ABSTRACT

The safety of traditional Chinese medicine is affected by many factors, and the influence of exogenous harmful substances has been concerned and become a hot spot in recent years, especially heavy metals, pesticide residues and some other harmful substances. In order to explore the effects of non-soil and non-pesticide treatment on residues of these two harmful substances, the heavy metals and agricultural residues of Lilii Bulbus were detected, and the correlation of the data was analyzed. In this experiment, heavy metals and pesticide residues of Scrophulariae Radix were detected, and correlation analysis was conducted for their data. The mechanism of transport phase was interpreted with statistical moment similarity tool of total fingerprint by supramolecular chemistry theory. A large number of experimental data in this paper showed that heavy metals and pesticide contents in Lilii Bulbus basaltifolia had a positive correlation, which was closely related to supramolecular phenomena. Moreover, the similarity of fingerprints between Lilii Bulbus and Scrophulariae Radix suggested that Lilii Bulbus and Scrophulariae Radixa had a high selectivity in absorption of agricultural residues, which proved that the absorption of pesticides in Lilii Bulbus and Scrophulariae Radix from different habitats had the function of supramolecular imprinting template. It was considered that medicinal plant was a giant complex supramolecule with various levels of " imprinted template" . Heavy metals and agricultural residues were also involved in plant growth, forming an " imprinted template" for the formation of supramolecules between agricultural residues and heavy metals. After heavy metals and agricultural residues formed supramolecules, their liposolubility and permeability changed in varying degrees, and their transport in medicinal plants was promoted. Finally, the heavy metal supramolecules of pesticides were absorbed, distributed, aggregated and accumulated in plants. The purpose of the study was to reveal the mechanism of heavy metal and pesticide supramolecule transport, provide a new direction for the treatment of heavy metals and pesticide residues, and ensure the safety of traditional Chinese medicine.

ABSTRACT

Objective@#To investigate the relationship between human cytomegalovirus (HCMV) infection and peripheral blood CD14 CD16 monocytes in the pathogenesis of coronary heart disease (CHD), and to elucidate the mechanism of pathogenesis in CHD by analyzing the correlation between infection, inflammation, and CHD, to provide a basis for the prevention, evaluation, and treatment of the disease.@*Methods@#In total, 192 patients with CHD were divided into three groups: latent CHD, angina pectoris, and myocardial infarction. HCMV-IgM and -IgG antibodies were assessed using ELISA; CD14 CD16 monocytes were counted using a five-type automated hematology analyzer; mononuclear cells were assessed using fluorescence-activated cell sorting; and an automatic biochemical analyzer was used to measure the levels of triglyceride, cholesterol, high- and low-density lipoprotein cholesterols, lipoprotein, hs-CRp and Hcy.@*Results@#The positive rates of HCMV-IgM and -IgG were significantly higher in the CHD groups than in the control group. HCMV infection affects lipid metabolism to promote immune and inflammatory responses.@*Conclusion@#HCMV infection has a specific correlation with the occurrence and development of CHD. The expression of CD14 CD16 mononuclear cells in the CHD group was increased accordingly and correlated with acute HCMV infection. Thus, HCMV antibody as well as peripheral blood CD14 CD16 mononuclear cells can be used to monitor the occurrence and development of CHD.

OBJECTIVES@#Investigate the influence of benazepril and amlodipine on the expression of secretin (PZ) and somatostatin (SS) in spontaneously hypertensive rats (SHR).@*METHODS@#Forty-five SHRs (14 weeks old, male) were randomly assigned into 3 groups (=15):SHR group, Benazepril group (which was given benazepril 0.90 mg·kg·d) and Amlodipine group (SHRs were given amlodipine 0.45 mg· kg·d), taking WistarKyoto(WKY) as normal control (=15), meanwhile, rats in SHR group and WKY group were given the same volume of distilled water. After 8 weeks of intervention, the expression of protein and mRNA of PZ in duodenum and SS in sinuses ventriculi was detected by enzyme-linked immunoassay and RT-PCR.@*RESULTS@#After 8 weeks of intervention, compared with the WKY group, the expression of protein and mRNA of PZ in duodenum and SS in sinuses ventriculi was increased significantly in SHR group (<0. 05). Compared with SHR group, the expression of PZ in duodenum and SS in sinuses ventriculi was decreased significantly in Benazepril group and Amlodipine group (<0.05). Compared with Benazepril group, in Amlodipine group the expression of PZ mRNA in duodenum and SS mRNA in sinuses ventriculi was decreased more significantly (<0.05).@*CONCLUSIONS@#The regulation disorder of PZ in duodenum and SS in sinuses ventriculi exists in SHR. The antihypertensive effect of benazepril and amlodipine may be realized by regulating the expression of PZ and SS, while the regulation of amlodipine is more obvious than benazepril.

