ABSTRACT

Objective: To observe the effects of heat-sensitive moxibustion plus Chinese medication on serum inflammatory indicators, T-lymphocyte subsets, and serum microRNAs in patients with multiple myeloma after chemotherapy. Methods: Eighty-two patients with multiple myeloma who had terminated chemotherapy were divided into an observation group and a control group using the random number table method, with 41 cases in each group. The control group received symptomatic treatment of Western medicine plus oral Chinese medication Yi Shen Qiang Gu Tang (decoction for benefiting the kidney to strengthen bones), and the observation group received additional heat-sensitive moxibustion treatment. After the intervention, the following measures were observed, including clinical efficacy, symptom scores of traditional Chinese medicine (TCM), and tumor patient's quality of life (QOL) scores [including Karnofsky performance status (KPS) score, performance scale (PS) score, and tumor-specific QOL questionnaire score] alongside the serum inflammatory indicators [tumor necrosis factor-α (TNF-α), C-reactive protein (CRP), and interleukin-6 (IL-6)], and the relative expression levels of T-lymphocyte subsets (CD3+, CD4+, and CD8+) and serum microRNA-302 (miR-302), microRNA-140-5p (miR-140-5p), and microRNA-125a (miR-125a). Results: After treatment, the total effective rate was 85.4% in the observation group, higher than 68.3% in the control group (P<0.05). The observation group was superior to the control group in improving the serum inflammatory factor levels, TCM symptom scores, tumor patient's QOL scores, and the relative expression levels of T-lymphocyte subsets and serum miR-302, miR-140-5p, and miR-125a, and the between-group differences were statistically significant (P<0.05). Conclusion: Heat-sensitive moxibustion plus Chinese medication can lower the serum inflammatory factor levels, strengthen immune function, mitigate TCM symptoms, enhance QOL, and raise the expression levels of serum miR-125a, miR-140-5p, and miR-302 in multiple myeloma patients after chemotherapy.

ABSTRACT

Objective:To observe the effect of modified Shengxian Decoction on extravascular lung water index (EVLWI) and lung injury prediction score (LIPS) in patients with acute respiratory distress syndrome (ARDS) caused by sepsis.Methods:Prospective cohort study. A total of 200 patients with ARDS caused by sepsis who were hospitalized in Baoshan Branch, Shuguang Hospital Affiliated to Shanghai University of Traditional Chinese Medicine from January 2019 to May 2021 were selected and divided into the observation group and control group by random number table method, with 100 in each group. The patients in the control group were given rountin western medicine treatment according to the guidelines, and the patients in the observation group were treated with modified Shengxian Decoction on the basis of the treatment in the control group. Both groups were treated for 7 days as a course of treatment. The PH value, oxygen volume index (FiO 2), oxygen partial pressure (PaO 2), arterial carbon dioxide partial pressure (PaCO 2) of the two groups before and after treatment, calculate the oxygenation index (PaO 2/FiO 2) were observed and compared. The C-reactive protein (hs-CRP), interleukin-6 (IL-6) levels were observed by ELISA, the procalcitonin (PCT) levels was detected by double antibody sandwich immunoluminescence method. The APACHE Ⅱ score and LIPS score, EVLWI and cardiac index (CI) of the two groups were observed and compared. The mechanical ventilation time and ICU hospitalization time of the two groups were compared. Results:After treatment, the PaCO 2 level [(37.15 ± 5.42) mmHg vs. (38.24 ± 3.24) mmHg, t=2.03] of the observation group was significantly lower than that of the control group, and the oxygenation index (292.34 ± 78.91 vs. 236.54 ± 70.58, t=5.27) was significantly higher than that of the control group ( P<0.05). After treatment, the levels of hs-CRP [(35.21 ± 6.73) mg/L vs. (48.97 ± 8.52) mg/L, t=12.67], IL-6 [(40.57 ± 8.51) ng/L vs. (47.61 ± 9.97) ng/L, t=5.37] and PCT [(0.75 ± 0.21) μg/L vs. (1.14 ± 0.38) μg/L, t=8.98] in the observation group were significantly lower than those in the control group ( P<0.01). After treatment, the APACHE Ⅱscore (11.14 ± 0.54 vs. 14.67 ± 0.89, t=33.91], LIPS score (2.21 ± 0.73 vs. 4.59 ± 0.88, t=20.82), and EVLWI [(6.19 ± 0.42) ml/kg vs. (8.24 ± 0.78) ml/kg, t=23.14) of the observation group were significantly lower than those in the control group, and the CI level [(4.49 ± 1.27) L/(min?m 2) vs. (3.61 ± 0.88) L/(min?m 2), t=5.70] was significantly higher than that of the control group ( P<0.01). The mechanical ventilation time and ICU stay in the observation group were shorter than those in the control group ( t=3.66, 5.74, P<0.01). Conclusion:The modified Shengxian Decoction can reduce the level of inflammation indexes in patients with ARDS caused by sepsis, reduce EVLWI and LIPS scores, improve blood gas analysis indexes, and shorten the time of mechanical ventilation and ICU hospitalization.

