ABSTRACT

Objective To investigate the relationship between the changes of plasma thromboxane B2 (TXB2),6-keto-prostaglandin 1 α (6k-PGF1 α) and positive platelet α-granule membrane glycoprotein (CD62P) in patients with acute cerebral infarction.Methods 160 patients with acute cerebral infarction (case group) and 80 healthy subjects were enrolled in our hospital from August 2016 to August 2018.The plasma levels of 6k-PGF1α,CD62P and TXB2 were measured and analyzed.Subgroup analysis was performed on patients with cerebral infarction with different trial of org 10172 in acute stroke treatment (TOAST) classification,National Institute of Health Stroke Scale (NIHSS) score,and prognosis outcome.Results The plasma levels of 6k-PGF1α,CD62P and TXB2 in the case group were significantly higher than those in the control group (P < 0.05).The plasma levels of 6k-PGF1 α,CD62P and TXB2 in mild,moderate and severe groups were gradually increased (P < 0.05).The plasma levels of 6k-PGF1 α,CD62P and TXB2 in patients with small infarction,mid-infarction and large infarction were also gradually increased,with statistically significant difference (P < 0.05);plasma 6k-PGF1 α,CD62P,TXB2 levels in patients with good prognosis were significantly lower than those in poor prognosis group (P < 0.05).Conclusions The levels of plasma TXB2,6k-PGF1α and CD62P in patients with acute cerebral infarction are elevated,and are closely related to the patient's condition and prognosis.

ABSTRACT

Objective To evaluate the efficacy of Tenghuang-Jiangu tablet combined with osteopeptide injection in the treatment of lumbar hyperosteogeny. Methods A total of 106 patients with lumbar hyperosteogeny were randomly divided into two groups, 53 in each group by digital table method. The control group was treated with osteopeptide injection, and the study group was treated with Tenghuang-Jiangu tablet on the basis of the control group. Both groups were treated for 45 days. The traditional Chinese medcine (TCM) symptoms were scored from pain, numbness, swelling, flexion and extension before and after treatment. The levels of thromboxane B2 (TXB2) and 6-ketone prostaglandin Flα ( 6-Keto-PGFlα ) in serum were detected by ELISA. Adverse reactions during treatment were recorded. Results The total effective rate was 86.8% (46/53) in the study group and 64.2% (34/53) in the control group. There were significant differences between the two groups (χ2=7.338, P=0.007). After treatment, the serum TXB2 (128.01 ± 23.68 pg/ml vs. 172.26 ± 19.33 pg/ml, t=10.539) level in the study group was significantly lower than control group (P<0.01); the serum 6-Keto-PGFlα (36.84 ± 4.96 pg/ml vs. 25.44 ± 4.21 pg/ml, t=12.757) level was significantly higher than control group (P<0.01). After treatment, the scores of pain, numbness, swelling, flexion and extension in the study group were significantly lower than those in the control group (t=10.061, 11.449, 14.921, 15.527, P<0.01). During the treatment period, the incidence of adverse reactions was 17.0% (9/53) in the control group and 22.6% (12/53) in the study group. There was no significant difference between the two groups (χ2=0.534, P=0.465). Conclusions The Tenghuang-Jiangu tablet combined with osteopeptide injection can improve the clinical symptoms of patients with lumbar hyperosteogeny, and regulate the serum levels of TXB2 and 6-Keto-PGFlα.