ABSTRACT

Objective To investigate the TCM syndrome type distribution of hypertensive patients; To analyze the correlation of blood pressure variability (BPV) parameters and its influence factors. Methods The data of 515 hypertensive patients including demographic information, laboratory test parameters, risk factors and clinical symptoms were collected for correlation analysis and the patients were divided to different TCM syndromes. BPV parameters of 515 hypertensive patients were acquired by 24 h ambulatory blood pressure, then our study analyzed and compared the differents of 24 h MSBP, 24 h MDBP, 24 h MSCV, 24 h MDCV, DMSBP, DMDBP, DMSCV, DMDCV, NMSBP, NMDBP, NMSCV, NMDCV between different TCM syndromes. Results There were 160 cases with hyperactivity of yang due to yin deficiency syndrome, 136 cases with turbid phlegm and blood stasis syndrome, 83 cases with overabundant liver-fire syndrome, 69 cases with deficiency of kidney qi, and 67 cases with abundant phlegm-dampness syndrome. The difference of gender and age was statistically significant between different TCM syndromes (P<0.05). Compared with the group of hyperactivity of yang due to yin deficiency syndrome, the level of 24 h MSBP, 24 h MDBP, DMSBP, DMDBP, NMSBP, NMDBP, 24 h MSCV, 24 h MDCV, DMSCV, DMDCV, NMSCV and NMDCV had statistical significance in the groups of turbid phlegm and blood stasis syndrome, overabundant liver-fire syndrome, deficiency of kidney qi and abundant phlegm-dampness syndrome (P<0.05). Logistic regression analysis showed that the factors including gender (female), insomnia (time<6 h), sodium salt intake, emotion and educational level were positively correlated to hyperactivity of yang due to yin deficiency, and other factors including gender (female), age, sodium salt intake and educational level positively correlated with turbid phlegm and blood stasis syndrome. And the study also showed that age, sodium salt intake, family history of hypertension and educational level were positively correlated to abundant phlegm-dampness syndrome. Conclusion The BPV parameters and blood pressure are significantly increased d in the group of hyperactivity of yang due to yin deficiency than other groups, but decrease in the group of abundant phlegm-dampness syndrome. Hyperactivity of yang due to yin deficiency syndrome, abundant phlegm-dampness syndrome and turbid phlegm and blood stasis syndrome are closely related to the influencing factors that lead to cardiovascular and cerebrovascular events.

OBJECTIVE@#To discuss the effect of dimethyl fumarate (DMF) on rats with l-arginine induced chronic pancreatitis (CP).@*METHODS@#Male Wistar rats were given DMF treatment (25 mg/kg) by oral lavage method; then Wistar rats were given the intraperitoneal injection of l-arginine for 5 times (250 mg/100 kg, twice per time, each interval of 1 h) for building of CP model. Rats were divided into control group, CP group, DMF group and CP + DMF group. Rats in CP + DMF group were given the oral intragastric administration of DMF (25 mg/kg), while rats in control group and CP group were given the equal volume of normal saline. The weight of rats was evaluated and the intraperitoneal glucose tolerance test was performed (IPGTT, 2 g/kg). The islet of rats was isolated and then flow cytometry was employed to evaluate the quality and activity of islets. Meanwhile, the histology of non-endocrine tissues and levels of myeloperoxidase (MPO) and malondialdehyde (MDA) were detected.@*RESULTS@#Compared with control group, the weight of rats in CP group was significantly reduced at week 2, 4 and 6; the blood glucose significantly increased, AUC increased, the histopathological scores of pancreatic atrophy, acinar injury, edema and cellular infiltration increased, levels of MDA and MPO increased, the islet equivalent and islet activity decreased at 0, 30, 60, 120 and 180 min. Compared with CP group, the weight of rats in CP + DMF group significantly increased at week 2, 4 and 6; the blood glucose significantly decreased, AUC decreased, the histopathological scores of pancreatic atrophy, acinar injury, edema and cellular infiltration decreased, levels of MDA and MPO decreased, the islet equivalent and islet activity increased at 0, 30, 60, 120 and 180 min.@*CONCLUSIONS@#DMF treatment can improve CP induced by l-arginine and islet function in rats.

