ABSTRACT

Abstract In the hospital environment, postoperative pain is a common occurrence that impairs patient recovery and rehabilitation and lengthens hospitalization time. Racemic bupivacaine hydrochloride (CBV) and Novabupi® (NBV) (S (-) 75% R (+) 25% bupivacaine hydrochloride) are two examples of local anesthetics used in pain management, the latter being an alternative with less deleterious effects. In the present study, biodegradable implants were developed using Poly(L-lactide-co-glycolide) through a hot molding technique, evaluating their physicochemical properties and their in vitro drug release. Different proportions of drugs and polymer were tested, and the proportion of 25%:75% was the most stable for molding the implants. Thermal and spectrometric analyses were performed, and they revealed no unwanted chemical interactions between drugs and polymer. They also confirmed that heating and freeze-drying used for manufacturing did not interfere with stability. The in vitro release results revealed drugs sustained release, reaching 64% for NBV-PLGA and 52% for CBV-PLGA up to 30 days. The drug release mechanism was confirmed by microscopy, which involved pores formation and polymeric erosion, visualized in the first 72 h of the in vitro release test. These findings suggest that the developed implants are interesting alternatives to control postoperative pain efficiently.

ABSTRACT Purpose To evaluate the use of barbed sutures over the surgical time, the leukogram, the tissue thickness in which the sutures were employed (ultrasonography), the costs, and the possible complications in bitches with pyometra submitted to ovariohysterectomy (OH). Methods Convectional 2.0 polyglyconate suture was used in the control group (CG n = 10) and 2.0 barbed polyglyconate suture in the barbed group (BG n = 10) to perform celiorrhaphy (simple continuous pattern) and subcutaneous closure (continuous intradermal pattern). Data were assessed using paired (leukogram between 24 and 48 h within the same group) and unpaired (leukogram, surgical time, tissue thickness, and costs) Student's t-test. The Fisher exact test was used to assess the occurrence of seroma between groups (p < 0.05). Results are shown as mean ± standard error of mean. Results The time spent to perform the celiorrhaphy (195.30 ± 17.37 s vs. 204 ± 16.00 s), subcutaneous closure (174.0 ± 15.86 s vs. 198.0 ± 15.62 s), and the total surgical time (24.30 ± 1.44 min vs. 23.00 ± 1.30 min) did not differ between BG and CG, respectively (p > 0.05). Leukogram at 48 h post-surgery did not differ between groups (p = 0.20). No differences were observed in the subcutaneous and the abdominal wall thickness (cm) assessed by ultrasonography at 48 h in BG (0.31 ± 0.04, 0.80 ± 0.05) and CG (0.34 ± 0.03, 0.72 ± 0.06), respectively. Similarly, 15 days post-surgery the same structures did not differ between BG (0.26 ± 0.02, 0.74 ± 0.08) and CG (0.26 ± 0.03, 0.64 ± 0.05) (p > 0.05). In one bitch from each group, a mild seroma was observed on one side of the surgical wound 48 h after surgery (p = 1.00). The procedures in which barbed sutures were used had an average additional cost of R$ 200.00 ± 11.66 (p < 0.0001). Conclusions Barbed suture has proven to be efficient and safe for abdominal and subcutaneous closure. However, considering its current high cost in addition thatthe surgical time of bitches with pyometra undergone OH was not reduced, no advantages were observed with theuse of barbed sutures for this type of surgery.

Objetivo: Avaliar o efeito do envelhecimento térmico por termociclagem na estabilidade de cor e rugosidade superficial de duas resinas compostas, avaliar grau de conversão (GC), sorção (SO), solubilidade (SL) e microdureza. Material e método: Foram confeccionados 10 espécimes (6 mm x 1 mm) para cada resina: Filtek Z350 XT (Z350XT) e Estelite Sigma Quick (ES). Foram avaliados: GC, utilizando um espectrofotômetro no infravermelho, SO e SL. A avaliação da microdureza Vickers (MV) e densidade de ligações cruzadas foi realizada antes e após a imersão em etanol por 24 h. A rugosidade superficial e a estabilidade de cor (CIEDE2000) foram avaliadas antes e após envelhecimento térmico por 2000 ciclos, alternando 30 s a 5°, 37° e 55° C. A estabilidade de cor e translucidez foram avaliadas com um espectrofotômetro. Os dados foram submetidos ao teste t (α=0,05). Resultados: ES apresentou resultados de GC maiores (62,5% ± 3,7) que Z350XT (48,7% ± 6,9) (p=0,004). Ambas resinas apresentaram resultados de SO e SL similares. A MV final da ES diminuiu 13% com relação a inicial (p=0,115). Z350XT apresentou diminuição de 17% (p=0,004). ES apresentou aumento de 115% na rugosidade superficial após a termociclagem (p=0,001), enquanto a Z350XT apresentou diminuição de 35% (p=0,299). ES apresentou menor alteração de cor (0,280 ± 0,093), quando comparada a Z350XT (0,549 ± 0,183). A translucidez foi similar para ambos materiais (ES=30,80; Z350XT=30,35). Conclusão: A termociclagem influenciou na alteração de cor e rugosidade superficial das resinas compostas, demonstrando que podem ser alteradas pela degradação na matriz polimérica.


