ABSTRACT

Objective To set up a living mice colonoscopy platform to establish an orthotopic model of colorectal cancer in mice under direct vision,and to observe its biological behavior such as metastasis.Methods Eighteen-week-old male C57/BL mice were anesthetized,and the intestinal lumen of the mice was examined by a self-developed living mice colonoscopy and Olympus URF-P5 ureteroscopy,respectively.The imaging effects of the two methods were compared.Human colon cancer HT-29 cells were injected into the colonic mucosa of BALB/c-nu mice under direct vision.The colonoscopy was performed on the 3rd,7th and 15th day after the injection to observe the tumor formation in the intestinal lumen.The mice were sacrificed when the body weight decreased significantly or cachexia appeared,and then the abdominal cavity was examined including the tumor formation and metastasis.Results The self-developed living mice colonoscopy platform can provide clear vision of enteric cavity,and no mice died in the colonoscopy examination.In vivo subcutaneous injection of HT-29 cells in mice was performed with a perforation rate of 15%,a mortality rate of 33.3%,a tumor formation rate of 62.5%,an abdominal metastasis rate of 60%,a liver metastasis rate of 25%,and an abdominal wall transfer rate of 25%.Conclusion The self-developed mice colonoscopy platform can be used for the study of colorectum in living mice.The imaging effect is no less than that of Olympus URF-P5 ureteroscopy.In addition,an orthotopic colorectal cancer model can be established by this platform combing with submucosal injection technology.

ABSTRACT

Objective@#To investigate the expression of syndecan-1 and syndecan-2 and their clinicopathological significance in patients with gallbladder squamous cell (SC)/adenosquamous carcinoma (ASC) and adenocarcinoma (AC).@*Methods@#A total of 126 patients with SC/ASC (n=46) and AC (n=80) were included in this study. The expression levels of syndecan-1 and syndecan-2 were detected by Envison™ immunohistochemistry assay. The clinical and prognostic significance of syndecan-1 and syndecan-2 were analyzed.@*Results@#In the 46 SC/ASC samples, syndecan-1 and syndecan-2 were positively expressed in 29 (63.0%) and 28 (60.9%) tumor tissues, respectively. (Positive expression was defined based on the staining in the component of squamous cell carcinoma. That is to say, the tissue which adenocarcinoma part was positively stained, but squamous cell carcinoma part was negatively stained is also regarded as negative.) In the 80 AC samples, 47 (58.8%) cases showed syndecan-1 positive expression, and 51 (63.8%) showed syndecan-2 positive expression. There was no significant difference in the positive rates of syndecan-1 and syndecan-2 between SC/ASC and AC groups (P>0.05 for all). The levels of syndecan-1 and syndecan-2 were associated with tumor size, TNM staging, lymph node metastasis, invasion of adjacent tissue, and surgical procedures in SC/ASC patients (P<0.05 for all). However, their expression was associated with tumor differentiation, tumor size, TNM staging, lymph node metastasis, invasion of adjacent tissue, and surgical procedures in AC patients (P<0.05 for all). The Kaplan-Meier survival analysis of SC/ASC and AC patients revealed that the average survival time for patients with positive syndecan-1 and syndecan-2 expression was significantly shorter than that of those with negative expression (P<0.01 for all). Cox multivariate analysis indicated that syndecan-1 and syndecan-2 expression were independent unfavorable prognostic factors for SC/ASC and AC patients (P<0.05 for all).@*Conclusion@#The syndecan-1 and syndecan-2 expression are associated with the tumor progression and poor prognosis in patients with gallbladder SC/ASC and AC.