ABSTRACT

OBJECTIVE: To discuss the methods for clinical rational use and state regulation of high risk Traditional Chinese Medicine Injections (TCMIs) based on our hospital TCMIs analysis and literature review. METHODS: By analyzing the prescription rates and amount percentage of TCMIs in emergency, outpatient and inpatient departments of our hospital as well as literature searching to learn the current state and conditions of TCMIs. RESULTS: Utilities of TCMIs from 2012 to 2014 shows an elevated tendency with percentages of 1.19%, 1.34% and 1.40% respectively. Emergency, ENT and internal neurology departments oi outpatient are the top 3 on prescription rates and amount percentage. Inpatients' order rates of TCMIs are 0.79%, 0.80% and 0.79% respectively. Oncology (20%-26%), emergency (12%-14%) and internal neurology (10%-11%) departments are the top 3 amount rates in ordering TCMIs,however, the department of TCM only account for 4%-5%. The percentages of TMCIs amount vs. annual total drug amount during 2012-2014 are 2.10%, 2.07% and 2.14%, which is lower than literature reports. CONCLUSION: Not all the TCMIs use in our hospital are rational, mainly reflects in non-TCM doctors having used the most amount of TCMIs. Therefore, the TCM consultation policy should be established to support TCMI rational utility, TCMIs labels should be strictly regulated and the quality standards of TCMIs should be elevated to ensure the safety and effectiveness. All above efforts should be done to protect and develop TCMI, an innovation of TCM.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the risk factors of prostate cancer in urban Qingdao and provide some theoretical evidence for the scientific prevention and treatment of the disease.</p><p><b>METHODS</b>We performed a hospital-based matched case-control study in Qingdao Municipal Hospital. The cases and controls were matched in age, gender, nationality and the place of residence. All the subjects were interviewed face to face in the hospital using a questionnaire, and the data analyzed by the conditional logistic regression method.</p><p><b>RESULTS</b>According to the 258 valid questionnaires collected, the prostate cancer risk was significantly higher in the cases with a family history of cancer than in those without (OR = 2.58), and so was it in the men with the first spermatorrhea at the age of < or = 15 years than in those at the age of > or = 18 years (OR = 2.27). A decreased risk of prostate cancer was found among the men with the first experience of sexual intercourse between 25 to 30 years of age (OR = 0.76). An increased risk was shown in those with sexual intercourses > or = 4 times per week before 35 years old (OR = 2.57), masturbations > or = 3 times per week (OR = 2.30) and a drinking history (alcohol > or = 150 g/d) of > or = 10 years (OR = 2.83).</p><p><b>CONCLUSION</b>Positive family history of cancer, earlier age of the first spermatorrhea, sexual intercourses > or = 4 times per week before 35 years old, frequent masturbations, and heavy drinking for more than 10 years are risk factors for prostate cancer.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the computed tomography (CT) manifestations of liver involvement in children with Langerhans cell histiocytosis (LCH).</p><p><b>METHODS</b>Retrospective analysis was performed on 9 LCH children with liver involvement confirmed by clinical, laboratory and pathological examinations to investigate the CT manifestations of this condition. These children, including 6 males and 3 females, had undergone both plain CT scan and dual-phase (the arterial and portal venous phases) contrast-enhanced CT scan.</p><p><b>RESULTS</b>The main CT manifestations included hepatomegaly (8 cases); periportal dendritic hypodense lesions or "periportal halo sign" (7 cases) which were mildly or moderately enhanced in the arterial phase; intrahepatic bile duct dilatation (5 cases); lymphadenopathy in the hepatic hilar or retroperitoneal region (4 cases); and diffuse small hypodense nodules (3 cases), which showed annular enhancement on the contrast-enhanced CT scan.</p><p><b>CONCLUSIONS</b>CT findings may be helpful in the early diagnosis and treatment of LCH in children.</p>