Objective: To evaluate the thermocycling effect on color stability and surface roughness of two resin composites and also evaluate the degree of conversion (GC), sorption (SO), solubility (SL) and microhardness. Material and method: 10 specimens (6 mm x 1 mm) were made for each resin: Filtek Z350 XT (Z350XT) and Estelite Sigma Quick (ES). The following were evaluated: GC, using an infrared spectroscopy, SO and SL. Vickers microhardness (MV) and cross-link density evaluation was performed before and after immersion in ethanol for 24 hours. Surface roughness and color stability (CIEDE2000) were evaluated before and after thermal aging for 2000 cycles, alternating between 30 s at 5°, 37° and 55° C. Color stability and translucency were assessed with a spectrophotometer. Data were submitted to t test (α= 0.05). Results: ES presented higher GC results (62.5% ± 3.7) than Z350XT (48.7% ± 6.9) (p= 0.004). Both resins showed similar SO and SL results. The final MV of ES decreased 13% in relation to the initial (p= 0.115). Z350XT decreased by 17% (p= 0.004). ES showed a 115% increase in surface roughness after thermocycling (p= 0.001), while the Z350XT showed a 35% decrease (p= 0.299). ES showed less color change (0.280 ± 0.093) when compared to Z350XT (0.549 ± 0.183). The translucency was similar for both materials (ES= 30.80; Z350XT= 30.35). Conclusion: Thermocycling influenced the change in color stability and surface roughness of composite resins, demonstrating that they may be altered by degradation in the polymer matrix.

ABSTRACT

ABSTRACT Sickle cell anemia (SCA) is a common genetic blood disorder, affecting millions worldwide. According to current evidence, individuals with SCA have more than 300 times greater risk to develop bacterial meningitis (BM) than the general population. Herein we have described the characteristics of a series of BM cases in SCA patients in Salvador, Brazil, during 13 years of hospital-based surveillance. Data on clinical presentation, laboratory parameters and outcomes were collected retrospectively by reviewing medical records. From 1999 to 2011, ten SCA patients were identified among the 2511 cases of BM (10/2511; 0.40%). These patients were more likely to be male (90%) and to be younger (median age 8.5 years). The causative agents were Streptococcus pneumoniae (n = 5) and Haemophilus influenzae (n = 1). The most frequent pneumococcal serotypes were 23 F (2 cases), 14, 18 F, 23B (one case each). Common medical complications were stroke (n = 3); heart failure (n = 2), respiratory problems (n = 2), renal dysfunctions (n = 2) and leg ulcers (n = 1). This study highlights the importance of S. pneumoniae as a causative agent of meningitis in individuals with SCA and shows the diversity of comorbidities associated with this condition.

Abstract When exposure of the pulp to external environment occurs, reparative dentinogenesis can be induced by direct pulp capping to maintain pulp tissue vitality and function. These clinical situations require the use of materials that induce dentin repair and, subsequently, formation of a mineralized tissue. Objective: This work aims to assess the effect of tricalcium silicate cements and mineral trioxide aggregate cements, including repairing dentin formation and inflammatory reactions over time after pulp exposure in Wistar rats. Methodology: These two biomaterials were compared with positive control groups (open cavity with pulp tissue exposure) and negative control groups (no intervention). The evaluations were performed in three stages; three, seven and twenty-one days, and consisted of an imaging (nuclear medicine) and histological evaluation (H&E staining, immunohistochemistry and Alizarin Red S). Results: The therapeutic effect of these biomaterials was confirmed. Nuclear medicine evaluation demonstrated that the uptake of 99mTc-Hydroxymethylene diphosphonate (HMDP) showed no significant differences between the different experimental groups and the control, revealing the non-occurrence of differences in the phosphocalcium metabolism. The histological study demonstrated that in mineral trioxide aggregate therapies, the presence of moderate inflammatory infiltration was found after three days, decreasing during follow-ups. The formation of mineralized tissue was only verified at 21 days of follow-up. The tricalcium silicate therapies demonstrated the presence of a slight inflammatory infiltration on the third day, increasing throughout the follow-up. The formation of mineralized tissue was observed in the seventh follow-up day, increasing over time. Conclusions: The mineral trioxide aggregate (WhiteProRoot®MTA) and tricalcium silicate (Biodentine™) present slight and reversible inflammatory signs in the pulp tissue, with the formation of mineralized tissue. However, the exacerbated induction of mineralized tissue formation with the tricalcium silicate biomaterial may lead to the formation of pulp calcifications

Abstract Candida infection is an important cause of morbidity and mortality in immunocompromised patients. The increase in its incidence has been associated with resistance to antimicrobial therapy and biofilm formation. The aim of this study was to evaluate the efficacy of tea tree oil (TTO) and its main component - terpinen-4-ol - against resistant Candida albicans strains (genotypes A and B) identified by molecular typing and against C. albicans ATCC 90028 and SC 5314 reference strains in planktonic and biofilm cultures. The minimum inhibitory concentration, minimum fungicidal concentration, and rate of biofilm development were used to evaluate antifungal activity. Results were obtained from analysis of the biofilm using the cell proliferation assay 2,3-Bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT) and confocal laser scanning microscopy (CLSM). Terpinen-4-ol and TTO inhibited C. albicans growth. CLSM confirmed that 17.92 mg/mL of TTO and 8.86 mg/mL of terpinen-4-ol applied for 60 s (rinse simulation) interfered with biofilm formation. Hence, this in vitro study revealed that natural substances such as TTO and terpinen-4-ol present promising results for the treatment of oral candidiasis.

