ABSTRACT

OBJETIVO: Analisar os achados citológicos de mulheres detectadas com adenocarcinoma do colo do útero, levando em conta o histórico da paciente no ano que antecedeu ao diagnóstico e a histopatologia das lesões. MÉTODOS: Este é um estudo comparativo, retrospectivo conduzido com dados de mulheres com adenocarcinoma ou com carcinoma escamoso do colo do útero detectados entre 2002 e 2008. Os laudos da citologia foram sintetizados de acordo com a terminologia Bethesda revisada em 2001 e foram comparados com a histopatologia de adenocarcinoma e de carcinoma escamoso. Foram verificadas as distribuições dos achados citológicos, a concordância global e corrigida pelo acaso com o uso do coeficiente Kappa de Cohen. Para isso, as alterações citológicas foram agregadas de acordo com a origem epitelial, formando os grupos de células glandulares e de células escamosas, tendo como padrão ouro os grupos de tumor histopatologicamente confirmados (adenocarcinoma versus carcinoma escamoso). RESULTADOS: No período, 284 casos de câncer do colo uterino foram diagnosticados. Os casos efetivamente estudados compreenderam 27 e 54 pacientes com adenocarcinoma e com carcinoma escamoso, respectivamente. O grupo de adenocarcinoma representou 9,5% do total diagnosticado, com 56% das mulheres com idade inferior a 50 anos. A coleta da citologia foi feita em média 92 dias antes do diagnóstico do câncer (variação: 19 dias a 310 dias). Em 41,6% dos casos, a citologia que precedeu o diagnóstico do adenocarcinoma foi indicativa de alterações glandulares do tipo adenocarcinoma e atipias de células glandulares. A concordância simples foi de 73,7% e o coeficiente Kappa de 48,7%, sugerindo moderada concordância. CONCLUSÃO: Nesta população, a citologia teve um importante papel no rastreio de mulheres ...


PURPOSE: To analyze the cytological findings of women with cervical adenocarcinoma, taking into account the patient's history in the year prior to diagnosis and the histopathological aspects of the lesions. METHODS: A retrospective comparative study was conducted using data from women with cervical adenocarcinoma or squamous carcinoma detected between 2002 and 2008. The cytological reports were synthesized according to the Bethesda System revised in 2001 and were compared to the histopathological findings of cervical adenocarcinoma and squamous carcinoma. The distributions of cytological findings were calculated, as well as the global agreement and chance-corrected agreement using the Cohen's Kappa Coefficient. For this purpose, the cytological findings were grouped according to the epithelial origin, forming the glandular cell and squamous cell groups, with the histopathologically confirmed tumor types (adenocarcinoma versus squamous carcinoma) being used as the gold standard. RESULTS: A total of 284 cases of cervical cancer were diagnosed during the study period. The effectively studied cases were 27 and 54 patients with adenocarcinoma and squamous carcinoma, respectively. The adenocarcinoma group represented 9.5% of the total cases diagnosed, and 56.0% of the women in this group were younger than 50 years. Cervical cytology was collected on average 92 days before the cancer diagnosis (range: 19 days to 310 days). In 41.6% of cases the cytological results were consistent with glandular alterations such as adenocarcinoma cells or atypical glandular cells. The global agreement and Cohen's Kappa Coefficient were 73.7 and 48.7%, suggesting substantial and moderate agreement, respectively. CONCLUSIONS: In this population, the cytological smears had an important role in screening women with adenocarcinoma, although some of them were referred to clarify the clinical symptoms. ABSTRACT

Objective To investigate the biological significance of differentially expressed proteins from human primary lung adenocarcinoma with lymph node metastasis adenocarcinoma (LNM AdC) and without metastasis (non-LNM AdC) according to clinical diagnosis of lymph node metastasis and distant metastasis,with bioinformatics approach.Methods Cytoscape software was used to analyze a functional enrichment analysis and a protein-protein interaction network from differentially expressed proteins from LNM AdC and non-LNM AdC.Results The top biological processes were related to glucose catabolic process,hexose catabolic process,monosaccharide catabolic process,alcohol catabolic process,and cellular carbohydrate catabolic process.The top molecular functions were related to phospholipase inhibitor activity,lipase inhibitor activity,calcium-dependent phospholipid binding,phosphlipase A2 inhibitor activity,and lipid binding.A protein-protein interaction network of differentially expressed proteins was generated with literature data.Conclusions This bioinformatics analysis demonstrated that glucose catabolic process,alcohol catabolic process,calcium-dependent phospholipid binding,phosphlipase A2 inhibitor activity,ACTB,ANXA1,ANXA2,ANXA3,VCP,NPM1,KRT1,and SUMO4 are significantly associated with a lung adenocarcinoma.These network data provide new insights into the metastasis mechanisms of human lung adenocarcinoma.

