery in 1 case(1.9%),revisional banded gastric bypass with GaBP ring in 1 case(1.9%), hiatal hernia repair in 1 case(1.9%). A total of 54 patients underwent reoperation with a follow-up period of 1 to 102 months. Among them, 42 patients were followed up,and 12 patients were lost to follow-up(follow-up rate of 77.8%). Among the 42 patients who were followed up, 41 of them achieved good results after surgery, and the symptoms and signs were relieved to various degrees. CONCLUSION: The most common cause of reoperative metabolic and bariatric surgery is inadequate weight loss/weight regain.The choice of reoperation method depends on the primary surgical procedure, the cause of failure, and the intraoperative condition.

Objective To investigate the sequence variations of mitochondrial D-loop region in chronic glomerular nephritis patients and family members and their possible associations with chronic glomerular nephritis.Methods 20 patients with chronic glomerular nephritis and 48 unaffected pedigree members,as well as 122 cases of normal control were recruited.mtDNA extracted from peripheral blood were examined by PCR-based assay for D-loop sequence variations,followed by sequencing analysis.Results Compared with the normal control and standard sequence,a total of 61 sequence variants were detected,among these,three high variations were found,and the base variation rate of patients with chronic glomerular nephritis(CGN)and unaffected pedigree members(UAPM)(0.88%,0.72%)significantly increased compared with the control group(0.61%).The distibutional difference of base variation rate in the experimental groups were significantly higher than those in the control group(x2=21.220,7.964,all P<0.05).Especially in microsatellite variation in D310 region,the mutation frequency and base variation rate in patients with CGN were 100.00% and 30.00%,respectively,which of UAPM were 81.30% and 17.95%,respectively,while those in the normal control group were 53.30% and 6.05%,respectively.Among them,the patients with CGN compared with the control group had statistically significant differences(x2=15.610,150.047,all P<0.05).Likewise,UAPM also had statistically significant difference compared with the control group(x2=11.347,66.188,all P<0.05).Conclusion D-Loop of mitochondrial genome mutations may be related to the development of chronic glomerular nephritis.

ABSTRACT

Objective To observe the clinical efficacy and prognosis of nimotuzumab combined with intensity modulated radiotherapy(IMRT) in the treatment of advanced local nasopharyngeal carcinoma.Methods 82 patients with advanced nasopharyngeal carcinoma were selected, and they were randomly divided into observation group(40 cases) and control group(42 cases) according to the digital table.The observation group was treated with nimotuzumab plus IMRT plus cisplatin, and the control group was treated with IMRT plus cisplatin.After 7 weeks of treatment, the clinical efficacy, adverse reactions and T cell subsets were observed.And the patients were followed up for 1 year, 2 years and 3 years to evaluate the survival rate.Results In the observation group, the total remission rate was 75.00%,which was higher than that in the control group(59.09%),the difference was statistically significant(x2=35.81,P<0.05).The CD+4,CD+8 and CD+4/CD+8 levels in the observation group were similar to those in the control group(tCD+4=0.59,P=0.56;tCD+8=1.70,P=0.01;tCD+4/CD+8=0.13,P=0.89).The incidence rates of Ⅰ degree and Ⅱ degree mucosal reaction and dermatitis in the observation group were 35.00%,40.00%,40.00%,35.00%,respectively,which were higher than 9.09%,9.09%,9.09%,9.09% of the control group,the differences were statistically significant(x2Ⅰmucous membrane=4.18,P=0.04;x2Ⅱmucous membrane=5.52,P=0.02;x2Ⅰdermatitis=5.52,P=0.02;x2Ⅱdermatitis=4.18,P=0.04).The 2-year and 3-year survival rates in the observation group were 65.50%,60.00%,respectively,which were significantly higher than 31.82%,27.27% in the control group, the differences were statistically significant(x2=4.11,P=0.04;x2=4.58,P=0.03).Conclusion Nimotuzumab combined with IMRT in the treatment of advanced nasopharyngeal carcinoma has good short-term clinical efficacy, good long-term efficacy and controllable adverse reactions.It is worthy of clinical application.