ABSTRACT

Objective To investigate the effect of targeted interruption of cyclooxygenase-2 (COX-2) gene by small interference RNA (siRNA) on the expression of COX-2, vascular endothelial growth factor (VEGF) and matrix metalloproteinase-9 (MMP-9) in eutopic and ectopic endometrial stromal cells (ESC) with endometriosis, and the effect on the content of 6-keto-prostaglandin-F1α (6-keto-PGF1α, metabolites of COX) and the apoptosis of eutopic and ectopic ESC with endometriosis. Methods Ectopic and eutopic ESC from 30 women with endometriosis were isolated and cultured respectively. Then, ESC were classified into three groups: interference group, negative control group and blank control group. ESC in interference group were injected into siRNA transfection complex while ESC in negative control group were injected into negative control transfection complex. ESC from 10 participants without endometriosis were the normal control group. The mRNA and protein expression of COX-2, VEGF, MMP-9 in pre-transfected and post-transfected eutopic and ectopic ESC were detected through real time reverse transcription PCR and western blot. The content of 6-keto-PGF1α was determined by ELISA, the apoptotic cells were detected by flow cytometry. Results After interruption of COX-2 gene, there were no significant difference in the mRNA and protein expression of COX-2, VEGF and MMP-9 between the negative control group and blank control group (P>0.05); the mRNA and protein expression of the three genes in interference group were significantly lower than those in negative control group and blank control group (P0.05);compared with those of negative control group and blank control group, the content of 6-keto-PGF1αin interference group decreased significantly (P0.05). The results from flow cytometry displayed that, there was no significant difference in apoptotic cells between the negative control group and blank control group (P>0.05);compared with those of negative control group and blank control group, more apoptotic cells were detected in interference group and the difference was significant (P<0.01);the apoptotic cells in ectopic ESC were significantly more than that in eutopic ESC (P<0.05); the apoptosis rate in interference group of eutopic and ectopic ESC were (33.76 ± 0.06)%, (47.18 ± 0.12)%. Conclusions Our results suggested the targeted interruption of COX-2 gene by siRNA effectively inhibited the mRNA and protein expression of COX-2, VEGF and MMP-9 in both eutopic ESC and ectopic ESC with endometriosis, greatly increased the apoptotic rate of cells and obviously reduced the content of 6-keto-PGF1αby inhibiting the activity of COX-2. And the changes in ectopic endometrium were more evident than those in eutopic endometrium.

ABSTRACT

Objective To explore the regularity of changes of plasma endothelin (ET), throm-boxame B2(TXB2) and 6-keto-prostaglandin F1α(6-keto-PGF1α) and restenosis after percutaneous transluminal angioplasty (PTCA) in patients with coronary heart disease. Methods Radioimmunoas-say was applied to measuring plasma levels of ET, TXB2and 6-keto-PGF1αat 0,30 min, 1 day and 3 days after PTCA in 41 patients with coronary heart disease. Results The level of ET in the patients with coronary heart disease was significantly decreased in 30 minutes after PTCA (P<0.05), but no significant difference was observed in 1 day and 3 days after PTCA (P>0.05). The level of plasma TXB2has no statistical difference after PTCA in 30minutes, 1 day and 3 days (P>0.05). The level of 6-keto-prostaglandin F1α(6-keto-PGF1α) of the patients with coronary heart disease was significantly declined in 30 minutes after PTCA (P<0.05) ,but no significant difference was observed in 1 day and 3 days after PTCA (P>0.05). Conclusion PTCA may lead to fluctuation of plasma levels of ET, TXB2and 6-keto-PGF1αThe clarification of changing regularity of these vasoactive substances contrib-utes to prevention of acute artery occlusion or restenosis after PTCA.

ABSTRACT

Objective To study the effects and clinical significance of plasma thromboxane B2(TXB2), platelet alpha granule membrance protein-140(GMP-140), 6-Keto-PGF1?(6-K-PGF1?), endothelins(ET), tissue type plasminogen activator(tPA) and plaminogen activator inhibitor-1(PAI-1) in patients with cerebral arteriosclerisis and cerebral thrombosis. Methods Plasma TXB2 ,GMP-140,6-K-PGFla and ET levels were determined by ra-dioimmuno-assay. Plasma tPA and PA1 were determined by the chromogenic peptide substrate method in controls and in patients with cerebral arteriosclerisis and cerebral thrombosis. Results The levels of plasma TXB2 and GMP-140 were significantly higher in patients with cerebral arteriosclerisis and cerebral thrombosis than in the control group(P0.05) .The levels of tPA and tPA/PAI were significantly lower in patients with cerebral arteriosclerisis and cerebral thrombosis than in controls. Although the PAI level increased in patients with cerebral arteriosclerisis and cerebral thrombosis,no difference was found in statistics. Conclusions Activation of platelets, injury of endothelial cell and decreased activity of fibrinolysis in patients with cerebrovascular disease may be involved in the development of these diseases. Appropriate treatment should be adopted on the above abnormality.

To study the clinical significance of the changes in PDGF and 6 keto PGF 1? in patients before and after cutting balloon angioplasty (CBA) and plain conventional balloon angioplasty (POBA).