Abstract This study examined the effects of liming on soil acidity and base saturation (V) in Italian ryegrass (ryegrass) and silage maize production systems over the medium-term. A split-plot design with four replications was employed on Humic Hapludox. The plots consisted of four soil management methods: conventional tillage (CT), minimum tillage (MT), no-tillage (NT) and chiselled NT (CNT). Within the split plots, ryegrass was used as cover crop (CC), silage (S), and integrated crop-livestock (ICL) system. Following ryegrass phytomass the maize was sowed. Once the maize had been cut with a silage machine, soil samples were collected from the layers 0-5, 5-10, 10-15, 15-20 and 20-30 cm at 24 and 60 months after liming. The attributes pH, H+Al, exchangeable aluminium (Al3+), and V were evaluated. The highest pH and V values were observed in the 0-5 cm layer in NT. The CNT did not result in improvement of acidity conditions or V. The ryegrass as S and ICL decreased soil acidity from 10 cm layer. NT combined with ICL provided soil acidity improvements 60 months after liming. Therefore, the exploration of winter with Italian ryegrass may elicit greater benefits in deep soil layers than cover crops.

Sweet sorghum is currently an important alternative for ethanol production in sugar cane off-season. In this study was to evaluate the effects of plant arrangements and the application of atrazine and S-metolachlor on growth and productivity of sweet sorghum. An experiment was conducted in randomized block design and, arranged in split plots with four repetitions. Plots consisted of spacing (0.25 m, 0.45 m 0.45-0.45-0.90 m) and sub-plots of weed control methods - atrazine (2000 g ha-1), atrazine + S-metolachlor (2000 + 720 g ha-1), atrazine + S-metolachlor (2000 + 960 g ha-1), atrazine + S-metolachlor (2000 + 1200 g ha-1), atrazine + S-metolachlor (2000 + 1440 g ha-1). Besides that, manual hoeing control was added. The intoxication of sorghum plants and weed control was evaluated at 7, 14 e 28 days after herbicides application (DAA). Plant height, stem diameter, total fresh matter, juice mass and total soluble solids (°Brix) were evaluated at 120 days after crop emergence (DAE). There was no interaction between the factors spacings and control method for variables total matter fresh (TMF), height (H), stem diameter (D), juice mass (JM), and Brix. Application of the mixture (atrazine + S-metolachlor) in the highest doses reduced total fresh matter and °Brix. The spacing of 0.25 m provided the highest productivity of fresh matter, juice mass and increased the °Brix. The application of mixture (atrazine + S-metolachlor) controlled 90% of the weeds at 28 days after application. The increasing in doses of S-metolachlor elevates the intoxication in sorghum plants. The mixture (atrazine + S-metolachlor) has a potential for use in weed control in sweet sorghum, but at doses below 960 g ha-1 of S-metolachlor.


O objetivo desse trabalho foi avaliar os efeitos de arranjos de plantas e dos herbicidas atrazine e S-metolachlor sobre o crescimento e a produtividade do sorgo sacarino. O experimento foi realizado no delineamento em blocos casualizados, com quatro repetições. Os tratamentos foram dispostos em esquema de parcelas subdivididas. Nas parcelas avaliaram-se os espaçamentos entrelinhas (0,25 m, 0,45 m e 0,45-0,45-0,90 m) e nas subparcelas os métodos de controle das plantas daninhas: atrazine (2000 g ha-1), atrazine + S-metolachlor (2000 + 720 g ha-1), atrazine + S-metolachlor (2000 + 960 g ha-1), atrazine + S-metolachlor (2000 + 1200 g ha-1), atrazine + S-metolachlor (2000 + 1440 g ha-1), capina manual e ausência de controle. Aos 7, 14 e 28 dias após a aplicação dos herbicidas foi avaliado a intoxicação das plantas de sorgo e o nível de controle de plantas daninhas na cultura. Aos 120 dias após a emergência da cultura avaliou-se altura de plantas, diâmetro de colmo, massa fresca total, massa de caldo e o teor de sólidos solúveis totais (Brix). Não houve interação entre os fatores espaçamentos e o método de controle para as variáveis massa fresca total (TMF), altura (H), diâmetro do caule (D), peso do suco (JM) e Brix. O controle das plantas daninhas e a intoxicação visual do sorgo aumentaram linearmente com o incremento das doses do S-metolachlor. O crescimento da cultura e os teores de Brix foram reduzidos pela aplicação da associação entre atrazine + S-metolachlor. A escolha do espaçamento de entrelinha influenciou nos componentes de rendimento da cultura. Conclui-se que, o espaçamento de 0,25 m de entrelinhas proporcionou maior produtividade de matéria fresca e de Brix do caldo. A associação entre atrazine e S-metolachlor tem potencial para o uso no controle de plantas daninhas em sorgo sacarino, em doses abaixo de 960 g ha-1 de S-metolachlor.