ABSTRACT

Aim To synthesize 8-bromo-ethoxy Rhein and investigate its mechanisms and inhibition effect on hepatitis B surface antigen ( HBsAg ) and e antigen ( HBeAg) in HepG2.2.15 cells.Methods 8-bromo-ethoxy Rhein was synthesized based on the chemical structure of Rhein , and its structure was identified by IR,1 H-NMR and 13 C-NMR spectra.MTT assay was used to test the inhibitory effect of 8-bromo-ethoxy Rhein on HepG2.2.15 cells.After the cells treatment by 8-bromo-ethoxy Rhein , the HBsAg and HBeAg in cell supernatant were detected by ELISA .The expres-sion of hepatitis B virus X gene ( HBx) was detected by Western blot .The cell cycles were examined with flow cytometry.The intracellular free calcium concentration was detected by laser scanning confocal microscopy . Results The structure of 8-bromo-ethoxy Rhein was confirmed by IR,1 H-NMR and 13 C-NMR.MTT results showed that synthetic product and Rhein could inhibit the cell proliferation in HepG2.2.15 cells.After trea-ted with 8-bromo-ethoxy Rhein and Rhein for 72 h,the half inhibitory concentration 50%( IC50 ) was 14.29 mg? L-1 and 11.59 mg? L-1 , respectively .Using non-cytotoxic dose of 8-bromo-ethoxy Rhein , the inhibitory effect on HBsAg and HBeAg was gradually enhanced with increasing 8-bromo-ethoxy Rhein concentration . The inhibitory effect of synthetic product on hepatitis B virus was better than that of Rhein .8-bromo-ethoxy Rhein could down-regulate the expression of HBx , in-tracellular calcium ion concentration and block the hepatitis B virus ( HBV ) replication.Flow cytometry results showed 8-bromo-ethoxy Rhein didn′t affect the cell cycle .Conclusions Compare with Rhein , the synthesis of 8-bromo-ethoxy Rhein shows stronger inhi-bition on hepatitis B virus in HepG2.2.15, and its mechanisms may involve down-regulating the expres-sion of HBx and reducing calcium ion concentration .

ABSTRACT

Objective: To investigate whether metformin can reverse drug-resistance of ovarian cancer cells to cisplatin, and explore its underlying mechanism. Methods: Cisplatin-resistant ovarian cancer cells C1 3K and CP70 were used in this study. The small interfering RNAs (siRNAs) targeting excision repair cross-complemention 1 (ERCC1) gene were synthesized and transfected into C1 3K and CP70 cells, respectively. The change of cisplatin-resistance of C1 3K and CP70 cells was detected by cell count kit-8 (CCK-8) assay after metformin treatment and ERCC1 siRNA transfection. The expressions of ERCC1 mRNA in C13K and CP70 cells treated with different concentrations of metformin were detected by real-time fluorescent quantitative-PCR. The activation of AMP-activated protein kinase (AMPK) and suppression of p38 mitogen-activated protein kinase (p38MAPK) in C1 3K and CP70 cells after metformin treatment were evaluated by Western blotting. Then the expression level of ERCC1 protein in C1 3K and CP70 cells treated with metformin (combined with Compound C, a blocking agent of AMPK signaling pathway, or p38MAPK siRNA transfection) or SB203580 (an inhibitor of p38MAPK signaling pathway) was detected by Western blotting. Results: Metformin could reverse drug-resistance of C13K and CP70 cells to cisplatin (P 0.05). Conclusion: Metformin may reverse the drug-resistance of cisplatin-resistant ovarian cancer cells to cisplatin. This effect of metformin may be due to the inhibition of p38MAPK signaling pathway and the downregulation of ERCC1 gene expression in ovarian cancer cells.