ABSTRACT

OBJETIVOS: Caracterizar o perfil de imunoexpressão de MLH1, MSH2, MSH6 e PMS2 em adenocarcinomas gástricos, explorando seu desempenho na identificação de características clínicas e patológicas, bem como sua influência prognóstica isolada e em relação aos demais parâmetros. MÉTODOS: Cento e trinta e três casos de adenocarcinomas gástricos esporádicos localmente avançados (pT2a ou mais) operados no Hospital das Clínicas/FMUSP foram incluídos pela ausência de metástases a distância ao diagnóstico (M0) e caracterizados clínica (idade, sexo e sobrevida) e patologicamente (tamanho, local, tipo de Borrmann, tipo histológico, infiltração vascular, perineural e variáveis de estadiamento locorregional). Amostras de 1,0 mm foram dispostas em micromatrizes teciduais (TMA) para pesquisa imuno-histoquímica das enzimas MLH1, MSH2, MSH6 e PMS2, com detecção por sistema de polímeros curtos conjugados a peroxidase. Casos com resultados negativos ou incertos nas amostras de TMA tiveram repetidas as reações em secções convencionais. A associação entre o estado de expressão dos marcadores com variáveis clínicopatológicas foi avaliada através do teste do qui-quadrado ou exato de Fisher. O impacto dos parâmetros clínico-patológicos e estado de expressão das enzimas na sobrevida foi explorado em modelos uni e multivariados de Cox, com construção de curvas de Kaplan-Meyer. Todas as análises estatísticas foram consideradas significativas ao nível de p<0,05. RESULTADOS: Quarenta e cinco casos (33,6%) exibiram perda de expressão de ao menos uma enzima, sendo frequente a perda de duas (9/45: 20%), três (14/45:31,2%) ou quatro enzimas (7/45:15,5%). Anormalidade mais frequente ocorreu com o MLH1 (26,7%), seguida de MSH6 (23%), PMS2 (21%) e MSH2 (20,8%). Quando avaliadas em conjunto, houve correlação entre o estado de expressão de todos os possíveis pares, com destaque para MLH1/PMS2 (rho=0,467, p<0,001) e MSH2/MSH6 (rho=0,666, p<0,001). OBJECTIVES: To characterize immunoexpression profile of MLH1, MSH2, MSH6 e PMS2 in gastric adenocarcinomas, exploring its performance to identify distinctive clinico-pathological features, as well as their prognostic implications in univariate and multivariate analyses. METHODS: A hundred and thirty three cases of locally advanced (pT2a or higher) sporadic gastric adenocarcinomas operated on Hospital das Clínicas/FMUSP were included due to absence of distant metastases at diagnosis (M0). Clinical (age, gender and survival) and pathological features (size, local, Borrmann´s type, histological type, vascular and perineural infiltration and locorregional staging parameters) were characterized. One millimeter samples were placed on tissue microarray blocks (TMA) and immunohistochemical detection of MLH1, MSH2, MSH6 and PMS2 performed on obtained sections, with revelation developed by peroxidase conjugated short-polymer based reagents. Negative or equivocal results obtained with TMA samples were repeated on conventional sections. Association between expression status for these markers and clinico-pathological features were evaluated by chisquare or Fisher´s exact test when appropriate. Impact of clinico-pathological features and expression status on disease specific survival were explored by Cox uni and multivariate models, with Kaplan-Meyer curves being fitted to illustrate these. All statistical results were considered significant at p<0,05. RESULTS: Forty five cases (33.6%) showed loss of at least one mismatchrepair enzyme, being frequent loss of two (9/45: 20%), three (14/45:31.2%) or even the four evaluated enzymes (7/45:15.5%). The most frequent abnormality addressed MLH1 (26.7%), followed by MSH6 (23%), PMS2 (21%) and MSH2 (20.8%). When analyses were performed in conjunction, correlation was identified for the expression status of all possible pairs, mainly the functional heterodimers MLH1/PMS2 (rho=0.467, p<0.001) and MSH2/MSH6 (rho=0.666, p<0.001)...

ABSTRACT

Objective To identify lung adenocarcinoma-associated antigens by using Serologic Proteome Analysis(SERPA),an approach which combined conventional proteome analysis with serological screening.Methods SERPA of four human lung adenocarcinoma tissues were performed.The Western blot imaging films which reacted with autologous patient serum and with control normal serum were obtained and the differential reacting protein spots were recognized.Results Well-resolved,reproducible 2-DE Western blot imaging films of human lung adenocarcinoma reacted with autologous patient sera and the control sera were obtained.Totally(27?5) differentially expressed proteins which only were reactive with lung adenocarcinoma patient sera were found.Some differentially expressed proteins were identified by peptide mass fingerprint(PMF).Some of the proteins were the products of oncogenes,and others were involved in the regulation of cell cycle and signal transduction.Conclusion These results will provide scientific foundation on screening the molecular biomarker for the diagnosis,treatment,and prognosis of the lung adenocarcinoma.