Abstract Introduction: Burnt sugarcane harvesting requires intense physical exertion in an environment of high temperature and exposure to particulate matter. Objective: To evaluate the effects of burnt sugarcane harvesting on rhinitis symptoms and inflammatory markers in sugarcane workers. Methods: A total of 32 male sugarcane workers were evaluated with questionnaire for rhinitis symptoms, and for inflammatory markers on peripheral blood and nasal lavage, in the non-harvesting, and 3 and 6 months into the sugarcane harvesting period. Weather data and particulate matter fine concentrations were measured in the same day. Results: The particulate matter concentrations in sugarcane harvesting were 27 (23-33 µg/m3), 112 (96-122 µg/m3), and 63 (17-263 µg/m3); 24 h temperatures were 32.6 (25.4-37.4 ºC), 32.3 (26.7-36.7 ºC) and 29.7 (24.1-34.0 ºC) and relative humidities were 45.4 (35.0-59.7%), 47.9 (39.1-63.0%), and 59.9 (34.7-63.2%) in the non-harvesting period, three and 6 months of the harvesting period. The age was 37.4 ± 10.9 years. The prevalence of rhinitis symptoms was significantly higher at 3 months of the harvesting period (53.4%), compared to non-harvesting period (26.7%; p = 0.039) and at 6 months into the harvesting period (20%; p = 0.006). Concentrations of interleukin 6 (IL-6) in nasal lavage increased after 3 months of the harvesting period compared to the non-harvesting period (p = 0.012). The presence of rhinitis symptoms, after 3 months of the harvesting period, was directly associated with blood eosinophils and inversely associated with neutrophils. Conclusions: After 3 months of work in burnt sugarcane harvesting the prevalence of rhinitis symptoms and IL-6 in nasal lavage increased. Furthermore, eosinophil counts were directly associated with the rhinitis symptoms in the period of higher concentration of particulate matter.


Resumo Introdução: A colheita de cana-de-açúcar queimada requer esforço físico intenso em um ambiente com altas temperaturas e exposição a material particulado. Objetivo: Avaliar os efeitos da colheita de cana-de-açúcar queimada nos sintomas de rinite e marcadores inflamatórios de cortadores de cana-de-açúcar. Método: Foram avaliados 32 cortadores de cana-de-açúcar do sexo masculino por meio de um questionário para sintomas de rinite, e marcadores inflamatórios em sangue periférico e lavado nasal, no período de entressafra, e em 3 e 6 meses após o início da colheita da cana-de-açúcar. Os dados climáticos e as concentrações de material particulado fino foram medidos no mesmo dia. Resultados: O material particulado fino na entressafra e em 3 e 6 meses de safra foi 27 (23-33 µg/m3), 112 (96-122 µg/m3) e 63 (17-263 µg/m3), respectivamente; a temperatura de 24 horas foi 32,6 (25,4º-37,4ºC), 32,3 (26,7º-36,7ºC) e 29,7 (24,1º-340ºC) e a umidade relativa do ar foi 45,4 (35,0%-59,7%), 47,9 (39,1%-63,0%), e 59,9 (34,7%-63,2%), na entressafra, 3 e 6 meses após o início da colheita. A idade foi de 37,4 ± 10,9 anos. A prevalência de sintomas de rinite foi significativamente maior em 3 meses da S (53,4%), comparado com a entressafra (26,7%; p = 0,039) e 6 meses da safra (20%; p = 0,006). As concentrações de interleucina 6 (IL-6) no lavado nasal aumentaram após 3 meses do início da colheita comparado com a entressafra (p = 0,012). A presença de sintomas de rinite, após 3 meses do início da colheita, foi diretamente associada com eosinófilos e inversamente associada com neutrófilos. Conclusões: Após 3 meses do início da colheita da cana-de-açúcar queimada, houve aumento na prevalência de sintomas de rinite e IL-6 em LN. Além disso, as contagens de eosinófilos foram diretamente associadas aos sintomas de rinite no período de maior concentração de material particulado.

Objective: To evaluate in vitro the effect of a red propolis ethanolic extract (RPE) in the prevention of growth of a cariogenic biofilm and its cytotoxic potential. Material and Methods: Minimum inhibitory and bactericidal concentrations (MIC and MBC) of RPE against Streptococcus mutans and Lactobacillus casei were determined. The cytotoxic potential of 0.4% RPE in oral fibroblasts was observed after 1, 3 and 5 min of contact. Cellulose membrane disks (13 mm, N=12) were used for biofilm formation (24 h) of S. mutans and L. casei, which were treated (1 min) with 0.4% RPE or 0.12% Chlorhexidine (CHX). The control group of biofilm formation was not submitted to any treatment. Serial dilutions were then made to evaluate microbial viability. Descriptive data analysis and, for microbial viability, Mann Whitney test were performed (p≤0.05). Results: RPE showed similar MIC and MBC (4.46 mg/mL) against S. mutans and, for L. casei, they were 8.92 mg/mL (MIC) and 17.85 mg/mL (MBC). CHX presented MIC and MBC <0.00002 mg/mL for S. mutans and 0.00047 mg/mL for L. casei. After 1, 3 and 5 min, the RPE exhibited, respectively, 69.38%, 43.91% and 40.36% of viable cells. The RPE (6.55) and CHX (6.87) presented similar efficacy to reduce the total number of viable bacteria (p>0.05). Regarding the total number of viable bacteria (Log10 CFU/mL), the RPE (6.55) and CHX (6.87) presented similar efficacy (p>0.05). Conclusion: Red propolis extract showed antibacterial activity against the tested strains, exhibited acceptable cytotoxicity and reduced the colonization of S. mutans and L. casei in a biofilm membrane model.