ABSTRACT

This study was designed to describe the clinical features and management outcomes of severe hand, foot and mouth disease [HFMD]. Data on 147 severe HFMD patients during an outbreak in 2009 were analyzed. Most patients were under 3 years of age; 102 [69.4%] were boys. All had skin rashes and fever of ?38°C. All [n = 147, 100%] showed signs of central nervous system involvement, such as lethargy [n = 124, 84.4%], myoclonic jerks [n = 76, 51.7%], or drowsiness [n = 34, 23.1%]. Respiratory symptoms were mainly tachypnea [n = 112, 76.2%] or bradypnea [n = 21, 14.3%]. Common cardiovascular symptoms included tachycardia [n = 134, 91.2%] and hypertension [n = 23, 15.5%]. Chest X-ray showed increased markings in 76 [51.7%] or consolidation in 44 [29.9%]. Hyperglycemia and elevated blood lactic acid levels were found in 127 [86.4%] and 130 [88.4%], respectively. Positive enterovirus EV71-PCR was found in 113 [76.9%]. All patients were treated with mechanical ventilation for 61.2 +/- 12.8 h [range, 40-96 h], as well as mannitol, dexamethasone, gamma globulin and ribavirin. Dopamine, dobutamine or amrinone was administered in 58.5, 51.0 and 21.8%, respectively. Three patients [2%] died during hospitalization. All others had a full recovery and were discharged after 14.2 +/- 1.6 days [range, 12-17 days]. Central nervous and cardiorespiratory systems were involved in the patients with severe HFMD. Fasting blood glucose and lactic acid levels increased in the majority of patients. Mechanical ventilation and supportive pharmacotherapy were associated with a good clinical outcome in these patients

Through introduction of the methodological mechanism and comparison with classic randomized controlled trial, the status and the applicability of the expertise-based randomized controlled trials in clinic are explored, and its characteristics in acupuncture clinical application are analyzed. It is held that expertise-based randomized controlled trial is more suitable for the acupuncture clinical research, especially for acupuncture practice which emphasizes manipulations and different schools.

ABSTRACT

During the infection of host cells, IE2 protein is one of the first and most abundantly expressed products of HCMV genome, which plays an important role in the controlling of cell cycle and apoptosis. But the correlation between expression level and anti-apoptotic activity of IE2 protein is still not clear. In this study, we successfully established a HCMV IE2 protein expression cell line that was controlled by Tet-On system. The effect of IE2 protein on cell apoptosis and the expression of p53 was detected under different condition of induction. Our results showed that the IE2 protein could inhibit cell apoptosis induced by TNF-alpha. Additionally, the anti-apoptotic activity of IE2 protein seemed to be relevant to its expression level. However, we failed to detect any difference of p53 expression between the IE2 protein expression and non-expression cells. These data indicated that the IE2 protein might inhibit cell apoptosis through regulating different signal pathways.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the change of endogenic nerve growth factor (NGF) expression in human glioma cells infected with human cytomegalovirus (HCMV).</p><p><b>METHODS</b>U251 cells were cultured in RPMI 1640 culture medium and infected with HCMV AD169 strain in vitro to establish a cell model of viral infection. Morphologic changes of U251 cells were observed under inverted microscope before and after infection with HCMV. Expression of NGF gene and protein of cells was detected by RT-PCR and Western blotting before and after infection with HCMV.</p><p><b>RESULTS</b>The cytopathic effects of HCMV-infected cells appeared on day 5 after infection. However, differential NGF expression was evident on day 7. NGF expression was decreased significantly in U251 cells on day 7 after infection in comparison with control group (P < 0.05).</p><p><b>CONCLUSION</b>HCMV can down-regulate endogenous NGF levels in human glioma cell line U251.</p>