ABSTRACT

Objective To investigate the effects of metformin on cell proliferation in differentiation degree of endometrial carcinoma cells and related mechanisms. Methods The endometrial cancer cell lines Ishikawa and AN3CA were used. Cell proliferation was assessed after exposure to metformin with or without epithelial growth factor receptor (EGFR) inhibitor AG1478 by cell counting kit-8 (CCK-8) method. EGFR mRNA was determined by reverse transcription (RT)-PCR. The expression of phosphorylation EGFR (p-EGFR) and total EGFR (t-EGFR) and phosphorylation extracellular signal-regulated kinase 1/2 (p-ERK1/2) and total ERK1/2 (t-ERK1/2) were examined by western blot. Results (1)CCK-8 experiment showed that metformin could inhibit the proliferation of endometrial cancer cells in a time-dependent manner and a dose-dependent manner (P0.05). But the expression levels of p-EGFR and p-ERK1/2 protein were significantly lower between two groups (P<0.01), which showed a time-dependent manner(P<0.01). Conclusion Metformin could inhibit the proliferation of endometrial cancer cells, the inhibition is associated with the differentiation degree of cancer cells. Metformin could enhance the EGFR signaling pathway inhibitor AG1478 inhibition of endometrial cancer cells, which may inhibit EGFR expression of phosphorylated proteins to inhibit the phosphorylation of ERK1/2 proteins and then inhibit proliferation of endometrial cancer cells.

ABSTRACT

Objective To explore the effect of the family-centered prenatal maternal health education on delivery modes and breastfeeding.Methods Two hundred and thirty pregnant women who accepted prenatal maternal health education were divided into the observation group and the control group according to whether their families involved in the prenatal education.There were 108 cases in the observation group and 122 in the control group.The observation group was given prenatal health education involving the pregnant women and their caregivers or their husbands.The control group was given prenatal health education according to the previously-designed course without presence of their families.The two groups were compared in delivery mode,extent of pain during production and breast feeding.Results The rates of vaginal delivery and the breast feeding at discharge in the observation group were higher than the control group(P<0.05 for both).The rate of breast swelling in 72 hours was significantly lower than the control group(P<0.01). Conclusion Family-centered prenatal maternmal health education is good for the natural birth rate,improvement of breastfeeding rate and reduction the breast swelling.

BACKGROUND: Effective drugs for improving vascular elasticity and revascularization are few for treating Leriche syndrome (chronic lower extremities ischemia) induced by abdominal aortic occlusion.Vascular outflow tract reconstruction in vascular prosthesis transplantation can rapidly improve blood supply in the distal extremities.OBJECTIVE: To evaluate the biocompatibility and outcome of vascular prosthesis transplantation for the treatment of Leriche syndrome.DESIGN: Controlled study before and after surgery.SETTING: Department of General Surgery of General Hospital of Chinese PLA and Department of Thoracic and Cardiovascular Surgery of Shijingshan Hospital.PARTICIPANTS: Sixty-two patients and sixteen patients with Leriche syndrome were enrolled in this study,who were respectively from Department of General Surgery of General Hospital of Chinese PLA between January 1995 and January 2007 and from Department of Thoracic and Cardiovascular Surgery of Shijingshan Hospital between January 2001 and January 2007.The subjects were 66 males and 12 females,whose age ranged from 46-75 years (averagely 58 years) and onset time from 6 months-8 years (averagely 2 years and 6 months).Informed consents were obtained from all patients.METHODS: After general anaesthesia or epidural anesthesia,vascular bypass was performed,including aortofemoral bypass in 63 cases and aortoiliac bypass in 15 cases.Vascular prostheses included polytetrafiuoroethylene (FIFE)vascular prosthesis in 18 cases and polyester vascular prosthesis in 60 cases.PTFE vascular prosthesis (Gore,USA) is a kind of inert material with low biological response.Its fibers with micropores arrange irregularly.Polyester vascular prosthesis (Intervascular,France) is a kind of knitting polyester products,which is made from high-purity cattle collagen Ⅰ with high biocompatibility.To inlay heparin molecules that can retain 4 weeks into the surface of vessels coated with collagen can prevent thrombogenesis and inhibit hyperplasy of smooth muscle cells (SMC).The type is IGK1608;internal diameter of the bole is 16 nun; internal diameter of the branch is 8 mm.Therapeutic prescriptions were approved by Hospital's Ethics Committee.Follow-up was performed for 3 months-5 years by vascular Color Doppler Sonography,spiral CT angiography (SCTA) three-dimensional reconstruction or digital subtraction angiography (DSA) in hospital.MAIN OUTCOME MEASURES: Biocompatibility of vascular prosthesis and outcome of vascular prosthesis transplantation.RESULTS: Three months after vascular prosthesis transplantation,thrombus was not detected on vascular anastomosis and in vascular prosthesis by Color Doppler Sonography and SCTA.Seventy-seven patients were followed up for over one year,and their stomas were unobstructed.Seventy-six patients were followed up for over five years,and the patency rate of stomas was 89% (68/76).No changes in blood plaque,lencocyte count,haematoglobin or liver and kidney function were found after transplantation.Seven days after surgery,affected extremities with ischemic symptom were improved.Three months later,ischemic symptom disappeared.CONCLUSION: With good biocompatibility of vascular prosthesis,vascular prosthesis transplantation in the treatment of Leriche syndrome has a good outcome following aortofemoral bypass and aortoiliac bypass.