Objetivo:O presente estudo piloto "in vitro" avaliou o efeito antimicrobiano do hipoclorito de sódio, clorexidina, terapia fotodinâmica e do óleo de girassol ozonizado utilizados como agentes irrigantes na eliminação do Enterococcus faecalis em canais radiculares de dentes humanos extraídos.Métodos:Sessenta raízes de dentes unirradiculares (n = 60) foram selecionadas e autoclavadas para realização do experimento em fluxo laminar. Para a realização do teste de infiltração, confeccionou-se um dispositivo para cada denteque permitiu a contaminação por Enterococcus faecalis e posterior preparo químico-mecânico. As amostras foram submetidas ao preparo biomecânico com diâmetro cirúrgico correspondente à lima #45 e mesma conicidade nos terços médio e cervical e receberam protocolo de desinfecção com hipoclorito de sódio a 2,5% (NaOCl), Clorexidina a 2% (CHX), óleo de girassol ozonizado a 2400ppm (OGO), PDT após irrigação com soro fisiológico (PDT+S) e PDT após irrigação com hipoclorito de sódio a 2,5% (PDT+H). As dez raízes restantes constituíram os controles positivo e negativo, sendo cinco dentes para cada grupo. Os controles positivos foram infectados e não foi utilizado nenhum agente irrigante. Os controles negativos foram compostos por dentes não contaminados pelo E. faecallis. Resultados: Os resultados da contagem de UFC/mL total e análise descritiva foram realizados. NaOCl e PDT+H apresentaram crescimento bacteriano inferiores aos valores de referência e foi considerado nulo. CHX e PDT+S apresentaram crescimento bacteriano baixo e OGO apresentou crescimento bacteriano moderado. Conclusão: NaOCl e PDT+H apresentaram melhor desempenho em relação aos demais protocolos de desinfecção utilizados. OGO apresentou crescimento bacteriano moderado sugerindo inviabilidade de sua utilização isolada em protocolos de desinfecção em endodontia. (AU)


Aim: The present "in vitro" pilot study evaluated the antimicrobial effects of sodium hypochlorite, chlorhexidine, photodynamic therapy, and ozonated sunflower oil, which were used as irrigating agents in the elimination of Enterococcus faecalis in the root canals of extracted human teeth. Methods: Sixty roots of single-root teeth (n = 60) were selected and autoclaved to perform the laminar flow experiment. To perform the infiltration test, a device was constructed for each tooth, which allowed for contamination by Enterococcus faecalis, and the tooth's subsequent chemical-mechanical preparation. The samples were submitted to biomechanical preparation with surgical diameter, corresponding to file #45 and the same taper in the middle and cervical thirds, and received a disinfection protocol with 2.5% sodium hypochlorite (NaOCl), chlorhexidine 2% (CHX), ozonized sunflower oil at 2400ppm (OGO), PDT after irrigation with saline solution (PDT+S), and PDT after irrigation with 2.5% sodium hypochlorite (PDT + H). The remaining ten roots were positive and negative controls, with five teeth in each group. Positive controls were infected, and no irrigating agent was used. Negative controls consisted of teeth that were not contaminated by E. faecallis. Results: The results of the total CFU count and descriptive analysis were performed. NaOCl and PDT+H presented a bacterial growth of much lower t than the reference values and was considered null. CHX and PDT+S presented low bacterial growth, while OGO presented moderate bacterial growth. Conclusion: NaOCl and PDT + H presented better performance in relation to the other disinfection protocols used in this study. OGO presented moderate bacterial growth, suggesting its unviable use in endodontic disinfection protocols. (AU)

ABSTRACT

Objetivo: Avaliar a incidência de sinais e sintomas de depressão entre acadêmicos de enfermagem de uma instituição do sul de Minas Gerais. Metodologia: estudo de natureza quantitativa, com abordagem exploratória, descritiva e transversal, com a participação de 116 acadêmicos de enfermagem selecionados entre o primeiro e o décimo período de graduação, avaliados por meio do inventário de Beck. Resultados: dos 21 itens avaliados pelo referido instrumento, evidenciamos que apenas 3 itens que compõem o inventário (autoacusação, inibição do sono e irritabilidade) se destacaram entre a amostra estudada. Conclusão: Por meio da aplicação do inventário de Beck, evidenciamos a probabilidade de se desenvolver entre estes acadêmicos um quadro depressivo futuramente, caso os mesmos não tenham um equilíbrio emocional, apoio de professores, família e comunidade para prevenir a manifestação da depressão.


Objective: To evaluate the incidence of signs and symptoms of depression among nursing students from an institution in the south of Minas Gerais. Methodology: a quantitative study, with an exploratory, descriptive and cross-sectional approach, with the participation of 116 nursing students selected between the first and the tenth graduation period, evaluated through the Beck inventory. Results: of the 21 items evaluated by said instrument, we showed that only 3 items that compose the inventory (selfaccusation, sleep inhibition and irritability) stood out among the sample studied. Conclusion: Using the Beck inventory, we show the probability of developing a future depressive state among these students, if they do not have an emotional balance, support of teachers, family and community to prevent the manifestation of depression.

Abstract Purpose: To assess the action of vitamin C on the expression of 84 oxidative stress related-genes in cultured skin fibroblasts from burn patients. Methods: Skin samples were obtained from ten burn patients. Human primary fibroblasts were isolated and cultured to be distributed into 2 groups: TF (n = 10, fibroblasts treated with vitamin C) and UF (n = 10, untreated fibroblasts). Gene expression analysis using quantitative polymerase chain reaction array was performed for comparisons between groups. Results: The comparison revealed 10 upregulated genes as follows: arachidonate 12-lipoxygenase (ALOX12), 24-dehydrocholesterol reductase (DHCR24), dual oxidase 1 (DUOX1), glutathione peroxidase 2 (GPX2), glutathione peroxidase 5 (GPX5), microsomal glutathione S-transferase 3 (MGST3), peroxiredoxin 4 (PRDX4), phosphatidylinositol-3,4,5-trisphosphate dependent Rac exchange factor 1 (P-REX1), prostaglandin-endoperoxide synthase 1 (PTGS1), and ring finger protein 7 (RNF7). Conclusion: Cultured fibroblasts obtained from burn patients and treated with vitamin C resulted in 10 differentially expressed genes, all overexpressed, with DUOX1, GPX5, GPX2 and PTGS1 being of most interest.