ObjectiveTo study the expression of ABCG2, SFRP2, BRMS1 and HPA and detect their clinicopathologicalsignificancesinthebenignandmalignatntlesionsofthegallbladder.MethodsEnVisiom immunohistochemical method for determining the expressions of ABCG2, SFRP2,BRMS1 and HPA was used in paraffin-embedded sections of surgical resected specimens from gallbladder adenocarcinoma (n =108), peritumoral tissues (n =46 ), adenomatous polyp (n =15 ), and chronic cholecystitis ( n =35 ).ResultsThe positive rates of ABCG2 and HPA expression were significantly higher in gallbladder adenocarcinoma than that in peritumoral tissues, adenomatous polyp and chronic cholecystitis (P < 0. 05 or P < 0. 01 ) ; The positive rates of SFRP2 and BRMS1 expression were significantly lower in gallbladder adenocarcinoma than that in peritumoral tissues, adenomatous polyp and chronic cholecystitis(P <0. 05 or P <0. 01 ). The positive cases of ABCG2 and/or HPA as well as negative ones of SFRP2 and/or BRMS1in the benign lesions showed moderately-or severely-atypical hyperplasia of gallbladder epithelium. The frequency of samples with positive staining for ABCG2 and/or HPA in cases with small tumor volume (diameter < 2 cm), no lymph node metastasis, and no invasion into surrounding tissues was significantly lower than that in cases with larger tumor volume (diameter> 2 cm ), lymph node metastasis, and invasion into surrounding tissues ( P < 0.05 or P < 0. 01 ). However, compared with ABCG2 and/or HPA, the expression of SFRP2 and/or BRMS1 showed an opposite correlation in these cases ( P < 0. 05 or P <0. 01 ). Unitivariate Kaplan-Meier analysis showed that increased expressions of ABCG2 (P =0. 019) and HPA ( P =0. 016) or decreased expression of SFRP2 ( P =0. 019) and BRMS1 ( P =0. 008 )were associated with poorer overall survival, respectively. Multivariate Cox regression analysis showed that increased expression of ABCG2 (P =0. 018 ) and HPA ( P =0. 019) and/or decreased expression of SFRP2 (P =0. 015 ) and BRMS1 ( P =0. 011 ) were independently poor-prognostic predictors in gallbladder adenocarcinoma.ConclusionsThe expression of ABCG2, SFRP2, BRMS1 or HPA might be closely related to the carcinogenesis, clinical biological behaviors, and prognosis of gallbladder adenocarcinoma.

Objective To study the expressive levels of galectin-3 (gal-3) and sambucus nigra agglutinin(SNA) and to detect their clinicopathological significances in benign and malignant lesions of the gallbladder. Methods EnVisonTM Immunohistochemistry for assaying gal-3 expressive levels and ABC cytochemistry for determining SNA expressive levels were used in conventional paraffin-embedded sections from the specimens of adenocarcinoma (n = 108) , peritumoral tissues (n=46) . Adenomatous polyp (n=15), and chronic cholecystitis (n = 35). Results The positive rates of gal-3 and SNA were significantly higher in adenocarcinoma (62. 0%, 66. 7%) than in peritumoral tissues (39. 1%,45.6%), adenomatous polyp (26.7%, 33.3%) and chronic cholecystitis (11.4%, 11.4%)(P<0. 05 or P<0. 01). The positive gal-3 and SNA in benign cases showed atypical hyperplasia of the epthelium. The positive rates of gal-3 and SNA expression were significantly lower in well-differentiated adenocarcioma, mass with a maximal diameter of <2 cm, absence of lymph node metastasis,and absence of invasion to adjacent tissues than in poor-differentiated adenocarcinoma, mass with a maximal diameter of ≥2 cm, lymph node metastasis and invasion to adjacent tissues. (P<0. 05 or P<0. 01). There was a significant correlation between the expressive levels of gal-3 and SNA in gallbladder adenocarcinoma (x2=9. 51, P<0. 01). Conclusions The expressive levels of gal-3 and SNA lectins had important effects on carcinogenesis, progression and biologic behaviors of gallbladder cancer. Patients with positive gal-3 and /or SNA expressions had poor prognosis.

ABSTRACT

Objective To study the expression and clinicopathological significance of Paxillin and CAⅨ in the benign and malignant lesions of gallbladder.Methods The surgical resected specimens of 108 cases of gallbladder adenocarcinoma, 46 cases of peritumoral tissue, 15 cases of adenomatous polyp and 35 cases of chronic cholecystitis were routinely made paraffin embedded sections.The expressions of Paxillin and CAⅨ were stained with Envision immunohistochemistry.Results The positive rates of Paxillin and CAⅨ expression was significantly higher in gallbladder adenocarcinoma (60.2% and 49.1%) than those in peritumoral tissues (26.1%, x2 =15.00, P <0.01 and 23.9%,x2=8.41,P <0.01), adenomatous polyp (20.0%,x2=8.60,P<0.01 and 20.0%,x2 =4.49,P<0.05) and chronic cholecystitis(14.3% ,x2 =22.89, P<0.01 and 11.4%,x2 =15.63,P <0.01).All the gallbladder epithelia of the benign cases with Paxillin and CA Ⅸ positive expression showed moderate to severe atypical hyperplasia.The positive expression rates of Paxillin and CA Ⅸ were significanctly lower in the cases of well-differentiated, maximal diameter of mass less than 2 cm, no lymph nodes metastasis and no surrounding tissues invasion than those of the cases with poorly differentiated, maximal diameter of mass over 2 cm, lymph nodes metastasis and surrounding tissues invasion.With Kaplan-Meier analysis, it suggested that the survival period after the surgery in Paxillin and CAⅨ positive expression cases was shorter than that of negative expression cases (x2 = 5.65,P<0.05,5.65=5.92, P<0.01).With multivariate Cox regression analysis, it indicated that both Paxillin and CAⅨ positive expression was an important indicator of the poor prognosis in gallbladder adenocarcinoma.Conclusion The expression of Paxillin and CA Ⅸ may be closely related to the carcinogenesis, tumor biological behaviors, and prognosis of gallbladder adenocarcinoma.The positive expression of Paxillin and/or CAⅨ is associated with poor prognosis.