Human herpes simplex virus 1 (HSV-1) causes facial,ocular,and encephalitic disease and is associated with latent infection and cancer.Here,we developed a means of studying the pathogenesis of HSV-1 infection at the protein level by using the SELDI Protein Chip to detect changes of protein expression in Vero cells cultured in vitro.After infection with HSV-1 and culture for 12,24 or 48 h,cells were harvested and lysed.IMAC3 arrays were applied to SELDI-TOF-MS to detect proteomic differences before and after infection.The chip detected a series of differentially expressed protein peaks.Interestingly,both peaks at 16 912 Da and 17 581 Da corresponded precisely with the molecular mass of ISG 15,which may participate in antiviral activity during the process of infection.Thus,the results we obtained can serve as a basis to study the pathogenesis of HSV-1 and the interaction between the virus and its host.In addition,they can help in the discovery of new therapeutic targets for treatment of HSV-1 infection.

ABSTRACT

<p><b>OBJECTIVE</b>Many studies have demonstrated that low levels of insulin-like growth factor-I (IGF-I) may be associated with the hypoxic-ischemic brain damage (HIBD) and that IGF-I has a neuroprotective effect. The role of IGF-II, a structurally and functionally homologous polypeptide with IGF-I, is unclear in HIBD. This study was designed to observe the changes of serum and cerebrospinal fluid (CSF) IGF-II levels in neonates with hypoxic-ischemic encephalopathy (HIE) and to investigate its effects on HIE.</p><p><b>METHODS</b>Serum and CSF IGF-II levels in 41 neonates with HIE were measured by radioimmunoassay in the acute phase (postnatal age 12-24 hrs) and the convalescence phase (postnatal age 10-12 days). The 41 HIE neonates included 10 cases of mild, 12 moderate, and 19 severe HIE. Serum samples of 10 normal neonates were used as controls.</p><p><b>RESULTS</b>In the acute phase, serum IGF-II levels in the Mild HIE group (203.28 +/- 40.09 ng/mL) and the Moderate HIE group (192.33 +/- 39.66 ng/mL) were not significantly reduced, but were obviously reduced in the Severe HIE group (116.72 +/- 39.50 ng/mL) compared with normal controls (229.38 +/- 43.39 ng/mL) (P<0.01). During the convalescence phase, serum IGF-II levels in the Mild HIE group (285.53 +/- 49.44 ng/mL) and in the Moderate HIE group (278.69 +/- 51.34 ng/mL) increased significantly (P < 0.01); CSF IGF-II levels increased in the Mild HIE group from 27.23 +/- 7.82 ng/mL (acute phase) to 81.58 +/- 9.77 ng/mL (convalescence phase) (P < 0.01) and also increased in the Moderate HIE group from 23.43 +/- 7.79 ng/mL (acute phase) to 78.48 +/- 10.44 ng/mL (convalescence phase) (P < 0.01). The patients from the severe HIE group whose neurological symptoms or signs were improved in the convalescence showed higher serum and CSF IGF-II levels than in the acute phase (254.08 +/- 48.50 ng/mL vs 122.21 +/- 46.26 ng/mL; 69.42 +/- 10.20 ng/mL vs 15.05 +/- 7.03 ng/mL; P < 0.01). A positive correlation was found between the serum and CSF IGF-II levels in the HIE group (r=0.69, P < 0.01).</p><p><b>CONCLUSIONS</b>IGF-II levels in serum and CSF are associated with the pathogenesis and the prognosis of neonatal HIE.</p>