Abstract Yeast infections have acquired great importance due to increasing frequency in immunocompromised patients or patients undergoing invasive diagnostic and therapeutic techniques, and also because of its high morbidity and mortality. At the same time, it has been seen an increase in the emergence of new pathogenic species difficult to diagnose and treat. The aim of this study was to determine the in vitro susceptibility of 89 yeasts from different sources against the antifungals amphotericin B, voriconazole, fluconazole and flucytosine, using the VITEK® 2 Compact system. The antifungal susceptibility was performed automatically by the Vitek® 2 Compact system. The origin of the yeasts was: Group 1 - microbiota of wild animals (W) (26/89), 2 - cow's milk with subclinical mastitis (M) (27/89) and 3 - hospital enviorment (H) (36/89). Of the 89 yeasts submitted to the Vitek® 2 test, 25 (20.9%) were resistant to fluconazole, 11 (12.36%) to amphotericin B, 3 (3.37%) to voriconazole, and no sample was resistant to flucytosine. Regarding the minimum inhibitory concentration (MIC), fluconazole showed an MIC between 1 and 64 mg/mL for the three groups, voriconazole had an MIC between 0.12 and 8 mg/mL, amphotericin B had an MIC between 0.25 and 4 mg/mL for group H and group W respectively, between 0.25 and 16 mg/mL for group M and flucytosine had an MIC equal to 1μg/mL for all groups. The yeasts isolated from the H group showed the highest resistance to fluconazole 12/89 (13.49%), followed by group W (7.87%) and group M (5.62%). The more resistant group to voriconazole was followed by the M and H groups, the W group showed no resistance to this antifungal. Group H was the least resistant (2.25%) to amphotericin.


Resumo As infecções por leveduras têm adquirido grande importância, devido ao aumento da sua frequência em pacientes imunocomprometidos ou pacientes submetidos a técnicas diagnosticas e terapêuticas agressivas, e devido sua alta morbidade e mortalidade. Paralelamente tem-se observado um incremento na aparição de novas espécies patógenas difíceis de diagnosticar e tratar. O objetivo desse estudo foi avaliar a suscetibilidade in vitro de 89 leveduras de diferentes origens frente aos antifúngicos Anfotericina B, Voriconazol, Fluconazol e Fluocitocina pelo Sistema Vitek® 2. O antifungigrama foi realizado automaticamente pelo Vitek® 2 Compact. A origem das leveduras foi: Grupo 1- Microbiota de Animais Silvestres (S) (26/89), 2- Leite com mastite bovina subclínica (L) (27/89) e 3- Ambiente Hospitalar (H) (36/89). Das 89 leveduras submetidas à carta Vitek®, 25 (20.09%) foram resistentes ao fluconazol, oito (8.99%) à anfotericina B, três (3.37%) ao voriconazol, e nenhuma amostra mostrou-se resistente a fluocitosina. O grupo três (H) foi mais resistente ao fluconazol que os demais, já o dois (L) foi mais resistente ao voriconazol e a anfotericina B que os outros dois. O fluconazol pode ter apresentado maior número de resistências devido ser um fármaco comumente usado principalmente em humanos. As leveduras isoladas de humanos apresentaram maior número de resistências aos fármacos testados do que as leveduras isoladas de animais silvestres. O que pode ocorrer devido a uma maior exposição dos humanos aos fármacos em relação aos animais que vivem isolados em ambientes selvagens e na maioria dos casos nunca teve contato com fármacos de qualquer origem.

Abstract Objective: To compare the effectiveness of a single intramuscular dose of bromopride, metoclopramide, or ondansetron for treating vomiting. Methods: Randomized controlled trial including children 1-12 years of age presenting with acute vomiting at the pediatric emergency department. Outcomes: Number of children that stopped vomiting at one, six, and 24 h following treatment; episodes of diarrhea; acceptance of oral liquids; intravenous rehydration; return to hospital and side effects. Results: There were 175 children who completed the study. Within the first hour after treatment, all drugs were equally effective, with ondansetron preventing vomiting in 100%, bromopride in 96.6%, and metoclopramide in 94.8% of children (p = 0.288). Within six hours, ondansetron was successful in preventing vomiting in 98.3% of children, compared to bromopride and metoclopramide, which were successful in 91.5% and 84.4% of patients, respectively (p = 0.023). Within 24 h, ondansetron was superior to both other agents, as it remained efficacious in reducing vomiting in 96.6% of children, as opposed to 67.8% and 67.2% with bromopride and metoclopramide, respectively (p = 0.001). The ondansetron group showed better acceptance of oral liquids (p = 0.05) when compared to the bromopride and metoclopramide. The ondansetron group did not show any side effects in 75.9% of cases, compared to 54.2% and 53.5% in the bromopride and metoclopramide groups, respectively. Somnolence was the most common side effect. Conclusions: A single dose of ondansetron is superior to bromopride and metoclopramide in preventing vomiting six hours and 24 h following treatment. Oral fluid intake after receiving medication was statistically better with Ondansetronwhile also having less side effects compared to the other two agents.


