ABSTRACT
Age estimation based on tissues or body fluids is an important task in forensic science. The changes of DNA methylation status with age have certain rules, which can be used to estimate the age of the individuals. Therefore, it is of great significance to discover specific DNA methylation sites and develop new age estimation models. At present, statistical models for age estimation have been developed based on the rule that DNA methylation status changes with age. The commonly used models include multiple linear regression model, multiple quantile regression model, support vector machine model, artificial neural network model, random forest model, etc. In addition, there are many factors that affect the level of DNA methylation, such as the tissue specificity of methylation. This paper reviews these modeling methods and influencing factors for age estimation based on DNA methylation, with a view to provide reference for the establishment of age estimation models.

Objective To evaluate the interactive effects of fine particulate matter and temperature on non-accidental mortality of residents in Pudong, Shanghai. Methods Daily mortality, air pollutants and meteorological data from Jan 1st.2016 to Dec 31st.2017 were collected.Generalized additive Poisson regression models was used to estimate the effects of PM2.5 pollution on daily mortality, bivariate response surface models and temperature stratified models were applied to examine the interaction of temperature with PM2.5 on mortality. Results A total of 43 345 non-accidental deaths were included, daily mean PM2.5 concentration was 39.1 μg/m3, daily mean temperature was 17.7 ℃.A 10 μg/m3 increase in the daily PM2.5 at lag 1 day was associated with a 0.56%(95%CI:0.11%-1.01%), 0.49%(95%CI:-0.19%-1.18%) and 0.22%(95%CI:-1.14%-1.60%) increase in non-accidental, cardiovascular and respiratory mortality, respectively.Higher risks were identified for males and the older (≥65 years).The effect estimates per 10 μg/m3 increase in PM2.5 in medium temperature level were 0.59%(95%CI:0.04%-1.14%)for non-accidental, 0.51%(95%CI:-0.32%-1.35%)for cardiovascular and 0.51%(95%CI:-0.32%-1.35%) for respiratory mortalities.The PM2.5 effects were approximately 2-4 times higher in higher temperature level for non-accidental and cardiovascular mortalities compared with other temperature levels; for respiratory mortality, the PM2.5 effects was approximately 2-fold higher in lower temperature levels than the medium, although the interactions between temperature and PM2.5 were statistically insignificant. Conclusions Temperature may modify the effects of PM2.5 on mortality in Pudong, Shanghai and the interactive pattern may be different across disease-specific mortalities.

OBJECTIVES@#To collect single piece of dandruff with microscopes to improve the regular EZ-tape method for DNA extraction and genotyping, increase the utilization of samples, reduce the miss rate as well as the proportion of genotyping results of mixed stains.@*METHODS@#The insides of the hats worn by two volunteers were stuck by EZ-tape and scotch tape respectively. DNA on EZ-tape was directly extracted using traditional method. Single piece of dandruff was collected from the scotch tapes under microscope. The two kinds of methods were both performed under continuous oscillation and standing digestion, respectively. DNA was extracted through Chelex-100 method, and STR genotypes were obtained after amplification and electrophoresis. The results of STR genotypes obtained by EZ-tape method and single piece of dandruff analytical method were compared.@*RESULTS@#Miss detections happened in 11 samples (45.8%) by EZ-tape method and only single-source typing results were obtained. Ten samples (41.7%) showed the genotype results of mixed stain and six of which showed allele insertions and deletions. The genotype results were obtained successfully using the single piece of dandruff analytical method, and two samples showed mixed stain genotype. The number of exact typing processed by oscillation was higher than that by standing digestion ( P<0.05).@*CONCLUSIONS@#The oscillation during the DNA extraction process is in favour of the DNA releasing. Single piece of dandruff analytical method can be used to obtain single-source STR genotype with high successful ratio and low miss rate. This method can be a collection method of special samples such as dandruff in forensic practice.