ABSTRACT

Objective To establish a model of pancreatic cancer induced by 7,12-dimethylbenzathracene (DMBA) in SD rats, and to detect the expression levels of RAD51 and Myc-associated factor X (MAX) and their effect on carcinogenesis of rat pancreas. Methods Ninety SD rats were randomly divided into 3 groups: a model group, an intervention group, and a control group. DMBA was directly implanted into the parenchyma of rat pancreas (the model group and the intervention group). Rats in the intervention group were treated with 1 mL trichostatin A (TSA) saline solution (1 μg/mL) via ip weekly. Rats within 3~5 months in the model group and the intervention group were executed and observed by macrograph and under microscope. Meanwhile, the rats in the control group were executed at 5th month. The EnVision~(TM) immunohistochemistry to assay the expression levels of RAD51 and MAX was used in conventional paraffin-embedded sections from the above pancreatic specimens.Results The incidence of pancreatic cancer in the model group within 3-5 months was 48.7% (18/37), including 17 ductal adenocarcinomas and 1 fibrosarcoma. The incidence of pancreatic cancer in the intervention group within 3-5 months was 33.3%(12/36), including 11 ductal adenocarcinomas and 1 fibrosarcoma. The maximal diameter of mass in the model group was significantly higher than that in the intervention group (P<0.05). No pathological changes were found in pancreas of the control group and other extra-pancreatic main organs of the model group and the intervention group (such as the liver, biliary tract, gastrointestine tract, kidney, and lung). The positive rate of RAD51 was significantly higher in ductal adenocarcinoma in the model group, the intervention group, and the model group +the intervention group than those in corresponding groups of non-cancerous pancreatic tissues (P<0.01), but the positive rate of MAX expression was opposite to RAD51 expression(P<0.01). The positive tissues of RAD51 expression and/or negative tissues of MAX expression in non-cancerous tissues showed atypical-hyperplasia of ductal epitheli. Pacncreas of the control group showed the negative expression of RAD51 and positive expression of MAX. Two cases of fibrosarcoma showed the negative expression of RAD51 and MAX.Conclusion DMBA directly implanted into the parenchyma of pancreas can obtain an ideal pancreatic cancer model with high incidence in a short time. The TSA might have an inhibitive effect on carcinogenesis and growth of rat pancreas. The over-expression of RAD51 and/or lose-expression might have important effect on carcinogenesis induced DMBA in rat pancreas.

ABSTRACT

Objective To study the expression of nuclear factor-kappaB (NF-κB) and the counts of lymph vessels in laryngeal squarnous cell carcinoma and polyps of vocal cord tissues, and explore their clinicopathologic significance and correlation in the course of laryngeal carcinoma. Methods SP immuno-histochemical method was used to detect the expression of NF-κB and the counts of lymph vessels on the routinely paraffln-embedded sections of the specimens from 50 cases laryngeal squamous cell carcinoma and 10 cases of polyps of vocal cord tissues. Results The positive rate of NF-κB and the counts of lymph ves-sels in laryngeal carcinoma[60. 0% ,( 13.3±3.4)/HP]were significantly higher ( P <0. 05 and P <0. 01respectively) than those in polyps of vocal cord tissues[10.0 % ,(6. 1±3. 8)/HP]. The positive rate of NF-κB and the counts of lymph vessels in well differentiated adenocarcinoma and cases without metastasis were significantly lower( P < 0. 05, P <0. 01 ), compared with poor-differentiated adenoearcinoma and ca-ses with metastasis. The counts of lymph vessels in the NF-κB positive cases were significantly higher than thoseinNF-κBnegativecases[(14.9±4.1)/HPvs (9.8±3.1)/ HP, P <0.01] . Conclusions The expression of NF-κB and the counts of lymph vessels might be important markers to be used to monitor the progression, biological behaviors, metastatic status and prognosis of laryngeal carcinoma. NF-κB might pro-mote lympoangiogenesis in laryngeal squnmous cell carcinoma tissues.