ABSTRACT

ObjectiveTo observe the effects of Suanzaoren Tang on the behavior, growth-associated protein-43 (GAP-43), postsynaptic density protein-95 (PSD-95), and synaptophysin Ⅰ (SynⅠ) of insomniac rats induced by p-chlorophenylalanine (PCPA), and to investigate the mechanism of Suanzaoren Tang in improving the behavior of the insomniac rats. MethodSeventy-two SD rats were randomly assigned into 6 groups (12 rats in each group): control group (normal saline), PCPA (0.35 g·kg-1·d-1) group, estazolam (2.7×10-4 g·kg-1·d-1) group, and low-, medium-, and high-dose (3.25, 7.5, 15 g·kg-1·d-1, respectively) Suanzaoren Tang groups. The rat model of insomnia was established by intraperitoneal injection of PCPA and then the rats were administrated with corresponding drugs for 7 continuous days. The Morris water maze and Y-maze were used to test the learning and memory functions, and the open field to test anxiety. Histopathological changes in the hippocampus were observed via hematoxylin-eosin (HE) staining. The mRNA and protein levels of GAP-43, PSD-95, and SynⅠ in hippocampus were determined by real-time fluorescence quantitative polymerase chain reaction (Real-time PCR), Western blot, and immunohistochemistry (IHC). ResultCompared with the control group, the PCPA group showcased long escape latency, shortened time in the quadrants, and decreased times of crossing the platform in Morris water maze, decreased alternation correct rate was significantly Y-maze, and increased distance, mean velocity, and time in center of the open field test (P<0.01). Furthermore, the PCPA-treated rats showed obvious pathological damage in the hippocampus and down-regulated mRNA and protein levels of GAP-43, PSD-95, and SynⅠ in hippocampus (P<0.01). Compared with the PCPA group, the treatments with estazolam and different doses of Suanzaoren Tang improved the rat performance in Morris water maze, Y-maze, and open field test (P<0.05, P<0.01), alleviated the hippocampal damage, and up-regulated the mRNA and protein levels of GAP-43, PSD-95, and SynⅠ (P<0.05, P<0.01). ConclusionSuanzaoren Tang may alleviate the learning and memory disorders and anxiety in PCPA-induced insomnia rat model by up-regulating the mRNA and protein levels of hippocampal synaptic plasticity-associated proteins GAP-43, PSD-95, and SynⅠ.

ABSTRACT

This paper compared and analyzed the relevant records of Chinese medicine pharmacy in the Han,Tang and Song dynasties,and summarized the changes of the dosage forms,preparation techniques and administration methods of Chinese medicine with the development of history.In this study,three classic medical works in the Han,Tang and Song dynasties,including Treatise on Febrile Diseases Caused by Cold,Valuable Prescriptions for Emergency and Formularies of the Bureau of People's Welfare Pharmacies,were taken as the research objects,and the development of the dosage forms,auxiliary materials,preparation technology and medication theory were summarized and explored by the ways of content analysis,comparative analysis and case analysis.The comparison showed that in the development process,the dosage forms gradually increased,but the liquid dosage forms gradually decreased,the solid dosage forms gradually increased.Not only the dosage forms varied in the number,types of excipients used more and more,but the level of preparation had been constantly improved while the methods of taking became more detailed.This evolution of dosage forms and pharmaceutical technologies in Chinese history is worth learning and thinking about it.Through exploring the traditional Chinese medicine(TCM) technologies and theories in the Han,Tang and Song dynasties,we can contribute to the inheritance of traditional preparations and it can provide the basis for the development of modern preparations with TCM characteristics.