Objective: To analyze the characteristics of the healthy state of TCM (Traditional Chinese Medicine) of female, and preliminarily constructs the model of TCM health status identification of female with menstrual cycle. Methods: The text of disease diagnosis and syndrome differentiation of menstrual-cycle-linked disorders from Guidelines for diagnosis and treatment of common disease of gynecology in Traditional Chinese Medicine was analyzed, and the symptoms related with viscera functions, the waxing and waning of qi and blood, and pathogenic properties were extracted. The questionnaires of diseases and syndrome elements differentiation of female by calculating the weight relationship between symptoms and health status were designed to estimate the interior state of waxing and waning of yin-yang by figuring out the different periods of menstrual, and in the last step, Constitution in Chinese medicine Questionnaire was used to identify the constitution. Through the above four identification methods, a comprehensive identification of the female's health status of TCM was formed. Results: Through the disease identification model, we distinguished the state of disease and yet disease. And through the syndrome elements differentiation model, we knew the viscera functions, the waxing and waning of qi-blood and yin-yang, and pathogenic properties. And by identifying the internal qi-blood and yin-yang status, we initially built the model that can identify health status of female integrally with the constitution. Conclusion:The model identification results can reflect the comprehensive health status of female in multiple dimensions. It can make a fine classification of women.s TCM health status. It can not only help to know the previous health status but also help to speculate the future health trend, which contributes to the formulation of personalized health interventions of follow-up.

Objective: To construct the evaluation index of follow-up assessments on TCM (traditional Chinese medicine) health management of female. Methods: The evaluation indicators and score criteria were extracted for the efficacy evaluation of diseases and syndromes related to irregular menstruation in The criteria of diagnosis and therapeutic effect of diseases and syndromes in Traditional Chinese Medicine and Guidelines of clinical research of new drugs of Traditional Chinese Medicine; The health behaviors related to the onset of irregular menstruation diseases, such as diet, emotion, exercise, etc. were collected to build index of health behavior of irregular menstruation. Results: A comprehensive index system of follow-up assessments on TCM health management of female composed of disease curative effect, syndrome curative effect and health behavior score was constructed. Conclusion: The evaluation index system helps to dynamically track the effectiveness of interventions and management, and helps users to adjust the intervention plan and avoid the unhealthy behaviors.

Objective: To explore pulse characteristic of the health female college students and their relationship with menstrual cycle. Methods: The subjects were 17 female college students of Jiangxi University of Traditional Chinese Medicine. Pulse test was performed with pulse bracelet. The detection time was every day in the wake and has not yet get up and wake up at any time before going to bed with a pulse-taking bracelet, and recorded the current menstrual cycle information. The pulses of the wake that has not yet get up and wake up at any time before going to bed took consistency analysis of the record with Kruskal Wallis Test. The pulse of menstruation stage analysis was compared with chi-square test. Results: The pulses of the wake that has not yet get up and wake up at any time before going to bed consistency was poor except rapid pulse (P> 0.05) . The occurrence frequency of slippery pulse and string-like pulse were 55.21% and26. 03% respectively. The pulses of premenstrual period, post menstrual period and menstrual period were not much different from others. The occurrence frequency of moderate pulse, string-like pulse, unsmooth pulse, slippery pulse in intermenstrual period respectively were 40.68%, 44.07%, 30.51%, and 40.68%. The occurrence frequency of moderate pulse, string-like pulse, unsmooth pulse, slippery pulse in-non intermenstrual period respectively were 17.38%, 24.69%, 10.45%, 56.30%. The difference was significant (P < 0.05) . Conclusion: The female college students are mainly in terms of slippery pulse. The occurrence frequency slippery pulse in intermenstrual period respectively are less common. The string-like pulse, moderate pulse, unsmooth pulse in intermenstrual period are more common than in-non intermenstrual period.

Objective: To fulfill the TCM (traditional Chinese medicine) health management needs of female through constructing the intervention knowledge base, explore the long-term development mechanism of intervention knowledge base. Methods: The structure of female TCM Health Intervention Knowledge was designed base according to the characteristics of TCM health status of females. The inferential mechanism by studying the relation between the female health status and the diet therapy scheme was explored based on syndrome differentiation. A Long-term development mechanism of TCM health intervention knowledge database was designed based on big data and effect feedback mechanism. Results: The classification standard of the TCM Health Intervention Knowledge base of female includes the state of the viscera functions, qi and blood, yin and yang, and pathogenic properties of the body under the illness and health condition. The dietary therapy formula inference engine can provide food material according to the region, natures, function of tonifying, evil eliminating, to match the female health status, and constantly adjusting and improving the Intervention plan according to the feedback. Conclusion: The study proves the construction plan of TCM Health Intervention Knowledge base of female is able to meet the needs of female TCM health management intervention, and the method of constantly adjusting and improving the intervention plan according to the feedback helps to realize the precise recommendation and long-term development of intervention knowledge base.