OBJECTIVE@#To detect the expression of galectin-3 (gal-3) and Sambucus nigra agglutinin (SNA) and determine their clinicopathological significance in breast cancers and benign breast lesions.@*METHODS@#Envison immunohistochemistry for staining gal-3 expression, and ABC affinity-cytochemistry to detect SNA expression were used in paraffin-embedded slides from specimens of breast cancers (n=60) and benign lesions (n=30).@*RESULTS@#The positive rates and scoring means of gal-3 and SNA were significantly higher in breast cancer (48.3%, 2.07 +/- 2.25, 2.12 +/- 2.26) than those in benign lesions (26.7%, 1.03 +/- 1.63, 1.07 +/- 1.59, P < 0.05). The scoring means of gal-3 and SNA expression were significantly lower in the positive cases of estrogen receptor (ER) and the negative ones of CA15-3 than those in the negative cases and the positive ones (P < 0.05).The survival analysis of Kaplan-Meier showed the 5-year survival rate and mean survival period were significantly lower in the gal-3 or SNA expression positive cases than those in the negative cases of breast cancer (P<0.01).@*CONCLUSION@#The expressive level of gal-3 and SNA lectins might have important effect on the carcinogenesis, progression and biologic behaviors of breast cancer. The positive cases of gal-3 and /or SNA expression might have poor prognosis.

OBJECTIVE@#To determine the expressive level of checkpoint kinase 1 (CHK1) and polo-like kinase 1 (PLK1) and to detect their clinicopathological significance in benign and malignant lesions of the stomach.@*METHODS@#Envision Tm immunohistochemistry was used to detect the expression level of CHK1 and PLK1 in conventional paraffin-embedded sections from specimens of primary foci (n=59)and metastatic foci of lymph node (n=42) of gastric cancer, peritumoral tissues (n=20), and benign lesions of the stomach (n=95).@*RESULTS@#The positive rates of CHK1 were significantly higher in gastric cancer than that in different types of benign lesions(P0.05). The positive rates of CHK1 and PLK1 were significantly lower in the non-metastatic lymph node than that in the metastatic lymph node (P<0.05). The positive rate of CHK1 was significantly lower in histologic grade II than that in the histologic grade III+IV (P<0.05). Positive correlation was found between the expression of CHK1 and PLK1 in gastric cancer tissues (P<0.01).@*CONCLUSION@#The expression level of CHK1 and /or PLK1 might be important biological markers of kinases to reflect the carcinogenesis, progression, biological behaviors, and guide clinical auxiliary treatment of gastric cancer.

ABSTRACT

Objective:To study the expression levels of tumor-associated glycoprotein (TAG72) and glucose-regulated protein 94 (GRP94) in the breast benign and malignant lesions and detect their clinicopathological significance. Methods:EnVisionTM immunohistochemistry was used for assaying expressive levels of TAG72 and GRP94 in conventional paraffin-embedded sections from specimens of breast cancer (n=60) and benign lesions (n=30). The survival analysis was performed by using Kaplan-Meier method.Results:The positive rates and scores of TAG72 and GRP94 were significantly higher in breast cancer than those in benign lesions (TAG72: 68.3% vs 10.0%, 2.53±1.86 vs 0.37±1.13; GRP94: 63.3% vs 13.3%, 2.73±2.23 vs 0.47±1.22; P=0.000). The positive rates and scores of TAG72 and GRP94 were significantly lower in the intra-ductal,histological grade Ⅰ or Ⅱ , with maximal diameter ≤3 cm, without lymph node metastasis, estrogen receptor (ER)-positive, p53 and CA15-3-negative breast cancer than those in the ductal invasive (P3 cm (P<0.05), with lymph node metastasis (P<0.01), ER-negative (P<0.05) , p53-positive (P<0.01), and CA15-3-positive (P<0.01) breast cancer tissues. Kaplan-Meier survival analysis showed that the median survival time were significantly shorter in the TAG72-positive, GRP94-positive or TAG72 and GRP94-positive breast cancer patients than those with negative TAG72 and/or GRP94 expression (P<0.01). Multivariate COX regression analysis showed that TAG72 and/or GRP94-positive expression were negatively associated with postoperative survival rate and positively related with death rate. Both of them were independent prognostic factors for breast cancer. Conclusion:The expressive levels of TAG72 and/or GRP94 might have important effects on the carcinogenesis, progression, biologic behaviors and prognosis of breast cancer. The breast cancer patients with over-expression of TAG72 and/or GRP94 might have poor prognosis.