ABSTRACT

Objective: To explore the changes and benefits of vascular endothelial cell function in rats with qi deficiency and blood stasis syndrome, and the effect of Yiqi Huoxue recipe (YQHXF) of such changes. Method: Rats were randomly divided into blank control group, Qi deficiency and blood stasis group, and YQHXF high and low dose groups (5.54,2.77 g·kg-1). A small platform of water environment was used to make the rats stand for a long-term with irregular and incomplete sleep deprivation, 16 h per day for six weeks, so that both mentality and labor of rats were consumed to establish qi deficiency and blood stasis rat models. From the fifth week, intragastric administration was given for 2 weeks, until end of the experiment. Then levels of endothelin-1(ET-1), von willebrand factor (vWF), vascular cell adhesion molecule-1(VCAM-1), intercellular adhesion molecule (ICAM-1), P-selectin,interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α) in rat plasma were detected by enzyme-linked immunosorbent assay (ELISA) and nitric oxide (NO) was detected by nitrate reductase assay. Result:Compared with blank control group, rats in Qi deficiency and blood stasis group showed rough and dark hair, with significantly decreased body weight and pulse amplitude (PPPα were abnormally increased after sleep deprivation (PPPPPPPConclusion:Sleep deprivation can induce the formation of Qi deficiency and blood stasis syndrome in rats, and lead to vascular endothelial dysfunction. YQHXF has the function of protecting the vascular endothelium. It can improve the Qi deficiency and blood stasis symptoms in rats with Qi deficiency and blood stasis syndrome by regulating the secretion of vascular endothelial active substances, reducing cell adhesion and inhibiting inflammation.

Objective To evaluate the effect of Ji Tang Zhi on glucose metabolism in insulin resistance (IR) HepG 2 cell line,and to explore the related mechanism.Methods The HepG2 cells were incubated in culture medium addition of 10-7 mol/L insulin for 24 h to establish the IR cell model.Effect of Ji Tang Zhi on rate of glucose absorption in HepG2 cell was detected by the method of glucose oxidase-peroxidase (GOD-POD).We performed an MTT assay to determine cytotoxicity effects of Ji Tang Zhi on HepG2 cell line.The expression of p-IRS-1 Ser307,PI3K and GLUT-4 were detected by Western blotting.Results Incubated with 10-7 mol/L insulin for 24 h,the insulin resistance cell model had been built.Compared with model group,the rate of glucose absorption of cell treated with JTZ (30 ~ 120 μg/mL) was significantly improved.According to model cells,the expression of GLUT-4 and PI3K decreased significantly compared to control cells.While the expression of p-IRS-1 Ser 307 was inhibited and GLUT-4 and PI3K expression were increased in IR cells after treated with JTZ (30 ~ 120 μtg/mL).Conclusion JTZ exert beneficial effects on hyperglycosemia in IR cell line possibly through regulating the levels of GLUT-4,p-IRS-1 Ser307 and PI3K in HepG2 cell.

ABSTRACT

The article was aimed to study the influence of low pressure and hypoxia on rat metabolism and evaluate the intervention effect of San-Guo-Tang-San (SGTS) on high altitude polycythemia (HAPC) rats. A total of 25 rats were divided into the plain control group, high altitude model group, Hong-Jing-Tian capsule (Nuo-Di-Kang cap-sule) group, high-dose SGTS group and low-dose SGTS group, with 5 rats in each group. After one week adaptation, rats in the model group and the medication groups were put into the hyperbaric chamber for 40 days (22 h/d) to simulate high altitude environment of 5 000 m. In the end of 40th day, the hemorheology and the dry/wet weight ratio of lung of rats were measured. And plasma samples were derivatized with ECF prior to GC-MS instrumental analysis. Principal component analysis (PCA) was used to find potential biomarkers, and evaluate the intervention effects of SGTS. The results showed that the low pressure and hypoxia changed the hemorheology and dry/wet weight ratio of lung of rats markedly. Metabolomics studies showed that the high altitude model group, high-dose SGTS group, low-dose SGTS group, and Hong-Jing-Tian capsule group can be obviously differentiated. Main markers such as 9-hexylheptadecane, glycine, N-methyl-N-methoxycarbonyl-ethyl ester, 2,4-Di-tert-butylphenol, were found to be the endogenous substances of SGTS which intervening the HAPC rats. It was concluded that SGTS can intervene low pressure and hypoxia induced HAPC.

ABSTRACT

Objective:To explore the influence ofDan Shou Tang to fetal loss induced by APA.Methods:One hundred of10-week pregnant SD rats were divided randomly into two groups, every group were divided randomly into nourishing kidney and promoting blood flow group A, nourishing kidney group B, promoting blood flow group C, group D-APS model group and group E--blank control group.From the first day ofpregnancy, the rats in group A were given Dan Shou Tang through intragastric administration;the rats in group B were given formula to nourish kidney and rats in group C were given formula to promote blood flow, while group D and group E, as the control were given the corresponding physiological brine through intragastric administration.Then on the eighth and the twelfth day ofpregnancy, all rats ofgroup A, B, C and D were given multi-site subcutaneous injecting ofpurified ACA-IgG or LA-IgG with a dosage of15mg/ml, fats ofgroup E were injected the corresponding physiological brine.On the 15th day ofpregnancy, the rats were killed for samples.Results:Compared with APS model group, fetal absorptivity, ACA and APTT level were dramatically decreased in group A, B and C(P