Objective: To construct a female TCM management platform that integrates pulse diagnosis information to facilitate women's health status identification and conditioning of menstrual cycle related health problems. Methods:Using cloud computing technology, taking the EHR and knowledge base as the core, designing the TCM health information collection questionnaire according to the characteristics of women.s menstrual cycle related health problems, and incorporating the data collected by the Chinese medicine pulse diagnosis bracelet, the knowledge base can assist the development of a health management plan, designing a platform framework for combining the PC side of the doctor with the patient mobile APP. Results: The platform was conducive to the realization of more objective TCM consultation and pulse diagnosis information collection and long-term continuous monitoring, and can assist in the identification of women's menstrual problems and the development of health conditioning plans. Conclusion: The platform has good operability and practicability for the implementation of TCM health management for women with menstrual problems.

Now it has become the leading cause of chronic liver diseases worldwide, which is closely related to the high incidence of type 2 diabetes and arteriosclerotic cardiovascular disease. It can progress into cirrhosis and hepatocellular carcinoma. The pathogenesis of non-alcoholic fatty liver disease is complicated and not yet fully elucidated. In recent years, studies have found that autophagy plays an important role in the development of non-alcoholic fatty liver disease, mainly through regulation of insulin resistance, endoplasmic reticulum stress, mitochondrial dysregulation, lipid-toxicity caused by lipid deposition and inflammation.

AIM:To investigate the effect of Fufang Zhenzhu Tiaozhi capsule(FTZ)on serum lipids and in-flammatory factors in rabbits with abdorminal aortic restenosis after balloon angioplasty.METHODS: New Zealand white rabbits(n=30)were divided into 5 groups.Except blank control group,the rabbits in other groups were used to establish abdominal aortic endothelium exfoliative vascular stenosis model.After 4 weeks of high-fat diet feeding,the animals in rest-enosis model group and drug treatment groups underwent percutaneous balloon dilatation in the stenosis.The angiographic stenosis was analyzed by a two-dimensional quantitative coronary angiography workstation with a digital subtraction X -ray machine.Blood samples were taken during angiography and the profiles of serum lipids and cytokines were measured.The expression of nuclear factor-κB(NF-κB)in the blood vessels was determined by immunohistochemistry.RESULTS:An-giography confirmed that the rates of area stenosis and diameter stenosis were significantly decreased in treatment groups compared with restenosis model group(P<0.01).Compared with restenosis model group,the serum lipid profiles and cy-tokine concentrations in drug treatment groups were significantly decreased(P<0.05).Immunohistochemistry showed the expression of NF-κB in restenosis model group was significantly higher than that in blank control group and drug treatment groups(P<0.05).CONCLUSION: FTZ significantly reduces the blood lipids and inflammatory factors in abdominal aortic restenosis model,and the anti-inflammatory effect may be related to the regulation of NF-κB pathway to inhibit the production of various inflammatory factors.

<p><b>OBJECTIVE</b>To assess the impact of delayed decompression on long-term neurological and bladder function recovery in patients with cauda equina syndrome (CES) secondary to lumbar disc herniation (LDH).</p><p><b>METHODS</b>The clinical data of 35 patients receiving delayed decompression surgery for CES secondary to LDH were reviewed. The bladder empty function, bowel control, sexual ability and neurological functions of the lower limbs were evaluated after the operation, and the urodynamic changes were assessed in 6 patients with urodynamic data before and after the operation.</p><p><b>RESULTS</b>Surgical decompression was performed at 4.1∓3.9 weeks in 12 patients with complete CES and at 5.5∓7.6 weeks in 23 patients with incomplete CES after the onset of symptoms. The patients were followed up for a mean of 43.0∓28.9 months (3-110 months). In the 23 patients with incomplete CES, 19 obtained full recovery, 4 had slight sensory alterations in the saddle area or the lower limbs. In the 12 patients with complete CES, 2 had full recovery, 4 reported slight sensory alterations in the saddle area or the lower limbs (including 2 with occasional constipation); 6 still had sense deficit in the saddle area and difficulties in bladder or bowl emptying, but they all reported significant improvements compared to the condition before operation. Urodynamic analysis in the 6 patients with pre- and postoperative urodynamic data showed increased abdominal pressure when voiding with significantly reduced residual urine in all the 6 patients; 4 patients with abnormal first desire volume before operation reported recovery after the operation.</p><p><b>CONCLUSION</b>Patients with LDH-induced CES who missed the chance of early decompression can still expect favorable functional recovery in the long term. The improvement of bladder function following decompression is probably a result of recovery of bladder sensation and the compensation by increased intra-abdominal pressure. The key strategy to promote bladder function recovery in these patients is to promote the detrusor recovery.</p>