Resumo Objetivo: Para comparar a eficacia de uma unica dose intramuscular de bromoprida, metoclopramida ou ondansetrona no tratamento de vomito. Métodos: Ensaio controlado randomizado incluindo crianc¸as de 1 a 12 anos de idade que apresentam vomito agudo no departamento de emergencia pediatrica. Desfechos: Numero de crianças que pararam de vomitar 1, 6 e 24 horas apos o tratamento; episodios de diarreia; aceitac¸ao de liquidos orais; reidratac¸ao intravenosa, retorno ao hospital e efeitos colaterais. Resultados: 175 crianças concluiram o estudo. Na primeira hora apos o tratamento, todos os medicamentos foram igualmente eficazes, sendo que a ondansetrona preveniu vomito em 100%, a bromoprida em 96,6% e metoclopramida em 94,8% das crianças (p = 0,288). Em 6 horas, a ondansetrona mostrou sucesso na prevençao do vomito em 98,3% das crianças, em comparac¸ao a bromoprida e a metoclopramida, que mostraram sucesso em 91,5% e 84,4% dos pacientes, respectivamente (p = 0,023). Em 24 horas, a ondansetrona foi superior aos dois outros agentes, pois ela continuou eficaz na reduçao do vomito em 96,6% das crianças, diferente de 67,8% e 67,2% com bromoprida e metoclopramida, respectivamente (p = 0,001). A ingestao de fluidos orais apos receber medicaçao foi estatisticamente melhor com ondansetrona, ao mesmo tempo em que tambem apresentando menos efeitos colaterais em comparaçao aos outros dois agentes.

ABSTRACT

This study aimed to assess the weight gain and the incidence of foot diseases in male, crossbred (Bos taurus x Bos indicus) bovines that were supplemented with biotin. An amount of 240 animals, supplemented or not with biotin, allocated in 12 groups of 20 animals, was assessed for a period of six months. The study was conducted during three years and the groups were divided according to the forage available, corn, corn residue and sorghum silage, and initial weights between 100 and 200 kg and between 200 and 300 kg. The statistical analyses used were the Tukey's Test, in triple factorial scheme (type of silage x use of biotin x initial body weight) and the Fisher´s Exact Test, both at 5% significance level. The biotin supplementation in bovines did not influence weight gain and the incidence of foot diseases, however, when comparing only the type of forage, corn and sorghum silage provided higher weight gain than silage made of corn residue.


Esse estudo teve como objetivo avaliar o ganho em peso e a ocorrência de enfermidades digitais em bovinos do sexo masculino, mestiços (Bos taurus x Bos indicus) que foram suplementados com biotina. Avaliaram-se, por um período de seis meses, 240 bovinos, suplementados ou não com biotina, alocados em 12 grupos de 20 animais. O estudo foi realizado durante três anos e os grupos foram divididos de acordo com o volumoso disponibilizado, silagem de milho, de resíduo de milho e de sorgo, e pesos iniciais entre 100 e 200 kg e entre 200 e 300 kg. As análises estatísticas empregadas foram o Teste de Tukey, em esquema de fatorial triplo (tipo de silagem x uso da biotina x peso corporal inicial) e o Teste Exato de Fisher, ambas em nível de significância de 5%. A suplementação com biotina nos bovinos não exerceu influência sobre o ganho em peso e a ocorrência de enfermidades digitais, mas, quando se comparou apenas o tipo de volumoso, a silagem de milho e sorgo, pode se observar um maior ganho em peso que a silagem confeccionada de resíduo de milho.

Abstract Mucoadhesive nanoparticles are particularly interesting for delivery through nasal or pulmonary routes, as an approach to overcome the mucociliary clearance. Moreover, these nanoparticles are attractive for peptide and protein delivery, particularly for insulin to treat diabetes, as an alternative to conventional parenteral administration. Thus, chitosan, a cationic mucoadhesive polysaccharide found in shells of crustaceans, and the negatively-charged dextran sulfate are able to form nanoparticles through ionic condensation, representing a potential insulin carrier. Herein, chitosan/dextran sulfate nanoparticles at various ratios were prepared for insulin loading. Formulations were characterized for particle size, zeta potential, encapsulation efficiency, scanning electron microscopy, differential scanning calorimetry, and in vitro drug release. Moreover, the interaction with mucin and the cytotoxicity against a lung cell line were studied, which altogether have not been addressed before. Results evidenced that a proper selection of polyelectrolytes is necessary for smaller particle size formation and also the composition and zeta potential impact encapsulation efficiency, which is benefited by the positive charge of chitosan. Insulin remained stable after encapsulation as evidenced by calorimetric assays, and was released in a sustained manner in the first 10 h. Positively-charged nanoparticles based on chitosan/dextran-sulfate at the ratio of 6:4 successfully interacted with mucin, which is a prerequisite for delivery to mucus-containing tissues. Finally, insulin-loaded nanoparticles displayed no cytotoxicity effect against lung cells at tested concentrations, suggesting the potential for further in vivo studies.

ABSTRACT

ABSTRACT Sinotaia quadrata is a snail native from Asia recorded for the first time in South America in 2009 in central Argentina. In 2015, this species was also found in a lowland stream with different water qualities. Our aims were to contribute to the knowledge of its population ecology and to compare the individuals from the two locations anatomically. Snails were searched at 6 sites, where physicochemical and hydraulic parameters were measured. Biological samples were also taken at two sites (S3 and S4) to study the population traits of S. cf quadrata (density, size structure, fecundity and sex ratio) and to assess the water quality through macroinvertebrates' biological indices (richness, diversity and IBPamp). Physicochemical and biological parameters allowed us classifying sites as "moderately polluted" (S3) and "heavily polluted" (S4). At S4, the population showed a lower density, larger individuals, higher fecundity and a scarce representation of young snails. The differences observed in the radula and mantle border of snails from the two geographical regions might be attributed to environmental differences. We conclude that this species is tolerant to a wide range of environmental variables which, along with its high fecundity and morphological plasticity, could allow this species to colonize neighbor streams.