ABSTRACT

<p><b>OBJECTIVE</b>To study the genetic diversity of medicinal Dendrobium by SRAP.</p><p><b>METHOD</b>The genetic diversity of 9 spices Dendrobium was studied by using the optimized SRAP reaction system. The NTSYS software was used to analyze the markers.</p><p><b>RESULT</b>Forty primer pairs were selected from 88 amplified 1 782 polymorphic bands with an average of 44.55 polymorphic bands per primer pair. Cluster analysis using UPGMA method based on the data of SRAP amplified bands by 40 primer pairs showed that 9 spices of could be distinguished into two main groups. Jaccard's similarity coefficient ranged from 0.330 2-0.789 2.</p><p><b>CONCLUSION</b>The results of this research indicate that SRAP molecular marker is efficient to study the medical Dendrobium genetic diversity.</p>

ABSTRACT

Sugarcane stem is an ideal organ for producing foreign pharmaceutical proteins and chemicals by genetic engineering.A perfect promoter driving foreign gene to express strongly and specifically in sugarcane stem is necessary for this purpose.In order to isolate a Sugarcane stem-specific promoter,a fragment of 1968bp nucleotide sequence(Ppst2a)upstream 5' of sugarcane pst2a gene,which was demonstrated to express specifically in sugarcane stem previously was isolated by using chromosomal walking.Bioinformatical analysis of this sequence shows that the sequence contains some typical elements of a promoter.To identify the stem-specific of this promoter,a construct was derived from pCAMBIA1301,which original CaMV 35S promoter was replaced by the 1968bp nucleotide sequence,and named as pCAMBIA1900.Transformations of pCAMBIA1900 and pCAMBIA1301 to leaves and stem pieces of sugarcane were carried out by using particle bombardment.The transient expression of gus showed that the gus expressed specifically in sugarcane with a little higher level compared with CaMV 35S.It is the first report that pst2a promoter is a potential stem-specific promoter which can further be used in transgenes into sugarcane.

ABSTRACT

Objective To explore whether gastrointestinal carcinoma could influence differentiation and maturation of dentritic cell (DC) in cultured peripheral blood mononuclear cells (PBMC). Methods PBMC of peripheral blood obtained either from healthy individuals or gastrointestinal carcinoma patients were incubated to induce into DC with the aid of addition of GM-CSF, IL-4 and TNF-?. The percentage of DC and expression rate of markers CD83, CD1a were determined by flow cytometric analysis. Results The quantity of PBMC isolated from gastrointestinal carcinoma patients was less than that from healthy individuals with same volume of blood. DC could be induced from PBMC of gastrointestinal carcinoma patients, and they could express B7 costimulatory molecules as those of healthy individuals. But the expression rates of CD83, CD1a were significantly different in two groups. Conclusion The quantity of PBMC and their ability to differentiate into DC seemed to be decreased in gastrointestinal carcinoma patients.