OBJECTIVE:To establish a method for the determination of drug loading and encapsulation efficiency of Epirubi-cin hydrochloride-sorafenib-loaded Polylactic Acid-glycolic Acid Polymer(PLGA)embolic microspheres. METHODS:HPLC meth-od was adopted to determine the contents of epirubicin hydrochloride and sorafenib in the preparation,and then drug loading and encapsulation efficiency were calculated by formula. The determination was performed on Phenomenex Luna 5u C8(2) 100A col-umn with mobile phase consisted of methanol-water(containing 0.05% trifluoroacetic acid and 0.14% dium dodecyl sulfate)(75:25,V/V)at the flow rate of 1.0 mL/min. The detection wavelength was set at 252 nm,and the column temperature was maintained at 25℃. The injection volume was 10μL. RESULTS:The linear ranges were 2.020-101.00μg/mL for epirubicin hydrochloride(r=0.9998)and 2.048-102.40 μg/mL for sorafenib(r=0.9997),respectively. The limits of quantification were 3.2970,2.5468 μg/mL, respectively. The detection limits were 0.9891,0.7641 μg/mL,respectively. RSDs of precision,stability and repeatability tests were all less than 2.0%. The recoveries were 96.41%-101.80%(RSD=1.64%,n=9),99.46%-101.45%(RSD=0.70%,n=9),re-spectively. Drug loading of two components in 3 batches of samples were no lower than 1.17%,encapsulation efficiency no lower than 58%. CONCLUSIONS:The method is simple,accurate,can be used to determine drug loading and encapsulation efficiency of Epirubicin hydrochloride-sorafenib PLGA embolic microspheres.

Objective:To explore the application value of health education in patients with coronary heart disease(CHD) complicated diabetes mellitus(DM).Methods:A total of 72 CHD+DM patients were selected from our hospital.According to order of archives,they were divided into routine education group(n=36)and CLT group(n=36,received CLT-based health education intervention based on routine health education).Both groups were followed up after three-month interven-tion.Levels of fasting blood glucose(FBG)and 2h postprandial glucose(2hPG),scores of self-efficacy and quality of life (QOL)were measured and compared between two groups before and after intervention.Results:Compared with before in-tervention,after intervention,there were significant reductions in levels of FBG and 2hPG,and significant rise in scores of self-efficacy and QOL in two groups,P=0.001 all.Compared with routine education group after intervention,there were significant reductions in levels of FBG[(7.74 ± 0.81)mmol/L vs.(5.94 ± 0.46)mmol/L]and 2hPG[(9.16 ± 0.89)mmol/L vs.(7.41 ± 0.40)mmol/L],and significant rise in scores of self-efficacy[(46.56 ± 3.71)scores vs.(58.67 ± 4.58) scores]and QOL[(63.88 ± 4.86)scores vs.(76.55 ± 5.22)scores]in CLT group,P=0.001 all.Conclusion:Health educa-tion can effectively reduce blood glucos level,improve self-efficacy and quality of life in patients with coronary heart disease complicated diabetes mellitus,which is worth extending.

OBJECTIVE To investigate the developmental toxicity of a new disinfectant trichloroiso-cyanuric acid (TCCA) on zebrafish embryos and larvae. METHODS Zebrafish embryos of 2 h post fertilization (hpf) were exposed to TCCA:①The embryos were exposed to a culture medium containing TCCA 0, 50, 100, 150, 200, 250 and 300 mg·L-1 for 48 h before 50%lethal concentration (LC50) was calcu-lated. ② The embryos were exposed to a culture medium containing TCCA 0, 10.4, 20.8 and 41.7 mg · L-1 for 96 h. The mortality, malformation rate and heart rate of embryos and larvae were measured at 48, 72 and 96 h after TCCA exposure. The expression of superoxide dismutase (SOD) was detected at 2, 4 and 6 h after TCCA exposure. The pathological changes in the head of zebrafish larvae were observed 7 d after exposure by HE staining. RESULTS LC50 of 48 h after TCCA exposure was 166.9 mg · L-1. Compared with normal control group, the mortality and malformation rate of zebrafish embryos were significantly increased (P<0.05) in TCCA 41.7 mg · L-1 group, but the heart rate was decreased significantly (P<0.05) in each dose of TCCA group at 96 h after TCCA exposure. At 72 h after TCCA exposure, the mortality rate of zebrafish embryos was significantly increased (P<0.05) in TCCA 41.7 mg·L-1 group, the malformation rate of zebrafish embryos was increased (P<0.05) and the heart rate of zebrafish embryos was decreased (P<0.05) in 20.8 and 41.7 mg · L-1 groups. At 48 h after TCCA exposure, the heart rate of zebrafish embryos was decreased significantly (P<0.05) in 41.7 mg · L-1 group. The SOD activity of zebrafish embryos in 20.8 and 41.7 mg · L-1 groups was significantly lower than that of control group (P<0.05). At 7 d after exposure, the brain and ocular space of larvae were enlarged and the ocular retina layers were not obvious in any dose of TCCA groups. CONCLUSION TCCA has toxic effect on zebrafish embryonic development, which can down-regulate SOD activity in the early developmental stage of embryos and damage the retina tissue of larvae.