Abstract This study evaluated the effects of three metal primers and one multi-mode adhesive system on the shear bond strength (SBS) of a flowable composite resin to nickel-chrome metal alloy (Ni-Cr). Ninety plates were cast from Ni-Cr and divided in nine groups (n=10). The surfaces were sandblasted with Al2O3 and primed with three adhesive primers: Alloy Primer (AP), Universal Primer (TP) and RelyX Ceramic Primer (CP), and a multi-mode adhesive (Scotchbond Universal, SU). The Adper Single Bond Plus (SB) and SU adhesives were also combined with adhesive primers. Control group did not have any surface treatment. The groups were: AP, AP+SB, AP+SU, TP+SB, TP+SU, CP+SB, CP+SU and SU. Composite cylinders were built on alloy surface. After 24 h, half the specimens were subjected to SBS and the other half to thermal cycling before testing. Data were analyzed by two-way ANOVA and Tukey’s test (a=0.05). Failure modes were assessed by SEM observation. Higher SBS were obtained with AP and TP combined with adhesives at 24 h and the lowest one for control group. Thermocycling reduced SBS for AP, CP+SU and SU. Combination between TP and SU resulted in the highest SBS after the thermocycling. TP groups showed all types of failures and high incidence of mixed failures. The use of AP and UP metal primers before application of SU and SB adhesive systems increased the SBS of composite to Ni-Cr. These combinations between metal primers and adhesives had the highest SBS after thermocycling.


Resumo Este estudo avaliou os efeitos de três primers para metais e um sistema adesivo multi-mode na resistência da união por cisalhamento (SBS) de um compósito resinoso de baixa viscosidade a uma liga niquel-cromo (Ni-Cr). Noventa placas Ni-Cr foram divididas em 9 grupos (n=10). As superfícies foram jateadas com Al2O3 e condicionadas com três primers adesivos: Alloy Primer (AP), Universal Primer (TP) e Rely X Ceramic Primer (CP) e um adesivo multi-mode (Scotchbond Universal, SU). Os adesivos Adper Single Bond Plus (SB) e SU foram combinados com os primers para metais. O grupo controle não recebeu nenhum tipo de tratamento superficial. Os grupos foram: AP, AP+SB, AP+SU, TP+SB, TP+SU, CP+SB, CP+SU e SU. Cilindros de resina foram construídos na superfície da liga. Após 24 h, metade dos espécimes foi submetido ao SBS, a outra metade foi termociclada previamente. Os dados foram analisados pelos testes ANOVA dois fatores e Tukey (a=0.05). Os tipos de fraturas foram determinados utilizando microscopia eletrônica de varredura (MEV). Os maiores valores de SBS foram obtidos com AP e TP combinados com adesivos após 24 h e o menor valor foi obtido no grupo controle. A ciclagem térmica reduziu a SBS para AP, CP+SU e SU. Combinação entre TP e SU resultou em altos valores de SBS após termociclagem. Os grupos TP apresentaram todos os modos de fratura e alta incidência de fraturas mistas. O uso dos primers AP e UP para metais antes da aplicação dos sistemas adesivos SU e SB aumentou a SBS entre o Ni-Cr e a resina composta. Essa combinação entre primers e adesivos mostrou os maiores valores de SBS após a termociclagem.

Abstract Saliva when compared to blood collection has the following advantages: it requires no specialized personnel for collection, allows for remote collection by the patient, is painless, well accepted by participants, has decreased risks of disease transmission, does not clot, can be frozen before DNA extraction and possibly has a longer storage time. Objective and Material and Methods This study aimed to compare the quantity and quality of human DNA extracted from saliva that was fresh or frozen for three, six and twelve months using five different DNA extraction protocols: protocol 1 – Oragene™ commercial kit, protocol 2 – QIAamp DNA mini kit, protocol 3 – DNA extraction using ammonium acetate, protocol 4 – Instagene™ Matrix and protocol 5 – Instagene™ Matrix diluted 1:1 using proteinase K and 1% SDS. Briefly, DNA was analyzed using spectrophotometry, electrophoresis and PCR. Results Results indicated that time spent in storage typically decreased the DNA quantity with the exception of protocol 1. The purity of DNA was generally not affected by storage times for the commercial based protocols, while the purity of the DNA samples extracted by the noncommercial protocols typically decreased when the saliva was stored longer. Only protocol 1 consistently extracted unfragmented DNA samples. In general, DNA samples extracted through protocols 1, 2, 3 and 4, regardless of storage time, were amplified by human specific primers whereas protocol 5 produced almost no samples that were able to be amplified by human specific primers. Depending on the protocol used, it was possible to extract DNA in high quantities and of good quality using whole saliva, and furthermore, for the purposes of DNA extraction, saliva can be reliably stored for relatively long time periods. Conclusions In summary, a complicated picture emerges when taking into account the extracted DNA’s quantity, purity and quality; depending on a given researchers needs, one protocol’s particular strengths and costs might be the deciding factor for its employment.