OBJECTIVE@#To evaluate the effect of health education on the symptoms, lung function and life quality in patients with stable chronic obstructive pulmonary diseases (COPD).@*METHODS@#Eighty-two patients were assigned into 2 groups randomly: The treatment group (n = 43) accepted health education for 6 months and the control group (n = 39) did not. Before and after the 6 months, we observed the number of smokers, Borg score, inhale treatment, times of acute episode, SGRQ score, lung function, and the therapeutic effect in the two groups.@*RESULTS@#Six months later, the ratio of smokers in the treatment group was 13.95%, much lower than that in the control group (35.90%, P = 0.021); the Borg score of the treatment group decreased from 4.86 +/- 1.21 to 3.38 +/- 0.94 (P =0.000), but there was no difference in the control group; the ratios of accepted inhale treatment in the treatment group and the control group were 100% (43/43) and 20.51 % (8/39) respectively (P = 0.000), and the accuracy rates were 100% (43/43 ) in the treatment group and 12. 82% (5/39) in the control group (P =0. 000); the times of acute episode in the treatment group was 1.51 +/- 1.53, much lower than that in the control group (4.46 +/- 5.17, P = 0. 000); the indexes of lung function before and after the 6 months between the two groups had no significant difference. The total score, the symptom section score, the activity section score, and the impact section score of SGRQ after the 6 months of health education were much lower than those of 6 months before, and those in the control group had no significant difference.@*CONCLUSION@#Six months of the health education can decrease the ratio of smokers and the times of acute episode, and to improve the life quality of patients with stable COPD.

ABSTRACT

<p><b>AIM</b>To study the mechanism of ginsenosides Rg1 and Rb1 promoting glutamic acid release from PC12 cells.</p><p><b>METHODS</b>The amount of glutamic acid released from PC12 cells was measured by high performance liquid chromatography (HPLC). The effect of Rg1 and Rb1 on the phosphorylation of synapsins was detected with immunofluorescent staining and Western blotting.</p><p><b>RESULTS</b>Both Rg1 (10 micromol x L(-1)) and Rb1 (10 micromol x L(-1)) increased glutamic acid release from PC12 cells. The release of glutamic acid was decreased by pre-incubating with the PKA inhibitor H89. H89 inhibited the release of glutamic acid induced by Rb1, but had no effect on the release of glutamic acid induced by Rg1. Moreover, Rb1 enhanced the phosphorylation of synapsins via PKA pathway, Rg1 was out of touch with this.</p><p><b>CONCLUSION</b>Rb1 may promote release of neurotransmitters by increasing the phosphorylation of synapsins via PKA pathway, whereas the up-regulation of neurotransmitters release induced by Rg1 is independent of the phosphorylation of synapsins.</p>

ABSTRACT

OBJECTIVE@#To investigate the changes of C-reactive protein (CRP) level in patients with chronic obstructive pulmonary diseases (COPD) and to analyze the correlation between the lung function and CRP levels.@*METHODS@#Sputum and serum specimens were obtained from 30 COPD patients diagnosed according to the national criteria. The CRP level was measured by enzyme-linked immunosorbent assays. The lung function indexes such as the forced expiratory volume in one second (FEV1) and the forced expiratory volume in one second/forced vital capacity (FEV1/FVC) were also determined.@*RESULTS@#The sputum CRP level in COPD patients was 30-50 times higher than that of the health controls. The sputum CRP level in COPD patients is significantly higher than that of serum in these patients (P < 0.05). The correlation between changes of serum and sputum CRP levels and sputum CRP levels was positive (r = 0.625, P < 0.05). The correlations between changes of serum CRP levels and FEV1 were negative (r = -0.610, -0.725, respectively, P < 0.001). And the correlations between changes of serum and sputum CRP levels and FEV1/FVC were negative (r = -0.639, -0.600, respectively, P < 0.05).@*CONCLUSION@#The CRP may be secreted from the local respiratory tract. The damage of lung function in COPD patients is associated with the increase of CRP level.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the therapeutic effect of Xiaobai Mixture (XBM) in treating vitiligo.</p><p><b>METHODS</b>Seventy-four patients with vitiligo were randomly divided into the XBM group treated with XBM and the control group treated with 8-MOP. The therapeutic effect, nail-fold microcirculation, plasma endothelin-1, serum immunoglobulin were observed and compared.</p><p><b>RESULTS</b>The therapeutic effect of XBM was better than that of 8-MOP (P < 0.05). XBM could also obviously improve the nail-fold microcirculation, elevate the plasma endothelin-1 level and lower the serum IgG (P < 0.01).</p><p><b>CONCLUSION</b>XBM has superiority in treating vitiligo.</p>
