ABSTRACT

Objective:To study the compatibility and stability of coenzyme A for injection, adenosine disodium triphosphate and inosine injection. Methods:By simulating the clinical medication, the three drugs and 5% glucose injection were mixed together. The contents and relative substances of coenzyme A, adenosine disodium triphosphate and inosine were measured by HPLC. The changes in appearance, pH and insoluble particles were observed or tested at ambient temperature. Results:The mixed solution showed no signifi-cant changes in appearance, pH, number of insoluble particles, contents and relative substances of coenzyme A, adenosine disodium triphosphate and inosine in 4 h, while the mixed solution became turbid and the pH, number of insoluble particles and contents of the three drugs showed significant changes after 24-h storage. Conclusion:The mixed solution of coenzyme A for injection, adenosine dis-odium triphosphate and inosine injection in 5% glucose injection should be used up in 4 h at ambient temperature.

ABSTRACT

OBJECTIVE: To establish bacterial endotoxin test for adenosine disodium triphosphate (ATP) raw material by TAL method. METHODS: The sensitivity of TAL and the limit of endotoxin were reviewed and confirmed. Preliminary interference test was used to determine the dilution ratio of samples and interference test was applied to determine maximum non-interference concentration. Bacterial endotoxin test was performed on 11 batches of samples according to the method stated in Chinese Pharmacopoeia (2005 edition). RESULTS: The sensitivity of TAL was up to the standard and the limit of endotoxin in samples were 2.0 EU?mg-1. The maximum non-interference concentration was 0.125 mg?mL-1. Results of bacterial endotoxins test of 11 batches of samples were all in line with the standard. CONCLUSIONS: It is feasible to detect the bacterial endotoxin of ATP raw material by TAL methods, which can replace pyrogen test.

ABSTRACT

Objective:Investigate whether adenosine disodium triphosphate for injection can reduce the systemic inflammatory response which induced by cardiopulmonary bypass.Methods:Forty-seven patients scheduled for cardiac surgery over 30 min following CPB were randomly divided into two groups. In treated group(n=23),adenosine disodium triphosphate for injection(2 mmol/L) was added into the prime during CPB, but in control group(n=24), no adenosine disodium triphosphate for injection was added. Blood samples were drawn from the radial artery at the following time points:(1)before CPB;(2)30 min after aortic clamping;(3)30 min after aortic declamping;(4) 12 h after aortic declamping;(5)24 h after aortic declamping.Enzyme-linked immunosorbent assays(ELISA) were used to measure the concentration of tumor necrosis factor-?(TNF-?), interleukin-6(IL-6), in plasma,CK-MB in plasma level were measured too.Results:The concentration of TNF-??IL-6?CK-MB levels in plasma level of two groups showed no difference before CPB.