ABSTRACT

Objective To investigate the relationship between the expression of regulated-upon activation normal T expressed and secreted(RANTES)and microvascular density(MVD)in hepatocarcinoma(PHC)tissues,and explore its clinical significance. Methods The samples of PHC tissues and adjacent tissues in 47 patients were collected.The expression of RANTES in tissues was detected by immunohistochemistry,and MVD was calculated.The relationship among the expression of RANTES,MVD and clinicopathological features was analysed. Results The positive expression rate and expression score of RANTES in PHC tissues were significantly higher than those in adjacent tissues(55.32% vs 19.15%,P<0.01;1.89±1.77 vs 0.77±1.29,P<0.01).MVD in PHC tissues was significantly higher than that in adjacent tissues(67.30±13.68 vs 37.20±10.58,P<0.01).MVD of PHC tissues in patients with metastasis was significantly higher than that in patients without metastasis[(73.50±13.77)/HP vs (64.10±12.68)/HP,P<0.05],while there were no correlations among MVD,expression of RANTES and the other clinicopathological features of PHC.MVD was positively correlated with the expression score of RANTES in PHC tissues(r=0.386,P<0.05). Conclusion RANTES might be closely related to the angiogenesis in PHC.

ABSTRACT

OBJECTIVE@#To investigate the expression of the matrix metalloproteinase-1, -9 (MMP-1,9), and tissue inhibitor of metalloproteinase-4 (TIMP-4), extracellular matrix metalloproteinase inducer (EMMPRIN) in the myocardium of congestive heart failure in patients with rheumatic heart diseases.@*METHODS@#The papillary muscle specimens of the left ventricle were obtained from 18 patients with heart failure during rheumatic heart valve replacement, and the normal specimens were obtained from the autopsy of 10 adults without heart disease. The specimens were stained to examine the expression of MMP-1, MMP-9, TIMP-4, and EMMPRIN by the EnVision immunohistochemical assays.@*RESULTS@#The expressions of MMP-1, MMP-9, and EMMPRIN in the myocardium of the patients with the cardiac function classes III and IV (NYHA III and IV) were significantly higher than those the normal cardiac function (NYHA I) (P<0.05) except MMP-1 expression between NYHA III and I. The TIMP-4 level was significantly lower in patients with NYHA III and IV than that of the NYHA I ( P<0.05). The expression of MMP-1, MMP-9, and EMMPRIN was significantly higher in patients with atrial fibrillation than those in the control group with regular sinus rhythm (P<0.05), whereas the TIMP-4 level was obviously lower in patients with atrial fibrillation than that in the control group with regular sinus rhythm (P<0.05). The expressions of MMP-1, MMP-9, and EMMPRIN were positively correlated with each other, but were negatively correlated with TIMP-4.@*CONCLUSION@#MMPs, TIMP, and EMMPRIN were significantly unbalanced in the myocardium of congestive heart failure patients with rheumatic heart diseases. They may play an important role in congestive heart failure through myocardial remodeling.

Objective To establish a pancreatic cancer model in Sprague Dawley(SD) rats,and to study the distribution and the counts of myofibroblast(MF) in pancreatic cancer and non-cancerous pancreatic tissues.MethodsDirectly implanted DMBA into pancreas parenchyma of SD rats and established TSA intervening group and control group. The carcinogenesis of rats executed within 3~5 months were inspected by HE stain and microscopy for pathomorphological changes.Myofibroblast(MF) was stained by Heidenains method. Objective To investigate therapeutic effect and mechanism of hyperbaric oxygen and ulinastatin respectively or combinatively used to treat acute necrotizing pancreatitis (ANP). Methods One hundred and twenty SD rats were divided into 6 groups randomly: group of normal control, group receiving sham operation, group of untreated acute necrotizing pancreatitis (ANP group), group of acute necrotizing pancreatitis treated with hyperbaric oxygen (HBO group), group of acute necrotizing pancreatitis treated with ulinastatin (ULT group), and group of acute necrotizing pancreatitis treated with combined hyperbaric oxygen and ulinastatin (HBO+ULT group). The rat model of acute necrotizing pancreatitis was established according to Aho HJ et al. Objective To study the expression of hMLH1, hMSH2 and their association with clinico-pathological characteristics in hepatocellular carcinoma(HCC). Methods The expression of hMLH1 and hMSH2 in 37 HCC tissues was detected by SABC immunohistochemistry.