AIM To establish an HPLC-ELSD method for the simultaneous content determination of euscaphic acid,tormentic acid,crategolic acid,corosolic acid,platycodin E,platycodin D3 and platycodin D in Chuanbei Luohan Zhike Granules (Fritillariae cirrhosae Bulbus,Eriobotryae Folium,Platycodonis Radix,etc.).METHODS The analysis of 60% methanol extract of this drug was performed on a 30 ℃ thermostatic Ultimate XB C18 column (4.6 mm ×250 mm,5 μm),with the mobile phase comprising of acetonitrile-0.5% ammonium acetate flowing at 0.6 mL/min in a gradient elution manner.RESULTS Seven constituents showed good linear relationships within their own ranges (r > 0.999 0),whose average recoveries were 96.99%-99.78% with the RSDs of 0.72%-1.72%.CONCLUSION This accurate,simple,specific and reproducible method can be used for the quality control of Chuanbei Luohan Zhike Granules.

<p><b>OBJECTIVE</b>To investigate the expressions of inflammation- and fibrosis-related genes in perinephric and subcutaneous adipose tissues in patients with adrenocorticotropic hormone (ACTH)-independent Cushing's syndrome.</p><p><b>METHODS</b>The perinephric and subcutaneous adipose tissues adipose tissues were obtained from 8 patients with ACTH-independent Cushing's syndrome undergoing laparoscopic retroperitoneal adrenalectomy. Real-time PCR was used to detect the mRNA expression levels of interleukin 6 (IL-6), tumor necrosis factor-α (TNF-α), matrix metallopeptidase 2 (MMP-2), TIMP metallopeptidase inhibitor 1 (TIMP-1), early growth response 1 (EGR1), CCAAT/enhancer binding protein β(CEBPβ), uncoupling protein 1(UCP-1), PPARγ coactivator 1 alpha (PGC1α) and cell death-inducing DFFA-like effector a (CIDEA).</p><p><b>RESULTS</b>The mRNA level of CIDEA was significantly higher in the perinephric adipose tissue (peri-N) than in the subcutaneous adipose tissue (subQ) (P<0.05). The expressions of CEBPβ, UCP-1, and PGC1α mRNA in the peri-N were similar with those in the subQ. The expressions of IL-6, TIMP1 and EGR1 mRNA in the subQ were significantly higher than those in the peri-N (P<0.05). No significant difference in TNF-α and MMP-2 mRNA levels was found between peri-N and subQ.</p><p><b>CONCLUSION</b>The expression levels of the inflammation- and fibrosis-related genes are higher in the subQ than in the peri-N of patients with ACTH-independent Cushing's syndrome, suggesting that chronic exposure to endogenous hypercortisolism may cause adipose tissue dysfunction.</p>

<p><b>OBJECTIVE</b>To determine the equilibrium solubility of pulchinenosiden D in different solvents and its n-octanol/water partition coefficients.</p><p><b>METHOD</b>Combining shaking flask method and high performance liquid chromatography (HPLC) to detect the n-octanol/water partition coefficients of pulchinenosiden D, the equilibrium solubility of pulchinenosiden D in six organic solvents and different pH buffer solution were determined by HPLC analysis.</p><p><b>RESULT</b>n-Octanol/water partition coefficients of pulchinenosiden D in different pH were greater than zero, the equilibrium solubility of pulchinenosiden D was increased with increase the pH of the buffer solution. The maximum equilibrium solubility of pulchinenosiden D was 255.89 g x L(-1) in methanol, and minimum equilibrium solubility of pulchinenosiden D was 0.20 g x L(-1) in acetonitrile.</p><p><b>CONCLUSION</b>Under gastrointestinal physiological conditions, pulchinenosiden D exists in molecular state and it has good absorption but poor water-solubility, so increasing the dissolution rate of pulchinenosiden D may enhance its bioavailability.</p>
